建立了一个新的非定常脑AVM血液动力学模型。模型克服了以往的定常AVM模型无法反映血流实际脉动,以及模型结构和血管性质描述上的缺陷,并依据临床实测数据提出了一种更为符合生理和病理实际的描述AVM供血动脉扩张现象的方法。模型模拟了AVM造成的脑血管压力、流量波形和脑血管输入阻抗的变化,并对脑AVM病人脑血管系统的输入阻抗进行了分析。为研究具有复杂结构的脑AVM血液动力学的特征,提供了一个有效的工具。

Malformation rates in embryos of dab, whiting, cod, flounder and plaice have been monitored for several years (1984-2006) in the Southern North Sea. For embryos of all species investigated trends for the fluctuation of malformation rates over the time were registered in the areas showing intermediate prevalences at the beginning of the studies in 1984 and maxima in 1987. Thereafter for all species a decrease of malformation rates was found until 2006 excepting an increase in 1996. A significant negative correlation existed between surface water temperature and prevalences of malformed embryos of dab and other species.

The trends of malformation prevalence in embryos of dab, Limanda limanda, in the southern North Sea after the year 1990 mirrored the drop in major pollutants in the rivers draining into the German Bight. Despite this general decline we detected a pollution event in the southern North Sea in winter 1995/1996 employing the prevalence of malformations in dab embryos as an indicator. An abrupt rise in malformation prevalence in the embryos of dab, corresponded to a dramatic increase in DDT levels in parent fish from the same area, indicating a hitherto unnoticed introduction of considerable quantities of DDT into the system. This input could be traced back to discharges of unknown origen into the River Elbe.

Malformation rates in fish embryos have been monitored for several years in the Southern North Sea. Their occurrence was interpreted to be related to pollution because malformation rates were highest in near coastal waters known to receive high pollution loads. For embryos of all species investigated synchronous trends for the fluctuation of malformation rates over the time were registered in the areas covered with intermediate prevalences at the beginning of the studies in 1984 and maxima in 1987. Thereafter malformation rates of all species decreased significantly followed by an increase in 1996. It was found that a significant negative correlation between surface water temperature and prevalences of malformed embryos of dab (Limanda limanda) and other species existed over time and space. These correlations became increasingly visible with decreasing concentrations of organochlorines in livers of dab. From these findings it is concluded that temperatures possibly predispose developing fish embryos to the impact of pollutants.

Esta pesquisa teve por objetivo compreender as vicissitudes da experiência de tornar-se mãe de um bebê em situação de risco neonatal. Emergiu da experiência da autora no acompanhamento psicológico prestado às mães e familiares de recém-nascidos de alto risco internados em uma Unidade de Terapia Intensiva Neonatal (UTI Neonatal). Foi possível perceber ali que a puérpera que tinha seu filho internado em situação de risco logo após o nascimento vivenciava uma experiência de intenso sofrimento, permeada por conflitos e angústias específicas, tais como: o sentimento de incapacidade pelo parto prematuro e/ou pela malformação fetal, a dor/luto diante da situação de risco e eminência de perda do filho, o medo e a ambivalência na relação afetiva com o bebê. Tais vivências podem representar uma ameaça para a construção do vínculo inicial entre pai/mãe/filho, bem como para a própria saúde psíquica da mulher e do bebê em constituição. Para uma aproximação da experiência subjetiva destas mulheres/mães, optou-se pela pesquisa qualitativa na abordagem psicanalítica. A compreensão do fenômeno foi possibilitada pela análise do discurso das mães que tiveram seus filhos internados na UTI Neonatal, através do método de observação participante dos atendimentos grupais prestados pelo Serviço de Psicologia da instituição, bem como pela análise documental das fichas de acompanhamento psicológico destas mulheres. Participaram do estudo, as mulheres/mães que acompanharam seus filhos internados na UTI Neonatal do Núcleo Perinatal do Hospital Universitário Pedro Ernesto (HUPE), da Universidade do Estado do Rio de Janeiro (UERJ) e que receberam atendimento psicológico grupal: Grupo Mães Presentes durante um período de três meses. A compreensão e interpretação dos aspectos essenciais do fenômeno foram fundamentadas nos pressupostos psicanalíticos da teoria do amadurecimento pessoal de Donald W. Winnicott e em outros autores atuais de referência no campo materno-infantil. Acredita-se que este trabalho servirá de reflexão e contribuição para a construção, na assistência neonatal, de um lugar de acolhimento para a experiência subjetiva dessas mulheres/mães e suas repercussões, a partir de uma perspectiva de cuidado humanizado e integral a saúde, tal como preconizado pelas políticas públicas atuais.

