The cores described in this report were taken on MIDPAC (Middle Pacific) Expedition in August-September 1950 by Scripps Institution of Oceanography from, the R/V Horizon. A total of 106 cores and dredges were recovered and are available at Scripps for sampling and study. The coring sites, all in the tropical central Pacific.

La resistencia al corte de las discontinuidades de un macizo rocoso es un tema que se ha venido estudiando desde años atrás por diversos autores. Adquiere una gran importancia, debido a que constituye la base de diseños de taludes, de obras subterráneas y de otras obras de ingeniería. Es por esto que se presenta esta tesis como un aporte a las investigaciones realizadas en este ámbito. Se basa en la comparación de cuatro (4) criterios de resistencia al corte conocidos: Patton (1966), Ladanyi y Archambault (1970), Barton & Choubey (1976) y Maksimovic (1996). Con los cuatro criterios mencionados, se analiza la influencia de cada parámetro dentro de cada uno. Además, de observar las diferencias que presentan al utilizar los mismos valores de los parámetros independientes correspondientes. Procediendo de esta manera, se pueda tener una base para saber cuáles son los parámetros que resultan determinantes dentro de cada criterio. Se aplican dichos criterios a un caso práctico. Posteriormente, se proporcionan las conclusiones para el momento de necesitar utilizar cada uno de los cuatro criterios que se estudian.

Nitrous oxide (N2O) is the main greenhouse gas (GHG) produced by agricultural soils due to microbial processes. The application of N fertilizers is associated with an increase of N2O losses. However, it is possible to mitigate these emissions by the introduction of adequate management practices (Snyder et al., 2009). Soil conservation practices (i.e.no tillage, NT) have recently become widespread because they promote several positive effects (increases in soil organic carbonand soil fertility, reduction of soil erosion, etc). In terms of GHG emissions, there is no consensus in the literature on the effects of tillage on N2O. Several studies found that NT can produce greater (Baggs et al., 2003), lower (Malhi et al., 2006) or similar (Grandey et al., 2006) N2O emissions compared to traditional tillage (TT). This large uncertainty is associated with the duration of tillage practices and climatic variability. Liming is widely use to solve problems of soil acidity (Al toxicity, yield penalties, etc). Several studies show a decrease in N2O emissions with liming (Barton et al., 2013) whereas no significant effects or increases were observed in others (Galbally et al., 2010). The aim of this work was to evaluate the effects of tillage (NT vs TT) and liming application or not of Ca-amendment) on N2O emissions from an acid soil during a rainfed crop.

The theoretical basis for evaluating shear strength in rock joints is presented and used to derive an equation that governs the relationship between tangential and normal stress on the joint during situations of slippage between the joint faces. The dependent variables include geometric dilatancy, the instantaneous friction angle, and a parameter that considers joint surface roughness. The effect roughness is studied, and the aforementioned formula is used to analyse joints under different conditions. A mathematical expression is deduced that explains Barton's value for the joint roughness coefficient (JRC) according to the roughness geometry. In particular, when the Hoek and Brown failure criterion is used for a rock in the contact with the surface roughness plane, it is possible to determine the shear strength of the joint as a function of the relationship between the uniaxial compressive strength of the wall with the normal stress acting on the wall. Finally, theoretical results obtained for the geometry of a three-dimensional joint are compared with those of the Barton's formulation

