Portugal é um dos países europeus com melhor cobertura espacial e populacional de rede de autoestradas (5º entre os 27 da UE). O acentuado crescimento desta rede nos últimos anos leva a que seja necessária a utilização de metodologias de análise e avaliação da qualidade do serviço que é prestado nestas infraestruturas, relativamente às condições de circulação. Usualmente, a avaliação da qualidade de serviço é efetuada por intermédio de metodologias internacionalmente aceites, das quais se destaca a preconizada no Highway Capacity Manual (HCM). É com esta última metodologia que são habitualmente determinados em Portugal, os níveis de serviço nas diversas componentes de uma autoestrada (secções correntes, ramos de ligação e segmentos de entrecruzamento). No entanto, a sua transposição direta para a realidade portuguesa levanta algumas reservas, uma vez que os elementos que compõem o ambiente rodoviário (infraestrutura, veículo e condutor) são distintos dos da realidade norte-americana para a qual foi desenvolvida. Assim, seria útil para os atores envolvidos no setor rodoviário dispor de metodologias desenvolvidas para as condições portuguesas, que possibilitassem uma caracterização mais realista da qualidade de serviço ao nível da operação em autoestradas. No entanto, importa referir que o desenvolvimento de metodologias deste género requer uma quantidade muito significativa de dados geométricos e de tráfego, o que acarreta uma enorme necessidade de meios, quer humanos, quer materiais. Esta abordagem é assim de difícil execução, sendo por isso necessário recorrer a metodologias alternativas para a persecução deste objetivo. Ultimamente tem-se verificado o uso cada vez mais generalizado de modelos de simulação microscópica de tráfego, que simulando o movimento individual dos veículos num ambiente virtual permitem realizar análises de tráfego. A presente dissertação apresenta os resultados obtidos no desenvolvimento de uma metodologia que procura recriar, através de simuladores microscópicos de tráfego, o comportamento das correntes de tráfego em secções correntes de autoestradas com o intuito de, posteriormente, se proceder à adaptação da metodologia preconizada no HCM (na sua edição de 2000) à realidade portuguesa. Para tal, com os simuladores microscópicos utilizados (AIMSUN e VISSIM) procurou-se reproduzir as condições de circulação numa autoestrada portuguesa, de modo a que fosse possível analisar as alterações sofridas no comportamento das correntes de tráfego após a modificação dos principais fatores geométricos e de tráfego envolvidos na metodologia do HCM 2000. Para o efeito, realizou-se uma análise de sensibilidade aos simuladores de forma a avaliar a sua capacidade para representar a influência desses fatores, com vista a, numa fase posterior, se quantificar o seu efeito para a realidade nacional e dessa forma se proceder à adequação da referida metodologia ao contexto português. Em resumo, o presente trabalho apresenta as principais vantagens e limitações dos microssimuladores AIMSUN e VISSIM na modelação do tráfego de uma autoestrada portuguesa, tendo-se concluído que estes simuladores não são capazes de representar de forma explícita alguns dos fatores considerados na metodologia do HCM 2000, o que impossibilita a sua utilização como ferramenta de quantificação dos seus efeitos e consequentemente inviabiliza a adaptação dessa metodologia à realidade nacional. São, no entanto, referidas algumas indicações de como essas limitações poderão vir a ser ultrapassadas, com vista à consecução futura dessa adequação.

