We investigated whether the protection from graft-versus-host disease (GVHD) afforded by donor treatment with granulocyte colony-stimulating factor (G-CSF) could be enhanced by dose escalation. Donor treatment with human G-CSIF prevented GVHD in the B6 --> B6D2F1 murine model in a dose-dependent fashion, and murine G-CSF provided equivalent protection from GVHD at 10-fold lower doses. Donor pretreatment with a single dose of pegylated G-CSF (peg-G-CSF) prevented GVHD to a significantly greater extent than standard G-CSIF (survival, 75% versus 11%, P < .001). Donor T cells from peg-G-CSF-treated donors failed to proliferate to alloantigen and inhibited the responses of control T cells in an interleukin 10 (IL-10)-dependent-fashion in vitro. T cells from peg-GCSF-treated IL-10(-/-) donors induced lethal GVHD; T cells from peg-G-CSF-treated wild-type (wt) donors promoted long-term survival. Whereas T cells from peg-G-CSF wt donors were able to regulate GVHD induced by T cells from control-treated donors, T cells from G-CSF-treated wt donors and peg-G-CSF-treated IL-10(-/-) donors did not prevent mortality. Thus, peg-G-CSF is markedly superior to standard G-CSF for the prevention of GVHD following allogeneic stem cell transplantation (SCT), due to the generation of IL-10-producing regulatory T cells. The initiation of graft-vs-host disease (GVHD) after stem cell transplantation is dependent on direct Ag presentation by host APCs, whereas the effect of donor APC populations is unclear. We studied the role of indirect Ag presentation in allogenic T cell responses by adding populations of cytokine-expanded donor APC to hemopoietic grafts that would otherwise induce lethal GVHD. Progenipoietin-1 (a synthetic G-CSF/Flt-3 ligand molecule) and G-CSF expanded myeloid dendritic cells (DC), plasmacytoid DC, and a novel granulocyte-monocyte precursor population (GM) that differentiate into class II+,CD80/CD86(+),CD40(-) APC during GVHD. Whereas addition of plasmacytoid and myeloid donor DC augmented GVHD, GM cells promoted transplant tolerance by MHC class II-restricted generation of IL-10-secreting, Ag-specific regulatory T cells. Importantly, although GM cells abrogated GVHD, graft-vs-leukemia effects were preserved. Thus, a population of cytokine-expanded GM precursors function as regulatory APCs, suggesting that G-CSF derivatives may have application in disorders characterized by a loss of self-tolerance.

Host antigen-presenting cells (APCs) are known to be critical for the induction of graft-versus-host disease (GVHD) after allogeneic bone marrow transplantation (BMT), but the relative contribution of specific APC subsets remains unclear. We have studied the role of host B cells in GVHD by using B-cell-deficient mu MT mice as BMT recipients in a model of CD4-dependent GVHD to major histocompatlibility complex antigens. We demonstrate that acute GVHD is initially augmented in mu MT recipients relative to wild-type recipients (mortality: 85% vs 44%, P < .01), and this is the result of an increase in donor T-cell proliferation, expansion, and inflammatory cytokine production early after BMT. Recipient B cells were depleted 28-fold at the time of BMT by total body irradiation (TBI) administered 24 hours earlier, and we demonstrate that TBI rapidly induces sustained interleukin-110 (IL-10) generation from B cells but not dendritic cells (DCs) or other cellular populations within the spleen. Finally, recipient mice in which B cells are unable to produce IL-10 due to homologous gene deletion develop more severe acute GVHD than recipient mice in which B cells are wild type. Thus, the induction of IL-10 in host B cells during conditioning attenuates experimental acute GVHD.

The health status of wild and captive Atlantic Bottlenose dolphins ( Tersiops truncatis) is difficult to ascertain. Mass strandings of these animals have been attributed to pollutants, as well as bacterial infections. Using human Enzyme Linked Immuno-Assays (ELISA) for immunological cytokines, I measured soluble cytokine levels with respect to their health status. In a retrospective analysis of dolphin sera, there was a trend of higher cytokine levels in “sick” animals. I cultured dolphin lymphocytes in the presence of a mitogen (PHA), a super antigen (Staph-A), Lipopolysaccharide (LPS), and a calcium flux inducer (PMA). Levels of messenger RNA, from these cultured cells, were assayed with Polymerase Chain Reaction (PCR) using primers for the human cytokines IL-2, IL-4, IL-6, IL-10, Tumor Necrosis Factor, and Interferon gamma. Only IL-4, IL-6, and IL-10 messages were obtained, inferring similar nucleotide homology to the human primer sequences. The PCR products were sequenced. Sixteen IL-4 sequences, twelve IL-6 sequences and seven IL-10 sequences were obtained and analyzed. Each cytokine exhibited the same nucleotide sequence in all dolphins examined. There was no difference in the cytokine profile in response to the various stimuli. The derived amino acid composition for each of the dolphin cytokines was used for molecular modeling, which showed that dolphin IL-4, IL-6, and IL-10 were structurally similar to the corresponding proteins of Perissodactyla. ^

