PROBING THE SYMMETRY ENERGY AT SUPRA-SATURATION DENSITIES FROM PION EMISSION IN HEAVY-ION COLLISIONS

Within the framework of the improved isospin dependent quantum molecular dynamics (ImIQMD) model, the emission of pion in heavy-ion collisions in the region 1 A GeV as a probe of nuclear symmetry energy at supra-saturation densities is investigated systematically, in which the pion is considered to be mainly produced by the decay of resonances Delta(1232) and N*(1440). The pi(-)/pi(+) yields are calculated for selected Skyrme parameters SkP, SLy6, Ska and SIII, and also for the cases of different stiffness of symmetry energy with the parameter SLy6. Preliminary results compared with the measured data by the FOPI collaboration favor a hard symmetry energy of the potential term proportional to (rho/rho(0))(gamma s) with gamma(s) = 2.

中能重离子碰撞中的核反应机制及其形成的高激发热核的性质是中能重离子物理研究的重要领域,而高激发热核性质的同位旋效应研究是这一领域的热点之一。选取了具有不同N亿比的反应体系以研究激发热核性质的同位旋效应。本论文涉及的反应系统三对共六个反应体系:55MeV/u~(40)Ar+~(58.64)Ni、30MeV/u~(40)Ar+~(112,124)Sn、35Mev/u~(36)Ar+~(112,124)Sn,这六个反应体系的N/z比分别为1.13,1.26、1.24,1.41、1.18,1.35。分别从带电粒子多重性、相对态布居核温度、关联函数等角度研究了这三对反应体系高激发热核性质的同位旋效应。在55MeV/u 40Ar+58,64Ni核反应中,用兰州4π带电粒子探测器阵列测量带电粒子多重性,研究了He和中等质量碎片的产额与反应系统的同位旋的关系,以及这种同位旋效应与反应系统的碰撞参数(即碰撞的激烈程度)、系统的激发能的变化关系。对两个反应系统,观察到带电粒子多重性中He的比分随带电粒子多重性的增加而增大,带电粒子多重性中IMF的比分随带电粒子多重性的增加而先增大,后减小的规律。两个反应系统虽然具有相同的核电荷数,但轻粒子He和中等质量碎片在多重性中的比分有明显的同位旋相关性。在30Mev/u40Sn、35MeV/u~(40)Ar~(112,124)Sn、35Mev/u 36Ar+112,124Sn反应中用13单元望远镜探测器阵列测量了小角关联粒子。由价a关联函数提取了30Mev/u 40Ar+112,12Sn反应系统中激发热核的态布居核温。对于不同同位旋反应系统舜UAr+112Sn和4VAr+124Sn,提取的相对杰布居核温度分别是4.18+0.28/0.21MEV和4.10士0.22/0.20MeV;考察态布居核温度和粒子能量的关薰时,观察到两个系统的发射温度均随着粒子能量的增加而降低,缺中子系统40Ar+l12Sn中由低能时的5.13士.30/0.26MEV降低到高能时的3.87士0.37/0.29MeV,丰中子系统40Ar+124Sn中由低能时的5.39士0.30/0.26MeV降低到高能时的3.32士MeV。讨论了这种布居态核温度的同位旋相关性。在35Mev/u 36Ar+112,124Sn反应中提取了洲F(3‘25)的约化速度关联函数。相对丰中子36Ar+124Sn系统的IMF关联函数在小约化速度处反关联程度更强,表明36Ai+124Sn系统的发射IMF的平均时间更短。用MENEKA程序提取了两个系统IMF的平均发射时间,36Ai+112sn反应中IMF的发射时间约为150fm/c,而36Ar十124Sn反应IME的发射时间稍短,约为120fm/c。以关联IMF的单核子总能量/动量为窗条件,发现低能IMF关联函数几乎没有差别,而高能IMF关联函数在小约化速度处的差别更为明显,表明两个系统IMF关联函数的同位旋效应可能来自于IMF的早期发射。为了得到进一步的信息,我们提取了高动量窗条件下的IMF发射时间,它们比平均发射时间短,36Ar+112Sn反应中高能IMF的发射时间约为100蒯c,而36Ai+124Sn反应中IMF的发射时间则更短,约为50fm/c。

Intracerebroventricular (ICV) administration of bombesin (BN) induces a
syndrome characterized by stereotypic locomotion and grooming,
hyperactivity and sleep elimination, hyperglycemia and hypothermia,
hyperhemodynamics, feeding inhibition, and gastrointestinal function
changes. Mammalian BN-like peptides (MBNs), e.g. gastrin-releasing
peptide (GRP), Neuromedin C (NMC), and Neuromedin B (NMB), have been
detected in the central nervous system. Radio-labeled BN binds to specific
sites in discrete cerebral regions. Two specific BN receptor subtypes (GRP
receptor and NMB receptor) have been identified in numerous brain regions.
The quantitative 2-[14C]deoxyglucose ([14C]20G) autoradiographic
method was used to map local cerebral glucose utilization (LCGU) in the
rat brain following ICV injection of BN (vehicle, BN O.1Jlg, O.5Jlg). At each
dose, experiments were conducted in freely moving or restrained
conditions to determine whether alterations in cerebral function were the
result of BN central administration, or were the result of BN-induced

