The rhizosphere constitutes a complex niche that may be exploited by a wide variety of bacteria. Bacterium-plant interactions in this niche can be influenced by factors such as the expression of heterologous genes in the plant. The objective of this work was to describe the bacterial communities associated with the rhizosphere and rhizoplane regions of tobacco plants, and to compare communities from transgenic tobacco lines (CAB1, CAB2 and TRP) with those found in wild-type (WT) plants. Samples were collected at two stages of plant development, the vegetative and flowering stages (1 and 3 months after germination). The diversity of the culturable microbial community was assessed by isolation and further characterization of isolates by amplified ribosomal RNA gene restriction analysis (ARDRA) and 16S rRNA sequencing. These analyses revealed the presence of fairly common rhizosphere organisms with the main groups Alphaproteobacteria, Betaproteobacteria, Actinobacteria and Bacilli. Analysis of the total bacterial communities using PCR-DGGE (denaturing gradient gel electrophoresis) revealed that shifts in bacterial communities occurred during early plant development, but the reestablishment of original community structure was observed over time. The effects were smaller in rhizosphere than in rhizoplane samples, where selection of specific bacterial groups by the different plant lines was demonstrated. Clustering patterns and principal components analysis (PCA) were used to distinguish the plant lines according to the fingerprint of their associated bacterial communities. Bands differentially detected in plant lines were found to be affiliated with the genera Pantoea, Bacillus and Burkholderia in WT, CAB and TRP plants, respectively. The data revealed that, although rhizosphere/rhizoplane microbial communities can be affected by the cultivation of transgenic plants, soil resilience may be able to restore the original bacterial diversity after one cycle of plant cultivation.

There is concern that the use of lower quality phosphate rock can result in elevated amounts of Fe-Al-P water-insoluble compounds in fertilizers and, consequently, low agronomic effectiveness. Therefore, studies were conducted to evaluate the effect of some of these compounds on plant growth. Four commercial superphosphates varying in chemical composition (two single and two triple superphosphates) were selected for the study. Fertilizer impurities were collected as water-insoluble residues by washing each P source with deionized water. A modal analysis, based primarily on elemental chemical analysis and x-ray diffractometry, was used to estimate the chemical composition of each P source. Water-soluble monocalcium phosphate (MCP) and the water-leached fertilizer residues were prepared to give a range of fertilizers in terms of water-soluble phosphorus (WSP) (0-100% of the available P as MCP). The water-leached fractions, MCP, and the mixtures of MCP with water-leached fractions were applied to supply 40 mg available P kg(1) to a thermic Rhodic Kanhapludult with pH values of 5.2 +/- 0.05 (unlimed) and 6.4 +/- 0.08 (limed). Wheat (Triticum aestivum L.) grown in a greenhouse for 101 d served as the test crop. The requirement for WSP was source and pH dependent. At a soil pH of 5.2, the fertilizers required 73 to 95% WSP to reach the maximum dry-matter yield, while they required 60 to 86% WSP at pH 6.4. To reach 90% of the maximum yield, all superphosphate fertilizers required <50% WSP. These results show that it is not always necessary to have high water solubility as required by legislation in many countries.

Italian ryegrass resistance to diclofop has been documented in several countries, including the United States. The purpose of this research was to screen selected putative resistant populations of Italian ryegrass for resistance to the acetyl-CoA carboxylase (ACCase)-inhibiting herbicides diclofop and pinoxaden and the acetolactate synthase (ALS)-inhibiting herbicides imazamox, pyroxsulam, and mesosulfuron in the greenhouse and to use field experiments to develop herbicide programs for Italian ryegrass control. Resistance to diclofop was confirmed in eight populations from Tennessee. These eight populations did not show cross-resistance to pinoxaden. One additional population (R1) from Union County, North Carolina, was found to be resistant to both diclofop and pinoxaden. The level of resistance to pinoxaden of the R1 population was 15 times that of the susceptible population. No resistance was confirmed to any of the ALS-inhibiting herbicides examined in this research. Field experiments demonstrated PRE Italian ryegrass control with chlorsulfuron (71 to 94%) and flufenacet + metribuzin (84 to 96%). Italian ryegrass control with pendimethalin applied PRE or delayed preemergence (DPRE) was variable (0 to 85%). POST control of Italian ryegrass was acceptable with pinoxaden, mesosulfuron, flufenacet + metribuzin, and chlorsulfuron + flucarbazone (> 80%). Application timing and herbicide treatment had no effect on wheat yield, except for diclofop and pendimethalin treatments, in which uncontrolled Italian ryegrass reduced wheat yield.

