Em Portugal, as indústrias corticeira e de pasta de papel constituem um importante sector económico, contudo, gerando elevadas quantidades de subprodutos. Estes subprodutos poderiam ser explorados em aplicações de alto valor acrescentado, como fonte de compostos fenólicos, por exemplo, em vez de serem apenas queimados para produção de energia. Estes compostos são conhecidos pelas suas inúmeras propriedades, entre as quais, antioxidante, anti-inflamatória e anti-trombótica. Neste estudo as frações fenólicas da maior parte dos subprodutos gerados nas indústrias corticeira e de pasta de papel foram caracterizados em detalhe, com vista à sua valorização. A fração fenólica das cascas de Eucalyptus globulus, E. grandis, E. urograndis e E. maidenii, bem como da cortiça de Quercus suber e resíduos provenientes da sua exploração, nomeadamente, o pó de cortiça e os condensados negros, foi obtida por processos convencionais de extração sólido-líquido. No caso da casca de E. globulus, foi ainda avaliado o potencial de metodologias “verdes” no processo de extração de compostos fenólicos, usando extração com CO2 supercrítico. Esta técnica foi otimizada com recurso a metodologias de superfície de resposta. Na identificação e quantificação dos compostos fenólicos foi usada cromatografia líquida de alta resolução aliada a técnicas de espectrometria de massa. O teor de fenólicos totais foi ainda determinado pelo método de Folin- Ciocalteu, essencialmente para efeitos comparativos. A caracterização da fração fenólica de cada extrato foi ainda complementada com a análise da atividade antioxidante, usando o radical 2,2-difenil-1-picrilhidrazilo (DPPH). Foram identificados trinta compostos fenólicos na casca de E. globulus, 17 deles referenciados pela primeira vez como seus constituintes, nomeadamente os ácidos quínico, di-hidroxifenilacétic, cafeico e metil-elágico, bis-hexahidroxidifenoil( HHDP)-glucose, galoil- bis-HHDP-glucose, galoil-HHDPglucose, isoramnetina—hexosídeo, quercetina-hexosídeo, ácido metil-elágicopentosídeo, miricetina-ramnosídeo, isoramnetina-ramnosídeo, mearnsetina, floridzina, mearnsetina-hexosídeo, luteolina e uma proantocianidina B. Neste trabalho, foi estudada pela primeira vez a composição fenólica das cascas de E. grandis, E. urograndis e E. maidenii. Treze, doze e vinte e quatro compostos fenólicos foram identificados nas cascas de E. grandis, E. urograndis e E. maidenii, respetivamente. Entre estes compostos encontram-se os ácidos quínico, gálico, metilgálico, protocatequínico, clorogénico e elágico, catequina, galoil-bis-HHDP-glucose, digaloilglucose, epicatequina, quercetina-glucoronídeo, di-hidroxiisopropilcromona- hexosídeo, isoramnetina-hexosídeo, ácido elágicoramnosídeo, taxifolina, quercetina-hexosídeo, di-hidroxi- (metilpropil)isopropilcromona-hexosídeo, ácido metil-elágico-pentosídeo, miricetina-ramnosídeo, isoramnetina-ramnosídeo, aromadendrina-ramnosídeo, mearnsetina, mearnsetina-hexosídeo, eriodictiol, quercetina, isoramnetina e naringenina. A análise da fração fenólica da cortiça permitiu identificar vinte e dois compostos fenólicos, dez deles referenciados pela primeira vez como seus constituintes, nomeadamente, os ácidos quínico, salicílico, p-hidroxifenillático e metilgálico, ácido carboxílico da brevifolina, eriodictiol, naringenina, um éster isoprenílico do ácido cafeico, isoramnetina-ramnosídeo e isoramnetina. No pó de cortiça industrial foram identificados dezasseis compostos fenólicos, nomeadamente os ácidos quínico, gálico, protocatequínico, cafeico, ferúlico, elágico e metilgálico, esculetina, ácido carboxílico da brevifolina, coniferaldeído, um éster isoprenílico do ácido cafeico, uma dilactona do ácido valoneico, ácido elágico-pentosídeo, ácido