O presente artigo descreve uma metodologia espectrofotométrica para a análise de paraformaldeído presente em amostras de saneantes comerciais e industriais, utilizando-se ácido cromotrópico (ACT), ácido clorídrico concentrado e peróxido de hidrogênio, produzindo um composto púrpura - avermelhado (lambda max = 575 nm). A lei de Beer é obedecida numa faixa de concentração de 0,8 - 4,8 mg L-1 em formaldeído, apresentando excelente coeficiente de correlação (r = 0,9999). Os valores obtidos nas análises concordaram muito favoravelmente com os obtidos pelo procedimento padrão recomendado pelo NIOSH (National Institute for Occupational Safety and Health).

Lipopolysacharide (LPS) present on the outer leaflet of Gram-negative bacteria is important for the adaptation of the bacteria to the environment. Structurally, LPS can be divided into three parts: lipid A, core and O-polysaccharide (OPS). OPS is the outermost and also the most diverse moiety. When OPS is composed of identical sugar residues it is called homopolymeric and when it is composed of repeating units of oligosaccharides it is called heteropolymeric. Bacteria synthesize LPS at the inner membrane via two separate pathways, Lipid A-core via one and OPS via the other. These are ligated together in the periplasmic space and the completed LPS molecule is translocated to the surface of the bacteria. The genes directing the OPS biosynthesis are often clustered and the clusters directing the biosynthesis of heteropolymeric OPS often contain genes for i) the biosynthesis of required NDP-sugar precursors, ii) glycosyltransferases needed to build up the repeating unit, iii) translocation of the completed O-unit to the periplasmic side of the inner membrane (flippase) and iv) polymerization of the repeating units to complete OPS. The aim of this thesis was to characterize the biosynthesis of the outer core (OC) of Yersinia enterocolitica serotype O:3 (YeO3). Y. enterocolitica is a member of the Gram-negative Yersinia genus and it causes diarrhea followed sometimes by reactive arthritis. The chemical structure of the OC and the nucleotide sequence of the gene cluster directing its biosynthesis were already known; however, no experimental evidence had been provided for the predicted functions of the gene products. The hypothesis was that the OC biosynthesis would follow the pathway described for heteropolymeric OPS, i.e. a Wzy-dependent pathway. In this work the biochemical activities of two enzymes involved in the NDP-sugar biosynthesis was established. Gne was determined to be a UDP-N-acetylglucosamine-4-epimerase catalyzing the conversion of UDP-GlcNAc to UDP-GalNAc and WbcP was shown to be a UDP-GlcNAc- 4,6-dehydratase catalyzing the reaction that converts UDP-GlcNAc to a rare UDP-2-acetamido- 2,6-dideoxy-d-xylo-hex-4-ulopyranose (UDP-Sugp). In this work, the linkage specificities and the order in which the different glycosyltransferases build up the OC onto the lipid carrier were also investigated. In addition, by using a site-directed mutagenesis approach the catalytically important amino acids of Gne and two of the characterized glycosyltranferases were identified. Also evidence to show the enzymes involved in the ligations of OC and OPS to the lipid A inner core was provided. The importance of the OC to the physiology of Y. enterocolitica O:3 was defined by determining the minimum requirements for the OC to be recognized by a bacteriophage, bacteriocin and monoclonal antibody. The biological importance of the rare keto sugar (Sugp) was also shown. As a conclusion this work provides an extensive overview of the biosynthesis of YeO3 OC as it provides a substantial amount of information of the stepwise and coordinated synthesis of the Ye O:3 OC hexasaccharide and detailed information of its properties as a receptor.