Aluminum (Al) toxicity is a major worldwide agricultural problem. At low pH, Al speciates into the soluble and phyto-toxic form Al(3+), inhibiting the root growth and affecting plant development. In Brazil, agriculture in acidic soils with elevated concentration of Al has significantly increased in the last decades. Therefore, in order to achieve efficient agriculture practices, the selection of plant cultivars with improved Al resistance has become crucial in this type of soils. In this work we have evaluated the Al resistance of six genotypes of grapevine rootstocks. The grapevine hardwood cuttings were grown in nutrient solution in the absence and presence of 250 and 500 mu M Al at pH 4.2. The phenotypic indexes of relative root growth, fresh and dry root weight, root area, hematoxylin staining profile, and Al content were evaluated for all six genotypes. These phenotypic indexes allowed us to identify the `Kober 5BB`, `Gravesac`, `Paulsen 1103`, and `IAC 766` grapevine rootstocks genotypes as the ones with the highest resistance to Al. Likewise, `IAC 572` and `R110` genotypes were the most Al-sensitive cultivars. We evaluated the root organic acid exudation profile in the most Al-resistant (`Kober 5BB`) and most Al-sensitive (`R110`) in plantlets cultivated in vitro in the absence and presence of 100, 200, and 400 mu M of Al. Among several compounds detected, citrate was the only organic acid related to the Al resistance phenotype observed in the `Kober 5BB` genotype. The high constitutive citrate exudation observed in `Kober 5BB` strongly suggests that exudation of this particular organic acid may impart Al-resistance/a melioration in grapevine.

Plants have multiple potassium (K+) uptake and efflux mechanisms that are expressed throughout plant tissues to fulfill different physiological functions. Several different classes of K+ channels and carriers have been identified at the molecular level in plants. K+ transporters of the HKT1 superfamily have been cloned from wheat (Triticum aestivum), Arabidopsis, and Eucalyptus camaldulensis. The functional characteristics as well as the primary structure of these transporters are diverse with orthologues found in bacterial and fungal genomes. In this report, we provide a detailed characterization of the functional characteristics, as expressed in Xenopus laevis oocytes, of two cDNAs isolated from E. camaldulensis that encode proteins belonging to the HKT1 superfamily of K+/Na+ transporters. The transport of K+ in EcHKT-expressing oocytes is enhanced by Na+, but K+ was also transported in the absence of Na+. Na+ is transported in the absence of K+ as has been demonstrated for HKT1 and AtHKT1. Overall, the E. camaldulensis transporters show some similarities and differences in ionic selectivity to HKT1 and AtHKT1. One striking difference between HKT1 and EcHKT is the sensitivity to changes in the external osmolarity of the solution. Hypotonic solutions increased EcHKT induced currents in oocytes by 100% as compared with no increased current in HKT1 expressing or uninjected oocytes. These osmotically sensitive currents were not enhanced by voltage and may mediate water flux. The physiological function of these osmotically induced increases in currents may be related to the ecological niches that E. camaldulensis inhabits, which are periodically flooded. Therefore, the osmosensing function of EcHKT may provide this species with a competitive advantage in maintaining K+ homeostasis under certain conditions.

