A novel RGD-independent cel adhesion pathway mediated by fibronectin-bound tissue transglutaminase rescues cells from anoikis

Specific association of tissue transglutaminase (tTG) with matrix fibronectin (FN) results in the formation of an extracellular complex (tTG-FN) with distinct adhesive and pro-survival characteristics. tTG-FN supports RGD-independent cell adhesion of different cell types and the formation of distinctive RhoA-dependent focal adhesions following inhibition of integrin function by competitive RGD peptides and function blocking anti-integrin antibodies alpha5beta1. Association of tTG with its binding site on the 70-kDa amino-terminal FN fragment does not support this cell adhesion process, which seems to involve the entire FN molecule. RGD-independent cell adhesion to tTG-FN does not require transamidating activity, is mediated by the binding of tTG to cell-surface heparan sulfate chains, is dependent on the function of protein kinase Calpha, and leads to activation of the cell survival focal adhesion kinase. The tTG-FN complex can maintain cell viability of tTG-null mouse dermal fibroblasts when apoptosis is induced by inhibition of RGD-dependent adhesion (anoikis), suggesting an extracellular survival role for tTG. We propose a novel RGD-independent cell adhesion mechanism that promotes cell survival when the anti-apoptotic role mediated by RGD-dependent integrin function is reduced as in tissue injury, which is consistent with the externalization and binding of tTG to fibronectin following cell damage/stress.

Shifting borders of European Union internal security:technology, externalisation and accountability

This edited volume analyzes recent key developments in EU border management. In light of the refugee crises in the Mediterranean and the responses on the part of EU member states, this volume presents an in-depth reflection on European border practices and their political, social and economic consequences. Approaching borders as concepts in flux, the authors identify three main trends: the rise of security technologies such as the EUROSUR system, the continued externalization of EU security governance such as border mission training in third states, and the unfolding dynamics of accountability. The contributions show that internal security cooperation in Europe is far from consolidated, since both political oversight mechanisms and the definition of borders remain in flux. This edited volume makes a timely and interdisciplinary contribution to the ongoing academic and political debate on the future of open borders and legitimate security governance in Europe. Correlação entre imagem corporal e autoestima em pessoas com ostomias intestinais

Ostomized is every individual that, because of a traumatic or clinical condition, it required a surgery resulting in the externalization of a hollow organ through the skin, and such temporary or permanent condition. This study has the general objective to investigate the relationship between body image and self-esteem in these individuals; as well as to verify their levels of satisfaction with their body image regarding the aspect of appearance and to evaluate the degree of self-esteem related to that condition. This is a correlational research, cross-sectional, which was accomplished in an institution in support of these users of Rio Grande do Norte state, with location in Natal. There were used three sampling instruments: a structured general questionnaire covering socio-demographic and clinical data; the Satisfaction Scale with Appearance (SSA) and the Rosenberg Self-Esteem Scale (RSS). The information obtained was analyzed with the aid of an electronic spreadsheet software. The project is according with the resolution 466/12 of the National Health Council, It was approved by the Research Ethics Committee of UFRN, under number CAAE 19159713.5.0000.5537 in August 2013. The sample consisted of 93 participants with an average age of 50.4 years (SD = 15.4). In general they had low satisfaction with their body image (M = 66.9), as regards the appearance, although maintained high levels of self-esteem (M = 34.8). Therefore, It was found a positive correlation, moderate (ρ = 0.426) and statistically significant was found (p <0.001) from the application of the Spearman correlation test. Therefore, dissatisfaction with body image is an important issue to be observed by professionals who attend ostomizeds. however, it seems, other aspects are also influencing the level of self-esteem of these individuals, who were not able to be determined in this study.

Induction of apoptosis by UV in the flat oyster, Ostrea edulis

Apoptosis is a fundamental feature in the development of many organisms and tissue systems. It is also a mechanism of host defense against environmental stress factors or pathogens by contributing to the elimination of infected cells. Hemocytes play a key role in defense mechanisms in invertebrates and previous studies have shown that physical or chemical stress can increase apoptosis in hemocytes in mollusks. However this phenomenon has rarely been investigated in bivalves especially in the flat oyster Ostrea edulis. The apoptotic response of hemocytes from flat oysters, O. edulis, was investigated after exposure to UV and dexamethasone, two agents known to induce apoptosis in vertebrates. Flow cytometry and microscopy were combined to demonstrate that apoptosis occurs in flat oyster hemocytes. Investigated parameters like intracytoplasmic calcium activity, mitochondrial membrane potential and phosphatidyl-serine externalization were significantly modulated in cells exposed to UV whereas dexamethasone only induced an increase of DNA fragmentation. Morphological changes were also observed on UV-treated cells using fluorescence microscopy and transmission electron microscopy. Our results confirm the apoptotic effect of UV on hemocytes of O. edulis and suggest that apoptosis is an important mechanism developed by the flat oyster against stress factors.