Metastases are the major cause of cancer deaths. Tumor cell dissemination from the primary tumor utilizes dysregulated cellular adhesion and upregulated proteolytic degradation of the extracellular matrix for progeny formation in distant organs. Integrins are transmembrane adhesive receptors mediating cellcell and cellmatrix interactions that are crucial for regulating cell migration, invasion, proliferation, and survival. Consequently, increased integrin activity is associated with augmented migration and invasion capacity in several cancer types. Heterodimeric integrins consist of an alpha - and beta-subunit that are held together in a bent conformation when the receptor is inactive, but extension and separation of subdomains is observed during receptor activation. Either inside-out or outside-in activation of receptors is possible through the intracellular molecule binding to an integrin cytoplasmic domain or extracellular ligand association with an integrin ectodomain, respectively. Several regulatory binding partners have been characterized for integrin cytoplasmic beta-domains, but the regulators interacting with the cytoplasmic alpha-domains have remained elusive. In this study, we performed yeast two-hybrid screens to identify novel binding partners for the cytoplasmic integrin alpha-domains. Further examination of two plausible candidates revealed a significant coregulatory role of an integrin alpha-subunit for cellular signaling processes. T-cell protein tyrosine phosphatase (TCPTP) showed a specific interaction with the cytoplasmic tail of integrin alpha1. This association stimulated TCPTP phosphatase activity, leading to negative regulation of epidermal growth factor receptor (EGFR) signaling and diminished anchorage-independent growth. Another candidate, mammary-derived growth inhibitor (MDGI), exhibited binding to several different integrin cytoplasmic alpha-tails through a conserved GFFKR sequence. MDGI overexpression in breast cancer cells altered EGFR trafficking and caused a remarkable accumulation of EGFR in the cytoplasm. We further demonstrated in vivo that MDGI expression induced a novel form of anti-EGFR therapy resistance. Moreover, MDGI binding to α-tails retained integrin in an inactive conformation attenuating integrin-mediated adhesion, migration, and invasion. In agreement with these results, sustained MDGI expression in breast cancer patients correlated with an increased 10-year distant disease-free survival. Taken together, the integrin signaling network is far from a complete view and future work will doubtless broaden our understanding further.

Maailman energian kulutuksen lisääntymisen ja ilmastonmuutoksen myötä energiantuotannossa joudutaan jatkuvasti sopeutumaan muuttuviin tilanteisiin ja haasteisiin. Polttoteknillisiä haasteita aiheuttavat pelto- ja kierrätyspolttoaineet ovat lisäämässä osuuttaan uusiutuvien polttoaineiden joukossa. Jotta kyseisiä haasteellisia polttoaineita pystytään hyödyntämään, täytyy niiden aiheuttamat ongelmat tuntea ja laitevalmistajien kehittää niiden hyödyntämiseen sopivaa tekniikkaa. Tässä diplomityössä käydään läpi tulevaisuudessa käytettävät polttoaineet, nykyiset päästörajat, kiinteiden polttoaineiden poltto- ja kaasutustekniikat sekä likaantumis-, kuonaantumis- ja korroosiomekanismit voimalaitoskattiloissa. Työssä tutkitaan, onko haasteellisten polttoaineiden käyttöön investoiminen järkevää ja mikä nykypäivän tekniikoista on kannattavin. Myös välitulistuksen, lauhdeperän ja apujäähdyttimen kannattavuuksia vertaillaan sähkön ja lämmön yhteistuotannossa. Tuloksiksi saatiin, että edullisten peltobiomassojen ja kierrätyspolttoaineiden käyttäminen, joko perinteisten polttoaineiden seassa tai pääpolttoaineena, on nykyhinnoilla perinteisiin polttoaineisiin verrattuna kannattavaa. Investoiminen kierrätyspolttoaineiden valmistuslaitteisiin maksimoi kierrätyspolttoaineista saatavaa hyötyä. Välitulistuksen todettiin soveltuvan huonosti vastapaineprosessiin, sillä siitä saatava sähköntuotannon lisäys on hyvin pieni. Lauhdeperän ja apujäähdyttimen vertailuissa huomattiin, että lauhdeperä on kannattava investointi, jos sähkön ja lämmön hintaero pysyy tarpeeksi suurena. Haasteellisilla polttoaineilla pystytään pienentämään kasvihuonepäästöjä ja korvaamaan fossiilisten polttoaineiden käyttöä.

O presente artigo apresenta um estudo sobre a variação da velocidade ultra-sônica longitudinal em função do comprimento da peça de madeira. As espécies utilizadas foram: pinus caribaea (Pinus caribaea var. caribaea) eucalipto citriodora (Eucalyptus citriodora), eucalipto grandis (Eucalyptus grandis) e jatobá (Hymenaea sp). Utilizou-se equipamento de ultra-som com transdutores exponenciais de 22 kHz. Foram feitas medições do tempo de propagação das ondas ultra-sônicas, com o comprimento variando de 300 cm a 10 cm e mantendo-se a seção transversal constante de 12 cm x 5 cm. Os resultados apontaram que ocorre variação da velocidade em função da distância percorrida e do comprimento de onda (lambda) utilizado. Conclui-se que adequar a freqüência do transdutor com o comprimento da peça é essencial para a determinação correta da velocidade ultra-sônica em madeiras.