The objective of this thesis is to study the properties of resistive switching effect based on bistable resistive memory which is fabricated in the form of Al2O3/polymer diodes and to contribute to the elucidation of resistive switching mechanisms. Resistive memories were characterized using a variety of electrical techniques, including current-voltage measurements, small-signal impedance, and electrical noise based techniques. All the measurements were carried out over a large temperature range. Fast voltage ramps were used to elucidate the dynamic response of the memory to rapid varying electric fields. The temperature dependence of the current provided insight into the role of trapped charges in resistive switching. The analysis of fast current fluctuations using electric noise techniques contributed to the elucidation of the kinetics involved in filament formation/rupture, the filament size and correspondent current capabilities. The results reported in this thesis provide insight into a number of issues namely: (i) The fundamental limitations on the speed of operation of a bi-layer resistive memory are the time and voltage dependences of the switch-on mechanism. (ii) The results explain the wide spread in switching times reported in the literature and the apparently anomalous behaviour of the high conductance state namely the disappearance of the negative differential resistance region at high voltage scan rates which is commonly attributed to a “dead time” phenomenon which had remained elusive since it was first reported in the ‘60s. (iii) Assuming that the current is filamentary, Comsol simulations were performed and used to explain the observed dynamic properties of the current-voltage characteristics. Furthermore, the simulations suggest that filaments can interact with each other. (iv) The current-voltage characteristics have been studied as a function of temperature. The findings indicate that creation and annihilation of filaments is controlled by filling and neutralizing traps localized at the oxide/polymer interface. (v) Resistive switching was also studied in small-molecule OLEDs. It was shown that the degradation that leads to a loss of light output during operation is caused by the presence of a resistive switching layer. A diagnostic tool that predicts premature failure of OLEDs was devised and proposed. Resistive switching is a property of oxides. These layers can grow in a number of devices including, organic light emitting diodes (OLEDs), spin-valve transistors and photovoltaic devices fabricated in different types of material. Under strong electric fields the oxides can undergo dielectric breakdown and become resistive switching layers. Resistive switching strongly modifies the charge injection causing a number of deleterious effects and eventually device failure. In this respect the findings in this thesis are relevant to understand reliability issues in devices across a very broad field.

Urban mobility is one of the main challenges facing urban areas due to the growing population and to traffic congestion, resulting in environmental pressures. The pathway to urban sustainable mobility involves strengthening of intermodal mobility. The integrated use of different transport modes is getting more and more important and intermodality has been mentioned as a way for public transport compete with private cars. The aim of the current dissertation is to define a set of strategies to improve urban mobility in Lisbon and by consequence reduce the environmental impacts of transports. In order to do that several intermodal practices over Europe were analysed and the transport systems of Brussels and Lisbon were studied and compared, giving special attention to intermodal systems. In the case study was gathered data from both cities in the field, by using and observing the different transport modes, and two surveys were done to the cities users. As concluded by the study, Brussels and Lisbon present significant differences. In Brussels the measures to promote intermodality are evident, while in Lisbon a lot still needs to be done. It also made clear the necessity for improvements in Lisbon’s public transports to a more intermodal passenger transport system, through integration of different transport modes and better information and ticketing system. Some of the points requiring developments are: interchanges waiting areas; integration of bicycle in public transport; information about correspondences with other transport modes; real-time information to passengers pre-trip and on-trip, especially in buses and trams. After the identification of the best practices in Brussels and the weaknesses in Lisbon the possibility of applying some of the practices in Brussels to Lisbon was evaluated. Brussels demonstrated to be a good example of intermodality and for that reason some of the recommendations to improve intermodal mobility in Lisbon can follow the practices in place in Brussels.

Regulatory light chain (RLC) phosphorylation in fast twitch muscle is catalyzed by skeletal myosin light chain kinase (skMLCK), a reaction known to increase muscle force, work, and power. The purpose of this study was to explore the contribution of RLC phosphorylation on the power of mouse fast muscle during high frequency (100 Hz) concentric contractions. To determine peak power shortening ramps (1.05 to 0.90 Lo) were applied to Wildtype (WT) and skMLCK knockout (skMLCK-/-) EDL muscles at a range of shortening velocities between 0.05-0.65 of maximal shortening velocity (Vmax), before and after a conditioning stimulus (CS). As a result, mean power was increased to 1.28 ± 0.05 and 1.11 ± .05 of pre-CS values, when collapsed for shortening velocity in WT and skMLCK-/-, respectively (n = 10). In addition, fitting each data set to a second order polynomial revealed that WT mice had significantly higher peak power output (27.67 ± 1.12 W/ kg-1) than skMLCK-/- (25.97 ± 1.02 W/ kg-1), (p < .05). No significant differences in optimal velocity for peak power were found between conditions and genotypes (p > .05). Analysis with Urea Glycerol PAGE determined that RLC phosphate content had been elevated in WT muscles from 8 to 63 % while minimal changes were observed in skMLCK-/- muscles: 3 and 8 %, respectively. Therefore, the lack of stimulation induced increase in RLC phosphate content resulted in a ~40 % smaller enhancement of mean power in skMLCK-/-. The increase in power output in WT mice suggests that RLC phosphorylation is a major potentiating component required for achieving peak muscle performance during brief high frequency concentric contractions.

