Antibody response to Salmonella typhi O and H antigens was evaluated in 24 individuals with either hepatointestinal or hepatosplenic schistosomiasis mansoni before and after typhoid vaccination, and compared with that of non-infected controls. Before vaccination, Schistosoma-infected patients showed a higher frequency of positive antibody to O antigen and the same frequency to H antigen when compared with that of healthy individuals. However, those with hepatosplenic schistosomiasis showed higher titres of antibody to H antigen than those with hepatointestinal disease or healthy individuals. Infected subjects, particularly those with hepatointestinal disease, showed a decreased response after typhoid vaccine. Tins diminished ability to mount an immune response towards typhoid antigens dining schistosomiasis may interfere ivith the clearance of the bacteria from blood stream and, therefore, play a role in the prolonged survival of salmonella as obsewed in some patients with chronic salmonellosis associated with schistosomiasis.

We investigated the influence of Salmonella typhimurium load and specific antibodies on phagocytosis in schistosomiasis. Macrophages from Schistosoma mansoni-infected mice showed depressed capacity to increase the phagocytosis in the presence of a high bacterial load, due to a reduced involvement of these cells in phagocytosis and to a deficient ability to increase the number of phagocytosed bacteria. Normal and Salmonella-infected mice increased their phagocytic capacity when exposed to a high bacterial load. Antibody to Salmonella increased the phagocytic capacity of macrophages from Schistosoma-infected mice due to an increase in the number of bacteria phagocytosed but caused no modification in the number of macrophages engaged in phagocytosis. Our data indicate that macrophages from Schistosoma-infected mice work close to their functional limit, since no increase in phagocytosis was observed after increasing the bacterial load. Specific antibodies can improve their phagocytic capacity and, therefore, could help clearing concurrent infection.

Optimization of the RAPD reaction for characterizing Salmonella enterica serovar Typhi strains was studied in order to ensure the reproducibility and the discriminatory power of this technique. Eight Salmonella serovar Typhi strains isolated from various regions in Brazil were examined for the fragment patterns produced using different concentrations of DNA template, primer, MgCl2 and Taq DNA polymerase. Using two different low stringency thermal cycle profiles, the RAPD fingerprints obtained were compared. A set of sixteen primers was evaluated for their ability to produce a high number of distinct fragments. We found that variations associated to all of the tested parameters modified the fingerprinting patterns. For the strains of Salmonella enterica serovar Typhi used in this experiment, we have defined a set of conditions for RAPD-PCR reaction, which result in a simple, fast and reproducible typing method.

INTRODUCTION: Salmonella sp infections have been reported over recent years in hospitals in Argentina and other countries due to multiresistant strains. The aim of this study was to characterize the extended-spectrum β-lactamases in third-generation cephalosporin-resistant strains of Salmonella enterica serovar Oranienburg. METHODS: We studied 60 strains isolated from children with gastroenteritis and/or extraintestinal complications. The antibiotic susceptibility patterns of the isolates were analyzed and the β-lactamases were characterized using phenotyping and genotyping methods. RESULTS: All the strains were resistant to ampicillin, cefotaxime, cefepime and aztreonam and partially susceptible to ceftazidime, thus corresponding well with the resistance phenotype conferred by CTX-M-type β-lactamases. An isoelectric point enzyme (pI = 7.9) was detected in all of the strains, and this was confirmed by PCR as a member of the CTX-M-2 group. CONCLUSIONS: This is the first report of Salmonella enterica serovar Oranienburg producing β-lactamases of the CTX-M-2 group in a pediatric hospital in Tucumán, Argentina.

Salmonella enterica serovars are Gram-negative facultative intracellular bacterial pathogens that infect a wide variety of animals. Salmonella infections are common in humans, causing usually typhoid fever and gastrointestinal diseases. Salmonella enterica serovar Typhimurium (S. Typhimurium), which is a leading cause of human gastroenteritis, has been extensively used to study the molecular pathogenesis of Salmonella, because of the availability of sophisticated genetic tools, and of suitable animal and tissue culture models mimicking different aspects of Salmonella infections.(...)