Devido à grande importância em se avaliar o destino e o transporte de íons no solo, o presente trabalho teve o objetivo de obter os parâmetros de transporte do nitrato no solo, como a velocidade da água nos poros (v), o fator de retardamento (R), a dispersividade (lambda) e o coeficiente de dispersão (D), em solos de diferentes texturas e em amostras deformadas e indeformadas. O ensaio foi conduzido empregando as amostras deformadas e indeformadas oriundas de dois perfis distintos de solos, coletadas à profundidade de 0-20 cm, com aplicação de solução de nitrato de cálcio contendo 50 mg L-1 de NO3-. Após a obtenção dos parâmetros de transporte, ajustados pelo programa computacional CXTFIT, notou-se que as curvas de efluentes preparadas com solos a partir de amostra deformada superestimaram os valores dos parâmetros avaliados, com exceção da dispersividade, quando comparados à amostra indeformada, para os dois tipos de solo.

The objective of this study was to evaluate growth and yield of papaya cv. Sunrise solo under trickle irrigation system configurations. A random block design was used with six treatments and four replications. Treatments were: T1- a 32 L h-1 micro sprinkler; T2 - a 43 L h-1 micro sprinkler and T3 - a 60 L h-1 micro sprinkler for four plants; T4 - Drip system with four emitters per plant on one lateral line per crop row; T5 - Drip system with eight emitters per plant on two laterals line per crop row; T6 - Drip system with four emitters per plant on one lateral line distributed as pig tail. The dripper flow rate was 4 L h-1and they were apart each other 0.50 m. Crop growth variables were measured every two weeks. Production variables were evaluated during harvest. The treatment that presented superiority was irrigated by micro sprinkler system with flow rate of 43 L h-1.

The papillary cystic and solid tumor of the pancreas is rare. It occurs predominantly in young women and most present a benign behavior. The most common clinical sign is a large palpable abdominal mass. The pathogenesis of this tumor has attracted a number of investigations but remains unclear. We present a 18 year old white woman with abdominal mass detected after cesarian. Clinical examination showed minimal tenderness. There was no history of weight loss or jaundice. Haematological parameters were normal, except anaemia. The computed tomography was performed and surprisingly showed a 10 cm mass in the region of the tail of the pancreas. An extended distal pancreatectomy was performed with splenic preservation. The patient had an uneventful recovery and two months later remains asymptomatic.

The purpose of this study is to examine how well risk parity works in terms of risk, return and diversification relative to more traditional minimum variance, 1/N and 60/40 portfolios. Risk parity portfolios were constituted of five risk sources; three common asset classes and two alternative beta investment strategies. The three common asset classes were equities, bonds and commodities, and the alternative beta investment strategies were carry trade and trend following. Risk parity portfolios were constructed using five different risk measures of which four were tail risk measures. The risk measures were standard deviation, Value-at-Risk, Expected Shortfall, modified Value-at-Risk and modified Expected Shortfall. We studied also how sensitive risk parity is to the choice of risk measure. The hypothesis is that risk parity portfolios provide better return with the same amount of risk and are better diversified than the benchmark portfolios. We used two data sets, monthly and weekly data. The monthly data was from the years 1989-2011 and the weekly data was from the years 2000-2011. Empirical studies showed that risk parity portfolios provide better diversification since the diversification is made at the risk level. Risk based portfolios provided superior return compared to the asset based portfolios. Using tail risk measures in risk parity portfolios do not necessarily provide better hedge from tail events than standard deviation.

Papyllary cystic tumor of the pancreas, so-called Frantz's tumor, is rare. Clinical presentation of this disease is usually a slowly growing abdominal mass with or without abdominal pain, affecting predominantly young females. Its pathogenesis is still unknown . Surgical resection is usually curative, and prognosis is excellent. The authors report two pancreatic tumor cases(Frantz's tumor) in women aged 26 and 31 years old. Pre operative assessment showed a solid-cystic tumor of the tail and body of the pancreas. An extended distal pancreatectomy was performed without splenic preservation.

