Folic acid (FA) supplementation during carcinogenesis is controversial. Considering the impact of liver cancer as a public health problem and mandatory FA fortification in several countries, the role of FA supplementation in hepatocarcinogenesis should be elucidated. We evaluated FA supplementation during early hepatocarcinogenesis. Rats received daily 0.08 mg (FA8 group) or 0.16 mg (FA16 group) of FA/100 g body weight or water (CO group, controls). After a 2-week treatment, animals were subjected to the ""resistant hepatocyte"" model of hepatocarcinogenesis (initiation with diethylnitrosamine, selection/promotion with 2-acetylaminofluorene and partial hepatectomy) and euthanized after 8 weeks of treatment. Compared to the CO group, the FA16 group presented: reduced (p < 0.05) number of persistent and increased (p < 0.05) number of remodeling glutathione S-transferase (GST-P) positive preneoplastic lesions (PNL); reduced (p < 0.05) cell proliferation in persistent GST-P positive PNL; decreased (p < 0.05) hepatic DNA damage; and a tendency (p < 0.10) for decreased c-myc expression in microdissected PNL. Regarding all these parameters, no differences (p > 0.05) were observed between CO and FA8 groups. FA-treated groups presented increased hepatic levels of S-adenosylmethionine but only FA16 group presented increased S-adenosylmethionine/S-adenosylhomocysteine ratio. No differences (p > 0.05) were observed between experimental groups regarding apoptosis in persistent and remodeling GST-P positive PNL, and global DNA methylation pattern in microdissected PNL. Altogether, the FA16 group, but not the FA8 group, presented chemopreventive activity. Reversion of PNL phenotype and inhibition of DNA damage and of c-myc expression represent relevant FA cellular and molecular effects.

Glutatione S-transferases (GSTs) are a family of enzymes involved in detoxification of xenobiotics. Placental GST, known as GST-P, has been detected in tissues following exposure to carcinogenic agents being regarded a reliable biomarker of exposure and susceptibility in early phases of carcinogenesis. The aim of this study was to investigate the expressivity of GST-P positive foci in the rat tongue mucosa exposed to cigarette smoke by means of immunohistochemistry. A total of twelve male Wistar rats were distributed into two groups: negative control and experimental group exposed to cigarette smoke during 75 days. After experimental period, no histopathological changes in the tongue mucosa were evidenced in the negative control and the experimental group. However, a total of five GST-P positive foci were detected in two out of six animals exposed to cigarrette smoke. None control animals were noticed GST-P positive foci. These data indicate that expression of GST-P may reflect the carcinogenic effect of cigarette smoke as well as the genetic susceptibility of animals in relation to continuous carcinogens exposure.

RESUMO: Antecendentes: Uma avaliação dos serviços de abuso de substâncias em Barbados identificou a necessidade de programas e serviços que são projetados especificamente para crianças e adolescentes. Objetivo: Realizar programa com base em evidências para reduzir a incidência de abuso de drogas entre crianças e adolescentes por meio do fortalecimento da unidade familiar através de parentalidade positiva, de maior funcionamento familiar e de resistência dos jovens. Método: Dois projetos-piloto foram realizadas com base no programa "Fortalecer as Famílias para Pais e Jovens de 12 a 16 anos (SFPY). O programa de nove semanas foi empregado como uma intervenção para criar laços familiares mais fortes, aumentar a resistência dos jovens e reduzir o abuso de drogas entre crianças e adolescentes de idades de 11 a 16 anos. A decisão foi tomada para incluir participantes de 11 anos desde que as crianças possam estar no primeiro ano da escola secundária nessa idade. IMPLEMENTATION OF SUBSTANCE ABUSE PILOT PROJECT FOR CHILDREN AND ADOLESCENTS 5 Resultados: Quinze famílias participaram em dois projetos-piloto e a avaliação final mostrou que os jovens após o programa, geralmente tornaram-se