B cell abnormalities contribute to the development and progress of autoimmune disease. Traditionally, the role of B cells in autoimmune disease was thought to be predominantly limited to the production of autoantibodies. Nevertheless, in addition to autoantibody production, B cells have other functions potentially relevant to autoimmunity. Such functions include antigen presentation to and activation of T cells, expression of costimulatory molecules and cytokine production. Recently, the ability of B cells to negatively regulate cellular immune responses and inflammation has been described and the concept of “regulatory B cells” has emerged. A variety of cytokines produced by regulatory B cell subsets have been reported with interleukin-10 (IL-10) being the most studied. IL-10-producing regulatory B cells predominantly localize within a rare CD1dhiCD5+ B cell subset in mice and the CD24hiCD27+ B cell subset in adult humans. This specific IL-10-producing subset of regulatory B cells have been named “B10 cells” to highlight that the regulatory function of these rare B cells is primarily mediated by IL-10, and to distinguish them from other regulatory B cell subsets that regulate immune responses through different mechanisms. B10 cells have been studies in a variety of animal models with autoimmune disease and clinical settings of human autoimmunity. There are many unsolved questions related to B10 cells including their surface phenotype, their origin and development in vivo, and their role in autoimmunity.

In Chapter 3 of this dissertation, the role of the B cell receptor (BCR) in B10 cell development is highlighted. First, the BCR repertoire of mouse peritoneal cavity B10 cells is examined by single cell sequencing; peritoneal cavity B10 cells have clonally diverse germline BCRs that are predominantly unmutated. Second, mouse B10 cells are shown to have higher frequencies of λ+ BCRs compared to non-B10 cells which may indicate the involvement of BCR light chain editing early in the process of B10 cell development in vivo. Third, human peripheral blood B10 cells are examined and are also found to express higher frequencies of λ chains compared to non-b10 cells. Therefore, B10 cell BCRs are clonally diverse and enriched for unmutated germline sequences and λ light chains.

In Chapter 4 of this dissertation, B10 cells are examined in the healthy developing human across the entire age range of infancy, childhood and adolescence, and in a large cohort of children with autoimmunity. The study of B10 cells in the developing human documents a massive transient expansion during middle childhood when up to 30% of blood B cells were competent to produce IL-10. The surface phenotype of pediatric B10 cells was variable and reflective of overall B cell development. B10 cells down-regulated CD4+ T cell interferon-gamma (IFN-γ) production through IL-10-dependent pathways and IFN-γ inhibited whereas interleukin-21 (IL-21) promoted B cell IL-10 competency in vitro. Children with autoimmunity had a contracted B10 cell compartment, along with increased IFN-γ and decreased IL-21 serum levels compared to age-matched healthy controls. The decreased B10 cell frequencies and numbers in children with autoimmunity may be partially explained by the differential regulation of B10 cell development by IFN-γ and IL-21 and alterations in serum cytokine levels. The age-related changes of the B10 cell compartment during normal human development provide new insights into immune tolerance mechanisms involved in inflammation and autoimmunity.

These studies collectively demonstrate that BCR signals are the most important early determinant of B10 cell development in vivo, that human B10 cells are not a surface phenotype defined developmental B cell subset but a functionally defined regulatory B cell subset that regulates CD4+ T IFN-γ production through IL-10-dependent pathways and that human B10 cell development can be regulated by soluble factors in vivo such as the cytokine milieu. The findings of these studies provide new insights into immune tolerance mechanisms involved in human autoimmunity and the potent effects of IL-21 on human B cell IL-10 competence in vitro open new horizons in the development of autologous B10 cell-based therapies as an approach to treat human autoimmune disease in the future.