En España existen del orden de 1,300 grandes presas, de las cuales un 20% fueron construidas antes de los años 60. El hecho de que existan actualmente una gran cantidad de presas antiguas aún en operación, ha producido un creciente interés en reevaluar su seguridad empleando herramientas nuevas o modificadas que incorporan modelos de fallo teóricos más completos, conceptos geotécnicos más complejos y nuevas técnicas de evaluación de la seguridad. Una manera muy común de abordar el análisis de estabilidad de presas de gravedad es, por ejemplo, considerar el deslizamiento a través de la interfase presa-cimiento empleando el criterio de rotura lineal de Mohr-Coulomb, en donde la cohesión y el ángulo de rozamiento son los parámetros que definen la resistencia al corte de la superficie de contacto. Sin embargo la influencia de aspectos como la presencia de planos de debilidad en el macizo rocoso de cimentación; la influencia de otros criterios de rotura para la junta y para el macizo rocoso (ej. el criterio de rotura de Hoek-Brown); las deformaciones volumétricas que ocurren durante la deformación plástica en el fallo del macizo rocoso (i.e., influencia de la dilatancia) no son usualmente consideradas durante el diseño original de la presa. En este contexto, en la presente tesis doctoral se propone una metodología analítica para el análisis de la estabilidad al deslizamiento de presas de hormigón, considerando un mecanismo de fallo en la cimentación caracterizado por la presencia de una familia de discontinuidades. En particular, se considera la posibilidad de que exista una junta sub-horizontal, preexistente y persistente en el macizo rocoso de la cimentación, con una superficie potencial de fallo que se extiende a través del macizo rocoso. El coeficiente de seguridad es entonces estimado usando una combinación de las resistencias a lo largo de los planos de rotura, cuyas resistencias son evaluadas empleando los criterios de rotura no lineales de Barton y Choubey (1977) y Barton y Bandis (1990), a lo largo del plano de deslizamiento de la junta; y el criterio de rotura de Hoek y Brown (1980) en su versión generalizada (Hoek et al. 2002), a lo largo del macizo rocoso. La metodología propuesta también considera la influencia del comportamiento del macizo rocoso cuando este sigue una ley de flujo no asociada con ángulo de dilatancia constante (Hoek y Brown 1997). La nueva metodología analítica propuesta es usada para evaluar las condiciones de estabilidad empleando dos modelos: un modelo determinista y un modelo probabilista, cuyos resultados son el valor del coeficiente de seguridad y la probabilidad de fallo al deslizamiento, respectivamente. El modelo determinista, implementado en MATLAB, es validado usando soluciones numéricas calculadas mediante el método de las diferencias finitas, empleando el código FLAC 6.0. El modelo propuesto proporciona resultados que son bastante similares a aquellos calculados con FLAC; sin embargo, los costos computacionales de la formulación propuesta son significativamente menores, facilitando el análisis de sensibilidad de la influencia de los diferentes parámetros de entrada sobre la seguridad de la presa, de cuyos resultados se obtienen los parámetros que más peso tienen en la estabilidad al deslizamiento de la estructura, manifestándose además la influencia de la ley de flujo en la rotura del macizo rocoso. La probabilidad de fallo es obtenida empleando el método de fiabilidad de primer orden (First Order Reliability Method; FORM), y los resultados de FORM son posteriormente validados mediante simulaciones de Monte Carlo. Los resultados obtenidos mediante ambas metodologías demuestran que, para el caso no asociado, los valores de probabilidad de fallo se ajustan de manera satisfactoria a los obtenidos mediante las simulaciones de Monte Carlo. Los resultados del caso asociado no son tan buenos, ya que producen resultados con errores del 0.7% al 66%, en los que no obstante se obtiene una buena concordancia cuando los casos se encuentran en, o cerca de, la situación de equilibrio límite. La eficiencia computacional es la principal ventaja que ofrece el método FORM para el análisis de la estabilidad de presas de hormigón, a diferencia de las simulaciones de Monte Carlo (que requiere de al menos 4 horas por cada ejecución) FORM requiere tan solo de 1 a 3 minutos en cada ejecución. There are 1,300 large dams in Spain, 20% of which were built before 1960. The fact that there are still many old dams in operation has produced an interest of reevaluate their safety using new or updated tools that incorporate state-of-the-art failure modes, geotechnical concepts and new safety assessment techniques. For instance, for gravity dams one common design approach considers the sliding through the dam-foundation interface, using a simple linear Mohr-Coulomb failure criterion with constant friction angle and cohesion parameters. But the influence of aspects such as the persistence of joint sets in the rock mass below the dam foundation; of the influence of others failure criteria proposed for rock joint and rock masses (e.g. the Hoek-Brown criterion); or the volumetric strains that occur during plastic failure of rock masses (i.e., the influence of dilatancy) are often no considered during the original dam design. In this context, an analytical methodology is proposed herein to assess the sliding stability of concrete dams, considering an extended failure mechanism in its rock foundation, which is characterized by the presence of an inclined, and impersistent joint set. In particular, the possibility of a preexisting sub-horizontal and impersistent joint set is considered, with a potential failure surface that could extend through the rock mass; the safety factor is therefore computed using a combination of strength along the rock joint (using the nonlinear Barton and Choubey (1977) and Barton and Bandis (1990) failure criteria) and along the rock mass (using the nonlinear failure criterion of Hoek and Brown (1980) in its generalized expression from Hoek et al. (2002)). The proposed methodology also considers the influence of a non-associative flow rule that has been incorporated using a (constant) dilation angle (Hoek and Brown 1997). The newly proposed analytical methodology is used to assess the dam stability conditions, employing for this purpose the deterministic and probabilistic models, resulting in the sliding safety factor and the probability of failure respectively. The deterministic model, implemented in MATLAB, is validated using numerical solution computed with the finite difference code FLAC 6.0. The proposed deterministic model provides results that are very similar to those computed with FLAC; however, since the new formulation can be implemented in a spreadsheet, the computational cost of the proposed model is significantly smaller, hence allowing to more easily conduct parametric analyses of the influence of the different input parameters on the dam’s safety. Once the model is validated, parametric analyses are conducting using the main parameters that describe the dam’s foundation. From this study, the impact of the more influential parameters on the sliding stability analysis is obtained and the error of considering the flow rule is assessed. The probability of failure is obtained employing the First Order Reliability Method (FORM). The probabilistic model is then validated using the Monte Carlo simulation method. Results obtained using both methodologies show good agreement for cases in which the rock mass has a nonassociate flow rule. For cases with an associated flow rule errors between 0.70% and 66% are obtained, so that the better adjustments are obtained for cases with, or close to, limit equilibrium conditions. The main advantage of FORM on sliding stability analyses of gravity dams is its computational efficiency, so that Monte Carlo simulations require at least 4 hours on each execution, whereas FORM requires only 1 to 3 minutes on each execution.

