In vascular plants, the best-known feature of a differentiated endodermal cell is the "Casparian Strip" (CS). This structure refers to a highly localized cell wall impregnation in the transversal and anticlinal walls of the cell, which surrounds the cell like a belt/ring and is tightly coordinated with respect to neighboring cells. Analogous to tight junctions in animal epithelia, CS in plants act as a diffusion barrier that controls the movement of water and ions from soil into the stele. Since its first description by Robert Caspary in 1865 there have been many attempts to identify the chemical nature of the cell wall deposition in CS. Suberin, lignin, or both have been claimed to be the important components of CS in a series of different species. However, the exact chemical composition of CS has remained enigmatic. This controversy was due to the confusion and lack of knowledge regarding the precise measurement of three developmental stages of the endodermis. The CS represent only the primary stage of endodermal differentiation, which is followed by the deposition of suberin lamellae all around the cellular surface of endodermal cells (secondary developmental stage). Therefore, chemical analysis of whole roots, or even of isolated endodermal tissues, will always find both of the polymers present. It was crucial to clarify this point because this will guide our efforts to understand which cell wall biosynthetic component becomes localized in order to form the CS. The main aim of my work was to find out the major components of (early) CS, as well as their spatial and temporal development, physiological roles and relationship to barrier formation. Employing the knowledge and tools that have been accumulated over the last few years in the model plant Arabidopsis thaliana, various histological and chemical assays were used in this study. A particular feature of my work was to completely degrade, or inhibit formation of lignin and suberin biopolymers by biochemical, classical genetic and molecular approaches and to investigate its effect on CS formation and the establishment of a functional diffusion barrier. Strikingly, interference with monolignol biosynthesis abrogates CS formation and delays the formation of function diffusion barrier. In contrast, transgenic plants devoid of any detectable suberin still develop a functional CS. The combination of all these assays clearly demonstrates that the early CS polymer is made from monolignol (lignin monomers) and is composed of lignin. By contrast, suberin is formed much later as a secondary wall during development of endodermis. These early CS are functionally sufficient to block extracellular diffusion and suberin does not play important role in the establishment of early endodermal diffusion barrier. Moreover, suberin biosynthetic machinery is not present at the time of CS formation. Our study finally concludes the long-standing debate about the chemical nature of CS and opens the door to a new approach in lignin research, specifically for the identification of the components of the CS biosynthetic pathway that mediates the localized deposition of cell walls. I also made some efforts to understand the patterning and differentiation of endodermal passage cells in young roots. In the literature, passage cells are defined as a non- suberized xylem pole associated endodermal cells. Since these cells only contain the CS but not the suberin lamellae, it has been assumed that these cells may offer a continued low-resistance pathway for water and minerals into the stele. Thus far, no genes have been found to be expressed specifically in passage cells. In order to understand the patterning, differentiation, and physiological role of passage it would be crucial to identify some genes that are exclusively expressed in these cells. In order to identify such genes, I first generated fluorescent marker lines of stele-expressed transporters that have been reported to be expressed in the passage cells. My aim was to first highlight the passage cells in a non-specific way. In order to find passage cell specific genes I then adapted a two-component system based on previously published methods for gene expression profiling of individual cell types. This approach will allow us to target only the passage cells and then to study gene expression specifically in this cell type. Taken together, this preparatory work will provide an entry point to understand the formation and role of endodermal passage cells. - Chez les plantes vasculaires, la caractéristique la plus commune des cellules différentiées de l'endoderme est la présence de cadres de Caspary. Cette structure correspond à une imprégnation localisée des parties transversales et anticlinales de la paroi cellulaire. Cela donne naissance, autour de la cellule, à un anneau/cadre qui est coordonné par rapport aux cellules voisines. De manière analogue aux jonctions serrées des épithéliums chez les animaux, les cadres de Caspary agissent chez les plantes comme barrière de diffusion, contrôlant le mouvement de l'eau et des ions à travers la racine entre le sol et la stèle. Depuis leur première description par Robert Caspary en 1865, beaucoup de tentatives ont eu pour but de définir la nature chimique de ces cadres de Caspary. Après l'étude de différentes espèces végétales, à la fois la subérine, la lignine ou les deux ont été revendiquées comme étant des composants importants de ces cadres. Malgré tout, leur nature chimique exacte est restée longtemps énigmatique. Cette controverse provient de la confusion et du manque de connaissance concernant la détermination précise des trois stades de développement de l'endoderme. Les cadres de Caspary représentent uniquement le stade primaire de différentiation de l'endoderme. Celui-ci est suivi par le second stade de différentiation, la déposition de lamelles de subérine tout autour de la cellule endodermal. De ce fait, l'analyse chimique de racines entières ou de cellules d'endoderme isolées ne permet pas de séparer les stades de différentiation primaire et secondaire et aboutit donc à la présence des deux polymères. Il est également crucial de clarifier ce point dans le but de connaître quelle machinerie cellulaire localisée à la paroi cellulaire permet l'élaboration des cadres de Caspary. En utilisant les connaissances et les outils accumulés récemment grâce à la plante modèle Arabidopsis thaliana, divers techniques histologiques et chimiques ont été utilisées dans cette étude. Un point particulier de mon travail a été de dégrader ou d'inhiber complètement la formation de lignine ou de subérine en utilisant des approches de génétique classique ou moléculaire. Le but étant d'observer l'effet de l'absence d'un de ces deux polymères sur la formation des cadres de Caspary et l'établissement d'une barrière de diffusion fonctionnelle. De manière frappante, le fait d'interférer avec la voie de biosynthèse de monolignol (monomères de lignine) abolit la formation des cadres de Caspary et retarde l'élaboration d'une barrière de diffusion fonctionnelle. Par contre, des plantes transgéniques dépourvues d'une quantité détectable de subérine sont quant à elles toujours capables de développer des cadres de Caspary fonctionnels. Mises en commun, ces expériences démontrent que le polymère formant les cadres de Caspary dans la partie jeune de la racine est fait de monolignol, et que de ce fait il s'agit de lignine. La subérine, quant à elle, est formée bien plus tard durant le développement de l'endoderme, de plus il s'agit d'une modification de la paroi secondaire. Ces cadres de Caspary précoces faits de lignine suffisent donc à bloquer la diffusion extracellulaire, contrairement à la subérine. De plus, la machinerie de biosynthèse de la subérine n'est pas encore présente au moment de la formation des cadres de Caspary. Notre étude permet donc de mettre un terme au long débat concernant la nature chimique des cadres de Caspary. De plus, elle ouvre la porte à de nouvelles approches dans la recherche sur la lignine, plus particulièrement pour identifier des composants permettant la déposition localisée de ce polymère dans la paroi cellulaire. J'ai aussi fais des efforts pour mettre en évidence la formation ainsi que le rôle des cellules de passage dans les jeunes racines. Dans la littérature, les cellules de passage sont définies comme de la cellule endodermal faisant face aux pôles xylèmes et dont la paroi n'est pas subérisée. Du fait que ces cellules contiennent uniquement des cadres de Caspary et pas de lamelle de subérine, il a été supposé qu'elles ne devraient offrir que peu de résistance au passage de l'eau et des nutriments entre le sol et la stèle. Le rôle de ces cellules de passage est toujours loin d'être clair, de plus aucun gène s'exprimant spécifiquement dans ces cellules n'a été découvert à ce jour. De manière à identifier de tels gènes, j'ai tout d'abord généré des marqueurs fluorescents pour des transporteurs exprimés dans la stèle mais dont l'expression avait également été signalée dans l'endoderme, uniquement dans les cellules de passage. J'ai ensuite développé un système à deux composants basé sur des méthodes déjà publiées, visant principalement à étudier le profil d'expression génique dans un type cellulaire donné. En recoupant les gènes exprimés spécifiquement dans l'endoderme à ceux exprimés dans la stèle et les cellules de passage, il nous sera possible d'identifier le transriptome spécifique de ces cellules. Pris dans leur ensemble, ces résultats devraient donner un bon point d'entrée dans la définition et la compréhension des cellules de passage.

