A short-term whole-skin organ culture model has been established to enable the investigation of cell cycle perturbations in epidermal layer cells following exposure to ultraviolet radiation (UVR). This model affords the opportunity to manipulate the growth and nutrient conditions, and to perform detailed biochemical and immunohistochemical analysis of skin cells in their normal epidermal layer microenvironment. The use of this model is described in this chapter.

Caucasian renal transplant recipients from Queensland, Australia have the highest non-melanoma skin cancer (NMSC) risk worldwide. Although ultraviolet light (UVR) exposure is critical, genetic factors also appear important. We and others have shown that polymorphism in the glutathione S-transferases (GST) is associated with NMSC in UK recipients. However, the effect of high UVR exposure and differences in immunosuppressive regimen on these associations is unknown. In this study, we examined allelism in GSTM1, GSTM3, GSTT1 and GSTP1 in 361 Queensland renal transplant recipients. Data on squamous (SCC) and basal cell carcinoma (BCC), UVR/tobacco exposure and genotype were obtained. Associations with both NMSC risk and numbers were examined using logistic and negative binomial regression, respectively. In the total group, GSTM1 AB [P = 0.049, rate ratio (RR) = 0.23] and GSTM3 AA (P = 0.015, RR = 0.50) were associated with fewer SCC. Recipients were then stratified by prednisolone dose (less than or equal to7 versus >7 mg/day). In the low-dose group, GSTT1 null (P = 0.006, RR = 0.20) and GSTP1 Val/Val (P = 0.021, RR = 0.20) were associated with SCC numbers. In contrast, in the high-dose group, GSTM1 AB (P = 0.009, RR = 0.05), GSTM3 AB (P = 0.042, RR = 2.29) and BB (P = 0.014, RR = 5.31) and GSTP1 Val/Val (P = 0.036, RR = 2.98) were associated with SCC numbers. GSTM1 AB (P = 0.016) and GSTP1 Val/Val (P = 0.046) were also associated with fewer BCC in this group. GSTP1 associations were strongest in recipients with lower UVR/tobacco exposure. The data confirm our UK findings, suggesting that protection against UVR-induced oxidative stress is important in NMSC development in recipients, but that this effect depends on the immunosuppressant regimen.

Human melanoma susceptibility is often characterized by germ-line inactivating CDKN2A (INK4A/ARF) mutations, or mutations that activate CDK4 by preventing its binding to and inhibition by INK4A. We have previously shown that a single neonatal UV radiation (UVR) dose delivered to mice that carry melanocyte-specific activation of Hras (TPras) increases melanoma penetrance from 0% to 57%. Here, we report that activated Cdk4 cooperates with activated Hras to enhance susceptibility to melanoma in mice. Whereas UVR treatment failed to induce melanomas in Cdk4(R24C/R24C) mice, it greatly increased the penetrance and decreased the age of onset of melanoma development in Cdk4(R24C/R24C)/TPras animals compared with TPras alone. This increased penetrance was dependent on the threshold of Cdk4 activation as Cdk4(R24C/+)/TPras animals did not show an increase in UVR-induced melanoma penetrance compared with TPras alone. In addition, Cdk4(R24C/R24C)/TPras mice invariably developed multiple lesions, which occurred rarely in TPras mice. These results indicate that germ-line defects abrogating the pRb pathway may enhance UVR-induced melanoma. TPras and Cdk4(R24C/R24C)/TPras tumors were comparable histopathologically but the latter were larger and more aggressive and cultured cells derived from such melanomas were also larger and had higher levels of nuclear atypia. Moreover, the melanomas in Cdk4(R24C/R24C)/TPras mice, but not in TPras mice, readily metastasized to regional lymph nodes. Thus, it seems that in the mouse, Hras activation initiates UVR-induced melanoma development whereas the cell cycle defect introduced by mutant Cdk4 contributes to tumor progression, producing more aggressive, metastatic tumors.

