Based on the multidomain structure of Pseudomonas aeruginosa exotoxin A, a fusion protein termed rPEA has been constructed, which is expected to serve as a gene carrier in vitro. The expression and purification of rPEA are described. The basal properties of rPEA as a gene carrier are evaluated by investigating its interaction with plasmid DNA and mimic biomembrane by surface plasmon resonance (SPR) and electrochemical methods. rPEA is proved to be able to bind with plasmid DNA with high affinity. It can also interact with lipid membrane and increase permeability of the membrane, so the probe molecules can easily reach the gold surface and exhibit the electrochemical response.

Neuromelanin is a complex polymer pigment found primarily in the dopaminergic neurons of the human substantia nigra, whose composition is complex including production of dopamine auto-oxidation, glutathione and a variety of amino acid. Neuromelanin forms stable complex with iron (111). We observed that 5,6-dihydroxyindole and its ramification possessed strong ability of chelating iron (111), and they are the production of dopamine auto-oxidation under physiological pH condition. In the present Of L-Cysteine, the relative yields of electrochemical oxidation of dopamine also had strong ability of chelating iron (111). The experimental results suggest that 5,6-dihydroxyindole and 5-S-cysteineldopamine play important roles in the process of synthetic neuromelanin chelating iron (111).

Single crystal of butyl branched polyethylene with various molecular weight formed from the melt in the presence of electric field was studied. It was found that electric field influenced morphology and structure of the butyl branched polyethylene single crystals formed. The lateral habits of the single crystals were circular shape, which was different from truncated lozenge or lenticular shape single crystals formed from the melt in the absence of electric field. The stems in the single crystals formed in the presence Of electric field were perpendicular to the basal plane of the single crystals, which was different from chain tilting in single crystals formed from the melt in the absence of electric field. The electron diffraction patterns showed that the structure of the circular single crystals was a quasi-hexagonal with looser chain packing. This looser chain packing was favorable to thickening growth of single crystals through chain sliding diffusion. The thickness of the single crystals was much larger and depended on molecular weight. It indicated that the single crystal in the presence of electric field should be an extended chain type Single crystal.

Platinum nanoparticles were electrocrystallized on a 4-aminophenyl monolayer-grafted carbon substrate. These Pt-modified surfaces were characterized by scanning tunneling microscopy (STM). The characterization by STM revealed that the platinum nanoparticles obtained had good size monodispersity and were well separated from one another on HOPG surfaces. (C) 2000 Elsevier Science S.A. Polyaniline nanoparticles were prepared on a highly oriented pyrolytic graphite (HOPG) surface from dilute polyaniline acidic solution (1 mM aniline + 1 M HClO4) using a pulsed potentiostatic method. Electrochemistry, Fourier transform infrared external reflection spectroscopy (FT-IR-ERS), X-ray photoelectron spectroscopy (XPS) and tapping-mode atomic force microscopy (TMAFM) were: used to characterize the composition and structure of the polyaniline nanoparticles. FT-IR-ERS and XPS results revealed that the polyaniline was in its emeraldine form. TMAFM measurement showed that the electropolymerized polyaniline nanoparticles dispersed on the:HOPG surface with a coverage of about 10(10) cm(-2). These nanoparticles were disk-shaped having a height of 10(-30) Angstrom and an apparent diameter varying from 200 to 600 Angstrom. The particle dimensions increased with the electropolymerization charge (Q) over the interval from 5.7 to 19.3 mu C cm(-2) (C) 2000 Elsevier Science S.A. All rights reserved.

In this paper, we present a new method of fabricating metal nanoparticles on carbon substrates through molecular design. Scanning tunneling microscopy measurements show that the electrochemically synthesized Ag nanoparticles are homogeneously dispersed on the modified highly oriented pyrolytic graphite (HOPG) surface with a narrow particle size distribution. Moreover, the size and number density of Ag nanoparticles on the grafted HOPG surface can be easily changed through control of the experimental conditions. (C) 2001 Elsevier Science B.V. Effects of La3+ and Gd3+ on Ca2+ influx were investigated in rat hepatoma H-35 cells by measuring the initial rate of Ca-45(2+) uptake. It was found that the maximum initial rate of Ca2+ uptake was increased six- to ten-fold at low concentrations of La3+ and Gd3+. Kinetic analyses by measuring the initial rate of Ca2+ influx at different external Ca2+ concentrations indicated the existence of two intracellular exchangeable components in the basal Ca2+ system, with low and high affinities for Ca2+, and only one class of Ca2+ binding sites was observed in the La3+- or Gd3+-treated cells. For high affinity, La3+ and Gc(3+) increased both kinetic parameters K-m and V-max of basal Ca2+ influx. La3+ and Gd3+ compete directly with Ca2+ for Ca2+ binding site for low affinity. The kinetics is competitive.

