We determined the effects of moxonidine and rilmenidine 20 mol (alpha(2)-adrenergic and imidazoline receptor agonists) injected into the medial septal area (MSA) on the pilocarpine-induced salivation, when injected intraperitoneally (i.p.), of male Holtzman rats weighing 250300 g, with stainless-steel cannula implanted into the MSA. The rats were anesthetized with zoletil 50 mg kg(-1) b.wt. (tiletamine chloridrate 125.0 mg and zolazepan chloridrate 125.0 mg) into quadriceps muscle intramuscularly (IM), saliva was collected using pre-weighed small cotton balls inserted in the animal's mouth. The pre-treatment with moxonidine injected into the MSA reduced the salivation induced by pilocarpine (1 mg kg(-1)) injected i.p. (12 +/- 3 mg min(-1)) vs. control (99 +/- 9 mg min(-1)). The pre-treatment with rilmenidine 40 nmol also reduced the salivation induce by pilocarpine injected i.p. (20 +/- 5 mg min(-1)) vs. control (94 +/- 7 mg min(-1)). Idazoxan 40 nmol (imidazoline receptor antagonist) injected into the MSA previous to moxonidine and rilmenidine partially blocked the effect of moxonidine and totally blocked the rilmenidine effect in pilocarpine-induced salivation injected i.p. (60 +/- 8 and 95 +/- 10 mg min(-1), respectively). Yohimbine 40 nmol (alpha(2)-adrenergic receptor antagonist) injected into the MSA previously to moxonidine and rilmenidine partially blocked the moxonidine effect but produced no change on the rilmenidine effect on i.p. pilocarpine-induced salivation (70 +/- 6 and 24 +/- 6 mg min(-1), respectively). Injection of these alpha(2)-adrenergic and imidazoline agonists and antagonists agents i.p. produced no change on i.p. pilocarpine-induced salivation. These results show that central, but not peripheral, injection of alpha(2)-adrenergic and imidazoline agonists' agents inhibit pilocarpine-induced salivation. Idazoxan, an imidazoline receptor antagonist, totally inhibits the rilmenidine effect and partially inhibits the moxonidine effect on pilocarpine-induced salivation. Yohimbine produced no change on rilmenidine effect but partially inhibited the moxonidine effect. Both of these antagonists when injected into the MSA previous to pilocarpine i.p. potentiated the sialogogue effect of pilocarpine. The results suggest that alpha(2)-adrenergic/imidazoline receptor of the MSA when stimulated blocked pilocarpine-induced salivation in rats when injected intraperitonially These receptors of the medial septal area have an inhibitory mechanism on salivary secretion. The aim of the present study was to analyze the role of alpha(1),alpha(2)-adrenoceptors, and the effects of losartan and PD123319 (selective ligands of the AT(1) and AT(2) angiotensin receptors, respectively) injected into the paraventricular nucleus (PVN) on the diuresis, natriuresis, and kaliuresis induced by administration of adrenaline into the medial septal area (MSA). Male Holtzman rats with a stainless steel cannula implanted into the MSA and bilaterally into the PVN were used. The administration of adrenaline into the MSA increased in a dose-dependent manner the urine, sodium, and potassium excretions. The previous administration of prazosin (an alpha(1)-adrenoceptor antagonist) injected into the PVN abolished the above effects of adrenaline, whereas yohimbine (an a-adrenoceptor antagonist) doesn't affect the diuresis, natriuresis, and kaliuresis induced by adrenaline. Pretreatment with losartan into the PVN decreased in a dose-dependent manner the urine, sodium, and potassium excretions induced by MSA administration of adrenaline (50 ng), while PVN PD123319 was without effect. These results indicate that urinary and electrolyte excretion effects induced by adrenaline into the MSA are mediated primarily by PVN AT, receptors. However, the doses of losartan were more effective when combined with the doses of PD123319 than given alone, suggesting that the urinary, natriuretic, and kaliuretic effects of MSA adrenaline may involve activation of multiple angiotensin II receptors subtypes into the PVN. The preventative intestinal anti-inflammatory activity of paepalantine, an isocoumarin isolated from the capitula of Paepalarithus bromelioides, was tested in the trinitrobenzenesulphonic acid (TNBS) model of rat colitis. This was performed in two different experimental settings, i. e. when the colonic mucosa is intact or when the mucosa is in process of recovery after an initial insult. The results obtained revealed that the paepalantine pretreatment, at doses of 5 and 10 mg/kg, significantly attenuated the colonic damage induced by TNBS in both situations, as it was evidenced both histologically and biochemically. This beneficial effect was associated with an improvement in the colonic oxidative status, since paepalantine prevented the glutathione depletion that occurred as a consequence of the colonic inflammation. In addition, the intestinal anti-inflammatory effect exerted by this isocoumarin was associated with an inhibition of colonic nitric oxide activity, which is upregulated as a consequence of the inflammatory process. In conclusion, the preventative effect exerted by paepalantine in the TNBS model of rat colitis is probably related with its antioxidant properties.

