Dentre os tumores que acometem o sistema nervoso, o glioblastoma multiforme (GBM), destaca-se por seu alto grau de agressividade e baixo prognóstico, apresentando em média uma sobrevida de 15 meses a partir do diagnóstico. O presente estudo objetivou investigar a atividade antiproliferativa e antineoplásica de quatro flavonoides isolados da espécie Brosimum acutifolium (Huber), duas flavanas: 4’-hidroxi-7,8-(2”,2”-dimetilpirano) flavana (BAS-1) e 7,4’-dihidroxi-8,(3,3-dimetilalil)-flavana, (BAS-4); e duas chalconas: 4,2’-dihidroxi-3’,4’-(2”,2”-dimetilpirano)-chalcona (BAS-6) e 4,2’,4’-trihidroxi-3’-(3,3-dimetilalil)-chalcona (BAS-7), em glioblastoma C6 de rato in vitro. Nossos resultados mostraram boa atividade citotóxica para as flavanas (BAS-1, -4) e para a chalcona BAS-7, com IC50 menor que 100 μM em teste de viabilidade pelo MTT, já a chalcona BAS-6, não demonstrou atividade citotóxica nas concentrações testadas. Estes flavonoides mostram ser menos citotóxico para célula não neoplásica (glia), com grau de segurança maior para a BAS-4 e BAS-7, uma vez que apresentaram menor efeito citotóxico à célula não neoplásica e menores índices hemolíticos. A análise de migração celular mostrou que o tratamento com BAS-1, BAS-4 e BAS-7 em baixas concentrações foi efetivo em promover inibição da migração celular. Estes três flavonoides também foram muito promissores em inibir a formação e o crescimento de colônia, além de promover parada no ciclo celular, com substancial aumento na população SubG0 para o tratamento com BAS-1 e BAS-4 com 100 μM. As flavanas BAS-1 e BAS-4 também mostraram maior capacidade de promover a perda na integridade do potencial de membrana mitocondrial (ΔΨm) e aumento para marcação com anexina V, indicativo de que estas drogas promovem morte por apoptose. No entanto a análise por microscopia eletrônica demonstrou marcantemente no tratamento com a BAS-4 a presença de vacúolos autofágicos, sugestivo que o processo de morte neste tratamento ocorre tanto por apoptose quanto autofagia. Com base nestes resultados pode-se concluir que dos flavonoides testados a BAS-1, BAS-4 e BAS-7 possuem potencial como agente antineoplásico na terapia do GBM, sendo a BAS-4 a mais promissora de todas.

Neuroblastoma é a neoplasia mais frequentemente diagnosticada na infância. O termo é comumente usado para se referir a uma ampla variedade de tumores neuroblásticos, incluindo os neuroblastomas, ganglioneuroblastomas e ganglioneuromas. Estimativas mostram que 8 milhões de crianças até 15 anos de idade por ano são atingidas por esta neoplasia, onde 80% dos casos são acometidos em até 4 anos de idade, o tumor é derivado de células malignas embrionárias advindas de células neuronais primordiais, desde gânglios simpáticos até medula adrenal e outros pontos. Neste estudo, foi avaliado o potencial citotóxico do composto 4,2´,3´,4´ tetrametoxi chalcona em modelo in vitro de neuroblastoma B103 de rato. Foram preparadas soluções estoques da droga a 50mM em dimetilsulfóxido (DMSO) e armazenadas a -20ºC para o preparo de novas concentrações (150μM, 100 μM, 75 μM e 50 μM). A viabilidade celular foi testada a partir de cultura de células da glia do córtex de rato e de neuroblastoma b103. Ensaios de migração celular e formação de colônias também foram realizados. Para a análise estatística foi realizado a análise de variância um critério (ANOVA) seguido pelo teste de Tukey, utilizando-se o programa BioEstat 5.0. Na avaliação do efeito citotóxico das chalconas, foi observado que o tratamento com o composto 4,2`3`4´- tetrametoxi chalcona não demonstrou nenhum efeito citotóxico contra células normais do córtex de rato para as concentrações testadas, enquanto que em culturas de células de neuroblastoma B103 foi demonstrado que esta droga promove a morte celular de forma significativa.

