Only a small fraction of contemporary nanoscience will ever emerge as a functioning and commercial nanotechnology. Far too little attention is paid to the hoops and hurdles presented by the transition from science to technology by those who assert that their work will find applications. 从新几内亚核桃木的树皮中分离得到的吲哚类喹诺里西定生物碱10-Desbromoarborescidine A,因发现其具有阻滞钙离子通道的活性而倍受关注。10-Desbromoarborescidine A由A、B、C、D四个环组成,只有一个手性中心,是吲哚生物碱中结构较简单的一种,常作为此类生物碱全合成方法的模型化合物。但迄今为止,能高效而简便的实现手性10-Desbromoarborescidine A不对称全合成方法线路不多,大多数以不对称诱导的方式建立其手性中心,手性催化的方式仅有一例金属催化。从逆合成分析可知,Desbromoarborescidine A的全合成可以通过亚胺不对称催化还原进行关键的手性中心构建,而本课题组在之前的研究中通过手性有机小分子催化剂的发展,已将三氯硅烷氢转移还原亚胺发展成了一类简便实用、高效、高对映选择性并具有优良底物适应范围的不对称催化反应,我们希望以这一反应作为关键手段,发展一条Desbromoarborescidine A及其类似物不对称合成新路线。 根据我们设计的新路线,首先成功合成了其关键中间体,然后我们进行了关键的不对称催化尝试。用本实验室已有的高性能有机小分子催化剂虽得到了较好的对应选择性,但是产率很低。同时,为了验证整条线路的可行性,我们也用消旋的中间体进行拉通线路的尝试。但不幸的是,在脱除保护基时遇到了很大困难。尝试换不同的保护基,或改变脱保护基的顺序,都未能成功合成目标产物。究其原因可能是由于吲哚的特殊性造成的,吲哚类亚胺与常规的芳香亚胺有较大的差异,其NH基团无论保护还是不保护,对与其2位相联接的C=N双键均有很大的影响,导致其不对称催化还原难以进行。另外,由于所设计的还原产物含有处在吲哚苄位的胺基,稳定性较差,造成保护基脱除困难。 烯胺C-亚磺酰化反应是本课题组最近发现的一个新反应,之前未见文献报道。本研究对该反应进行了反应条件优化和底物扩展,发现带Cbz,Ac,COt-Bu,CO2Et,Bz等保护基的一系列环状和非环状烯胺在亚磺酸钠、DMAc和MeSiCl3的共同作用下能高效高产率生成β-胺基烯基亚砜类新化合物,为合成多官能团化的烯基亚砜新化合物提供了一条简便实用的途径。 The main constituent of Dracontomelum mangiferum B1, indoloquinolizidine alkaloid 10-Desbromoarborescidine A, has drawn great attention due to its calcium channel blocking activity. Its molecular structure is relatively simple compared with the other alkaloids of the same type, which has only one chiral center, albeit with four cycles A, B, C, and D. This compound is often used as a model target for exploring different strategies for the total synthesis of indole alkaloids. Nevertheless, so far there still lack practical and highly efficient methods for the asymmetric total synthesis of 10-Desbromoarborescidine A. Most of the current available methods rely on stoichiometric asymmetric synthesis for the construction of the chiral center. There is only one example reporting utilization of asymmetric catalysis, but with transition metal complex as the catalyst. Our retrosynthetic analysis shows that catalytic asymmetric reduction of imine could be used as the key step for the construction of the chiral center of Desbromoarborescidine A. Since in the previous studies our group has developed the asymmetric reduction of imines by trichlorosilane into a practical and highly efficient and enantioselective method using newly designed chiral organocatalysts, we hope to apply this method to develop a novel synthetic route for the total synthesis of Desbromoarborescidine A and its analogues in this study. According to the newly designed synthetic route, we first accomplished the synthesis of the key intermediates which was then examined for the critical asymmetric catalysis. The asymmetric reduction using the highly efficient organocatalysts, developed in our lab afforded high ee but poor yield. We tried different reaction conditions to improve the yield, but failed to get any good results. Simultaneously, to vertify the feasibility of the synthetic route we designed, we also tired to go through the route toward the racemic synthesis of Desbromoarborescidine A. But unfortunately, protection and deprotection proved to be big hurdles. All the different protection groups and different sequences of protection and deprotection we tried failed to get us through the designed synthetic sequence and furnish the final product. Most likely, the indole part is the culprit behind the failures.The NH group of the indole, no matter protected or not, may impact the catalytic asymmetric reduction of C-N double bond connected with 2-C. Additionally, the reduction product we designed contains an amino group in the β-position of the indole, which may cause problems due to its instability. C-sulfenylation of enamines is a novel reaction discovered recently by our group, which has not been seen before in the literature. In this study, optimization of the reaction conditions and exploration of the substrate scope were further undertaken for this reaction, which reveal that a series of enamines with N-Cbz, Ac, COt-Bu, CO2Et protection groups could all undergo smooth C-sulfinylations with the comined use of sodium benzene sulphinate, DAMc and MeSiCl3, efficiently furnishing the β-amino vinylsulfoxide products in high yield, affording a practical and highly efficient methods for synthesis of functional vinylsulfoxides.