The application of an antiserum to ultraviolet radiation (UVR)-damaged DNA is presented. A novel experimental system was employed to ascertain the limits of detection for this antiserum. Using a DNA standard containing a known amount of dimer, the limits of detection were found to be 0.9 fmol of dimer. This was compared to a limit of 20-50 fmol dimer using gas chromatography-mass spectrometry (GC-MS). Induction of thymine dimers in DNA following UVR exposure, as assessed using this antiserum in an enzyme-linked immunosorbent assay (ELISA), was compared with GC-MS measurements. The ELISA method successfully demonstrated the induction of lesions in DNA irradiated either with UVC or UVB, although despite high sensitivity, no discernible binding was seen to UVA-irradiated DNA. The antiserum was also shown to be applicable to immunocytochemistry, localising damage in the nuclei of UVR exposed keratinocytes in culture. The ability of the antiserum to detect DNA damage in skin biopsies of individuals exposed to sub-erythemal doses of UVR was also demonstrated. Moreover, the subsequent removal of this damage, as evidenced by a reduction in antiserum staining, was noted in sections of biopsies taken in the hours following irradiation. © 2003 Elsevier B.V. While knowledge about standardization of skin protection against ultraviolet radiation (UVR) has progressed over the past few decades, there is no uniform and generally accepted standardized measurement for UV eye protection. The literature provides solid evidence that UV can induce considerable damage to structures of the eye. As well as damaging the eyelids and periorbital skin, chronic UV exposure may also affect the conjunctiva and lens. Clinically, this damage can manifest as skin cancer and premature skin ageing as well as the development of pterygia and premature cortical cataracts. Modern eye protection, used daily, offers the opportunity to prevent these adverse sequelae of lifelong UV exposure. A standardized, reliable and comprehensive label for consumers and professionals is currently lacking. In this review we (i) summarize the existing literature about UV radiation-induced damage to the eye and surrounding skin; (ii) review the recent technological advances in UV protection by means of lenses; (iii) review the definition of the Eye-Sun Protection Factor (E-SPF®), which describes the intrinsic UV protection properties of lenses and lens coating materials based on their capacity to absorb or reflect UV radiation; and (iv) propose a strategy for establishing the biological relevance of the E-SPF. © 2013 John Wiley & Sons A/S.

Previous studies have shown that increasing atmospheric CO2 concentrations affect calcification in some planktonic and macroalgal calcifiers due to the changed carbonate chemistry of seawater. However, little is known regarding how calcifying algae respond to solar UV radiation (UVR, UVA+UVB, 280-400 nm). UVR may act synergistically, antagonistically or independently with ocean acidification (high CO2/low pH of seawater) to affect their calcification processes. We cultured the articulated coralline alga Corallina sessilis Yendo at 380 ppmv (low) and 1000 ppmv (high) CO2 levels while exposing the alga to solar radiation treatments with or without UVR. The presence of UVR inhibited the growth, photosynthetic O2evolution and calcification rates by13%, 6% and 3% in the low and by 47%, 20% and 8% in the high CO2 concentrations, respectively, reflecting a synergistic effect of CO2 enrichment with UVR. UVR induced significant decline of pH in the CO2-enriched cultures. The contents of key photosynthetic pigments, chlorophyll a and phycobiliproteins decreased, while UV-absorptivity increased under the highpCO2/low pH condition. Nevertheless, UV-induced inhibition of photosynthesis increased when the ratio of particulate inorganic carbon/particulate organic carbon decreased under the influence of CO2-acidified seawater, suggesting that the calcified layer played a UV-protective role. Both UVA and UVB negatively impacted photosynthesis and calcification, but the inhibition caused by UVB was about 2.5-2.6 times that caused by UVA. The results imply that coralline algae suffer from more damage caused by UVB as they calcify less and less with progressing ocean acidification.