The length of the post-partum anoestrous interval affects reproductive efficiency in many tropical beef cattle herds. In this study, results from genome-wide association studies (Experiment 1: GWAS) and gene expression (Experiment 2: microarray) were combined in a systems approach to reveal genetic markers, genes and pathways underlying the physiology of post-partum anoestrus in tropically adapted cattle. The microarray study measured the expression of 13,964 genes in the hypothalamus of Brahman cows. A total of 366 genes were differentially expressed (DE) in the post-partum period, when acyclic cows were compared to cows that had resumed ovarian cycles. Associated markers (P < 0.05) from a high density GWAS pointed to 2829 genes that were associated with post-partum anoestrous interval (PPAI) in two populations of beef cattle: Brahman and Tropical composite. Together the experiments provided evidence for 63 genes that are likely to influence the resumption of ovulation post-partum in tropically adapted beef cattle. Functional annotation analysis revealed that some of the 63 genes have known roles in hormonal activity, energy balance and neuronal synapse plasticity. Polymorphisms within candidate genes identified by this systems approach could have biological significance in post-partum anoestrus and help select Zebu (Bos indicus) influenced cattle with genetic potential for shorter post-partum anoestrus. In birds, neurons of the isthmo-optic nucleus (ION), as well as ''ectopic'' neurons, send axons to the retina, where they synapse on cells in the inner nuclear layer (INL). Previous work has shown that centrifugal axons can be divided into two anatomically distinct types depending on their mode of termination: either ''convergent'' or ''divergent'' (Ramon y Cajal, 1889; Maturana and Frenk, 1965). We show that cytochrome-oxidase histochemistry specifically labels ''convergent'' centrifugal axons and target neurons which appear to be amacrine cells, as well as three ''types'' of ganglion cells: two types found in the INL (displaced ganglion cells) and one in the ganglion cell layer. Labeled target amacrine cells have distinct darkly labeled ''nests'' of boutons enveloping the somas, are associated with labeled centrifugal fibers, and are confined to central retina. Lesions of the isthmo-optic tract abolish the cytochrome-oxidase labeling in the centrifugal axons and in the target amacrine cells but not in the ganglion cells. Cytochromeoxidase-labeled ganglion cells in the INL are large; one type is oval and similar to the classical displaced ganglion cells of Dogiel, which have been reported to receive centrifugal input; the other type is rounder. Rhodamine beads injected into the accessory optic system results in retrograde label in both types of cells, showing that two distinct types of displaced ganglion cells project to the accessory optic system in chickens. The ganglion cells in the ganglion cell layer that label for cytochrome oxidase also project to the accessory optic system. These have proximal dendrites that ramify in the outer inner plexiform layer. Neither the target amacrine cells nor either of the displaced ganglion cells are immunoreactive for the inhibitory transmitter gamma aminobutyric acid. At least some of the target amacrine cells may, however, be cholinoceptive: we found that the antibody to the alpha-7 subunit of the nicotinic ACh receptor labels a population of cells in the INL that are similar in location, size, and the presence of labeled bouton-like structures to those we find labeled with cytochrome oxidase. This antibody also labels neurons in the ION proper but not ectopic cells. In conclusion, it appears that cytochrome oxidase may be a marker for ''convergent'' centrifugal axons and at least one of their target cells in the INL.

The present paper introduces a new model of fuzzy neuron, one which increases the computational power of the artificial neuron, turning it also into a symbolic processing device. This model proposes the synapsis to be symbolically and numerically defined, by means of the assignment of tokens to the presynaptic and postsynaptic neurons. The matching or concatenation compatibility between these tokens is used to decided about the possible connections among neurons of a given net. The strength of the compatible synapsis is made dependent on the amount of the available presynaptic and post synaptic tokens. The symbolic and numeric processing capacity of the new fuzzy neuron is used here to build a neural net (JARGON) to disclose the existing knowledge in natural language data bases such as medical files, set of interviews, and reports about engineering operations.

In the LTP (Long-Term Potentiation) model of learning and memory formation, elaborated in the context of molecular neurobiology, the opening of NMDA (N-Metyl-D-Aspartate) channels to entry of calcium ions into the post-synaptic neuron depends on two excitatory events: the repeated activation of the post-synaptic neuron by only one pre-synaptic neuron (Alternative 1), or its concomitant activation by two or more pre-synaptic neurons (Alternative 2). With the purpose of testing these alternatives, in the context of Cognitive Psychology, we presented to 73 university students a sequence of slides, with the duration of 6 seconds each, containing sentences (one for each slide) considered as being relevant or irrelevant for the subjects. Relevant sentences (R1) were presented only one time, while irrelevant ones were divided in three groups: the first one with sentences presented only one time (I1), the second with sentences presented three times (I3) and the third with sentences presented five times (I5). We conjectured that relevant sentences presented only one time would mobilize two or more brain excitatory pathways (corresponding to Alternative 2 above), while repeated irrelevant sentences would progressively activate the same sensory pathway. After the presentation of the sentences, the subjects answered a written questionnaire with questions about each presented sentence. The results indicate a prevalence of correct answers to R1 over I1, I3 and I5, suggesting that the relevance factor has greater weight than repetition in the induction of declarative memories

To assess the influence of anatomic location on the relationship between computed tomography (CT) number and X-ray attenuation in limited and medium field-of-view (FOV) scans. Materials and Methods Tubes containing solutions with different concentrations of K2HPO4 were placed in the tooth sockets of a human head phantom. Cone-beam computed tomography (CBCT) scans were acquired, and CT numbers of the K2HPO4 solutions were measured. The relationship between CT number and K2HPO4 concentration was examined by linear regression analyses. Then, the variation in CT number according to anatomic location was examined. Results The relationship between K2HPO4 concentration and CT number was strongly linear. The slopes of the linear regressions for the limited FOVs were almost 2-fold lower than those for the medium FOVs. The absolute CT number differed between imaging protocols and anatomic locations. Conclusion There is a strong linear relationship between X-ray attenuation and CT number. The specific imaging protocol and anatomic location of the object strongly influence this relationship.

The aim of this work was to analyze the neuron morphology and morphometry of cervical, thoracic and lumbar areas of nonsymptomatic seropositive dogs’ spinal cord for toxoplasmosis. Twenty indefinite-breed adult dogs were used; ten dogs were healthy, with negative serology for toxoplasmosis, and were used as the control group (group 1), and ten dogs were nonsymptomatic but seropositive for toxoplasmosis (group 2). After the microtomy, with interval of 100 micrometers (µm), the histological 5-µm-thick cuts were dyed by hematoxylin-eosin and Masson's trichrome techniques. The glass slides were analyzed under light microscope to examine the neuron morphology. The parameters considered for the morphometric analysis were area, perimeter, maximum diameter, minimum diameter and shape factor of cytoplasm and nucleus of neuron. The results were statistically analyzed by Student’s t test at 5% probability level. The morphological characteristics between the two groups were similar and according to literature. The morphometric results showed that there were changes in neurons size and structure, and increase and loss of star shape were noticed in seropositive animals. The results suggest that the neurons of these dogs, yet nonsymptomatic, can have lost their conductor function.
