With the increasing environmental awareness, maximizing biodegradability and minimizing ecotoxicity is the main driving force for new technological developments. Thus, can be developed new biodegradable lubricants for use in environmentally sensitive areas. The aim of this study was to obtain new bio-lubricants from passion fruit (Passiflora edulis Sims f. flavicarpa Degener) and moringa (Moringa oleifera Lamarck) epoxidized oils and develop a new additive package using experimental design for their use as a hydraulic fluid. In the first stage of this work was performed the optimization of the epoxidation process of the oils using fractional experimental design 24-1 , varying the temperature, reaction time, ratio of formic acid and hydrogen peroxide. In the second step was investigated the selectivity, thermodynamics and kinetics of the reaction for obtaining the two epoxides at 30, 50 and 70 °C. The result of the experimental design confirmed that the epoxidation of passion fruit oil requires 2 hours of reaction, 50 °C and a ratio H2O2/C=C/HCOOH (1:1:1). For moringa oil were required 2 hours reaction, 50 °C and a ratio of H2O2/C=C/HCOOH (1:1:1.5). The results of the final conversions were equal to 83.09% (± 0.3) for passion fruit oil epoxide and 91.02 (±0,4) for moringa oil epoxide. Following was made the 23 factorial design to evaluate which are the best concentrations of corrosion inhibitor and anti-wear (IC), antioxidant (BHA) and extreme pressure (EP) additives. The bio-lubricants obtained in this step were characterized according to DIN 51524 (Part 2 HLP) and DIN 51517 (Part 3 CLP) standards. The epoxidation process of the oils was able to improve the oxidative stability and reduce the total acid number, when compared to the in natura oils. Moreover, the epoxidized oils best solubilized additives, resulting in increased performance as a lubricant. In terms of physicochemical performance, the best lubricant fluid was the epoxidized moringa oil with additives (EMO-ADI), followed by the epoxidized passion fruit oil with additives (EPF-ADI) and, finally, the passion fruit in natura oil without additives (PFO). Lastly, was made the investigation of the tribological behavior under conditions of boundary lubrication for these lubricants. The tribological performance of the developed lubricants was analyzed on a HFRR equipment (High Frequency Reciprocating Rig) and the coefficient of friction, which occurs during the contact and the formation of the lubricating film, was measured. The wear was evaluated through optical microscopy and scanning electron microscopy (SEM). The results showed that the addition of extreme pressure (EP) and anti-wear and corrosion inhibitor (CI) additives significantly improve the tribological properties of the fluids. In all assays, was formed a lubricating film that is responsible for reducing the coefficient of metal-to-metal wear. It was observed that the addition of EP and IC additives in the in natura vegetable oils of passion fruit and moringa did not favor a significant reduction in wear. The bio-lubricants developed from passion fruit and moringa oils modified via epoxidation presented satisfactory tribological properties and shown to be potential lubricants for replacement of commercial mineral-based fluids.

The thermodynamic properties of anchovy fillets and enzymatic modified pastes in two hydrolysis degrees (3% HD and 14% HD), at 50, 60 and 70 C were evaluated. The GAB model was used to calculate the values of the monolayer moisture content and the thermodynamic properties of the samples. The enzymatic modification led to the increases of the superficial area and differential enthalpies, and decrease of the differential entropies in relation the samples in natura. The enthalpy–entropy compensation showed that the process was controlled by the enthalpy, it was only spontaneous for the samples in natura. Pore size decreased with enzymatic modification, and all samples were in the limit of region between micropores and mesopores (<2 nm) for moisture content of 15%, and mesopores (from 2 to 50 nm) to moisture content above 15%.

An effective hygiene and sanitation inspection of meat and meat products is essential for its production and commercialization. For this reason, the national and international standards responsible for these products quality control employs microbiological analyses methods as quality control tools. In December of 2012, it was included in the Ministério da Agricultura Pecuária e Abastecimento (MAPA) website, a Microbiological Scope of food and water, which presents the replacement of some methods proposed by the Normative Instruction 62. Some of these methodologies are considered rapid, practical and convenient. However, other methodologies were still replaced by conventional ones, which presents disadvantages as incorrect interpretations of the microorganism phenotypical and biochemical characteristics, leading to the misinterpretation of test results. Therefore, the objective of this study is to develop a comprehensive, practical and illustrative guidebook of microbiological analysis for in natura poultry cuts. The methods addressed in this guide are the official standards analysis required by the poultry cuts legislation, which are the Escherichia coli count, the thermotolerant coliforms count, the aerobic plate count and the detection of Salmonella spp. The approached methodologies for these analysis will be the AOAC 998.08, the Normative Instruction 62 and the ISO 4833-1:2013 and ISO 6579:2002, respectively. In these events, it is expected to obtain an enlightening and approved guidebook evaluated by laboratory technicians, which will help reduce the analytical subjectivity leading to a more reliable interpretation of the test results.

