Uma bactéria identificada como Bacillus licheniformis P40 isolada de intestino de peixe (Leporinus sp.) da bacia amazônica foi estudada quanto à sua capacidade de produzir antimicrobianos. O sobrenadante da cultura obtido em caldo de cérebro e coração (BHI) foi caracterizado, sendo ativo contra importantes bactérias patogênicas e deteriorantes como L. monocytogenes, B. cereus, E. carotovora e isolados clínicos de Streptococcus. Este foi parcialmente purificado através de precipitação com sulfato de amônio e cromatografia de gel filtração e de troca iônica. Foram assim isoladas duas substâncias com atividade antimicrobiana, sendo uma delas de natureza protéica. Esta foi estável a altas temperaturas (100o C), numa ampla faixa de pH e mostrou propriedades de biosurfactante. O sobrenadante parcialmente purificado foi utilizado para o combate a um importante fitopatógeno: Erwinia carotovora. Uma dose de 6400 UA/mL foi bactericida para uma concentração de 107 UFC/mL em 20 minutos in vitro. A substância foi capaz de evitar a formação da podridão mole em batatas (in vivo). Foi estudada a produção da atividade antimicrobiana em resíduos e sub-produtos da indústria de alimentos, sendo escolhido o soro de queijo para otimizar a produção através de um experimento fatorial 23 variando as condições de temperatura, pH e concentração de soro de queijo em pó. As melhores condições foram para temperaturas entre 26 e 37o C e pH entre 6,5 e 7,5 para uma concentração de soro de 7%, sendo que aumentos na concentração levaram a aumentos na produção.

Pouco se conhece sobre a estrutura dos ácidos naftênicos presentes no petróleo brasileiro e derivados. Existem alguns trabalhos realizados com a fração ácida, dos óleos crus, direcionados à aplicação geológica, visto que os ácidos hopanóicos, constituintes dessa fração, podem ser indicadores de migração e maturação. Entretanto, além do interesse geológico, a fração ácida, também está relacionada à corrosão nos equipamentos de refino desses óleos, motivando pesquisas para separar e identificar os ácidos responsáveis pelo problema corrosivo nas refinarias. Os ácidos naftênicos (AN) representam um teor muito pequeno, tanto nos óleos crus, quanto nos derivados, dos quais são constituintes. A concentração bastante baixa destes ácidos, em matrizes com composição ampla e variada, dificulta sua extração. Portanto, as etapas de separação, concentração e purificação da amostra, além da análise, demandam tempo e trabalho extensivos. Os objetivos deste trabalho de pesquisa podem ser resumidos em, otimizar o método de extração e análise, para os ácidos presentes no gasóleo pesado (GOP) e verificar seu efeito corrosivo. Neste trabalho, utilizou-se extração líquido-líquido (ELL), cromatografia líquida preparativa com sílica, cromatografia de troca iônica com as resinas A-21e A-27 e extração em fase sólida (SPE) com dessorção em banho de ultra-som (US), aplicada no extrato da ELL, para extrair a fração ácida do (GOP) do petróleo Marlim (Campos, Rio de Janeiro, Brasil). A análise foi realizada, através de cromatografia em fase gasosa com detector de espectrometria de massas e ionização por impacto de elétrons, após uma etapa de derivatização com metanol/BF3 ou com N-metil-N-(terc-butildimetilsilil)trifluoracetamida. Os resultados apresentados indicam a presença de ácidos carboxílicos, pertencentes a famílias de compostos alicíclicos e cíclicos, com até quatro anéis na molécula. Também foi verificado o efeito corrosivo da amostra de GOP sobre amostras de aço ASTM 1010, associado à presença de ácidos naftênicos nesta amostra.