The development of a new bioprocess requires several steps from initial concept to a practical and feasible application. Industrial applications of fungal pigments will depend on: (i) safety of consumption, (ii) stability of the pigments to the food processing conditions required by the products where they will be incorporated and (iii) high production yields so that production costs are reasonable. Of these requirements the first involves the highest research costs and the practical application of this type of processes may face several hurdles until final regulatory approval as a new food ingredient. Therefore, before going through expensive research to have them accepted as new products, the process potential should be assessed early on, and this brings forward pigment stability studies and process optimisation goals. Only ingredients that are usable in economically feasible conditions should progress to regulatory approval. This thesis covers these two aspects, stability and process optimisation, for a potential new ingredient; natural red colour, produced by microbial fermentation. The main goal was to design, optimise and scale-up the production process of red pigments by Penicillium purpurogenum GH2. The approach followed to reach this objective was first to establish that pigments produced by Penicillium purpurogenum GH2 are sufficiently stable under different processing conditions (thermal and non-thermal) that can be found in food and textile industries. Once defined that pigments were stable enough, the work progressed towards process optimisation, aiming for the highest productivity using submerged fermentation as production culture. Optimum production conditions defined at flask scale were used to scale up the pigment production process to a pilot reactor scale. Finally, the potential applications of the pigments were assessed. Based on this sequence of specific targets, the thesis was structured in six parts, containing a total of nine chapters. Engineering design of a bioprocess for the production of natural red colourants by submerged fermentation of the thermophilic fungus Penicillium purpurogenum GH2.

Composers from all eras and of all ethnicities explore spirituality and prayer by using one or a combination of the following ideas: having a spiritual concept in mind when composing certain pieces, quoting hymns, being influenced by their own personal beliefs, or portraying spiritual figures and ideas in their works. Some musical works are inspired by spirituality; others, as in the case of Bloch's Nigun, even serve as prayers themselves. These recitals gave me the opportunity to approach a wide variety of musical styles while discovering my own mode for expression. The unaccompanied violin works throughout this project trace a distinct lineage from the baroque to the twentieth century. Biber's appendix to the Rosary Sonatas, the Passacaglia for solo violin, is a crucial predecessor to Bach's monumental Chaconne. Eugene Ysaye was inspired to write the Six Sonatas, Op. 27 after he attended a performance of Bach's Sonatas and Partitas given by Josef Szigeti. Ysaye's second solo sonata blatantly quotes Bach's Partita No.3 in E major throughout the first movement. Every movement also contains quotations from and variations on the plainchant Dies Irae. Although each of the solo violin works presented in this project may be viewed as virtuosic concert pieces, each piece allows the performer to transcend the technical hurdles-and perhaps even utilize them-to serve a higher, artistic and spiritual purpose while alone on the concert stage. Each of the sonata works in this project requires a close, equal collaboration between violinist and pianist, rather than displaying the violinist as soloist.

A clinical trial using human embryonic stem cell (hESC) therapy for an inherited retinal degenerative disease is about to commence. The Advanced Cell Technology (ACT) trial will treat patients with Stargardt's macular dystrophy using transplanted retinal pigment epithelium derived from hESCs. Currently, no effective treatment is available for Stargardt's disease so a stem cell-based therapy that can slow progression of this blinding condition could represent a significant breakthrough. While there are some hurdles to clear, the ACT trial is a fine example of translational research that could eventually pave the way for a range of stem cell therapies for the retina and other tissues.

We examine the impact of the Great Depression on the share of votes for right-wing extremists in elections in the 1920s and 1930s. We confirm the existence of a link between political extremism and economic hard times as captured by growth or contraction of the economy. What mattered was not simply growth at the time of the election, but cumulative growth performance. The impact was greatest in countries with relatively short histories of democracy, with electoral systems that created low hurdles to parliamentary representation, and which had been on the losing side in World War I.

