The carbohydrate storage is necessary to support the plant growth in periods of stress, during the dormancy, in the beginning of the vegetative development and during the fruiting time. In this context, this work intended to evaluate the carbohydrate concentrations of the peach (Prunus persica (L.) Batsch) tree ‘BRS Rubimel’, cultivated under subtropical conditions. The experiment was performed at the experimental farm Lageado, of the Faculty of Agricultural Sciences of UNESP at Botucatu/SP. The evaluated peach trees were two years old and were cultivated in the spacing of 6.0 x 4.0 m. The adopted experimental delineation was in randomized blocks, making use of four plants per parcel with four repetitions. The treatments corresponded to the period of the collection of leaves and branches, during the annual cycle, corresponding to January to May and July to December 2012. The sample collection of the roots was performed in January, April, August, November and December 2012. Four fruits per plant were collected. The concentration of starch in the roots of the peach tree were superior to the ones from the branches, from August to December. The carbohydrate with the higher storage level in the peach tree ‘BRS Rubimel’ was starch.

Phenolic compounds play a central role in peach fruit colour, flavour and health attributes. Phenolic profiles of several peaches and nectarines and most of the structural genes leading to the anthocyanin synthesis in peach fruit have been studied. Moreover, crosses of red and non-red peaches suggested that a major gene controls skin colour of the extreme phenotypes ‘highlighter’ and ‘full-red’. However, there is no data about either the ‘flavan-3-ols specific genes’ (ANR and LAR) or the regulation of the flavonoid metabolism in this crop. In the present study, we determined the concentration of phenolic compounds in the yellowfleshed nectarine Prunus persica cv. ‘Stark Red Gold’ during fruit growth and ripening. We examined the transcript levels of the main structural genes of the flavonoid pathway. Gene expression of the biosynthetic genes correlated well with the concentration of flavan-3-ols, which was very low at the beginning of fruit development, strongly increased at mid-development and finally decreased again during ripening. In contrast, the only gene transcript which correlated with anthocyanin concentration was PpUFGT, which was high at the beginning and end of fruit growth, remaining low during the other developmental stages. These patterns of gene expression could be explained by the involvement of different transcription factors, which up-regulate anthocyanin biosynthesis (PpMYB10 and PpbHLH3), or repress (PpMYBL2) the transcription of the structural genes. These transcription factors appeared to be involved also in the regulation of the lightinduced anthocyanin accumulation in ‘Stark Red Gold’ nectarines, suggesting that they play a critical role in the regulation of flavonoid biosynthesis in peaches and nectarines in response to both developmental and environmental stimuli. Phenolic profiles and expression patterns of the main flavonoid structural and regulatory genes were also determined for the extreme phenotypes denominated ‘highlighter’ and ‘full-red’ and hypotheses about the control of phenolic compounds content in these fruit are discussed.

