Tidal stream turbines could have several direct impacts upon pursuit-diving seabirds foraging within tidal stream environments (mean horizontal current speeds > 2 ms−1), including collisions and displacement. Understanding how foraging seabirds respond to temporally variable but predictable hydrodynamic conditions immediately around devices could identify when interactions between seabirds and devices are most likely to occur; information which would quantify the magnitude of potential impacts, and also facilitate the development of suitable mitigation measures. This study uses shore-based observational surveys and Finite Volume Community Ocean Model outputs to test whether temporally predictable hydrodynamic conditions (horizontal current speeds, water elevation, turbulence) influenced the density of foraging black guillemots Cepphus grylle and European shags Phalacrocorax aristotelis in a tidal stream environment in Orkney, United Kingdom, during the breeding season. These species are particularly vulnerable to interactions with devices due to their tendency to exploit benthic and epi-benthic prey on or near the seabed. The density of both species decreased as a function of horizontal current speeds, whereas the density of black guillemots also decreased as a function of water elevation. These relationships could be linked to higher energetic costs of dives in particularly fast horizontal current speeds (>3 ms−1) and deeper water. Therefore, interactions between these species and moving components seem unlikely at particularly high horizontal current speeds. Combining this information, with that on the rotation rates of moving components at lower horizontal current speeds, could be used to assess collision risk in this site during breeding seasons. It is also likely that moderating any device operation during both lowest water elevation and lowest horizontal current speeds could reduce the risk of collisions for these species in this site during this season. The approaches used in this study could have useful applications within Environmental Impact Assessments, and should be considered when assessing and mitigating negative impacts from specific devices within development sites.

Although anti−cancer immuno−based combinatorial therapeutic approaches have shown promising results, efficient tumour eradication demands further intensification of anti−tumour immune response. With the emerging field of nanovaccinology, multi−walled carbon nanotubes (MWNTs) have manifested prominent potentials as tumour antigen nanocarriers. Nevertheless, the utilization of MWNTs in co−delivering antigen along with different types of immunoadjuvants to antigen presenting cells (APCs) has not been investigated yet. We hypothesized that harnessing MWNT for concurrent delivery of cytosine−phosphate−guanine oligodeoxynucleotide (CpG) and anti-CD40 Ig (αCD40), as immunoadjuvants, along with the model antigen ovalbumin (OVA) could potentiate immune response induced against OVA−expressing tumour cells. We initially investigated the effective method to co−deliver OVA and CpG using MWNT to the APC. Covalent conjugation of OVA and CpG prior to loading onto MWNTs markedly augmented the CpG−mediated adjuvanticity, as demonstrated by the significantly increased OVA−specific T cell responses in vitro and in C57BL/6 mice. αCD40 was then included as a second immunoadjuvant to further intensify the immune response. Immune response elicited in vitro and in vivo by OVA, CpG and αCD40 was significantly potentiated by their co−incorporation onto the MWNTs. Furthermore, MWNT remarkably improved the ability of co−loaded OVA, CpG and αCD40 in inhibiting the growth of OVA−expressing B16F10 melanoma cells in subcutaneous or lung pseudo−metastatic tumour models. Therefore, this study suggests that the utilization of MWNTs for the co−delivery of tumour−derived antigen, CpG and αCD40 could be a competent approach for efficient tumours eradication.

