Laminin has been shown to promote the malignant phenotype and the level of the 32/67 Kd laminin receptor has been found to correlate with Dukes' staging of colon cancer. A biopsy of a Dukes' stage B2 human colon carcinoma formed a tumor in a nude mouse after coinjection with Matrigel. The parental tumor and the murine tumor appeared identical at the histological level. A cell line LCC-C1 was established from the murine tumor. The cell line appeared moderately differentiated although it did not produce mucin in vitro; however, the xenograft in vivo did produce low levels of mucin. Laminin adherent and non-adherent cell lines were selected. The parental and the laminin-selected cell subclones adhered equally well to plastic and to fibronectin and showed similar growth rates on plastic. When injected subcutaneously into nude mice, the laminin-adherent cells formed relatively undifferentiated tumors that were twice as large as the parental cell tumors whereas the laminin non adherent cells formed very small, but highly differentiated tumors. These data demonstrate that subpopulations of tumor cells which differ in their tumorigenic properties can be selected based on their adhesion to laminin and thus provide models for studying the mechanisms of tumor growth.

Background Adherence to evidence based medicines in patients who have experienced a myocardial infarction remains low. Individual’s beliefs towards their medicines are a strong predictor of adherence and may influence other factors that impact on adherence. Objective To investigate if community pharmacists discussing patients’ beliefs about their medicines improved medication adherence at 12 months post myocardial infarction. Setting This study included 200 patients discharged from a public teaching hospital in Queensland, Australia, following a myocardial infarction. Patients were randomised into intervention (n = 100) and control groups (n = 100) and followed for 12 months. Method All patients were interviewed between 5 to 6 weeks, at 6 and 12 months post discharge by the researcher using the repertory grid technique. This technique was used to elicit the patient’s individualised beliefs about their medicines for their myocardial infarction. In the intervention group, patients’ beliefs about their medicines were communicated by the researcher to their community pharmacist. The pharmacist used this information to tailor their discussion with the patient about their medication beliefs at designated time points (3 and 6 months post discharge). The control group was provided with usual care. Main outcome measure The difference in non-adherence measured using a medication possession ratio between the intervention and control groups at 12 months post myocardial infarction. Results There were 137 patients remaining in the study (intervention group n = 72, control group n = 65) at 12 months. In the intervention group 29 % (n = 20) of patients were non-adherent compared to 25 % (n = 16) of patients in control group. Conclusion Discussing patients’ beliefs about their medicines for their myocardial infarction did not improve medication adherence. Further research on patients beliefs should focus on targeting non-adherent patients whose reasons for their non-adherence is driven by their medication beliefs.

An early molecular response to DNA double-strand breaks (DSBs) is phosphorylation of the Ser-139 residue within the terminal SQEY motif of the histone H2AX1,2. This phosphorylation of H2AX is mediated by the phosphatidyl-inosito 3-kinase (PI3K) family of proteins, ataxia telangiectasia mutated (ATM), DNA-protein kinase catalytic subunit and ATM and RAD3-related (ATR)3. The phosphorylated form of H2AX, referred to as γH2AX, spreads to adjacent regions of chromatin from the site of the DSB, forming discrete foci, which are easily visualized by immunofluorecence microscopy3. Analysis and quantitation of γH2AX foci has been widely used to evaluate DSB formation and repair, particularly in response to ionizing radiation and for evaluating the efficacy of various radiation modifying compounds and cytotoxic compounds Given the exquisite specificity and sensitivity of this de novo marker of DSBs, it has provided new insights into the processes of DNA damage and repair in the context of chromatin. For example, in radiation biology the central paradigm is that the nuclear DNA is the critical target with respect to radiation sensitivity. Indeed, the general consensus in the field has largely been to view chromatin as a homogeneous template for DNA damage and repair. However, with the use of γH2AX as molecular marker of DSBs, a disparity in γ-irradiation-induced γH2AX foci formation in euchromatin and heterochromatin has been observed5-7. Recently, we used a panel of antibodies to either mono-, di- or tri- methylated histone H3 at lysine 9 (H3K9me1, H3K9me2, H3K9me3) which are epigenetic imprints of constitutive heterochromatin and transcriptional silencing and lysine 4 (H3K4me1, H3K4me2, H3K4me3), which are tightly correlated actively transcribing euchromatic regions, to investigate the spatial distribution of γH2AX following ionizing radiation8. In accordance with the prevailing ideas regarding chromatin biology, our findings indicated a close correlation between γH2AX formation and active transcription9. Here we demonstrate our immunofluorescence method for detection and quantitation of γH2AX foci in non-adherent cells, with a particular focus on co-localization with other epigenetic markers, image analysis and 3Dmodeling.