elágico-ramnosídeo, isoramnetinaramnosídeo e isoramnetina. Destes, apenas o ácido elágico foi previamente referenciado como componente do pó de cortiça. Do mesmo modo, treze compostos fenólicos foram identificados no condensado negro, doze deles referenciados pela primeira vez como seus constituintes. São eles os ácidos quínico, gálico, p-hidroxifenil-láctico, protocatequínico, p-coumarico, cafeico e elágico, vanilina, esculetina, coniferaldeído, um éster isoprenílico do ácido cafeico e o eriodictiol. A extração supercrítica de compostos fenólicos da casca de eucalipto permitiu não só verificar os parâmetros que afetam a qualidade e quantidade finais dos extratos, como também obter os valores ótimos para estes parâmetros. Esta extração mostrou ainda ser bastante seletiva para determinados grupos de compostos fenólicos, como as flavanonas eriodictiol e naringenina e para o flavonol O-metilado isoramnetina. Este é também o primeiro estudo envolvendo a determinação da atividade antioxidante de extratos da cortiça e dos resíduos da sua exploração, bem como da casca de E. grandis, E. urograndis e E. maidenii. A vasta gama de compostos fenólicos identificados em cada extrato analisado, assim como as prominentes atividades antioxidantes, todas na mesma gama de valores do bem conhecido antioxidante comercial, ácido ascórbico, são claramente um grande contributo para a valorização destes subprodutos industriais.

The work presented in this thesis was developed in collaboration with a Portuguese company, BeyonDevices, devoted to pharmaceutical packaging, medical technology and device industry. Specifically, the composition impact and surface modification of two polymeric medical devices from the company were studied: inhalers and vaginal applicators. The polyethylene-based vaginal applicator was modified using supercritical fluid technology to acquire self-cleaning properties and prevent the transport of bacteria and yeasts to vaginal flora. For that, in-situ polymerization of 2-substituted oxazolines was performed within the polyethylene matrix using supercritical carbon dioxide. The cationic ring-opening polymerization process was followed by end-capping with N,N-dimethyldodecylamine. Furthermore, for the same propose, the polyethylene matrix was impregnated with lavender oil in supercritical medium. The obtained materials were characterized physical and morphologically and the antimicrobial activity against bacteria and yeasts was accessed. Materials modified using 2-substituted oxazolines showed an effective killing ability for all the tested microorganisms, while the materials modified with lavender oil did not show antimicrobial activity. Only materials modified with oligo(2-ethyl-2-oxazoline) maintain the activity during the long term stability. Furthermore, the cytotoxicity of the materials was tested, confirming their biocompatibilty. Regarding the inhaler, its surface was modified in order to improve powder flowability and consequently, to reduce powder retention in the inhaler´s nozzle. New dry powder inhalers (DPIs), with different needle’s diameters, were evaluated in terms of internal resistance and uniformity of the emitted dose. It was observed that they present a mean resistance of 0.06 cmH2O0.5/(L/min) and the maximum emitted dose obtained was 68.9% for the inhaler with higher needle´s diameter (2 mm). Thus, this inhaler was used as a test and modified by the coating with a commonly-used force control agent, magnesium stearate, dried with supercritical carbon dioxide (scCO2) and the uniformity of delivered dose tests were repeated. The modified inhaler showed an increase in emitted dose from 68.9% to 71.3% for lactose and from 30.0% to 33.7% for Foradil.