Liddle's syndrome is a genetic form of hypertension linked to Na(+) retention caused by activating mutations in the COOH terminus of the beta or gamma subunit of the epithelial sodium channel (ENaC). In this study, we used the short-circuit current (I(sc)) method to investigate the effects of deamino-8-d-arginine vasopressin (dDAVP) on Na(+) and Cl(-) fluxes in primary cultures of cortical collecting ducts (CCDs) microdissected from the kidneys of mice with Liddle's syndrome carrying a stop codon mutation, corresponding to the beta-ENaC R(566) stop mutation (L) found in the original pedigree. Compared to wild-type (+/+) CCD cells, untreated L/+ and L/L CCD cells exhibited 2.7- and 4.2-fold increases, respectively, in amiloride-sensitive (Ams) I(sc), reflecting ENaC-dependent Na(+) absorption. Short-term incubation with dDAVP caused a rapid and significant increase (approximately 2-fold) in Ams I(sc) in +/+, but not in L/+ or L/L CCD cells. In sharp contrast, dDAVP induced a greater increase in 5-nitro-2-(3-phenylpropamino)benzoate (NPPB)-inhibited apical Cl(-) currents in amiloride-treated L/L and L/+ cells than in their +/+ counterparts. I(sc) recordings performed under apical ion substituted conditions revealed that the dDAVP-stimulated apical secretion of Cl(-), which was absent in cultured CCDs lacking CFTR, was 1.8-fold greater in L/+ and 3.7-fold greater in L/L CCD cells than in their +/+ CCD counterparts. After the basal membrane had been permeabilized with nystatin and a basal-to-apical Cl(-) gradient had been imposed, dDAVP also stimulated larger Cl(-) currents across L/L and L/+ CCD layers than +/+ CCD layers. These findings demonstrate that vasopressin stimulates greater apical CFTR Cl(-) conductance in the renal CCD cells of mice with Liddle's syndrome than in wild-type mice. This effect could contribute to the enhanced NaCl reabsorption observed in the distal nephron of patients with Liddle's syndrome.