O aumento da prevalência da obesidade e osteoporose, bem como a identificação de mecanismos comuns que ligam a osteogênese e a adipogênese, sugerem que a obesidade e osteoporose podem ser distúrbios relacionados, e além disso, ambos podem ter suas origens no início da vida. Em 3 modelos diferentes de plasticidade ontogenética foi observado obesidade na vida adulta. Sendo assim, o objetivo deste trabalho foi investigar o impacto desses 3 modelos, o desmame precoce mecânico (DPM) e o farmacológico (DPF), e a supernutrição neonatal (SN) no tecido ósseo da prole durante o desenvolvimento. Para tanto, 2 experimentos foram realizados. No experimento 1, ratas lactantes foram divididas em 3 grupos: controle - os filhotes tiveram livre acesso ao leite durante toda a lactação; DPM - as mães foram envolvidas com uma atadura nos últimos 3 dias de lactação; DPF - as mães foram tratadas com bromocriptina (0,5 mg/duas vezes/dia) 3 dias antes do desmame padrão. No experimento 2, o tamanho da ninhada foi reduzido para 3 filhotes machos no 3o dia de lactação até o desmame (SN); o grupo controle permaneceu com 10 filhotes durante toda a lactação. Realizou-se absorciometria de raios-x de dupla energia, tomografia computadorizada, microtomografia computadorizada, teste biomecânico e análises séricas. Os dados foram considerados significativos quando P<0,05. No experimento 1, ao desmame, os filhotes DPM e DPF apresentaram menor massa corporal, massa gorda, densidade mineral óssea total (DMO), conteúdo mineral ósseo total (CMO), área óssea e osteocalcina sérica, e maior telopeptídeo carboxi-terminal do colágeno tipo I (CTX-I). O cálcio ionizado sérico foi menor apenas na prole DPM, a 25-hidroxivitamina D (25(OH)D) foi maior e o PTH menor apenas na prole DPF. Aos 180 dias, as proles DPM e DPF apresentaram maior massa corporal, maior massa de gordura visceral, hiperleptinemia, maior 25(OH)D e menor CTX-I. Ambos os grupos apresentaram aumento da DMO total, do CMO, da DMO da coluna vertebral e da área óssea aos 150 e 180 dias de idade. Nas avaliações ósseas individuais, as proles DPM e DPF também apresentaram aumento da DMO do fêmur e da vértebra lombar, da radiodensidade da cabeça femoral e do corpo vertebral; melhora da microarquitetura trabecular óssea e da resistência óssea. No experimento 2, observamos aumento da massa corporal, da massa gorda e da massa magra, do CMO e da área óssea no grupo SN desde o desmame até a idade adulta. Aos 180 dias, a prole SN também apresentou aumento da DMO total, da DMO do fêmur e da vértebra lombar, da radiodensidade da cabeça femoral e do corpo vertebral; melhora da microarquitetura trabecular óssea e da resistência óssea, maior osteocalcina e menor CTX-I. Demonstramos que, apesar de fatores de imprinting opostos, ambos os modelos causam melhora da massa, do metabolismo, da qualidade e da resistência óssea. Porém, parece que este efeito protetor sobre o tecido ósseo não é um resultado direto da programação deste tecido, mas sim consequência das alterações fisiopatológicas da obesidade programada pelos três modelos.