Background: The -819C/T polymorphism in interleukin 10 (IL-10) gene has been reported to be associated with inflammatory bowel disease (IBD) ,but the previous results are conflicting. Materials and Methods: The present study aimed at investigating the association between this polymorphism and risk of IBD using a meta-analysis.PubMed,Web of Science,EMBASE,google scholar and China National Knowledge Infrastructure (CNKI) databases were systematically searched to identify relevant publications from their inception to April 2016.Pooled odds ratio (OR) with 95% confidence interval (CI) was calculated using fixed- or random-effects models. Results: A total of 7 case-control studies containing 1890 patients and 2929 controls were enrolled into this meta-analysis, and our results showed no association between IL-10 gene -819C/T polymorphism and IBD risk(TT vs. CC:OR=0.81,95%CI 0.64- 1.04;CT vs. CC:OR=0.92,95%CI 0.81-1.05; Dominant model: OR=0.90,95%CI 0.80-1.02; Recessive model: OR=0.84,95%CI 0.66-1.06). In a subgroup analysis by nationality, the -819C/T polymorphism was not associated with IBD in both Asians and Caucasians. In the subgroup analysis stratified by IBD type, significant association was found in Crohn’s disease(CD)(CT vs. CC:OR=0.68,95%CI 0.48-0.97). Conclusion: In summary, the present meta-analysis suggests that the IL-10 gene -819C/T polymorphism may be associated with CD risk.

CD4+CD25+调节性T细胞是1995年才发现的一个具有免疫抑制功能的T细胞亚群,主要通过细胞与细胞间直接接触和分泌抑制性细胞因子发挥作用,在维持机体免疫自稳、防止自身免疫以及肿瘤免疫、移植免疫等方面起着重要作用。有关Treg HIV/SIV病毒感染及AIDS的进展关系密切,但却有两种不同的观点。一种认为Treg的数量和功能受到损伤,从而导致宿主免疫系统过度活化。另一种则认为Treg在早期抑制了针对HIV/SIV的特异性的免疫反应,从而导致机体无法清除感染的病毒。本文利用动物模型对CD4+CD25+调节性T细胞在SIV感染后的数量和功能做了动态的检测,并对其中的机制做了初步的探讨。我们首先建立了SIVmac239病毒株对中国起源的恒河猴感染的动物模型,建立了前病毒的检测方法、血浆病毒载量的测定方法、血浆病毒特异性抗体的测定方法,以及病毒的分离方法,并获得了早期感染的相关数据。在研究中我们发现,SIV感染后的1周后即可在恒河猴外周单个核细胞DNA中检测到前病毒。病毒血症也在1周后出现,并很快达到高峰。不同的个体对病毒感染的体液免疫不尽相同,血浆抗体很快出现,但是99003猴抗体下降很快,而99083猴则保持了一定数量的抗体。同时,伴随SIV的感染进程的还有T淋巴细胞的数量变化,CD4+T细胞数量持续下降,而CD8+T细胞数量则在增加,出现CD4/CD8倒置的现象。以上说明恒河猴被SIV所成功感染。在该动物模型的基础上,我们利用体内传代的SIVmac239病毒株,对4只健康恒河猴进行了感染,并对CD4+CD25+调节性T细胞(Treg)亚群在数量上的变化进行了检测,并对其中的机制做了初步探讨。我们在研究中发现,外周血中的Treg在SIV感染后无论是绝对数量还是在占CD4+T细胞中的相对数量均有增加,而且Treg仍然保持了对靶细胞的抑制功能。对腹部淋巴结的分析显示,SIV感染后的一段时期内,该部位FoxP3 mRNA的表达水平也在上升,TGF-β、IL-10的转录也显著增加。前者可以通过抑制树突状细胞间接抑制效应细胞,而后者则是一个抑制性的细胞因子,可以直接作用于靶细胞。因此,我们推测SIV引起免疫系统的过度活化可能不是由于Treg功能的受损,其中的机制需要深入研究。 Treg表达CCR5表面分子(HIV辅助受体之一),同时也有CD4分子的表达,因此推测HIV/SIV可以感染Treg。但是国内外这方面的文献很少。我们对Treg中前病毒的检测发现,SIV可以感染Treg,而且对Treg的感染比例高于CD4+CD25-T细胞。这个结果与Treg绝对数量的上升的结果说明,SIV感染Treg但可能却没有杀伤Treg,因此Treg在数量上有所增加。不过,其中的机制仍有待于进一步的研究。在对SIV引起的体液免疫的研究中还发现,机体针对SIV不同抗原的抗体有不同的模式。部分抗原很快就产生了比较强的反应,但是却不能维持高水平的表达。而针对p27蛋白的抗体产生比较晚,但却长时间维持在比较高的水平。是否这样高水平的抗体有助于控制病毒复制是个值得探讨的问题。

