We have previously reported successful trans-complementation of defective Kunjin virus genomic RNAs with a range of large lethal deletions in the nonstructural genes NSI, NS3, and NS5 (A. A. Khromykh et al., J. Virol. 74:3253-3263, 2000). In this study we have mapped further the minimal region in the NS5 gene essential for efficient trans-complementation of genome-length RNAs in repBHK cells to the first 316 of the 905 codons. To allow amplification and easy detection of complemented defective RNAs with deletions apparently affecting virus assembly, we have developed a dual replicon complementation system. In this system defective replicon RNAs with a deletion(s) in the nonstructural genes also encoded the puromycin resistance gene (PAC gene) and the reporter gene for beta-galactosidase (beta-Gal). Complementation of these defective replicon RNAs in repBHK cells resulted in expression of PAC and beta-Gal which allowed establishment of cell lines stably producing replicating defective RNAs by selection with puromycin and comparison of replication efficiencies of complemented defective RNAs by beta-Gal assay. Using this system we demonstrated that deletions in the C-terminal 434 codons of NS3 (codons 178 to 611) were complemented for RNA replication, while any deletions in the first 178 codons were not. None of the genome-length RNAs containing deletions in NS3 shown to be complementable for RNA replication produced secreted defective viruses during complementation in repBHK cells. In contrast, structural proteins produced from these complemented defective RNAs were able to package helper replicon RNA. The results define minimal regions in the NS3 and NS5 genes essential for the formation of complementable replication complex and show a requirement of NS3 in cis for virus assembly.

The approach to maintenance management has changed over the last one hundred years. Over the last few years, the Reliability Engineering and Risk Management Group (RERMG) at the University of Queensland has developed an approach called the strategic maintenance management (SMM) approach. The paper outlines the approach and contrasts it with the current approaches. It then discusses the industry-university partnership in the implementation of this approach and the current activities at the University of Queensland to assist industry in the implementation of the SMM approach.

When products are sold under warranty, the manufacturer incurs additional costs for warranty servicing. Preventive maintenance actions can be used to reduce this cost and these are worthwhile only if the extra cost incurred is less than the reduction achieved. In this paper we propose a new preventive maintenance policy where the parameters are selected optimally to minimize the total warranty servicing cost.

OBJECTIVE: To investigate the prevalence and predictors of weight maintenance over time in a large sample of young Australian women. DESIGN: This population study examined baseline and 4y follow-up data from the cohort of young women participating in the Australian Longitudinal Study on Women's Health. SUBJECTS: A total of 8726 young women aged 18 - 23y at baseline. MEASURES: Height, weight and body mass index (BMI); physical activity; time spent sitting; selected eating behaviours (eg dieting, disordered eating, takeaway food consumption); cigarette smoking, alcohol consumption; parity; and socio-demographic characteristics. RESULTS: Only 44% of the women reported their BMI at follow-up to be within 5% of their baseline BMI (maintainers); 41% had gained weight and 15% had lost weight. Weight maintainers were more likely to be in managerial or professional occupations; to have never married; to be currently studying; and not to be mothers. Controlling for sociodemographic factors, weight maintainers were more likely to be in a healthy weight range at baseline, and to report that they spent less time sitting, and consumed less takeaway food, than women who gained weight. CONCLUSIONS: Fewer than half the young women in this community sample maintained their weight over this 4y period in their early twenties. Findings of widespread weight gain, particularly among those already overweight, suggest that early adulthood, which is a time of significant life changes for many women, may be an important time for implementing strategies to promote maintenance of healthy weight. Strategies which encourage decreased sitting time and less takeaway food consumption may be effective for encouraging weight maintenance at this life stage.

The widespread adoption of soil conservation technologies by farmers (notably contour hedgerows) observed in Guba, Cebu City, Philippines, is not often observed elsewhere In the country. Adoption of these technologies was because of the interaction of such phenomena as site-specific factors, appropriate extension systems, and technologies. However, lack of hedgerow maintenance, decreasing hedgerow quality, and disappearance of hedgerows raised concerns about sustainability. The dynamic nature of upland farming systems suggests the need for a location-specific farming system development framework, which provides farmers with ongoing extension for continual promotion of appropriate conservation practices.