The aim of the present thesis was to better understand the physiological role of the phytohormones jasmonates (JAs) and abscisic acid (ABA) during fruit ripening in prospect of a possible field application of JAs and ABA to improve fruit yield and quality. In particular, the effects of exogenous application of these substances at different fruit developmental stages and under different experimental conditions were evaluated. Some aspects of the water relations upon ABA treatment were also analysed. Three fruit species, peach (Prunus persica L. Batsch), golden (Actinidia chinensis) and green kiwifruit (Actinidia deliciosa), and several of their cvs, were used for the trials. Different experimental models were adopted: fruits in planta, detached fruit, detached branches with fruit, girdled branches and micropropagated plants. The work was structured into four sets of experiments as follows: (i) Pre-harvest methyl jasmonate (MJ) application was performed at S3/S4 transition under field conditions in Redhaven peach; ethylene production, ripening index, fruit quality and shelf-life were assessed showing that MJ-treated fruit were firmer and thus less ripe than controls as confirmed by the Index of Absorbance Difference (IAD), but exhibited a shorter shelf-life due to an increase in ethylene production. Moreover, the time course of the expression of ethylene-, auxin- and other ripening-related genes was determined. Ripening-related ACO1 and ACS1 transcript accumulation was inhibited though transiently by MJ, and gene expression of the ethylene receptor ETR2 and of the ethylene-related transcription factor ERF2 was also altered. The time course of the expression of several auxin-related genes was strongly affected by MJ suggesting an increase in auxin biosynthesis, altered auxin conjugation and release as well as perception and transport; the need for a correct ethylene/auxin balance during ripening was confirmed. (ii) Pre- and post-harvest ABA applications were carried out under field conditions in Flaminia and O’Henry peach and Stark Red Gold nectarine fruit; ethylene production, ripening index, fruit quality and shelf-life were assessed. Results show that pre-harvest ABA applications increase fruit size and skin color intensity. Also post-harvest ABA treatments alter ripening-related parameters; in particular, while ethylene production is impaired in ABA-treated fruit soluble solids concentration (SSC) is enhanced. Following field ABA applications stem water potential was modified since ABA-treated peach trees retain more water. (iii) Pre- and post-harvest ABA and PDJ treatments were carried out in both kiwifruit species under field conditions at different fruit developmental stages and in post-harvest. Ripening index, fruit quality, plant transpiration, photosynthesis and stomatal conductance were assessed. Pre-harvest treatments enhance SSC in the two cvs and flesh color development in golden kiwifruit. Post-harvest applications of either ABA or ABA plus PDJ lead to increased SSC. In addition, ABA reduces gas exchanges in A. deliciosa. (iv) Spray, drench and dipping ABA treatments were performed in micropropagated peach plants and in peach and nectarine detached branches; plant water use and transpiration, biomass production and fruit dehydration were determined. In both plants and branches ABA significantly reduces water use and fruit dehydration. No negative effects on biomass production were detected. The present information, mainly arising from plant growth regulator application in a field environment, where plants have to cope with multiple biotic and abiotic stresses, may implement the perspectives for the use of these substances in the control of fruit ripening.

Antimicrobial peptides (AMPs) are an important component of the innate immune system of the plants. Plant defensins are a large family of antimicrobial peptides with several interesting features, such as small dimension, high stability and broad spectrum of action. The discovery of new molecules and the study of their mechanism of action allow to consider them attractive for biotechnological applications. In this PhD thesis a defensin from Prunus persica (PpDFN1) and four novel DEFensin Like (DEFL) peptides from Vitis vinifera have been studied. In order to characterize the antimicrobial activity of these molecules, the recombinant mature peptides have been expressed in Escherichia coli and purified to homogeneity by chromatography techniques. PpDFN1 is able to inhibit the growth of B. cinerea, P. expansum and M. laxa with different intensity. The recombinant peptide is capable of membrane permeabilization as demonstrated by SYTOX green fluorescence uptake in treated mycelia. Its interaction with membranes containing sphingolipid species has been shown by artificial lipid monolayers. Furthermore, PpDFN1 displays stronger interaction with monolayers composed by lipids extracted from sensitive fungi with the highest interaction against P. expansum, the most sensitive fungi to PpDFN1 action. DEFL 13, a defensin from grapevine, resulted the strongest antibotrytis peptides. It is electrostatically attracted to the fungal membranes as shown by the antagonist effect of the cations and is able to membrane permeabilization in B. cinerea hyphae. DEFL 13 is internalized in fungal cells and leads to fungal death by activation of some signaling pathways as demonstrated by screening of a mutant collection of B. cinerea

Grapholita molesta (Lepidoptera: Olethreutidae) es plaga principal del duraznero en Mendoza. El desarrollo de crías artificiales permite interpretar el comportamiento de dicha plaga y consecuentemente, aplicar estrategias para su control. El objetivo del trabajo fue evaluar distintas relaciones de sexos de la especie mediante la fecundidad, viabilidad y longevidad La cría se desarrolló en una cámara con temperatura, humedad relativa y fotoperíodo controlados. El diseño experimental fue en bloques al azar para eliminar la influencia de la luz dentro de la cámara y el desarrollo generacional. Se realizaron cuatro repeticiones. Los resultados se analizaron mediante el programa Statistica. El análisis de los resultados entre bloque y tratamiento no tuvo diferencias significativas en la incidencia de la luz y en el desarrollo generacional. La fecundidad tuvo diferencias significativas entre los tratamientos. Por el contrario, la viabilidad y la longevidad de ambos sexos no tuvieron diferencias significativas, pero los valores de la primera fueron en todos los tratamientos más altos que el máximo citado por otros autores. La relación de sexos influyó positivamente sobre la fecundidad cuando la proporción de los machos fue mayor o igual que el sexo contrario.