This study presents a review of theories of the so-called post-industrial society, and proposes that the concept of post-industrial society can be used to understand the recent developments of the World Wide Web, often described as Web 2.0 or social Web. The study combines theories ranging from post-war management science and cultural studies to software development, and tries to build a holistic view of the development of the post-industrial society, and especially the Internet. The discourse on the emergence of a post-industrial society after the World Wars has addressed the ways in which the growing importance of information, and innovations in digital communications technology, are changing our society. It is furthermore deeply connected with the discourse on the postmodern society, which emphasizes cultural fragmentation, intertextuality, and pluralism. The Internet age is characterized by increasing masses of information that are managed through various technologies. While 1990s Internet technologies often used the network as a traditional broadcasting channel with added interactivity, Web 2.0 technologies are specifically designed to utilize the network model by facilitating communication between various services and devices, and analyzing the relationships between users and objects in order to produce intelligent insight. The wide adoption of the Internet, and recently of Internet-enabled mobile devices, is furthermore continuously producing new ways of communicating, consuming, and producing. Applications of the social Web, such as social media or social networking services, are permanently changing our traditional social, cultural, and economic practices. The study first presents an overview of the post-industrial society, the Internet, and the concept of Web 2.0. Then the concept of social Web is described with an analysis of the term social media, the brief histories of the interactive Web and social networking services, and a description of the concept ―long tail‖, used to represent the masses of information available in the Web that do not receive mainstream attention. Finally, methods for retrieving and filtering information, modeling social and cultural relationships, and communicating with customers, are presented.

Tämän tutkimuksen tavoitteena oli nykyisin tuotantokäytössä olevan rullaoikaisukoneen automatisointi ja uuden oikaisukonekokonaisuuden mitoitus tulevaisuuden materiaaliominaisuuksien perusteella. Automatisoinnilla on tarkoitus nykyisessä käytössä helpottaa nauhahitsaajan työtä ja lähitulevaisuudessa optimoida romutustarvetta kelojen keula- ja häntäalueilla. Uuden oikaisukonekokonaisuuden osalta työssä on määritetty rullaoikaisukoneen rullakoot, rullien keskiöetäisyys, vaadittava moottoriteho ja rullien tuennan tarve sekä erillisen hilseenmurskaimen/rullavenytysoikaisukoneen tarve peittauslinjan toiminnassa. Rullaoikaisukoneen asetusarvojen laskeminen toteutettiin ympyräkaarimenetelmää ja seurantatutkimusta käyttäen. Ensimmäisen ylärullan asetteluarvot on laskettu suorilla kaavasovelluksilla. Toisen ylärullan kohdalla oli tarpeellista tehdä laajempaa seurantatutkimusta, jotta löydettiin sellainen tapa, jolla myös sen arvot saataisiin riippuvaiseksi ensimmäisen ylärullan asetteluarvoista ja ympyränkaarimenetelmästä. Uutta oikaisukonekokonaisuutta varten suoritettujen laskelmien perusteella rullaoikaisukoneen telakokoa ja keskiöetäisyyttä on pienennettävä, moottoritehoa on kasvatettava ja työrullien tuentaa on parannettava verrattuna nykyiseen tuotantokoneeseen. Esioikaisukonetyyppisen rullaoikaisukoneen rajoittuneesta oikaisutuloksesta ja tietyillä kelalaaduilla esiintyvä erittäin luja wüstiittihilse puoltaa vahvasti erillisen hilseenmurskaimen lisäämistä peittauslinjan konekantaan.

The aim of this study was to compare different staining methods for the evaluation of sperm morphology by light microscopy and also to describe the morphometry of the entire sperm in collared peccaries (Pecari tajacu). Semen from 10 males was obtained by electroejaculation and evaluated for sperm motility, vigor, and concentration. Semen smears were prepared through three different staining methods: Bengal rose, brome-phenol blue, and eosin-nigrosin. Smears were evaluated under light microscopy and sperm morphologic alterations were determined in percentage. In addition, sperm morphometric analysis was conducted by light microscopy coupled to image analyzer software. The smears stained with Bengal Rose provide the best results for the visualization of the sperm tail, midpiece, and head. The use of eosin-nigrosin stain did not allow an adequate impregnation, and some sperm presented a few contrasts with the background. A higher incidence of bent coiled tails was verified in the use of brome-phenol blue staining (P<0.05). Through morphometric evaluation, it was observed that the tail occupies the greatest proportion (89%) of the sperm which presents a discretely elongated head. According to the results, the use of the Bengal Rose stain is recommended for the morphologic evaluation of the collared peccary sperm.

