The objective of this work was to assess the time-related action of probiotic Lactobacillus plantarum in the bacterial microbiota of the digestive tract of Litopenaeus vannamei, and the relation of total haemocyte count and serum phenol oxidase activity of shrimp challenged with Vibrio harveyi. Shrimps were fed with a probiotic-supplemented diet, for eight days, then shifted to a commercial diet. Shrimps fed only with the commercial diet served as control. Evaluations were made on the 8th day of experiment and repeated two, four, six and eight days later. Total lactic bacteria in the digestive tract was higher until the 4th day of evaluation in the probiotic-supplemented group. Vibrio spp. counts were higher in the control at days zero and two. Until the 4th day of evaluation, the total haemocyte counts in shrimps after challenge with V. harveyi were higher in probiotic-supplemented group than in control group. Significant difference was not observed in phenol oxidase activity. On the 6th day after shifting from supplemented to control diet, all parameters were equal in both groups, suggesting that the time-related action of L. plantarum in shrimp is short.

The bacterial microbiota from the whole gut of soldier and worker castes of the termite Reticulitermes grassei was isolated and studied. In addition, the 16S rDNA bacterial genes from gut DNA were PCR-amplified using Bacteria-selective primers, and the 16S rDNA amplicons subsequently cloned into Escherichia coli. Sequences of the cloned inserts were then used to determine closest relatives by comparison with published sequences and with sequences from our previous work. The clones were found to be affiliated with the phyla Spirochaetes, Proteobacteria, Firmicutes, Bacteroidetes, Actinobacteria, Synergistetes, Verrucomicrobia, and candidate phyla Termite Group 1 (TG1) and Termite Group 2 (TG2). No significant differences were observed with respect to the relative bacterial abundances between soldier and worker phylotypes. The phylotypes obtained in this study were compared with reported sequences from other termites, especially those of phylotypes related to Spirochaetes, Wolbachia (an Alphaproteobacteria), Actinobacteria, and TG1. Many of the clone phylotypes detected in soldiers grouped with those of workers. Moreover, clones CRgS91 (soldiers) and CRgW68 (workers), both affiliated with"Endomicrobia", were the same phylotype. Soldiers and workers also seemed to have similar relative protist abundances. Heterotrophic, poly-β-hydroxyalkanoate-accumulating bacteria were isolated from the gut of soldiers and shown to be affiliated with Actinobacteria and Gammaproteobacteria. We noted that Wolbachia was detected in soldiers but not in workers. Overall, the maintenance by soldiers and workers of comparable axial and radial redox gradients in the gut is consistent with the similarities in the prokaryotes and protists comprising their microbiota.

Procurou-se determinar a menor concentração eficiente de hipoclorito de sódio (NaOCl) para desinfecção da casca, bem como avaliar a necessidade de utilização do mesmo agente sanitizante no banho de imersão da polpa do abacaxi 'Pérola' minimamente processado. Frutos previamente lavados foram desinfectados com NaOCl 100; 150 ou 200mg.L-1, durante 2 minutos. Após aproximadamente 24 horas de armazenamento refrigerado, os frutos foram descascados e fatiados mecanicamente. As fatias foram imersas em solução de NaOCl 20mg.L-1 ou em água (controle), durante 30 segundos. Após período de repouso, para drenagem do excesso de líquido, foram acondicionadas em embalagens de polietileno tereftalato e mantidas à temperatura de 4 ± 1°C, durante 16 dias. As análises microbiológicas, realizadas em intervalos de três dias, foram: coliformes a 35ºC, coliformes a 45ºC, bactérias aeróbias mesófilas e de bolores, e leveduras. Não foram detectados coliformes totais e fecais em nenhum dos tratamentos, durante 16 dias de armazenamento refrigerado. A desinfecção da casca com NaOCl 200mg.L-1, associada à sanitização da polpa do abacaxi com NaOCl 20mg.L-1, proporcionou menores populações de microrganismos aeróbios mesófilos e de bolores, e leveduras, tornando tais operações imprescindíveis na obtenção de produtos minimamente processados que ofereçam garantia de sanidade ao consumidor.

