La toxine thermostable d’E.coli (STb) est une cause de diarrhée chez l’homme et l’animal. STb se lie au sulfatide, son récepteur, puis s’internalise. Dans le cytoplasme, par une cascade d’événements, STb déclenche l’ouverture des canaux ioniques permettant la sécrétion des ions et la perte d’eau menant à la diarrhée. Les jonctions serrées forment une barrière physique intercellulaire dans les cellules épithéliales intestinales, contrôlant ainsi le flux paracellulaire des ions et de l’eau. Les jonctions serrées sont affectées par divers pathogènes et par leurs toxines. À ce jour, l’effet de STb sur les jonctions serrées n’a pas été étudié. L’étude entreprise visait à explorer l’effet de STb sur les jonctions serrées et la barrière épithéliale des cellules intestinales. Des cellules épithéliales intestinales du colon humain (T84) ont été traitées pendant 24h soit avec la toxine STb purifiée soit avec une souche d’E.coli exprimant STb. La résistance transépithéliale (TER), le flux de marqueurs paracellulaires et la microscopie confocale ont été utilisés pour analyser les effets de STb sur les jonctions serrées. Les monocouches traitées par la souche E.coli exprimant STb et la toxine STb purifiée ont manifesté une forte réduction de TER (p<0.0001) parallèlement à une augmentation significative de la perméabilité paracellulaire à l’Albumine de Sérum Bovin marqué avec l’IsoThioCyanate Fluoroscéine, BSA-FITC (p<0.0001) comparativement aux cellules non traitées et aux cellules traitées par une souche d’E.coli commensale non-toxinogène. L’augmentation de la perméabilité paracellulaire induite par STb a été associée à une dissolution générale et une condensation des fibres de stress centrales des filaments d’actine. Le réarrangement des filaments d’actine a été accompagné par une redistribution et une fragmentation des protéines des jonctions serrées dont l’occludine, la claudine-1 et la Zonula Occludens-1. Les mêmes modifications on été observées après l’intoxication des cellules T84 avec un octapeptide synthétique retrouvé dans la séquence de STb correspondant à une séquence consensus de la toxine ZOT de Vibrio cholerae, impliquée dans la réorganisation des jonctions serrées. Cet effet n’a pas été observé lorsque les cellules ont été traitées avec un octapeptide synthétique comportant les mêmes acides aminés mais distribués de façon aléatoire ou avec la toxine mutée (D30V). Nos résultats montrent pour la première fois que STb induit le dysfonctionnement de la barrière épithéliale intestinale en modifiant la distribution des protéines des jonctions serrées. Ces résultats ouvrent une nouvelle voie pour la compréhension de la pathogenèse de diarrhée causée par la toxine STb.

Le risque de chute est une problématique bien présente chez les personnes âgées ou ayant une atteinte neurologique et reflète un déficit des mécanismes neuronaux assurant l’équilibre. De précédentes études démontrent que l’intégration des informations sensorielles est essentielle au contrôle de l’équilibre et que l’inhibition présynaptique (IP) serait un mécanisme important dans le contrôle de la transmission sensorielle. Ainsi, le but de cette étude était d’identifier la contribution du mécanisme d’IP à l’induction de réponses posturales efficaces suite à une perturbation d’équilibre. Notre hypothèse est qu’une diminution d’IP contribuerait à l’induction des ces réponses, en augmentant l’influence de la rétroaction sensorielle sur les réseaux de neurones spinaux. Afin de démontrer cette hypothèse, nous avons d’abord évalué l’excitabilité spinale pendant les perturbations vers l’avant ou vers l’arrière, à l’aide du réflexe H. L’excitabilité spinale était modulée selon la direction de la perturbation et cette modulation survenait dès 75 ou 100 ms (p<0.05), soit avant l’induction des réactions posturales. Puis, à l’aide de techniques plus précises de convergence spinale, nous avons démontré que l’IP était diminuée dès 75 et 100 ms dans les deux directions, suggérant que la transmission des informations sensorielles vers la moelle épinière est accrue juste avant le déclenchement de la réponse posturale. Cette étude met en évidence un mécanisme-clé permettant d’augmenter la rétroaction des informations sensorielles nécessaires à l’induction de réponses posturales appropriées. L’évaluation de ce mécanisme pourrait mener à une meilleure identification des individus à risque de chute.

