The aim of the present work was to study how the information on product labels influences consumer expectations and their acceptance and purchase intention of dark chocolate. Six samples of dark choco- late, varying in brand (premium and store brand) and in type of product (regular dark chocolate, single cocoa origin dark chocolate and high percentage of cocoa dark chocolate), were evaluated by 109 con- sumers who scored their liking and purchase intention under three conditions: blind (only tasting the products), expected (observing product label information) and informed (tasting the products together with provision of the label information). In the expected condition, consumer liking was mainly affected by the brand. In the blind condition, differences in liking were due to the type of product; the samples with a high percentage of cocoa were those less preferred by consumers. Under the informed condition, liking of dark chocolates varied depending on both brand and type of product. Premium brand chocolates generated high consumer expectations of chocolate acceptability, which were fulfilled by the sensory characteristics of the products. Store brand chocolates created lower expectations, but when they were tasted they were as acceptable as premium chocolates. Claims of a high percentage of cocoa and single cocoa origin on labels did not generate higher expectations than regular dark chocolates.

Os objetivos deste trabalho foram caracterizar a resistência à vassoura-de-bruxa de plantas de cacau originadas do cruzamento entre TSH 1188 e CCN 51 (população segregante), por meio de dois métodos de inoculação em condições de campo, e identificar marcadores microssatélites específicos para grupos de plantas resistentes e suscetíveis. As plantas-controle avaliadas pelos métodos de inoculação natural e inoculação artificial em campo produziram os mesmos padrões de sintomas. As plantas da população segregante também coincidiram os padrões de sintomas em 90%, por esses dois métodos. O método de inoculação artificial em campo permite detectar falso-resistentes. Dos 18 pares de primers microssatélites amplificados, 15 foram polimórficos entre os genitores, e seis entre os grupos de plantas segregantes contrastantes quanto à resistência à vassoura-de-bruxa. Foram confirmadas três marcas previamente associadas a QTL (locos para características quantitativas) relacionados com a resistência à vassoura-de-bruxa, comuns a outras populações. Também foram identificados três novos QTL para esta característica, típicos desta população, o que comprova sua utilidade para o melhoramento genético do cacaueiro.

O objetivo deste trabalho foi avaliar o uso de decepa de plantas jovens de eucalipto, para produção de árvores de menor diâmetro, com colheita facilitada por pequenos produtores; ou para recuperação de povoamentos jovens severamente danificados e produção de biomassa para energia em ciclos curtos. O experimento foi conduzido em sistema agroflorestal, com espaçamento entre plantas de 9,5x4 m. O crescimento de plantas intactas foi comparado ao de brotações de decepas realizadas aos nove ou doze meses após plantio. Aos seis ou nove meses após a decepa, realizou-se a desbrota, para deixar dois ou três brotos por cepa. Quando a desbrota foi realizada aos 12 meses após a decepa, todos os brotos dominantes foram deixados. Um tratamento sem desbrota também foi avaliado. Aos 42 meses após plantio, o tratamento sem desbrota apresentou o mesmo número de brotos por cepa que o de plantas desbrotadas para três brotos. O diâmetro das brotações foi equivalente a 69% do valor estimado para as plantas intactas. O valor assintótico de produção das brotações, da maioria dos tratamentos de decepa aos nove meses, foi igual ao das plantas intactas, o que evidencia a viabilidade da decepa sem a realização da desbrota, em sistemas agroflorestais.

