A asma brônquica é uma desordem inflamatória crônica, complexa, na qual estão envolvidos fatores genéticos e ambientais. A inflamação das vias aéreas na asma é regulada, predominantemente, por células do sistema imunológico e por uma vasta rede de citocinas que interagem mutuamente e com as vias aéreas. O exato desempenho funcional de cada citocina na fisiopatologia da asma ainda necessita ser completamente estabelecido. A presente investigação teve como objetivo comparar a distribuição dos alelos S e Z do gene da A1AT e do polimorfismo do gene do TNF-α (-308 G/A) em uma população de 110 asmáticos, divididos em dois níveis de severidade da asma (com 54 pacientes no nível da doença moderada persistente e 56 pacientes no nível da doença severa persistente). Os genótipos da A1AT e do TNF-α (-308 G/A) foram determinados pela técnica de digestão enzimática (polimorfismo no comprimento dos fragmentos de restrição). O polimorfismo no promotor do gene do fator de necrose tumoral TNF-α (-308G/A), citocina pro-inflamatória que participa da reação inflamatória em pacientes com asma, contribuindo para a hiperreatividade brônquica, não foi na presente investigação, associado com a doença ou com o aumento da hiperreatividade brônquica, nos níveis de severidade sintomática mais graves da doença.

O carcinoma hepatocelular corresponde à neoplasia maligna primária mais comum do fígado e ao quinto tumor sólido mais frequente no mundo. Altamente letal, permanece como um grave problema de saúde pública em virtude das dificuldades no diagnóstico precoce e na elaboração de medidas terapêuticas efetivas. Estudos recentes no ramo da biologia molecular sugerem que o perfil de miRNAs no carcinoma hepatocelular pode influir consideravelmente na identificação de fatores de riscos associados a oncogenes ou genes supressores. Objetivou-se avaliar a expressão de miRNA 135b, miRNA 181a-5p e miRNA 181a-3p em amostras de Carcinoma Hepatocelular e de Hepatite C Crônica e correlacioná-las de maneira a buscar prováveis biomarcadores relacionados ao mecanismo de carcinogenese. A investigação foi feita em seis pacientes com carcinoma hepatocelular e vinte e quatro casos de Hepatite C Crônica, procedentes do Pará, Norte do Brasil. Todas as amostras de Carcinoma Hepatocelular foram submetidas à microdissecção, para posterior extração do RNA. Para a extração do RNA total e do microRNA foi utilizado o kit AllPrep DNA/RNA FFPE Kit (Qiagen), quantificados pelo equipamento Qubit® 2.0 Fluorometer (Invitrogen) para concentração padrão final de 5ng/μL. Em seguida cDNA foi obtido, utilizando-se TaqMan® MicroRNA Reverse Transcription(AppliedBiosystems). As análises estatísticas foram realizadas nos softwares SPSS 17.0, usando o teste de Mann-Whitney, considerando como significantes valores de p<0,05. Os resultados demonstraram diferenças significativas dos níveis de expressão do miR181a-3p e do miR181a-5p no carcinoma hepatocelular (médias 3,94 e 17,9, respectivamente) em relação à hepatite C crônica (médias de 1,18 e 1,8, respectivamente) com P valor de 0,005 e 0,003. Nesse estudo, observou-se que os miRNAs 181a-3p e 181a-5p, especialmente o 181a-5p, foram significativamente mais expressos nas amostras de carcinoma hepatocelular, quando comparados ao tecido hepático não tumoral com hepatite C crônica. Portanto, os microRNAs possuem características interessantes que os favorecem como possíveis marcadores biológicos no rastreamento de tumores para diagnóstico precoce e terapias alvo selecionadas.

Com o objetivo de contribuir para um melhor conhecimento do envolvimento das infecções pelos vírus das hepatites B e C, na etioepidemiologia do CHC na Amazônia Oriental, estudou-se 36 pacientes em Belém-PA. Foram avaliados marcadores sorológicos e a pesquisa do HBV-DNA e HCV-RNA pela reação em cadeia da polimerase. Observou-se etilismo em 33,3% e cirrose em 83,3%. Marcadores sorológicos das infecções pelo HBV e HCV foram encontrados respectivamente em 88,9% e 8,3%. O HBsAg foi encontrado em 58,3%; anti-HBc em 86%; anti-HBe em 85,7; HBeAg em 9,5%; anti-HBc IgM em 57,1%. O HBV-DNA foi detectado em 37,7% e em 65% dos HBsAg positivos; o HCV-RNA em 8,5% e em 100% dos anti-HCV positivos. AFP esteve alterada em 88,9% e acima de 400ng/ml em 75% dos casos. Conclui-se que a infecção pelo HBV parece ter importância na etiologia do CHC e ressalta-se a importância de implementar programas de vacinação e detecção precoce do tumor.

