The stingless bee Partamona helleri in southeast Brazil shows the regular chromosome number 2n = 34 and a variable number of up to four minute B1 or B2 chromosomes. Previous cytogenetic analyses have indicated morphological similarities between the B1 chromosome and chromosome segments in the regular karyotype. In this study, microdissection and chromosome painting were employed along with C banding, NOR banding, and base-specific fluorochrome staining to investigate the origin of the B1 chromosome in P. helleri. B1-generated probe hybridized exclusively to B1 chromosomes. This result suggests an independent origin from the regular karyotype or, alternatively, that the B chromosome may have suffered substantial genetic alterations along its independent evolution. The absence of higher dosages of these small B chromosomes in this population of P. helleri may be related to the existence of either a genetic or cytogenetic constraint in the establishment of such high numbered karyotypes. © 2012 INRA, DIB and Springer-Verlag, France.

Studies of sub-lethal effects of pesticide residues on stingless bees are scarce and morphological analysis of organs would add information to toxicological analysis in order to clarify the continuous exposure of Scaptotrigona postica to insecticides. The aim of this study was to evaluate the morphology and histochemistry of the Malpighian tubules (excretory organ) of S. postica exposed to fipronil or boric acid to detect cellular responses that indicate toxicity or adaptative mechanisms to stress induced by exposure of worker bees to low doses of these chemical compounds. Newly emerged bees were submitted to toxicological bioassays and morphological analyses by optical microscopy and Transmission Electron Microscopy, as well as histochemical methods, were performed to detect proteins and glycoconjugates. Additionally, immunohistochemical detection of DNA fragmentation and HSP70 (70-kDa Heat shock protein) were performed to detect cell death and stress response, respectively. Statistical analysis, for the bioassays conducted with ingestion of contaminated diet with boric acid at 0.75% (w/w) or with fipronil at 0.1μg/kg of food, showed that the survival of bees that ingested the contaminated diets were significantly different to the survival rate presented by the control group (P<0.0001). Although some characteristics indicative of initiation of cell death were observed, the cells remained metabolically active in the processes of excretion and inactivation of chemical compounds. The data from this study reinforce the importance of research on sublethal effects of low doses of pesticides on bees in an attempt to assess a possible realistic dose and evaluate the risk assessment of stingless bee S. postica foraging in the vicinity of cultivated fields and/or in green urban areas. © 2013 Elsevier Ltd.

A criação de abelhas sem ferrão é antiga nas Américas. No entanto, a atividade ainda esta subdesenvolvida, necessitando de melhorias nas técnicas de manejo. Este estudo avaliou alternativas nutricionais para a abelha amazônica Melipona flavolineata, visando substituição da alimentação com mel e pólen fermentado. As seguintes alternativas nutricionais foram comparadas com o mel (T1): xarope de açúcar invertido com minerais (T2) e este xarope acrescido de solução de aminoácidos e vitaminas nas seguintes concentrações: 0,5% (T3), 1,0% (T4) e 10% (T5). Ao pólen fermentado (T1) foram comparados alimentos fermentados com inóculo de pólen fermentado de M. flavolineata, constituídos de: pólen apícola (T2); levedo (T3); e extrato de soja, em duas diferentes concentrações de proteínas (T4, 12% e T5, 18%). O valor nutricional das dietas foi baseado no consumo diário, no peso das operárias, no tamanho dos acinos das glândulas hipofaringeas e no tamanho dos oócitos. Avaliaram-se, tambem, os custos dos ingredientes das dietas. Os resultados obtidos indicaram que dentre as alternativas para o mel, não houve diferenças estatísticas entre o controle e todos os tratamentos. Foram encontradas diferenças apenas entre os tamanhos dos acinos das operárias alimentadas com os tratamentos 2 e 4, sendo o tratamento 4 superior. Os custos dos ingredientes indicaram o tratamento 2 como sendo mais barato que os demais. Dentre as alternativas para o sabura foram encontradas maiores diferenças. O consumo do tratamento 4 foi significativamente superior ao do T3, indicando maior aceitação do extrato de soja em relação ao levedo. Em relação aos demais parâmetros avaliados, o tratamento 5 foi superior a todos os demais e seu custo foi muito inferior ao do T2 e semelhante ao dos T3 e T4. Concluiu-se que as melhores alternativas para a nutrição de M. flavolineata sao o xarope de acucar invertido com minerais, e o pólen fermentado semi-artificial à base de extrato de soja. Colônias recém-formadas, com 500-600 abelhas, devem receber 10 a 20 ml de xarope e 25 g de pólen fermentado semi-artificial semanalmente.

