Moderate wine intake (i.e., 1-2 glasses of wine a day) is associated with a reduced risk of morbidity and mortality from cardiovascular disease. The aim of this study was to evaluate the anti-atherosclerotic effects of a nonalcoholic ethyl acetate fraction (EAF) from a South Brazilian red wine obtained from Vitis labrusca grapes. Experiments were carried out on low-density lipoprotein (LDL) receptor knockout (LDLr-/-) mice, which were subjected to a hypercholesterolemic diet and treated with doses of EAF (3, 10, and 30 mg/kg) for 12 weeks. At the end of the treatment, the level of plasma lipids, the vascular reactivity, and the atherosclerotic lesions were evaluated. Our results demonstrated that the treatment with EAF at 3 mg/kg significantly decreased total cholesterol, triglycerides, and LDL plus very low-density lipoprotein levels compared with control hypercholesterolemic mice. The treatment of mice with EAF at 3 mg/kg also preserved the vasodilatation induced by acetylcholine on isolated thoracic aorta from hypercholesterolemic LDLr-/- mice. This result is in agreement with the degree of lipid deposit on arteries. Taken together, the results show for the first time that the lowest concentration of an EAF obtained from a red wine produced in southern Brazil significantly reduced the progression of atherosclerosis in mice.

A greenhouse experiment was installed with bait cultures to extract the AMF species present in a rhizosphere soil sample of a native Araucaria angustifolia forest in Campos do Jordao, Brazil. The experimental design was completely randomized, with four increasing phosphorus doses (0, 20, 50, and 150 mg kg(-1), as triple superphosphate), with five replicates, the bait plant was araucaria, and all pots were inoculated with 100 g of rhizospheric soil collected in an araucaria forest. After twelve months the spores were extracted, counted and identified, and the percent root colonization was also determined. When taking all four P doses into account, eleven AMF species could be identified: Acaulospora bireticulata, Acaulospora morrowiae, Acaulospora sp., Entrophospora colombiana, Gigaspora margarita, Glomus diaphanum, Glomus etunicatum, Glomus macrocarpum, Scutellospora calospora, Scutellospora gilmorei, and Scutellospora pellucida. There was no effect of the P dose on the total amount of spores neither on the percent root colonization. However, the correspondence analysis showed that the different AMF species were selectively associated mostly to either one or another P dose.

Araucaria angustifolia, commonly named Araucaria, is a Brazilian native species that is intensively exploited due to its timber quality. Therefore, Araucaria is on the list of species threatened by extinction. Despite the importance of soil for forest production, little is known about the soil properties of the highly fragmented Araucaria forests. This study was designed to investigate the use of chemical and biological properties as indicators of conservation and anthropogenic disturbance of Araucaria forests in different sampling periods. The research was carried out in two State parks of Sao Paulo: Parque Estadual Turistico do Alto do Ribeira and Parque Estadual de Campos de Jordao. The biochemical properties carbon and nitrogen in microbial biomass (MB-C and MB-N), basal respiration (BR), the metabolic quotient (qCO(2)) and the following enzyme activities: beta-glucosidase, urease, and fluorescein diacetate hydrolysis (FDA) were evaluated. The sampling period (dry or rainy season) influenced the results of mainly MB-C, MB-N, BR, and qCO(2). The chemical and biochemical properties, except K content, were sensitive indicators of differences in the conservation and anthropogenic disturbance stages of Araucaria forests. Although these forests differ in biochemical and chemical properties, they are efficient in energy use and conservation, which is shown by their low qCO(2), suggesting an advanced stage of succession.