Les cellules endothéliales (EC) constituent une première barrière physique à la dissémination de virus pléiotropiques circulant par voie hématogène mais leur contribution à la défense innée anti-virale est peu connue. Des dysfonctions des EC de la barrière hémato-encéphalique (BMEC) et des sinusoïdes hépatiques (LSEC) ont été rapportées dans des neuropathologies et des hépatites aiguës ou chroniques d’origine virale, suggérant que des atteintes à leur intégrité contribuent à la pathogenèse. Les sérotypes de coronavirus de l’hépatite murine (MHV), se différenciant par leur capacité à induire des hépatites et des maladies neurologiques de sévérité variable et/ou leur tropisme pour les EC, représentent des modèles viraux privilégiés pour déterminer les conséquences de l’infection des EC sur la pathogenèse virale. Lors d’infection par voie hématogène, le sérotype MHV3, le plus virulent des MHV, induit une hépatite fulminante, caractérisée par une réponse inflammatoire sévère, et des lésions neurologiques secondaires alors que le sérotype moins virulent, MHV-A59, induit une hépatite modérée sans atteintes secondaires du système nerveux central (SNC). Par ailleurs, le sérotype MHV3, à la différence du MHV-A59, démontre une capacité à stimuler la production de cytokines par la voie TLR2. Les variants atténués du MHV3, les virus 51.6-MHV3 et YAC-MHV3, sont caractérisés par un faible tropisme pour les LSEC et induisent respectivement une hépatite modérée et subclinique. Compte tenu de l’importance des LSEC dans le maintien de la tolérance hépatique et de l’élimination des pathogènes circulants, il a été postulé que la sévérité de l’hépatite et de la réponse inflammatoire lors d’infections par les MHV est associée à la réplication virale et à l’altération des propriétés tolérogéniques et vasculaires des LSEC. Les désordres inflammatoires hépatiques pourraient résulter d’une activation différentielle du TLR2, plutôt que des autres TLR et des hélicases, selon les sérotypes. D’autre part, compte tenu du rôle des BMEC dans la prévention des infections du SNC, il a été postulé que l’invasion cérébrale secondaire par les coronavirus est reliée à l’infection des BMEC et le bris subséquent de la barrière hémato-encéphalique (BHE). À l’aide d’infections in vivo et in vitro par les différents sérotypes MHV, chez des souris ou des cultures de BMEC et de LSEC, nous avons démontré, d’une part, que l’infection in vitro des LSEC par le sétotype MHV3, à la différence des variants 51.6- et YAC-MHV3, altérait la production du facteur vasodilatant NO et renversait leur phénotype tolérogénique en favorisant la production de cytokines et de chimiokines inflammatoires. Ces dysfonctions se traduisaient in vivo par une réponse inflammatoire incontrôlée et une dérégulation du recrutement intrahépatique de leucocytes, favorisant la réplication virale et les dommages hépatiques. Nous avons aussi démontré, à l’aide de souris TLR2 KO et de LSEC dont l’expression du TLR2 a été abrogée par des siRNA, que la sévérité de l’hépatite et de la réponse inflammatoire induite par le sérotype MHV3, dépendait en partie de l’induction et de l’activation préférentielle du TLR2 par le virus dans le foie. D’autre part, la sévérité de la réplication virale au foie et des désordres dans le recrutement leucocytaire intrahépatique induits par le MHV3, et non par le MHV-A59 et le 51.6-MHV3, corrélaient avec une invasion virale subséquente du SNC, au niveau de la BHE. Nous avons démontré que l’invasion cérébrale du MHV3 était associée à une infection productive des BMEC et l’altération subséquente des protéines de jonctions serrées occludine, VE-cadhérine et ZO-1 se traduisant par une augmentation de la perméabilité de la BHE et l’entrée consécutive du virus dans le cerveau. Dans l’ensemble, les résultats de cette étude mettent en lumière l’importance du maintien de l’intégrité structurale et fonctionnelle des LSEC et des BMEC lors d’infections virales aigües par des MHV afin de limiter les dommages hépatiques associés à l’induction d’une réponse inflammatoire exagérée et de prévenir le passage des virus au cerveau suite à une dissémination par voie hématogène. Ils révèlent en outre un nouveau rôle aggravant pour le TLR2 dans l’évolution de l’hépatite virale aigüe ouvrant la voie à de nouvelles avenues thérapeutiques visant à moduler l’activité inflammatoire du TLR2.