Del total de la superficie implantada con duraznos conserveros en la provincia de Mendoza, el 27% corresponde a cultivares de maduración temprana, en los que el raleo de frutos es un factor crítico de manejo. Para determinar la incidencia de diferentes épocas de raleo sobre la rentabilidad del cultivo se realizó un ensayo en un monte comercial de durazneros cvs. Fortuna y Carson. Se utilizaron árboles injertados sobre portainjerto Nemaguard, conducidos en vaso tipo californiano e implantados a 6 m entre hileras y a 5 m entre plantas, que fueron raleados en tres épocas diferentes: a los 40 días después de plena floración, en inicio de endurecimiento de carozo y 10 días después, en la fecha de referencia. Se realizaron evaluaciones de producción por planta y calidad de frutos. También se determinó el valor de la producción y la relación entre éste y las distintas fechas de raleo. En ambos cultivares, los árboles raleados en época temprana obtuvieron mayor peso de frutos, producción total y producción destinada a mitades. En el cv. Fortuna, este tratamiento produjo un aumento significativo del porcentaje de frutos con carozo partido.El raleo temprano de frutos aumentó significativamente el valor de la cosecha, logrando un incremento de $ 3636.ha-1 y $ 3441.ha-1en los cvs. Fortuna y Carson, respectivamente, en relación con el tratamiento en fecha de referencia.

Para evaluar la influencia de la tela antigranizo sobre los rendimientos y la coloración de piel de los frutos de duraznero Prunus persicae L. Batsch variedad Dr. Davis se utilizaron plantas testigo y bajo tela antigranizo seleccionadas por uniformidad de copa y de carga frutal. La cosecha se realizó en dos pasadas. En cada recolección se determinó peso individual y color de piel de los frutos. Se calcularon los rendimientos totales y por pasadas y los rendimientos por categoría de color de piel. La tela antigranizo no afectó los rendimientos totales. En cambio retrasó el desarrollo de color de fondo y, por lo tanto, redujo el rendimiento de la primera pasada de cosecha. Además aumentó la proporción de frutos con madurez mínima en la segunda recolección.

Para determinar el efecto de la carga frutal sobre el peso de frutos y la distribución de calibres se realizó un ensayo en durazneros variedades Bowen, Andross y Ross. Árboles en plena producción, conducidos en vaso tipo californiano e implantados a 6x5 m fueron raleados manualmente 20 días después de iniciado el endurecimiento de carozo. La carga frutal quedó establecida en el rango de 750 a 1 350 frutos/planta en Bowen y Ross y entre 850 y 1400 frutos/planta en Andross. Se realizaron evaluaciones de tamaño final de frutos, producción total y producción destinada a mitades. En las tres variedades se estableció un alto grado de correlación entre la carga frutal y los tres parámetros de evaluación y se observó una respuesta similar a diferentes niveles de carga frutal. A medida que se incrementó la carga frutal, se lograron aumentos en la producción total, aunque disminuyó el peso de frutos. La producción destinada a mitades aumentó en relación con la carga frutal, hasta alcanzar valores máximos con cargas de 1 150 frutos/planta. Luego se estabilizó y, superado un valor crítico en la carga frutal, empezó a disminuir. Para todo el rango de carga utilizado, el peso de frutos y la producción total fueron mayores en Ross. Esta variedad también logró los máximos valores de producción destinada a mitades. Andross superó a Bowen en los tres parámetros evaluados.