Calcitonin receptor-like receptor (CLR) and receptor activity modifying protein 1 (RAMP1) comprise a receptor for calcitonin gene related peptide (CGRP) and intermedin. Although CGRP is widely expressed in the nervous system, less is known about the localization of CLR and RAMP1. To localize these proteins, we raised antibodies to CLR and RAMP1. Antibodies specifically interacted with CLR and RAMP1 in HEK cells coexpressing rat CLR and RAMP1, determined by Western blotting and immunofluorescence. Fluorescent CGRP specifically bound to the surface of these cells and CGRP, CLR, and RAMP1 internalized into the same endosomes. CLR was prominently localized in nerve fibers of the myenteric and submucosal plexuses, muscularis externa and lamina propria of the gastrointestinal tract, and in the dorsal horn of the spinal cord of rats. CLR was detected at low levels in the soma of enteric, dorsal root ganglia (DRG), and spinal neurons. RAMP1 was also localized to enteric and DRG neurons and the dorsal horn. CLR and RAMP1 were detected in perivascular nerves and arterial smooth muscle. Nerve fibers containing CGRP and intermedin were closely associated with CLR fibers in the gastrointestinal tract and dorsal horn, and CGRP and CLR colocalized in DRG neurons. Thus, CLR and RAMP1 may mediate the effects of CGRP and intermedin in the nervous system. However, mRNA encoding RAMP2 and RAMP3 was also detected in the gastrointestinal tract, DRG, and dorsal horn, suggesting that CLR may associate with other RAMPs in these tissues to form a receptor for additional peptides such as adrenomedullin.

With a rapidly increasing fraction of electricity generation being sourced from wind, extreme wind power generation events such as prolonged periods of low (or high) generation and ramps in generation, are a growing concern for the efficient and secure operation of national power systems. As extreme events occur infrequently, long and reliable meteorological records are required to accurately estimate their characteristics. Recent publications have begun to investigate the use of global meteorological “reanalysis” data sets for power system applications, many of which focus on long-term average statistics such as monthly-mean generation. Here we demonstrate that reanalysis data can also be used to estimate the frequency of relatively short-lived extreme events (including ramping on sub-daily time scales). Verification against 328 surface observation stations across the United Kingdom suggests that near-surface wind variability over spatiotemporal scales greater than around 300 km and 6 h can be faithfully reproduced using reanalysis, with no need for costly dynamical downscaling. A case study is presented in which a state-of-the-art, 33 year reanalysis data set (MERRA, from NASA-GMAO), is used to construct an hourly time series of nationally-aggregated wind power generation in Great Britain (GB), assuming a fixed, modern distribution of wind farms. The resultant generation estimates are highly correlated with recorded data from National Grid in the recent period, both for instantaneous hourly values and for variability over time intervals greater than around 6 h. This 33 year time series is then used to quantify the frequency with which different extreme GB-wide wind power generation events occur, as well as their seasonal and inter-annual variability. Several novel insights into the nature of extreme wind power generation events are described, including (i) that the number of prolonged low or high generation events is well approximated by a Poission-like random process, and (ii) whilst in general there is large seasonal variability, the magnitude of the most extreme ramps is similar in both summer and winter. Peculiar Early Permian palaeontological and sedimentological features are reviewed from South China, including characteristic Early Permian cold-water Gondwanan brachiopod taxa and faunas from Sichuan and Guizhou provinces, widespread rosettes and irregular aggregates of calcite prisms ('Chrysanthemum Stones') within the Qixia limestones, and lack of significant Early Permian reef buildups. The occurrences of these features are at odds with the currently widely held view that South China was located in a palaeotropical, warm-water setting throughout the Permian and hence harboured a highly diverse shallow marine biota. In this paper, I propose a working hypothesis, suggesting that influence of at least cool water masses may have intermittently occurred in South China during the Early Permian, which facilitated the formation of the cool water-influenced palaeontological and sedimentological features and promoted the interchanges of cool to cold water marine faunas between the Gondwanan and Boreal Realms. These cool water masses may have been transported to low-latitude regions as deep currents from northern and eastern shelves of Gondwanaland and upwelled along the western coast of South China as well as within the relatively deep-water basins of central South China. Prevalence of these meridional, north-directed deep cold water currents during the Early Permian may have been related to the glaciation event of Gondwanaland. An alternative and/or additional source of cooling may have also originated from strong easterly palaeoequatorial boundary currents operating within the Palaeotethys at times during the Early Permian, inducing and/or enhancing upwelling of cool to cold water masses in the eastern Palaeotethys. This latter scenario is analogous to the occasional 'La Nina' effect (opposite to the 'El Nino' effect) at the equatorial belt of the modern Pacific Ocean.