There is a need to measure energy expenditure in man for a period of 24 h or even several days. The respiration chamber offers a unique opportunity to reach this goal. It allows the study of energy and nutrient balance; from the latter, acute changes in body composition can be obtained. The respiration chamber built in Lausanne is an air-tight room (5 m long, 2.5 m wide, and 2.5 m high) which forms an open circuit ventilated indirect calorimeter. The physical activity of the subject inside the chamber is continuously measured using a radar system based on the Doppler effect. Energy expenditure of obese and lean women was continuously measured over 24 h and diet-induced thermogenesis was assessed by using an approach which allows one to subtract the energy expended for physical activity from the total energy expenditure. Expressed in absolute terms, total energy expenditure was more elevated in the obese than in the lean controls. Basal metabolic rate was also higher in the obese than in the controls, but diet-induced thermogenesis was found to be blunted in the obese. In a second study, the effect of changing the carbohydrate/lipid content of the diet on fuel utilization was assessed in young healthy subjects with the respiration chamber. After a 7-day adaptation to a high-carbohydrate low-fat diet, the fuel mixture oxidized matched the change in nutrient intake. A last example of the use of the respiration chamber is the thermogenic response and changes in body composition due to a 7-day overfeeding of carbohydrate. Diet-induced thermogenesis was found to be 27%; on the last day of overfeeding, carbohydrate balance was reached by oxidation of 50% of the carbohydrate intake, the remaining 50% being converted into lipid.

Boundaries for delta, representing a "quantitatively significant" or "substantively impressive" distinction, have not been established, analogous to the boundary of alpha, usually set at 0.05, for the stochastic or probabilistic component of "statistical significance". To determine what boundaries are being used for the "quantitative" decisions, we reviewed pertinent articles in three general medical journals. For each contrast of two means, contrast of two rates, or correlation coefficient, we noted the investigators' decisions about stochastic significance, stated in P values or confidence intervals, and about quantitative significance, indicated by interpretive comments. The boundaries between impressive and unimpressive distinctions were best formed by a ratio of greater than or equal to 1.2 for the smaller to the larger mean in 546 comparisons, by a standardized increment of greater than or equal to 0.28 and odds ratio of greater than or equal to 2.2 in 392 comparisons of two rates; and by an r value of greater than or equal to 0.32 in 154 correlation coefficients. Additional boundaries were also identified for "substantially" and "highly" significant quantitative distinctions. Although the proposed boundaries should be kept flexible, indexes and boundaries for decisions about "quantitative significance" are particularly useful when a value of delta must be chosen for calculating sample size before the research is done, and when the "statistical significance" of completed research is appraised for its quantitative as well as stochastic components.

La finalitat d'aquest projecte és la creació d'una eina colaborativa de workflow (circuit de treball) que permeti gestionar de manera eficaç les peticions de serveis o productes rebudes pel departament de tecnologies d'una empresa mitjana/gran, informant en cada moment del seu cicle de vida les persones involucrades en la mateixa.