Previous studies have reported that a diet containing 10% cocoa, a rich source of flavonoids, has immunomodulatory effects on rats and, among others effects, is able to attenuate the immunoglobulin (Ig) synthesis in both systemic and intestinal compartments. The purpose of the present study was focused on investigating whether these effects were attributed exclusively to the flavonoid content or to other compounds present in cocoa. To this end, eight-week-old Lewis rats were fed, for two weeks, either a standard diet or three isoenergetic diets containing increasing proportions of cocoa flavonoids from different sources: one with 0.2% polyphenols from conventional defatted cocoa, and two others with 0.4% and 0.8% polyphenols, respectively, from non-fermented cocoa. Diet intake and body weight were monitored and fecal samples were obtained throughout the study to determine fecal pH, IgA, bacteria proportions, and IgA-coated bacteria. Moreover, IgG and IgM concentrations in serum samples collected during the study were quantified. At the end of the dietary intervention no clear changes of serum IgG or IgM concentrations were quantified, showing few effects of cocoa polyphenol diets at the systemic level. However, in the intestine, all cocoa polyphenol-enriched diets attenuated the age-related increase of both fecal IgA and IgA-coated bacteria, as well as the proportion of bacteria in feces. As these effects were not dependent on the dose of polyphenol present in the diets, other compounds and/or the precise polyphenol composition present in cocoa raw material used for the diets could be key factors in this effect.

Numerous health benefits have been attributed to cocoa and its derived products in the last decade including antioxidant, anti-platelet and positive effects on lipid metabolism and vascular function. Inflammation plays a key role in the initiation and progression of atherosclerosis. However, cocoa feeding trials focused on inflammation are still rare and the results yielded are controversial. Health effects derived from cocoa consumption have been partly attributed to its polyphenol content, in particular of flavanols. Bioavailability is a key issue for cocoa polyphenols in order to be able to exert their biological activities. In the case of flavanols, bioavailability is strongly influenced by several factors, such as their degree of polymerization and the food matrix in which the polyphenols are delivered. Furthermore, gut has become an active site for the metabolism of procyanidins (oligomeric and polymeric flavanols). Estimation of polyphenol consumption or exposure is also a very challenging task in Food and Nutrition Science in order to correlate the intake of phytochemicals with in vivo health effects. In the area of nutrition, modern analytical techniques based on mass spectrometry are leading to considerable advances in targeted metabolite analysis and particularly in Metabolomics or global metabolite analysis. In this chapter we have summarized the most relevant results of our recent research on the bioavailability of cocoa polyphenols in humans and the effect of the matrix in which cocoa polyphenols are delivered considering both targeted analysis and a metabolomic approach. Furthermore, we have also summarized the effect of long-term consumption of cocoa powder in patients at high risk of cardiovascular disease (CVD) on the inflammatory biomarkers of atherosclerosis.

O objetivo deste trabalho foi realizar monitoramento químico de seis substratos utilizados na produção de mudas de cacaueiros. Os substratos receberam 10 volumes (100mL, cada) de água destilada, e no lixiviado coletado foram determinados o pH, a condutividade elétrica (CE) e os teores de sódio (Na) e potássio (K) em solução. Os valores de pH variaram de 5,0 a 8,0 em Plantmax® e composto do tegumento da amêndoa do cacau (CTAC), respectivamente. Verificou-se que, a partir do primeiro volume lixiviado, a CE de todos os substratos foi reduzida. Foram encontradas correlações positivas para a CE em relação ao K e Na da fibra de coco (FC) (r=0,83 e 0,72 P<0,05) e CTAC (r=0,98 e 0,99 P<0,05). Os resultados indicam que a FC e o CTAC deveriam ser usados após pré-lavagens. Verificou-se que, embora a FC apresente na primeira lixiviação valor de CE superior aos demais substratos, o valor ideal da 2,0 dSm-1 é alcançado na segunda lixiviação. Os teores de K e Na são um indicativo do valor da CE em FC e CTAC.