The pH(i) (intracellular pH) is an important physiological parameter which is altered during hypoxia and ischaemia, pathological conditions accompanied by a dramatic decrease in pH(i). Sensors of pH(i) include ion transport systems which control intracellular Ca2+ gradients and link changes in pH(i) to functions as diverse as proliferation and apoptosis. The annexins are a protein family characterized by Ca2+-dependent interactions with cellular membranes. Additionally, in vitro evidence points to the existence of pH-dependent, Ca(2+)-independent membrane association of several annexins. We show that hypoxia promotes the interaction of the recombinant annexin A2-S100A10 (p11) and annexin A6 with the plasma membrane. We have investigated in vivo the influence of the pH(i) on the membrane association of human annexins A1, A2, A4, A5 and A6 tagged with fluorescent proteins, and characterized this interaction for endogenous annexins present in smooth muscle and HEK (human embryonic kidney)-293 cells biochemically and by immunofluorescence microscopy. Our results show that annexin A6 and the heterotetramer A2-S100A10 (but not annexins A1, A4 and A5) interact independently of Ca2+ with the plasma membrane at pH 6.2 and 6.6. The dimerization of annexin A2 within the annexin A2-S100A10 complex is essential for the pH-dependent membrane interaction at this pH range. The pH-induced membrane binding of annexins A6 and A2-S100A10 might have consequences for their functions as membrane organizers and channel modulators.

Friedreich's ataxia is caused by the expansion of the GAA•TTC trinucleotide repeat sequence located in intron 1 of the frataxin gene. The long GAA•TTC repeats are known to form several non-B DNA structures including hairpins, triplexes, parallel DNA and sticky DNA. Therefore it is believed that alternative DNA structures play a role in the loss of mRNA transcript and functional frataxin protein in FRDA patients. We wanted to further elucidate the characteristics for formation and stability of sticky DNA by evaluating the structure in a plasmid based system in vitro and in vivo in Escherichia coli. The negative supercoil density of plasmids harboring different lengths of GAA•TTC repeats, as well as either one or two repeat tracts were studied in E. coli to determine if plasmids containing two long tracts (≥60 repeats) in a direct repeat orientation would have a different topological effect in vivo compared to plasmids that harbored only one GAA•TTC tract or two tracts of < 60 repeats. The experiments revealed that, in fact, sticky DNA forming plasmids had a lower average negative supercoil density (-σ) compared to all other control plasmids used that had the potential to form other non-B DNA structures such as triplexes or Z-DNA. Also, the requirements for in vitro dissociation and reconstitution of the DNA•DNA associated region of sticky DNA were evaluated. Results conclude that the two repeat tracts associate in the presence of negative supercoiling and MgCl 2 or MnCl2 in a time and concentration-dependent manner. Interaction of the repeat sequences was not observed in the absence of negative supercoiling and/or MgCl2 or in the presence of other monovalent or divalent cations, indicating that supercoiling and quite specific cations are needed for the association of sticky DNA. These are the first experiments studying a more specific role of supercoiling and cation influence on this DNA conformation. To support our model of the topological effects of sticky DNA in plasmids, changes in sticky DNA band migration was measured with reference to the linear DNA after treatment with increasing concentrations of ethidium bromide (EtBr). The presence of independent negative supercoil domains was confirmed by this method and found to be segregated by the DNA-DNA associated region. Sequence-specific polyamide molecules were used to test the effect of binding of the ligands to the GAA•TTC repeats on the inhibition of sticky DNA. The destabilization of the sticky DNA conformation in vitro through this binding of the polyamides demonstrated the first conceptual therapeutic approach for the treatment of FRDA at the DNA molecular level. ^ Thus, examining the properties of sticky DNA formed by these long repeat tracts is important in the elucidation of the possible role of sticky DNA in Friedreich's ataxia. ^

The infected cell protein 0 (ICP0) of herpes simplex virus 1, a promiscuous transactivator shown to enhance the expression of genes introduced into cells by infection or transfection, interacts with numerous cellular proteins and has been linked to the disruption of ND10 and degradation of several proteins. ICP0 contains a RING finger domain characteristic of a class of E3 ubiquitin ligases. We report that: (i) in infected cells, ICP0 interacts dynamically with proteasomes and is bound to proteasomes in the presence of the proteasome inhibitor MG132. Also in infected cells, cdc34, a polyubiquitinated E2 ubiquitin-conjugating enzyme, exhibits increased ICP0-dependent dynamic interaction with proteasomes. (ii) In an in vitro substrate-independent ubiquitination system, the RING finger domain encoded by exon 2 of ICP0 binds cdc34, whereas the carboxyl-terminal domain of ICP0 functions as an E3 ligase independent of the RING finger domain. The results indicate that ICP0 can act as a unimolecular E3 ubiquitin ligase and that it promotes ubiquitin-protein ligation and binds the E2 cdc34. It differs from other unimolecular E3 ligases in that the domain containing the RING finger binds E2, whereas the ligase activity maps to a different domain of the protein. The results also suggest that ICP0 shuttles between nucleus and cytoplasm as a function of its dynamic interactions with proteasomes.