Pearl millet landraces from Rajasthan, India, yield significantly less than improved cultivars under optimum growing conditions, but not under stressed conditions. To successfully develop a simulation model for pearl millet, capable of capturing such genotype x environment (G x E) interactions for grain yield, we need to understand the causes of the observed yield interaction. The aim of this paper is to quantify the key parameters that determine the accumulation and partitioning of biomass: the,light extinction coefficient, radiation use efficiency (RUE), pattern of dry matter allocation to the leaf blades, the determination of grain number, and the rate and duration of dry matter accumulation into individual grains. We used data on improved cultivars and landraces, obtained from both published and unpublished sources collected at ICRISAT, Patancheru, India. Where possible, the effects of cultivar and axis (main shoot vs. tillers) on these parameters were analysed, as previous research suggested that G x E interactions for grain yield are associated with differences in tillering habit. Our results indicated there were no cultivar differences in extinction coefficient, RUE, and biomass partitioning before anthesis, and differences between axes in biomass partitioning were negligible. This indicates there was no basis for cultivar differences in the potential grain yield. Landraces, however, produced consistently less grain yield for a given rate of dry matter accumulation at anthesis than did improved cultivars. This was caused by a combination of low grain number and small grain size. The latter was predominantly due to a lower grain growth rate, as genotypic differences in the duration of grain filling were relatively small. Main shoot and tillers also had a similar duration of grain filling. The low grain yield of the landraces was associated with profuse nodal tillering, supporting the hypothesis that grain yield was below the potential yield that could be supported by assimilate availability. A field experiment was conducted to study the effect of micronutrients, zinc (Zn), copper (Cu), iron (Fe), manganese (Mn), boron (13) and a commercial fritted micronutrient product called Zarzameen, on the yield and the yield components of wheat (Triticum aestivum L.), in the Peshawar valley, Pakistan. Different combinations of Zn, Cu. Fe. Mn, B, and Zarzameen were applied at the rate of 4.0, 2.0, 5.0, 2.0, 1.0 kg ha(-1) and 1.0 kg ha(-1), respectively, along with a basal dose of 100 kg ha(-1) nitrogen(N), 75 kg ha(-1) phosphorus (P) and 50 kg ha(-1) potassium (K). The fertilizer treatments (macro- and micronutrients) increased wheat dry matter, grain yield, and straw yield significantly over an unfertilized control. Soil tests for B and Zn were increased both at boot and harvesting stage, and Fe at boot stage, with the addition of micronutrients. Plants without B had showed classical B deficiency symptoms at grain formation stage, but not at vegetative stage. Boron concentration in the dry matter of wheat plants increased with the addition of the B fertilizer in the soil. Boron deficiency was not observed in plants containing >4 mg B kg(-1) at the boot stage, or in soils containing > 1.4 mg kg(-1) hot water soluble B.

Caustis blakei is an attractive cut foliage plant harvested from the wild in Australia and marketed under the name of koala fern. Previous attempts to propagate large numbers of this plant have been unsuccessful. The effect of four light irradiances on organogenesis from compact and friable callus of C. blakei was studied for 21 wk. Both callus types produced numerous primordial shoots but many failed to develop into green plantlets. However, significantly more primordial shoots and green plantlets developed on the friable callus than on the compact callus, and significantly more green plantlets were regenerated under the higher photon irradiances of 200 and 300 mumol m(-2) s(-1) than under the lower irradiances of 100 and 150 mumol m(-2) s(-1). The compact callus produced its maximum number of green plantlets early in the experiment (after 9 wk), while the friable callus continued to produce primordial shoots and green plantlets throughout the period of the experiment, and reached its maximum production of green plantlets at 21 wk under the irradiance of 300 mumol m(-2) s(-1). Organogenesis from friable callus under high irradiance (300 mumol m(-2) s(-1)) offers an efficient propagation method for C. blakei.

O nitrogênio é o nutriente acumulado em maior quantidade e de mais alto custo na produção do trigo. Sendo assim, este trabalho foi realizado com o objetivo de determinar a viabilidade econômica (em 2007 e 2008) da utilização de diferentes fontes, doses e épocas de aplicação de nitrogênio na produtividade de grãos de dois cultivares de trigo irrigados, em Selvíria-MS. Para tal, um único experimento foi desenvolvido em 2007, na área experimental pertencente à Faculdade de Engenharia - UNESP, em um Latossolo Vermelho Distrófico. Para análise econômica foram considerados os custos (2007 e 2008) e os acréscimos de produção dos diferentes manejos da adubação nitrogenada, correspondentes a cada tratamento do fatorial. Ou seja, cinco doses de N (0, 50, 100, 150 e 200 kg ha-1), três fontes de N (Entec®, sulfato de amônio e uréia) e duas épocas de aplicação (totalmente na semeadura, ao lado das linhas ou em cobertura), em dois cultivares de trigo (E 21 e IAC 370), com quatro repetições. Concluiu-se que: o acréscimo no valor de produção para o produtor foi maior em junho de 2008. A aplicação da dose de 50 kg ha-1 de N apresentou sempre maior vantagem econômica, independentemente da fonte e época de aplicação. A ureia aplicada em cobertura, independentemente da dose de N, proporcionou benefícios líquidos positivos para o cultivar E 21 e IAC 370 em 2008. Recomenda-se para obtenção da maior margem de ganho a aplicação de 50 kg ha-1 de N, de sulfato de amônio, em cobertura, para ambos os cultivares.