ABSTRACT: Honey produced by three stingless bee species (Melipona flavolineata, M. fasciculata and Apis mellifera) from different regions of the Amazon was analyzed by separating phenolic acids and flavonoids using the HPLC technique. Data were subjected to multivariate statistical analysis (PCA, HCA and DA). Results showed the three species of honey samples could be distinguished by phenolic composition. Antioxidant activity of the honeys was determined by studying the capacity of inhibiting radicals using DPPH assay. Honeys with higher phenolic compound contents had greater antioxidant capacity and darker color.

Vitellogenin (Vg) is an egg yolk protein that is produced primarily in the fat body of most female insects. In the advanced social structure of eusocial honeybees, the presence of the queen inhibits egg maturation in the workers ovaries. However in the stingless bee Melipona quadrifasciata, the workers always develop ovaries and lay a certain amount of eggs while provisioning the brood cells with larval food during what is known as the worker nurse phase. The present work is a comparative study of the presence of Vg in homogenates of the fat bodies and ovaries of the nurse workers, and the virgin and physogastric queens of M. quadrifasciata. The presence of Vg was determined by sodium dodecyl sulfate polyacrylamide gel electrophoresis and immunoblotting using Apis mellifera anti-egg antibody. Vg was not detected in the fat bodies or ovaries of the workers, but it was found in the ovaries of virgin and physogastric queens and in the fat body of physogastric queens. The results are discussed, taking into account the reproductive state of the individuals and the other possible roles of Vg, such as a storage protein for metoabolism of other organs.

Flight activity of foragers of four colonies of Plebeia remota (Holmberg, 1903) was registered from December 1998 to December 1999, using an automated system (photocells and PLC system). The colonies originated from two different regions: Cunha, state of Sao Paulo, and Prudentopolis, state of Parana, Brazil. Flight activity was influenced by different climatic factors in each season. In the summer, the intensity of the correlations between flight activity and climatic factors was smaller than in the other seasons. During the autumn and winter, solar radiation was the factor that most influenced flight activity, while in the spring, this activity was influenced mainly by temperature. Except in the summer, the various climatic factors similarly influenced flight activity of all of the colonies. Flight activity was not affected by geographic origin of the colonies. Information concerning seasonal differences in flight activity of P. remota will be useful for prediction of geographic distribution scenarios under climatic changes.

Ethnopharmacological relevance: The pharmacological activity of geopropolis collected by stingless bees (important and threatened pollinators), a product widely used in folk medicine by several communities in Brazil, especially in the Northeast Region, needs to be studied. Objective: The aim of this study was to evaluate the antinociceptive activity of Melipona scutellaris geopropolis (stingless bee) using different models of nociception. Material and methods: The antinociceptive activity of the ethanolic extract of geopropolis (EEGP) and fractions was evaluated using writhing induced by acetic acid, formalin test, carrageenan-induced hypernociception, and quantification of IL-1 beta and TNF-alpha. The chemical composition was assessed by quantification of total flavonoids and phenolic compounds. Results: EEGP and its hexane and aqueous fractions showed antinociceptive activity. Both EEGP and its aqueous fraction presented activity in the mechanical inflammatory hypernociception induced by the carrageenan model, an effect mediated by the inhibition of IL-1 beta and TNF-alpha. The chemical composition of EEGP and its hexane and aqueous fractions showed a significant presence of phenolic compounds and absence of flavonoids. Conclusion: Our data indicate that geopropolis is a natural source of bioactive substances with promising antinociceptive activity. (C) 2012 Elsevier Ireland Ltd. All rights reserved.

