Solvent influences on the crystallization of polymorph and hydrate forms of the nootropic drug piracetam (2-oxo-pyrrolidineacetamide) were investigated from water, methanol, 2-propanol, isobutanol, and nitromethane. Crystal growth profiles of piracetam polymorphs were constructed using time-resolved diffraction snapshots collected for each solvent system. Measurements were performed by in situ energy dispersive X-ray diffraction recorded in Station 16.4 at the synchrotron radiation source (SRS) at Daresbury Laboratory, CCLRC UK. Crystallizations from methanol, 2-propanol, isobutanol, and nitromethane progressed in a similar fashion with the initial formation of form I which then converted relatively quickly to form II with form III being generated upon further cooling. However, considerable differences were observed for the polymorphs lifetime and both the rate and temperature of conversion using the different solvents. The thermodynamically unstable form I was kinetically favored in isobutanol and nitromethane where traces of this polymorph were observed below 10 degrees C. In contrast, the transformation of form II and subsequent growth of form III were inhibited in 2-propanol and nitromethane solutions. Aqueous solutions produced hydrate forms of piracetam which are different from the reported monohydrate; this crystallization evolved through successive generation of transient structures which transformed upon exchange of intramolecular water between the liquid and crystalline phases. Rifaximin, a rifamycin derivative, has been reported to induce clinical remission of active Crohn's disease (CD), a chronic inflammatory bowel disorder. In order to understand how rifaximin affects the colonic microbiota and its metabolism, an in vitro human colonic model system was used in this study. We investigated the impact of the administration of 1800 mg/day of rifaximin on the faecal microbiota of four patients affected by colonic active CD [Crohn's disease activity index (CDAI > 200)] using a continuous culture colonic model system. We studied the effect of rifaximin on the human gut microbiota using fluorescence in situ hybridization, quantitative PCR and PCR–denaturing gradient gel electrophoresis. Furthermore, we investigated the effect of the antibiotic on microbial metabolic profiles, using 1H-NMR and solid phase microextraction coupled with gas chromatography/mass spectrometry, and its potential genotoxicity and cytotoxicity, using Comet and growth curve assays. Rifaximin did not affect the overall composition of the gut microbiota, whereas it caused an increase in concentration of Bifidobacterium, Atopobium and Faecalibacterium prausnitzii. A shift in microbial metabolism was observed, as shown by increases in short-chain fatty acids, propanol, decanol, nonanone and aromatic organic compounds, and decreases in ethanol, methanol and glutamate. No genotoxicity or cytotoxicity was attributed to rifaximin, and conversely rifaximin was shown to have a chemopreventive role by protecting against hydrogen peroxide-induced DNA damage. We demonstrated that rifaximin, while not altering the overall structure of the human colonic microbiota, increased bifidobacteria and led to variation of metabolic profiles associated with potential beneficial effects on the host.

Time-resolved kinetic studies of silylene, SiH2, generated by laser flash photolysis of 1-silacyclopent-3-ene and phenylsilane, have been carried out to obtain rate constants for its bimolecular reactions with methanol, ethanol, 1-propanol, 1-butanol and 2-methyl-1-butanol. The reactions were studied in the gas phase over the pressure range 1-100 Torr in SF6 bath gas, at room temperature. In the study with methanol several buffer gases were used. All five reactions showed pressure dependences characteristic of third body assisted association reactions. The rate constant pressure dependences were modelled using RRKM theory, based on Eo values of the association complexes obtained by ab initio calculation (G3 level). Transition state models were adjusted to fit experimental fall-off curves and extrapolated to obtain k∞ values in the range 1.9 to 4.5 × 10-10 cm3 molecule-1 s-1. These numbers, corresponding to the true bimolecular rate constants, indicate efficiencies of between 16 and 67% of the collision rates for these reactions. In the reaction of SiH2 + MeOH there is a small kinetic component to the rate which is second order in MeOH (at low total pressures). This suggests an additional catalysed reaction pathway, which is supported by the ab initio calculations. These calculations have been used to define specific MeOH-for-H2O substitution effects on this catalytic pathway. Where possible our experimental and theoretical results are compared with those of previous studies.

