Cells of the bacterial symbiont Xenorhabdus nematophila from the entomopathogenic nematode, Steinernema carpocapsae entered the pupae of Plutella xylostella after 15 minutes treatment with suspensions containing the bacterial cells. Secretions of Xenorhabdus nematophila, in either broth or water, were found lethal to the pupae of P. xylostella when applied in moist sand. The bacterial symbiont Xenorhabdus nematophila was found lethal to the pupae of greater wax moth (Galleria mellonella), beet armyworm (Spodoptera exigua), diamondback moth (Plutella xylostella) and black vine weevil (Otiorhynchus sulcatus) in the absence of the nematode vector and the cells of X. nematophila entered the haemocoele of the pupae.

Plants produce volatile organic compounds (VOCs) in response to herbivore attack, and these VOCs can be used by parasitoids of the herbivore as host location cues. We investigated the behavioural responses of the parasitoid Cotesia vestalis to VOCs from a plant–herbivore complex consisting of cabbage plants (Brassica oleracea) and the parasitoids host caterpillar, Plutella xylostella. A Y-tube olfactometer was used to compare the parasitoids' responses to VOCs produced as a result of different levels of attack by the caterpillar and equivalent levels of mechanical damage. Headspace VOC production by these plant treatments was examined using gas chromatography–mass spectrometry. Cotesia vestalis were able to exploit quantitative and qualitative differences in volatile emissions, from the plant–herbivore complex, produced as a result of different numbers of herbivores feeding. Cotesia vestalis showed a preference for plants with more herbivores and herbivore damage, but did not distinguish between different levels of mechanical damage. Volatile profiles of plants with different levels of herbivores/herbivore damage could also be separated by canonical discriminant analyses. Analyses revealed a number of compounds whose emission increased significantly with herbivore load, and these VOCs may be particularly good indicators of herbivore number, as the parasitoid processes cues from its external environment

Interest in sustainable farming methods that rely on alternatives to conventional synthetic fertilizers and pesticides is increasing. Sustainable farming methods often utilize natural populations of predatory and parasitic species to control populations of herbivores, which may be potential pest species. We investigated the effects of several types of fertilizer, including those typical of sustainable and conventional farming systems, on the interaction between a herbivore and parasitoid. The effects of fertilizer type on percentage parasitism, parasitoid performance, parasitoid attack behaviour and responses to plant volatiles were examined using a model Brassica system, consisting of Brassica oleracea var capitata, Plutella xylostella (Lepidoptera) larvae and Cotesia vestalis (parasitoid). Percentage parasitism was greatest for P. xylostella larvae feeding on plants that had received either a synthetic ammonium nitrate fertilizer or were unfertilized, in comparison to those receiving a composite fertilizer containing hoof and horn. Parasitism was intermediate on plants fertilized with an organically produced animal manure. Male parasitoid tibia length showed the same pattern as percentage parasitism, an indication that offspring performance was maximized on the treatments preferred by female parasitoids for oviposition. Percentage parasitism and parasitoid size were not correlated with foliar nitrogen concentration. The parasitoids did not discriminate between hosts feeding on plants in the four fertilizer treatments in parasitoid behaviour assays, but showed a preference for unfertilized plants in olfactometer experiments. The percentage parasitism and tibia length results provide support for the preference–performance hypothesis

The characterization and identification of proteolytic bacteria from the gut of the velvetbean caterpillar (Anticarsia gemmatalis) were the objectives of this study. Twelve aerobic and anaerobic isolates of proteolytic bacteria were obtained from the caterpillar gut in calcium caseinate agar. The number of colony forming units (CFUs) of proteolytic bacteria was higher when the bacteria were extracted from caterpillars reared on artificial diet rather than on soybean leaves (1.73 +/- 0.35 X 10(3) and 0.55 +/- 0.22 X 10(3) CFU/mg gut, respectively). The isolated bacteria were divided into five distinct groups, according to their polymerase chain reaction restriction fragment-length polymorphism profiles. After molecular analysis, biochemical tests and fatty acid profile determination, the bacteria were identified as Bacillus subtilis, Bacillus cereus, Enterococcus gallinarum, Enterococcus mundtii, and Staphylococcus xylosus. Bacterial proteolytic activity was assessed through in vitro colorimetric assays for (general) proteases, serine proteases, and cysteine proteases. The isolated bacteria were able of hydrolyzing all tested substrates, except Staphylococcus xylosus, which did not exhibit serine protease activity. This study provides support for the hypothesis that gut proteases from velvetbean caterpillar are not exclusively secreted by the insect cells but also by their symbiotic gut bacteria. The proteolytic activity from gut symbionts of the velvetbean caterpillar is suggestive of their potential role minimizing the potentially harmful consequences of protease inhibitors from some of this insect host plants, such as soybean, with implications for the management of this insect pest species.