mais positivos sobre o seu lugar na unidade familiar e sentiram que sua participação no programa foi benéfica. Os pais, da mesma forma, relataram que eles conquistaram, com o programa uma relação mais positiva, uma melhor compreensão das necessidades, e consciência das mudanças de desenvolvimento de seus jovens. Desta forma, considera-se que o programa atingiu o resultado desejado de criar unidades familiares mais fortes. Conclusão: O Projeto Piloto “SFPY” foi bem sucedido em fazer pais e jovens mais conscientes de suas necessidades individuais e de responsabilidades dentro da unidade familiar. Como resultado, o relacionamentos das respectivas famílias melhorou. Estudos baseados em evidências têm demonstrado que um relação familiar mais forte diminui a incidência de uso e abuso de drogas na população adolescente, aumentando os fatores de proteção e diminuindo os fatores de risco. A implementação do programa, que foi desenvolvido e testado no ambiente norte-americano, demonstrou que era transferível para a sociedade de Barbados. No entanto, seu impacto total só pode ser determinado através de um estudo comparativo envolvendo um grupo de controle e / ou uma intervenção alternativa ao abuso de substâncias. Portanto, é recomendável que um estudo comparativo da intervenção SFPY deve envolver uma amostra representativa de adolescentes que estão em estágio de desenvolvimento anterior mais cedo. Evidências já demonstram que o programa é mais eficaz, com impacto mais longo sobre os jovens que participam em uma idade maisABSTRACT:Background: An evaluation of substance abuse services in Barbados has identified the need for programmes and services that are specifically designed for children and adolescents. Aim: To conduct an evidence-based programme to reduce the incidence of substance abuse among children and adolescents by strengthening the family unit through positive parenting, enhanced family functioning and youth resilience. Method: Two pilot projects were conducted based on the ‘Strengthening Families for Parents and Youths 12– 16’ (SFPY) programme. The nine-week programme was employed as an intervention to create stronger family connections, increase youth resiliency and reduce drug abuse among children and adolescents between the ages of 11 to 16. The decision was made to include participants from age 11 since children may be in the first year of secondary school at this age. IMPLEMENTATION OF SUBSTANCE ABUSE PILOT PROJECT FOR CHILDREN AND ADOLESCENTS 3 Results: Fifteen families participated in two pilot projects and an evaluation conducted at the conclusion showed that the youth were generally more positive about their perceived place in the family unit and felt that the being in the programme was generally beneficial. The parents similarly reported they had a more positive relationship with their youths and also had a better understanding of their needs, and an awareness of their developmental changes. This affirmed that the programme had achieved its desired outcome to create stronger family units. Conclusion: The SFPY Pilot Project was successful in making parents and youths more aware of their individual needs and responsibilities within the family unit. As a result relationships within their respective families were strengthened. Evidence-based studies have shown that enhanced family functioning decreases the incidence of substance use and abuse in the adolescent population by increasing protective factors and decreasing risk factors. The implementation of the programme, which was developed and tested in the North American environment, demonstrated that it was transferable to the Barbadian society. However, its full impact can only be determined through a comparative study involving a control group and/or an alternative substance abuse intervention. It is therefore recommended that a comparative study of the SFPY intervention should be delivered to a representative sample of adolescents who are at an earlier developmental stage. Evidence has shown that the programme is more effective, with longer impact on youths who participate at a younger age.