A proposição do presente trabalho foi avaliar o potencial das enzimas das NSLAB do queijo Serrano quanto às atividades aminopeptidásicas. Seis bateladas de queijo Serrano foram elaboradas (3 no verão e 3 no inverno) por três diferentes produtores (A, B e C) de Caxias do Sul (RS), utilizando leite cru, sem adição de culturas iniciadoras. A atividade das aminopeptidases foi avaliada nas FPS dos queijos e nos extratos aquosos dos microrganismos através da hidrólise dos substratos sintéticos Gly- NA, Arg- NA, Tyr- NA, Leu- NA e Asp- NA, Glu- NA, Lys- NA, e Pro- NA. Nos queijos do produtor A, que apresentaram as maiores atividades (19 a 74 U/g de queijo), foram analisados os amino ácidos livres (AAL) e os resultados revelaram que eles aumentam continuamente, estão presentes em quantidades elevadas (4051 mg/100g de ES do queijo aos 60 d), e os AAL com maiores concentrações nos queijos foram Glu, Arg, Lys, Pro e Leu (juntos representam 58% a 61% do total de AAL). Extratos aquosos das NSLAB dos queijos de verão foram parcialmente purificados por cromatografia de troca iônica de fluxo rápido, utilizando Q-sepharose e incubados com os substratos Glu- NA, Arg- NA, Lys- NA, Leu- NA e Pro- NA, indicando atividade para todos os substratos e que a atividade nas frações mudam nas NSLAB para queijos com diferentes tempos de maturação. Uma comparação das atividades aminopeptidases recuperadas após o zimograma do gel de eletroforese-PAGE das FPS dos queijos e dos microrganismos, sugerem que as PepN, PepC e PepL ativas nas FPS têm suas origens na lise das NSLAB presentes nos mesmos.

Metarhizium anisopliae é um fungo cosmopolita com capacidade de infectar uma grande variedade de hospedeiros, estando entre eles o carrapato Boophilus microplus. A penetração de M. anisopliae em seus hospedeiros ocorre de forma ativa onde a cutícula constitui a principal barreira. A penetração é um processo multifatorial, porém, o emprego de pressão mecânica e a secreção de enzimas hidrolíticas parecem ser fundamentais para o seu sucesso. M. anisopliae, quando cultivado em meios com fontes de carbono que mimetizam a cutícula de seus hospedeiros, secreta enzimas como proteases, quitinases e lipases. Atualmente, o emprego de técnicas que identificam genes diferencialmente expressos (RDA) mostrou o possível envolvimento de outras enzimas, como as β-glicanases, durante o processo de penetração. A descoberta da ocorrência de modificações morfológicas como espessamento e perda da definição da parede celular nas extremidades das hifas que penetram na cutícula do carrapato sustentam ainda mais o possível envolvimento de enzimas que degradam as β-glicanas nas etapas iniciais da infecção. Neste trabalho, foi investigada a produção de β-1,3- glicanases pela linhagem E6 de M. anisopliae como também, buscou-se purificar as enzimas produzidas. A síntese e secreção de β-1,3-glicanases foram verificadas em meio contendo diferentes fontes de carbono sendo a secreção diferenciada dependendo da condição testada. A utilização de glicose em determinadas concentrações pareceu inibir a secreção enzimática. Duas das condições testadas, N-acetilglicosamina (NAG) 0,5% e parede celular de Rizoctonia solani 0,5%, foram utilizadas para a produção enzimática em larga escala. O sobrenadante dos cultivos em fermentador foi submetido ao processo de purificação que constou de três etapas: concentração por ultrafiltração com membrana de celulose regenerada, aplicação em coluna de troca iônica QSepharose Fast Flow e aplicação em coluna de filtração em gel Superdex 75. O emprego deste protocolo permitiu a purificação parcial de uma β-1,3-glicanase com aproximadamente 95kDa, secretada durante a fermentação em presença de parede celular de Rizoctonia solani, e de outra, com aparentemente a mesma massa molecular secretada em fermentação utilizando NAG 0,5% como fonte de carbono. Durante este trabalho, também foi confirmada a presença de pelo menos um gene que codifica uma exo-β-1,3-glicanase no genoma da linhagem E6 de M. anisopliae. Por fim, o estudo das β-1,3 glicanases em M. anisopliae é justificado pela importância destas enzimas em variados aspectos do desenvolvimento do fungo bem como, pelo seu possível envolvimento na infecção de hospedeiros.