Introduction. Cardiovascular disease (CVD) represents the main cause of morbidity and mortality in kidney recipients. This study was undertaken to assess the impact of functional polymorphisms located in cytokine and apoptosis genes on CVD after kidney transplantation. Cytokine polymorphisms, generally located in gene regulatory regions, are associated with high and low cytokine production and are likely to modulate the magnitude of inflammatory responses following transplantation, depending on the balance between the levels of pro-inflammatory and antiinflammatory cytokines. The role of apoptosis in atherosclerosis has not been completely elucidated, and here we explored the hypothesis that the heterogeneity in cardiovascular risk in kidney recipients may also be linked to functional polymorphisms involved in apoptosis induction. Purpose. In the search for relevant genetic markers of predisposition to CVD after renal transplant, the present investigation was undertaken to identify the clinical impact of polymorphisms of cytokines TNF-α, TGF-β, IL-10, IL-6, IFN-γ and IL-8 and of apoptosis genes Fas and Caspase 9 in a population of kidney transplant recipients. Materials and methods. The study involved 167 patients who received cadaveric kidney transplantation at our centre between 1997 and 2005 (minimum follow-up of 12 months); 35 of them had experienced cardiovascular events (CVD group) and 132 had no cardiovascular complications (non-CVD group). Genotyping was performed using RFLP (Restriction Fragment Length Polymorphism) for RFLP per IL-8/T-251A, Fas/G-670A e Casp9/R221Q polymorphism and SSP (Sequence Specific Primer) for TNF-α/G-308A, TGF-β/L10P, TGF-β/R25P, IL-10/G-1082A, IL- 10/C-819T, IL-10/C-592A, IL-6/G-174C, IFN-γ/T+874A polymorphisms.Results. We found a significant difference in TNF-α and IL-10 genotype frequencies between the patients who had suffered cardiovascular events and those with no CVD history. The high producer genotype for proflogistic cytokine TNF-α appeared to have a significantly superior prevalence in the CVD group compared to the non-CVD group (40.0% vs 21.2%) and it resulted in a 2.4-fold increased cardiovascular risk (OR=2.361; p=0.0289). On the other hand, the high producer genotype for the antiinflammatory cytokine IL-10 was found in 2.8% of the CVD group and in 16.7% of non-CVD group; logistic regression showed a 0.3-fold reduced risk of CVD associated with genetically determined high IL-10 production (OR=0.278; p<0.0001). The other polymorphisms did not prove to have any impact on CVD. Conclusions. TNF-α and IL-10 gene polymorphisms might represent cardiovascular risk markers in renal transplant recipients.

Patienten mit akuter, selbstlimitierender HCV-Infektion und solche mit IFN-a Therapieresponse (SCR) zeigten eine hochregulierte NS3-spezifische T-Helferzellantwort.Über die funktionelle Bedeutung der T-Helferzellantwort und der Antikörpersynthese besteht jedoch noch Unklarheit.In dieser Studie wurde die proliferative Antwort der PBMC von Patienten mit anhaltender Therapieresponse (SCR; n=8), Non-Respondern (NR; n=13) und unbehandelten HCV-Patienten(UTR; n=10) sowie gesunden Kontrollen (HC; n=5) auf die rekombinanten HCV-Antigene Core, Helicase, NS3, NS4 und NS5-4 bestimmt und ihre Sekretion von IFN-.