Highly oriented pyrolytic graphite (HOPG) is the substrate often used in scanning tunneling microscopy (STM). It is well known that STM images of the basal plane of HOPG show some unusual structural patterns. In this letter, we present in situ STM images of some unusual features on HOPG in solutions, including normal or abnormal chain-like features and hexagonal or oblique superperiodic structures. These features emerge both next to and apart from the step of HOPG.

The surface topography of highly oriented pyrolytic graphite (HOPG) which has been subjected to anodized treatment at a low potential (less positive) has been exhaustively studied using scanning tunneling microscopy (STM). Characterization of graphite surface has revealed that a small percentage of the surface (5%-10%) shows superperiodic features. In this case, the typical topographical features are triangular networks with the observed periodicities in the plane ranging from approximately 26 to 240 nm with peak-to-peak amplitudes out of the plane extending from 7 to 35 Angstrom. We show that this triangular network can be attributed to the injection of electrolyte and solvent in the earliest stages of oxidation of the basal HOPG electrode surface. A simple model is proposed to explain this phenomenon.

Prussian blue has been formed by cyclic voltammetry onto the basal pyrolytic graphite surface to prepare a chemically modified electrode which provides excellent electrocatalysis for both oxidation and reduction of hydrogen peroxide. It is found for the first time that glucose oxidase or D-amino oxidase can be incorporated into a Prussian blue film during its electrochemical growth process. Two amperometric biosensors were fabricated by electrochemical codeposition, and the resulting sensors were protected by coverage with a thin film of Nafion. The influence of various experimental conditions was examined for optimum analytical performance. The glucose sensor responds rapidly to substrates with a detection limit of 2 x 10(-6) M and a linear concentration range of 0.01-3 mM. There was no interference from 2 mM ascorbic acid or uric acid. Another (D-amino acid) sensor gave a detection limit of 3 x 10(-5) M D-alanine, injected with a linear concentration range of 7.0 x 10(-5)-1.4 x 10(-2) M. Glucose and D-amino acid sensors remain relatively stable for 20 and 15 days, respectively. There is no obvious interference from anion electroactive species due to a low operating potential and excellent permselectivity of Nafion.

The title complex was synthesized and characterized by H-1, C-13, Sn-119 NMR and IR spectra. A single crystal X-ray diffraction study confirmed its molecular structure and revealed that 3,4,5-trimethoxy-benzoyl salicylahydrazone was a tridentate and approximately planar ligand. The complex crystallizes in the triclinic space group P1BAR with a = 9.208(3), b = 12.536(2), c = 12.187(4) angstrom, alpha = 113.12(2), beta = 90.58(2), gamma = 81.42(2), V = 1277.5(6) angstrom, Z = 2. The structure was refined to R = 0.033 and R(w) = 0.041 for 3944 observed independent reflections. The tin atom has a distorted trigonal bipyramidal coordination. The Sn-C bond lengths are 2.129(5) and 2.113(5) angstrom (av. 2.121(5) angstrom), the C-Sn-C angle is 123.3(2); the bond length between the tin atom and the chelating nitrogen is 2.173(3) angstrom. Two chain carbon atoms and the chelating nitrogen atom occupy the basal plane. The skeleton of two erect oxygen atoms and the tin atom is bent (O-Sn-O angle = 153.5(1)). In the complex, the ligand exists in the enol-form.

The effects of three non-nutrient additives on nonspecific immunity and growth of juvenile turbot (Scophthalmus maximus L.) were studied in this feeding experiment. The five treatments are basal diet alone, basal diets containing three different additives [0.4 g kg(-1) of xylo-oligosaccharides (XOS), 1.3 g kg (-1) of yeast cell wall and 0.8 g kg (-1) of bile acids] individually or in combination. Two hundred and twenty-five turbots (average initial weight 151.3 +/- 11.3 g) were randomly allotted in five treatments with three replicates within each treatment in a 72-day period. Comparing with basal diet group, activities of C3, C4, phagocyte, lysozyme, specific growth rate and feed conversion rate in yeast cell wall, XOS and the combined groups was enhanced significantly (P < 0.05); however, these parameters in bile acid groups were increased slightly (P > 0.05) except for phagocyte (P < 0.05); superoxide dismutase activity in additive groups was not significantly increased (P > 0.05) except for the combined group (P < 0.05). In conclusion, supplementation of yeast cell wall and XOS enhanced the nonspecific immunity of juvenile turbot. Synergistic or additive effect of the three additives was not observed.