O alagamento do solo pode promover alterações no metabolismo celular e causar desvios nas condições ótimas de crescimento das plantas, gerando uma condição de estresse. Objetivou-se no presente trabalho avaliar os efeitos do alagamento no crescimento e na nutrição mineral de seis clones de T. cacao (CP-49, CCN-10, CP-06, CEPEC-2007, CEPEC-2008 e PS-1319), para elucidar possíveis mecanismos de tolerância ao alagamento. Mudas clonais de T. cacao, com 6 meses de idade, foram submetidas ao alagamento, juntamente com o tratamento-controle (não alagado), por 30 dias. Observou-se, no final desse período, que o alagamento promoveu diminuição nas taxas de crescimento relativo radicular (exceto para CP-06) e de área foliar, acúmulo de matéria seca e incremento nas taxas de crescimento relativo caulinar (exceto para os clones CP-06 e CEPEC-2008) e assimilatória líquida (exceto para o clone alagado PS-1319) e de massa foliar específica para os clones CP-49, CCN-10 e CP-06. Os clones que sobreviveram aos 30 dias de alagamento apresentaram baixos valores de razão de área foliar. De modo geral, o alagamento do substrato acarretou deficiência na absorção de macro e micronutrientes minerais, exceto de Fe. Dentre os clones avaliados, o CP-49 foi tolerante e o CEPEC-2008 não tolerante ao alagamento. Os demais clones ficaram numa posição intermediária. Logo, o clone CP-49 tem grande potencial para ser cultivado em condições de alagamento.

Previous studies in young rats reported the impact of cocoa intake on healthy immune status and allow suggesting it may have a role in the prevention of some immune-mediated diseases. The aim of this study was to ascertain the effect of a cocoa diet in a model of allergy in young rats. Three-week-old Brown Norway rats were immunized by i.p. injection of ovalbumin (OVA) with alum as adjuvant and Bordetella pertussis toxin. During the next 4 weeks rats received either a cocoa diet (containing 0.2% polyphenols, w/w) or a standard diet. Animals fed a standard diet showed high concentrations of anti-OVA IgG1, IgG2a, IgG2b and high anti-OVA IgE titres, which is the antibody involved in allergic response. In contrast, animals fed a cocoa diet showed significantly lower concentrations of anti-OVA IgG1 and IgG2a antibodies. Interestingly, the cocoa diet prevented anti-OVA IgE synthesis and decreased total serum IgE concentration. Analysis of cytokine production in lymph node cells at the end of the study revealed that, in this compartment, the cocoa diet decreased the tumor necrosis factor (TNF) - alpha and the interleukin (IL) -10 secretion but not IL-4 production. In conclusion, a cocoa-enriched diet in young rats produces an immunomodulatory effect that prevents anti-allergen IgE synthesis, suggesting a potential role for cocoa flavonoids in the prevention or treatment of allergic diseases.

The effect of different food matrices on the metabolism and excretion of polyphenols is uncertain. The objective of the study was to evaluate the possible effect of milk on the excretion of (2)-epicatechin metabolites from cocoa powder after its ingestion with and without milk. Twenty-one volunteers received the following three test meals each in a randomised cross-over design with a 1-week interval between meals: (1) 250 ml whole milk as a control; (2) 40 g cocoa powder dissolved in 250 ml whole milk (CC-M); (3) 40 g cocoa powder dissolved in 250 ml water (CC-W). Urine was collected before consumption and during the 0-6, 6-12 and 12-24 h periods after consumption. (2)-Epicatechin metabolite excretion was measured using liquid chromatography-MS. One (2)-epicatechin glucuronide and three (2)-epicatechin sulfates were detected in urine excreted after the intake of the two cocoa beverages (CC-M and CC-W). The results show that milk does not significantly affect the total amount of metabolites excreted in urine. However, differences in metabolite excretion profiles were observed; there were changes in the glucuronide and sulfate excretion rates, and the sulfation position between the period of excretion and the matrix. The matrix in which polyphenols are consumed can affect their metabolism and excretion, and this may affect their biological activity. Thus, more studies are needed to evaluate the effect of these different metabolite profiles on the body.