The ever-growing energy consumption in mobile networks stimulated by the expected growth in data tra ffic has provided the impetus for mobile operators to refocus network design, planning and deployment towards reducing the cost per bit, whilst at the same time providing a signifi cant step towards reducing their operational expenditure. As a step towards incorporating cost-eff ective mobile system, 3GPP LTE-Advanced has adopted the coordinated multi-point (CoMP) transmission technique due to its ability to mitigate and manage inter-cell interference (ICI). Using CoMP the cell average and cell edge throughput are boosted. However, there is room for reducing energy consumption further by exploiting the inherent exibility of dynamic resource allocation protocols. To this end packet scheduler plays the central role in determining the overall performance of the 3GPP longterm evolution (LTE) based on packet-switching operation and provide a potential research playground for optimizing energy consumption in future networks. In this thesis we investigate the baseline performance for down link CoMP using traditional scheduling approaches, and subsequently go beyond and propose novel energy e fficient scheduling (EES) strategies that can achieve power-e fficient transmission to the UEs whilst enabling both system energy effi ciency gain and fairness improvement. However, ICI can still be prominent when multiple nodes use common resources with di fferent power levels inside the cell, as in the so called heterogeneous networks (Het- Net) environment. HetNets are comprised of two or more tiers of cells. The rst, or higher tier, is a traditional deployment of cell sites, often referred to in this context as macrocells. The lower tiers are termed small cells, and can appear as microcell, picocells or femtocells. The HetNet has attracted signiffi cant interest by key manufacturers as one of the enablers for high speed data at low cost. Research until now has revealed several key hurdles that must be overcome before HetNets can achieve their full potential: bottlenecks in the backhaul must be alleviated, as well as their seamless interworking with CoMP. In this thesis we explore exactly the latter hurdle, and present innovative ideas on advancing CoMP to work in synergy with HetNet deployment, complemented by a novel resource allocation policy for HetNet tighter interference management. As system level simulator has been used to analyze the proposed algorithm/protocols, and results have concluded that up to 20% energy gain can be observed.

This research thesis analyses the motivation behind the cross-border mergers and acquisitions deals. How mergers and acquisitions of new knowledge and assets, enhance business with expansion into new streams and international markets. Also, how mega deals help them to gain a power in the international markets. The research focuses on understanding the interrelation between motivations which are contributing to M&A activities and how issues like cultural differences and different management styles are overcome by these firms in cross-border settings. Chapter 1, gives a background knowledge on cross-border M&A as popular internationalization strategy choice, continuing with describing the process in Finnish and Japanese cultural context, and how these deals are proceeding in particular cases. Chapter 2, reviews the important findings and touches the common gaps or aspects those are not studied extensively, does play a key role in the success and failure of M&A deals. A methodology is presented in chapter 3, presenting the hurdles faced by many in this research field. Chapter 4, present the case study is presented to show how M&A can play an important role in structuring the entire economy of Japan. At last chapter 5, presents the evidence, if cultural, HRM and geographical aspects really contribute to the success of M&A, based on which managerial implications are suggested and propositions are built for future research references.

Les oligonucléotides (ONs) antisens présentent un fort potentiel en tant qu’agents thérapeutiques. Toutefois, leurs propriétés physicochimiques limitent leur utilisation en thérapie génique. Pour pallier aux divers obstacles, des systèmes de vectorisation, tels que les micelles polyioniques (PICMs), ont été développés. Grâce à leur structure unique, les micelles protégent l’ON contre une dégradation prématurée et le couplage d’un ligand à leur surface augmente leur spécificité et leur internalisation. Dans d’autres systèmes, un polymère adjuvant aux propriétés pH-sensibles peut être ajouté pour faciliter la sortie de l’endosome et augmenter l’efficacité de l’ON. L’objectif général de ce mémoire était de mettre au point des PICMs ternaires ciblées pour l’administration d’ONs. Ces micelles assureraient à la fois l’internalisation cellulaire de leur cargaison en interagissant avec des récepteurs cellulaires et sa fuite de l’endosome grâce à un mécanisme de déstabilisation de la membrane endosomale. Pour cela, des PICMs composées d’un copolymère cationique de type poly(éthylène glycol)-bloc-poly(méthacrylate d’(alkylamino)éthyle) et d’un copolymère d’acide méthacrylique ont été préparées. Les propriétés physicochimiques de ces vecteurs ont démontré qu’ils permettaient une condensation efficace de l’acide nucléique et ce, indépendamment de la nature du polymère cationique et de l’acide nucléique. Finalement, une approche de couplage par pont disulfure a été développée afin de greffer au copolymère un fragment d’anticorps dirigé contre les récepteurs de la transferrine. En conclusion, ces travaux démontrent la versatilité et le potentiel des PICMs ternaires en tant que vecteurs d’acide nucléique, et proposent une méthodologie de couplage d’un ligand afin de formuler des PICMs ciblées.