A structurally related series of fluorinated nonionic oxyethylene glycol surfactants of the type C(m)F(2m+1)(CH(2))(n)O[(CH(2)CH(2)O)(p)H], denoted C(m.n)E(p) (where m=4, 6, or 7, m=1 or 2, and p=4 or 6) were synthesized and their surface behavior in aqueous solution was characterized. The ability of these surfactants to form water-in-hydrofluorocarbon (HFC) propellant 134a microemulsions suitable for use in the aerosolized delivery of water-soluble drugs has been investigated. Phase studies showed that, regardless of the composition used, clear one-phase systems could not be prepared if a fluorinated nonionic surfactant was used alone, or in combination with a short or medium fluorocarbon alcohol cosurfactant. Clear one-phase systems could, however, be prepared if a short-chain hydrocarbon alcohol, such as ethanol, n-propanol, or n-pentanol, was used as cosurfactant, with the extent of the one-phase region increasing with decreased chain length of the alcohol cosurfactant. Light-scattering studies on a number of the hydrocarbon-alcoholcontaining systems in the propellant-rich part of the phase diagram showed that only systems prepared with C(4.2)E(6) and propanol contained microemulsion droplets (all other systems investigated were considered to be cosolvent systems).

Phase studies have been performed for quaternary systems composed of egg lecithin, cosurfactant, water and oil. The lecithin used was the commercially available egg lecithin Ovothin 200 (which comprises ≥ 92% phosphatidylcholine). The cosurfactants employed were propanol and butanol, and these were used at lecithin/cosurfactant mixing ratios (Km) of 1:1 and 1.94:1 (weight basis). Six polar oils were investigated, including the alkanoic acids, octanoic and oleic, their corresponding ethyl esters and the medium and long chain triglycerides, Miglyol 812 and soybean oil. All oils, irrespective of the alcohol and the Km used, gave rise to systems that produced a stable isotropic region along the surfactant/oil axis (designated as a reverse microemulsion system). In addition, the systems incorporating propanol at both Km and butanol at a Km of 1.94: 1, generally gave rise to a liquid crystalline region and, in some cases, a second isotropic non-birefingent area (designated as a normal microemulsion system). The phase behaviour observed was largely dependent upon the alcohol and Km used and the size and the polarity of the oil present.

Catalytic properties of copper thin films deposited in small channels and cavities were tested using Raman microscopy and mass spectroscopy (MS) techniques, mainly. The catalytic surface conditions were addressed visually and chemically by optical microscopy and X-ray photoelectron spectroscopy (XPS), respectively. The experimental conditions of present work induced copper oxidation; eventually a number of carbon species and graphite remained on the catalytic surface. Quartz crystal microbalance and mass spectroscopy data support both adsorption and catalysis phenomena. MS showed CO2 formation during n-hexane heating process but not to 2-propanol, probably due to redox reactions. XPS of copper surface present in the cavity after catalysis tests detected Cu2O and a range of possible carbon species. The adsorption and catalytic performance of copper films deposited in cavities and microchannels were quite similar. A simple miniaturized device for microanalysis was proposed. The catalytic hydrodechlorination (HDC) reaction, which is an attractive abatement process for chlorinated organic wastes, was studied over a magnetically recoverable supported Pd(0) catalyst. We investigated the most favorable reaction conditions under which to obtain the highest substrate conversion rates while preserving the catalyst properties and morphology. Sodium hydroxide, triethylamine and buffered solutions were used as proton scavengers in the HDC of chlorobenzene under mild conditions. It was observed that sodium hydroxide caused corrosion of the silica support, triethylamine in 2-propanol preserved the morphology of the catalyst which could be recycled for up to five successive H DC reactions, and aqueous buffer solutions preserved the catalyst morphology and the catalytic activity for up to four successive HDC reactions. The use of buffer solutions to neutralize the HCl formed during the HDC reaction is an interesting, less aggressive, alternative approach to HDC reactions. (C) 2010 Elsevier B.V. Lipid transfer proteins (LTPs) were thus named because they facilitate the transfer of lipids between membranes in vitro. This study was triggered by the characterization of a 9-kDa LTP from Capsicum annuum seeds that we call Ca-LTP(1). Ca-LTP(1) was repurified, and in the last chromatographic purification step, propanol was used as the solvent in place of acetonitrile to maintain the protein`s biological activity. Bidimensional electrophoresis of the 9-kDa band, which corresponds to the purified Ca-LTP(1), showed the presence of three isoforms with isoelectric points (pIs) of 6.0, 8.5 and 9.5. Circular dichroism (CD) analysis suggested a predominance of alpha-helices, as expected for the structure of an LTP family member. LTPs immunorelated to Ca-LTP(1) from C. annuum were also detected by western blotting in exudates released from C. annuum seeds and also in other Capsicum species. The tissue and subcellular localization of Ca-LTP(1) indicated that it was mainly localized within dense vesicles. In addition, isolated Ca-LTP(1) exhibited antifungal activity against Colletotrichum lindemunthianum, and especially against Candida tropicalis, causing several morphological changes to the cells including the formation of pseudohyphae. Ca-LTP(1) also caused the yeast plasma membrane to be permeable to the dye SYTOX green, as verified by fluorescence microscopy. We also found that Ca-LTP(1) is able to inhibit mammalian alpha-amylase activity in vitro.