Stromules are motile extensions of the plastid envelope membrane, whose roles are not fully understood. They are present on all plastid types but are more common and extensive on non-green plastids that are sparsely distributed within the cell. During tomato fruit ripening, chloroplasts in the mesocarp tissue differentiate into chromoplasts and undergo major shifts in morphology. In order to understand what factors regulate stromule formation, we analysed stromule biogenesis in tobacco hypocotyls and in two distinct plastid populations in tomato mesocarp. We show that increases in stromule length and frequency are correlated with chromoplast differentiation, but only in one plastid population where the plastids are larger and less numerous. We used tobacco hypocotyls to confirm that stromule length increases as plastids become further apart, suggesting that stromules optimise the plastid-cytoplasm contact area. Furthermore, we demonstrate that ectopic chloroplast components decrease stromule formation on tomato fruit chromoplasts, whereas preventing chloroplast development leads to increased numbers of stromules. Inhibition of fruit ripening has a dramatic impact on plastid and stromule morphology, underlining that plastid differentiation status, and not cell type, is a significant factor in determining the extent of plastid stromules. By modifying the plastid surface area, we propose that stromules enhance the specific metabolic activities of plastids. Systematic low-temperature measurements of the thermal conductivity, specific heat, dielectric constant, and temperature-dependent ultrasound velocity have been made on a single piece of vitreous silica. These measurements were repeated after fast neutron irradiation of the material. It was found that the irradiation produced changes of the same relative magnitude in the low-temperature excess specific heat C , the thermal conductivity K, ex and the anomalous temperature dependence of the ultrasound velocity Deltav/v. A corresponding change in the temperature dependent dielectric constant was not observed. It is therefore likely that K and Deltav/v are determined by the same localized excitations responsible for C , but the temperature dependence of the dielectric constant may have a different, though possibly related, origin. Furthermore, a consistent account for the measured C , K, ex and Deltav/v of unirradiated silica is given by the tunneling-state model with a single, energy-dependent density of states. Changes in these three properties due to irradiation can be explained by altering only the density of tunneling states incorporated in the model.

Hydrodynamism is an environmental stressor on marine communities with effects on populations of intertidal organisms. Microphrys bicornutus is a crab species associated with secondary substrates on rocky shores and little is known about its population dynamics and its relation to the shore hydrodynamics. The aim of this study is to describe the population structure of Microphrys bicornutus on intertidal rocky shores from the north coast of the São Paulo State, Brazil, and the influence of wave exposure on the density of this decorator crab. There was a greater density of M. bicornutus on sheltered than on exposed rocky shores, which is probably due to the hydrodynamic influence on M. bicornutus populations on rocky shores: directly ( by the physical stress caused by wave impact) and indirectly ( by determining the dominance of the secondary substrate) acting on the density of these crabs. Our results also suggest a vertical distribution of juveniles and adults, where the juveniles might be more abundant on the infralittoral fringe and, thus, more influenced by hydrodynamism than adults in the infralittoral.

The vapor pressure of four liquid 1H,1H-perfluoroalcohols (CF3(CF2)n(CH2)OH, n ¼ 1, 2, 3, 4), often called odd-fluorotelomer alcohols, was measured as a function of temperature between 278 K and 328 K. Liquid densities were also measured for a temperature range between 278 K and 353 K. Molar enthalpies of vaporization were calculated from the experimental data. The results are compared with data from the literature for other perfluoroalcohols as well as with the equivalent hydrogenated alcohols. The results were modeled and interpreted using molecular dynamics simulations and the GC-SAFT-VR equation of state.

Suite à la rencontre d’un antigène (Ag) présenté à la surface des cellules présentatrice de l’Ag (CPA), les lymphocytes T naïfs, ayant un récepteur des cellules T (RCT) spécifique de l’Ag, vont proliférer et se différencier en LT effecteurs (1). Suite à l’élimination de l’Ag la majorité des LTe vont mourir par apoptose alors que les restants vont se différencier en LT mémoire (LTm) protégeant l’organisme à long terme. Les mécanismes qui permettent la différenciation des LTe en LTm sont encore inconnus. Pour comprendre comment les LTm CD8+ sont générés à partir des LTe, nous avons émis l’hypothèse que la densité de l’Ag présenté par les CPA peut avoir un impact sur la sélection des LT CD8+ répondant l’Ag à se différencier en LTm. De manière intéressante, nos résultats montrent qu’une immunisation avec des cellules dendritiques (DCs) exprimant un haut niveau de complexe CMH/peptide à sa surface permet le développement de LTm. À l’inverse, le développement des LTm est fortement réduit (10-20X) lorsque les souris sont immunisées avec des DCs exprimant un niveau faible de complexes CMH/peptide à leur surface. De plus, la quantité d’Ag n’a aucune influence ni sur l’expansion des LT CD8+ ni sur l’acquisition de leurs fonctions effectrices, mais affecte de manière critique la génération des LTm. Nos résultats suggèrent que le nombre de RCT engagé lors de la reconnaissance de l’Ag est important pour la formation des LTm. Pour cela nous avons observé par vidéo-microscopie le temps d’interaction entre des LTn et des DCs. Nos résultats montrent que le temps et la qualité de l’interaction sont dépendants de la densité d’Ag présenté par les DCs. Effectivement, nous observons une diminution dans le pourcentage de LT faisant une interaction prolongée avec les DCs quand le niveau d’Ag est faible. De plus, nous observons des variations de l’expression des facteurs de transcription clefs impliqués dans la différenciation des LTm tels qu’Eomes, Bcl-6 et Blimp-1. Par ailleurs, la densité d’Ag fait varier l’expression du Neuron-derived orphan nuclear receptor 1 (Nor-1). Nor-1 est impliqué dans la conversion de Bcl-2 en molécule pro-apoptotique et contribue à la mort par apoptose des LTe pendant la phase de contraction. Notre modèle propose que la densité de l’épitope contrôle la génération des CD8+ LTm. Une meilleure compréhension des mécanismes impliqués dans la génération des LTm permettra le développement de meilleures stratégies pour la génération de vaccin. Dans un second temps, nous avons évalué le rôle du signal RCT dans l’homéostasie des LTm. Pour ce faire, nous avons utilisé un modèle de souris transgénique pour le RCT dont son expression peut être modulée par un traitement à la tétracycline. Ce système nous a permis d’abolir l’expression du RCT à la surface des LTm. De manière intéressante, en absence de RCT exprimé, les LTm CD8+ peuvent survivre à long terme dans l’organisme et rester fonctionnels. De plus, une sous population des LTm CD4+ a la capacité de survivre sans RCT exprimé dans un hôte lymphopénique alors que l’autre sous population nécessite l’expression du RCT.