O cultivo de trigo de sequeiro constitui ótima opção para o Estado de Minas Gerais em regiões com altitude superior a 800 m. Neste estudo objetivou-se estimar a divergência genética entre genótipos de trigo com o auxílio de métodos de análise multivariada, objetivando identificar potenciais genótipos para obtenção de combinações híbridas promissoras. Foi utilizado o delineamento experimental em blocos ao acaso, com quatro repetições. Avaliaram-se seis caracteres morfoagronômicos em 11 genótipos de trigo cultivados em condições de sequeiro no município de Patos de Minas-MG, nos anos de 2007 e 2008. Houve grande variabilidade fenotípica entre os genótipos de trigo avaliados em cultivo de sequeiro. Os genótipos IAC 289-L22 e IAC 289-L4 foram os mais similares, enquanto os CD 113 e EP 011106, os maisdivergentes, promissores nos cruzamentos para obtenção de materiais adaptados e com bons atributos fenotípicos em cultivo de sequeiro.

Com o uso da análise de trilha, é possível avaliar as associações entre caracteres e desdobrar a correlação existente em efeitos diretos e indiretos. O objetivo deste estudo foi estimar correlações genotípicas e desdobrar os efeitos diretos e indiretos dos componentes do rendimento de grãos sobre a produtividade de cultivares de trigo. O experimento foi conduzido no ano de 2008, no campo experimental da Cooperativa Central de Pesquisa Agrícola (Coodetec) no município de Palotina/PR. O delineamento experimental utilizado foi de blocos casualizados com três repetições. As características avaliadas foram: tamanho da espiga, número de espiguetas por espiga, número de grãos por espiga, número de espigas por metro, massa de mil grãos e rendimento de grãos. Os resultados obtidos foram submetidos à análise de variância e à análise de trilha. A análise de variância mostrou diferença entre os genótipos avaliados, indicando a presença de variabilidade genética para os caracteres avaliados. A seleção indireta, por meio do número de grãos por espiga, levando em consideração a massa de mil grãos, é a melhor estratégia para a obtenção de genótipos superiores em rendimento de grãos.

A utilização de fungicida para o controle de doenças foliares na cultura do trigo é prática comum entre os triticultores. Contudo, devido ao impacto ambiental provocado pelo seu uso generalizado, buscam-se alternativas para controle dos fitopatógenos. Sendo assim, este trabalho teve como objetivo avaliar o efeito da nutrição mineral foliar sobre as doenças foliares do trigo, bem como seu efeito sobre o desenvolvimento da planta. O delineamento experimental foi em blocos casualizados com quatro repetições e quatro tratamentos: 1- testemunha; 2- adubação foliar aos 30 dias após a emergência (DAE) e aos 75 DAE; 3- fungicida aos 30 e 75 DAE; e 4- adubação foliar aos 30 DAE e adubação foliar mais fungicida aos 75 DAE. Foram realizadas nove avaliações da incidência e da severidade do oídio, cujos dados foram utilizados para o cálculo das áreas abaixo das curvas de progresso da doença; três avaliações da severidade da ferrugem e da mancha da gluma; e duas avaliações da mancha amarela. Foram determinadas a massa de matéria fresca e seca da parte aérea e do sistema radicular e a produtividade da cultura. Foi observada redução da incidência e da severidade de oídio em função da adubação foliar, apenas na primeira avaliação de incidência e nas segunda e quinta avaliações de severidade da doença. O efeito complementar dos nutrientes com o fungicida para redução das doenças foi observado apenas para a mancha amarela, com redução de aproximadamente 11,16 % da severidade. Os maiores valores de produtividade foram observados para os tratamentos apenas com fungicida e, com fungicida mais nutrientes. A adubação foliar proporcionou aumento da massa fresca de raízes e massa seca de raízes. Apesar disso, não houve aumento da massa fresca da parte aérea em função da adubação foliar.

