Los hidrocarburos aromáticos policíclicos (PAHs) son un grupo de compuestos mutagénicos a los que los seres vivos estamos expuestos continuamente. En la primera fase del metabolismo de estos xenobióticos se generan epóxidos, intermediarios reactivos, capaces de generar aductos con el DNA produciendo lesiones en el material genético. Además, también se generan especies reactivas de oxígeno (ROS) que causan estrés oxidativo dañando las células. Este tipo de lesiones puede conducir a la aparición de cáncer y enfermedades neurodegenerativas como el Alzehimer, la enfermedad de Parkinson y la distrofia lateral amiotrófica. Ciertos compuestos denominados antioxidantes tienen capacidad de combatir estos radicales reactivos. Este estudio tiene como objetivo analizar el efecto de seis antioxidantes (Coenzima Q10, butil hidroxianisol, silibin, licopeno, turmérico y 6-gingerol) frente a la toxicidad de uno de estos intermediarios reactivos, el (±)-anti-11, 12-dihidróxido-13,14-epóxido- 11,12,13,14-tetrahidrodibenzo[a, l]pireno (DBPDE) en células XEM2 de mamífero. Se empleó el ensayo de mutación HPRT para determinar la tasa de supervivencia y la frecuencia de mutación de las células después de ser tratadas con el DBPDE y los antioxidantes. La toxicidad del DBPDE se comprobó y se observó dos posibles efectos para los antioxidantes. Por un lado, la CoQ10 y el 6-gingerol mostraron un efecto protector frente al PAH. Sin embargo, los otros antioxidantes no presentaron efecto protector. El BHA, el silibin, el licopeno y el turmérico presentaron una toxicidad similar a la del DBPDE. Esto puede ser debido a que los antioxidantes son específicos en el tipo de radicales que neutralizan y a la dosis empleada. Los antioxidantes solo tienen efecto protector cuando se emplea su dosis óptima. En otras concentraciones pueden ser incluso dañinos.

A depressão é uma doença grave que vem se tornando mais prevalente na população mundial e no Brasil. Segundo a Organização Mundial de Saúde (OMS), é a quarta doença mais incapacitante e estima-se que em 2020 ocupe o segundo lugar, ficando atrás apenas das doenças cardiovasculares (DCV), que são a principal causa de morte no mundo. O Transtorno depressivo maior (TDM) se caracteriza por humor deprimido, tristeza intensa ou desânimo ou perda de interesse ou de prazer por quase todas as atividades por, pelo menos, duas semanas. Além disso, tem um elevado índice de mortalidade cardiovascular, e esta associação parece ser multifatorial e altamente complexa, e ainda não está completamente elucidada. Recentes estudos sugerem que a ocorrência de aterotrombose e eventos cardiovasculares no TDM está associada a uma diminuição na biodisponibilidade do óxido nítrico (NO), um potente vasodilatador, anti-agregante plaquetário e neurotransmissor. O NO é um gás formado a partir da L-arginina, pela ação da família de enzimas NO sintases (NOS), e vai ocasionar um aumento de guanosina monofosfato cíclica (GMPc), que é posteriormente degradada pelas fosfodiesterases (PDE). A L-arginina participa em outras vias além da produção de NO, como a arginase. O estresse oxidativo também tem uma participação no desenvolvimento dos transtornos psiquiátricos e nas DCV, e pode reduzir a meia-vida do NO. O objetivo deste estudo é investigar a via NO-GMPc, o ciclo da uréia, marcadores de estresse oxidativo e de inflamação em plaquetas e a sua associação com a função plaquetária no TDM. Participaram da pesquisa nove pacientes com diagnóstico de depressão leve a moderada do Serviço de Psicologia Aplicada (SPA/UERJ) e onze indivíduos saudáveis pareados por idade como controles. Este projeto foi aprovado pelo Comitê de Ética e Pesquisa do Hospital Universitário Pedro Ernesto (1436-CEP/HUPE). A agregação plaquetária, a expressão e atividade da arginase II, a expressão da PDE 5, marcadores de estresse oxidativo (níveis de TBARS, carbonilação de proteínas, expressão da NADPH oxidase e da glutationa peroxidase (GPx) e atividade desta e da catalase (CAT), ambas enzimas anti-oxidantes) nas plaquetas e no soro, e o fibrinogênio sistêmico foram investigados. No presente estudo observou-se um aumento da agregação plaquetária induzida por ADP em pacientes com TDM comparados aos controles. Uma ativação da arginase II em plaquetas sem qualquer alteração na sua expressão foi demonstrada em pacientes com TDM. Além disso, um aumento na carbonilação de proteínas e na expressão de GPx, de NADPH e de PDE5 foi observado em plaquetas de pacientes com TDM. A produção de TBARS, a atividade de GPx e CAT nas plaquetas e no soro não foram afetados pelo TDM. Não houve diferença nos níveis de fibrinogênio entre pacientes com TDM e controles. A ativação da arginase, somada ao estresse oxidativo, reduziria a biodisponibilidade de NO levando à disfunção plaquetária nos pacientes com TDM. O presente estudo acrescenta dados importantes para a compreensão dos mecanismos celulares envolvidos na relação TDM e DCV. Além disso, abre caminho para a utilização de novas ferramentas farmacológicas, como os antioxidantes, para o tratamento do TDM.