Araucaria angustifolia, a unique species of this genus that occurs naturally in Brazil, has a high socio-economic and environmental value and is critically endangered of extinction, since it has been submitted to intense predatory exploitation during the last century. Root-associated bacteria from A. angustifolia were isolated, selected and characterized for their biotechnological potential of growth promotion and biocontrol of plant pathogenic fungi. Ninety-seven strains were isolated and subjected to chemical tests. All isolates presented at least one positive feature, characterizing them as potential PGPR. Eighteen isolates produced indole-3-acetic acid (IAA), 27 were able to solubilize inorganic phosphate, 21 isolates were presumable diazotrophs, with pellicle formation in nitrogen-free culture medium, 83 were phosphatases producers, 37 were positive for siderophores and 45 endospore-forming isolates were antagonistic to Fusarium oxysporum, a pathogen of conifers. We also observed the presence of bacterial strains with multiple beneficial mechanisms of action. Analyzing the fatty acid methyl ester (FAME) and partial sequencing of the 16S rRNA gene of these isolates, it was possible to characterize the most effective isolates as belonging to Bacillaceae (9 isolates), Enterobacteriaceae (11) and Pseudomonadaceae (1). The impact of tannery sludge application on soil microbial community and diversity is poorly understood. We studied the microbial community in an agricultural soil following two applications (2006 and 2007) of tannery sludge with annual application rates of 0.0,2.3 and 22.6 Mg ha(-1). The soil was sampled 12 and 271 days after the second (2007) application. Community structure was assessed via a phospholipid fatty acid analysis, and the physiological profile of the soil microbial community via the Biolog method. Tannery sludge application changed soil chemical properties, increasing the soil pH and electrical conductivity as well as available P and mineral N concentrations. The higher sludge application rate changed the community structure and the physiological profile of the microbial community at both sampling dates. However, there is no clear link between community structure and carbon substrate utilization. According to the Distance Based Linear Models Analysis, the fatty acids 16:0 and 117:0 together contributed 84% to the observed PLFA patterns, whereas the chemical properties available P, mineral N, and Ca, and pH together contributed 54%. At 12 days, tannery sludge application increased the average well color development from 0.46 to 0.87 after 48 h, and reduced the time elapsed before reaching the midpoint carbon substrate utilization (s) from 71 to 44 h, an effect still apparent nine months after application of the higher sludge application rate. Testosterone has been implicated in vascular remodeling associated with hypertension. Molecular mechanisms underlying this are elusive, but oxidative stress may be important. We hypothesized that testosterone stimulates generation of reactive oxygen species (ROS) and migration of vascular smooth muscle cells (VSMCs), with enhanced effects in cells from spontaneously hypertensive rats (SHRs). The mechanisms (genomic and nongenomic) whereby testosterone induces ROS generation and the role of c-Src, a regulator of redox-sensitive migration, were determined. VSMCs from male Wistar-Kyoto rats and SHRs were stimulated with testosterone (10(-7) mol/L, 0-120 minutes). Testosterone increased ROS generation, assessed by dihydroethidium fluorescence and lucigenin-enhanced chemiluminescence (30 minutes [SHR] and 60 minutes [both strains]). Flutamide (androgen receptor antagonist) and actinomycin D (gene transcription inhibitor) diminished ROS production (60 minutes). Testosterone increased Nox1 and Nox4 mRNA levels and p47phox protein expression, determined by real-time PCR and immunoblotting, respectively. Flutamide, actinomycin D, and cycloheximide (protein synthesis inhibitor) diminished testosterone effects on p47phox. c-Src phosphorylation was observed at 30 minutes (SHR) and 120 minutes (Wistar-Kyoto rat). Testosterone-induced ROS generation was repressed by 3-(4-chlorophenyl) 1-(1,1-dimethylethyl)-1H-pyrazolo[3,4-day]pyrimidin-4-amine (c-Src inhibitor) in SHRs and reduced by apocynin (antioxidant/NADPH oxidase inhibitor) in both strains. Testosterone stimulated