Sensory information is an important factor in shaping neuronal circuits during development and adulthood. In the barrel cortex of adult rodents, cells from layer IV are able to adapt their functional state to an increased flow of sensory information from the mystacial whisker follicles. Previous studies in our group have shown that whisker stimulation induces the formation of inhibitory synapses in the corresponding barrel (Knott et al., 2002) and decreases neuronal responses toward the deflection of the stimulated whisker (Quairiaux et al., 2007). Together these observations have turned the barrel cortex into a model to study homeostatic plasticity. At the cellular level, neuronal activity triggers intracellular signaling cascades leading to a transcriptional response. To further characterize the molecular pathways involved in the synaptic changes after whisker stimulation in the adult mouse, a previous doctoral student in our group performed a microarray analysis on laser-dissected barrels in sections through layer IV. This study identified the regulation (up and down) of a series of genes in the stimulated barrels (thesis of Johnston-Wenger, 2010). We here focused on ten genes that presented the highest fold change according to the microarray analysis. Out of these genes, 7 are known as neuronal activity-dependent genes (Tnncl, Nptx2, Sorcs3, Ptgs2, Nr4a2, Npas4 and Adcyapl) whereas three have so far not been related to neuronal plasticity (Scn7a, Pcdhl5 and Cede3). The study aimed at confirming the results of the microarray analysis and localizing molecular modifications in the stimulated barrel column at the cellular level. In situ hybridization for Pcdhl5 after different periods of whisker stimulation (3, 6, 9, 15, 24 hrs) allowed us to confirm that the 1.25 fold change used for the microarray analysis is an appropriate threshold for considering a regulation significant after sensory-stimulation. Moreover, we confirmed with in situ hybridization a significant upregulation of the genes of interest in the stimulated barrels. In situ hybridization and immunohistochemistry allowed us to observe the distribution of the genes of interest and the corresponding protein products at the cellular level. Three observations were made: 1) alterations of the expression was restricted to the stimulated barrels for all genes tested; 2) within a barrel column not all cells responded to whisker stimulation with an altered gene expression; 3) in the stimulated barrels, two different patterns of mRNA and protein expression can be distinguished. We hypothesize that this segregation of the activity-induced gene expression reflects the segregation of the two principal thalamocortical pathways conveying the sensory information to the barrel cortex. Moreover, only neurons reaching the critical threshold will modify their gene expression program resulting in structural as well as physiological modifications that prevent the subsequent propagation of the excess of excitation to the postsynaptic targets. The activity-induced gene expression is therefore adapted in a cell-type-specific manner to induce a homeostatic response to the entire neuronal network involved in the integration of the sensory information. This to our knowledge the first study showing the distinct, but complementary contribution of the two thalamocortical pathways in experience-dependent plasticity in the adult mouse barrel cortex. -- L'information sensorielle nous permet de continuellement façonner nos circuits neuronaux autant durant le développement qu'à l'âge adulte. Chez le rongeur l'information sensorielle perçue par les vibrisses est intégrée au niveau du cortex somatosensoriel primaire (appelé en anglais « barrel cortex ») dont les cellules de la couche IV sont capables d'adapter leur état fonctionnel en réponse à une augmentation d'activité neuronale. Ce modèle expérimental a permis à notre groupe de recherche d'observer des changements rapides du circuit neuronal en fonction de l'activité sensorielle. En effet, la stimulation continue d'une vibrisse d'une souris adulte pendant 24 heures induit non seulement un remaniement synaptique (Knott et al., 2002), mais également des changements physiologiques au niveau des neurones du tonneau correspondant (Quairiaux et al., 2007). Ces observations nous permettent d'affirmer que le « barrel cortex » est un modèle approprié pour y étudier la plasticité synaptique. Au niveau cellulaire, l'activité neuronale déclenche des cascades de signalisation intracellulaire résultant en une réponse transcriptionnelle. Afin de caractériser les voies moléculaires impliquées dans la plasticité synaptique, une puce à ARN nous a permis de comparer l'expression de gènes entre un tonneau correspondant à une vibrisse stimulée et un tonneau d'une vibrisse non-stimulée (Nathalie). Cette analyse a révélé un certain nombre de gènes régulés de manière positive ou négative par l'augmentation de l'activité neuronale. Nous nous sommes concentrés sur 10 gènes dont l'expression est fortement régulée. L'expression de sept d'entre eux a déjà été démontrée comme dépendante de l'activité neuronale (Tnncl, Nptx2, Sorcs3, Ptgs2, Nr4a2, Npas4 otAdcyapl) alors que l'expression des trois autres (Scn7a, Pcdhl5 et Cedei) n'a pour le moment pas encore été liée à la plasticité neuronale. Le but de cette thèse est de confirmer les résultats de la puce à ARN et de déterminer dans quel type cellulaire ces gènes sont exprimés. L'hybridation in situ pour le gène Pcdhl5, après différentes périodes de stimulation des vibrisses (3, 6, 9, 15 et 24 heures), nous a permis de confirmer que le seuil de 1.25x utilisé dans l'analyse de la puce à ARN est approprié pour considérer qu'un gène est régulé de manière significative par la stimulation sensorielle. Nous avons également pu confirmer à l'aide de cette technique que la stimulation sensorielle augmente significativement l'expression de ces dix gènes. L'expression de ces gènes au niveau cellulaire a été observée à l'aide des techniques d'hybridation in situ et d'immunohistochimie. Trois observations ont été faites : 1) la régulation de ces gènes est restreinte aux tonneaux correspondants aux vibrisses stimulées ; 2) au niveau d'une colonne corticale correspondant aux vibrisses stimulées, seules certaines cellules présentent une altération de leur expression génique ; 3) au niveau des tonneaux stimulés, deux profils d'expression d'ARNm et de protéines sont observés. Notre hypothèse est que cette distribution pourrait correspondre à la terminaison ségrégée des deux voies thalamocortical qui amènent l'information sensorielle dans le cortex cérébral. De plus, seul les neurones atteignant le seuil critique d'activation modifient leur expression génique en réponse à la stimulation sensorielle. Ces changements d'expression géniques vont permettre à la cellule de modifier ses propriétés structurales et physiologiques de manière a prevenir la propagation d'un excès d'activité neuronale au niveau de ses cibles postsynaptics. L'activité neuronale agit donc spécifiquement sur certains types cellulaires de maniere a induire une réponse homéostatique au niveau du réseau neuronal impliqué dans l'integration de l'information sensorielle. Nos travaux démontrent pour une première fois que les deux voies sensorielles contribuent d'une manière distincte et complémentaire à la plasticité corticale induite par un changement de l'activité sensorielle chez la souris adulte.