The proline-rich N-terminal domain of gamma-zein has been reported in relevant process, which include its ability to cross the cell membranes. Evidences indicate that synthetic hexapeptide (PPPVHL), naturally found in N-terminal portion of gamma-zein, can adopt the polyproline II (PPII) conformation in aqueous solution. The secondary structure of gamma-zein in maize protein bodies had been analyzed by solid state Fourier transform infrared and nuclear magnetic resonance spectroscopies. However, it was not possible to measure PPII content in physiological environment since the beta-sheet and PPII signals overlap in both solid state techniques. Here, the secondary structure of gamma-zein has been analyzed by circular dichroism in SDS aqueous solution with and without ditiothreitol (DTT), and in 60% of 2-propanol and water with DTT The results show that gamma-zein has high helical content in all solutions. The PPII conformation was present at about 7% only in water/DTT solution. (c) 2007 Wiley Periodicals, Inc.

Glycine-rich proteins (GRP), serve a variety of biological functions. Acanthoscurrin is an antimicrobial GRP isolated front hemocytes-of the Brazilian spider Acanthoscurria gomesiana. Aiming to contribute to the knowledge of the secondary structure and stepwise solid-phase synthesis of GRPs` glycine-rich domains, we attempted to prepare G(101)GGLGGGRGGGYG(113) GGGGYGGGYG(123)GGy(126)GGGKYK(132)-NH(2), acanthoscurrin C-terminal amidated fragment. Although a theoretical prediction did not indicate high aggregation potential for this peptide, repetitive incomplete aminoacylations were observed after incorporating Tyr(126) to the growing peptide-MBHA resin (Boc chemistry) at 60 degrees C. The problem was not solved by varying the coupling reagents or solvents, adding chaotropic salts to the reaction media or changing the resin/chemistry (Rink amide resin/Fmoc chemistry). Some improvement was mode when CLEAR amide resin (Fmoc chemistry) was 32 used, as it allowed for obtaining fragment (G(113)-K(132) NIR-FT-Raman spectra collected for samples of the growing peptide-MBHA, -Rink amide resin and -CLEAR amide resin revealed the presence of beta-sheet structures. Only the combination of CLEAR-amide resin, 60 degrees C, Fmoc-(Fmoc-Hmb)Gly-OH and LiCl (the last two used alternately) was able to inhibit the phenomenon, as proven by NIR-FT-Raman analysis of the growing peptide-resin, allowing the total synthesis of desired 132 fragment Gly(101)-K(132). In summary, this work describes a new difficult sequence, contributes to understanding stepwise solid-phase synthesis of this type of peptide and shows that, at least while protected and linked to a resin, this GRPs glycine-rich motif presents all early tendency to assume beta-sheet structures. The question raised in the title has been answered by comparing the solvatochromism of two series of polarity probes, the lipophilicities of which were increased either by increasing the length of an alkyl group (R) attached to a fixed pyridine-based structure or through annelation (i.e., by fusing benzene rings onto a central pyridine-based structure). The following novel solvatochromic probes were synthesized: 2,6-dibromo-4-[(E)-2-(1-methylquinolinium-4-yl)ethenyl]-phenolate (MeQMBr(2)) and 2,6-dibromo-4-[(E)-2-(1-methyl-acridinium-4- yl) ethenyl)]phenolate (MeAMBr(2) The solvatochromic behavior of these probes, along with that of 2,6dibromo-4-[(E)-2-(1-methylpyridinium-4-yl)ethenyl]phenol-ate(MePMBr(2)) was analyzed in terms of increasing probe lipophilicity, through annelation. Values of the empirical solvent polarity scale [E(T)(MePMBr(2))] in kcalmol(-1) correlated linearly with ET(30), the corresponding values for the extensively employed probe 2,6-diphenyl-4-(2,4,6-triphenylpyridinium-1-yl)phenolate (RB). On the other hand, the nonlinear correlations of ET(MeQMBr(2)) or ET(MeAMBr(2)) with E(T)(30) are described by second-order polynomials. Possible reasons for this behavior include: i) self-aggregation of the probe, ii) photoinduced cis/trans isomerization of the dye, and iii) probe structure- and solvent-dependent contributions of the quinonoid and zwitterionic limiting formulas to the ground and excited states of the probe. We show that mechanisms (i) and (ii) are not operative under the experimental conditions employed; experimental evidence (NMR) and theoretical calculations are presented to support the conjecture that the length of the central ethenylic bond in the dye increases in the order MeAMBr(2) > MeQMBr(2) > MePMBr(2), That is, the contribution of the zwitterionic limiting formula predominates for the latter probe, as is also the case for RB, this being the reason for the observed linear correlation between the ET(MePMBr2) and the ET(30) scales. The effect of increasing probe lipophilicity on solvatochromic behavior therefore depends on the strategy employed. Increasing the length of R affects solvatochromism much less than annelation, because the former structural change hardly perturbs the energy of the intramolecular charge-transfer transition responsible for solvatochromism. The thermo-solvatochromic behavior (effect of temperature on solvatochromism) of the three probes was studied in mixtures of water with propanol and/or with DMSO. The solvation model used explicitly considers the presence of three ""species"" in the system: bulk solution and probe solvation shell [namely, water (W), organic solvent (Solv)], and solvent-water hydrogen-bonded aggregate (Solv-W). For aqueous propanol, the probe is efficiently solvated by Solv-W; the strong interaction of DMSO with W drastically decreases the efficiency of Solv-W in solvating the probe, relative to its precursor solvents. Temperature increases resulted in desolvation of the probes, due to the concomitant reduction in the structured characters of the components of the binary mixtures.