The work presented in this thesis explores novel routes for the processing of bio-based polymers, developing a sustainable approach based on the use of alternative solvents such as supercritical carbon dioxide (scCO2), ionic liquids (ILs) and deep eutectic solvents (DES). The feasibility to produce polymeric foams via supercritical fluid (SCF) foaming, combined with these solvents was assessed, in order to replace conventional foaming techniques that use toxic and harmful solvents. A polymer processing methodology is presented, based on SCF foaming and using scCO2 as a foaming agent. The SCF foaming of different starch based polymeric blends was performed, namely starch/poly(lactic acid) (SPLA) and starch/poly(ε-caprolactone) (SPCL). The foaming process is based on the fact that CO2 molecules can dissolve in the polymer, changing their mechanical properties and after suitable depressurization, are able to create a foamed (porous) material. In these polymer blends, CO2 presents limited solubility and in order to enhance the foaming effect, two different imidazolium based ILs (IBILs) were combined with this process, by doping the blends with IL. The use of ILs proved useful and improved the foaming effect in these starch-based polymer blends. Infrared spectroscopy (FTIR-ATR) proved the existence of interactions between the polymer blend SPLA and ILs, which in turn diminish the forces that hold the polymeric structure. This is directly related with the ability of ILs to dissolve more CO2. This is also clear from the sorption experiments results, where the obtained apparent sorption coefficients in presence of IL are higher compared to the ones of the blend SPLA without IL. The doping of SPCL with ILs was also performed. The foaming of the blend was achieved and resulted in porous materials with conductivity values close to the ones of pure ILs. This can open doors to applications as self-supported conductive materials. A different type of solvents were also used in the previously presented processing method. If different applications of the bio-based polymers are envisaged, replacing ILs must be considered, especially due to the poor sustainability of some ILs and the fact that there is not a well-established toxicity profile. In this work natural DES – NADES – were the solvents of choice. They present some advantages relatively to ILs since they are easy to produce, cheaper, biodegradable and often biocompatible, mainly due to the fact that they are composed of primary metabolites such as sugars, carboxylic acids and amino-acids. NADES were prepared and their physicochemical properties were assessed, namely the thermal behavior, conductivity, density, viscosity and polarity. With this study, it became clear that these properties can vary with the composition of NADES, as well as with their initial water content. The use of NADES in the SCF foaming of SPCL, acting as foaming agent, was also performed and proved successful. The SPCL structure obtained after SCF foaming presented enhanced characteristics (such as porosity) when compared with the ones obtained using ILs as foaming enhancers. DES constituted by therapeutic compounds (THEDES) were also prepared. The combination of choline chloride-mandelic acid, and menthol-ibuprofen, resulted in THEDES with thermal behavior very distinct from the one of their components. The foaming of SPCL with THEDES was successful, and the impregnation of THEDES in SPCL matrices via SCF foaming was successful, and a controlled release system was obtained in the case of menthol-ibuprofen THEDES.

The enantiomers of sulfoxide proton pump inhibitors - omeprazole, lansoprazole, rabeprazole and Ro 18-5364 - were enantiomerically separated by liquid chromatography at multimilligram scale on a poly saccharide-based chiral stationary phase using normal and polar organic conditions as mobile phase. The values of the recovery and production rate were significant for each enantiomer; better results were achieved using a solid-phase injection system. However, this system was applied just for the enantionteric separation of omeprazole to demonstrate the applicability of this injection mode at milligram scale. The chiroptical characterization of the compounds was performed using a polarimeter and a circular dichroism detector. The higher enantiomeric purity obtained for the isolated enantiomers suggests that the methods here described should be considered as a simple and rapid way to obtain enantiomeric pure standards for analytical purpose. Sonohydrolysis of mixtures of tetraethoxysilane (TEOS) and tetramethoxysilane (TMOS) with different TMOS/(TMOS + TEOS) molar ratio R was carried out to obtain similar to 2.0 x 10(-3) mol SiO2/cm(3) and similar to 86%-volume liquid phase wet gels. Aerogels were obtained by supercritical CO2 extraction in autoclave. The samples were analyzed by small-angle X-ray scattering (SAXS) and nitrogen adsorption. The structure of the wet gels can be described as a mass fractal