VSMCs migration, assessed by the wound healing technique, with greater effects in SHRs. Flutamide, apocynin, and 3-(4-chlorophenyl) 1-(1,1-dimethylethyl)-1H-pyrazolo[3,4-day] pyrimidin-4-amine blocked testosterone-induced VSMCs migration in both strains. Our study demonstrates that testosterone induces VSMCs migration via NADPH oxidase-derived ROS and c-Src-dependent pathways by genomic and nongenomic mechanisms, which are differentially regulated in VSMCs from Wistar-Kyoto rats and SHRs. The use of sewage sludge is a highly promising practice for the development of sustainable agricultural systems. The objective of this study was to evaluate doses of sewage sludge composted with and without Rhizobium inoculation in leaf N content, nodule number, nodule dry weight and plant during flowering. The experiment was conducted in the greenhouse of the Department of Soil Science and Natural Resources College of Agricultural Sciences of Botucatu, using as substrate used in vessels of 30 liters a Red Yelow Latosol sandy texture with experimental design adopted was randomized blocks constituted for 10 treatments and five doses of composted sewage sludge (0, 10, 20, 30, 40 t ha(-1)) with or without inoculation Bradyrhizobium japonic with three replications. There was an increase in the number and dry weight of nodules and shoot dry mass of soybeans due to the increase of the dose of sludge up to a dose of 20 t ha(-1) and after this dose there was a decrease of these parameters. At a dose of 10 t ha(-1) sludge compost inoculated seeds showed higher for foliar concentrations of N and number of nodules compared with uninoculated seeds. At a dose of 30 t ha(-1) inoculated seeds were higher compared to uninoculated in all parameters.

PURPOSE: To evaluate the sulcus anatomy and possible correlations between sulcus diameter and white-to-white (WTW) diameter in pseudophakic eyes, data that may be important in the stability of add-on intraocular lenses (IOLs). SETTING: University Eye Hospital, Tuebingen, Germany. DESIGN: Case series. METHODS: In pseudophakic eyes, the axial length (AL) and horizontal WTW were measured by the IOLMaster device. Cross-sectional images were obtained with a 50 MHz ultrasound biomicroscope on the 4 meridians: vertical, horizontal (180 degrees), temporal oblique, and nasal oblique. Sulcus-to-sulcus (STS), angle-to-angle (ATA), and sclera-to-sclera (ScTSc) diameters were measured. The IOL optic diameter (6.0 mm) served as a control. To test reliability, optic measurements were repeated 5 times in a subset of eyes. RESULTS: The vertical ATA and STS diameters were statistically significantly larger than the horizontal diameter (P=.0328 and P=.0216, respectively). There was no statistically significant difference in ScTSc diameters. A weak correlation was found between WTW and horizontal ATA (r = 0.5766, P<.0001) and between WTW and horizontal STS (r = 0.5040, P=.0002). No correlation was found between WTW and horizontal ScTSc (r = 0.2217, P=.1217). CONCLUSIONS: The sulcus anatomy had a vertical oval shape with the vertical meridian being the largest, but it also had variation in the direction of the largest meridian. The WTW measurements showed a weak correlation with STS. In pseudophakic eyes, Soemmerring ring or a bulky haptic may affect the ciliary sulcus anatomy.

The liver was among the first organs in which connexin proteins have been identified. Hepatocytes harbor connexin32 and connexin26, while non-parenchymal liver cells typically express connexin43. Connexins give rise to hemichannels, which dock with counterparts on adjacent cells to form gap junctions. Both hemichannels and gap junctions provide pathways for communication, via paracrine signaling or direct intercellular coupling, respectively. Over the years, hepatocellular gap junctions have been shown to regulate a number of liver-specific functions and to drive liver cell growth. In the last few years, it has become clear that connexin hemichannels are involved in liver cell death, particularly in hepatocyte apoptosis. This also holds true for hemichannels composed of pannexin1, a connexin-like protein recently identified in the liver. Moreover, pannexin1 hemichannels are key players in the regulation of hepatic inflammatory processes. The current paper provides a concise overview of the features of connexins, pannexins and their channels in the liver.