O objetivo deste trabalho foi detectar uma possível especificidade alimentar de duas espécies crípticas de cochonilhas do gênero Planococcus, refletida em seu desenvolvimento em frutos de cacaueiro, cafeeiro e citros. A cochonilha Planococcus minor (Maskell), obtida de frutos de cacau (Theobroma cacao L. cv. Comum), e Planococcus citri (Risso), de lavoura de café (Coffea arabica L. cv. Mundo Novo) e de mudas de citros (Citrus sinensis L. Osbeck cv. Bahia), foram criadas em abóboras (Cucurbita maxima L.), em laboratório. Rosetas com frutos de café foram mantidas sobre uma lâmina de 5 mm de ágar-água em placas de Petri, vedadas com filme plástico de polietileno. Em frutos de citros e cacau foram utilizadas gaiolas plásticas cilíndricas (1,5 cm x 3,0 cm), vedadas com voile na parte superior, as quais foram fixadas nos frutos por meio de um elástico. Os bioensaios foram conduzidos em câmaras climatizadas a 25 ± 1ºC, 70 ± 10% UR e 12 horas de fotofase, utilizando-se de 30 repetições. Para as cochonilhas provenientes de cafeeiro, o substrato café proporcionou o maior período ninfal de fêmeas (20,8 dias) e maior longevidade (31,7 dias). Para fêmeas oriundas de cacau, o substrato cacau promoveu o menor período ninfal (21,1 dias) e maior longevidade (25,0 dias). Para aquelas oriundas de citros, o substrato cacau promoveu o menor período ninfal (18,4 dias), e o substrato citros, a maior longevidade (32,0 dias). As maiores porcentagens de mortalidade (50%) foram obtidas das ninfas oriundas de frutos de cacau e citros, criadas em café e cacau, e as menores foram das ninfas oriundas de frutos de café, independentemente do substrato em que foram criadas. A cochonilha P. minor mostra uma associação mais estreita com o cacau e, eventualmente, café, em relação ao citros, o que explicaria sua maior ocorrência em cacau. No entanto, P. citri não evidencia nenhuma especificidade para os três substratos testados.

Em função da importância da casca do fruto como principal resíduo da cacauicultura e as possibilidades de seu uso, este trabalho teve como objetivos realizar análise de nutrientes do extrato orgânico obtido por lavagem do composto de casca e avaliar seu efeito como fertilizante potássico no solo e no crescimento de mudas de cacaueiro. O ensaio foi realizado em casa de vegetação, com aplicação de doses de extrato via solo, cultivo em tubetes e mudas seminais de cacaueiro. O delineamento experimental foi em blocos casualizados, com três repetições, e a unidade experimental formada por nove plantas crescidas individualmente em tubetes. Os tratamentos foram 5 doses de K: zero; 125; 250; 500 e 1.000 mg de K dm-3 de solo. Após 120 dias da aplicação do extrato no solo, verificou-se que o pH, a saturação de bases e os teores disponíveis de Ca, Mg, K, Zn e Mn aumentaram, e o teor de Al e Fe diminuíram. Os resultados também evidenciaram que a dose de 1.000 mg K dm-3 de solo desequilibrou as relações K/Mg e K/Ca nas folhas com redução no crescimento das mudas. Mudas de cacaueiro apresentaram crescimento significativo em função de doses de K e, considerando a facilidade de produzir e aplicar o extrato da casca do fruto do cacaueiro, é possível usá-lo como fonte de K na produção de mudas de cacaueiro.

Cocoa is a food relatively rich in polyphenols, which makes it a potent antioxidant. Due to its activity as an antioxidant, as well as through other mechanisms, cocoa consumption has been reported to be beneficial for cardiovascular health, brain functions, and cancer prevention. Furthermore, cocoa influences the immune system, in particular the inflammatory innate response and the systemic and intestinal adaptive immune response. Preclinical studies have demonstrated that a cocoa-enriched diet modifies T-cell functions that conduce to a modulation of the synthesis of systemic and gut antibodies. In this regard, it seems that a cocoa diet in rats produces changes in the lymphocyte composition of secondary lymphoid tissues and the cytokines secreted by T cells. These results suggest that it is possible that cocoa could inhibit the function of Th2 cells, and in line with this, the preventive effect of cocoa on IgE synthesis in a rat allergy model has been reported, which opens up new perspectives when considering the beneficial effects of cocoa compounds. On the other hand, cocoa intake modifies the functionality of gut-associated lymphoid tissue by means of modulating IgA secretion and intestinal microbiota. The mechanisms involved in these influences are discussed here. Further research may elucidate the cocoa compounds involved in such an effect and also the possible medical approaches to these repercussions

