To newly identify loci for age at natural menopause, we carried out a meta-analysis of 22 genome-wide association studies (GWAS) in 38,968 women of European descent, with replication in up to 14,435 women. In addition to four known loci, we identified 13 loci newly associated with age at natural menopause (at P < 5 x 10(-8)). Candidate genes located at these newly associated loci include genes implicated in DNA repair (EXO1, HELQ, UIMC1, FAM175A, FANCI, TLK1, POLG and PRIM1) and immune function (IL11, NLRP11 and PRRC2A (also known as BAT2)). Gene-set enrichment pathway analyses using the full GWAS data set identified exoDNase, NF-kappaB signaling and mitochondrial dysfunction as biological processes related to timing of menopause.

The ethylene biosynthesis inhibitor, 1-methylcyclopropene (1-MCP), has been commercially used to extend the storage life of European pear fruit and to allow shipment to distant markets. However, the influence of 1-MCP on the ability of fruit to ripen to an acceptable sensory quality has not been investigated in 'Bartlett' pear, one of the most aromatic of pear fruit. In the current study, early-, mid- and late-season 'Bartlett' pear fruit were treated with 0.6μLL-1 1-MCP at 0°C for 24h, 100μLL-1 ethylene at 20°C for 24h, or untreated before immediate transfer to 20°C for ripening until eating soft (13N firmness). Fruit from each treatment were subjected to objective and sensory quality evaluation once ripe. Sensory quality showed differences related to harvest maturity; the importance of harvest maturity depended on the treatment. Treatment with 1-MCP effectively slowed fruit ripening and therefore extended postharvest life, but also resulted in fully ripened pears with more desirable sensory traits, as compared with ethylene-treated and untreated pears. The enhanced sensory quality was related to higher sweetness, juiciness and pear aroma and reduced fermented aroma, gritty texture and tart taste. In addition, the sensory profiles of 1-MCP-treated pears were more stable among fruit from the three harvest dates than the ethylene-treated pears, which exhibited very different sensory profiles between early- and late-season fruits. © 2015 Elsevier B.V.

Quantification of pyridoxal-5´-phosphate (PLP) in biological samples is challenging due to the presence of endogenous PLP in matrices used for preparation of calibrators and quality control samples (QCs). Hence, we have developed an LC-MS/MS method for accurate and precise measurement of the concentrations of PLP in samples (20 µL) of human whole blood that addresses this issue by using a surrogate matrix and minimizing the matrix effect. We used a surrogate matrix comprising 2% bovine serum albumin (BSA) in phosphate buffer saline (PBS) for making calibrators, QCs and the concentrations were adjusted to include the endogenous PLP concentrations in the surrogate matrix according to the method of standard addition. PLP was separated from the other components of the sample matrix using protein precipitation with trichloroacetic acid 10% w/v. After centrifugation, supernatant were injected directly into the LC-MS/MS system. Calibration curves were linear and recovery was > 92%. QCs were accurate, precise, stable for four freeze-thaw cycles, and following storage at room temperature for 17h or at -80 °C for 3 months. There was no significant matrix effect using 9 different individual human blood samples. Our novel LC-MS/MS method has satisfied all of the criteria specified in the 2012 EMEA guideline on bioanalytical method validation.

The max-coloring problem is to compute a legal coloring of the vertices of a graph G = (V, E) with a non-negative weight function w on V such that Sigma(k)(i=1) max(v epsilon Ci) w(v(i)) is minimized, where C-1, ... , C-k are the various color classes. Max-coloring general graphs is as hard as the classical vertex coloring problem, a special case where vertices have unit weight. In fact, in some cases it can even be harder: for example, no polynomial time algorithm is known for max-coloring trees. In this paper we consider the problem of max-coloring paths and its generalization, max-coloring abroad class of trees and show it can be solved in time O(vertical bar V vertical bar+time for sorting the vertex weights). When vertex weights belong to R, we show a matching lower bound of Omega(vertical bar V vertical bar log vertical bar V vertical bar) in the algebraic computation tree model.

Hydrolysis of p-nitrophenyl-beta-D-glucoside by the beta-glucosidase of a thermophilic and cellulolytic fungus, Humicola insolens was stimulated by two-fold in the presence of high concentrations of beta-mercaptoethanol. This enzyme did not have any free sulfhydryl groups and high concentrations of beta-mercaptoethanol (5% v/v) reduced all of the three disulfide bonds present in the enzyme. In contrast, the hydrolysis of cellobiose and cellulose polymers was inhibited by 50% under the same conditions. Sodium dodecyl sulfate (1% w/v) even in combination with beta-mercaptoethanol did not show any significant effects on this enzyme. These unusual properties suggest that this enzyme may be of significant importance for understanding the structure of the enzyme.