This study’s goal was to analyze whether the quality of university students’ relationship with their parents mediated the association between mental health and physical symptoms and health behavior. Participants were 250 university students (66% female and 34% male), aged between 17 and 29 years old (M = 20.88, SD = 2.03) that answered the Father/Mother Attachment Questionnaire (FMAQ), the Physical Symptoms Scale from the Rotterdam Symptom Checklist (RSCL), the Health Behavior Questionnaire (HBQ), and the Hospital Anxiety and Depression Scale (HADS). The results showed that the indirect effect of physical symptoms on health behavior was significantly mediated by the father’s and mother’s inhibition of exploration and individuality (IEI). Also the indirect effect of psychological distress on health behavior was significantly mediated by the father’s and mother’s IEI. These results suggest that young adults who had more restrictions to their individuality show worse health behaviors. Separation Anxiety and Dependence (SAD) and Quality of Emotional Bond (QEB), the other 2 attachment scales, were not mediators of the relationship between physical symptoms/ psychological distress and health behavior. This study shows the importance of promoting positive parenting practices that contribute to healthier behavior choices and less risky behaviors, as well as the need for more studies that clearly identify these practices in young adult populations.

Résumé Cette étude quasi expérimentale consistait à élaborer et à mettre à l’essai une mesure de soutien à l’intention d’enseignants débutants ainsi qu’à évaluer l’efficacité de celle-ci. L’une des particularités de cette mesure, appelée Dispositif de soutien en gestion de classe, était qu’elle était centrée essentiellement sur le développement de la compétence à gérer la classe. L’application du dispositif, échelonnée sur une année scolaire, portait sur une trentaine d’enseignants débutants œuvrant au primaire, en milieu défavorisé, à Montréal. Basé sur les trois phases du modèle théorique d’Archambault et Chouinard (2003), le dispositif se déclinait selon trois cycles de formation : l’établissement du fonctionnement de la classe, le maintien de celui-ci et le soutien à la motivation scolaire, ainsi que l’intervention pour résoudre des problèmes de comportement. Chaque cycle commençait par une journée de formation et d’appropriation (JFA) durant laquelle il y avait présentation d’un contenu théorique puis des ateliers d’appropriation. Par la suite, les enseignants effectuaient des mises en pratique dans leur classe. Pour terminer le cycle, un autre type de rencontre, la rencontre de suivi (RS), servait entre autres à objectiver la pratique. L’aspect original de cette mesure de soutien était que la première rencontre de formation était offerte une semaine avant la rentrée scolaire. Sur le thème « Commencer l’année du bon pied en gestion de classe », cette journée avait pour objectif de soutenir les enseignants débutants dans l’installation du fonctionnement de leur classe. L’efficacité du dispositif a été évaluée sur la base de trois dimensions : l’établissement et le maintien de l’ordre et de la discipline, le sentiment d’efficacité personnelle ainsi que la motivation professionnelle. Les perceptions du groupe d’enseignants débutants ayant pris part aux activités du dispositif (n = 27) ont été comparées à celles d’un groupe témoin (n = 44). Les participants avaient, en moyenne, 2,9 années d’expérience et leur âge variait de 23 à 56 ans. Les données ont été recueillies à l’aide d’un questionnaire auto rapporté rempli en deux temps, soit au deuxième et au huitième mois de l’année scolaire. Les scores des enseignants débutants du dispositif ont augmenté dans le temps pour l’ensemble des variables à l’étude. De plus, les analyses de variance à mesures répétées ont révélé que le dispositif a eu une triple incidence positive, attestée par des effets d’interaction. Les enseignants débutants engagés dans la démarche ont connu une augmentation de leur capacité à implanter les règles de classe, de leur sentiment d’efficacité personnelle à gérer les situations d’apprentissage et de leur motivation professionnelle. En effet, alors que, au début de l’étude, ils rapportaient des scores significativement inférieurs à ceux du groupe témoin, à la fin, les scores étaient équivalents. Les résultats ont aussi montré que les participants du groupe expérimental se distinguaient en affichant un meilleur sentiment d’efficacité à faire apprendre leurs élèves. L’étude nous apprend également que le sentiment d’efficacité personnelle à faire face aux problèmes de comportement et la capacité à gérer les comportements se sont renforcés de façon significative dans le temps chez l’ensemble des enseignants débutants. Finalement, aucun changement significatif n’a été détecté pour deux des huit variables à l’étude : le sentiment d’efficacité personnelle à avoir un effet sur le comportement des élèves et l’application des règles de classe. En définitive, ces résultats sont encourageants. Ils montrent l’enrichissement professionnel que les enseignants débutants peuvent retirer lorsqu’ils sont soutenus adéquatement. Nous croyons que la journée de formation portant sur l’installation du fonctionnement de la classe, avant la rentrée scolaire, a joué un rôle central dans les succès vécus par les enseignants débutants participants. C’est pourquoi nous recommandons ce type de formation assorti d’un suivi à long terme, où d’autres composantes entrent en jeu, afin de nourrir le sentiment d’efficacité personnelle et la motivation professionnelle des nouveaux enseignants.