Seit Beginn der Waldzustandserhebungen im Jahr 1984 verschlechterte sich der Zustand der Eiche sowohl auf Bundesebene als auch im Land Rheinland-Pfalz deutlich. 1998 konnten nur noch 5 % der rheinland-pfälzischen Eichen in die Kategorie "ohne Schadensmerkmale" eingestuft werden. Vor diesem Hintergrund ergab sich die Notwendigkeit, die biotischen Stress- und Schadfaktoren näher zu untersuchen. Im Rahmen der vorliegenden Arbeit lag der Fokus auf den holzbewohnenden Käfern von Traubeneichen (Quercus petraea) aus dem Pfälzerwald. Zu diesem Zweck wurden für die erste Probenserie Untersuchungsbäume aus den Vitalitätsstufen 'vital', 'geschädigt', 'ein Jahr tot' und 'zwei Jahre tot' ausgewählt und in die drei Straten Stammfuß, Kronenansatz und Derbholz unterteilt. In der zweiten Probenserie kamen keine 'vitalen' Stämme mehr zum Einsatz. Die einzelnen Proben wurden in Fasseklektoren überführt, in denen die xylobionten Käfer ihre Entwicklung beenden und schlüpfen konnten. Die erste Probenserie wurde im Herbst 1998 entnommen, die zweite Serie im darauffolgenden Herbst. Zusätzlich zu diesen Laboruntersuchungen wurden Freilanduntersuchungen mit Stammeklektoren an vier stehenden Eichen im Wald durchgeführt. Die gefangenen Tiere wurden nach Ordnungen sortiert und gezählt, die Käfer nach Möglichkeit bis zur Art bestimmt. Die Ergebnisse der ersten und zweiten Serie wurden in Abundanzen (Ind./m² Rindenoberfläche) umgerechnet, um einen Vergleich der Proben untereinander möglich zu machen. Insgesamt wurden aus den Fasseklektoren beider Serien Käfer mit einer Abundanz von 36.990 Ind./m² ausgewertet. In den Fallen der Stammeklektoren wurden insgesamt 1.487 Käfer gefunden. Den weitaus größten Teil der Käfer der Fasseklektoren stellen die Borkenkäfer (Scolytidae). Dieses Ergebnis schlägt sich auch in der Betrachtung der Dominanz der einzelnen Arten nieder. In nahezu allen Fällen gehörten die Hauptarten in die Familie Scolytidae. Der mit den Absterbeerscheinungen der Eichen in Verbindung gebrachte Prachtkäfer Agrilus biguttatus (Buprestidae) trat in deutlich geringeren Abundanzen auf. Aufgrund seiner Fraßtätigkeit (Ringelung der Larven im Kambialbereich der Bäume) gehört er aber zu den potentiell stark schädigenden Käfern. Neben A. biguttatus sind auch A. sulcicollis und die gefundenen Borkenkäfer in der Lage, vorgeschädigte und geschwächte Eichen zu befallen und noch weiter zu schwächen. Aus waldhygienischen Gründen sollten deshalb regelmäßige Kontrollen durchgeführt werden. Bei erkennbarem Befall sollten die betroffenen Bäume gefällt und aus dem Bestand entfernt werden. Langfristig können die Vermeidung von nicht-standortgerechtem Eichenanbau und das Anlegen von naturnahen Mischbeständen zu den waldbaulichen Maßnahmen gerechnet werden, die eine Reduktion des Infektionsrisikos zur Folge haben. Die 'ein Jahr toten' Bäume wiesen die mit Abstand höchste Abundanz an Lebendholzbesiedlern auf. Bäume, die bereits ein Jahr länger tot im Bestand standen, wurden von deutlich weniger Lebendholzbesiedlern befallen, d.h. von 'zwei Jahre toten' Bäumen ging ein potentiell geringerer Infektionsdruck aus als von 'ein Jahr toten' Eichen. Im Laufe des Verrottungsprozesses verringert sich diese Gefahr noch weiter, da die Holzfeuchte weiter abnimmt und die Lebendholzbesiedler keine Nahrungsgrundlage mehr vorfinden. Besonders die Gefahr des Neubefalls durch Agrilus von mindestens zweijährig toten Bäumen besteht kaum, weil zumindest die Larven der ersten Stadien des Prachtkäfers auf lebendes Gewebe angewiesen sind.