Esta tese discute o impacto do Diagnóstico de Malformação fetal na experiência das gestantes usuárias do SUS na Bahia, destacando as noções de dia-gnosis e pro-gnosis desenvolvidas por Gross e Shuval (2008) de forma associada à medicina do risco no encontro médico-paciente. Destaca o discurso biomédico na formatação diagnóstica, as diferentes percepções de risco e o forte engajamento das usuárias frente às tecnologias pré-natais e intervenções cirúrgicas neonatais, caucionado na esperança de que o avanço da ciência seja capaz de reverter ou abrandar a condição do seu feto/bebê. È diante da responsabilização da mulher por não ter produzido um feto/bebê saudável, mas um feto/bebê malformado, que se observa a prevalência de normas culturais e de gênero que conferem à maternidade um lugar de autossacrificio, de dedicação e criação dos filhos, como também status social O espaço pré-natal é marcado pela ausência de discussão a respeito do prognóstico de tais condições, com a consequente busca pelas gestantes do conhecimento por meio da internet, da opinião do marido e da crença religiosa que servem de alicerce para lidar com a antecipação da deficiência. As gestantes acreditam ser este um desígnio de Deus, uma espécie de provação e uma prova de amor incondicional ao futuro filho com deficiência (que poderá ou não sobreviver). A maioria das gestantes, 20 entrevistadas, prefere, contudo, ter um filho com deficiência do que sofrer sua perda. Em outra vertente, a tese analisa a forma como se organiza o sistema de saúde quanto à detecção de uma malformação congênita, apontando a precariedade da rede de atenção básica quanto à qualificação dos profissionais e o devido encaminhamento referente ao serviço especializado. A tecnologia de visualização o ultrassom obstétrico é a primordial ferramenta para detecção de alguma alteração fetal, porém somente ocorre o esclarecimento do diagnóstico de malformação fetal no serviço público de referência em medicina fetal em Salvador, Bahia. Destaca-se a falta de uma política pública do Ministério da Saúde que norteie o desenvolvimento da medicina fetal no Brasil, haja vista os diferentes impactos diante das tecnologias de inovação em saúde que geram vulnerabilidades e desigualdades sociais. Enfatiza-se a necessidade de uma revisão quanto à regulamentação do uso do ultrassom obstétrico que impeça o uso abusivo ou sua omissão diante dos crescentes casos de anomalias congênitas.

Climbing perch locally known as koi (Anabas testudineus) is a popular food fish in our country. Thai climbing perch was introduced in Bangladesh from Thailand. To explore the variation in growth performance and orphological features of local and Thai climbing perch a study was undertaken. The highest gain in length, weight and SGR were found in Thai koi 12.23±0.38 cm, 55.83±0.53 g and 7.92±0.11 %/day respectively. Fourteen morphometric characters were studied where eleven (TL, SL, HL, HBD, LBD, DFL, PECFL, PELFL, AFL, UJL and LJL) showed significant difference (p<0.01) in Thai koi from the local ones. Of the meristic characters no. of dorsal fin rays (hard), anal fin rays (hard), caudal fin rays and scale along lateral line (upper and lower) as recorded from the Thai koi were significantly higher (p<0.01) than that of the local koi. The no. of dorsal fin rays (soft) in Thai koi were also significantly higher (p<0.05) from that of local koi. The number of vertebra were also variable in local and Thai climbing perch (25 in case of local koi and 26 in case of Thai koi). Hence, the results obtained form the present study satisfy the characteristics of A. testudineus which reveals that both the local and Thai koi belongs to the same species. Growth performance of Thai koi was better compared to local koi reared in same conditions.