Objective To determine rates of adherence to antipsychotic medication in first episode patients and the correlates of adherence in this group. Method Subjects were the first 200 admissions to an Early Psychosis Program. Adherence was determined on a three-point scale. Symptoms, medication side-effects, quality of life, substance use and family involvement were examined longitudinally. Results In their first year in the program 39% were non-adherent, 20% inadequately adherent, and 41% adherent. Non-adherent patients demonstrated more positive symptoms, more relapses, more alcohol and cannabis use, reduced insight, and poorer quality of life. They were younger, had an earlier age of onset and less likely to have a family member involved in treatment. Conclusion Results for this group are similar to those reported in the literature. Correlates are often the consequence of non-adherence. Non-compliance has to be anticipated and relationships maintained with patients and families to intervene as soon as possible to minimize the consequence of non-compliance.

Clinical utility of biodegradable magnesium implants is undermined by the untimely degradation of these materials in vivo. Their high corrosion rate leads to loss of mechanical integrity, peri–implant alkalization and localised accumulation of hydrogen gas. Biodegradable coatings were produced on pure magnesium using RF plasma polymerisation. A monoterpene alcohol with known anti-inflammatory and antibacterial properties was used as a polymer precursor. The addition of the polymeric layer was found to reduce the degradation rate of magnesium in simulated body fluid. The in vitro studies indicated good cytocompatibility of non-adherent THP–1 cells and mouse macrophage cells with the polymer, and the polymer coated sample. The viability of THP–1 cells was significantly improved when in contact with polymer encapsulated magnesium compared to unmodified samples. Collectively, these results suggest plasma enhanced polymer encapsulation of magnesium as a suitable method to control degradation kinetics of this biomaterial.