structure with fractal dimension D similar to 2.2 and characteristic length increasing from similar to 4.6 nm for pure TEOS to similar to 6.4 nm for pure TMOS. A fraction of the porosity is eliminated with the supercritical process. The fundamental role of the TMOS/(TMOS + TEOS) molar ratio on the structure of the aerogels is to increase the porosity and the pore mean size as R changes from pure TEOS to pure TMOS. The supercritical process increases the mass fractal dimension and shortens the fractality domain in the mesopore region. A secondary structure appearing in the micropore region of the aerogels can be described as a mass/surface fractal structure with correlated mass fractal dimension D-m similar to 2.6 and surface fractal dimension D-s similar to 2.3. About similar to 2.1 x 10(-3) Mol SiO2 cm(-3) and similar to 88%-volume liquid-phase silica wet gels were prepared from oxalic-acid-catalyzed tetraethoxysilane (TEOS) sonohydrolysis. Aerogels were obtained by supercritical CO2 extraction. The samples were analyzed by thermogravimetry, small-angle X-ray scattering and nitrogen adsorption. Wet gels can be described as mass fractal structures with fractal dimension D similar to 1.94 and structural characteristic length zeta changing between similar to 3.3 to similar to 3.0 nm in the studied range of the catalyst concentration. A fraction of the porosity is apparently eliminated in the supercritical process. The values of the BET specific surface S-BET, the total pore volume V-p and the mean pore size l(p) of the aerogels were found to change almost randomly around the mean values S-BET = 874 m(2) g(-1), V-p = 0.961 cm(3) g(-1) and l(p) = 4.4 nm with catalyst concentration variation. Wet silica gels with similar to 1.4 x 10(-3) mol SiO2/cm(3) and similar to 92 vol% liquid phase were obtained from sonohydrolysis of tetraethoxysilane (TEOS) with different additions of isopropyl alcohol ( IPA). The IPA/TEOS molar ratio R was changed from 0 to 4. Aerogels were obtained by supercritical CO2 extraction. The samples were analyzed by small-angle X-ray scattering (SAXS) and nitrogen adsorption. The wet gels exhibit mass fractal structure with fractal dimension increasing from D similar to 2.10 to D similar to 2.22, characteristic length xi decreasing from similar to 9.5 to similar to 6.9 nm, as R increases from 0 to 4, and an estimated characteristic length for the primary silica particles lower than similar to 0.3 nm. The supercritical process apparently eliminates a fraction of the porosity, increasing the mass fractal dimension and shortening the fractality domain in the mesopore region. The fundamental role of isopropyl alcohol on the structure of the resulting aerogels is to decrease the porosity and the pore mean size as R changes from pure TEOS to R = 4. A secondary structure appearing in the micropore region of the aerogels can be described as a mass/surface fractal structure, with correlated mass fractal dimension D-m similar to 2.7 and surface fractal dimension D-s similar to 2.3, as inferred from SAXS and nitrogen adsorption data.

The sol-gel process is a method for obtaining vitreous or vitro-ceramic materials which, are prepared a sol and by drying the liquid phase. This technique has been used extensively for the preparation of glassy gels, films, fibers and particles from the hydrolysis and polycondensation reactions of metal alkoxides. The usual methods for drying are: evaporation drying (xerogels), freeze drying (criogéis) and via supercritical CO2 extraction (aerogels). In the present work, we studied the preparation of silica gels by the sol-gel process from the hydrolysis of alkoxides tetraethylorthosilicate (TEOS) and 3-glycidoxypropyltrimethoxysilane (GPTS). The hydrolysis was promoted from GPTS and TEOS in proportion (4: 1) under acidic conditions. The hydrolysis reaction was promoted inside a Becker at room temperature. After hydrolyses the prepared sol had pH 2, and kept under mechanical agitation for a period of 1 hour. In order to accelerate the polycondensation reaction, the pH was corrected to a value near 5 by slowly adding NH4OH. Then the sols were leaked in sealed polycarbonate containers and maintained for 20 days at 40°C for gelation. Silica aerogels were prepared via supercritical CO2 extraction of the wet gel at temperature and pressure higher than 31°C and 74 atm, in an autoclave specially developed for the process. The structural characteristics were studied in the dry gel (aerogel). Aerogels were then characterized by nitrogen adsorption and small angle light scattering. The nitrogen adsorption data were analyzed for the determination of the BET specific surface (SBET), the total pore volume Vp, the pore mean size (lP=4Vp/SBET), the particle mean size (lS) and the pore size distribution (PSD). And the data from small angle light scattering were analyzed to determine the correlation function (γ'), the area per unit volume (S/V), average pore size (l ) and the average particle size...