Preeclampsia is a specific disorder of pregnancy, characterized by arterial hypertension and proteinuria detected after 20 weeks of gestation. This pathology is associated with hyperuricemia, higher levels of pro-inflammatory cytokines, enhanced leukocyte activation and oxidative stress. Adenosine deaminase (ADA) is an enzyme present in all human tissues and, it is involved with the maturation of the immune system. Although its function is not fully understood, ADA is considered an indicator of cellular inflammation and, its increased serum concentration is observed in inflammatory diseases, such as tuberculosis and rheumatoid arthritis. This study aimed to assess serum ADA levels in preeclamptic patients (PE) compared with normotensive pregnant (NT) and non-pregnant women (NP), and to correlate these values with TNF-α and IL-1β production. Ninety pregnant women were included: 60 were pre-eclamptic and 30 were normotensive matched for gestational age. As control group 20 healthy non-pregnant women matched with pregnant for age were included. Peripheral blood mononuclear cells (PMMC) obtained from the three groups studied were cultured with or without lipopolysaccharide (LPS) for 18h at 37oC, and TNF-α and IL-1β production was assessed in the supernatant of cultures by enzyme immunoassay (ELISA). ADA plasmatic concentration was determined by colorimetric method. The results show that ADA plasma levels were significantly higher in PE group compared with NT and NP groups. A positive correlation between ADA and uric acid levels was detected in preeclamptic women. There was no significant difference in relation to ADA levels when PE patients were classified in early and late-onset PE. The endogenous production of IL-1β and TNF-α by PBMC was significantly higher in PE group than in NT and NP women, showing the activation state of these cells in PE. Legg-Calvé-Perthes disease is a non-inflammatory aseptic necrosis of the head of the femur that is found in both young animals and humans before the gap in the femur head closes. In the fields of both human and veterinary medicine the cause of this condition is not known for certain. Various factors have been put forward in the literature as being responsible for the incidence of this condition such as: abnormalities in coagulation, changes in blood flow in the arteries, a septic obstruction in the draining of the epiphysis or the upper parts of the femur, trauma, growth cycle, hyperactivity in a child, genetic influences and dietary factors. Case histories in dogs show that the first stages of the condition progress slowly but that limping or putting weight on the limb worsens at 6 to 8 weeks. Some owners talk about a sharp onset in clinical lameness. Other clinical symptoms may include irritability, loss of appetite and knawing at the hair surrounding the affected hip. In the course of physical examination manipulating the hip joint will cause pain to the animal. The advanced stages of the disease may result in restricted amplitude of movement, muscular atrophy and fracturing. In humans the clinical signs are similar, although progression of the disease is slower so that it can be diagnosed at an earlier stage. In veterinary medicine the diagnosis is, in the main, based on case history, clinical symptoms, physical examination and certain related procedures such as radiography. The various diagnoses include physical trauma and dislocation of the medial patella. In human medicine many people have been correctly diagnosed. It is know that endotoxin and various matrix metalloproteinases (MMPs) are involved in the development of periapical lesions. The purpose of this study was to evaluate and correlate the presence of endotoxins and MMP- 3, MMP-8 and MMP-9 in root canals of teeth with necrotic pulp and periapical lesion before, during and after the biomechanical preparation (PBM) using a combination of different irrigations solutions and intracanal dressing. Thirty-three single-root teeth with a diagnosis of pulp necrosis and periapical lesion radiographically visible were selected. Immediately after the coronal opening was collected the first sample from the root canal content. Then, all canals were prepared (cervical and middle thirds) by oscillatory instruments (EndoEze) and irrigated by 2.5% NaOCl. After, a manual preparation was made for the apical third and the teeth were divided into three groups according to the irrigation protocol: G1) 2.5% NaOCl (4 manual files); G2) 2.5% NaOCl (2 manual files) + [Ca (OH)2 0.14%] (2 manual files) and G3) 2.5% NaOCl (2 manual files) + polymyxin B (2 manual files). After the PBM, the second sample was collected; then the third collect was performed after using EDTA final flush. The fourth sample was collected 14 days after placing the dressing [2% chlorhexidine gel + Ca(OH)2]. Quantification of endotoxins was performed by a kinetic chromogenic lysate from amoebocytes of Limulus (LAL) and quantification of MMPs by ELISA assay. The results were analyzed statistically by Kruskal-Wallis and Dunn's test (5%) and ordinal Spearman correlation. Presence of endotoxin was observed in 100% of cases and G3 showed the greatest reduction of endotoxins from the 1st to the 2nd samples (97%), being statistically similar to G2 (84.2%) and different from G1 (49.4%) (p<0.05).