After more than twenty years of basic and applied research, the use of nanotechnology in the design and manufacture of nanoscale materials is rapidly increasing, particularly in commercial applications that span from electronics across renewable energy areas, and biomedical devices. Novel polymers are attracting significant attention for they promise to provide a low−cost high−performance alternative to existing materials. Furthermore, these polymers have the potential to overcome limitations imposed by currently available materials thus enabling the development of new technologies and applications that are currently beyond our reach. This work focuses on the development of a range of new low−cost environmentally−friendly polymer materials for applications in areas of organic (flexible) electronics, optics, and biomaterials. The choice of the monomer reflects the environmentally−conscious focus of this project. Terpinen−4−ol is a major constituent of Australian grown Melaleuca alternifolia (tea tree) oil, attributed with the oil's antimicrobial and anti−inflammatory properties. Plasma polymerisation was chosen as a deposition technique for it requires minimal use of harmful chemicals and produces no hazardous by−products. Polymer thin films were fabricated under varied process conditions to attain materials with distinct physico−chemical, optoelectrical, biological and degradation characteristics. The resultant materials, named polyterpenol, were extensively characterised using a number of well−accepted and novel techniques, and their fundamental properties were defined. Polyterpenol films were demonstrated to be hydrocarbon rich, with variable content of oxygen moieties, primarily in the form of hydroxyl and carboxyl functionalities. The level of preservation of original monomer functionality was shown to be strongly dependent on the deposition energy, with higher applied power increasing the molecular fragmentation and substrate temperature. Polyterpenol water contact angle contact angle increased from 62.7° for the 10 W samples to 76.3° for the films deposited at 100 W. Polymers were determined to resist solubilisation by water, due to the extensive intermolecular and intramolecular hydrogen bonds present, and other solvents commonly employed in electronics and biomedical processing. Independent of deposition power, the surface topography of the polymers was shown to be smooth (Rq <0.5 nm), uniform and defect free. Hardness of polyterpenol coatings increased from 0.33 GPa for 10 W to 0.51 GPa for 100 W (at 500 μN load). Coatings deposited at higher input RF powers showed less mechanical deformation during nanoscratch testing, with no considerable damage, cracking or delamination observed. Independent of the substrate, the quality of film adhesion improved with RF power, suggesting these coatings are likely to be more stable and less susceptible to wear. Independent of fabrication conditions, polyterpenol thin films were optically transparent, with refractive index approximating that of glass. Refractive index increased slightly with deposition power, from 1.54 (10 W) to 1.56 (100 W) at 500 nm. The optical band gap values declined with increasing power, from 2.95 eV to 2.64 eV, placing the material within the range for semiconductors. Introduction of iodine impurity reduced the band gap of polyterpenol, from 2.8 eV to 1.64 eV, by extending the density of states more into the visible region of the electromagnetic spectrum. Doping decreased the transparency and increased the refractive index from 1.54 to 1.70 (at 500 nm). At optical frequencies, the real part of permittivity (k) was determined to be between 2.34 and 2.65, indicating a potential low-k material. These permittivity values were confirmed at microwave frequencies, where permittivity increased with input RF energy – from 2.32 to 2.53 (at 10 GHz ) and from 2.65 to 2.83 (at 20 GHz). At low frequencies, the dielectric constant was determined from current−voltage characteristics of Al−polyterpenol−Al devices. At frequencies below 100 kHz, the dielectric constant varied with RF power, from 3.86 to 4.42 at 1 kHz. For all samples, the resistivity was in order of 10⁸−10⁹ _m (at 6 V), confirming the insulating nature of polyterpenol material. In situ iodine doping was demonstrated to increase the conductivity of polyterpenol, from 5.05 × 10⁻⁸ S/cm to 1.20 × 10⁻⁶ S/cm (at 20 V). Exposed to ambient conditions over extended period of time, polyterpenol thin films were demonstrated to be optically, physically and chemically stable. The bulk of ageing occurred within first 150 h after deposition and was attributed to oxidation and volumetric relaxation. Thermal ageing studies indicated thermal stability increased for the films manufactured at higher RF powers, with degradation onset temperature associated with weight loss shifting from 150 ºC to 205 ºC for 10 W and 100 W polyterpenol, respectively. Annealing the films to 405 °C resulted in full dissociation of the polymer, with minimal residue. Given the outcomes of the fundamental characterisation, a number of potential applications for polyterpenol have been identified. Flexibility, tunable permittivity and loss tangent properties of polyterpenol suggest the material can be used as an insulating layer in plastic electronics. Implementation of polyterpenol as a surface modification of the gate insulator in pentacene-based Field Effect Transistor resulted in significant improvements, shifting the threshold voltage from + 20 V to –3 V, enhancing the effective mobility from 0.012 to 0.021 cm²/Vs, and improving the switching property of the device from 10⁷ to 10⁴. Polyterpenol was demonstrated to have a hole transport electron blocking property, with potential applications in many organic devices, such as organic light emitting diodes. Encapsulation of biomedical devices is also proposed, given that under favourable conditions, the original chemical and biological functionality of terpinen−4−ol molecule can be preserved. Films deposited at low RF power were shown to successfully prevent adhesion and retention of several important human pathogens, including P. aeruginosa, S. aureus, and S. epidermidis, whereas films deposited at higher RF power promoted bacterial cell adhesion and biofilm formation. Preliminary investigations into in vitro biocompatibility of polyterpenol demonstrated the coating to be non−toxic for several types of eukaryotic cells, including Balb/c mice macrophage and human monocyte type (HTP−1 non-adherent) cells. Applied to magnesium substrates, polyterpenol encapsulating layer significantly slowed down in vitro biodegradation of the metal, thus increasing the viability and growth of HTP−1 cells. Recently, applied to varied nanostructured titanium surfaces, polyterpenol thin films successfully reduced attachment, growth, and viability of P. aeruginosa and S. aureus.