Supercritical Emulsion Extraction technology (SEE-C) was proposed for the production of poly-lactic-co-glycolic acid microcarriers. SEE-C operating parameters as pressure, temperature and flow rate ratios were analyzed and the process performance was optimized in terms of size distribution and encapsulation efficiency. Microdevices loaded with bovine serum insulin were produced with different sizes (2 and 3 µm) or insulin charges (3 and 6 mg/g) and with an encapsulation efficiency of 60%. The microcarriers were characterized in terms of insulin release profile in two different media (PBS and DMEM) and the diffusion and degradation constants were also estimated by using a mathematical model. PLGA microdevices were also used in a cultivation of embryonic ventricular myoblasts (cell line H9c2 obtained from rat) in a FBS serum free medium to monitor cell viability and growth in dependence of insulin released. Good cell viability and growth were observed on 3 µm microdevices loaded with 3 mg/g of insulin. PLGA microspheres loaded with growth factors (GFs) were charged into alginate scaffold with human Mesenchimal Steam Cells (hMSC) for bone tissue engineering with the aim of monitoring the effect of the local release of these signals on cells differentiation. These “living” 3D scaffolds were incubated in a direct perfusion tubular bioreactor to enhance nutrient transport and exposing the cells to a given shear stress. Different GFs such as, h-VEGF, h-BMP2 and a mix of two (ratio 1:1) were loaded and alginate beads were recovered from dynamic (tubular perfusion system bioreactor) and static culture at different time points (1st, 7th, 21st days) for the analytical assays such as, live/dead; alkaline phosphatase; osteocalcin; osteopontin and Van Kossa Immunoassay. The immunoassay confirmed always a better cells differentiation in the bioreactor with respect to the static culture and revealed a great influence of the BMP-2 released in the scaffold on cell differentiation.

Results of direct geological and geochemical observations of the modern Rainbow hydrothermal field (Mid-Atlantic Ridge, 36°14'N; 33°54'W) carried out from the deep-sea manned Mir submersibles during Cruises 41 and 42 of the R/V Akademik Mstislav Keldysh in 1998-1999 and data of laboratory studies of collected samples are under consideration in the paper. The field lacks neovolcanic rocks and the axial part of the rift is filled in with a serpentinite protrusion. In this field there occur metalliferous sediments, as well as active and relict sulfide edifices composed of sulfide minerals; pyrrhotite, chalcopyrite, isocubanite, sphalerite, marcasite, pyrite, bornite, chalcosine, digenite, magnetite, anhydrite, rare troilite, wurtzite, millerite, and pentlandite have been determined. Sulfide ores are characterized by concentric-zoned textures. During in situ measurements during 35 minutes temperature of hydrothermal fluids was varying within a range from 250 to 350°C. Calculated chemical and isotopic composition of hydrothermal fluid shows elevated concentrations of Cl, Ni, Co, CH4, and H2. Values of d34S of H2S range from +2.4 to +3.1 per mil, of d13C of CH4 from -15.2 to -11.2 per mil, and d13C of CO2 from +1.0 to -4.0 per mil. Fluid inclusions are homogenized at temperatures from 140 to 360°C, whereas salinity of the fluid varies from 4.2 to 8.5 wt %. d34S values of sulfides range from +1.3 to +12.5 per mil. 3He/4He ratio in mineral-forming fluid contained in the fluid inclusions from sulfides of the Rainbow field varies from 0.00000374 to 0.0000101. It is shown that hydrothermal activity in the area continues approximately during 100 ka. It is assumed that the fluid and sulfide edifices contain components from the upper mantle. A hypothesis of phase separation of a supercritical fluid that results in formation of brines is proposed. Hydrothermal activity is related to the tectonic, not volcanic, phase of the Mid-Atlantic Ridge evolution.