El problema consiste básicamente en realizar un estudio de sensibilidad a la hora de analizar la estabilidad frente a rotura por cimiento a través de macizo rocoso y discontinuidad en el caso de presas de gravedad de tamaño medio. El presente documento muestra una comparativa de diferentes metodologías para estimar la seguridad de una presa de gravedad frente al deslizamiento por macizo rocoso y discontinuidad. Para ello se ha empleado un modelo de cálculo que computa la seguridad frente al deslizamiento a traves de un estudio paramétrico en función de las principales variables geométricas y geomecánicas que intervienen en el mismo. Se utilizan los criterios de resistencia de Hoek & Brown (1980) y de Barton & Choubey (1974) para caracterizar la resistencia del macizo rocoso y de las discontinuidades, respectivamente. El modelo permite definir tanto las variables de entrada (acciones y parámetros de los materiales), y el calculo del coeficiente de seguridad, (FS), a partir del cual se presenta el criterio de fallo a emplear en los cálculos de fiabilidad mediante técnicas estadísticas. Las diferentes metodologías utilizadas se agrupan en distintos niveles, que van desde: a) los niveles básicos asociados al determinismo, trabajando con FS, y/o con FS parciales; b) cálculos de segundo nivel, como el método de los momentos de segundo orden, los cuales proporcionan una probabilidad de fallo a partir de la aproximación de la función de densidad de probabilidad conjunta mediante sus dos primeros momentos (media y desviación típica) y la definición de una región de fallo con el modelo propuesto; c) finalmente, los métodos exactos o de nivel III, tales como el método de simulación de Monte Carlo, que generan histogramas de parámetros y proporcionan probabilidades de fallo. Con ello se pretende profundizar en las técnicas de cálculo de fiabilidad en presas, además de estimar la influencia de los distintos parámetros que definen la cimentación. Este modelo se puede emplear para otras estructuras y tipologías. A modo de ejemplo, se aplica un cálculo para el caso de una presa bóveda y se analizan los resultados obtenidos.