Apresenta a descrição da metodologia para determinação analítica de alumínio trocável do solo. A partir da extração de alumínio do solo através de mecanismos de troca-iônica com uma solução de KCl concentrado, seguido de titulação ácido-base com NaOH, determina-se a quantidade de alumínio trocável do solo. Todas as etapas do procedimento são reproduzidas detalhadamente.

-D-glucosidase (EC 3.2.1.21) is one of the most interesting glycosidases, especially for hydrolysis cellobiose releasing glucose, is last step degradation of cellulose. This function makes the -D-glucosidase is of great interest as a versatile industrial biocatalyst, being critical to various bio-treatment / biorefinery processes, such as bioethanol production. Hen in the report, a -D-glucosidase was extracts from protein extracted of the invertebrate marine Artemia franciscana was purified and characterized with a combination of precipitation with ammonium sulfate (0 - 30%, 30 to 50%, 50 to 80%), the fraction saturated in the range of 30 to 50% (called F-II) was applied in a molecular exclusion chromatography, in Sephacryl S-200, the fractions corresponding to the first peak of activity of -D-glucosidase were gathered and applied in a chromatography of ion exchange in Mono Q; the third peak this protein obtained chromatography, which coincides with the peak of activity of -D-glucosidase was held and applied in a gel filtration chromatography Superose 12 where the first peak protein, which has activity of -D-glucosidase was rechromatography on Superose 12. This enzyme is probably multimerica, consisting of three subunit molecular mass of 52.7 kDa (determined by SDS-PAGE) with native molecular mass of 157 kDa (determined by gel filtration chromatography on Superose 12 under the system FPLC). The enzyme was purified 44.09 times with a recovery of 1.01%. Using up p-nitrophenyl-β-D-glucopiranoside as substrate obtained a Km apparent of 0.229 mM and a Vmax of 1.109 mM.60min-1.mL-1mM. The optimum pH and optimum temperature of catalysis of the synthetic substrate were 5.0 and 45 °C, respectively. The activity of the -D-glucosidase was strongly, inhibited by silver nitrate and N- etylmaleimide, this inhibition indicates the involvement of radical sulfidrila the hydrolysis of synthetic substrate. The -D-glucosidase of Artemia franciscana presented degradativa action on celobiose, lactose and on the synthetic substrate -nitrophenyl-β-D-glucopiranoside indicating potential use of this enzyme in the industry mainly for the production of bioethanol (production of alcohol from the participating cellulose), and production hydrolysate milk (devoid of milk lactose)

Since the first description of sulfated polysaccharides from seaweeds, the biological activities of these compounds have been evaluated under different aspects and experimental procedures. Among the broad biological activities presented by seaweed polysaccharides, anticoagulant action appears as a promising function. In this present study we have obtained sulfated polysaccharides from the green seaweed Codium isthmocladium by proteolytic digestion, followed by separation into five fractions (0.3, 0.5, 0.7, 0.9 and 1.2) by sequential acetone precipitation. The chemical analyses have demonstrated that all fractions are composed mainly by sulfated polysaccharides. The anticoagulant activity of these fractions was determined by activated partial thromboplastin time (aPTT) and prothrombin time test (PT) using citrate normal human plasma. None fraction has shown anticoagulant activity by PT test. Furthermore, all of them have shown anticoagulant activity by aPTT test. These results indicated that the molecular targets of these sulfated polysaccharides are mainly in the intrinsic via of the coagulation cascade. Agarose gel electrophoresis in 1,3-diaminopropane acetate buffer, pH 9.0, stained with 0.1% toluidine blue showed the presence of two or three bands in several fractions while the fraction 0.9 showed a single spot. By anion exchange chromatography, the acid polysaccharides from the 0.9 acetone fraction were separated into two new fractions eluted respectively with 2.0 and 3.0 M NaCl. These compounds showed a molecular weight of 6.4 and 7.4 kDa respectively. Chemical analyses and infrared spectroscopy showed that Gal 1 and Gal 2 are sulfated homogalactans and differ one from the other in degree and localization of sulfate groups. aPPT test demonstrated that fractions 2,0 and 3,0M (Gal1 and Gal 2, respectively) have anticoagulant activity. This is the first time that anticoagulant sulfated homogalatans have been isolated from green algae. To prolong the coagulation time to double the baseline value in the aPTT, the required amount of sulfated galactan 1 (6,3mg) was similar to low molecular heparin Clexane®, whereas only 0,7mg of sulfated galactan 2 was needed to obtain the same effect. Sulfated galactan 2 in high doses (250mg) induces platelet aggregation. These results suggest that these galactans from C. isthmocladum have a potential application as an anticoagulant drug