Para determinar la relación entre la carga frutal y la productividad, medida en términos de producción destinada a mitades (PDM) y producción para pulpa, se realizó un ensayo en un monte comercial de duraznero variedad Bowen, Andross y Ross. Se utilizaron árboles injertados sobre patrón Nemaguard, conducidos en vaso tipo californiano e implantados a 6 m entre hileras y a 5 m entre plantas (333 árboles/ha), los que fueron raleados 20 días después del inicio de endurecimiento de carozo. La carga frutal quedó establecida en el rango de 750 a 1350 frutos/planta en Bowen y Ross y entre 850 y 1400 frutos/planta en Andross. A la cosecha se evaluó: tamaño de frutos; producción de los árboles, a través de las variables producción total, producción destinada a mitades (PDM) y producción para pulpa; y valor de la producción. La carga frutal se vinculó, con un alto grado de correlación, con todas las variables analizadas y se observó una respuesta a la carga de frutos muy similar en las tres variedades en estudio. A medida que se incrementó la carga frutal, aumentó la producción total, aunque disminuyó el peso medio de frutos. La PDM aumentó en función de la carga frutal, hasta alcanzar valores máximos con 1200 frutos/planta; por encima de un valor crítico en la carga frutal, la PDM empezó a disminuir. Los rendimientos de pulpa, en fábrica, no se relacionaron linealmente con los rendimientos totales del monte frutal y respondieron a una función similar a la que relaciona carga frutal y PDM. Los resultados indicaron que, con independencia del destino de la fruta, el raleo de frutos aseguró los rendimientos y la calidad de frutos que permitieron maximizar la rentabilidad del productor, convirtiéndose en una tarea cultural imprescindible, desde el punto de vista técnico y económico.

El presente ensayo fue establecido en el Alto Valle del Río Negro, Argentina (38°55´ Sur), sobre durazneros cv. Elegant Lady conducidos en vaso, de 5 m de altura, con un distanciamiento de 4 m entre plantas y 4,8 m entre filas. Se realizaron tres tratamientos en un diseño totalmente aleatorizado, simulando diferentes intensidades de luz: restricción lumínica con mallas de sombreo del 80%, poda en verde y un control sin intervención. En cada una de las plantas se diferenciaron 4 sectores orientados hacia los 4 puntos cardinales y 3 alturas distintas de la copa del árbol. La radiación fotosintéticamente activa (RFA) fue medida en cada sector 25, 15 y 6 días antes de la cosecha. La RFA interceptada estuvo influenciada por la restricción lumínica y por la altura. Las variables vegetativas y de producción se relacionaron entre sí linealmente, y ambas dependieron principalmente de la RFA interceptada. Los modelos que explican el comportamiento entre la RFA y las variables de calidad son de tipo asintóticos. A partir de los 25 días anteriores a la cosecha, la RFA necesaria para alcanzar frutos con un peso y color adecuados para su comercialización debe ser del 30%. En el rango de 0 a 15% de RFA interceptada, pequeñas variaciones de RFA dan como resultado grandes cambios en las variables de peso, color de cobertura e intensidad de color del fruto.

Experiments to evaluate the effect of in-season calcium (Ca) sprays on late-season peach (Prunus persica L. Batsch cv. Calrico) were carried out for a 2-year period. Calcium formulations (0.5% and 1.0% in 2008 and only 0.5% tested in 2009) supplied either as CaCl2 or Ca propionate in combination with two or three adjuvants (0.05% of the nonionic surfactants Tween 20 and Break Thru, and 0.5% carboxymethylcellulose, CMC) were sprayed four to five times over the growing season. Peach mesocarp and endocarp Ca concentrations were determined on a 15-day basis from the beginning of May until the end of June. Further tissue analyses were performed at harvest. A decreasing trend in fruit Ca concentrations over the growing season was always observed regardless of the Ca treatments. Both in 2008 and 2009, significant tissue Ca increments associated with the application of Ca-containing sprays in combination with adjuvants were only observed in June, which may be coincident with the period of pit hardening. In 2008, both at harvest and after cold storage, the total soluble-solids concentration (° Brix) of fruits supplied with Ca propionate (0.5% and 1.0% Ca) was always lower as compared to the rest of treatments. The application of multiple Ca-containing sprays increased firmness at harvest and after cold storage, especially when CaCl2 was the active ingredient used. Supplying the adjuvants Tween 20 and CMC increased fruit acidity both at harvest and after cold storage. Evaluation of the development of physiological disorders after cold storage (2 weeks at 0°C) indicated a lower susceptibility of Ca-treated fruits to internal browning. Fruits treated with multiple CaCl2-, CMC-, and Break Thru®-containing sprays during the growing season were significantly less prone to the development of chilling injuries as compared to untreated peaches.
