A nonpathogenic mutant of Colletotrichum magna (path-1) was previously shown to protect watermelon (Citrullus lanatus) and cucumber (Cucumis sativus) seedlings from anthracnose disease elicited by wild-type C. magna. Disease protection was observed in stems of path-1-colonized cucurbits but not in cotyledons, indicating that path-1 conferred tissue-specific and/or localized protection. Plant biochemical indicators of a localized and systemic (peroxidase, phenylalanine ammonia-lyase, lignin, and salicylic acid) “plant-defense” response were investigated in anthracnose-resistant and -susceptible cultivars of cucurbit seedlings exposed to four treatments: (1) water (control), (2) path-1 conidia, (3) wild-type conidia, and (4) challenge conditions (inoculation into path-1 conidia for 48 h and then exposure to wild-type conidia). Collectively, these analyses indicated that disease protection in path-1-colonized plants was correlated with the ability of these plants to mount a defense response more rapidly and to equal or greater levels than plants exposed to wild-type C. magna alone. Watermelon plants colonized with path-1 were also protected against disease caused by Colletotrichum orbiculare and Fusarium oxysporum. A model based on the kinetics of plant-defense activation is presented to explain the mechanism of path-1-conferred disease protection.

These diaries of Benjamin Guild document his travels as a Presbyterian pastor in Massachusetts and Rhode Island. The daily entries describe people Guild met and dined with, the food he ate (including strawberries, currants, watermelon, English cherries, and lobster), the funerals he attended, and the sermons he gave. Many entries relate to his health concerns (the ague and eye trouble), sleeping habits, and widespread public health concerns (including smallpox, dysentery, "nervous fevers," consumption, and "putrid fever"). The diaries also contain passing references to the activities of American, British, French, and German soldiers during the American Revolution; the invasion of Canada and battles occurring in New York are noted. In August 1778, after visiting Providence, Rhode Island, Guild comments on the disordered state of the city after American soldiers passed through it. He also recounts a visit by officers of the French fleet to the Harvard College library in September 1778 and describes his dinner on board the French man-of-war, Sagitaire. One entry describes an elaborate ball sponsored by John Hancock, held for French soldiers and "Boston ladies," and another refers to the "incursion" of Indians. Many of Guild's diary entries pertain to his work as a Harvard College Tutor; these entries describe his lectures at the College, meetings with colleagues, personnel decisions, and the examination of students. He also describes books he is reading and his opinions of them, the purchase and sale of books, and his desire to learn Hebrew and French. In addition, multiple entries refer to a man named Prince, who was perhaps Guild's slave. Prince sometimes accompanied Guild on his travels.

CHAPTER 1 - The gummy stem blight, caused by the fungus D. bryoniae, is a disease commonly found in watermelon cultivated in several countries. In Brazil, there are numerous studies related to the disease, but there are not uniform methods for quantifying of disease severity in the field. Thus, we developed a diagrammatic scale based on scanned photos of watermelon leaves infected with D. bryoniae. The scale developed showed levels of 0; 10; 20; 45; 65 and 90% of severity. The scale validation was divided into two parts: initially, 10 evaluators (half with experienced and other half without experience) estimated the disease severity based on the initial observation of 100 photos of watermelon leaves with symptoms of the disease at different severity levels. Before, the same evaluators estimated the disease severity with the support of the scale prepared from the Quant program. Data were analyzed using linear regression and were obtained angular, linear, and correlation coefficients. Based on these data, we determined the accuracy and precision of the evaluations. The correlation coefficients (R2) ranged from 0.88 - 0.97 for the experienced evaluators and from 0.55 - 0.95 for the inexperienced evaluators. The average angular coefficient (A) for inexperienced evaluators was 20.42 and 8.61 with and without the support of diagrammatic scale, respectively. Experienced evaluators showed values of average linear coefficient of 5.30 and 1.68 with and without the support of diagrammatic scale, respectively. The absolute errors analysis indicated that the use of diagrammatic scale contributed to minimize the flaws in the severity levels estimation. The diagrammatic scale proposed shown adequate for gummy stem blight severity evaluation in watermelon. CHAPTER 2 - The gummy stem blight (Didymella bryoniae) is a disease that affects the productivity of watermelon leading to losses over 40%. This study aimed to evaluate the efficiency of different production systems in control of gummy stem blight in watermelon for to establish efficient methods to combat the disease. There were applied the following treatments: conventional tillage (T1), integrated management (T2) and organic management (T3). In T1 and T2 were applied mineral fertilization and T3 was used bovine manure. There was application of fungicides and insecticides in commercial dose in T1 and T2, being after soil chemical analysis in T2. Disease severity was assessed by grading scale. The experimental design was randomized blocks. The severity of gummy stem blight has increased substantially during the fruit formation. Watermelon plants grown with integrated management (T2) showed lower levels of disease severity, while plants in organic management (T3) exhibited higher levels of severity. We conclude that management based on judicious accompaniments in field represents best way to achieve the phytosanitary aspect adequate for cultivation of watermelon in Tocantins.

A cultura da melancia, variedade Charleston Gray, vem se comportando muito bem no submedio Sao Francisco, no Nordeste do Brasil, a producao estilar vem limitando o desenvolvimento da cultura na regiao, provocando perdas que podem atingir 40% da produtividade. Este trabalho apresenta os resultados de um estudo conduzido com dois tratamentos, um modificado e outro tradicional. Este representou a temologia dos produtores, aquele utilizou as inovacoes tecnologicas recomendades pela pesquisa. O principal resultado obtido com o tratamento diz respeito a reducao da podridao estilar.