One Kunitz-type trypsin inhibitors (PmTI) was purified from Piptadenia moniliformis seeds, a tree of the sub-family Mimosoideae, by TCA precipitation, affinity chromatography on immobilized trypsin-Sepharose, DEAE cellulose (ion exchange) and Superose 12 (molecular exclusion) column FPLC/AKTA. The inhibitor has Mr of 25 kDa by SDS-PAGE and chromatography molecular exclusion. The N-terminal sequence of this inhibitor showed high homology with other family Kunitz inhibitors. This also stable variations in temperature and pH and showed a small decrease in its activity when incubated with DDT in the concentration of 100mM for 120 minutes. The inhibition of trypsin by PmTI was competitive, with Ki of 1.57 x10-11 M. The activity of trypsin was effectively inhibited by percentage of inhibition of 100%, among enzymes tested, was not detected inhibition for the bromelain, was weak inhibitor of pancreatic elastase (3.17% of inhibition) and inhibited by 76.42% elastase of neutrophils, and inhibited in a moderate, chymotrypsin and papain with percentage of inhibition of 42.96% and 23.10% respectively. In vitro assays against digestive proteinases from Lepidoptera, Diptera and Coleoptera pests were carried out. Several degrees of inhibition were found. For Anthonomus grandis and Ceratitis capitata the inhibition was 89.93% and 70.52%, respectively, and the enzymes of Zabrotes subfasciatus and Callosobruchus maculatus were inhibited by 5.96% and 9.41%, respectively, and the enzymes of Plodia. interpunctella and Castnia licus were inhibited by 59.94% and 23.67, respectively. In vivo assays, was observed reduction in the development of larvae in 4rd instar of C. capitata, when PmTI was added to the artificial diet, getting WD50 and LD50 of 0.30% and 0.33%, respectively. Heparan sulfate (HS) and Heparin (Hep) glycosaminoglycans (GAGs) are heterogeneous and highly charged polysaccharides. HS is structurally related to Hep but is much less substituted with sulfo groups than heparin and has a more varied structure (or sequence). Because of structural similiarities between these two polymers, they have been described together as heparinoids . Both chains bind a variety of proteins and mediate various physiologically important processes including, blood coagulation, cell adhesion and growth factor regulation. Heparinoids with structural characteristics similar to these described from HS and/or Hep from mammalian tissues have been isolated from different species of invertebrates, although only a few heparinoids from unusual sources have been characterized. The present study describes the presence of unusual heparinoids population from Artemia franciscana, isolated after proteolysis and fractionation by ion exchange resin and named, F-3.0M. The study model in vivo were hemostasis (rat tail scarification) and inflamatoty activity. The tests in vitro were used for coagulations assays (PT and APTT). The analyse of the heparinoids eluted with 3,0M NaCl showed electrophoretic migration in different buffer systems a single band with a behaviour intermediate between those of mammalian HEP and HS. The main products obtained from Artemia heparinoids after enzymatic degradation with heparitinases I and II from F. heparinum were N-sulphated disaccharides (∆U-GlcNS,6S/ ∆U,2S-GlcNS and ∆U-GlcNS) and N-acetylated disaccharides (∆U, GlcNAc). This heparinoid had a lower hemorrhagic effect (400μg/ml) when compared to unfractiionated heparins(25μg/ml).The results also suggest a negligible APTT activity of this heparinoid (62.2s). No action was observed on PT indicating that F-3.0M haven t action on the extrinsic pathway. The results showed that the fraction F- 3.0M have inhibitory effect on migration of leukocytes, 64.5% in the concentration of 10 μg/ml (P<0.001). The search for new heparin and/or heparan sulphates analogs devoid of anticoagulant activity is an atractive alternative and may open up a wide variety of new therapeutic applications