The physicochemical characteristics of three Brazilian pears were investigated using elemental analysis, scanning electron microscopy (SEM), X-ray diffractometry (XRD) and studies of Cr(III) biosorption based on adsorption isotherms. Adsorption of Cr(III) by in natura peat from Santo Amaro das Brotas (Sergipe State) was much greater than by peats from either Ribeirao Preto (São Paulo State) or Itabaiana (Sergipe State), with adsorption capacities (q) of 4.90 +/- 0.01, 1.70 +/- 0.01 and 1.40 +/- 0.01 mgg (1), respectively. Pre-treatments with HCl and NaOH + HCl reduced adsorption by the Santo Amaro clas Brotas peat, showing that adsorption efficiency was associated with the amount of organic matter present. Conversely, increase in the mineral content following pre-treatment increased adsorption of Cr(III) by the Ribeirao Preto and Itabaiana peats. Highest adsorption (retention >95.0%) was achieved at equilibrium pH 4.0 using the Santo Amaro das Brotas peat. Experimental data for the adsorption of Cr(III) from aqueous solution onto this peat were fitted to the Langmuir equation, from which an equilibrium adsorption capacity, q(max), of 5.60 mgg(-1) was obtained, which was close to the experimentally determined value. O preparo de amostra é frequentemente a fase crítica de um método multirresíduo, devido à diversidade de substâncias que podem ser extraídas e a ineficiência da extração dos analitos de interesse, aplicados em matrizes com características e composições próprias. Os agrotóxicos são moléculas de estruturas complexas, com características químicas diferentes, como polaridade e estabilidade, e muitas vezes se degradam rapidamente durante o procedimento analítico a seus metabólitos ou a outros produtos. Por estas razões, se faz necessário o desenvolvimento de métodos de preparo de amostras, cada vez mais eficientes e rápidos para determinações de analitos em matrizes complexas. A Extração Líquido-Líquido (ELL) apresenta recuperações satisfatórias para compostos apolares e de média polaridade, porém tem algumas desvantagens relevantes, como pode formar emulsões, requer grandes volumes de solventes, demanda longo tempo de extração e é de difícil automação. A Extração em Fase Sólida (EFS), uma alternativa à ELL, exige pequenos volumes de solvente e é de fácil operação e automação. Dependendo do sorvente, pode ser empregada na análise de compostos polares e apolares. O método QuEChERS apresenta como vantagens rapidez, simplicidade, baixo custo, eficiência, robustez, segurança, baixo consumo de solventes, além de cobrir uma ampla variedade de classes de agrotóxicos (com caráter ácido, básico ou polar). Neste trabalho, foi feito um estudo comparativo entre as técnicas de preparo de amostra: ELL modificada, EFS e o método QuEChERS na extração dos agrotóxicos, ciproconazol, difenoconazol, fenarimol, tebuconazol e triadimefom, aplicados em culturas de uva, para posterior determinação por Cromatografia Líquida de Alta Eficiência (CLAE).

Este trabalho propõe um modelo de prospecção de cenários e avaliação de projetos de atividades econômicas de produção agrícola de mamona caracterizada como agricultura familiar, extração de óleo vegetal in natura de sementes de mamona e produção de biodiesel (transesterificação), na forma de cadeia produtiva integrada. O modelo utiliza indicadores de avaliação econômico-flnanceiro, ambiental e social, como por exemplo, VPL, TIR, PBS, PBD, custo de produção, renda familiar, número de famílias assentadas e funcionários empregados, e outros, para identificar o desempenho de cada proposta de projeto. Para tanto, foi desenvolvido um ambiente computacional "AGRIFIS" para abrigar o modelo proposto nesse trabalho, que é uma ferramenta de apoio à tomada de decisão para investimentos em cadeia produtiva de mamona, pois se pode estimar o fluxo de caixa futuro de uma determinada proposta de projeto, incluindo as lucratividades, as despesas, a rentabilidade das famílias assentadas para determinadas condições pré-estabelecidas.