We carried out short term pCO2/pH perturbation experiments in the coastal waters of the South China Sea to evaluate the combined effects of seawater acidification (low pH/high pCO2) and solar UV radiation (UVR, 280-400 nm) on photosynthetic carbon fixation of phytoplankton assemblages. Under photosynthetically active radiation (PAR) alone treatments, reduced pCO2 (190 ppmv) with increased pH resulted in a significant decrease in the photosynthetic carbon fixation rate (about 23%), while enriched pCO2 (700 ppmv) with lowered pH had no significant effect on the photosynthetic performance compared to the ambient level. The apparent photosynthetic efficiency decreased under the reduced pCO2 level, probably due to C-limitation as well as energy being diverged for up-regulation of carbon concentrating mechanisms (CCMs). In the presence of UVR, both UV-A and UV-B caused photosynthetic inhibition, though UV-A appeared to enhance the photosynthetic efficiency under lower PAR levels. UV-B caused less inhibition of photosynthesis under the reduced pCO2 level, probably because of its contribution to the inorganic carbon (Ci)-acquisition processes. Under the seawater acidification conditions (enriched pCO2), both UV-A and UV-B reduced the photosynthetic carbon fixation to higher extents compared to the ambient pCO2 conditions. We conclude that solar UV and seawater acidification could synergistically inhibit photosynthesis.

The calcifying phytoplankton species, coccolithophores, have their calcified coccoliths around the cells, however, their physiological roles are still unknown. Here, we hypothesized that the coccoliths may play a certain role in reducing solar UV radiation (UVR, 280-400 nm) and protect the cells from being harmed. Cells of Emiliania huxleyi with different thicknesses of the coccoliths were obtained by culturing them at different levels of dissolved inorganic carbon and their photophysiological responses to UVR were investigated. Although increased dissolved inorganic carbon decreased the specific growth rate, the increased coccolith thickness significantly ameliorated the photoinhibition of PSII photochemical efficiency caused by UVR. Increase by 91% in the coccolith thickness led to 35% increase of the PSII yield and 22% decrease of the photoinhibition of the effective quantum yield by UVR. The coccolith cover reduced more UVA (320-400 nm) than UVB (280-315 nm), leading to less inhibition per energy at the UV-A band.

The combination of elevated CO2 and the increased acidity in surface oceans is likely to have an impact on photosynthesis via its effects on inorganic carbon speciation and on the overall energetics of phytoplankton. Exposure to UV radiation (UVR) may also have a role in the response to elevated CO2 and acidification, due to the fact that UVR may variously impact on photosynthesis and because of the energy demand of UVR defense. The cell may gain energy by down-regulating the CO2 concentrating mechanism, which may lead to a greater ability to cope with UVR and/or higher growth rates. In order to clarify the interplay of cell responses to increasing CO2 and UVR, we investigated the photosynthetic response of the marine and estuarine diatom Cylindrotheca closterium f. minutissima cultured at either 390 (ambient) or 800 (elevated) ppmv CO2, while exposed to solar radiation with or without UVR (UVR, 280-400 nm). After a 6 day acclimation period, the growth rate of cells was little affected by elevated CO2 and no obvious correlation with the radiation dose (for both PAR and PAR + UV treatments) could be detected. However, the relative electron transport rate was reduced and was more sensitive to UVR in cells main - tained at elevated CO2 as compared to cells cultured at ambient CO2. The CO2 concentrating mechanism was down regulated at 800 ppmv CO2, but was apparently not completely switched off. These data are discussed with respect to their significance in the context of global climate change.

