Coccidiosis of the domestic fowl is a worldwide disease caused by seven species of protozoan parasites of the genus Eimeria. The genome of the model species, Eimeria tenella, presents a complexity of 55-60 MB distributed in 14 chromosomes. Relatively few studies have been undertaken to unravel the complexity of the transcriptome of Eimeria parasites. We report here the generation of more than 45,000 open reading frame expressed sequence tag (ORESTES) cDNA reads of E. tenella, Eimeria maxima and Eimeria acervulina, covering several developmental stages: unsporulated oocysts, sporoblastic oocysts, sporulated oocysts, sporozoites and second generation merozoites. All reads were assembled to constitute gene indices and submitted to a comprehensive functional annotation pipeline. In the case of E. tenella, we also incorporated publicly available ESTs to generate an integrated body of information. Orthology analyses have identified genes conserved across different apicomplexan parasites, as well as genes restricted to the genus Eimeria. Digital expression profiles obtained from ORESTES/EST countings, submitted to clustering analyses, revealed a high conservation pattern across the three Eimeria spp. Distance trees showed that unsporulated and sporoblastic oocysts constitute a distinct clade in all species, with sporulated oocysts forming a more external branch. This latter stage also shows a close relationship with sporozoites, whereas first and second generation merozoites are more closely related to each other than to sporozoites. The profiles were unambiguously associated with the distinct developmental stages and strongly correlated with the order of the stages in the parasite life cycle. Finally, we present The Eimeria Transcript Database (http://www.coccidia.icb.usp.br/eimeriatdb), a website that provides open access to all sequencing data, annotation and comparative analysis. We expect this repository to represent a useful resource to the Eimeria scientific community, helping to define potential candidates for the development of new strategies to control coccidiosis of the domestic fowl. Atmosphärische Aerosole haben einen starken Einfluss auf das Klima, der bisher nur grundlegend verstanden ist und weiterer Forschung bedarf. Das atmosphärische Verhalten der Aerosolpartikel hängt maßgeblich von ihrer Größe und chemischen Zusammensetzung ab. Durch Reflexion, Absorption und Streuung des Sonnenlichtes verändern sie den Strahlungshaushalt der Erde direkt und durch ihre Einflussnahme auf die Wolkenbildung indirekt. Besonders gealterte, stark oxidierte organische Aerosole mit großem Sauerstoff-zu-Kohlenstoff-Verhältnis wirken als effektive Wolkenkondensationskeime. Neben primären Aerosolpartikeln, die direkt partikelförmig in die Atmosphäre gelangen, spielen sekundäre Aerosolpartikel eine große Rolle, die aus Vorläufergasen in der Atmosphäre entstehen. Aktuelle Forschungsergebnisse legen nahe, dass kurzkettige aliphatische Amine bei Nukleationsprozessen beteiligt sind und somit die Partikelneubildung vielerorts mitsteuern. Um die Rolle von Aminen in der Atmosphäre besser erforschen und industrielle Emissionen kontrollieren zu können, bedarf es einer zuverlässigen Methode zur Echtzeitquantifizierung gasförmiger Amine mit hoher Zeitauflösung und niedriger Nachweisgrenze.rnDas hochauflösende Flugzeit-Aerosolmassenspektrometer (HR-ToF-AMS) bietet die Möglichkeit, atmosphärische Partikel in Echtzeit zu analysieren. Dabei werden Größe, Menge und grundlegende chemische Zusammensetzung erfasst. Anorganische Aerosolbestandteile können eindeutig zugeordnet werden. Es ist jedoch kaum möglich, einzelne organische Verbindungen in den komplizierten Massenspektren atmosphärischer Aerosole zu identifizieren und quantifizieren.rnIn dieser Arbeit wird atmosphärisches Aerosol untersucht, das im Westen Zyperns während der CYPHEX-Kampagne mit einem HR-ToF-AMS gemessen wurde. An diesem Standort ist vor allem stark gealtertes Aerosol vorzufinden, das aus Zentral- und Westeuropa stammt. Lokale Einflüsse spielen fast keine Rolle. Es wurde eine durchschnittliche Massenkonzentration von 10,98 μg/m3 gefunden, zusammengesetzt aus 57 % Sulfat, 30 % organischen Bestandteilen, 12 % Ammonium, < 1 % Nitrat und < 1 % Chlorid, bezogen auf das Gewicht. Der Median des vakuum-aerodynamischen Durchmessers betrug 446,25 nm. Es wurde sehr acides Aerosol gefunden, dessen anorganische Bestandteile weitgehend der Zusammensetzung von Ammoniumhydrogensulfat entsprachen. Tag-Nacht-Schwankungen in der Zusammensetzung wurden beobachtet. Die Sulfatkonzentration und die Acidität zeigten tagsüber Maxima und nachts Minima. Konzentrationsschwankungen an Nitrat und Chlorid zeigten einen weniger ausgeprägten Rhythmus, Maxima fallen aber immer mit Minima der Sulfatkonzentration, Aerosolacidität und Umgebungstemperatur zusammen. Organische Aerosolbestandteile entsprachen stark gealtertem, schwerflüchtigem oxidiertem organischem Aerosol. Es wurde eine interne Mischung der Partikel beobachtet, die ebenfalls meist bei alten Aerosolen auftritt.rnUm mit dem HR-ToF-AMS auch einzelne organische Verbindungen identifizieren und quantifizieren zu können, wurde eine Methode entwickelt, mit der man Amine der Gasphase selektiv in künstlich erzeugte Phosphorsäurepartikel aufnimmt und so für die HR-ToF-AMS-Messung zugänglich macht. Dadurch kombiniert man die Vorteile der Online-Messung des HR-ToF-AMS mit den Vorteilen klassischer Offline-Probenahmen. So können in Echtzeit sehr einfache Massenspektren gemessen werden, in denen störende Komponenten abgetrennt sind, während die Analyten eindeutig identifiziert werden können. Systeme dieser Art wurden GTRAP-AMS (Gaseous compound TRapping in Artificially-generated Particles – Aerosol Mass Spectrometry) genannt. Kalibrierungen für (Mono)Methylamin, Dimethylamin, Trimethylamin, Diethylamin und Triethylamin ergaben Nachweisgrenzen im ppt-Bereich bei einer Zeitauflösung von 3 min. Kammerexperimente zur Aminemission von Pflanzen zeigten eine gute Übereinstimmung des neu entwickelten Systems mit einer Gasdiffusionsabscheider-Offline-Probenahme und anschließender ionenchromatographischer Analyse. Beide Methoden zeigten Reaktionen der Pflanzen auf eine Veränderung der Lichtverhältnisse, während erhöhte Ozonkonzentrationen die Aminemission nicht veränderten. Die GTRAP-AMS-Methode eignet sich bereits für die Messung von Umgebungsluftkonzentrationen an einigen Orten, für die meisten Orte reicht die Nachweisgrenze allerdings noch nicht aus. Die Technik könnte bereits zur Echtzeitkontrolle industrieller Abgasemissionen eingesetzt werden.