The methanolic extract of the leaves of the medicinal plant Byrsonima crassa (Malpighiaceae) contain flavonoids with antioxidant activity. They were separated in a preparative scale using high-speed counter-current chromatography. The optimum solvent system used was composed of a mixture of ethyl acetate-n-propanol-water (140:8:80 (v/v/v)) and led to a successful separation between monoglucosilated flavonoids (quercetin-3-O-alpha-L-arabinoside, quercetin-3-O-beta-D-galactoside) and the biflavonoid amentoflavone in only 3.5 h. The purities of quercetin-3-O-alpha-L-arabinoside (95 mg), quercetin-3-O-beta-D-galactoside (16 mg) and the biflavonoid amentoflavone (114 mg) were all isolated at purity over 95%. A Pavlovian conditioning paradigm was used to induce a connection between a conditioned stimulus, light (CS), associated with an unconditioned stimulus, confinement (US) in Nile tilapia Oreochromis niloticus, which resulted in a conditioned endocrine response (CR) to the CS alone manifested as an increase in plasma cortisol. Individual isolated Nile tilapia were submitted for 10 days to the conditioning treatment consisting of turning on a light (CS) for I min with subsequent 30 min confinement (US). on the 10th day of the experiment, plasma cortisol was not increased when fish were subjected to no handling at all, or only light, or even a daily stressor for the 9 days. on the other hand, at the 10th day cortisol was significantly increased only when light was presented either with or without pairing with the stressor. These results confirmed that the cue, light (CS), was not stressful in itself, but when given as the CS in the absence of the US post conditioning the hypothalamus-pituitary-interrenal axis was activated. Therefore, it was concluded that memory of a previous experience with a stressor can be recalled by a conditioned stimulus and induce stress, which is the first demonstration of a memory-induced stress in fishes. This study tested the use of ventilatory frequency (VF) as an indicator of stress in the Nile tilapia, Oreochromis niloticus (L.). Firstly, we tested the relationship between VF and plasma cortisol after confinement. Confined fish showed higher VF and plasma cortisol levels, but the latter continued to increase significantly for longer time than VF. Secondly, we conducted another experiment to test the use of VF as indicator of fish stress. In four out of six treatment, we confined the fish for different intervals (30 s, 5, 15 or 30 min). The others were used as control. In one, no handling was imposed. The other control consisted of introducing the partition (the same used to perform the confinement) into the aquarium for less than 4 s, without confinement and immediately removing the partition (partition control). Ventilatory frequency was increased for the partition control as much as for the longer duration of confinement. This clearly indicates that VF is a very sensitivity response to disturbance, but of limited use because this parameter does not reflect the severity of the stimulus. Thus, although VF is a non-invasive technique that does not require sophisticated recording equipment, its usefulness is limited. (C) 2004 Elsevier B.V. All rights reserved.

The abundance and ecologic distribution of the shrimp R. constrictus were compared among three different embayments within the northern coast of São Paulo State (23degreesS, 45degreesW); i.e. Mar Virado (MV), Ubatuba (UBA) and Ubatumirim (UBM). Key environmental factors were monitored for a better understanding of the distributional patterns of this species, namely temperature, salinity, depth, organic matter content and texture of sediments. In each bay, six transects were delimited, four parallel to the coastline and the other two in areas adjacent to rocky shores. Monthly samples were taken over a 2-year period (1998 and 1999), using a shrimp fishing boat equipped with double-rig nets. Abundance data were treated separately according to environmental characteristics. A total of 4978 specimens was obtained; 921 at MV, 1948 at UBA and 2109 at UBM. The spatial distribution of R. constrictus differed among bays. Higher abundance values were recorded at areas where silt and clay comprised less than 70% of the sediment. A more diversified sediment at UBM and UBA apparently favours the establishment of this species in the region. Its abundance also followed a seasonal trend, higher during spring and summer when intrusions of cold South Atlantic Coastal Waters (SACW) are frequent, promoting the migration of this shrimp to more sheltered areas. These results suggest that sediment type and temperature are the most important variables affecting the spatial and seasonal distribution of this species.

The venom of the neotropical social wasp Agelaia pallipes pallipes was fractionated by RP-HPLC resulting in the elution of seven fractions; the last two were re-fractionated under RP-HPLC by using isocratic elution conditions and the purity of the fractions were confirmed by using ESI-MS analysis. Both fractions are constituted of peptide components, which were sequenced by Edman degradation chemistry, resulting in the following sequences:Protonectin I-L-G-T-I-L-G-L-L-K-G-L-NH2Agelaia-MP I-N-W-L-K-L-G-K-A-I-I-D-A-L-NH2Both peptides are manually synthesized on solid-phase and functionally characterized by using Wistar rats cells. Protonectin is a non-hemolytic chemotactic peptide for polymorphonucleated leukocytes (PMNL), presenting some mast cell degranulating activity and potent antimicrobial action both against Gram-positive and Gram-negative bacteria. Agelaia-MP was characterized as a hemolytic mast cell degranulator toxin, presenting a poor antimicrobial action and no chemotaxis for PMNL. (C) 2004 Elsevier Ltd. All rights reserved.