A 140,0 kDa lectin was purified and characterized from the mushroom Clavaria cristata. The purification procedures from the crude extract of the mushroom comprised gel filtration chromatography on Sephacryl s200 and ion exchange on Resource Q column. The purified lectin agglutinated all types of human erythrocytes with preference for trypsinized type O erythrocytes. The haemagglutinating activity is dependent of Ca 2+ ions and was strongly inhibited by the glycoprotein bovine submaxillary mucin (BSM) up to the concentration of 0, 125 mg/mL. The C. cristata lectin (CcL) was stable in the pH range of 2,5-11,5 and termostable up to 80 °C. CcL molecular mass determined by gel filtration on a Superose 6 10 300 column was approximately 140,3 kDa. SDS polyacrilamide gel electrophoresis revealed a single band with a molecular mass of approximately 14,5 kDa, when the lectin was heated at 100 ⁰C in the presence or absence of β-mercaptoethanol. CcL induced activation of murine peritoneal macrophages in vitro resulting in the release of nitric oxide (NO), reaching the maximum production at 24 h. In experimental paw oedema model in mice, CcL showed proinflammatory activity being able to induce oedema formation. Cell viability of HepG2, MDA 435 e 3T3 cell lines was examined after 72 h of incubation with CcL in different concentrations (0,5-50 μg/mL). CcL inhibited HepG2 cells growth with an IC50 value of 50 μg/mL. In the present work, the observed immunomodulatory and antiproliferative effects indicate CcL as a possible immunomodulator compound, interfering in the macrophages immune response, taking possible anti-parasitic, anti-tumoral effects or diagnostic and/or therapeutic

Studies indicate that several components were isolated from medicinal plants, which have antibacterial, antifungal, antitumor and anti-inflammatory properties. Sepsis is characterized by a systemic inflammation which leads to the production of inflammatory mediators exacerbated by excessive activation of inflammatory cells and disseminated intravascular coagulation (DIC), in which the human neutrophil elastase plays an important role in its pathogenesis. Several epidemiological studies suggest that components of plants, especially legumes, can play a beneficial role in reducing the incidence of different cancers. A chymotrypsin inhibitor of Kunitz (Varela, 2010) was purified from seeds of Erythrina velutina (Mulungu) by fractionation with ammonium sulfate, affinity chromatography on Trypsin-Sepharose, Chymotrypsin-Sepharose and ion exchange chromatography on Resource Q 1 ml (GE Healthcare) in system FPLC / AKTA. The inhibitor, called EvCI, had a molecular mass of 17 kDa determined by SDS-PAGE. The purified protein was able to inhibit human neutrophil elastase (HNE), with an IC50 of 3.12 nM. The EvCI was able to inhibit both pathways of HNE release stimulated by PAF and fMLP (75.6% and 65% respectively). The inhibitor also inhibited leukocyte migration in septic mice about 87% and prolonged the time of coagulation and inhibition factor Xa. EvCI showed neither hemolytic activity nor cytotoxicity. EvCI showed a selective antiproliferative effect to HepG2 cell lines with IC50 of 0.5 micrograms per milliliter. These results suggest EvCI as a molecule antagonist of PAF / fMLP and a potential use in fighting inflammation related disorders, disseminated intravascular coagulation (DIC) and cancer