Kan det vara så att extramuralt lärande i forskarmiljö för elever vid gymnasiets naturvetenskapliga program kan öka motivationen till vidare studier inom naturvetenskap? Extramuralt lärande innebär allt lärande som sker under aktiviteter utanför skolan. Jag har i den här studien följt och intervjuat en klass i åk 2 som tillbringat en vecka på forskarskola i Garpenberg. En av frågorna i denna studie var om arbetssättet påverkat dem, och i så fall hur, om det har lockat eller avskräckt? Resultatet visar att alla elever i den här studien tyckte att forskarveckan var rolig och intressant. Ungefär 80% hade redan innan bestämt sig för att läsa vidare, men ca 45% ansåg att deras motivation till att läsa vidare hade ökat i och med forskarskolan. Höstens resultat ligger lite över genomsnittet som visar att ca 31% av gymnasieeleverna som under de sex åren som forskarskolan funnits har upplevt en ökad studiemotivation. En internationell studie från England inom extramuralt lärande visar på att många elever tycker att skolundervisningen är tråkig, irrelevant och för gammal och att den har en design som endast tilltalat ett fåtal elever. Däremot tycker eleverna att extramuralt lärande vid s k Science centers, botaniska trädgårdar och museum m fl presenterar vetenskapen på ett intressant och inspirerande sätt. Jag försöker i den här studien att se lärandet ur ett sociokulturellt perspektiv samt dra paralleller mellan tidigare studier om Science centers och min undersökning om forskarskolan som båda erbjuder extramuralt lärande. Jag anser dock att forskarskolan gör det lite mer konkret eftersom eleverna får genomföra ett projekt som de sedan redovisar inför alla forskare och övriga elever. Upplägget för ett besök på ett museum eller vetenskapscentra ser lite annorlunda ut. De allra flesta eleverna i studien insåg nyttan med att läsa ämnen som matematik, fysik, kemi och biologi, och de tyckte att arbetsformen att kombinera fältstudier med teori och laboratorieverksamhet var mycket givande. Eleverna ansåg att de under forskarskolan fått en inblick i hur det kan vara att arbeta som forskare men det var endast en elev som kunde tänka sig det yrket i framtiden. Studiens resultat gäller endast för de elever som deltagit i enkätundersökningen och kan inte sägas vara generaliserbart för alla elever på gymnasiets NV-program.

The influences of fasting on DEN-initiation and of intermittent fasting (IF) on the rat liver chemical carcinogenesis process were evaluated in a 52-week long assay. Three groups of adult male Wistar rats were used: Groups I to 3 were treated with a single i.p. injection of 200 mg/kg of diethylnitrosamine (DEN). Group 2 was submitted to 48 h fasting prior to DEN treatment. After the 4th week, Group 3 was submitted to IF, established as 48 h weekly fasting during 48 weeks, while Groups I and 2 were fed ad libitum until the 52nd week. All animals were submitted to 70% partial hepatectomy and sacrificed at the 3rd and 52nd weeks, respectively. Fasting prior to DEN-initiation did not influence the development of altered foci of hepatocytes (AFHs) and of hepatic nodules (Group 2 vs. Group G1). IF inhibited the development of preneoplastic lesions, since this dietary regimen decreased the number and the size of glutathione S-transferase (GST-P) positive foci and the number and size of liver nodules (Group G3 vs. Group G1), the inhibitory effect of IF was also reflected in the development of clear and basophilic cell foci. These results indicate that long-term IF regimen exerts an anti-promoting effect on rat hepatocarcinogenesis induced by DEN. (C) 2002 Wiley-Liss, Inc.

