One of the main components of snake venoms are the Asp49-phospholipases A(2), also known as svPLA(2)s. The study of these toxins is a matter of great scientific interest due to their wide variety of biological effects. In this work we present strong evidences found in literature and other aspects which strengthen the importance of quaternary assembly for understanding the activities and molecular evolution of svPLA(2)s.

The 1.7 angstrom resolution crystal structure of recombinant family G/11 beta-1,4-xylanase (rXynA) from Bacillus subtilis 1A1 shows a jellyroll fold in which two curved P-sheets form the active-site and substrate-binding cleft. The onset of thermal denaturation of rXynA occurs at 328 K, in excellent agreement with the optimum catalytic temperature. Molecular dynamics simulations at temperatures of 298-328 K demonstrate that below the optimum temperature the thumb loop and palm domain adopt a closed conformation. However, at 328 K these two domains separate facilitating substrate access to the active-site pocket, thereby accounting for the optimum catalytic temperature of the rXynA. Neste experimento, avaliou-se o efeito de sexo (macho e fêmea) e de quatro distintos pesos de abate (28, 32, 36, e 40 kg) sobre a morfometria da carcaça, os pesos dos cortes, a composição tecidual e os componentes não constituintes da carcaça, em cordeiros mestiços Ile de France x Corriedale, terminados em confinamento. Utilizaram-se 40 animais (20 machos e 20 fêmeas), desmamados aos 60 dias e alimentados, à vontade, com uma ração com 16,46% de PB e 67,63% de NDT. Foram divididos em quatro grupos de machos (G1M, G2M, G3M e G4M), sacrificados, respectivamente, com os pesos supracitados, e, da mesma forma, quatro grupos de fêmeas (G1F, G2F, G3F e G4F). Determinaram-se as medidas da carcaça e, de forma subjetiva, a condição corporal, o grau de conformação e o grau de gordura de cobertura. Calcularam-se os índices de compacidade da carcaça e da perna. Foram registrados os pesos e as porcentagens de sete cortes da meia carcaça, bem como as porcentagens de osso, músculo e tecido adiposo, avaliadas por meio de dissecção do lombo. A espessura da gordura de cobertura foi determinada na região lombar. O estudo morfométrico confirmou maior alongamento ósseo dos machos. de maneira geral, as carcaças apresentaram-se com bom acabamento, segundo os índices de compacidade verificados. Os cortes das carcaças das fêmeas foram mais pesados que os dos machos (exceto para o peso de 28 kg ao abate), principalmente em função dos maiores teores de gordura. Destaca-se a representabilidade dos pesos da pele e do conteúdo gastrintestinal na determinação do rendimento da carcaça. Pelos resultados obtidos, recomenda-se 28 kg como peso referência para sacrifício.

The effect of anions on the stability of different functional conformations of Hb is examined through the determination of the dependence of O-2 affinity on water activity (a(w)). The control of a(w) is effected by varying the sucrose osmolal concentration in the bathing solution according to the osmotic stress method. Thus, the hydration change following Hb oxygenation is determined as a function of Cl- and of DPG concentration. We find that only similar to 25 additional water molecules bind to human Hb during the deoxy-to-oxy conformation transition in the absence of anions, in contrast with similar to 72 that bind in the presence of more than 50 mM Cl- or more than 15 mu M DPG. We demonstrate that the increase in the hydration change linked with oxygenation is coupled with anion binding to the deoxy-Hb. Hence, we propose that the deoxy-Hb coexists in two allosteric conformations which depend on whether anion is bound or not: the tense T-state, with low oxygen affinity and anion bound, or a new allosteric P-state, with intermediate oxygen affinity and free of bound anions. The intrinsic oxygen affinity of this unforeseen P-state and the differential binding of Cl-, DPG, and H2O between states P and T and P and R are characteristics which are consistent with those expected for a putative intermediate allosteric state of Hb. These findings represent a new opportunity to explore the structure-function relationships of hemoglobin regulation.

The objective of this work was to estimate the correlations among measurements taken in vivo with ultrasound equipment with some carcass traits measured after slaughter. Twenty eight Mediterranean bulls, with average shrunk body weight of 330 kg and 14 months of age, were fed by 120 days with high concentrate diets. The shrunk body weight, the ribeye area (REAU), the back fat thickness (FTU) over the Longissimus dorsi muscle between 12(a) and 13(a) ribs and rump fat (EGP8U), were measured at 28 days intervals. Real-time ultrasound equipment Piemedical Scanner 200 VET, with 18 cm linear array transducer was utilized. After the slaughter, the hot carcass weight (PCQ) and the kidney, pelvic and inguinal fat (GRPI) were weighted and the dressing percentage (DP) calculated. After 24 hours of cooling the ribeye area (REAC), backfat thickness (FTC) and rump fat (EGP8C) were measured. Both the REAC, FTC and EGP8C were underestimated by ultrasound measurements. The Pearson correlation coefficients for ribeye area, backfat thickness and rump fat measured in the carcass and with ultrasound, were 0.96, 0.99 and 0.91, respectively. The coefficient between DP and REAU was 0.47; 0.45 between DP and REAC, 0.56 between DP and FTU and 0.58 between DP and FTC. DP presented a 0.59 correlation coefficient with EGP8U. The Spearman correlation was estimated between REAU and REAC, FTU and FTC, EGP8U and EGP8C, and the values were 0.96, 0.99 and 0.91,respectively. The ultrasound measures could be used to estimate carcass traits in buffaloes with good accuracy.