Chitinases are enzymes involved in degradation of chitin and are present in a range of organisms, including those that do not contain chitin, such as bacteria, viruses, plants and animals, and play important physiological and ecological roles. Chitin is hydrolyzed by a chitinolytic system classified as: endo-chitinases, exo-chitinases and N-acetyl-b-D-glucosaminidases. In this study a Litochitinase1 extracted from the cephalotorax of the shrimp Litopenaeus Schmitt was purified 987.32 times using ionexchange chromatography DEAE-Biogel and molecular exclusion Sephacryl S-200. These enzyme presented a molecular mass of about 28.5 kDa. The results, after kinetic assay with the Litochitinase1 using as substrate p-nitrophenyl-N-acetyl-b-Dglucosaminideo, showed apparent Km of 0.51 mM, optimal activity at pH ranging from 5.0 to 6.0, optimum temperature at 55°C and stability when pre-incubated at temperatures of 25, 37, 45, 50 and 55°C. The enzyme showed a range of stability at pH 4.0 to 5.5. HgCl2 inhibited Litochitinase1 while MgCl2 enhances its activity. Antimicrobial tests showed that Litochitinase1 present activity against gram-negative bacterium Escherichia coli in the 800 μg/mL concentration. The larvicidal activity against Aedes aegypti was investigated using crude extracts, F-III (50-80%) and Litochitinase1 at 24 and 48 hours. The results showed larvicidal activity in all these samples with EC50 values of 6.59 mg/mL for crude extract, 5.36 mg/mL for F-III and 0.71 mg/mL for Litochitinase1 at 24 hours and 3.22 and 0.49 mg/mL for the F-III and Litochitinase1 at 48 hours, respectively. Other experiments confirmed the presence of chitin in the midgut of Aedes aegypti larvae, which may be suffering the action of Litochitinase1 killing the larvae, but also the absence of contaminating proteins as serine proteinase inhibitors and lectins in the crude extract, F-III and Litochitinase1, indicating that the death of the larvae is by action of the Litochitinase1. We also observed that the enzymes extracted from intestinal homogenate of the larvae no have activity on Litochitinase1. These results indicate that the enzyme can be used as an alternative to control of infections caused by Escherichia coli and reducing the infestation of the mosquito vector of dengue.

Heparin is a pharmaceutical animal widely used in medicine due to its potent anticoagulant effect. Furthermore, it has the ability to inhibit the proliferation, invasion and adhesion of cancer cells to vascular endothelium. However, its clinical applicability can be compromised by side effects such as bleeding. Thus, the search for natural compounds with low bleeding risk and possible therapeutic applicability has been targeted by several research groups. From this perspective, this study aims to evaluate the hemorrhagic and anticoagulant activities and citotoxic effect for different tumor cell lines (HeLa, B16-F10, HepG2, HS-5,) and fibroblast cells (3T3) of the Heparin-like from the crab Chaceon fenneri (HEP-like). The HEP-like was purified after proteolysis, ion-exchange chromatography, fractionation with acetone and characterized by electrophoresis (agarose gel) and enzymatic degradation. Hep-like showed eletroforetic behavior similar to mammalian heparin, and high trisulfated /Nacetylated disaccharides ratio. In addition, HEP-like presented low in vitro anticoagulant activity using aPTT and a minor hemorrhagic effect when compared to mammalian heparin. Furthermore, the HEP-like showed significant cytotoxic effect (p<0.001) on HeLa, HepG2 and B16-F10 tumor cells with IC50 values of 1000 ug/mL, after incubation for 72 hours. To assess the influence of heparin-like on the cell cycle in HeLa cells, analysis was performed by flow cytometry. The results of this analysis showed that HEP-like influence on the cell cycle increasing S phase and decreasing phase G2. Thus, these properties of HEP-like make these compounds potential therapeutic agents