Frequent reviews of teacher education in recent times seem to indicate that those charged with the responsibility for teacher education have little understanding of the contemporary needs of teachers. Just how does educational research inform teachers in their day-to-day practice and how is its relevance tested? As an educational researcher Professor Shirley Grundy challenges her own notions of 'relevance' by returning from academia to the school system in Western Australia. This experience expanded her understanding of how conceptual frameworks and the realities of schools complemented each other. She reflects on the agenda for change faced by education systems and the ensuing transformation of curriculum planning, pedagogy, assessment and reporting procedures in some schools while not in others.

Her reference to Habermas' work relating to 'system' and 'lifeworld' provided her with an explanation for the social conditions of schooling, while her exploration of discourse theory provides some understanding of the practices related to the exercise of power in school settings. Interchanges such as that experienced by Grundy are to the mutual benefit of both parties.

A new brachiopod fauna is described from the Early and Middle Permian of Zadoi and Zhidoi counties, southern Qinghai (Changdu block), northwest China. This fauna includes 13 species in nine genera with Spinomarginifera concentrica n. sp. and Transennatia waterhousei n. sp. The Early to Middle Permian brachiopod fauna from southern Qinghai is very similar to the contemporary Cathaysian faunas of South China with which the new fauna shares 70 per cent of its species. On the other hand, the Qinghai brachiopods also demonstrate a significant link with the Permian brachiopod fauna of the Sino-Mongolian-Japanese Province in northeast China, as suggested by Marginifera septentrionalis and Attenuatella. In particular, the occurrence of the bipolar brachiopod genus Attenuatella suggests that southern Qinghai may have played an important role as a biogeographic stepping stone in the marine faunal interchanges between the northern and southern hemispheres during the Early and Middle Permian.

Receptor activity-modifying proteins (RAMPs) interact with and modify the behavior of the calcitonin receptor (CTR) and calcitonin receptor-like receptor (CLR). We have examined the contribution of the short intracellular C terminus, using constructs that delete the last eight amino acids of each RAMP. C-Terminal deletion of individual RAMPs had little effect on the signaling profile induced when complexed with CLR in COS-7 or human embryonic kidney (HEK)293 cells. Likewise, confocal microscopy revealed each of the mutant RAMPs translocated hemagglutinin-tagged CLR to the cell surface. In contrast, a pronounced effect of RAMP C-terminal truncation was seen for RAMP/CTRa complexes, studied in COS-7 cells, with significant attenuation of amylin receptor phenotype induction that was stronger for RAMP1 and -2 than RAMP3. The loss of amylin binding upon C-terminal deletion could be partially recovered with overexpression of Gαs, suggesting an impact of the RAMP C terminus on coupling of G proteins to the receptor complex. In HEK293 cells the c-Myc-RAMP1 C-terminal deletion mutant showed high receptor-independent cell surface expression; however, this construct showed low cell surface expression when expressed alone in COS-7 cells, indicating interaction of RAMPs with other cellular components via the C terminus. This mutant also had reduced cell surface expression when coexpressed with CTR. Thus, this study reveals important functionality of the RAMP C-terminal domain and identifies key differences in the role of the RAMP C terminus for CTR versus CLR-based receptors.