Actiaomycin-D (actD) binds to natural DNA at two different classes of binding sites, weak and strong. The affinity for these sites is highly dependent on DNA se(sequence and solution conditions, and the interaction appears to be purely entropic driven Although the entropic character of this reaction has been attributed to the release of water molecules upon drug to DNA complex formation, the mechanism by which hydration regulates actD binding and discrimination between different classes of binding sites on natural DNA is still unknown. In this work, we investigate the role of hydration on this reaction using the osmotic stress method. We skew that the decrease of solution water activity, due to the addition of sucrose, glycerol ethylene glycol, and betaine, favors drug binding to the strong binding sites on DNA by increasing both the apparent binding affinity Delta G, and the number of DNA base pairs apparently occupied by the bound drug n(bp/actD). These binding parameters vary linearly with the logarithm of the molar fraction of water in solution log(X-w), which indicates the contribution of water binding to the energetic of the reaction. It is demonstrated that the hydration change measured upon binding increases proportionally to the apparent size of the binding site n(bp/uctD). This indicates that n(bp/actD) measured from the Scatchard plod is a measure of the size of the DNA molecule changing conformation due to ligand binding. We also find that the contribution of DNA deformation, gauged by n(bp/act) to the total free energy of binding Delta G, is given by Delta G = Delta G(local) + n(bp/actD) x delta G(DNA), where Delta G(local), = -8020 +/- 51 cal/mol of actD bound and delta G(DNa) = -24.1 +/- 1.7cal/mol of base pair at 25 degrees C. We interpret Delta G(local), as the energetic contribution due to the direct interactions of actD with the actual tetranucleotide binding site, and it n(bp/actB) X delta G(DNA) as that due to change inconformation, induced by binding, of it n(bp/actD) DNA base pairs flanking the local site. This interpretation is supported by the agreement found between the value of delta G(DNA) and the torsional free energy change measured independently. We conclude suggesting an allosteric model for ligand binding to DNA, such that the increase in binding affinity is achieved by increasing the relaxation of the unfavorable free energy of binding storage at the local site through a larger number of DNA base pairs. The new aspect on this model is that the size of the complex is not fixed but determined by solutions conditions, such as water activity, which modulate the energetic barrier to change helix conformation. These results may suggest that long-range allosteric transitions of duplex DNA are involved in the inhibition of RNA synthesis by actD, and more generally, in the regulation of transcription. Based on in vitro experiments, Bos indicus embryos were more resistant to heat stress (HS) than Bos taurus embryos. To increase knowledge regarding differences between Bos indicus and Bos taurus in resistance to HS, the primary objective of this study was to determine if tolerance to HS is due to the breed, origin of the oocyte, sperm, or both. Additionally, the influence of the interval between ovary acquisition (in the abattoir) and oocyte aspiration in the laboratory, on early embryo development was ascertained. Oocytes were collected from Nelore and Holstein cows in an abattoir; 4.0 or 6.5 h later, oocytes were aspired in the laboratory, and then matured and fertilized using semen from Nelore (N), Gir (GIR), or Holstein (H) bulls. Ninety-six h post insemination (hpi), embryos with >= 16 cells were divided in two groups: control and HS. In the control group, embryos were cultured at 39 degrees C, whereas in the HS group, embryos were subjected to 41 degrees C for 12 h, and then returned to 39 degrees C. Rates of cleavage, and formation of morula and blastocysts were higher (P < 0.05) for oocytes aspirated at 4.0 versus 6.5 h after ovaries were acquired. Heat stress decreased rates of blastocyst formation for all breeds (N X N; H x H; and H X GIR) and in both time intervals (4.0 and 6.5 h). However, N X N had higher cleavage rate (P < 0.05) in both time intervals when compared with H X H and H X GIR. In addition, Nelore oocytes fertilized with Nelore semen (N X N) had higher blastocyst yields (P < 0.05) in the control and HS group, when compared with the other two breeds (H X H and H X GIR). We concluded that the breed of origin of the oocyte was more important than that of the sperm for development of thermotolerance, because bull breed did not influence embryo development after HS, and in vitro early embryonic development was impaired by increasing (from 4 to 6.5 h) the interval between ovary acquisition and oocyte aspiration. The present study aimed to compare testicular histology and the testicular cell population as well as spermatogenic efficiency in goats with different scrotal conformations. Eighteen goats were divided into 3 groups: Group I - goats without bipartition of the scrotum, Group II - animals with bipartition of the scrotum up to 50% of the testicular length, Group III - goats with scrotal bipartition more than 50% of the testicular length. In goats in Groups I, II and III, the values for the volume density of seminiferous epithelium were 68.9 +/- 0.6%, 71.5 +/- 2.8% and 73.4 +/- 4.7% (P < 0.05), the height of the seminiferous epithelium were 60.2 +/- 4.9 mu m, 61.0 +/- 5.0 mu m and 73.1 +/- 6.6 mu m (P < 0.05), total length of seminiferous tubules found for Groups I, II and III were 2091.9 +/- 27 m, 2172.5 +/- 24.1 m, and 2340.1 14 m (P < 0.05), number of Sertoli and Leydig cells were 1.8 +/- 0.4 x 10(9) and 1.4 +/- 0.1 x 10(9), 2.2 +/- 0.4 and 2.2 +/- 0.7 x 10(9), and 2.5 +/- 0.1 10(9) and 2.3 +/- 0.510(9)(P < 0.05)and daily sperm production observed were 2.1 0.3 x 109, 2.8 0.4 x 109, and 3.1 0.7 x 109 (P < 0.05). In conclusion, goats with greater scrotal bipartition have a greater capacity to produce reproductive cells that is reflected in a greater reproductive potential. (C) 2011 Elsevier B.V.