O objetivo deste trabalho foi avaliar a eficiência, a estabilidade, a forma de aplicação e a compatibilidade do óleo de mamona com o fungo entomopatogênico Beauveria bassiana no controle da traça-das-crucíferas, Plutella xylostella. No primeiro experimento, foram avaliadas a eficiência, a estabilidade após 70 dias de armazenamento e a forma de aplicação do óleo de mamona. No segundo experimento, foi avaliada a compatibilidade do óleo de mamona com B. bassiana. A atividade inseticida do óleo de mamona é instável ao longo do tempo. O óleo de mamona é compatível com B. bassiana no controle de P. xylostella.

O objetivo desta pesquisa foi comparar diferentes cultivares de brassicáceas em relação à capacidade reprodutiva e preferência para alimentação e oviposição da traça-das-crucíferas. Os experimentos foram realizados utilizando-se as cultivares de repolho Midori, Chato-de-Quintal, híbridos da Top Seed® - Agristar (TPC 308, TPC 681 e TPC 668), couve-flor Bola de Neve, couve brócolis Ramoso Piracicaba Precoce e couve Manteiga da Geórgia, sendo esta última utilizada como padrão de suscetibilidade. Por meio dos dados biológicos de P. xylostella foram estimados os parâmetros necessários para a construção de tabela de vida de fertilidade, para comparação das cultivares testadas em relação à capacidade reprodutiva da praga. em gaiolas de criação da traça-das-crucíferas foram colocados quatro quartos de folha, dois a dois, que justapostos formavam um círculo de 8 cm de diâmetro, confrontando-se os materiais dois a dois. As partes foram dispostas equidistantemente, para realização do teste de dupla chance de escolha (preferência para alimentação e oviposição) e de múltipla chance de escolha, confrontando todos os substratos (preferência para alimentação). As cultivares que proporcionaram melhor desenvolvimento e reprodução para a traça-das-crucíferas foram couve Manteiga da Geórgia e couve brócolis. Para preferência alimentar constatou-se alta suscetibilidade em couve Manteiga e TPC 681 e para preferência de oviposição alta preferência para couve-flor Bola de Neve. Com isso, sugere-se a divisão das cultivares estudadas em quatro classes distintas: repolho Midori como moderadamente resistente (MR); couve-flor Bola de Neve e repolho Chato-de-Quintal como suscetíveis (S); couve brócolis, TPC308, TPC681 e TPC668 como moderadamente suscetíveis (MS); e couve Manteiga como altamente suscetível (AS).

A traça-das-crucíferas, Plutella xylostella (Lepidoptera: Plutellidae), é considerada a principal praga das brássicas no mundo, sendo o uso de inseticidas o método mais usado para o seu controle. Assim, o objetivo desta pesquisa foi selecionar isolados de Beauveria bassiana com viabilidade para utilização no controle da traça-das-crucíferas. Dezessete isolados e um produto comercial de B. bassiana foram testados. Lagartas de segundo ínstar da traça-das-crucíferas foram pulverizadas com suspensão de conídios na concentração de 10(7) conídios mL-1. Para o bioensaio de concentração letal (CL) sete concentrações espaçadas em escala logarítmica foram testadas. Os isolados CCA/UFES-4, 18, 31 e 35 foram selecionados para o bioensaio de CL por causarem mortalidade confirmada superior a 90%. O isolado padrão ESALQ-447 e o produto comercial tiveram resultados semelhantes e também foram selecionados para o bioensaio de CL. Com base nas estimativas da CL50, os isolados CCA/UFES-4, 18, 31, ESALQ-447 e o produto comercial podem ser selecionados para utilização no controle da traça-das-crucíferas.

The Diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae), is one of the main plague-insect specie of Brassicaceae plants in Brazil and all over the world. The resistant genotypes use to its control is a promising alternative. This work aimed evaluates the eggs distribution along the plant, the adults' density per plant, and determine the cauliflower genotypes effect in the P. xylostella oviposition. The experiment was carried out at FCAV/UNESP-Jaboticabal Campus Phytossanity Department (Departamento de Fitossanidade). It was evaluated the eggs distribution, the P. xylostella adults density effect using Sharon hybrid, and tests with or without choose choice to determine the P. xylostella nonpreference in the Teresopolis Gigante, Verona, Barcelona, Sharon, Silver Streak, and Piracicaba Precoce genotypes. It is possible conclude that P. xylostella has higher willingness to oviposits in the stem than in the basal leaves. The three couple density of P. xylostella per plant is the best to discriminate cauliflower genotypes regarding the resistance grade to nonpreference choose choice to oviposition. During the P. xylostella oviposition preference tests with choose choice, the genotypes Sharon, Piracicaba Precoce, Barcelona, Verona e Teresopolis Gigante are less desirable to oviposition; while during the no choose choice tests the genotypes did not differ among them.

The aim of this study was to evaluate the selectivity of pesticides to T. pretiosum and T. exiguum in different hosts. The tests were carried out using the LBCI (Laboratorio de Biologia e Criacao de Insetos) methodology based on the IOBC/WPRS methodology for selectivity studies. The hosts used were Anagasta kuehniella, Spodoptera frugiperda and Plutella xylostella eggs. The analysis of the selectivity was carried out for the products triflumuron (20 mL/100 L), etofenproxi (47 mL/100 L) and endosulfan (750 mL/100 L), with distilled water used as a control. Parasitized eggs, percentage of parasitism, percentage of emergence, longevity and sex ratio were evaluated in generations F1 and F2. Endosulfan was the most harmful, inhibiting the parasitism in all hosts. Etofenproxi showed low selectivity to parasitoids. Triflumuron was selective to parasitoid species when eggs of natural hosts were used.

BACKGROUND: The use of Trichogramma species is a potential key strategy in integrated pest management. However, its effectiveness depends on the use of chemicals that do not interfere with parasitism and parasite population growth. Here, a study was made of the effects of synthetic insecticides on Trichogramma pretiosum and Trichogramma exiguum in different hosts (Ephestia kuehniella, Plutella xylostella and Spodoptera frugiperda) and the influence of International Organisation for Biological Control (IOBC/WPRS) methodology in selectivity studies using different Trichogramma species. The insecticides used were commercial formulations (triflumuron at a concentration of 0.2 mL L-1 water, etofenprox at a concentration of 0.47 mL L-1 water and endosulfan at a concentration of 7.5 mL L-1 water); the control treatment consisted of distilled water. Eggs attached to cardboard cards were offered to parasitoids inside glass cages. Parasitised eggs, parasitism and adult emergence rates and parasitism reduction were evaluated.RESULTS: Endosulfan and etofenprox, classified as class-4 toxic products, were extremely toxic to the parasitoids. Triflumuron, classified as a non-toxic product, was selective to the parasitoids in eggs of all hosts.CONCLUSIONS: The methodology recommended by IOBC/WPRS influenced results regarding the use of different species of parasitoids, and the use of a single parasitoid species in their experiment is questionable. The feeding nonpreference of Plutella xylostella (L.) larvae was tested in regard to the cauliflower genotypes Barcelona, Verona, Piracicaba Precoce, Sharon, Silver Streak (hybrids) and Teresopolis Gigante (cultivar). Choice and no-choice tests were carried out at the laboratories of the Departamento de Fitossanidade, FCAV/UNESP, Jaboticabal, state of São Paulo, Brazil, to study attractiveness and dry mass consumption by 1st-instar (just hatched) and 4th-instar larvae for 24 hours. No significant difference was found for attractiveness in choice and no-choice tests, for both 1st-and 4th-instar larvae. The consumption of dry mass in both tests, with 1st-and 4th-instar larvae, showed a significant difference between genotypes only for 1st-instar larvae, the hybrid Silver Streak being the most consumed (3.72 mg) by P. xylostella larvae. The cauliflower genotypes tested presented no variation in attractiveness for 1st-instar (just hatched) and 4th-instar larvae and did not present nonpreference feeding resistance for P. xylostella larvae.