Background:  Several authors have highlighted areas of overlap in symptoms and impairment among children with autism spectrum disorder (ASD) and children with specific language impairment (SLI). By contrast, loss of language and broadly defined regression have been reported as relatively specific to autism. We compare the incidence of language loss and language progression of children with autism and SLI. Methods:  We used two complementary studies: the Special Needs and Autism Project (SNAP) and the Manchester Language Study (MLS) involving children with SLI. This yielded a combined sample of 368 children (305 males and 63 females) assessed in late childhood for autism, history of language loss, epilepsy, language abilities and nonverbal IQ. Results:  language loss occurred in just 1% of children with SLI but in 15% of children classified as having autism or autism spectrum disorder. Loss was more common among children with autism rather than milder ASD and is much less frequently reported when language development is delayed. For children who lost language skills before their first phrases, the phrased speech milestone was postponed but long-term language skills were not significantly lower than children with autism but without loss. For the few who experienced language loss after acquiring phrased speech, subsequent cognitive performance is more uncertain. Conclusions:  Language loss is highly specific to ASD. The underlying developmental abnormality may be more prevalent than raw data might suggest, its possible presence being hidden for children whose language development is delayed.

Tight junctions between intestinal epithelial cells prevent ingress of luminal macromolecules and bacteria and protect against inflammation and infection. During stress and inflammation, mast cells mediate increased mucosal permeability by unknown mechanisms. We hypothesized that mast cell tryptase cleaves protease-activated receptor 2 (PAR2) on colonocytes to increase paracellular permeability. Colonocytes expressed PAR2 mRNA and responded to PAR2 agonists with increased [Ca2+]i. Supernatant from degranulated mast cells increased [Ca2+]i in colonocytes, which was prevented by a tryptase inhibitor, and desensitized responses to PAR2 agonist, suggesting PAR2 cleavage. When applied to the basolateral surface of colonocytes, PAR2 agonists and mast cell supernatant decreased transepithelial resistance, increased transepithelial flux of macromolecules, and induced redistribution of tight junction ZO-1 and occludin and perijunctional F-actin. When mast cells were co-cultured with colonocytes, mast cell degranulation increased paracellular permeability of colonocytes. This was prevented by a tryptase inhibitor. We determined the role of ERK1/2 and of beta-arrestins, which recruit ERK1/2 to PAR2 in endosomes and retain ERK1/2 in the cytosol, on PAR2-mediated alterations in permeability. An ERK1/2 inhibitor abolished the effects of PAR2 agonist on permeability and redistribution of F-actin. Down-regulation of beta-arrestins with small interfering RNA inhibited PAR2-induced activation of ERK1/2 and suppressed PAR2-induced changes in permeability. Thus, mast cells signal to colonocytes in a paracrine manner by release of tryptase and activation of PAR2. PAR2 couples to beta-arrestin-dependent activation of ERK1/2, which regulates reorganization of perijunctional F-actin to increase epithelial permeability. These mechanisms may explain the increased epithelial permeability of the intestine during stress and inflammation.

Este estudo foi desenvolvido dentro do projeto de pesquisa “Micrometeorologia da Mata Atlântica alagoana (MICROMA)” do Departamento de Meteorologia da Universidade Federal de Alagoas, que busca avaliar modificações no sistema solo-planta-atmosfera provocadas pela substituição da Mata Atlântica pela cultura da cana-de-açúcar, na Zona da Mata alagoana. O objetivo principal do presente estudo foi o ajuste do modelo de Penman-Monteith para o cálculo da evapotranspiração da cultura da cana-de-açúcar, na zona canavieira de Alagoas, a partir de determinações micrometeorológicas pelo método do balanço de energia, em diferentes estádios de desenvolvimento da cultura e em diferentes condições de demanda evaporativa atmosférica. A evapotranspiração foi estimada pelo método de Penman-Monteith e comparada com valores calculados através do balanço de energia, pela razão de Bowen, considerando este como método padrão. Medições micrometeorológicas foram feitas nos anos agrícolas de 1997/98 e 1998/99, em um área de 12,5ha de cultivo contínuo de cana-deaçúcar, cultivar SP 70-1143, sob condições de lavoura comercial, na Fazenda Vila Nova, município de Pilar, Alagoas (9o36´S, 35o53´W, 107m de altitude). Durante o período experimental, foram tomadas amostras quinzenais para determinar altura do dossel, comprimento de colmo, índice de área foliar e acúmulo de matéria seca (este apenas em 1998/99). Os parâmetros aerodinâmicos da cultura zo (comprimento de rugosidade) e d (deslocamento do plano zero) foram determinados a partir de perfis de vento, em condições de atmosfera neutra, e relacionados com a altura do dossel. Foram ajustados modelos para estimar o índice de área foliar e a altura do dossel da cultura em função do comprimento do colmo. O fracionamento do saldo de radiação no chamado “período potencial”, sem deficiência hídrica e com a cultura cobrindo completamente o solo, foi de 78% para o fluxo de calor latente de evaporação, 21% para o fluxo de calor sensível na atmosfera e aproximadamente 0,1% para o luxo de calor no solo. A estimativas pelo método de Penman-Monteith se ajustaram ao valores calculados pelo método do balanço de energia, tanto para médias qüinqüidiais (r = 0,93) como para valores diários (r = 0,91). Assim, dados confiáveis de evapotranspiração da cultura de cana-de-açúcar podem ser obtidos pelo método de Penman-Monteith ajustado neste trabalho, para a zona canavieira de Alagoas, em períodos sem deficiência hídrica e com cobertura completa do solo. O uso deste método apresenta a vantagem prática de requerer apenas dados meteorológicos, medidos em apenas um nível, em estações meteorológicas convencionais.