We present a case study of formal verification of full-wave rectifier for analog and mixed signal designs. We have used the Checkmate tool from CMU [1], which is a public domain formal verification tool for hybrid systems. Due to the restriction imposed by Checkmate it necessitates to make the changes in the Checkmate implementation to implement the complex and non-linear system. Full-wave rectifier has been implemented by using the Checkmate custom blocks and the Simulink blocks from MATLAB from Math works. After establishing the required changes in the Checkmate implementation we are able to efficiently verify, the safety properties of the full-wave rectifier.

Sequence design and resource allocation for a symbol-asynchronous chip-synchronous code division multiple access (CDMA) system is considered in this paper. A simple lower bound on the minimum sum-power required for a non-oversized system, based on the best achievable for a non-spread system, and an analogous upper bound on the sum rate are first summarised. Subsequently, an algorithm of Sundaresan and Padakandla is shown to achieve the lower bound on minimum sum power (upper bound on sum rate, respectively). Analogous to the synchronous case, by splitting oversized users in a system with processing gain N, a system with no oversized users is easily obtained, and the lower bound on sum power (upper bound on sum rate, respectively) is shown to be achieved by using N orthogonal sequences. The total number of splits is at most N - 1.

In this paper, the study of a third-order mechanical oscillator is presented by demonstrating its equivalence to the well-known R.C. multivibrator with two additional reactive elements. The conditions for the oscillator's possession of periodic solutions are presented. It is also shown that under certain conditions, the study of the given third-order autonomous system can be reduced to the study of an equivalent second-order, non-autonomous system.

A common and practical paradigm in cooperative communications is the use of a dynamically selected 'best' relay to decode and forward information from a source to a destination. Such a system consists of two core phases: a relay selection phase, in which the system expends resources to select the best relay, and a data transmission phase, in which it uses the selected relay to forward data to the destination. In this paper, we study and optimize the trade-off between the selection and data transmission phase durations. We derive closed-form expressions for the overall throughput of a non-adaptive system that includes the selection phase overhead, and then optimize the selection and data transmission phase durations. Corresponding results are also derived for an adaptive system in which the relays can vary their transmission rates. Our results show that the optimal selection phase overhead can be significant even for fast selection algorithms. Furthermore, the optimal selection phase duration depends on the number of relays and whether adaptation is used.

Such systems use two phases - a relay selection phase, in which the system uses transmission time and energy to select the best relay, and a data transmission phase, in which it uses the spatial diversity benefits of selection to transmit data. In this paper, we derive closed-form expressions for the overall throughput and energy consumption, and study the time and energy trade-off between the selection and data transmission phases. To this end, we analyze a baseline non-adaptive system and several adaptive systems that adapt the selection phase, relay transmission power, or transmission time. Our results show that while selection yields significant benefits, the selection phase's time and energy overhead can be significant. In fact, at the optimal point, the selection can be far from perfect, and depends on the number of relays and the mode of adaptation. The results also provide guidelines about the optimal system operating point for different modes of adaptation. The analysis also sheds new insights on the fast splitting-based algorithm considered in this paper for relay selection.

A limit stop is placed at the elbow of an L-shaped beam whose linear natural frequencies are nearly commensurable. As a result of this hardening device the non-linear system exhibits multiple internal resonances, which involve various degree of coupling between the first five modes of the beam in free vibration. A point load is so placed as to excite several modes and the resulting forced vibration is examined. In the undamped case, three in-phase and two out-of-phase solution branches have been found. The resonance curve is extremely complicated, with multiple branches and interactions between the first four modes. The amplitudes of the higher harmonics are highly influenced by damping, the presence of which can effectively attenuate internal resonances. Consequently parts of the resonance curve may be eliminated, with the resulting response comprising different distinctive branches. (C) 1996 Academic Press Limited