Stejnulxin, a novel snake C-type lectin-like protein with potent platelet activating activity, was purified and characterized from Trimeresurus stejnegeri venom. Under non-reducing conditions, it migrated on a SDS-polyacrylamide gel with an apparent molecular mass of 120 kDa. On reduction, it separated into three polypeptide subunits with apparent molecular masses of 16 kDa (alpha), 20 kDa (beta(1)) and 22 kDa (beta(2)), respectively. The complete amino acid sequences of its subunits were deduced from cloned cDNAs. The N-terminal sequencing and cDNA cloning indicated that beta(1) and beta(2) subunits of stejnulxin have identical amino acid sequences and each contains two N-glycosylation sites. Accordingly, the molecular mass difference between 1 and 2 is caused by glycosylation heterogenity. The subunit amino acid sequences of stejnulxin are similar to those of convulxin, with sequence identities of 52.6% and 66.4% for the U. and beta, respectively. Stejnulxin induced human platelet aggregation in a dose-dependent manner. Antibodies against UNA inhibited the aggregation response to stejnulxin, indicating that activation of alpha(IIb)beta(3) and binding of fibrinogen are involved in stejnulxin-induced platelet aggregation. Antibodies against GPIbalpha or alpha(2)beta(1) as well as echicetin or rhodocetin had no significant effect on stejnulxin-induced platelet aggregation. However, platelet activation induced by stejnulxin was blocked by anti-GPVI antibodies. In addition, stejnulxin induced a tyrosine phosphorylation profile in platelets that resembled that produced by convulxin. Biotinylated stejnulxin bound specifically to platelet membrane GPVI.

A novel potent trypsin inhibitor was purified and characterized from frog Bombina maxima skin. A full-length cDNA encoding the protein was obtained from a cDNA library constructed from the skin. Sequence analysis established that the protein actually comprises three conserved albumin domains. B. maxima serum albumin was subsequently purified, and its coding cDNA was further obtained by PCR-based cloning from the frog liver. Only two amino acid variations were found in the albumin sequences from the skin and the serum. However, the skin protein is distinct from the serum protein by binding of a haem b (0.95 mol/mol protein). Different from bovine serum albumin, B. maxima albumin potently inhibited trypsin. It bound tightly with trypsin in a 1: 1 molar ratio. The equilibrium dissociation constants (K-D) obtained for the skin and the serum proteins were 1.92 x 10(-9) M and 1.55 x 10(-9) M, respectively. B. maxima albumin formed a noncovalent complex with trypsin through an exposed loop formed by a disulfide bond (Cys(53)-Cys(62)), which comprises the scissile bond Arg(58)(P-1)-His(59)(P-1'). No inhibitory effects on thrombin, chymotrypsin, elastase, and subtilisin were observed under the assay conditions. Immunohistochemical study showed that B. maxima albumin is widely distributed around the membranes of epithelial layer cells and within the stratum spongiosum of dermis in the skin, suggesting that it plays important roles in skin physiological functions, such as water economy, metabolite exchange, and osmoregulation.