Chen and Popova [Res. Engng Syst. Saf. 77 (2002) 61] discuss maintenance policies for items sold with a two-dimensional warranty. However, their paper fails to give a proper review of the literature and it also contains errors. Objectives: The aim was to determine whether methadone maintenance treatment reduced heroin use, syringe sharing and HIV or hepatitis C incidence among prisoners. Methods: All eligible prisoners seeking drug treatment were randomised to methadone or a waitlist control group from 1997 to 1998 and followed up after 4 months. Heroin use was measured by hair analysis and self report; drugs used and injected and syringe sharing were measured by self report. Hepatitis C and HIV incidence was measured by serology. Results: Of 593 eligible prisoners, 382 (64%) were randomised to MMT (n = 191) or control (n = 191). 129 treated and 124 control subjects were followed up at 5 months. Heroin use was significantly lower among treated than control subjects at follow up. Treated subjects reported lower levels of drug injection and syringe sharing at follow up. There was no difference in HIV or hepatitis C incidence. Conclusions: Consideration should be given to the introduction of prison methadone programs particular where community based programs exist. Although it is the best characterized in vitro model of GH action, the mechanisms used by GH to induce differentiation of murine 3T3-F442A preadipocytes remain unclear. Here we have examined the role of three transcriptional regulators in adipogenesis. These regulators are either rapidly induced in response to GH [Stra13, signal transducer and activator of transcription (Stat) 3] or of central importance to GH signaling (Stat5). Retroviral transfection of 3T3-F442A preadipocytes was used to increase expression of Stra13, Stat3, and Stat5a. Only Stat5a transfection increased the expression of adipogenic markers peroxisome proliferator-activated receptor gamma, CCAAT enhancer binding protein (C/EBP)alpha, and adipose protein 2/fatty acid-binding protein in response to GH, as determined by quantitative RT-PCR. Transfection with constitutively active Stat3 and Stat5a revealed that constitutively active Stat5a but not Stat3 was able to replace the GH requirement for adipogenesis. Constitutively active Stat5a but not Stat3 was able to increase the formation of lipid droplets and expression of alpha-glycerol phosphate dehydrogenase toward levels seen in mature adipocytes. Constitutively active Stat5a was also able to increase the expression of transcripts for C/EBPalpha to similar levels as GH, and of C/EBPbeta, peroxisome proliferator-activated receptor gamma, and adipose protein 2/fatty acid-binding protein transcripts to a lesser extent. An in vivo role for GH in murine adipogenesis is supported by significantly decreased epididymal fat depot size in young GH receptor-deleted mice, before manifestation of the lipolytic actions of GH. We conclude that Stat5 is a critical factor in GH-induced, and potentially prolactin-induced, murine adipogenesis.

The maintenance of arterial pressure at levels adequate to perfuse the tissues is a basic requirement for the constancy of the internal environment and survival. The objective of the present review was to provide information about the basic reflex mechanisms that are responsible for the moment-to-moment regulation of the cardiovascular system. We demonstrate that this control is largely provided by the action of arterial and non-arterial reflexes that detect and correct changes in arterial pressure (baroreflex), blood volume or chemical composition (mechano- and chemosensitive cardiopulmonary reflexes), and changes in blood-gas composition (chemoreceptor reflex). The importance of the integration of these cardiovascular reflexes is well understood and it is clear that processing mainly occurs in the nucleus tractus solitarii, although the mechanism is poorly understood. There are several indications that the interactions of baroreflex, chemoreflex and Bezold-Jarisch reflex inputs, and the central nervous system control the activity of autonomic preganglionic neurons through parallel afferent and efferent pathways to achieve cardiovascular homeostasis. It is surprising that so little appears in the literature about the integration of these neural reflexes in cardiovascular function. Thus, our purpose was to review the interplay between peripheral neural reflex mechanisms of arterial blood pressure and blood volume regulation in physiological and pathophysiological states. Special emphasis is placed on the experimental model of arterial hypertension induced by N-nitro-L-arginine methyl ester (L-NAME) in which the interplay of these three reflexes is demonstrable.

In subtropical climate areas, the models and methods proposed to evaluate the chilling requirement of temperate fruit crops often do not provide satisfactory results, thus calling for the development of alternative techniques. The aim of this study was to evaluate the correlations between some phonological traits and chilling requirement for seed germination of 18 peach cultivars and one nectarine cultivar. Two experiments were installed separately for the correlation studies. In experiment 1, the phenological traits were observed in the field, while in experiment 2, the chilling requirement for 50 and 100% seed germination of each cultivar was assessed. The number of days for beginning of bloom (r = 0.70**, 0.61**) and full bloom (r = 0.72**, 0.76**) were both significantly correlated with the number of chilling units for 50% and 100% germination of seeds. The number of days for beginning of budding and dormancy break were both significantly correlated with the number of chilling units for 50% and 100% germination (r = 0.48*, 0.50*, respectively). However, the same significant effect for these phenological traits was not found between chilling units and 50% germination of seeds, as well as between chilling units and harvest dates.