Background: Epidemiologic studies have suggested that flavonoid intake plays a critical role in the prevention of coronary heart disease. Because atherosclerosis is considered a low-grade inflammatory disease, some feeding trials have analyzed the effects of cocoa (an important source of flavonoids) on inflammatory biomarkers, but the results have been controversial. Objective: The objective was to evaluate the effects of chronic cocoa consumption on cellular and serum biomarkers related to atherosclerosis in high-risk patients. Design: Forty-two high-risk volunteers (19 men and 23 women; mean 6 SD age: 69.7 6 11.5 y) were included in a randomized crossover feeding trial. All subjects received 40 g cocoa powder with 500 mL skim milk/d (C+M) or only 500 mL skim milk/d (M) for 4 wk. Before and after each intervention period, cellular and serum inflammatory biomarkers related to atherosclerosis were evaluated. Results: Adherence to the dietary protocol was excellent. No significant changes in the expression of adhesion molecules on T lymphocyte surfaces were found between the C+M and M groups. However, in monocytes, the expression of VLA-4, CD40, and CD36 was significantly lower (P = 0.005, 0.028, and 0.001, respectively) after C+M intake than after M intake. In addition, serum concentrations of the soluble endothelium-derived adhesion molecules P-selectin and intercellular adhesion molecule-1 were significantly lower (both P = 0.007) after C+M intake than after M intake. Conclusions: These results suggest that the intake of cocoa polyphenols may modulate inflammatory mediators in patients at high risk of cardiovascular disease. These antiinflammatory effects may contribute to the overall benefits of cocoa consumption against atherosclerosis. Previous studies have shown that rat intestinal immunoglobulin A (IgA) concentration and lymphocyte composition of the intestinal immune system were influenced by a highly enriched cocoa diet. The aim of this study was to dissect the mechanisms by which a long-term high cocoa intake was capable of modifying gut secretory IgA in Wistar rats. After 7 weeks of nutritional intervention, Peyer's patches, mesenteric lymph nodes and the small intestine were excised for gene expression assessment of IgA, transforming growth factor ß, C-C chemokine receptor-9 (CCR9), interleukin (IL)-6, CD40, retinoic acid receptors (RAR¿ and RARß), C-C chemokine ligand (CCL)-25 and CCL28 chemokines, polymeric immunoglobulin receptor and toll-like receptors (TLR) expression by real-time polymerase chain reaction. As in previous studies, secretory IgA concentration decreased in intestinal wash and fecal samples after cocoa intake. Results from the gene expression showed that cocoa intake reduced IgA and IL¿6 in Peyer's patches and mesenteric lymph nodes, whereas in small intestine, cocoa decreased IgA, CCR9, CCL28, RAR¿ and RARß. Moreover, cocoa-fed animals presented an altered TLR expression pattern in the three compartments studied. In conclusion, a high-cocoa diet down-regulated cytokines such as IL-6, which is required for the activation of B cells to become IgA-secreting cells, chemokines and chemokine receptors, such as CCL28 and CCR9 together with RAR¿ and RARß, which are involved in the gut homing of IgA-secreting cells. Moreover, cocoa modified the cross-talk between microbiota and intestinal cells as was detected by an altered TLR pattern. These overall effects in the intestine may explain the intestinal IgA down-regulatory effect after the consumption of a long-term cocoa-enriched diet.