This thesis focuses on the role of B cells in mCMV and Leishmania major infection. B cells are an essential component of the adaptive immune system and play a key role in the humoral immune response. In mCMV infection we analyzed the influence of B cells on the virus-specific CD8 T cell response, in detail the role of B cells as IL-10 secreting cells, as source of immunoglobulin (Ig) and as antigen presenting cells. In Leishmania major infection we investigated the role of Ig in Th1 and Th2 directed disease.rnWe found in mCMV infection that the B cell secreted IL-10 suppresses effectively the acute virus-specific CD8 T cell response, while the IL-10 secreted by dendritic cell has no obvious effect. Ig has no effect in the acute virus-specific CD8 T cell response, but in memory response Ig is essential. If Ig is missing the CD8 T cell population remains high in memory response 135 days post infection. The complete absence of B cells dramatically reduces the acute virus-specific CD8 T cell response, while B cell reconstitution just partially rescues this dramatic reduction. A comparison of this reduction in a B cell free organism to an organism with depleted dendritic cells gave a similar result. To exclude a malfunction of the CD8 T cells in the B cell deficient mice, the decreased virus-specific CD8 T cell population was confirmed in a B cell depletion model. Further, bone marrow chimeras with a B cell compartment deficient for CD40-/- showed a decrease of the virus-specific response and an involvement of CD40 on B cells. Taken together these results suggest a role for B cells in antigen presentation during mCMV infection.rnFurther we took advantage of the altered mCMV specific CD8 T cell memory response in mice without Ig to investigate the memory inflation of CD8 T cells specific for distinct mCMV specifc peptides. Using a SIINFEKL-presenting virus system, we were able to shorten the time until the memory inflation occurs and show that direct presentation stimulates the memory inflation. rnIn Leishmania major infection, Ig of Th2 balanced BALB/c mice has a central role in preventing a systemic infection, although the ear lesions are smaller in IgMi mice without specific Ig. Here the parasite loads of ears and spleen are elevated, and an IMS-reconstitution does not affect the parasite load. In contrast in Th1 balanced C57BL/6 mice, reconstitution of IgMi mice with serum of either untreated or immunized mice decreased the parasite load of spleen and ear, further IMS treatment reduces the size of the spleen and the cytokine-levels of IL-10, IL-4, IL-2 and IFN-γ to a level comparable to wt mice. rn

Foxp3 es un marcador clave para identificación y función células T reguladoras, además su expresión se ha observado en diferentes líneas celulares de cáncer. El objetivo de este estudio fue determinar si la expresión de Foxp3 en células de melanoma murino actúa como un mecanismo de evasión de la respuesta inmune tumoral, modificando citocinas involucradas en la fase de inmunoedición de cáncer y promoviendo la generación de células Treg. En este estudio se determinó por primera vez la expresión de Foxp3 en las células melanoma murino B16F10 wt, y diseñamos RNA de interferencia en contra de Foxp3, además de analizar la expresión de CD25 y producción de IL-2, INF-γ, TGF-β e IL-10 para determinar su papel in vitro. Para la evaluación del efecto de Foxp3 durante el desarrollo tumoral in vivo, se estableció una línea celular con silenciamiento de Foxp3 la cuál identificamos como B16F10.DMH1 y se montaron dos modelos de melanoma murino, uno inducido con células B16F10 wt y otro inducido con células B16F10.DMH1, y se analizó progresión tumoral, producción de citocinas, expresión de CD25, Foxp3 y poblaciones celulares CD4+ , CD4+CD25+ y CD4+CD25+ Foxp3+ en TIL’s y células de bazo. Nuestros resultados in vitro demuestran que las células B16F10 wt expresan Foxp3 a nivel de RNAm y proteína, y su localización celular es principalmente perinuclear, además se encontró que estas células expresan CD25, y una producción de citocinas del tipo INF-γ, TGF-β, IL-10 e IL-2. Se encontró que la expresión de Foxp3 afecta la proliferación en células B16F10, encontrando una correlación positiva entre la expresión de Foxp3, CD25 e IL-2. In vivo, el silenciamiento de Foxp3 en las células B16F10.DMH1 afectó el desarrollo del melanoma incrementando el tiempo de aparición de tumor, sobrevida y disminuyendo el peso de los tumores, encontrando una correlación positiva entre Foxp3, CD25, IL-2 e IL-10 y negativa con la producción de IFN-γ, además se determinó que Foxp3 intratumoral está correlacionado con la expresión y presencia de células Treg con fenotipo CD4+CD25+ Foxp3+ en el microambiente tumoral y con una disminución de células T CD4+ a nivel periférico, sin afectar a linfocitos T activados (CD4+CD25+ ). Estos datos sugieren que Foxp3, participa en el desarrollo de la tumorogénesis en melanoma murino in vitro e in vivo, con la capacidad de modular a citocinas, moléculas involucradas en el desarrollo tumoral, así como poblaciones celulares con fenotipo regulador en el tumor, pero no en periferia.