Objetivou-se foi avaliar a fertilização artificial e a duração da motilidade espermática em pacus com diferentes doses inseminantes, volumes de água e preservação do sêmen in natura. Foram realizados quatro experimentos para avaliação do efeito de doses inseminantes (7x10³, 7x10(4), 7x10(5), 7x10(6) e 7x10(7) espermatozoides ovócito-1) sobre a fertilização artificial dos ovócitos; do efeito do volume de água (0,5; 15,0; 30,0; 45,0 e 60,0 mL de água mL-1 de ovócitos) com doses inseminantes de 105.481 e 210.963 espermatozoides ovócito-1; do efeito de diluição do sêmen (0,005; 0,05; 0,5 e 5,0 µL de sêmen mL-1 de água) sobre a duração da motilidade espermática; e do efeito do armazenamento a 15 ºC por 9 h sobre a duração da motilidade espermática e o índice de sobrevivência espermática. Os maiores resultados obtidos foram: doses inseminantes entre 7x10³ e 7x10(7) espermatozoides ovócito-1; 15 a 60 mL de água mL-1 de ovócitos; diluição de 0.005 µL sêmen mL-1 de água e 98,65% de sobrevivência espermática até o tempo de preservação de 2h45min36s. A preservação a 15ºC por 9 horas não influencia a duração da motilidade espermática. As maiores taxas de fertilização podem ser observadas no emprego de 0,27 a 270 µL de sêmen mL-1 de ovócitos, com 15 a 60 mL de água para ativação.

O BRS PC apresenta frutos ovoides e globosos, com massa de 50 a 60g e com 5,3 cm de comprimento por 3,8cm de diâmetro, casca verde-amarelada e rajadas, apresentando 5 listras longitudinais da base ao ápice do fruto, polpa amarelo creme e sementes pequenas e escuras, fruto aromático, suco doce-acidulado e saboroso (COSTA et al. 2015). Dadas as suas características sensoriais a Passiflora setacea vem ganhando espaço nos mercados dos centros urbanos e constitui-se em uma espécie com potencial comercial notável. O presente trabalho teve por objetivo principal a análise da aceitabilidade do fruto de Passiflora setacea- BRS Pérola do Cerrado.

La pelle dello squalo è stata validata come un efficace sistema di controllo della resistenza di attrito privilegiato in regime turbolento. Essa è stata oggetto di numerosi studi volti a rendere realizzabili repliche quanto più simili alla sua reale struttura superficiale. In questa trattazione, partendo da una breve digressione storica sull’evoluzione dello studio delle rugosità superficiali, sono state analizzate le principali campagne sperimentali svolte in merito alla replica di superfici con Riblets 2D. Di queste ultime, sono state stimate le prestazioni per sezioni triangolari, smerlate, a lama e, per ciascuna di esse, è stata fornita la rispettiva ottimizzazione dei parametri caratteristici della geometria. Inoltre, è stato presentato uno studio sperimentale, in cui sono state impiegate Riblets 3D a lama trapezoidale, mostrando le criticità rispetto alle Riblets 2D. Successivamente, è stata fornita un’analisi che ha evidenziato il ruolo dell’orientazione delle superfici Riblet e delle nervature tridimensionali disposte sopra di esse, rispetto al flusso; in quanto è stato supposto che, nella reale pelle di squalo, ciò avrebbe potuto compensare la perdita di prestazioni rispetto al caso 2D. Infine, sono state discusse le tecniche di fabbricazione delle superfici Riblet tramite micro-goffratura e micro-stampaggio, considerando qulle più efficaci per la produzione di superfici ad alte prestazioni, commercializzabili in ambito industriale. In particolare, in questa trattazione è stata discussa la produzione di pellicole di rivestimento per la superficie esterna dei velivoli, stimando la riduzione della resistenza su diversi profili alari in differenti regimi di moto e sono state confrontate le prestazioni delle riblets su tre diversi tessuti impiegati per il nuoto competitivo.