The modifying potential of crude extracts of the mushroom Agaricus blazei Murrill (Himematsutake) on the development and growth of glutathione S-transferase placental form (GST-P)-positive liver foci (liver preneoplastic lesion) was investigated in adult male Wistar rats. Six groups of animals were used. Groups 2 to 5 were given a single i.p. injection of 200 mg/kg b.w. of diethylnitrosamine (DEN) and groups 1 and 6 were treated with saline at the beginning of the experiment. After 2 weeks, animals of groups 3 to 6 were orally treated with three dose levels of aqueous extracts of the mushroom A. blazei (1.2, 5.6, 11.5, and 11.5 mg/ml of dry weight of solids) for 6 weeks. All animals were subjected to two-thirds partial hepatectomy at week 3 and sacrificed at week 8. Two hours before sacrifice, ten animals of each group were administered a single i.p injection of 100 mg/kg of bromodeoxyuridine (BrdU). Apoptotic bodies and BrdU-positive hepatocyte nuclei were quantified in liver sections stained for hematoxylin and eosin (H&E) (eosinophilic foci) and simultaneously stained for GST-P expression (GST-P-positive foci), respectively. The 6-week treatment with A. blazei did not alter the development (number and size) of GST-P-positive foci and did not affect the growth kinetics of liver normal parenchyma or foci in DEN-initiated animals. Our results indicate that the treatment with aqueous extracts of the mushroom A. blazei during the post-initiation stage of rat liver carcinogenesis does not exert any protective effect against the development of GST-P-positive foci induced by DEN. Annatto (Bixa orellana L.) is a natural food colorant extensively used in many processed foods, especially dairy products. The lower cost of production and the low toxicity, make annatto a very attractive and convenient pigment in substitution to the many synthetic colorants. In the present study we investigate the carcinogenic and anticarcinogenic effects of dietary annatto in Wistar rat liver using the preneoplastic glutathione S-transferase (GST-P) foci and DNA damage biomarkers. Annatto, containing 5% bixin, was administered in the diet at concentrations of 20, 200, and 1000 ppm (0.07; 0.80 and 4.23 bixin/kg body wt/day, respectively), continuously during 2 weeks before, or 8 weeks after DEN treatment (200 mg/kg body wt, i.p.), to evaluate its effect on the liver-carcinogenesis medium-term bioassay. The comet assay was used to investigate the modifying potential of annatto on DEN (20 mg/kg body wt)-induced DNA damage. The results showed that annatto was neither genotoxic nor carcinogenic at the highest concentration tested (1000 ppm). No protective effects were also observed in both GST-P foci development and comet assays. In conclusion, in such experimental conditions, annatto shows no hepatocarcinogenic effect or modifying potential against DEN-induced DNA damage and preneoplastic foci in the rat liver. The interaction between dietary energy restriction and low dose of the fungicide hexachlorobenzene (HCB) was evaluated in a rat liver medium-term bioassay for carcinogenesis. Male Wistar rats were fed a control or a 50% energy-restricted diet, both added or not with 50 ppm HCB, for 6 weeks. HCB exposure or energy restriction separately did not exert any influence on the development of glutathione S-transferase placental form (GST-P+) foci of hepatocytes. Simultaneous HCB exposure and energy restriction induced a significant increase in liver centrilobular hypertrophy and GST-P+ foci development. Our findings suggest that energy restriction increases liver response to low dose of HCB, unmasking the promoting potential of this fungicide. (C) 2000 Elsevier B.V. Ireland Ltd. All rights reserved.