A wide range of molecules acting as apoptotic cell-associated ligands, phagocyte-associated receptors or soluble bridging molecules have been implicated within the complex sequential processes that result in phagocytosis and degradation of apoptotic cells. Intercellular adhesion molecule 3 (ICAM-3, also known as CD50), a human leukocyte-restricted immunoglobulin super-family (IgSF) member, has previously been implicated in apoptotic cell clearance, although its precise role in the clearance process is ill defined. The main objective of this work is to further characterise the function of ICAM-3 in the removal of apoptotic cells. Using a range of novel anti-ICAM-3 monoclonal antibodies (mAbs), including one (MA4) that blocks apoptotic cell clearance by macrophages, alongside apoptotic human leukocytes that are normal or deficient for ICAM-3, we demonstrate that ICAM-3 promotes a domain 1-2-dependent tethering interaction with phagocytes. Furthermore, we demonstrate an apoptosis-associated reduction in ICAM-3 that results from release of ICAM-3 within microparticles that potently attract macrophages to apoptotic cells. Taken together, these data suggest that apoptotic cell-derived microparticles bearing ICAM-3 promote macrophage chemoattraction to sites of leukocyte cell death and that ICAM-3 mediates subsequent cell corpse tethering to macrophages. The defined function of ICAM-3 in these processes and profound defect in chemotaxis noted to ICAM-3-deficient microparticles suggest that ICAM-3 may be an important adhesion molecule involved in chemotaxis to apoptotic human leukocytes. © 2012 Macmillan Publishers Limited All rights reserved.

Diversity of T cell receptors (TCR) and immunoglobulins (Ig) is generated by V(D)J recombination of antigen receptor (AgR) loci. The Tcra-Tcrd locus is of particular interest because it displays a nested organization of Tcrd and Tcra gene segments and V(D)J recombination follows an intricate developmental program to assemble both TCRδ and TCRα repertoires. However, the mechanisms that dictate the developmental regulation of V(D)J recombination of the Tcra-Tcrd locus remain unclear.
We have previously shown that CCCTC-binding factor (CTCF) regulates Tcra gene transcription and rearrangement through organizing chromatin looping between CTCF- binding elements (CBEs). This study is one of many showing that CTCF functions as a chromatin organizer and transcriptional regulator genome-wide. However, detailed understanding of the impact of specific CBEs is needed to fully comprehend the biological function of CTCF and how CTCF influences the generation of the TCR repertoire during thymocyte development. Thus, we generated several mouse models with genetically modified CBEs to gain insight into the CTCF-dependent regulation of the Tcra-Tcrd locus. We revealed a CTCF-dependent chromatin interaction network at the Tcra-Tcrd locus in double-negative thymocytes. Disruption of a discrete chromatin loop encompassing Dδ, Jδ and Cδ gene segments allowed a single Vδ segment to frequently contact and rearrange to diversity and joining gene segments and dominate the adult TCRδ repertoire. Disruption of this loop also narrowed the TCRα repertoire, which, we believe, followed as a consequence of the restricted TCRδ repertoire. Hence, a single CTCF-mediated chromatin loop directly regulates TCRδ diversity and indirectly regulates TCRα diversity. In addition, we showed that insertion of an ectopic CBE can modify chromatin interactions and disrupt the rearrangement of particular Vδ gene segments. Finally, we investigated the role of YY1 in early T cell development by conditionally deleting YY1 in developing thymocytes. We found that early ablation of YY1 caused severe developmental defects in the DN compartment due to a dramatic increase in DN thymocyte apoptosis. Furthermore, late ablation of YY1 resulted in increased apoptosis of DP thymocytes and a restricted TCRα repertoire. Mechanistically, we showed that p53 was upregulated in both DN and DP YY1-deficient thymocytes. Eliminating p53 in YY1-deficient thymocytes rescued the survival and developmental defects, indicating that these YY1-dependent defects were p53-mediated. We conclude that YY1 is required to maintain cell viability during thymocyte development by thwarting the accumulation of p53.
Overall, this thesis work has shown that CTCF-dependent looping provides a central framework for lineage- and developmental stage-specific regulation of Tcra-Tcrd gene expression and rearrangements. In addition, we identified YY1 as a novel regulator of thymocyte viability.