PURPOSE: This study evaluated the quality of DNA obtained from stored human saliva and its applicability to human identification. METHODS: The saliva samples of 20 subjects, collected in the form of saliva in natura and from mouth swabs and stored at -20ºC, were analyzed. After 7 days, the DNA was extracted from the 40 saliva samples and subjected to PCR and electrophoresis. After 180 days, the technique was repeated with the 20 swab samples. RESULTS: The first-stage results indicated that DNA was successfully extracted in 97.5% of reactions, 95% of saliva in natura and 100% of swab saliva samples, with no statistically significant difference between the forms of saliva. In the second phase, the result was positive for all 20 analyzed samples (100%). Subsequently, in order to analyze the quality of the DNA obtained from human saliva, the SIX3-2 gene was tested on the 20 mouth swab samples, and the PCR products were digested using the MbO1 restriction enzyme to evaluate polymorphisms in the ADRA-2 gene, with positive results for most samples. CONCLUSION: It was concluded that the quantity and quality of DNA from saliva and the techniques employed are adequate for forensic analysis of DNA.

The objective of this study was to evaluate the use of fat sources in rations for lactating cows on the productive performance and composition of milk protein fraction. Twelve Holstein cows were used, grouped in three balanced 4 × 4 Latin squares, fed with the following rations: control; refined soybean oil; whole raw soybean; and calcium salts of unsaturated fatty acid (Megalac-E). Dry matter and nutrient intake, and daily milk production were evaluated. The samples used to analyze milk composition were collected in two alternate days and were obtained from two daily milking. Milk composition and total nitrogen, non-protein nitrogen and non-casein nitrogen ratios were analyzed. The casein, serum protein and true protein ratios were obtained by difference. Dry matter and nutrient intakes were lower when cows received the diet containing calcium salts of fatty acids, in relation to the control diet. Among the diets with fat sources, the one with whole raw soybean and calcium salts decreased milk production. There was no effect of fat sources added to the diet on crude protein, non-protein nitrogen, non-casein nitrogen, true protein, casein, casein/milk true protein ratio and serum protein. Similarly, the experimental diets did not influence the protein fractions when expressed in percentage of milk crude protein. The utilization of fat sources in diets changes milk production and composition of lactating cows, but does not influence the composition of milk protein fractions.

Aim of the study: Anacardium occidentale Linn. (cashew) is a Brazilian plant that is usually consumed in natura and is used in folk medicine. Anacardic acids (AAs) in the cashew nut shell liquid are biologically active as gastroprotectors, inhibitors of the activity of various deleterious enzymes, antitumor agents and antioxidants. Yet, there are no reports of toxicity testing to guarantee their use in vivo models. Materials and methods: We evaluated AAs biosafety by measuring the acute, subacute and mutagenic effects of AAs administration in BALB/c mice. In acute tests, BALB/c mice received a single oral dose of 2000 mg/kg, whereas animals in subacute tests received 300, 600 and 1000 mg/kg for 30 days. Hematological, biochemical and histological analyses were performed in all animals. Mutagenicity was measured with the acute micronucleus test 24 h after oral administration of 250 mg/kg AAs. Results: Our results showed that the AAs acute minimum lethal dose in BALB/c mice is higher than 2000 mg/kg since this concentration did not produce any symptoms. In subacute tests, females which received the highest doses (600 or 1000 mg/kg) were more susceptible, which was seen by slightly decreased hematocrit and hemoglobin levels coupled with a moderate increase in urea. Anacardic acids did not produce any mutagenic effects. Conclusions: The data indicate that doses less than 300 mg/kg did not produce biochemical and hematological alterations in BALB/c mice. Additional studies must be conducted to investigate the pharmacological potential of this natural substance in order to ensure their safe use in vivo. (C) 2011 Elsevier Ireland Ltd. This work aimed to study the in vitro colonic fermentation profile of unavailable carbohydrates of two different kinds of unripe banana flour and to evaluate their postprandial glycemic responses. The unripe banana mass (UBM), obtained from the cooked pulp of unripe bananas (Musa acuminata, Nanico variety), and the unripe banana starch (UBS), obtained from isolated starch of unripe banana, plantain type (Musa paradisiaca) in natura, were studied. The fermentability of the flours was evaluated by different parameters, using rat inoculum, as well as the glycemic response produced after the ingestion by healthy volunteers. The flours presented high concentration of unavailable carbohydrates, which varied in the content of resistant starch, dietary fiber and indigestible fraction (IF). The in vitro colonic fermentation of the flours was high, 98% for the UBS and 75% for the UBM when expressed by the total amount of SCFA such as acetate, butyrate and propionate in relation to lactulose. The increase in the area under the glycemic curve after ingestion of the flours was 90% lower for the UBS and 40% lower for the UBM than the increase produced after bread intake. These characteristics highlight the potential of UBM and UBS as functional ingredients. However, in vivo studies are necessary in order to evaluate the possible benefic effects of the fermentation on intestinal health.