Hexabromocyclododecane (HBCD) is widely used as a brominated flame retardant, and has been detected in the aquatic environment, wild animals, and humans. However, details of the environmental health risk of HBCD are not well known. In this study, zebrafish embryos were used to assess the developmental toxicity of the chemical. Four-hour post-fertilization (hpf) zebrafish embryos were exposed to various concentrations of HBCD (0, 0.05, 0.1, 0.5, and 1.0 mg L-1) until 96 h. Exposure to 0.1, 0.5, and 1.0 mg L-1 HBCD significantly increased the malformation rate and reduced survival in the 0.5 and 1.0 mg L-1 HBCD exposure groups. Acridine orange (AO) staining showed that HBCD exposure resulted in cell apoptosis. Reactive oxygen species (ROS) was significantly induced at exposures of 0.1, 0.5, and 1.0 mg L-1 HBCD. To test the apoptotic pathway, several genes related to cell apoptosis, such as p53, Puma, Apaf-1, caspase-9, and caspase-3, were examined using real-time PCR. The expression patterns of these genes were up-regulated to some extent. Two anti-apoptotic genes, Mdm2 (antagonist of p53) and Bcl-2 (inhibitor of Bax), were down-regulated, and the activity of capspase-9 and caspase-3 was significantly increased. The overall results demonstrate that waterborne HBCD is able to produce oxidative stress and induce apoptosis through the involvement of caspases in zebrafish embryos. The results also indicate that zebrafish embryos can serve as a reliable model for the developmental toxicity of HBCD. (C) 2009 Elsevier B.V. All rights reserved.

Perfluorooctane sulfonate (PFOS) is widely distributed and persistent in the environment and wildlife. The main aim of this study was to investigate the impact of long-term exposure to low concentrations of PFOS in zebrafish. Zebrafish fry (F-0, 14d post-fertilization, dpf) were exposed via the water for 70d to 0 (control), 10, 50 and 250 mu g L-1 PFOS, followed by a further 30d to assess recovery in clean water. The effects on survival and growth parameters and liver histopathology were assessed. Although growth suppression (weight and length) was observed in fish treated with high concentrations PFOS during the exposure period, no mortality was observed throughout the 70d experiment. Embryos and larvae (F-1) derived from maternal exposure suffered malformation and mortality. Exposure to 50 and 250 mu g L-1 PFOS could inhibit the growth of the gonads (GSI) in the female zebrafish. Histopathological alterations, primary with lipid droplets accumulation, were most prominently seen in the liver of males and the changes were not reversible, even after the fish were allowed to recover for 30d in clean water. The triiodothyronine (T-3)) levels were not significantly changed in any of the exposure groups. Hepatic vitellogenin (VTG) gene expression was significantly up-regulated in both male and female zebrafish, but the sex ratio was not altered. The overall results suggested that lower concentrations of PFOS in maternal exposure could result in offspring deformation and mortality. (c) 2008 Elsevier Ltd. All rights reserved.