One of the quickest plant movements ever known is made by the ´explosive´ style in Marantaceae in the service of secondary pollen presentation – herewith showing a striking apomorphy to the sister Cannaceae that might be of high evolutionary consequence. Though known already since the beginning of the 19th century the underlying mechanism of the movement has hitherto not been clarified. The present study reports about the biomechanics of the style-staminode complex and the hydraulic principles of the movement. For the first time it is shown by experiment that in Maranta noctiflora through longitudinal growth of the maturing style in the ´straitjacket´ of the hooded staminode both the hold of the style prior to its release and its tensioning for the movement are brought about. The longer the style grows in relation to the enclosing hooded staminode the more does its capacity for curling up for pollen transfer increase. Hereby I distinguish between the ´basic tension´ that a growing style builds up anyway, even when the hooded staminode is removed beforehand, and the ´induced tension´ which comes about only under the pressure of a ´too short´ hooded staminode and which enables the movement. The results of these investigations are discussed in view of previous interpretations ranging from possible biomechanical to electrophysiological mechanisms. To understand furthermore by which means the style gives way to the strong bending movement without suffering outwardly visible damage I examined its anatomical structure in several genera for its mechanical and hydraulic properties and for the determination of the entire curvature after release. The actual bending part contains tubulate cells whose walls are extraordinarily porous and large longitudinal intercellular spaces. SEM indicates the starting points of cell-wall loosening in primary walls and lysis of middle lamellae - probably through an intense pectinase activity in the maturing style. Fluorescence pictures of macerated and living style-tissue confirm cell-wall perforations that do apparently connect neighbouring cells, which leads to an extremely permeable parenchyma. The ´water-body´ can be shifted from central to dorsal cell layers to support the bending. The geometrical form of the curvature is determined by the vascular bundles. I conclude that the style in Marantaceae contains no ´antagonistic´ motile tissues as in Mimosa or Dionaea. Instead, through self-maceration it develops to a ´hydraulic tissue´ which carries out an irreversible movement through a sudden reshaping. To ascertain the evolutionary consequence of this apomorphic pollination mechanism the diversity and systematic value of hooded staminodes are examined. For this hooded staminodes of 24 genera are sorted according to a minimalistic selection of shape characters and eight morphological types are abstracted from the resulting groups. These types are mapped onto an already available maximally parsimonious tree comprising five major clades. An amazing correspondence is found between the morphological types and the clades; several sister-relationships are confirmed and in cases of uncertain position possible evolutionary pathways, such as convergence, dispersal or re-migration, are discussed, as well as the great evolutionary tendencies for the entire family in which – at least as regards the shape of hooded staminodes – there is obviously a tendency from complicated to strongly simplified forms. It suggests itself that such simplifying derivations may very likely have taken place as adaptations to pollinating animals about which at present too little is known. The value of morphological characters in relation to modern phylogenetic analysis is discussed and conditions for the selection of morphological characters valuable for a systematic grouping are proposed. Altogether, in view of the evolutionary success of Marantaceae compared with Cannaceae the movement mechanism of the style-staminode complex can safely be considered a key innovation within the order Zingiberales.