Antigen-kodierende RNA wird als eine sichere und effiziente Alternative zu traditionellen Impfstoff-Formulierungen, wie Peptid-, Protein-, rekombinanten viralen oder DNA basierten Impfstoffen betrachtet. Der endgültige klinische Nutzen RNA-basierter Impfstoffe wird von der Optimierung verschiedener Parameter abhängig sein, die zur Induktion und effizienten Expansion der humoralen und zellvermittelten Immunantwort beitragen. Vor diesem Hintergrund war die Zielsetzung der vorliegenden Arbeit, die Etablierung pharmakologischer und immunologischer Parameter für die Generierung effektiver Immunantworten durch RNA-Impfstoffe sowie deren Wirksamkeit in vitro und im Mausmodell unter Nutzung von Modellantigenen zu testen. Zur Untersuchung und Optimierung der RNA-Pharmakokinetik, als einem Schlüsselaspekt der klinischen Medikamentenentwicklung, wurde der Einfluss von strukturellen Modifikationen auf die Transkriptstabilität und Translationseffizienz von Reporter-Proteinen in einer zeitabhängigen Kinetik evaluiert. Es wurde gezeigt, dass ein poly(A) Schwanz von 120 Adenosinen, verglichen mit einem kürzeren, ein freies 3´ poly(A) Ende, verglichen mit einem verdeckten und eine doppelte β-globin 3´ UTR, unabhängig voneinander zu einer Erhöhung der IVT-RNA Stabilität und zu einer Verbesserung der Translationseffizienz beitrugen und dadurch insgesamt zu einer erhöhten Proteinexpression führten. Antigen-kodierende IVT-RNA mit diesen molekularen Merkmalen in Kombination führte, im Vergleich zur Standard IVT-RNA, zu einer erhöhten Dichte und Stabilität von Peptid/MHC-Komplexen auf der Zelloberfläche transfizierter DCs und dadurch zu einer verbesserten Stimulation von CD4+ und CD8+ T-Zellen im murinen und humanen System. Mit dem Ziel, die RNA kodierte Antigenform für die Induktion einer verstärkten Antikörperantwort zu modifizieren, wurde im zweiten Teil der Arbeit ein Antigen-IgM Fusionskonstrukt hergestellt und hinsichtlich seiner Eignung als neues Impfstoff-Format untersucht. Die Ausgangshypothese, dass die RNA kodierten Antigen-IgM Fusionsproteine polymerisieren, von transfizierten Zellen sezerniert werden und aufgrund der repetitiven Antigenstruktur im Vergleich mit dem monomeren Antigen zu einer Verstärkung der Antikörperantwort führen, wurde in vitro und in vivo im Mausmodell bestätigt. Die Entwicklung und Evaluierung von Zytokinfusionsproteinen zur selektiven Verstärkung der antigenspezifischen Immunantworten bildeten den dritten Schwerpunkt der vorliegenden Arbeit. Zur weiteren Verstärkung der Antikörperantwort wurde basierend auf den Resultaten aus dem zweiten Teil ein IL2-IgM Fusionskonstrukt hergestellt. Die Ko-Transfektion von Antigen-IgM und IL2-IgM kodierender IVT-RNA führte zu einer signifikant stärkeren Antikörperantwort als die Ko-Transfektion von Antigen-IgM und IL2. Für die Initiierung einer erfolgreichen anti-Tumor-Immunantwort ist das Priming antigenspezifischer T-Zellen essentiell. Um die Effizienz dieses Prozesses zu steigern, wurde ein bifunktionelles IL2-mCD40L Fusionskonstrukt hergestellt und sein Einfluss auf die Effektorfunktion von DCs in vitro und in vivo untersucht. Es wurde gezeigt, dass ein RNA kodiertes IL2-mCD40L Fusionsprotein als genetisches Adjuvanz zu einer Effizienzsteigerung des Priming zytotoxischer T-Zellen führt. Somit wurden in dieser Arbeit durch die Optimierung der Pharmakokinetik, die Modifikation der Antigenform und die Herstellung und Evaluierung von Zytokinfusionskonstrukten als genetische Adjuvantien, RNA-basierte Impfstoffe für eine optimierte Induktion von antigenspezifischen Immunantworten weiter verbessert.