BACKGROUND: Drugs are routinely combined in anesthesia and pain management to obtain an enhancement of the desired effects. However, a parallel enhancement of the undesired effects might take place as well, resulting in a limited therapeutic usefulness. Therefore, when addressing the question of optimal drug combinations, side effects must be taken into account. METHODS: By extension of a previously published interaction model, the authors propose a method to study drug interactions considering also their side effects. A general outcome parameter identified as patient's well-being is defined by superposition of positive and negative effects. Well-being response surfaces are computed and analyzed for varying drugs pharmacodynamics and interaction types. In particular, the existence of multiple maxima and of optimal drug combinations is investigated for the combination of two drugs. RESULTS: Both drug pharmacodynamics and interaction type affect the well-being surface and the deriving optimal combinations. The effect of the interaction parameters can be explained in terms of synergy and antagonism and remains unchanged for varying pharmacodynamics. For all simulations performed for the combination of two drugs, the presence of more than one maximum was never observed. CONCLUSIONS: The model is consistent with clinical knowledge and supports previously published experimental results on optimal drug combinations. This new framework improves understanding of the characteristics of drug combinations used in clinical practice and can be used in clinical research to identify optimal drug dosing.

The evolution of oceanographic conditions in the upwelling region off northern Chile (18 degrees-24 degrees S) between 1996 and 1998 (including the 1997-1998 El Niño) is presented using hydrographic measurements acquired on quarterly cruises of the Chilean Fisheries Institute, with sea surface temperature (SST), sea level, and wind speeds from Arica (18.5 degrees S), Iquique (20.5 degrees S), and Antofagasta (23.5 degrees S) and a time series of vertical temperature profiles off Iquique. Spatial patterns of sea surface temperature and salinity from May 1996 to March 1997 followed a normal seasonal progression, though conditions were anomalously cool and fresh. Starting in March 1997, positive anomalies in sea level and sea surface temperature propagated along the South American coast to 37 degrees S. Maximum sea level anomalies occurred in two peaks in May-July 1997 and October 1997 to February 1998, separated by a relaxation period. Maximum anomalies (2 degrees C and 0.1 practical salinity units (psu)) extended to 400 m in December 1997 within 50 km of the coast. March 1998 presented the largest surface anomalies (> 4 degrees C and 0.6 psu). Strong poleward flow (20-35 cm s(-1) ) occurred to 400 m or deeper during both sea level maxima and weaker (10 cm s(-1) ) equatorward flow followed each peak. By May 1998, SST had returned to the climatological mean, and flow was equatorward next to the coast. However, offshore salinity remained anomalously high owing to a tongue of subtropical water extending southeast along the Peruvian coast. Conditions off northern Chile returned to normal between August and December 1998. The timing of the anomalies suggests a connection to equatorial waves. The progression of the 1997-1998 El Niño was very similar to that of 1982-1983, though with different timing with respect to seasons.