O presente estudo propõe uma metodologia para dissolução de tecidos biológicos em sistema de micro-ondas com radiação focalizada e posterior análise elementar por espectrometria de massas com plasma acoplado indutivamente (ICPMS). Foram determinados os elementos arsênio (As), cádmio (Cd), cobre (Cu), chumbo (Pb), níquel (Ni), vanádio (V) e zinco (Zn). A matriz nitro-peroxo-sulfúrica foi comparada com outra sem o ácido sulfúrico e apresentou melhor padrão de recuperação (acima de 90%) na quantificação dos elementos certificados no material de referência TORT-2. As interferências causadas pela presença do ácido sulfúrico na matriz digestora foram contornadas com o uso da célula de reação/colisão com gás hélio (He) e adição deste ácido na composição da curva de quantificação. O ganho analítico proporcionado pelo método de digestão em matriz sulfúrica, em sistema não pressurizado, baseia-se no incremento da temperatura reacional e na degradação completa da matéria orgânica. As digestões tradicionalmente realizadas para quantificação por ICPMS, compostas unicamente de ácido nítrico (HNO3) e peróxido de hidrogênio (H2O2), não apresentam a mesma eficiência na degradação da matéria orgânica em sistemas abertos. Por fim, este método foi aplicado satisfatoriamente em amostras de tecido hepático de peixe Mugil liza (tainha), comprovando sua eficiência em monitorar a bioacumulação, utilizando-se da sensibilidade da técnica multielementar de ICPMS

No presente trabalho, foram processados compósitos de polietileno de alta densidade (PEAD) com hidroxiapatita deficiente de cálcio (HA), com o objetivo de obter materiais com melhores propriedades mecânicas e bioatividade. A adição da HA deficiente de cálcio proporcionou um aumento no módulo de elasticidade (maior rigidez), menor resistência ao impacto e decréscimo do grau de cristalinidade do PEAD, proporcionando uma maior bioatividade ao material. A análise térmica exploratória (sistema não isotérmico) foi realizada por meio da técnica de calorimetria exploratória diferencial (DSC) e foram avaliados os teores de fosfato de cálcio e a velocidade de rotação da rosca no processamento dos materiais. No estudo da cristalização não-isotérmica observou-se uma diminuição da temperatura de cristalização com o aumento da taxa de resfriamento para todos os materiais sintetizados. A energia de ativação (Ea) da cristalização dos materiais foi avaliada por meio dos métodos Kissinger e Ozawa. A amostra com 5% de HA deficiente de cálcio e velocidade de processamento de 200 rpm foi a que apresentou menor valor de energia de ativação, 262 kJ/mol, menor desvio da linearidade e a que mais se assemelhou à matriz de PEAD sem HA. O teor de hidroxiapatita deficiente de cálcio não favorece o processo de cristalização devido à alta energia de ativação determinada pelos métodos descritos. Provavelmente, a velocidade de rotação, favorece a dispersão da carga na matriz de PEAD, dificultando o processo de cristalização. Na aplicação do método de Osawa-Avrami, os coeficientes de correlação indicaram perda na correlação linear. Estas perdas podem estar associadas a uma pequena percentagem de cristalização secundária e/ou à escolha das temperaturas utilizadas para determinar a velocidade de cristalização. Na determinação dos parâmetros pelo método de Mo, as menores percentagens de cristalização apresentaram um grande desvio da linearidade, com coeficiente de correlação bem menor que 1 e com o aumento da percentagem de cristalização, o desvio da linearidade diminui, ficando próximo de 1. Os resultados obtidos mostraram que o modelo de Mo e de Osawa-Avrami não foram capazes de definir o comportamento cinético dos materiais produzidos neste trabalho.

As simulações computacionais tem sido amplamente empregadas no estudo do escoamento darciano e não-darciano em meios porosos consolidados e não-consolidados. Neste trabalho, através de uma nova formulação para a equação de Forchheimer, foram identificadas duas novas propriedades denominados fator de comportamento do fluido, que atua paralelamente a permeabilidade, e permeabilidade equivalente global, resultado da relação anterior. Este comportamento foi estudado e validado através da implementação de um aparato experimental e um código computacional baseado no modelo de regressão-linear que, além disso, demonstrou que o escoamento, ainda que em regime não darciano, comporta-se linearmente como a equação de Darcy, ainda que o coeficiente angular desta diminuiu de acordo com a faixa do número de Reynolds atingida, sendo esta dependente do tipo de leito empregado. Ainda neste trabalho, foi implementado o método de otimização R2W para estimar os parâmetros da equação de Kozeny-Carman a partir de dados experimentais obtidos por Dias et al, a fim de simular o escoamento darciano em meios porosos. Por fim, foi alcançada excelente concordância entre os dados simulados pelo método R2W / equação de Kozeny-Carman e os dados reais.