Antibody orientation and its antigen binding efficiency at interface are of particular interest in many immunoassays and biosensor applications. In this paper, spectroscopic ellipsometry (SE), neutron reflection (NR), and dual polarization interferometry (DPI) have been used to investigate interfacial assembly of the antibody [mouse monoclonal anti-human prostate-specific antigen (anti-hPSA)] at the silicon oxide/water interface and subsequent antigen binding. It was found that the mass density of antibody adsorbed at the interface increased with solution concentration and adsorption time while the antigen binding efficiency showed a steady decline with increasing antibody amount at the interface over the concentration range studied. The amount of antigen bound to the interfacial immobilized antibody reached a maximum when the surface-adsorbed amount of antibody was around 1.5 mg/m(2). This phenomenon is well interpreted by the interfacial structural packing or crowding. NR revealed that the Y-shaped antibody laid flat on the interface at low surface mass density with a thickness around 40 Å, equivalent to the short axial length of the antibody molecule. The loose packing of the antibody within this range resulted in better antigen binding efficiency, while the subsequent increase of surface-adsorbed amount led to the crowding or overlapping of antibody fragments, hence reducing the antigen binding due to the steric hindrance. In situ studies of antigen binding by both NR and DPI demonstrated that the antigen inserted into the antibody layer rather than forming an additional layer on the top. Stability assaying revealed that the antibody immobilized at the silica surface remained stable and active over the monitoring period of 4 months. These results are useful in forming a general understanding of antibody interfacial behavior and particularly relevant to the control of their activity and stability in biosensor development.

Purpose: To evaluate the microvessel density by comparing the performance of anti-factor VIII-related antigen, anti-CD31 and, anti-CD34 monoclonal antibodies in breast cancer. Methods: Twenty-three postmenopausal women diagnosed with Stage II breast cancer submitted to definitive surgical treatment were evaluated. The monoclonal antibodies used were anti-factor VIII, anti-CD31 and anti-CD34. Microvessels were counted in the areas of highest microvessel density in ten random fields (200 x). The data were analyzed using the Kruskal-Wallis nonparametric test (p < 0.05). Results: Mean microvessel densities with anti-factor VIII, anti-CD31 and anti-CD34 were 4.16 +/- 0.38, 4.09 +/- 0.23 and 6.59 +/- 0.42, respectively. Microvessel density as assessed by anti-CD34 was significantly greater than that detected by anti-CD31 or anti-factor VIII (p < 0.0001). There was no statistically significant difference between anti-CD31 and anti-factor VIII (p = 0.4889). Conclusion: The density of stained microvessels was greater and staining was more intense with anti-CD34 compared to anti-CD31 and anti-factor VII-related antigen.