Conventional coronary magnetic resonance angiography (MRA) techniques display the coronary blood-pool along with the surrounding structures, including the myocardium, the ventricular and atrial blood-pool, and the great vessels. This representation of the coronary lumen is not directly analogous to the information provided by x-ray coronary angiography, in which the coronary lumen displayed by iodinated contrast agent is seen. Analogous "luminographic" data may be obtained using MR arterial spin tagging (projection coronary MRA) techniques. Such an approach was implemented using a 2D selective "pencil" excitation for aortic spin tagging in concert with a 3D interleaved segmented spiral imaging sequence with free-breathing, and real-time navigator technology. This technique allows for selective 3D visualization of the coronary lumen blood-pool, while signal from the surrounding structures is suppressed.

The functional interaction between fibroblast growth factor 23 (FGF-23) and Klotho in the control of vitamin D and phosphate homeostasis is manifested by the largely overlapping phenotypes of Fgf23- and Klotho-deficient mouse models. However, to date, targeted inactivation of FGF receptors (FGFRs) has not provided clear evidence for an analogous function of FGFRs in this process. Here, by means of pharmacologic inhibition of FGFRs, we demonstrate their involvement in renal FGF-23/Klotho signaling and elicit their role in the control of phosphate and vitamin D homeostasis. Specifically, FGFR loss of function counteracts renal FGF-23/Klotho signaling, leading to deregulation of Cyp27b1 and Cyp24a1 and the induction of hypervitaminosis D and hyperphosphatemia. In turn, this initiates a feedback response leading to high serum levels of FGF-23. Further, we show that FGFR inhibition blocks Fgf23 transcription in bone and that this is dominant over vitamin D-induced Fgf23 expression, ultimately impinging on systemic FGF-23 protein levels. Additionally, we identify Fgf23 as a specific target gene of FGF signaling in vitro. Thus, in line with Fgf23- and Klotho-deficient mouse models, our study illustrates the essential function of FGFRs in the regulation of vitamin D and phosphate levels. Further, we reveal FGFR signaling as a novel in vivo control mechanism for Fgf23 expression in bone, suggesting a dual function of FGFRs in the FGF-23/Klotho pathway leading to vitamin D and phosphate homeostasis.