INTRODUCTION:In Venezuela, acute diarrheic syndrome (ADS) is a primary cause of morbi-mortality, often involving the Salmonella genus. Salmonella infections are associated with acute gastroenteritis, one of the most common alimentary intoxications, and caused by the consumption of contaminated water and food, especially meat. METHODS: Conventional and molecular methods were used to detect Salmonella strains from 330 fecal samples from individuals of different ages and both sexes with ADS. Polymerase chain reaction (PCR) was used for the molecular characterization of Salmonella, using invA, sefA, and fliC genes for the identification of this genus and the serotypes Enteritidis and Typhimurium, respectively. RESULTS: The highest frequency of individuals with ADS was found in children 0-2 years old (39.4%), and the overall frequency of positive coprocultures was 76.9%. A total of 14 (4.2%) strains were biochemically and immunologically identified as Salmonella enterica subsp. enterica, of which 7 were classified as belonging to the Enteritidis serotype, 4 to the Typhimurium serotype, and 3 to other serotypes. The S. enterica strains were distributed more frequently in the age groups 3-4 and 9-10 years old. CONCLUSIONS: The molecular characterization method used proved to be highly specific for the typing of S. enterica strains using DNA extracted from both the isolated colonies and selective enrichment broths directly inoculated with fecal samples, thus representing a complementary tool for the detection and identification of ADS-causing bacteria.

INTRODUCTION: In December 2001, an outbreak of foodborne gastroenteritis infected 114 of 161 people who ate at a restaurant in Aracaju, State of Sergipe, Brazil. METHODS: The epidemiological and microbiological aspects of the outbreak were characterized. RESULTS: Potato salad made with homemade mayonnaise and stored at unsuitable temperatures was associated with increased risk of foodborne infection. Salmonella Enteritidis was isolated from the diarrheal stools of the hospitalized patients, and genotyping of the fecal samples generated identical randomly amplified polymorphic deoxyribonucleic acid (DNA) profiles. CONCLUSIONS : To the best of our knowledge, this is the first and the only record of a gastrointestinal outbreak in Sergipe.

A presente pesquisa avaliou a contaminação em carcaças de frango por salmonelas. O estudo foi realizado em feiras e mercados nas diferentes zonas da Cidade de Manaus-AM, por um período de 11 semanas (março a maio de 1998). Sessenta amostras foram examinadas; destas, trinta (50%) resultaram positivas para Salmonella spp., das quais foram isoladas 67 cepas, incluídas em 11 sorotipos diversos. Entre os sorotipos identificados, a S. panama, S. mbandaka, S. schwarzengrund, S. typhimurium, S. albany e S. agona, foram as predominantes, representando 85% do total isolado. A contaminação de Salmonella é dependente de vários fatores, e a sua ocorrência pode estar relacionada com as condições de higiene da granja. A metodologia empregada para a detecção microbiológica foi a recomendada pela "Food and Drug Administration", dos Estados Unidos da América.

Na presente investigação, os autores estudaram a distribuição dos lisótipos Ví, tipos fermentativos e tetrationato-redutase, em 110 amostras de Salmonella typhi, isoladas no período de 1966-1970, no Estado da Guanabara. Para estabelecer uma melhor configuração epidemiológica dos resultados, fez-se a divisão do Estado em seis regiões, respectivamente as zonas Sul, Centro, Norte, da E. F. Central, da E. F. Leopoldina e rural. Obteve-se no cômputo geral, a prevalência dos lisótipos A (59,09%), E 1a (19,09%), T (7,27%) e amostras Vi negativas (4,55%). Verificou-se ainda, que cerca de 70,90% das culturas foram classificadas no tipo fermentativo II, segundo esquema de Kristensen e 95,42% das amostras revelaram-se positivas na prova de detecção da tetrationato-redutase.

Examinando-se 158 amostras de águas cloacais, provenientes de uma estação de tratamento e de uma elevatória de esgotos, localizadas na cidade do Rio de Janeiro, fêz-se uma avaliação da eficiência de quatro meios de enriquecimento para Salmonella. Os meios empregados foram o caldo tetrationato, segundo Kauffmann, o meio de Rappaport e duas modificações deste meio, previamente descritas. Os resultados encontrados, evidenciaram a superioridade de um dos meios modificados da Rappaport, em que se fez a substituição do verde malaquiata por outro corante bacteriostático (Metachromgelb II RD) e com a inclusão de tetrationato, à fórmula original. A maior eficiência assinalada para este meio se baseou nos resultados obtidos do enriquecimento dos diferentes grupos sorológicos e particularmente dos quinze sorotipos mais incidentes de Salmonella enteritidis, na presente investigação.

Durante o período de 1962 a 1971, foram identificadas 164 amostras de salmonelas, isoladas a partir de fezes de crianças e de adultos, possuidores ou não, de problemas entéricos e residentes na cidade do Rio de Janeiro. Os resultados obtidos na caracterização sorológica dessas culturas, evidenciaram uma nítida predominância de Salmonella enteritidis, pertencentes ao grupo sorológico B. Dentre os sorotipos de Salmonella enteritidis, que se destacaram pela maior frequência de isolamentos, citam-se os seguintes: Typhimurium Newport, Anatum e Thompson.