Instead of a time-invariant voltammetric profile, many electrochemical systems display a cycle-dependent current-potential response. This phenomenon has been referred to as complex voltammetric response and it has been observed during the electro-oxidation of several molecules such as methanol, ethanol, propanol and hydrogen. There are currently two explanations for the surface mechanism underlying this behavior. In one scenario, the complex voltammogram would result from the specific kinetic pathway taken during the forward sweep. In the other explanation, the phenomenon is discussed in terms of the interplay among the surface roughening and subsequent relaxation, and the ohmic drop coupled to a negative differential resistance. We report in this paper a nanogravimetric investigation of the complex voltammetric response in the electro-oxidation of methanol on platinum electrode in both acidic and alkaline media. Different periodic patterns composed of intercalated small and large hysteresis cycles were observed as a function of the applied voltage and the series resistance between the working electrode and the potentiostat. Independently, nanogravimetric results indicated no detectable difference in the delta-frequency versus voltage profile between small and large hysteresis cycles. These findings were interpreted as experimental evidence of the secondary, if any, role played by the very electrochemical reaction on the emergence of complex voltammetric response. Australian clandestine drug laboratories are constantly utilising alternative methods to produce methamphetamine, in part as restrictions are placed by Government on, for example, chemicals such as phenyl-2-propanone (P2P) (in the early 1980s), or on pseudoephedrine-containing pharmaceuticals, from the mid-2000s. This paper discusses the nitro-aldol reaction occurring between nitroethane and benzaldehyde, which can be utilised in a number of differing routes, in the presence of different bases. The resulting products, namely phenyl-2-nitropropene (P2P pathway) and 2-nitro-1-phenyl-1-propanol (ephedrine pathway) are directly dependant on which base is used; as such, the base may be used to provide an indication of a possible manufacture pathway of methamphetamine at a clandestine laboratory.