Background and purpose of the study: Herbal enhancers compared to the synthetic ones have shown less toxis effects. Coumarins have been shown at concentrations inhibiting phospoliphase C-Y (Phc-Y) are able to enhance tight junction (TJ) permeability due to hyperpoalation of Zonolous Occludense-1 (ZO-1) proteins. The purpose of this study was to evaluate the influence of ethanolic extract of Angelica archengelica (AA-E) which contain coumarin on permeation of repaglinide across rat epidermis and on the tight junction plaque protein ZO-1 in HaCaT cells. Methods: Transepidermal water loss (TEWL) from the rat skin treated with different concentrations of AA-E was assessed by Tewameter. Scanning and Transmission Electron Microscopy (TEM) on were performed on AA-E treated rat skin portions. The possibility of AA-E influence on the architecture of tight junctions by adverse effect on the cytoplasmic ZO-1 in HaCaT cells was investigated. Finally, the systemic delivery of repaglinide from the optimized transdermal formulation was investigated in rats. Results: The permeation of repaglinide across excised rat epidermis was 7-fold higher in the presence of AA-E (5% w/v) as compared to propylene glycol:ethanol (7:3) mixture. The extract was found to perturb the lipid microconstituents in both excised and viable rat skin, although, the effect was less intense in the later. The enhanced permeation of repaglinide across rat epidermis excised after treatment with AA-E (5% w/v) for different periods was in concordance with the high TEWL values of similarly treated viable rat skin. Further, the observed increase in intercellular space, disordering of lipid structure and corneocyte detachment indicated considerable effect on the ultrastructure of rat epidermis. Treatment of HaCaT cell line with AA-E (0.16% w/v) for 6 hrs influenced ZO-1 as evidenced by reduced immunofluorescence of anti-TJP1 (ZO-1) antibody in Confocal Laser Scanning Microscopy studies (CLSM) studies. The plasma concentration of repaglinide from transdermal formulation was maintained higher and for longer time as compared to oral administration of repaglinide. Major conclusion: Results suggest the overwhelming influence of Angelica archengelica in enhancing the percutaneous permeation of repaglinide to be mediated through perturbation of skin lipids and tight junction protein (ZO-1).

A wide variety of novel compounds obtained by combining two types of known organogelators, viz., bile acid alkyl amides and pyrene alkanoic acids, were synthesized and screened for their gelation ability. The 3 alpha esters of 1-pyrene butyric acid (PBA) of alkylamides of deoxycholic acid (DCA) turned out to be effective in the gel formation with many organic solvents although the gelation has to be triggered by the addition of a charge transfer (CT) agent 2,4,7-trinitrofluorenone (TNF). The special feature of these molecules is that the organogelation is achieved only after derivatizing the acid moiety of the 1-pyrenealkanoic acids. Additionally, the gelation properties can be fine-tuned by inserting different functional groups at the bile acid side chain. The gels obtained are deep red in colour and optically transparent up to 2% w/v. The SEM studies of the obtained xerogels revealed bundled rod-like morphology without specialized branching.

The gelation property of these derivatives is a function of the bile acid moiety, the spacer and the fluoroalkyl chain length. By varying these parameters, gels were obtained in aromatic hydrocarbons, DMSO and DMSO/DMF-H(2)O mixtures of different proportions. Several derivatives of deoxycholic and lithocholic acids were found to be efficient organogelators, while the reported bile-acid based organogelators are mostly derived from the cholic acid moiety. The efficient gelators among these compounds formed gels well below 1.0% (w/v) and hence they can be termed as supergelators. The mechanical properties of these gels could be modulated by changing either the bile acid moiety or by varying the length of the fluoroalkyl segment. The presence of CO(2)-philic perfluoroalkyl groups is also expected to enhance their solubility in supercritical CO(2) and hence these compounds are promising candidates for making aerogels.

Organic acids are important constituents of fruit juices. They render tartness, flavour and specific taste to fruit juices. Shelf life and stability of fruit juices are important factors, which determine their nutritional quality and freshness. In this view, the effect of storage on the concentration of organic acids in commercially packed fruit juices is studied by reverse phase high performance liquid chromatography (RP-HPLC). Ten packed fruit juices from two different brands are stored at 30 C for 24, 48 and 72 hours. A reverse phase high performance liquid chromatographic method is used to determine the concentration of oxalic, tartaric, malic, ascorbic and citric acid in the fruit juices during storage. The chromatographic analysis of organic acids is carried out using mobile phase 0.5% (w/v) ammonium dihydrogen orthophosphate buffer (pH 2.8) on C18 column with UV-Vis detector. The results show that the concentration of organic acids generally decreases in juices under study with the increase in storage time. All the fruit juices belonging to tropicana brand underwent less organic acid degradation in comparison to juices of real brand. Orange fruit juice is found to be least stable among the juices under study, after the span of 72 hours. Amongst all the organic acids under investigation minimum stability is shown by ascorbic acid followed by malic and citric acid.