Este proyecto de investigación construye y evalúa la asignación de activos para el portafolio de Pensión Obligatoria de Retiro Programado, el cual atiende los retiros a los que un pensionado tiene derecho a través de su mesada pensional, utilizando el modelo de Frontera Eficiente de Markowitz, en combinación con la teoría de Momentum -- Para la ejecución del modelo se determinaron los activos de inversión admisibles en el Régimen de Inversiones -- Posteriormente, se construyen las matrices de rentabilidades, de restricciones, de varianzas y covarianzas, las cuales constituyen los insumos para ejecutar el modelo de optimización de portafolios de Markowitz -- A continuación, se realiza la selección de los portafolios obtenidos, teniendo en cuenta el nivel de volatilidad que el portafolio de Obligatorias Retiro Programado debe presentar; lo anterior, con el fin de cumplir con el objetivo de preservación del capital en la cuenta individual del pensionado, de manera que se pueda atender, de acuerdo a su esperanza de vida y la de sus beneficiarios, el pago de las mesadas pensionales que le correspondan -- El resultado obtenido corresponde a una asignación, en gran parte, en activos de Renta Fija expedidos por el Gobierno Nacional (TES), tanto en tasa fija como en tasa indexada a la UVR -- Adicionalmente, el modelo de optimización sugiere participaciones en activos de renta variable y, particularmente, no asigna recursos representativos en títulos de deuda privada indexados al IPC -- Esta investigación puede ser útil al momento de diseñar un portafolio base para Obligatorias Retiro Programado que, bajo una administración pasiva, permita cumplir el objetivo de otorgar a los pensionados una mesada para satisfacer las necesidades básicas

En el mundo de las inversiones y las finanzas, las tasas de interés juegan un papel muy importante, ya que es por medio de estas que el inversionista mide y compara la rentabilidad de diferentes proyectos de inversión, con el fin de escoger la mejor alternativa. Esta obra contiene los fundamentos para el estudio, análisis y comprensión del manejo financiero de las tasas de interés. Los conceptos se exponen de una forma clara y sencilla, de tal modo que le permita al lector el aprendizaje rápido, aun sin tener conocimientos sobre finanzas, ya que se exponen con matemáticas simples o convencionales. Cada uno de los temas tratados va acompañado de ejercicios de común ocurrencia en el mercado financiero y de capitales, tanto en el contexto nacional como internacional. El texto está acompañado de la aplicación de algunos temas en Excel, al igual que de un simulador interactivo, el cual le permite al lector comprobar y resolver de manera rápida pequeños problemas del acontecer financiero en el campo de las tasas de interés. Al terminar de leer y comprender el texto, el lector tendrá la capacidad de analizar, interpretar y aplicar a la vida cotidiana los diferentes temas concernientes a las tasas de interés. Podrá solucionar de manera ágil problemas financieros presentes a la hora de adquirir un crédito o de realizar una inversión, sea en moneda nacional o extranjera. El lector también tendrá la capacidad de comprender los diferentes conceptos dentro del tópico de las tasas de interés: conversión de tasas, DTF, IPC, UVR, tasas futuras implícitas, netas, reales, devaluación, 4 por mil, TRM, entre otros.

Souvent conçues comme le vecteur de changements majeurs, les pratiques numériques constituent une occasion de mieux comprendre le fait littéraire en faisant apparaître de manière plus explicite que jamais des aspects ontologiques qui, en tant que tels, ont une valeur atemporelle. En particulier, et c’est l’objet de cet article, le fait numérique donne l’occasion de réinvestir une problématique qui parcourt l’ensemble de la réflexion sur le statut de la littérature depuis Platon et Aristote : celle du rapport entre littérature et réalité, dont Sartre et Derrida avaient déjà œuvré à déconstruire l’opposition au XXe siècle. Selon nous, le numérique souligne l’absence de séparation entre symbolique et non-symbolique, nous empêchant de penser une rupture entre imaginaire et réel. Pour rendre compte de cette structure, nous nous appuierons sur le concept d’éditorialisation, qui vient désigner l’ensemble des dispositifs permettant la production de contenus dans l’espace numérique en tenant compte de la fusion entre espace numérique et espace non numérique. À travers des exemples littéraires – Traque Traces de Cécile Portier et Laisse venir d’Anne Savelli et Pierre Ménard – nous démontrerons comment la littérature participe aujourd’hui à l’éditorialisation du monde, enterrant ainsi définitivement le dualisme imaginaire-réel pour lui préférer une structure anamorphique.

À travers des exemples littéraires – Traque Traces de Cécile Portier et Laisse venir d’Anne Savelli et Pierre Ménard – nous démontrerons comment la littérature participe aujourd’hui à l’éditorialisation du monde, enterrant ainsi définitivement le dualisme imaginaire-réel pour lui préférer une structure anamorphique.