Com o objetivo de selecionar genótipos de trigo tolerantes à seca, por meio da indução de déficit hídrico por diferentes agentes osmóticos, sementes e embriões imaturos de oito genótipos foram submetidos a diferentes gradientes osmóticos e avaliados quanto ao comprimento da parte aérea e da raiz principal e à produção de biomassa total. As sementes foram submetidas aos agentes polietilenoglicol-6000 e manitol, em sete gradientes osmóticos, e avaliadas após dez dias. Os embriões foram cultivados em meio macrobatata, suplementado com PEG 6000, manitol e maltose nas concentrações de 10, 20 e 30%, além do controle, e avaliados após 30 dias de incubação. O delineamento foi inteiramente casualizado e os dados foram submetidos à análise de regressão, utilizando-se o teste "t", para comparar os valores de Beta, e análise de variância, com teste de médias pelo teste "t". Houve efeito do estresse hídrico em todos os genótipos testados para as plântulas oriundas de sementes, com interação significativa entre agente e cultivar e efeito negativo maior para PEG 6000. Os genótipos BH 1146 e Ocepar 14 apresentaram maior tolerância à seca, redução média no crescimento, na presença dos agentes estressantes, de 58 e 62%, respectivamente. Em contraste, o Aliança foi o mais sensível, com redução média de 92%. Para as plântulas oriundas de embriões imaturos, o genótipo BH 1146 também apresentou relativa tolerância; já o Ocepar 14 e Aliança foram altamente sensíveis. Porém, estes genótipos também apresentaram baixo desempenho no meio sem a presença dos agentes estressantes, podendo o efeito negativo estar condicionado ao cultivo in vitro.

The search for higher profitability in wheat crop with cost reduction technologies that may promote sustainability is an important matter in Brazilian agriculture. This study evaluated the profitability of no-tilled wheat, reducing nitrogen topdressing doses with the cultivation of green manure before the wheat crop. The experiment was carried out in Selvíria (MS), Brazil, in 2009/10. The experiment was arranged in a randomized block design with 36 treatments in splitplots and four replicates. The plots were formed by six types of green manure: Cajanus cajan L. BRS Mandarin, Crotalaria juncea L., Pennisetum americanum L. BRS 1501, fallow area and mixed cropping of Pennisetum americanum L. + Cajanus cajan L. and Pennisetum americanum L. + crotalaria which provided straw for no-tilled wheat in the winter, following the rice crop in the summer. The subplots were formed by six levels of topdressing nitrogen (0, 25, 50, 75, 100 and 125 kg N ha-1) using urea as a nitrogen source. The wheat grown after green manure in the previous winter crop, with no nitrogen topdressing and a rate of 25 kg ha-1 N, had more frequently production costs above the gross income. Wheat production cost after the mixed cropping Pennisetum americanum L. + Cajanus cajan L. and Pennisetum americanum L. + Crotalaria juncea L. from the previous winter crop, combined with nitrogen rates of 50 and 75 kg N ha-1, provided better profitability compared with the other green manures evaluated.

NBPT (N-(n-butyl) thiophosphoric triamide), a urease inhibitor, has been reported as one of the most promising compounds to maximize urea nitrogen use in agricultural systems. The objective of this study was to evaluate the performance of irrigated wheat fertilized with urea or urea + NBPT as single or split application. The experiment was conducted from June to October 2006 in Viçosa, MG, Brazil. The experimental design followed a 2×2 factorial scheme, in which urea or urea + NBPT were combined with two modes of application: full dose at sowing (60kg ha-1) or split (20kg ha-1 at sowing + 40kg ha-1 as topdressing at tillering), in randomized blocks with ten replications. The split application of nitrogen fertilization does not improve the yield wheat under used conditions. The use of urease inhibitor improves the grain yield of wheat crop when urea is applied in topdressing at tillering, but its use does not promote difference when urea is applied in the furrow at planting.