A bactéria B. thuringiensis caracteriza-se pela produção de proteínas tóxicas a representantes de diversas ordens de insetos, as quais são codificadas por genes cry. Este trabalho foi realizado com objetivo de selecionar isolados de B. thuringiensis, por meio da caracterização morfológica e molecular, identificando as diferentes subclasses dos genes cry3 e cry35 e determinar a patogenicidade contra Sphenophorus levis, uma das mais importantes pragas da cultura da cana-de-açúcar. Foram utilizados 1163 isolados de B. thuringiensis e com a observação em microscópio com contraste de fases foram confirmadas como pertencentes à espécie de B. thuringiensis. O material genético foi purificado pela matriz de troca iônica Instagene Matrix e submetido a PCR com iniciadores gerais cry3 e cry35 identificando-se 30 isolados contendo genes com potencial para o controle de coleópteros, os quais juntamente com as linhagens-padrão de B. thuringiensis var. tenebrionis, B. thuringiensis var. morrissone e B. thuringiensis var. tolworthi foram utilizados para a realização do bioensaio. Através de análise discriminante alocaram-se os isolados em quatro grupos quanto à toxicidade de B. thuringiensis. Os grupos ficaram assim definidos: um grupo que promovem até 10% de mortalidade contendo as testemunhas e duas linhagens;um grupo que causou 39% de mortalidade contendo três linhagens padrão e dez isolados; um grupo com 52% de mortalidade contendo treze isolados e um grupo com 70% de mortalidade contendo cinco isolados, os quais devem ser considerados promissores no controle biológico de S. levis.

The crude glycerine is a raw material that can be used in a wide variety of products. Even with all the impurities inherent in the process of being obtained, the crude glycerin is already in a marketable product. However, the market is much more favorable to the commercialization of purified glycerine. The glycerin is a byproduct gotten from the process of transesterification of waste oils and fats in the production of biodiesel. More recently, the deployment of the new Federal Law of Brazil, related to the implementation of energy resources, forces, from 2008, the increase of 2% biodiesel in diesel common with prospects for 5% (B5). Therefore, it is indispensable that new routes of purification as well as new markets are developed. The objective of this work was to purify, through ion exchange, the crude glycerin, obtained from the reaction of transesterification of cottonseed oil. The cottonseed oil was characterized as the fatty acid composition and physical-chemical properties. The process of ion exchange was conducted in batch. In this process were used strong cation, low anion resins and a mixed resin used to de-ionize water. The purified glycerin was characterized as the content of metals. Tests were performed with activated charcoal adsorption, and for this, it was made tests of time contact with coal as well as quantity of coal used. The time of activation, the amount of the activation solution, the contact time of the glycerol solution in resins, the amount and type of resin applied were evaluated. Considering the analysis made with activated charcoal, when the glycerin solution was treated using the resins individually it was observed that in the conditions for treatment with 10 g of resin, 5 hours of contact with each resin and 50 mL of glycerin solution, its conductivity decreased to a cationic resin, increased to the anionic resin and had a variable value with respect to resin mixed. In the treatment in series, there was a constant decrease in the conductivity of the solution of glycerin. Considering two types of treatment, in series and individually, the content of glycerol in glycerin pre-purified solution with the different resins varied from 12,46 to 29.51% (diluted solution). In analysis performed without the use of activated charcoal, the behavior of the conductivity of the solution of glycerin were similar to results for treatment with activated charcoal, both in series as individually. The solution of glycerin pre-purified had a glycerol content varying from 8.3 to 25.7% (diluted solution). In relation to pH, it had a behavior in accordance with the expected: acid for the glycerin solution treated with cationic resin, basic when the glycerin solution was treated with the anionic resin and neutral when treated with the mixed resin, independent of the kind of procedure used (with or without coal, resins individually or in series). In relation to the color of the glycerin pre-purified solution, the resin that showed the best result was the anionic (colorless), however this does not mean that the solution is more in pure glycerol. The chromatographic analysis of the solutions obtained after the passage through the resins indicated that the treatment was effective by the presence of only one component (glycerol), not considering the solvent of the analysis