A Mg e Mn-Ftalocianina (Mg e Mn-Pc) foram solubilizados à 25°C em dimetilsulfóxido (DMSO); N, N-dimetilacetamida (DMA); N,N-dimetilformamida (DMF); N-metil-formamida, formamida, piridina, o-diclorobenzeno, monoclorobenzeno, tolueno, metanol, etanol, propanol-1, propanol-2, butanol-1 e octanol-1. Alguns valores representativos obtidos para o logarítimo da absortividade molar (E) da Mn-Pc, são os seguintes: o-diclorobenzeno (E = 4,94); DMSO (E = 4,39); octanol-1 (E = 3,90). Valores correspondentes para Mg-Pc são: o-diclarobenzeno (E = 4,93); DMSO (E = 5,22) e Octanol-1 ( E = 5,06). Em função de interação com solventes, pode-se classificar a Mg-Pc como um indicador básico e a Mn-Pc como indicador ácido. Os pigmentos Mg e Mn-Pc foram também solubillzados em soluções aquosas contendo vários surfatantes à 25°C. A Mg-Pc apresentou solubilidade significativa em água contendo brometo de cetiltrimetilamônio (CTAB), Brij-35, cloreto de cetilpiridinio (CPC1), brometo de cetilpiridínio (CPBr,) Triton X-100, cloreto de metildodecilbenziltrimetilamônio, brometo de cetildimetiletilamõnio e brometo de laurilisoquinolínio. A Mn-Pc foi solúvel em soluções aquosas de Brij-35 e Triton X-100. Em função de sua interação com surfatantes a Mg-Pc é classificada como corante catiônico e a Mn-Pc como corante aniônico. O corante comercial quinóide Oil Blue A [1,4-di(isopropilarnina)-antraquinona - 9,10 foi solubilizado à 25°C em DMF, DMSO, DMA, monoclorobenzeno, benzeno, tolueno, piridina, metanol, etanol, propanol-1, propanol-2, butanol-1 e octanol-1. Foi também solubilizado em soluções aquosas de surfatantes, tais como sódio lauril-sulfato (NaLS), cloreto de cetiltrimetilamônio (CTAB), brometo de cetildimetiletilamônio, Triton X-100, cloreto de cetilpiridínio (CPCl), Brij-35, cloreto de rnetildodecilbenziltrimetilamônio e brometo de laurilisoquinolínio. Em função de suas interações com os solventes o corante é um indicador ácido-básico pouco sensível e em função de sua interação com surfatantes é um corante catiônico. 0s resultados experimentais apresentam importância teórica e prática considerando sistemas que envolvem armazenamento e transferência de energia, compostos porfirínicos, fotossíntese, fotocondutores, coletores solares, semi-condutores e processos de embelezamento e proteção de superficies de vários materiais.

Amostras de óleo vegetal foram preparadas na forma de microemulsão de água em óleo (w/o) utilizando dodecil sulfato de sódio (SDS) como surfactante e um álcool como co-surfactante. As microemulsões foram caracterizadas através de medidas de viscosidade, índice de refração, condutividade elétrica, espalhamento de luz dinâmico e voltametria. Ensaios preliminares para a quantificação de analitos por análise direta foram realizados por voltametria linear e eletroforese capilar. As microemulsões de água em óleo de soja apresentaram gotículas de dimensões nanométricas e estabilidade termodinâmica dependente da temperatura, da concentração dos eletrólitos dissolvidos e da natureza do co-surfactante empregado. Por outro lado, o raio hidrodinâmico das gotículas (Rh) diminui com o aumento da temperatura. Quanto aos valores de condutividade, maiores do que os obtidos para o óleo de soja e para a água deionizada, aumentam com a temperatura, na faixa de 20 a 65 oC, e com o teor de água na microemulsão, entre 5 e 7,5 % de água. A maior estabilidade termodinâmica foi alcançada para uma microemulsão contendo 40,0 % de óleo, 43,2 % de pentanol, 10,8 % de SDS e 6,0 % de água, em massa, na razão 1:4 [SDS]:[álcool] Medidas voltamétricas com um ultramicroeletrodo de Pt em microemulsões contendo ferroceno ou ácido oléico dissolvidos evidenciaram a dependência linear das correntes limite anódica e catódica com a concentração da espécie eletroativa. Já a oxidação do ferroceno por voltametria cíclica, usando o mesmo ultramicroeletrodo, mostrou que a diminuição dos coeficientes de difusão nestes meios permite realizar medidas em estado transiente, empregando velocidades convencionais de varredura em potencial. Experimentos por cromatografia eletrocinética em microemulsão reversa (RMEEKC) com n-pentanol como fase contínua permitiram a separação de solutos neutros e aniônicos em amostras de óleo vegetal e gordura animal. Finalmente, foi desenvolvido um procedimento rápido para a separação e identificação dos biofenóis presentes em óleos, utilizando como eletrólito de corrida uma mistura de metanol e 1-propanol contendo KOH. Os resultados evidenciam a possibilidade de análise direta de óleos vegetais empregando métodos eletroanalíticos em microemulsões w/o, e métodos por eletroforese capilar, quer em microemulsões, quer em meio não aquoso.