A new class of steroid dimers (bile acid derivatives) linked through ester functionalities were synthesized, which gelled various aromatic solvents. The organogels formed by the three dimeric ester molecules showed birefringent textures and fibrous nature by polarizing optical microscopy and scanning electron microscopy, respectively. A detailed rheological study was performed to estimate the mechanical strengths of two sets of organogels. In these systems, the storage modulus varied in the range of 0.8-3.5 X 10(4) at 1% w/v of the organogelators. The exponents of scaling of the storage modulus and yield stress of the two systems agreed well with those expected for viscoelastic soft colloidal gels with fibrillar flocs. The nanofibers in the organogel were utilized to engineer gold nanoparticles of different sizes and shapes and generate new gel-nanoparticle hybrid materials.

Hydrogen peroxide (H2O2) level in biological samples is used as an important index in various studies. Quantification of H2O2 level in tissue fractions in presence of H2O2 metabolizing enzymes may always provide an incorrect result. A modification is proposed for the spectrofluorimetric determination of H2O2 in homovanillic acid (HVA) oxidation method. The modification was included to precipitate biological samples with cold trichloroacetic acid (TCA, 5% w/v) followed by its neutralization with K2HPO4 before the fluorimetric estimation of H2O2 is performed. TCA was used to precipitate the protein portions contained in the tissue fractions. After employing the above modification, it was observed that H2O2 content in tissue samples was >= 2 fold higher than the content observed in unmodified method. Minimum 2 h incubation of samples in reaction mixture was required for completion of the reaction. The stability of the HVA dimer as reaction product was found to be > 12 h. The method was validated by using known concentrations of H2O2 and catalase enzyme that quenches H2O2 as substrate. This method can be used efficiently to determine more accurate tissue H2O2 level without using internal standard and multiple samples can be processed at a time with additional low cost reagents such as TCA and K2HPO4.

[EN] The aims of this work were (i) to evaluate the potential of nanostructured lipid carriers (NLCs) as a tool to 24 enhance the oral bioavailability of poorly soluble compounds using saquinavir (SQV), a BCS class IV drug 25 and P-gp substrate as a model drug, and (ii) to study NLC transport mechanisms across the intestinal barrier. 26 Three different NLC formulations were evaluated. SQV transport across Caco-2 monolayers was enhanced up 27 to 3.5-fold by NLCs compared to SQV suspension. M cells did not enhance the transport of NLCs loaded with 28 SQV. The size and amount of surfactant in the NLCs influenced SQV's permeability, the transcytosis pathway 29 and the efflux of SQV by P-gp. An NLC of size 247 nm and 1.5% (w/v) surfactant content circumvented P-gp 30 efflux and used both caveolae- and clathrin-mediated transcytosis, in contrast to the other NLC formulations, 31 which used only caveolae-mediated transcytosis. By modifying critical physicochemical parameters of the 32 NLC formulation, we were thus able to overcome the P-gp drug efflux and alter the transcytosis mechanism 33 of the nanoparticles. These findings support the use of NLCs approaches for oral delivery of poorly 34 water-soluble P-gp substrates.