CO2 capture and storage (CCS) projects are presently developed to reduce the emission of anthropogenic CO2 into the atmosphere. CCS technologies are expected to account for the 20% of the CO2 reduction by 2050. One of the main concerns of CCS is whether CO2 may remain confined within the geological formation into which it is injected since post-injection CO2 migration in the time scale of years, decades and centuries is not well understood. Theoretically, CO2 can be retained at depth i) as a supercritical fluid (physical trapping), ii) as a fluid slowly migrating in an aquifer due to long flow path (hydrodynamic trapping), iii) dissolved into ground waters (solubility trapping) and iv) precipitated secondary carbonates. Carbon dioxide will be injected in the near future (2012) at Hontomín (Burgos, Spain) in the frame of the Compostilla EEPR project, led by the Fundación Ciudad de la Energía (CIUDEN). In order to detect leakage in the operational stage, a pre-injection geochemical baseline is presently being developed. In this work a geochemical monitoring design is presented to provide information about the feasibility of CO2 storage at depth.

Coal fired power generation will continue to provide energy to the world for the foreseeable future. However, this energy use is a significant contributor to increased atmospheric CO2 concentration and, hence, global warming. Capture and disposal Of CO2 has received increased R&D attention in the last decade as the technology promises to be the most cost effective for large scale reductions in CO2 emissions. This paper addresses CO2 transport via pipeline from capture site to disposal site, in terms of system optimization, energy efficiency and overall economics. Technically, CO2 can be transported through pipelines in the form of a gas, a supercritical. fluid or in the subcooled liquid state. Operationally, most CO2 pipelines used for enhanced oil recovery transport CO2 as a supercritical fluid. In this paper, supercritical fluid and subcooled liquid transport are examined and compared, including their impacts on energy efficiency and cost. Using a commercially available process simulator, ASPEN PLUS 10.1, the results show that subcooled liquid transport maximizes the energy efficiency and minimizes the Cost Of CO2 transport over long distances under both isothermal and adiabatic conditions. Pipeline transport of subcooled liquid CO2 can be ideally used in areas of cold climate or by burying and insulating the pipeline. In very warm climates, periodic refrigeration to cool the CO2 below its critical point of 31.1 degrees C, may prove economical. Simulations have been used to determine the maximum safe pipeline distances to subsequent booster stations as a function of inlet pressure, environmental temperature and ground level heat flux conditions. The ability to tune the structural and chemical properties of colloidal nanoparticles (NPs), make them highly advantageous for studying activity and selectivity dependent catalytic behaviour. Incorporating pre-synthesized colloidal NPs into porous supports materials remains a challenge due to poor wetting and pore permeability. In this report monodisperse, composition controlled AgPd alloy NPs were synthesised and embedded into SBA-15 using supercritical carbon dioxide and hexane. Supercritical fluid impregnation resulted in high metal loading without the requirement for surface pre-treatments. The catalytic activity, reaction profiles and recyclability of the alloy NPs embedded in SBA-15 and immobilised on non-porous SiO2 are evaluated. The NPs incorporated within the SBA-15 porous network showed significantly greater recyclability performance compared to non-porous SiO2.

Recent studies have evaluated closed-loop supercritical carbon dioxide (s-CO2) Brayton cycles to be a higher energy density system in comparison to conventional superheated steam Rankine systems. At turbine inlet conditions of 923K and 25 MPa, high thermal efficiency (similar to 50%) can be achieved. Achieving these high efficiencies will make concentrating solar power (CSP) technologies a competitive alternative to current power generation methods. To incorporate a s-CO2 Brayton power cycle in a solar power tower system, the development of a solar receiver capable of providing an outlet temperature of 923 K (at 25 MPa) is necessary. The s-CO2 will need to increase in temperature by similar to 200 K as it passes through the solar receiver to satisfy the temperature requirements of a s-CO2 Brayton cycle with recuperation and recompression. In this study, an optical-thermal-fluid model was developed to design and evaluate a tubular receiver that will receive a heat input similar to 2 MWth from a heliostat field. The ray-tracing tool SolTrace was used to obtain the heat-flux distribution on the surfaces of the receiver. Computational fluid dynamics (CFD) modeling using the Discrete Ordinates (DO) radiation model was used to predict the temperature distribution and the resulting receiver efficiency. The effect of flow parameters, receiver geometry and radiation absorption by s-CO2 were studied. The receiver surface temperatures were found to be within the safe operational limit while exhibiting a receiver efficiency of similar to 85%.