A field population of Ulva pseudocurvata Koeman et C. Hoek (hereafter termed Ulva) at Sylt Island (North Sea, Germany) exhibited biweekly peaks of gametophytic reproduction during the colder seasons and approximately weekly peaks during summer. The reproductive events lasted 1-5 d and were separated from each other by purely vegetative phases. Under constant conditions in the laboratory, a free-running rhythm was observed with reproductive peaks occurring approximately every 7 d. When artificial moonlight was provided every 4 weeks, fewer reproductive events occurred, and the reproductive rhythm became synchronized to the environmental artificial moonlight rhythm. In the laboratory, apical disks were entirely converted into reproductive tissue after 8 d cultivation, while almost all basal disks stayed vegetative, which prevented the entire loss of the vegetative thallus during reproductive events. Seasonal size reduction of the thallus occurred from late autumn onward and was determined to be controlled by a genuine photoperiodic response, since size reduction could be induced from May onward by experimental short-day (SD) treatment but was prevented in a long-day (LD) or night-break regime (NB). A daily fine-tuning occurred with gamete release early in the morning at the first sign of daylight, following an obligatory dark ("night") period of at least 1 h duration. No release took place if the overnight dark phase was replaced by continuous light. Blue, green, or red light all triggered gamete release after a dark phase at an irradiance of 0.1 mu mol photons . m(-2) .s(-1), while 0.001 mu mol photons . m(-2) . s(-1) was equivalent to a dark control.

Background: Red algae are primitive photosynthetic eukaryotes, whose spores are ideal subjects for studies of photosynthesis and development. Although the development of red alga spores has received considerable research attention, few studies have focused on the detailed morphological and photosynthetic changes that occur during the early development of tetraspores of Gracilaria vermiculophylla (Ohmi) Papenfuss (Gracilariales, Rhodophyta). Herein, we documented these changes in this species of red algae. Results: In the tetraspores, we observed two types of division, cruciate and zonate, and both could develop into multicellular bodies (disks). During the first 84 hours, tetraspores divided several times, but the diameter of the disks changed very little; thereafter, the diameter increased significantly. Scanning electron microscopy observations and analysis of histological sections revealed that the natural shape of the disk remains tapered over time, and the erect frond grows from the central protrusion of the disk. Cultivation of tissue from excised disks demonstrated that the central protrusion of the disk is essential for initiation of the erect frond. Photosynthetic (i.e., PSII) activities were measured using chlorophyll fluorescence analysis. The results indicated that freshly released tetraspores retained limited PSII photosynthetic capabilities; when the tetraspores attached to a substrate, those capabilities increased significantly. In the disk, the PSII activity of both marginal and central cells was similar, although some degree of morphological polarity was present; the PSII photosynthetic capabilities in young germling exhibited an apico-basal gradient. Conclusions: Attachment of tetraspores to a substrate significantly enhanced their PSII photosynthetic capabilities, and triggered further development. The central protrusion of the disk is the growth point, may have transfer of nutritive material with the marginal cells. Within the young germling, the hetero-distribution of PSII photosynthetic capabilities might be due to the differences in cell functions.

A molecular phylogeny is presented for the subfamily Dorippinae (including 9 individuals, representing 5 species and 4 genera), based on the sequence data from 16S rRNA gene. Two-cluster test between lineages in these phylogenetic trees has been performed. On the basis of rate constancy, the rate of nucleotide substitutions of 16S rDNA sequence data is estimated as 0.27% per million years. The analysis strongly supports the recognition of the Dorippinae as a monophyletic subfamily. Phylogenetic tree indicates that the subfamily Dorippinae is divided into two main clades, and genus Dorippe appears basal in the subfamily, diverging from other species 36.6 Ma ago. It is also clear that the Heikea is closely related to the genus Neodorippe. The divergence time between them is 15.8 Ma.