This research project is a contribution to the global field of information retrieval, specifically, to develop tools to enable information access in digital documents. We recognize the need to provide the user with flexible access to the contents of large, potentially complex digital documents, with means other than a search function or a handful of metadata elements. The goal is to produce a text browsing tool offering a maximum of information based on a fairly superficial linguistic analysis. We are concerned with a type of extensive single-document indexing, and not indexing by a set of keywords (see Klement, 2002, for a clear distinction between the two). The desired browsing tool would not only give at a glance the main topics discussed in the document, but would also present relationships between these topics. It would also give direct access to the text (via hypertext links to specific passages). The present paper, after reviewing previous research on this and similar topics, discusses the methodology and the main characteristics of a prototype we have devised. Experimental results are presented, as well as an analysis of remaining hurdles and potential applications.

Des lois sont adoptées, des politiques implantées, des centaines de milliers de dollars investis dans la formation des employés, et puis ? Cette recherche de type exploratoire tente de jeter la lumière sur ce qui se passe actuellement dans les points d'entrée canadiens situés au Québec en matière de lutte contre la traite de personnes migrantes et de présenter, s'il y a lieu, les incohérences entre l'engagement du Canada à lutter contre la traite de personnes et les pratiques des agents des services frontaliers du Canada sur le terrain. Pour ce faire, nous avons interviewé sept agents des services frontaliers du Canada travaillant à l'Aéroport international Pierre-Elliott-Trudeau de Montréal, quatrième poste frontalier aéroportuaire en importance au Canada et premier au Québec, afin d'en apprendre davantage sur les connaissances ainsi que sur les expériences de ceux-ci en matière de traite de personnes migrantes. Nous avons aussi rencontré quatre intervenants venant en aide aux victimes migrantes de la traite de personnes au Québec, principalement dans la région de Montréal, dans le but d'en savoir un peu plus sur l'expérience des victimes en lien avec l'Agence des services frontaliers du Canada et les impacts de ces expériences sur celles-ci. Si les agents interviewés semblent avoir une assez bonne idée, même si celle-ci paraît largement influencée par les médias, de ce qu'est la traite de personnes migrantes, rares sont ceux qui, dans le cadre de leurs fonctions, ont une expérience pratique auprès des victimes. L'absence de formation substantielle et continue, la perception du rôle de l'ASFC par ses agents ainsi que le manque de leadership de la direction en ce qui a trait à la traite de personnes migrantes semblent être des accrocs majeurs à la volonté politique du gouvernement canadien, plus particulièrement de l'Agence des services frontaliers du Canada, de combattre la traite de personnes migrantes sur son propre territoire. Les obstacles sont nombreux dans la lutte contre la traite de personnes aux frontières canadiennes et représentent un défi de taille à la fois complexe et compliqué. Une approche centrée sur la prévention et une plus grande sensibilisation des agents des services frontaliers du Canada proposent un début de solution.

In a global context of climate change and energy transition, Quebec seems to be privileged, producing a large amount of cheap hydroelectricity. But aside from the established popular belief that Quebec’s energy is abundant, clean and inexpensive, Quebec’s energy future is still precarious. Within a few decades, Quebec will have to import a significant amount of electricity at a higher price than it actually produces it; the cheap exploitable hydro resources will not only get scarcer if not nonexistent; and the national hydroelectric ``cultural`` heritage even seems to quell the development of alternative energies, letting few space for local innovation coming from municipalities. While in many countries, municipalities are recognised as key figures in the energy sector, here, in Quebec, their role in the national energy system seems marginal. As main actors responsible for territorial planning, it seams that municipalities could play a more important role on Quebec’s energy scene. So they can densify their territory, develop active and collective solutions to transportation issues, they can adopt exemplary energetic habits, they can produce their own energy with wind, solar or even district heating systems. District heating and heat networks being less well know and documented in Quebec, the present study aims at explaining their low penetration level in the Quebec energy landscape. The study also attempts to understand what are the main hurdles to the implementation of district heating in Quebec’s particular energetic context. Finally, the research tries to open a discussion on the motives that could incite municipalities to adopt district heating as an energy alternative. Based on some twenty interviews with key actors of the energy and municipal sectors, the findings give some indications that the low penetration level of district heating in the Quebec municipalities could explain itself in part by : the low priced hydroelectricity, the presence of a comfortable, sufficient and pervasive Hydro-Quebec(er) culture, and also by organizational dynamic and a certain political inertia which limit the appropriation of an energy competence by local governments. In turn, the study shows that district heating solutions are more likely to develop in contexts in which : there are minimum urban or energy density levels; the development of district heating coincides with the local or regional economic structure; and where exist a mobilising local leader or local visions from a community in favor of the implementation of alternative energy systems.