A presente pesquisa busca avaliar exegeticamente o texto que se encontra na Bíblia, especificamente no livro de Números capítulos 22-24 que relata sobre um personagem conhecido como Balaão. A pesquisa tem também como objeto o estudo sobre o panteão de divindades relatado no mesmo texto, assim como também o estudo dos textos descobertos em Deir Alla, na Jordânia, que apresentam um personagem designado como Balaão, possivelmente o mesmo personagem de Nm 22-24. A motivação que levou ao desenvolvimento dessa pesquisa foi o fato de se ter deparado com os conceitos dos diversos nomes divinos exibidos no texto, além da questão do profetismo fora de Israel, assim como as possibilidades hermenêuticas que se abrem para a leitura desse texto bíblico. O conceito geral sempre foi o de que Israel era a única nação onde existiam “verdadeiros” profetas e uma adoração a um único Deus, o “monoteísmo”. O que despertou interesse foi perceber, especialmente por meio da leitura dos livros bíblicos, que o profetismo não se restringiu somente a Israel. Ele antecede à formação do antigo Israel e já existia no âmbito das terras do antigo Oriente Médio, e que Israel ainda demorou muito tempo para ser monoteísta. Quem é esse Balaão, filho de Beor? Estudaremos sobre sua pessoa e sua missão. Examinaremos os textos de Deir Alla sobre Balaão e sua natureza de personagem mediador entre o divino e o humano. Esse personagem é apresentado como um grande profeta e que era famoso como intérprete de presságios divinos. Analisaremos a importante questão sobre o panteão de deuses que são apresentados na narrativa de Balaão nomeados como: El, Elyon Elohim e Shaddai, além de Yahweh. Entendemos, a princípio, que o texto possui uma conexão com a sociedade na qual foi criado e usando da metodologia exegética, faremos uma análise da narrativa em questão, buscando compreender o sentido do texto, dentro de seu cenário histórico e social. Cenário este, que nos apresentou esse profeta, não israelita, que profere bênçãos dos deuses sobre Israel e que, além disso, pronuncia maldições sobre os inimigos desse mesmo Israel. Percebemos que, parte do texto pesquisado é apresentado sob a ótica de Israel sobre as outras nações. A pesquisa defende, portanto, que o texto de Nm 22-24, além de nos apresentar um profeta fora de Israel igual aos profetas da Bíblia, defende que, o panteão de divindades também era adorado por Israel e que tais nomes são epítetos de uma mesma divindade, no caso YHWH. Defende, também, um delineamento de um projeto de domínio político e militar de Israel sobre as nações circunvizinhas.

The cytokine interleukin (IL) 18 (formerly interferon γ-inducing factor) induces the T helper type 1 response. In the present studies, IL-18 increased HIV type 1 (HIV-1) production from 5- to 30-fold in the chronically infected U1 monocytic cell line. Inhibition of tumor necrosis factor (TNF) activity by the addition of TNF-binding protein reduced IL-18-stimulated HIV-1 production by 48%. In the same cultures, IL-18-induced IL-8 was inhibited by 96%. Also, a neutralizing anti-IL-6 mAb reduced IL-18-induced HIV-1 by 63%. Stimulation of U1 cells with IL-18 resulted in increased production of IL-6, and exogenous IL-6 added to U1 cells increased HIV-1 production 4-fold over control. A specific inhibitor of the p38 mitogen-activated protein kinase reduced IL-18-induced HIV-1 by 73%, and a 50% inhibition was observed at 0.05 μM. In the same cultures, IL-8 was inhibited by 87%. By gel-shift and supershift analyses, increased binding activity of the transcription factor NF-κB was measured in nuclear extracts from U1 cells 1 h after exposure to IL-18. These results demonstrate induction of HIV-1 by IL-18 in a monocyte target associated with an intermediate role for TNF and IL-6, activation of p38 mitogen-activated protein kinase, and nuclear translocation of NF-κB.

Long-range promoter–enhancer interactions are a crucial regulatory feature of many eukaryotic genes yet little is known about the mechanisms involved. Using cloned chicken βA-globin genes, either individually or within the natural chromosomal locus, enhancer-dependent transcription is achieved in vitro at a distance of 2 kb with developmentally staged erythroid extracts. This occurs by promoter derepression and is critically dependent upon DNA topology. In the presence of the enhancer, genes must exist in a supercoiled conformation to be actively transcribed, whereas relaxed or linear templates are inactive. Distal protein–protein interactions in vitro may be favored on supercoiled DNA because of topological constraints. In this system, enhancers act primarily to increase the probability of rapid and efficient transcription complex formation and initiation. Repressor and activator proteins binding within the promoter, including erythroid-specific GATA-1, mediate this process.