Ameixas, de modo geral, têm curto período de conservação pós-colheita, havendo necessidade de otimizar as condições de colheita e de armazenamento. Este trabalho objetivou avaliar o efeito de diferentes modificadores de atmosfera, durante o armazenamento refrigerado, na qualidade pós-colheita de ameixas cv. Reubennel. Frutas no estádio de maturação meio-maduro foram submetidas aos seguintes modificadores de atmosfera: filmes de polietileno de 12,5 e 15µm de espessura e cera à base de carnaúba e avaliadas após 10; 20; 30 e 40 dias de armazenamento a 0ºC e UR de 90-95%. Avaliaram-se a perda de massa, cor, firmeza da polpa, pH, sólidos solúveis (SS), acidez titulável (AT), relação SS/AT, incidência de podridões, escurecimento interno e características sensoriais. Essas avaliações foram realizadas 3 dias após a retirada das frutas da câmara fria. Verificou-se que a cera à base de carnaúba GARFRESH N (BL9,5), sem diluição, e filmes de polietileno de 12,5 e 15µm reduziram a perda de massa, porém são impróprios para a modificação da atmosfera, em ameixas cv. Reubennel durante o armazenamento refrigerado. Ameixas cv. Reubennel mantêm a qualidade quando armazenadas durante 30 dias a 0ºC sem atmosfera modificada.

Intestinal helminths are potent regulators of their host's immune system and can ameliorate inflammatory diseases such as allergic asthma. In the present study we have assessed whether this anti-inflammatory activity was purely intrinsic to helminths, or whether it also involved crosstalk with the local microbiota. We report that chronic infection with the murine helminth Heligmosomoides polygyrus bakeri (Hpb) altered the intestinal habitat, allowing increased short chain fatty acid (SCFA) production. Transfer of the Hpb-modified microbiota alone was sufficient to mediate protection against allergic asthma. The helminth-induced anti-inflammatory cytokine secretion and regulatory T cell suppressor activity that mediated the protection required the G protein-coupled receptor (GPR)-41. A similar alteration in the metabolic potential of intestinal bacterial communities was observed with diverse parasitic and host species, suggesting that this represents an evolutionary conserved mechanism of host-microbe-helminth interactions.

Piqui (Caryocar brasiliense Camb) oil was transformed into a cocoa butter-like fat through an enzymatic interesterification reaction using Lipozyme in the presence of stearic acid that was incorporated in the sn 1,3-3,1 position into triglyceride. Stearic acid incorporation was determined by HPLC, based on the quantification of the principal triglycerides (POP, POS e SOS) found in cocoa butter. The proposed process was feasible with a reaction time of 240 minutes with 10% of Lipozyme at 70ºC and substract weight ratio of 0,33 (stearic acid:piqui oil).

In the present paper we discuss, based in our experience, some experimental procedures which may be employed for isolation of active compounds from medicinal plants. We have also emphasized some insights about the way to obtain more active and selective compounds from natural products through structural modifications oriented for analysis of structure-activity relationships.

Two models of a glass micro-extractor were constructed after modification of a model proposed in the literature. The two models were used for the simultaneous extraction and enrichment of organochlorinated pesticides from aqueous matrixes to an organic solution appropriate for the gas chromatographic analysis of the pesticide. It was established that the performance of one of the modified micro-extractors permits the pre-concentration of the pesticides to a level that allow their quantitation at the trace level with electron capture detection. It is thus concluded that the glass micro-extractor is a suitable tool for the sample preparation step in the gas chromatographic analysis of environmental pollutants of the class of pesticides.