The only long-term and cost-effective solution to the human immunodeficiency virus (HIV) epidemic in the developing world is a vaccine that prevents individuals from becoming infected or, once infected, from passing the virus on to others. There is currently little hope for an AIDS vaccine. Conventional attempts to induce protective antibody and CD8+ lymphocyte responses against HIV and simian immunodeficiency virus (SIV) have failed. The enormous diversity of the virus has only recently been appreciated by vaccinologists, and our assays to determine CD8+ lymphocyte antiviral efficacy are inadequate. The central hypothesis of a CTL-based vaccine is that particularly effective CD8+ lymphocytes directed against at least five epitopes that are derived from regions under functional and structural constraints will control replication of pathogenic SIV. This would be somewhat analogous to control of virus replication by triple drug therapy or neutralizing antibodies.

A promising approach to adoptive transfer therapy of tumors is to reprogram autologous T lymphocytes by TCR gene transfer of defined Ag specificity. An obstacle, however, is the undesired pairing of introduced TCRalpha- and TCRbeta-chains with the endogenous TCR chains. These events vary depending on the individual endogenous TCR and they not only may reduce the levels of cell surface-introduced TCR but also may generate hybrid TCR with unknown Ag specificities. We show that such hybrid heterodimers can be generated even by the pairing of human and mouse TCRalpha- and TCRbeta-chains. To overcome this hurdle, we have identified a pair of amino acid residues in the crystal structure of a TCR that lie at the interface of associated TCR Calpha and Cbeta domains and are related to each other by both a complementary steric interaction analogous to a "knob-into-hole" configuration and the electrostatic environment. We mutated the two residues so as to invert the sense of this interaction analogous to a charged "hole-into-knob" configuration. We show that this inversion in the CalphaCbeta interface promotes selective assembly of the introduced TCR while preserving its specificity and avidity for Ag ligand. Noteworthily, this TCR modification was equally efficient on both a Mu and a Hu TCR. Our data suggest that this approach is generally applicable to TCR independently of their Ag specificity and affinity, subset distribution, and species of origin. Thus, this strategy may optimize TCR gene transfer to efficiently and safely reprogram random T cells into tumor-reactive T cells.

The current drug options for the treatment of chronic Chagas disease have not been sufficient and high hopes have been placed on the use of genomic data from the human parasite Trypanosoma cruzi to identify new drug targets and develop appropriate treatments for both acute and chronic Chagas disease. However, the lack of a complete assembly of the genomic sequence and the presence of many predicted proteins with unknown or unsure functions has hampered our complete view of the parasite's metabolic pathways. Moreover, pinpointing new drug targets has proven to be more complex than anticipated and has revealed large holes in our understanding of metabolic pathways and their integrated regulation, not only for this parasite, but for many other similar pathogens. Using an in silicocomparative study on pathway annotation and searching for analogous and specific enzymes, we have been able to predict a considerable number of additional enzymatic functions in T. cruzi. Here we focus on the energetic pathways, such as glycolysis, the pentose phosphate shunt, the Krebs cycle and lipid metabolism. We point out many enzymes that are analogous to those of the human host, which could be potential new therapeutic targets.

Trypanosoma cruzi infection of the adipose tissue of mice triggers the local expression of inflammatory mediators and a reduction in the expression of the adipokine adiponectin. T. cruzi can be detected in adipose tissue by PCR 300 days post-infection. Infection of cultured adipocytes results in increased expression of cytokines and chemokines and a reduction in the expression of adiponectin and the peroxisome proliferator-activated receptor ³, both of which are negative regulators of inflammation. Infection also results in the upregulation of cyclin D1, the Notch pathway, and extracellular signal-regulated kinase and a reduction in the expression of caveolin-1. Thus, T. cruzi infection of cultured adipocytes leads to an upregulation of the inflammatory process. Since adiponectin null mice have a cardiomyopathic phenotype, it is possible that the reduction in adiponectin contributes to the pathogenesis of chagasic cardiomyopathy. Adipose tissue may serve as a reservoir for T. cruzi from which parasites can become reactivated during periods of immunosuppression. T. cruzi infection of mice often results in hypoglycemia. In contrast, hyperglycemia as observed in diabetes results in increased parasitemia and mortality. Adipose tissue is an important target tissue of T. cruzi and the infection of this tissue is associated with a profound impact on systemic metabolism, increasing the risk of metabolic syndrome.