Objective: The purpose of this study was to evaluate, by shear bond strength (SBS) testing, the influence of different types of temporary cements on the final cementation using conventional and self-etching resin-based luting cements. Material and Methods: Forty human teeth divided in two halves were assigned to 8 groups (n=10): I and V (no temporary cementation); II and VI: Ca(OH)(2)-based cement; III and VII: zinc oxide (ZO)based cement; IV and VIII: ZO-eugenol (ZOE)-based cement. Final cementation was done with RelyX ARC cement (groups I to IV) and RelyX Unicem cement (groups V to VIII). Data were analyzed statistically by ANOVA and Tukey's test at 5% significance level. Results: Means were (MPa): I - 3.80 (+/- 1.481); II - 5.24 (+/- 2.297); III - 6.98 (+/- 1.885); IV - 6.54 (+/- 1.459); V - 5.22 (+/- 2.465); VI - 4.48 (+/- 1.705); VII - 6.29 (+/- 2.280); VIII - 2.47 (+/- 2.076). Comparison of the groups that had the same temporary cementation (Groups II and VI; III and VII; IV and VIII) showed statistically significant difference (p<0.001) only between Groups IV and VIII, in which ZOE-based cements were used. The use of either Ca(OH) 2 based (Groups II and VI) or ZO-based (Groups III and VII) cements showed no statistically significant difference (p>0.05) for the different luting cements (RelyX (TM) ARC and RelyX (TM) Unicem). The groups that had no temporary cementation (Groups I and V) did not differ significantly from each other either (p>0.05). Conclusion: When temporary cementation was done with ZO- or ZOE-based cements and final cementation was done with RelyX ARC, there was an increase in the SBS compared to the control. In the groups cemented with RelyX Unicem, however, the use of a ZOE-based temporary cement affected negatively the SBS of the luting agent used for final cementation.

JUSTIFICATIVA E OBJETIVOS: Verificar o efeito sedativo da clonidina, um a2-agonista, e do midazolam, um benzodiazepínico, quando utilizados na medicação pré-anestésica, empregando-se avaliação clínica e eletroencefalográfica bispectral. MÉTODO: Após aprovação institucional e consentimento escrito fornecido, 45 pacientes de 18 a 65 anos, estado físico ASA I, foram aleatoriamente distribuídos nos grupos placebo (P), clonidina (C) ou midazolam (M), em que receberam, respectivamente placebo, 150 µg de clonidina ou 15 mg de midazolam por via oral, 60 minutos antes da indução da anestesia (n = 15 por grupo). A monitorização constituiu-se de eletrocardiograma (D II), pressão arterial não invasiva, freqüência cardíaca, saturação de pulso de oxigênio, freqüência respiratória, temperatura axilar e da sala de cirurgia e eletroencefalograma bispectral para determinação do índice bispectral (BIS). Esses atributos e a escala de sedações (1 - ansioso, 2 - calmo, 3 - sonolento, 4 - dormindo com reflexo, 5 - dormindo sem reflexo) foram obtidos aos 0 (M0), 15 (M15), 30 (M30), 40 (M40), 50 (M50) e 60 (M60) minutos após a medicação. RESULTADOS: Nos grupos não houve alteração significante dos parâmetros respiratórios, hemodinâmicos e de temperatura. Houve diferença significante entre os grupos na ES (M60: M=C>P) e no BIS (M50 e M60: M=C>P). CONCLUSÕES: Nas condições utilizadas, a clonidina e o midazolam determinaram níveis de sedação adequados e semelhantes na medicação pré-anestésica de pacientes estado físico ASA I, quando avaliados pela escala de sedação e pelo índice bispectral, sem determinarem alterações hemodinâmicas e respiratórias.