UV radiation is one of many harmful factors found in space that are detrimental to organisms on earth in space exploration. In the present work, we examined the role of antioxidant system in Nostoc sphaeroides Kutz (Cyanobacterium) and the effects of exogenously applied antioxidant molecules on its photosynthetic rate under UV-B radiation. It was found that UV-B radiation promoted the activity of antioxidant system to protect photosystem 11 (PSII) and exogenously applied antioxidant: sodium nitroprusside (SNP) and N-acetylcysteine (NAC) had an obvious protection on PSII activity under UV-B radiation. The activity of superoxide dismutase (SOD, EC 1.15.1.1), catalase (CAT, EC 1.11.1.6), peroxidase (POD, EC 1.11.1.7) and content of NIDA (malondialdehyde) and ASC (ascorbate) were improved by 0.5 mM and 1 mM SNP, but 0.1 mM SNP decreased the activity of antioxidant system. Addition of exogenous NAC decreased the activity of SOD, POD, CAT and the content MDA and ASC. In contrast, exogenously applied NAC increased GSH content. The results suggest that exogenous SNP and NAC may protect algae by different mechanisms: SNP may play double roles as both sources of reactive free radicals as well as ROS scavengers in mediating the protective role of PSII on algae under UV-B radiation. On the other hand, NAC functions as an antioxidant or precursor of glutathione, which could protect PSII directly from UV-B radiation. (c) 2007 COSPAR, Published by Elsevier Ltd. All rights reserved.

In this study, we found that UV-B radiation decreased photosynthetic activity and boosted lipid peroxidation of desert Nostoc sp., and exogenous chemicals (ascorbate acid (ASC), N-acetylcysteine (NAC), and sodium nitroprusside (SNP)) had obvious protective effects on photosynthesis and membranes under UV-B radiation. High-concentration SNP boosted the activities of antioxidant enzymes, but low-concentration SNP reduced the activities of antioxidant enzymes. Both NAC and ASC treatments of cells decreased activities of antioxidant enzymes. The results suggested that those chemicals possibly had different mechanisms of protection of algae cells against UV-B radiation. SNP might play double roles as a signal molecule in the formation of algae cell protection of Photosystem 11 under UV-B radiation and as a (reactive oxygen species) scavenger, while NAC and ASC might function as antioxidant reagents or precursors of other antioxidant molecules, which could protect cells directly against ROS initiated by UV-B radiation. HR212, a recombinant protein expressed in Escherichia coli, has been previously reported to inhibit HIV-1 membrane fusion at low nanomolar level. Here we report that HR212 is effective in blocking laboratory strain HIV-1IIIB entry and replication with EC50 values of 3.92±0.62 and 6.59±1.74 nM, respectively, and inhibiting infection by clinic isolate HIV-1KM018 with EC50 values of 44.44±10.20 nM, as well as suppressing HIV-1- induced cytopathic effect with an EC50 value of 3.04±1.20 nM. It also inhibited HIV-2ROD and HIV-2CBL-20 entry and replication in the μM range. Notably, HR212 was highly effective against T20-resistant strains with EC50 values ranging from 5.09 to 7.75 nM. Unlike T20, HR212 showed stability sufficient to inhibit syncytia formation in a time-of-addition assay, and was insensitive to proteinase K digestion. These results suggest that HR212 has great potential to be further developed as novel HIV-1 fusion inhibitor for treatment of HIV/ AIDS patients, particularly for those infected by T20-resistant variants.

A series of 3,4-dimethyl-4-(3-hydroxyphenyl) piperidine opioid antagonists with varying substituents on the nitrogen were evaluated for their effect on food consumption in obese Zucker rats. In developing three-dimensional quantitative structure-activity relationship (3D-QSAR) studies for this series of opioid antagonists, different structure alignments have been tested to predict the anorectant activities. The interaction energies between molecules and the probe atom were then correlated with anorectant activity using partial least squares (PLS) method. The steric and electrostatic features of the 3D-QSAR were presented in the form of standard deviation coefficient contour maps of steric and electrostatic fields. The results showed that 3D-QSAR results are much better than the results obtained by 2D-QSAR.