The applicability of the silylant agents of the general formula Y3Si-R-X, depends on the reactivity of Y group (halide or alcoxide) attached to silicon and the organic function X (halide, amine, thiol, cyanide, etc) in the extreme position of the chain. Both groups are linked together by an organic chain R, containing usually three methylene groups. A series of these agents can be covalently bonded to an inorganic matrix, since the available OH groups are distributed on the surface, making silica gel the most common support. However, other inorganic oxides, zeolites, lamellar inorganic phosphates and chrysotile can also have these agents anchored. Some illustration are presented for immobilized surface in the use as extractors of cations from dilute aqueous or non-aqueous solutions, catalysts agents, ionic exchanged materials, support for enzyme immobilization, chromatographic applications, use in some industrial features and in many other areas. The evolution of this exciting research field to produce new materials, for many tecnological applications, is strongly dependent on the development of a sensible systematic process for the synthesis of a series of new specific silylant agents.

A flow cell assembled on the original geometry of a graphite tube to achieve permanent chemical modifier is proposed. The graphite tube operates as the working electrode. A stainless steel tube, positioned downstream from the working electrode, was used as the auxiliary electrode. The potential value applied on the graphite electrode was measured against a micro reference electrode (Ag/AgCl) inserted into the auxiliary electrode. Palladium solutions in acetate buffer (100 mmol L-1, pH = 4.8), flowing at 0.5 mL min-1 for 60 min was used to perform the electrochemical modification. A mercury solution (1 ng) was used to evaluate the performance of the permanent palladium modifier.

The use of zeolites and other molecular sieves as catalysts is discussed at an introductory level. The text includes a brief historic background on the use of zeolites in catalysis, and a discussion of some chemical and physical properties of silicalite, aluminosilicate, and aluminophosphate molecular sieves. The strategies currently used to chemically modify zeolites and related materials to produce catalysts with increased activity and selectivity are discussed, including the use of redox molecular sieves for hydrocarbon oxidation and the leaching of the active metals from the support.

Interaction between host cells and microbes is known as crosstalk. Among other mechanisms, this takes place when certain molecules of the micro-organisms are recognized by the toll-like receptors (TLRs) in the body cells, mainly in the intestinal epithelial cells and in the immune cells. TLRs belong to the pattern-recognition receptors and represent the first line of defense against pathogens, playing a pivotal role in both innate and adaptive immunity. Dysregulation in the activity of such receptors can lead to the development of chronic and severe inflammation as well as immunological disorders. Among components present in the diet, flavonoids have been suggested as antioxidant dietary factors able to modulate TLR-mediated signaling pathways. This review focuses on the molecular targets involved in the modulatory action of flavonoids on TLR-mediated signaling pathways, providing an overview of the mechanisms involved in such action. Particular flavonoids have been able to modify the composition of the microbiota, to modulate TLR gene and protein expression, and to regulate the downstream signaling molecules involved in the TLR pathway. These synergistic mechanisms suggest the role of some flavonoids in the preventive effect on certain chronic diseases.

The most relevant advances on the analytical applications of chemically modified electrodes (CME) are presented. CME have received great attention due to the possibility of electrode surface modification including chemisorption, composite generation and polymer coating. In recent years, the interest in CME has increased overall to improve the sensitivity and selectivity of the electroanalytical probes, considering the electron mediator incorporation and the new conducting polymers development. The general procedures employed for the electrode modification and the operational characteristics of some electrochemical sensors are discussed.

The oleochemical industry has a permanent interested in controlling the physical, functional and organoleptical properties of their products and in producing useful derivatives from their raw materials. The potential of biotechnology for developing novel or well-known products at more competitive costs meets the need of this industrial segment in expanding their goals. In this work some technical aspects, problems and perspectives related to the production of oil and fat derivatives using biotransformation techniques are discussed. Particular emphasis is given to the description of biotransformation processes using lipase as catalyst, in view of the great versatility of this enzyme class to mediate typical reactions in this technological sector.