The observation that mice with a selective ablation of the androgen receptor (AR) in Sertoli cells (SC) (SCARKO mice) display a complete block in meiosis supports the contention that SC play a pivotal role in the control of germ cell development by androgens. To delineate the physiological and molecular mechanism responsible for this control, we compared tubular development in pubertal SCARKO mice and littermate controls. Particular attention was paid to differences in SC maturation, SC barrier formation and cytoskeletal organization and to the molecular mediators potentially involved. Functional analysis of SC barrier development by hypertonic perfusion and lanthanum permeation techniques and immunohistochemical analysis of junction formation showed that SCARKO mice still attempt to produce a barrier separating basal and adluminal compartment but that barrier formation is delayed and defective. Defective barrier formation was accompanied by disturbances in SC nuclear maturation (immature shape, absence of prominent, tripartite nucleoli) and SC polarization (aberrant positioning of SC nuclei and cytoskeletal elements such as vimentin). Quantitative RT-PCR was used to study the transcript levels of genes potentially related to the described phenomena between day 8 and 35. Differences in the expression of SC genes known to play a role in junction formation could be shown from day 8 for Cldn11, from day 15 for Cldn3 and Espn, from day 20 for Cdh2 and Jam3 and from day 35 for ZO-1. Marked differences were also noted in the transcript levels of several genes that are also related to cell adhesion and cytoskeletal dynamics but that have not yet been studied in SC (Actn3, Ank3, Anxa9, Scin, Emb, Mpzl2). It is concluded that absence of a functional AR in SC impedes the remodeling of testicular tubules expected at the onset of spermatogenesis and interferes with the creation of the specific environment needed for germ cell development.

Introduction: The aim of this study was to evaluate the pH, calcium ion release, setting time, and solubility of white mineral trioxide aggregate (WMTA) and white Portland cement (WPC) combined with the following radiopacifying agents: bismuth oxide (BO), calcium tungstate (CT), and zirconium oxide (ZO). Methods: Fifty acrylic teeth with root-end filling material were immersed in ultrapure water for measurement of pH and calcium release (atomic absorption spectrophotometry) at 3, 24, 72, and 168 hours. For evaluation of setting time, each material was analyzed according to the American Society for Testing and Materials guidelines 266/08. The solubility test was performed according to American National Standards Institute/American Dental Association specification no. 57/2000. Solubility, setting time, and pH values were compared by using analysis of variance and Tukey test, and the values of calcium release were compared by the Kruskal-Wallis and Miller tests. The significance level was set at 5%. Results: The pH and calcium release were higher at 3 and 24 hours. WPC was the material with the higher values for both properties. WMTA had the greatest solubility among all materials (P <.05). All radiopacifiers increased the setting time of WPC, and WMTA had the shortest setting time among all materials (P < .05). Conclusions: All materials released calcium ions. Except for WPC/CT at 168 hours, all materials promoted an alkaline pH. On the basis of the obtained results, ZO and CT can be considered as potential radiopacifying agents to be used in combination with Portland cement. The aim of this study was to evaluate the interference of the radiopacifiers bismuth oxide (BO), bismuth carbonate (BC), bismuth subnitrate (BS), and zirconiun oxide (ZO) on the solubility, alkalinity and antimicrobial properties of white Portland cement (WPC). The substances were incorporated to PC, at a ratio of 1:4 (v/v) and subjected to a solubility test. To evaluate the pH, the cements were inserted into retrograde cavities prepared in simulated acrylic teeth and immediately immersed in deionized water. The pH of the solution was measured at 3, 24, 72 and 168 h. The antimicrobial activity was evaluated by a radial diffusion method against the microorganisms S. aureus (ATCC 25923), P. aeruginosa (ATCC 27853), E. faecalis (ATCC 29212) and C. albicans (ATCC 10231). The zone of microbial growth inhibition was measured after 24 h. The addition of BS and BC increased the solubility of the cement. The pH values demonstrated that all materials produced alkaline levels. At 3 h, BS showed lower pH than WPC (p<0.05). At 168 h, all materials showed similar pHs (p>0.05). The materials did not present antimicrobial activity for S. aureus, P. aeruginosas and E. faecalis (p>0.05). With regards to C. albicans, all materials formed an inhibition zone, mainly the mixture of WPC with ZO (p<0.05). The type of radiopacifier incorporated into WPC interfered with its physical and antimicrobial properties. ZO was found to be a viable radiopacifier that can be used with WPC.