Nesta dissertação se propõe a analisar a política cultural do Brasil no exterior e em especial sua participação no processo de criação do Mercosul Cultural. O presente ensaio verifica até que ponto o Brasil conta com uma política cultural no exterior que possa realmente promover difundir e intercambiar a cultura brasileira em âmbito internacional. Buscou-se dentre os vários modelos existentes de política pública marcos teóricos que pudessem servir como parâmetros na análise da política cultural externa brasileira, utilizando-se principalmente as fases de uma política pública e o modelo ótimo normativo de Dror. De maneira geral o autor procurou examinar ao lado da análise da política cultural do Brasil no exterior o processo de integração internacional e a participação do Brasil na globalização enfatizando a cultura como fator fundamental no processo de regionalização promovido atualmente pela Argentina, Brasil, Paraguai e Uruguai.

Esta monografia é um estudo de desenvolvimento institucional que proporciona conhecer como uma organização da administração pública brasileira, criada com um fim especifico e transitório, sobreviveu, se transformou, criou raizes, expandiu sua área de influência e tornou-se uma instituição relativamente bem sucedida. Trata-se da Comissão Executiva do Plano da Lavoura Cacaueira - CEPLAC, órgão autônomo vinculado ao Ministério da Agricultura, criado na década de cinqüenta pelo Governo Federal, como solução de emergência para o apoio financeiro aos produtores de cacau do sul da Bahia, numa das maiores crises da economia cacaueira. O autor valeu-se, para sua dissertação, do quadro conceitual formulado por Milton J. Esman e Hans C. Blaise, da Inter-University Research Program in Institution Building. Este é um modelo que parte da premissa de que as instituições são sistema abertos e, como tal, mantêm padrões de relações e intercâmbio com o meio-ambiente em que operam. Importam energia do ambiente (inputs on entradas) e processam tais energias, transformando-as em produtos (outputs ou saldas) desejados e valorizados pelo ambiente. Dentro deste quadro, têm-se três elementos básicos: a organização; as transações; e o meio-ambiente. O trabalho foi dividido em cinco capítulos. No primeiro o autor discorreu sobre o tema e sua importância, objetivos do estudo, metodologia utilizada e o conteúdo da monografia. No segundo, fez uma discussão em torno do que é desenvolvimento institucional, sua evolução como estratégia de desenvolvimento; procedeu à revisão de literatura e descreveu o modelo que serviu de guia à pesquisa. Aí, registrou disfunções, limitações e restrições ao modelo de Esman e Blaise, e/ou aos efeitos da institucionalização, apontados por alguns estudiosos. No terceiro capítulo narrou os antecedentes, origem e evolução da CEPLAC, oportunidade em que identificou três fases da organização; no quarto, mostrou a CEPLAC através do modelo de desenvolvimento institucional, quando a organização foi analisada em função de suas variáveis institucionais (Liderança, Doutrina, Programa, Estrutura I nterna e Recursos), suas t ra n sações com o ambiente e suas variáveis institucionais-ambientais ou simplesmente elos institucionais. Finalmente, no qu into cap(tulo, alinhou as conclusões. A pesquisa objetivou atender aos questionamentos estabelecidos em cinco tópicos que buscavam investigar o seguinte: se houve um planejamento prévio, consciente e deliberado no sentido de transformar a CEPLAC de um órgão transitório, com atividade financeira, para uma instituição técnico-cientl'fica como é hoje. Em caso negativo, o que tornou isso poss(vel? Discorrer sobre a forma pela qual seu quadro dirigente foi estruturado e reuniu recursos, bem como que influências contribuíram para moldar seus programas; constatar como o quadro dirigente da CEPLAC manteve-se estável em um per(odo de mudanças no panorama político brasileiro; e especular sobre o futuro da institu ição. Entre outras coisas, o autor conclu iu que duas variáveis contribu iram particu larmente, de forma decisiva, para a sobrevivência, autonomia e desenvolvimento da instituição, com a obtenção de resultados valorizados pela sua clientela: 1) a existência de uma liderança institucional que soube definir o papel e a missão da CEPLAC, mantendo sua integridade ao longo do tempo; e, 2) a garantia de um fluxo sistemático de recursos financeiros através da contribuição cambial retirada das exportações de cacau. Além disso, constatou que três fatores foram importantes para sua estabilidade: o pleno enquadramento da instituição na filosofia modernizadora contida no bojo dos governos pós-54; a relevância de seus elos institucionais; e o reconhecimento da cl ientela e de setores governamentais importantes pela sua participação na evolução havida na economia cacaueira, de uma situação de crise para resultados satisfatórios.