In this paper we discuss interesting developments of expert systems for machine diagnosis and condition-based maintenance. We review some elements of condition-based maintenance and its applications, expert systems for machine diagnosis, and an example of machine diagnosis. In the last section we note some problems to be resolved so that expert systems for machine diagnosis may gain wider acceptance in the future.

No âmbito do Ciclo de Estudos conducente ao grau de Mestre em Engenharia Mecânica, Per-fil de Manutenção e Produção, foi desenvolvida esta Dissertação, que consiste no estudo do estado actual de implementação da “Total Productive Maintenance – TPM” na Área de Pren-sas da Volkswagen Autoeuropa, identificando os benefícios colhidos com a referida imple-mentação, e também algumas dificuldades. No final desta Dissertação, são enunciadas as considerações resultantes do presente estudo e são emitidas algumas sugestões, no sentido de que este trabalho se torne uma mais-valia para a Área de Prensas da Volkswagen Autoeuropa, contribuindo positivamente na busca da melhoria contínua, dos zero defeitos e das zero falhas e dos zero desperdícios e, finalmente propor um plano de melhoria, ou seja, uma adaptação do actual modelo funcional face ao modelo teórico.

Este relatório, escrito no âmbito do estágio profissional realizado no departamento de Manutenção e Engenharia da TAP Portugal, com o tema “Análise do Line Maintenance Manual numa perspectiva de melhoria contínua do Programa de Manutenção Avião”, tem como objectivo descrever a análise efectuada e as oportunidades de melhoria identificadas no Line Maintenance Manual, documento que serve de suporte às actividades da Manutenção de Linha. Espera-se que o trabalho realizado traga benefícios para a TAP Manutenção e Engenharia a médio e longo prazo, tais como a agilização das futuras revisões ao referido documento, a redução de recursos envolvidos nas inspecções de trânsito e, quiçá, a correcção de aspectos com implicações directas na operação segura das aeronaves. O trabalho desenvolvido no estágio implicou um estudo aprofundado do processo que resulta no estabelecimento dos programas de manutenção das aeronaves actuais, bem como dos documentos nele envolvidos, pelo que a análise específica do Line Maintenance Manual foi realizada no contexto muito mais abrangente da teoria da Manutenção Aeronáutica.

A utilização de recursos energéticos renováveis apresenta-se como um caminho vital para a humanidade alcançar um desenvolvimento sustentável. Nesta campanha, a energia eólica surge como um dos principais vectores de orientação tendo evoluído de forma quase exponencial nos últimos anos. No entanto, apesar da sua relativa maturidade, esta tecnologia enfrenta ainda alguns problemas e desafios. Não obstante a experiência empírica da indústria eólica, adquirida nos últimos trinta anos e dos esforços para melhorar a fiabilidade operacional das turbinas, as taxas de falha ainda se apresentam elevadas. Face às correntes práticas de Manutenção das turbinas e parques eólicos e às características de falha, (por vezes catastróficas), existe a necessidade de optimizar as estratégias de manutenção das turbinas eólicas e reduzir os custos durante o ciclo de vida, de modo a maximizar o retorno do investimento. Descreve-se neste trabalho o estado do conhecimento actual face ao objectivo pretendido, a recolha de dados reais da operação e Manutenção, a aplicabilidade dos modelos escolhidos para obtenção da probabilidade de falha, e as consequências e avaliação do risco. Assim, desenvolveu-se uma ferramenta de apoio à decisão, baseada em Modelos de RBI (Risk Based Inspection) e RBIM (Risk Based Inspection and Maintenance) aplicados a turbinas eólicas.

Longevity risk is one of the major risks that an insurance company or a pension fund has to deal with and it is expected that its importance will grow in the near future. In agreement with these considerations, in Solvency II regulation the Standard formula furnished for calculating the Solvency Capital Requirement explicitly considers this kind of risk. According to the new European rules in our paper we suggest a multiperiod approach to evaluate the SCR for longevity risk. We propose a backtesting framework for measuring the consistency of SCR calculations for life insurance policies.