O empilhamento por superfície de reflexão comum (ou empilhamento SRC), conhecido como empilhamento CRS, do inglês Commom reflection surface, constitui-se em um novo método para o processamento sísmico na simulação de seções afastamento nulo (AN) e afastamento comum (AC). Este método é baseado em uma aproximação paraxial hiperbólica de segunda ordem dos tempos de trânsito de reflexão na vizinhança de um raio central. Para a simulação de seção AN, o raio central é um raio normal, enquanto que para a simulação de uma seção AC o raio central é um raio de afastamento finito. Em adição à seção AN, o método de empilhamento SRC também fornece estimativas dos atributos cinemáticos do campo de onda, sendo aplicados, por exemplo, na determinação (por um processo de inversão) da velocidade intervalar, no cálculo do espalhamento geométrico, na estimativa da zona de Fresnel, e também na simulação de eventos de tempos de difrações, este último tendo uma grande importância para a migração pré-empilhamento. Neste trabalho é proposta uma nova estratégia para fazer uma migração em profundidade pré-empilhamento, que usa os atributos cinemáticos do campo de onda derivados do empilhamento SRC, conhecido por método CRS-PSDM, do inglês CRS based pre-stack depth migration. O método CRS-PSDM usa os resultados obtidos do método SRC, isto é, as seções dos atributos cinemáticos do campo de onda, para construir uma superfície de tempos de trânsito de empilhamento, ao longo da qual as amplitudes do dado sísmico de múltipla cobertura são somadas, sendo o resultado da soma atribuído a um dado ponto em profundidade, na zona alvo de migração que é definida por uma malha regular. Similarmente ao método convencional de migração tipo Kirchhoff (K-PSDM), o método CRS-PSDM precisa de um modelo de velocidade de migração. Contrário ao método K-PSDM, o método CRS-PSDM necessita apenas computar os tempos de trânsito afastamento nulo, ao seja, ao longo de um único raio ligando o ponto considerado em profundidade a uma dada posição de fonte e receptor coincidentes na superfície. O resultado final deste procedimento é uma imagem sísmica em profundidade dos refletores a partir do dado de múltipla cobertura.

The distribution of b-globin gene haplotypes was studied in 209 Amerindians from eight tribes of the Brazilian Amazon: Asurini from Xingú, Awá-Guajá, Parakanã, Urubú-Kaapór, Zoé, Kayapó (Xikrin from the Bacajá village), Katuena, and Tiriyó. Nine different haplotypes were found, two of which (n. 11 and 13) had not been previously identified in Brazilian indigenous populations. Haplotype 2 (+ - - - -) was the most common in all groups studied, with frequencies varying from 70% to 100%, followed by haplotype 6 (- + + - +), with frequencies between 7% and 18%. The frequency distribution of the b-globin gene haplotypes in the eighteen Brazilian Amerindian populations studied to date is characterized by a reduced number of haplotypes (average of 3.5) and low levels of heterozygosity and intrapopulational differentiation, with a single clearly predominant haplotype in most tribes (haplotype 2). The Parakanã, Urubú-Kaapór, Tiriyó and Xavante tribes constitute exceptions, presenting at least four haplotypes with relatively high frequencies. The closest genetic relationships were observed between the Brazilian and the Colombian Amerindians (Wayuu, Kamsa and Inga), and, to a lesser extent, with the Huichol of Mexico. North-American Amerindians are more differentiated and clearly separated from all other tribes, except the Xavante, from Brazil, and the Mapuche, from Argentina. A restricted pool of ancestral haplotypes may explain the low diversity observed among most present-day Brazilian and Colombian Amerindian groups, while interethnic admixture could be the most important factor to explain the high number of haplotypes and high levels of diversity observed in some South-American and most North-American tribes.