O objetivo deste trabalho foi avaliar o desempenho agronômico em termos de produtividade e características de frutos em 32 genótipos de melancia. O ensaio foi conduzido na Embrapa Semi-Árido, em Petrolina-PE, durante o período de agosto a outubro de 2005, utilizando o delineamento experimental de blocos ao acaso, com quatro repetições e parcelas com 10 plantas. As mudas foram transplantadas para o campo no espaçamento de 3 x 0,8 m. A cv ?C. Sweet? e as 31 linhas originadas do cruzamento entre ?C. Sweet? e ?CPATSA 85-030? foram avaliadas quanto a rendimento e aspectos externos e internos dos frutos. Verificou-se alto rendimento dos genótipos (33,6 a 66,7 t/ha). Na maioria das linhas, predominaram os frutos com casca rajada, com listras verde-claras alternadas com verde-escuras (96,8%), polpa vermelha (83,9%), alto teor de sólidos solúveis (10 a 12,3o Brix) (93,5%), frutos grandes (8,60 a 11,70 kg) (54,8%) e formato arredondado. Algumas linhas (9,7%) apresentaram alta prolificidade (2,9 a 2,5 frutos/planta), frutos pequenos e arredondados (4,30 a 4,60 kg) e casca rajada verde escura (3,2%). Entre os genótipos de melancia, 05.1168.003, 05.1172.004, 05.1185.001, 05.1189.003, 05.1203.007/1, 05.1194.001/1, 05.1194.005 e 05.1194.006 foram os mais promissores para a obtenção de cultivares resistentes ao oídio e com padrões de frutos para os mercados interno e externo.

This paper presents the results of four field experiments carried out with the objetive of evaluating the feaibility of irrigation by porous capsule method, from 1979 to 1983, at Bebedouro Experiment Station, EMBRAPA-CPATSA, Petrolina, PE, Brazil. The irrigation system consisted of fulcrum of cone shaped porous capsules, interconnected with conduit pipe and installed in the soil at equidistance and 0.10 m deep along contour lines. The hydrostatic pressures studies did not significantly influence the crop yield, but influenced, at 0.10 level, the daily water release from porous unit. The mean yields for watermelon (Citrullus vulgaris Shard), var. Charleston Gray, for muskmelon (Cucumis melo L.) var. Valenciano Amarelo, and for maize (Zea mays L.), var. Centralmex, estimated in ton/2,500 units/ha or in cobs/2,500 units/ha, were 28.5, 10 and 17,500, respectively. The water consumption for watermelon, musk melon and maize was, respectively, 60 mm, 60 mm and 100 mm in a deep sandy yellow-red latosol. The cost of the system was US$ 1.677,41/ha.

O controle de predadores em plantio de melancia deve seguir o manejo integrado de pragas (MIP). No entanto, como as informações não existem ou são muito escassas para esse cultivo, a sugestão é adotar os indicadores do manejo integrado do melão, considerando-se que é a mesma família botânica da melancia. Para adoção do MIP, é fundamental o conhecimento da fenologia da cultura, a identificação e o monitoramento das populações de insetos-pragas.

The availability of fresh-cut fruit (FCF) in the marketplace has been increasing in Portugal, although reports of its microbial quality are not known. Due to the growing concerns of these commodities over their microbial safety, the objectives of this work were to study the microbiological quality and prevalence of Salmonella and Listeria monocytogenes on fresh-cut fruits sold in southern Portugal. A study to examine the changes in pH and microbial counts, before and after the expiration dates, was also made. A total of 160 samples was purchased in the local grocery stores between September 2011 and August 2014, before their sell-by date. These samples were assayed for aerobic mesophilic (AM) and psychrotrophic (AP) microorganisms, yeasts and molds (YM), lactic-acid bacteria (LAB), coliforms (TC), Escherichia coli and coagulase positive staphylococci as well as L. monocytogenes and Salmonella. The microbiological counts ranged from 3.0-9.2 lg cfu/g (AM); 2.2–10.7 lg cfu/g (AP); 2.3–10.4 lg cfu/g (YM); 1.9–9.0 lg cfu/g (LAB) and less than 1–9.1 lg cfu/g (TC). The melons and watermelon presented the highest levels of the microbial quality parameters studied. However, no E. coli, staphylococci, Salmonella and L. monocytogenes were detected in any of the samples. After the sell-by date, an increase of the AM, AP, LAB and YM values was observed in all fruits. Conversely, the differences found in TC counts before and after the best-before date had no statistical significance. A decrease in pH was observed in all fruits except pineapple whose pH slightly increased after 14 days of storage. The results highlight the importance of preventing contamination and cross contamination, selecting adequate decontamination technologies and maintaining a strict temperature control during processing, distribution and selling of FCF.

Mixed infections in cucurbits are frequently observed in natural conditions between viruses from the Potyvirus genus and Cucumber mosaic virus (CMV), which significantly decreases productivity. The objectives of the present study was to compare the host range of PRSV-W, WMV, and ZYMV isolates and evaluate the effects of mixed infections with CMV in zucchini plants (Cucurbita pepo L.). Host range studies comprising 23 plant species confirmed some similarities and biological differences among the isolates of PRSV-W, ZYMV, and WMV. RT-PCR confirmed the amplification of DNA fragments of the PRSV-W, WMV, and ZYMV coat protein gene (cp) and cytoplasm inclusion gene (ci). The virus interaction studies in zucchini Caserta plants indicated synergistic interactions, particularly among species from the Potyvirus genus, and some CMV interference with some virus combinations.