The aim of the present study was to characterize the variation of the chemical profiles among workers in different colonies of the stingless bee Melipona marginata. We used gas chromatography and mass spectrometry (CG-MS) and multivariate analysis of the bees' chemical from three colonies of two localities in southeast Brazil. The results showed that cuticular hydrocarbon profiles clearly separated distinct colonies. We show here the importance of using the chemical analyses for characterization of colony membership, in addition of the traditional techniques of diversity analyses.

O Brasil é um dos maiores produtores mundiais de mel, no qual sua produção é baseada principalmente na criação da espécie exótica Apis mellifera. A produção de mel da Apis mellifera é cerca de 10 vezes maior que das espécies de abelhas sem ferrão, contudo, o mel de abelhas nativas possui maior valor comercial. Embora pouco explorado, o mel de abelhas sem ferrão desperta interesse em indústrias de cosméticos e medicinas naturais. A sua produção se apresenta como uma ferramenta com grande potencial para agregar valor econômico aos ecossistemas brasileiros, em especial os florestais, de forma sustentável e com menor potencial de influências de contaminantes traços. A qualidade química do mel é um importante requisito comercial, principalmente o destinado à exportação. Como exemplo, a União Européia em 2006 decidiu suspender a importação do mel produzido no Brasil sob alegação de que o país não possuía equivalência ao bloco quanto as diretrizes para o controle de resíduos e qualidade do produto. Diante do potencial de produção comercial sustentável do mel de abelhas nativas brasileiras e a falta de conhecimento sobre possíveis resíduos encontrados em sua composição, em especial os elementos traços, como objetivo principal deste trabalho pretendeu-se caracterizar a composição de elementos químicos do mel de abelhas sem ferrão, comparar com o de Apis mellifera e verificar as possíveis variações causadas pelo ambiente. Este estudo investigou a composição química dos méis de abelhas sem ferrão de cinco estados brasileiros: Bahia, Minas Gerais, Rio Grande do Norte, Santa Catarina e São Paulo; compreendendo um total de 70 colméias de diferentes espécies: Melipona quadrifasciata, Melipona scutelaris, Melipona mandacaia, Melipona capixaba, Melipona rufiventris, Melipona compressipes, Melipona bicolor, Nannotrigona testaceicornis, Tetragona clavipes, Tetragonisca angustula e Scaptotrigona sp.. Pólen, a principal fonte de minerais para a colméia, e as próprias abelhas foram também coletadas para estudos de composição e correlação com os méis. A análise por ativação neutrônica instrumental permitiu a determinação de Br, Ca, Co, Cs, Fe, La, Na, Rb, Sc e Zn nos méis, Br, Ca, Co, Cs, Fe, K, La, Na, Rb, Sc, Se e Zn nas amostras de pólen e As, Br, Co, Cr, Cs, Fe, K, La, Na, Rb, Sb, Sc, Se e Zn em abelhas. Méis das abelhas da subtribo trigonina apresentaram maiores concentrações dos elementos alcalinos. Alta razão K/Na foram observadas nas amostras de mel e pólen. Pólen se apresentou como uma grande fonte de P e Se. Análises quimiométricas indicaram os méis e abelhas como bons indicadores de atividades antrópicas. Arsênio apareceu nas abelhas coletadas em áreas de maior atividade antrópica. Como resultado, este estudo tem demonstrado o potencial nutracêutico do mel e pólen meliponícola e o potencial das abelhas nativas como ferramentas de avaliação da qualidade ambiental. A proximidade a atividades antrópicas mostrou-se fator decisivo para concentrações mais elevadas de As mas abelhas