The consequences of introducing Cichla cf. monoculus Spix & Agassiz, Astronotus ocellatus (Agassiz) and Pygocentrus nattereri Kner into lakes in the River Doce basin, Brazil, on richness, diversity and efficiency of aquatic macrophytes as natural refugia to native fishes was investigated. Samples were taken from lakes with and without alien fishes in areas with and without aquatic macrophytes. The presence of alien fishes reduced richness and diversity of the native fish community. The refugia function, which could be attributed to the clustering of aquatic macrophytes, does not exist in these lakes probably because the alien fishes exploit such habitats for reproduction. Since introductions threaten the native fish diversity of the region, studies on regional dispersion and factors that minimise the spread of alien fishes are needed.

O desempenho na canoagem de velocidade depende da capacidade de o organismo regenerar ATP em grandes quantidades e a altas taxas, a partir das diferentes vias metabólicas. Assim, o objetivo do presente estudo foi combinar dois modelos bioenergéticos, um genérico denominado de potência crítica e outro específico para a canoagem, proposto por Zamparo et al. (1999), na tentativa de produzir estimativas de aptidão aeróbia e anaeróbia para essa modalidade, bem como estabelecer estimativas não-invasivas da contribuição dos sistemas aeróbio e anaeróbio para diferentes distâncias percorridas. Para tanto, 11 atletas de canoagem (16,0 ± 1,2 anos; 174,0 ± 2,4cm; 65,2 ± 4,4kg), do sexo masculino, percorreram diferentes distâncias (500, 1.000 e 1.790m), na máxima velocidade possível, em embarcações do tipo K-1, em um lago com águas calmas. As informações obtidas foram inicialmente convertidas em quantidades geradas de trabalho (kJ) e potência interna (W). Posteriormente, os valores individuais estimados foram aplicados às três equações preditivas da potência crítica (PCrit) e capacidade de trabalho anaeróbio (CTAnaer). Por fim, os valores produzidos foram transformados em unidades de equivalentes de oxigênio para a estimativa da contribuição aeróbia (equivalente de O2 para a PCrit x tempo para a distância) e anaeróbia (equivalente de O2 para a CTAnaer x tempo para a distância), nas diferentes distâncias. A contribuição aeróbia relativa encontrada para as diferentes distâncias analisadas (500, 1.000 e 1.790m) foi de 60,6, 78,6 e 89,4%, respectivamente. Os resultados encontrados confirmaram as informações produzidas anteriormente por outras investigações, o que sugere que os procedimentos adotados neste estudo podem fornecer estimativas confiáveis sobre a participação das vias energéticas no desempenho de canoagem.

The flutamide antiandrogenic effects oil the Guinea pig male prostate morphology in puberal, post-puberal and adult ages were evaluated in the present study. Daily-treated group animals received flutamide Subcutaneous injection at a dose of 10 mg/Kg body weight for 10 days. The control group animals received a pharmacological vehicle under the same conditions. The lateral prostate was removed, fixed and processed for light and transmission electron microscopy. The results revealed all increase of the acinus diameter in the treated puberal animals and straitness in the stromal compartment around the acini. The epithelial cells exhibited cubic phenotype. In the post-puberal and adult animals, a decrease of the acinus diameter was observed, as well as an increase of the smooth muscle layer and presence of the folds at epithelium. The ultrastructural evaluation of the secretory cells in the treated group demonstrated endomembrane enlargement, mainly in the rough endoplasmic reticulum and Golgi apparatus. In addition, a decrease of the microvilli and alterations in the distribution patterns and density of the stromal fibrillar components were observed. In Conclusion, the flutamide treatment exerts tissue effects oil the lateral prostate, promoting stroma/epithelium alterations.

O presente trabalho caracteriza o gene codificador da proteína capsidial do isolado do Grapevine virus A (GVA) encontrado no Estado de São Paulo (GVA-SP). RNA total foi extraído de folhas e pecíolos de plantas de videira (Vitis spp.) da variedade 'Kober 5BB' e submetido a RT-PCR usando oligonucleotídeos desenhados para amplificar um fragmento entre as posições 6409 e 7175 do RNA do GVA (GenBank, acesso X75433). Foi obtido um fragmento de tamanho esperado (767 nt) que inclui o gene da proteína capsidial, codificando 198 aminoácidos. A seqüência do GVA-SP apresentou similaridade de nucleotídeos e aminoácidos de, respectivamente, 86-92,3% e 94,5-98% com isolados do GVA da Europa, África e Japão (Acessos X75433, AF441234, AF007415, AB039841) e da região Sul do Brasil (Acesso AF494187), sendo, entretanto, mais similar aos isolados africano e italiano.

A systematic study of the root-mean-square distance between the constituents of weakly-bound nuclei consisting of two halo neutrons and a core is performed using a renormalized zero-range model. The radii are obtained from a universal scaling function that depends on the mass ratio of the neutron and the core, as well as on the nature of the subsystems, bound or virtual. Our calculations are qualitatively consistent with recent data for the neutron-neutron root-mean-square distance in the halo of Li-11 and Be-14 nuclei. (C) 2004 Published by Elsevier B.V.