The objective of this study was to evaluate the culture of equine bone marrow mononuclear fraction and adipose tissue - derived stromal vascular fraction cells in two different cell culture media. Five adult horses were submitted to bone marrow aspiration from the sternum, and then from the adipose tissue of the gluteal region near the base of the tail. Mononuclear fraction and stromal vascular fraction were isolated from the samples and cultivated in DMEM medium supplemented with 10% fetal bovine serum or in AIM-V medium. The cultures were observed once a week with an inverted microscope, to perform a qualitative analysis of the morphology of the cells as well as the general appearance of the cell culture. Colony-forming units (CFU) were counted on days 5, 15 and 25 of cell culture. During the first week of culture, differences were observed between the samples from the same source maintained in different culture media. The number of colonies was significantly higher in samples of bone marrow in relation to samples of adipose tissue.

The number of molecular diagnostic assays has increased tremendously in recent years.Nucleic acid diagnostic assays have been developed, especially for the detection of human pathogenic microbes and genetic markers predisposing to certain diseases. Closed-tube methods are preferred because they are usually faster and easier to perform than heterogenous methods and in addition, target nucleic acids are commonly amplified leading to risk of contamination of the following reactions by the amplification product if the reactions are opened. The present study introduces a new closed-tube switchable complementation probes based PCR assay concept where two non-fluorescent probes form a fluorescent lanthanide chelate complex in the presence of the target DNA. In this dual-probe PCR assay method one oligonucleotide probe carries a non-fluorescent lanthanide chelate and another probe a light absorbing antenna ligand. The fluorescent lanthanide chelate complex is formed only when the non-fluorescent probes are hybridized to adjacent positions into the target DNA bringing the reporter moieties in close proximity. The complex is formed by self-assembled lanthanide chelate complementation where the antenna ligand is coordinated to the lanthanide ion captured in the chelate. The complementation probes based assays with time-resolved fluorescence measurement showed low background signal level and hence, relatively high nucleic acid detection sensitivity (low picomolar target concentration). Different lanthanide chelate structures were explored and a new cyclic seven dentate lanthanide chelate was found suitable for complementation probe method. It was also found to resist relatively high PCR reaction temperatures, which was essential for the PCR assay applications. A seven-dentate chelate with two unoccupied coordination sites must be used instead of a more stable eight- or nine-dentate chelate because the antenna ligand needs to be coordinated to the free coordination sites of the lanthanide ion. The previously used linear seven-dentate lanthanide chelate was found to be unstable in PCR conditions and hence, the new cyclic chelate was needed. The complementation probe PCR assay method showed high signal-to-background ratio up to 300 due to a low background fluorescence level and the results (threshold cycles) in real-time PCR were reached approximately 6 amplification cycles earlier compared to the commonly used FRET-based closed-tube PCR method. The suitability of the complementation probe method for different nucleic acid assay applications was studied. 1) A duplex complementation probe C. trachomatis PCR assay with a simple 10-minute urine sample preparation was developed to study suitability of the method for clinical diagnostics. The performance of the C. trachomatis assay was equal to the commercial C. trachomatis nucleic acid amplification assay containing more complex sample preparation based on DNA extraction. 2) A PCR assay for the detection of HLA-DQA1*05 allele, that is used to predict the risk of type 1 diabetes, was developed to study the performance of the method in genotyping. A simple blood sample preparation was used where the nucleic acids were released from dried blood sample punches using high temperature and alkaline reaction conditions. The complementation probe HLA-DQA1*05 PCR assay showed good genotyping performance correlating 100% with the routinely used heterogenous reference assay. 3) To study the suitability of the complementation probe method for direct measurement of the target organism, e.g., in the culture media, the complementation probes were applied to amplificationfree closed-tube bacteriophage quantification by measuring M13 bacteriophage ssDNA. A low picomolar bacteriophage concentration was detected in a rapid 20- minute assay. The assay provides a quick and reliable alternative to the commonly used and relatively unreliable UV-photometry and time-consuming culture based bacteriophage detection methods and indicates that the method could also be used for direct measurement of other micro-organisms. The complementation probe PCR method has a low background signal level leading to a high signal-to-background ratio and relatively sensitive nucleic acid detection. The method is compatible with simple sample preparation and it was shown to tolerate residues of urine, blood, bacteria and bacterial culture media. The common trend in nucleic acid diagnostics is to create easy-to-use assays suitable for rapid near patient analysis. The complementation probe PCR assays with a brief sample preparation should be relatively easy to automate and hence, would allow the development of highperformance nucleic acid amplification assays with a short overall assay time.