A biossorção é o termo aplicado à tecnologia de adsorção de íons metálicos em solução, por meio de materiais de natureza biológica inativa, comparável à adsorção utilizando-se adsorventes convencionais. A biossorção depende da temperatura, pH, concentração inicial do íon metálico, e tipo de biosorvente. O objetivo principal dessa dissertação é estudar e comparar os processos de adsorção de cobre iônico a partir de soluções aquosas utilizando Sargassum sp., alginato de cálcio úmido e desidratado e car£o ativo em pó e granular, em regime de batelada e contínuo. O alginato de cálcio foi preparado atra©s de uma solução 4% (m/v) de alginato de sódio gotejada em solução 37% (m/v) de cloreto de cálcio dihidratado. O alginato de cálcio apresentou uma umidade de 89% após secagem em estufa a 100C por 24 horas. Os parâmetros da biossorção foram fixados em: a dose de adsorventes de 2 g/L; a temperatura em 30C; pH da solução em 5,0; e velocidade de rotação da chapa rotatória em 150 rpm. Para os estudos de cinética e equilíbrio, os ensaios foram feitos em regime de batelada. Nos ensaios cinéticos para duas concentrações medias (0,16 e 3,15 mmol/L) variou-se o tempo de contato (1 a 120 min) para se atingir o equilíbrio. Nos ensaios de equilíbrio variou-se as concentrações (0,16 a 15,72 mmol/L) utilizando o tempo de equilíbrio determinado nos ensaios cinéticos. Foram utilizadas duas modelagens cinéticas (a de pseudo-primeira ordem e a de segunda ordem) e duas modelagens do equilíbrio (Langmuir e Freundlich). O modelo cinético de segunda ordem ajustou melhor os resultados. O tempo de equilíbrio para adsorção do cobre foi de 60 minutos para Sargassum sp. e pellets de alginatos de cálcio úmido e desidratado. Para os carµes ativos os tempos de equilíbrio para a adsorção do cobre foram mais rápidos, mas a capacidade de remoção do cobre foram muito baixas. Com base nos resultados desfavoráveis obtidos para os carµes ativos eles foram descartados para se prosseguir com os ensaios de equilíbrio. A isoterma de Freundlich melhor ajusta os dados experimentais para Sargassum sp. e alginato de cálcio úmido. A capacidade de adsorção máxima calculada pelo modelo de Freundlich: na concentração de equilíbrio de 11,66 0,06 mmol/L foi de 1,97 0,07 mmol/g para Sargassum sp.; e na concentração de equilíbrio de 12,12 0,03 mmol/L foi de 1,69 0,04 mmol/g para alginato de cálcio úmido. O processo de biossorção em regime contínuo, com alturas de leito variável de 10 a 40 cm, teve melhor desempenho com a altura de 40 cm. Em regime de batelada, o desempenho da Sargassum sp. foi superior ao dos alginatos de cálcio (úmido melhor que o desidratado), que por sua vez foram superiores ao desempenho dos carµes ativos em pó e granular (em pó melhor que o granular). O sistema contínuo com concentração inicial de cobre de 8,5 mmol/L deve ser operado com altura de leito igual ou superior a 40 cm, ou com sistemas multicolunas para soluções mais concentradas

A matriz energética mundial é baseada em fontes fósseis e reno¡veis. No Brasil, o bioetanol é gerado principalmente a partir da cana-de-açúcar. Resíduos agroindustriais (fontes celulósicas ou amiláceas) despontam como biomassas alternativas à cana-de-açúcar, para aumentar a competitividade deste combustível reno¡vel frente aos de origem fóssil e também favorecer a sustentabilidade e a segurança alimentar e energética, pois são ricos em polissacarídeos não diretamente fermentescíveis, abundantes (problema ambiental) e apresentam baixo valor comercial. O farelo de mandioca é um exemplo de resíduo sólido gerado na produção de fécula (amido) e farinha de mandioca que ainda contém, em média, 75% de amido. Consequentemente, deve ser previamente hidrolisado e posteriormente fermentado por leveduras do gênero Saccharomyces para gerar etanol. O objetivo deste estudo foi produzir bioetanol a partir de hidrolisados enzimáticos de farelo de mandioca, usando levedura álcool resistente (AR). Primeiramente, a concentração de açúcares obtida a partir da hidrólise enzimática foi verificada atra©s de um planejamento fatorial completo (24), com triplicata no ponto central, a fim de investigar a influência dos seguintes fatores na hidrólise: concentração de α-amilase (Termamyl 2X), tempo de liquefação, concentração de glucoamilase (AMG 300L) e o tempo sacarificação. A condição de hidrólise mais favorável foi a do ensaio com 0,517 mL de AMG/g amido, 0,270 mL de Termamyl/g amido, 1h de tempo de liquefação e 2h de tempo de sacarificação. O caldo resultante da condição escolhida alcançou altas concentrações de glicose (160 g/L). Os ensaios de fermentação alcoólica foram realizados em duplicata em biorreator de 3L, em regime de batelada, a 30C, 100 rpm e pH 5,5. Cerca de 3 g/L (massa seca) de uma linhagem de levedura álcool tolerante, Saccharomyces cerevisiae Hansen BY4741, crescida por 12h em meio YEDP (2% de glicose) foram usados como inóculo. O mosto consistiu de um litro de hidrolisado (160 g/L de glicose) fortificado com extrato de levedura (1%) e peptona de carne (1%), além da adição de um antiespumante (Tween 80) na concentração de 0,05% (m/v). Em 30 horas de fermentação, a média da concentração de etanol obtida foi de 65 g/L. A eficiência foi de 87,6% e o rendimento e a produtividade foram 0,448 e 2,16 g/L.h, respectivamente.