Perfluorooetanesulfonate (PFOS) is a persistent organic pollutant, the potential toxicity of which is causing great concern. In the present study, we employed zebrafish embryos to investigate the developmental toxicity of this compound. Four-hour post-fertilization (hpf) zebrafish embryos were exposed to 0.1, 0.5, 1, 3 and 5 mg/L PFOS. Hatching was delayed and hatching rates as well as larval survivorship, were significantly reduced after the embryos were exposed to 1, 3 and 5 mg/L PFOS until 132 hpf. The fry displayed gross developmental malformations, including epiboly deformities, hypopigmentation, yolk sac edema, tail and heart malformations and spinal curvature upon exposure to PFOS concentrations of I mg/L or greater. Growth (body length) was significantly reduced in the 3 and 5 mg/L PFOS-treated groups. To test whether developmental malformation was mediated via apoptosis, flow cytometry analysis of DNA content, acridine orange staining and TUNEL assay was used. These techniques indicated that more apoptotic cells were present in the PFOS-treated embryos than in the control embryos. Certain genes related to cell apoptosis, p53 and Bax, were both significantly up-regulated upon exposure to all the concentrations tested. In addition, we investigated the effects of PFOS on marker genes related to early thyroid development (hhex and pax8) and genes regulating the balance of androgens and estrogens (cyp19a and cyp19b). For thyroid development, the expression of hhex was significantly up-regulated at all concentrations tested, whereas pax8 expression was significantly up-regulated only upon exposure to lower concentrations of PFOS (0.1, 0.5, 1 mg/L). The expression of cyp19a and of cyp19b was significantly down-regulated at all exposure concentrations. The overall results indicated that zebrafish embryos constitute a reliable model for testing the developmental toxicity of PFOS, and the gene expression patterns in the embryos were able to reveal some potential mechanisms of developmental toxicity. A laboratory toxic experiment was conducted to examine dose-dependent effects of extracted microcystins (MCs) on embryonic development, larval growth and histopathological changes of southern catfish (Silurus meridionalis). Fertilized eggs were incubated in solutions with four concentrations of MCs (0, 1, 10, 100 mu g MC-LReq l(-1)). Higher MCs retarded egg development (2-10 h delays) and larval growth, reduced hatching rate (up to 45%), and caused high malformation rate (up to 15%) and hepatocytes damage (characterized by disorganization of cell structure and a loss of adherence between hepatocytes, cellular degeneration with vacuolar hepatocytes and marginal nuclei, even hepatocellular necrosis). A 10 mu g MC-LReql(-1) is close to a high concentration in natural cyanobacterial blooms, suggesting a possible existence of such toxic effects in eutrophic waters. (c) 2007 Elsevier Ltd. Mature female and male zebrafish were separated and exposed to nonylphenol (NP) at 0.1, 1, 10, 50, 100 and 500 mu g/L, respectively, for 3 weeks. Gonadosomatic index (GSI) in both sexes and vitellogenin (VTG) induction in males was measured as the bioindicators for the impairment to the parents. The results indicated that 50 mu g/L of NP was the non-observed effect concentration (NOEC) for GSI and VTG induction. Afterwards, the 50 mu g/L NP exposed females and males, and the control females and males were cross-wise pair-bred in the control water for one week to examine the reproductive effects. The embryonic cathepsin D (CAT D) activity, eggshell thickness, fecundity, hatching rate and malformation (vertebral column flexure) rate of offspring were determined in the four pair-bred groups. While endpoints remained unchanged in the groups with exposed males, prenatal exposure of females to 50 mu g/L of NP resulted in the impairment of reproduction in groups with exposed females including inhibition of CAT D activity (P < 0.05), decrease of eggshell thickness (by 23.6%) and elevation of malformation rate (P < 0.001). These results suggested NP could induce reproductive damage to zebrafish at NOEC for parents. The results also imply that alterations of CAT D activity and eggshell thickness may be more sensitive biomarkers to indicate the reproductive effects caused by endocrine disrupting chemicals. The effect of C-12(6+) heavy ions bombardment on mutagenesis in Salvia splendens Ker-Gawl. was studied. Dose-response studies indicated that there was a peak of malformation frequency of S. splendens at 200 Gy. Abnormal leaf mutants of the bileaf, trileaf and tetraleaf conglutination were selected. Meanwhile, a bicolor flower chimera with dark red and fresh red flower was isolated in M1 generation of S. splendens. Random amplified polymorphic DNA (RAPD) analysis demonstrated that DNA variations existed among the wild-type, fresh and dark red flower shoots of the chimera. The dark red flower shoots of the chimera were conserved and cultivated at a large-scale through micropropagation. MS supplemented with 2.0 mg/L BA and 0.3 mg/L NAA was the optimal medium in which the maximum proliferation ratio (5.2-fold) and rooting rate (88%) were achieved after 6 weeks. Our findings provide an important method to improve the ornamental quality of S. splendens.