This study investigated whether endothelin-1 (ET-1), a potent vasoconstrictor, which also stimulates cell proliferation, contributes to endothelial dysfunction and atherosclerosis. Apolipoprotein E (apoE)-deficient mice and C57BL/6 control mice were treated with a Western-type diet to accelerate atherosclerosis with or without ETA receptor antagonist LU135252 (50 mg/kg/d) for 30 wk. Systolic blood pressure, plasma lipid profile, and plasma nitrate levels were determined. In the aorta, NO-mediated endothelium-dependent relaxation, atheroma formation, ET receptor-binding capacity, and vascular ET-1 protein content were assessed. In apoE-deficient but not C57BL/6 mice, severe atherosclerosis developed within 30 wk. Aortic ET-1 protein content (P < 0.0001) and binding capacity for ETA receptors was increased as compared with C57BL/6 mice. In contrast, NO-mediated, endothelium-dependent relaxation to acetylcholine (56 ± 3 vs. 99 ± 2%, P < 0.0001) and plasma nitrate were reduced (57.9 ± 4 vs. 93 ± 10 μmol/liter, P < 0.01). Treatment with the ETA receptor antagonist LU135252 for 30 wk had no effect on the lipid profile or systolic blood pressure in apoE-deficient mice, but increased NO-mediated endothelium-dependent relaxation (from 56 ± 3 to 93 ± 2%, P < 0.0001 vs. untreated) as well as circulating nitrate levels (from 57.9 ± 4 to 80 ± 8.3 μmol/liter, P < 0.05). Chronic ETA receptor blockade reduced elevated tissue ET-1 levels comparable with those found in C57BL/6 mice and inhibited atherosclerosis in the aorta by 31% without affecting plaque morphology or ET receptor-binding capacity. Thus, chronic ETA receptor blockade normalizes NO-mediated endothelial dysfunction and reduces atheroma formation independent of plasma cholesterol and blood pressure in a mouse model of human atherosclerosis. ETA receptor blockade may have therapeutic potential in patients with atherosclerosis.

We have developed a noninvasive detection method for expression of viral-mediated gene transfer. A recombinant adenovirus was constructed by using the gene for arginine kinase (AK), which is the invertebrate correlate to the vertebrate ATP-buffering enzyme, creatine kinase. Gene expression was noninvasively monitored using 31P-magnetic resonance spectroscopy (31P-MRS). The product of the AK enzyme, phosphoarginine (PArg), served as an MRS-visible reporter of AK expression. The recombinant adenovirus coding for arginine kinase (rAdCMVAK) was injected into the right hindlimbs of neonatal mice. Two weeks after injection of rAdCMVAK, a unique 31P-MRS resonance was observed. It was observable in all rAdCMVAK injected hindlimbs and was not present in the contralateral control or the vehicle injected limb. PArg and phosphocreatine (PCr) concentrations were calculated to be 11.6 ± 0.90 and 13.6 ± 1.1 mM respectively in rAdCMVAK injected limbs. AK activity was demonstrated in vivo by monitoring the decreases in PArg and ATP resonances during prolonged ischemia. After 1 h of ischemia intracellular pH was 6.73 ± 0.06, PCr/ATP was decreased by 77 ± 8%, whereas PArg/ATP was decreased by 50 ± 15% of basal levels. PArg and PCr returned to basal levels within 5 min of the restoration of blood flow. AK activity persisted for at least 8 mo after injection, indicating that adenoviral-mediated gene transfer can produce stable expression for long periods of time. Therefore, the cDNA encoding AK provides a useful reporter gene that allows noninvasive and repeated monitoring of gene expression after viral mediated gene transfer to muscle.

Diverse biophysical and biochemical studies have sought to understand electron transfer (ET) in DNA in part because of its importance to DNA damage and its repair. However, the dynamics and mechanisms of the elementary processes of ET in this medium are not fully understood and have been heavily debated. Two fundamental issues are the distance over which charge is transported and the time-scale on which the transport through the π-stack of the DNA base pairs may occur. With femtosecond resolution, we report direct observation in DNA of ultrafast ET, initiated by excitation of tethered ethidium (E), the intercalated electron acceptor (A); the electron donor (D) is 7-deazaguanine (Z), a modified base, placed at different, fixed distances from A. The ultrafast ET between these reactants in DNA has been observed with time constants of 5 ps and 75 ps and was found to be essentially independent of the D–A separation (10–17 Å). However, the ET efficiency does depend on the D–A distance. The 5-ps decay corresponds to direct ET observed from 7-deazaguanine but not guanine to E. From measurements of orientation anisotropies, we conclude that the slower 75-ps process requires the reorientation of E before ET, similar to E/nucleotide complexes in water. These results reveal the nature of ultrafast ET and its mechanism: in DNA, ET cannot be described as in proteins simply by a phenomenological parameter, β. Instead, the involvement of the base pairs controls the time scale and the degree of coherent transport.