”compositions” is a new R-package for the analysis of compositional and positive data.It contains four classes corresponding to the four different types of compositional andpositive geometry (including the Aitchison geometry). It provides means for computation,plotting and high-level multivariate statistical analysis in all four geometries.These geometries are treated in an fully analogous way, based on the principle of workingin coordinates, and the object-oriented programming paradigm of R. In this way,called functions automatically select the most appropriate type of analysis as a functionof the geometry. The graphical capabilities include ternary diagrams and tetrahedrons,various compositional plots (boxplots, barplots, piecharts) and extensive graphical toolsfor principal components. Afterwards, ortion and proportion lines, straight lines andellipses in all geometries can be added to plots. The package is accompanied by ahands-on-introduction, documentation for every function, demos of the graphical capabilitiesand plenty of usage examples. It allows direct and parallel computation inall four vector spaces and provides the beginner with a copy-and-paste style of dataanalysis, while letting advanced users keep the functionality and customizability theydemand of R, as well as all necessary tools to add own analysis routines. A la física o a l’enginyeria es defineix com fregament o fricció a la resistència que s'oposa a la rotació o al lliscament d'un cos sobre un altre, o també a la força que apareix en la superfície de contacte de dos cossos quan s'intenta lliscar un sobre un altre. El fregament ha estat fins avui dia un gran problema físic, això és degut a que genera imperfeccions, especialment microscòpiques, entre les superfícies en contacte. Aleshores és quan apareix la tribologia. La tribologia és la ciència que estudia la fricció, el desgast i la lubricació de superfícies en contacte. El lliscament entre superfícies sòlides es caracteritza generalment per un alt coeficient de fricció i un gran desgast a causa de les propietats específiques de lessuperfícies. La lubricació consisteix en la introducció d'una capa intermèdia d'un material aliè entre les superfícies en moviment. En aquest projecte hem intentat aplicar tècniques i mètodes de control els quals puguin arribar a millorar i perfeccionar els sistemes de mesura i control a nivell industrial i/o particular. En el nostre cas ha estat l’estudi dels olis lubricants usats en motors de combustió interna de variats vehicles com motocicletes, automòbils, camions o vaixells. Hem introduït una millora d’automatització mitjançant un circuit pneumàtic al captador de partícules, hi hem introduït una tècnica per captar partícules a partir de filtres de membrana, hem estudiat les mostres i llurs ferrogrames i membranes amb la finalitat de detectar les anomalies dels motors. L’objectiu del treball és l’estudi del desgast originat en motors de combustió interna, majoritàriament, Dièsel de camions, automòbils i vaixells. Això, comprèn la captació de partícules en ferrografies, la seva observació i anàlisi en microscopi, la seva classificació, comparació i la detecció d’anomalies en els motors. Per altra banda, també s’aprofundirà en les tècniques d’anàlisi, la lubricació i manteniment dels motors i el nou disseny i validació d’un captador de partícules automatitzat

Oenocytes are ectodermic cells present in the fat body of several insect species and these cells are considered to be analogous to the mammalian liver, based on their role in lipid storage, metabolism and secretion. Although oenocytes were identified over a century ago, little is known about their messenger RNA expression profiles. In this study, we investigated the transcriptome of Aedes aegypti oenocytes. We constructed a cDNA library from Ae. aegypti MOYO-R strain oenocytes collected from pupae and randomly sequenced 687 clones. After sequences editing and assembly, 326 high-quality contigs were generated. The most abundant transcripts identified corresponded to the cytochrome P450 superfamily, whose members have roles primarily related to detoxification and lipid metabolism. In addition, we identified 18 other transcripts with putative functions associated with lipid metabolism. One such transcript, a fatty acid synthase, is highly represented in the cDNA library of oenocytes. Moreover, oenocytes expressed several immunity-related genes and the majority of these genes were lysozymes. The transcriptional profile suggests that oenocytes play diverse roles, such as detoxification and lipid metabolism, and increase our understanding of the importance of oenocytes in Ae. aegypti homeostasis and immune competence.