Die schlechte Prognose des Nierenzellkarzinoms (NZK) kommt nicht durch den Primärtumor an sich zustande, sondern durch das Vorhandensein von Fernmetastasen. Obwohl bereits vieles über die Mechanismen der Metastasierung bekannt ist, sind die Hintergründe der Organspezifität metastasierender Tumorzellen weitgehend ungeklärt. In 30% der Fälle kommt es zur Entstehung von Knochenmetastasen. Diese hohe Frequenz deutet darauf hin, dass NZK-Zellen bevorzugt in dieses Organ metastasieren, da die Knochenmatrix ein günstiges Mikromilieu für ihr Wachstum bietet. Hierbei könnte extrazellulärem Calcium und dem für die Detektion zuständigen Calcium-sensitiven Rezeptor (CaSR) eine entscheidende Rolle zukommen, da sich Knochen durch ihren hohen Gehalt an Calcium auszeichnen und von anderen Organen unterscheiden. Das Ziel der vorliegenden Dissertation lag in der Aufklärung der Mechanismen, die zu einer Knochenmetastasierung des NZK führen.rnrnIn ersten Analysen konnte gezeigt werden, dass sich bereits der Primärtumor durch eine von Calcium unabhängige charakteristische Expression bestimmter Signalmediatoren auszeichnet, die Metastasierungspotenzial und –ort bestimmen. So wurden in Gewebeproben und primären Tumorzellen von NZK-Patienten, die innerhalb von fünf Jahren nach Nephrektomie Knochenmetastasen entwickelten, in Westernblot-Analysen eine sehr hohe Expression der α5-Integrine, eine starke Aktivität von Akt, FAK und eine Reduktion der PTEN-Expression detektiert. Diese Veränderungen begünstigten die chemotaktische Migration in Richtung Fibronektin (bestimmt in einer Boyden-Kammer) und die Adhäsion dieser NZK-Zellen an Komponenten der Extrazellularmatrix (Fibronektin und Kollagen I – beides ist Bestandteil der Knochenmatrix). Migration und Adhäsion sind essentielle Schritte beim Austreten der Tumorzellen aus dem Primärtumor und Infiltration des Knochens. In NZK-Zellen von Patienten, die keine Metastasen oder Lungenmetastasen entwickelten, waren diese Charakteristika nicht oder deutlich schwächer ausgeprägt. Bestimmte Primärtumore sind somit prädestiniert Knochenmetastasen auszubilden.rnrnUm die Bedeutung von extrazellulärem Calcium und dem CaSR darzustellen, wurde die Expression des CaSR mittels Real-Time PCR, Westernblot-Analysen und durchflusszytometrisch in NZK-Gewebeproben und –Zellen von Patienten untersucht, die innerhalb von fünf Jahren nach Nephrektomie keine bzw. Lungen- oder Knochenmetastasen ausbildeten. Proben von Patienten mit Knochenmetastasen zeigten die stärkste Expression von CaSR-mRNA und CaSR-Protein. Durch eine Behandlung der NZK-Zellen mit Calcium in physiologischen Konzentrationen, konnte Calcium als möglicher Regulator der CaSR-Expression ausgeschlossen werden. Der Einfluss von Calcium auf die Metastasierungsfähigkeit der primären NZK-Zellen wurde anhand eines weiteren chemotaktischen Migrationsversuchs mit Calcium als Chemotaxin analysiert. Die Zellproliferationsrate konnte nach Behandlung der Zellen mit Calcium mittels BrdU-Inkorporation gemessen werden. NZK-Zellen, die aus dem Primärtumor von Patienten mit Knochenmetastasen kultiviert wurden, konnten durch eine erhöhte extrazelluläre Calcium-Konzentration verstärkt zu Migration und Proliferation (Konzentrations-abhängige Steigerung) angeregt werden. Diese stellen weitere essentielle Schritte bei der Infiltration und Vermehrung der NZK-Zellen in den Knochen dar. Die Effekte traten bei NZK-Zellen aus Patienten, die keine oder Lungenmetastasen ausbildeten, nicht auf. Die Identifizierung der beteiligten Signalwege erfolgte in Westernblot-Analysen und einem Phospho-Kinase Array. Hierdurch konnten eine verstärkte Aktivierung des Akt-, JNK-, p38α- und PLCγ-1-Signalwegs und eine beinahe vollständige Reduktion der PTEN-Expression nach Calcium-Behandlung in Knochen-metastasierenden NZK-Zellen aufgedeckt werden. Durch Blockierung des CaSR mittels des Inhibitors NPS 2143 konnte bestätigt werden, dass die metastasierungs-fördernde Wirkung von Calcium über den CaSR zustande kommt. rnrnNZK-Zellen zeichnen sich somit bereits im Primärtumor durch eine charakteristische Expression verschiedener Signalmediatoren aus, die ihr Metastasierungspotenzial und die mögliche Lokalisation der Metastase bestimmen. Gelangen metastasierende NZK-Zellen auf ihrem Weg durch den Blutkreislauf in das Knochenmilieu, verhilft ihnen hier eine hohe Expression des CaSR zu einem wichtigen Überlebensvorteil. Extrazelluläres Calcium wirkt über den CaSR, verstärkt ihre metastatischen Eigenschaften und fördert schließlich die Ausbildung einer Knochenmetastase. Aus diesem Grund kommt dem CaSR eine Rolle als möglicher prognostischer Marker hinsichtlich der Knochenmetastasierung beim NZK zu. Die Charakteristika des Primärtumors sollten daher die Auswahl des adjuvanten Therapeutikums und die Nachsorgeuntersuchungen beeinflussen. um die Medizin dem Ziel einer individualisierten Therapie näher zu bringen.rn

An interdisciplinary European group of clinical experts in the field of movement disorders and experienced Botulinum toxin users has updated the consensus for the use of Botulinum toxin in the treatment of children with cerebral palsy (CP). A problem-orientated approach was used focussing on both published and practice-based evidence. In part I of the consensus the authors have tabulated the supporting evidence to produce a concise but comprehensive information base, pooling data and experience from 36 institutions in 9 European countries which involves more than 10,000 patients and over 45,000 treatment sessions during a period of more than 280 treatment years. In part II of the consensus the Gross Motor Function Measure (GMFM) and Gross Motor Function Classification System (GMFCS) based Motor Development Curves have been expanded to provide a graphical framework on how to treat the motor disorders in children with CP. This graph is named "CP(Graph) Treatment Modalities - Gross Motor Function" and is intended to facilitate communication between parents, therapists and medical doctors concerning (1) achievable motor function, (2) realistic goal-setting and (3) treatment perspectives for children with CP. The updated European consensus 2009 summarises the current understanding regarding an integrated, multidisciplinary treatment approach using Botulinum toxin for the treatment of children with CP.

Adaptive immune responses are characterized by substantial restructuring of secondary lymphoid organs. The molecular and cellular factors responsible for virus-induced lymphoid remodeling are not well known to date. Here we applied optical projection tomography, a mesoscopic imaging technique, for a global analysis of the entire 3-dimensional structure of mouse peripheral lymph nodes (PLNs), focusing on B-cell areas and high endothelial venule (HEV) networks. Structural homeostasis of PLNs was characterized by a strict correlation between total PLN volume, B-cell volume, B-cell follicle number, and HEV length. After infection with lymphocytic choriomeningitis virus, we observed a substantial, lymphotoxin (LT) beta-receptor-dependent reorganization of the PLN microarchitecture, in which an initial B-cell influx was followed by 3-fold increases in PLN volume and HEV network length on day 8 after infection. Adoptive transfer experiments revealed that virus-induced PLN and HEV network remodeling required LTalpha(1)beta(2)-expressing B cells, whereas the inhibition of vascular endothelial growth factor-A signaling pathways had no significant effect on PLN expansion. In summary, lymphocytic choriomeningitis virus-induced PLN growth depends on a vascular endothelial growth factor-A-independent, LT- and B cell-dependent morphogenic pathway, as revealed by an in-depth mesoscopic analysis of the global PLN structure.