Cores from Deep Sea Drilling Project Holes 501, 504B and 505B have an unusual near-vein zonation in basalts. Megascopically, zonation occurs as differently colored strips and zones whose typical thickness does not exceed 6 to 7 cm. Microscopically, the color of zones depends on variably colored clay minerals which are the products of low-temperature hydrothermal alteration in basalt. These differently colored zones form the so called "oxidative" type of alteration of basalts. Another "background," or, less precisely termed, "non-oxidative," type of alteration in basalts is characterized by large-scale, homogeneous replacement of olivine, and filling of vesicles and cracks by an olive-brown or olive-green clay mineral. The compositions of clay minerals of the "background" type of alteration, as well as the composition of co-existing titanomagnetites, were determined with an electron microprobe. There are sharp maxima in potassium and iron content, and minima in alumina, silica, and magnesia in clay minerals in the colored zones near veins. Coloring of clay and rock-forming minerals by iron hydroxides and a decrease of the amount of titanomagnetite, which apparently was the source of redeposited iron, occur frequently in colored zones. We assume that the large-scale "background" alteration in the basalts occurred under the effect of pore waters slowly penetrating through bottom sediments. Faulting can facilitate access of fresh sea water to basalts; thus above the general homogeneous background arise zones of "oxidative" alteration along fractures in basalts. The main factors controlling these processes are time (age of basalt), grain size, temperature, thickness of sedimentary cover, and heat flow.

Acetate and hydrogen concentrations in pore fluids were measured in samples taken at seven sites from southern Hydrate Ridge (SHR) offshore Oregon, USA. Acetate concentrations ranged from 3.17 to 2515 µM. The maximum acetate concentrations occurred at Site 1251, which was drilled on a slope basin to the east of SHR at depths just above the bottom-simulating reflector (BSR) that marks the boundary of gas hydrate stability. Acetate maxima and localized high acetate concentrations occurred at the BSR at all sites and frequently corresponded with areas of gas hydrate accumulation, suggesting an empirical relationship. Acetate concentrations were typically at a minimum near the seafloor and above the sulfate/methane interface, where sulfate-reducing bacteria may consume acetate. Hydrogen concentrations in pressure core samples ranged from 16.45 to 1036 parts per million by volume (ppmv). In some cases, hydrogen and acetate concentrations were elevated concurrently, suggesting a positive correlation. However, sampling of hydrogen was limited in comparison to acetate, so any relationships between the two analytes, if present, were difficult to discern.

By analogy with the present-day ocean, primary productivity of paleoceans can be reconstructed using calculations based on content of organic carbon in sediments and their accumulation rates. Results of calculations based on published data show that primary productivity of organic carbon, mass of phosphorus involved in the process, and content of phosphorus in ocean waters were relatively stable during Cenozoic and Late Mesozoic. Prior to precipitation on the seafloor together with biogenic detritus, dissolved phosphorus could repeatedly be involved in the biogeochemical cycle. Therefore, only less than 0.1% of phosphorus is retained in bottom sediments. Bulk phosphorus accumulation rate in ocean sediments is partly consistent with calculated primary productivity. Some epochs of phosphate accumulation also coincide with maxima of primary productivity and minima of the fossilization coefficient of organic carbon. The latter fact can testify to episodes of acceleration of organic matter mineralization and release of phosphorus from sediments leading to increase in the phosphorus reserve in paleoceans and phosphate accumulation in some places.

A sediment core from the Lofoten Contourite Drift on the continental slope off Northern Norway, proximal to the former Vestfjorden-Trsnadjupet Ice Stream, details the development, variability and decline of marine margins of the northwestern Fennoscandian Ice Sheet during the time interval 25.3-14 cal ka BP, including the Last Glacial Maximum and onset of the deglaciation based on high-resolution IRD records. From the core interval between 25.3 and 17.7 cal ka BP we report data points with a mean time step of 10 years, between 17.7 cal ka BP and the Holocene time steps are typically 50 years. The core is divided into 7 informal ice-rafted debris (IRD) zones based on the variations in IRD including 7 major IRD maxima (A-G), inferred to represent periods of high iceberg production. Petrological identification reveals dominance of crystalline IRD (monocrystalline, plutonic and metamorphic rock fragments) accounting for 75-80% of total IRD assemblages, while sedimentary fragments generally account for 15-20%. The crystalline fragments (including eclogite and mangerite from a nearby terrestrial source) increase across the IRD peaks while the sedimentary fragments remain constant. This points to the importance of erosional products from icebergs originating from fast-flowing paleo-ice streams including the Vestfjorden-Trsnadjupet Ice Stream draining from the Fennoscandian mainland during the IRD maxima periods. Increased temperature of the adjacent surface water masses was probably an important external forcing factor on the Fennoscandian Ice Sheet behavior because some IRD maxima and plumite deposition from meltwater plumes post-date periods of increased sea surface temperatures. The peak IRD depositions occur in centennial and millennial time cycles (~200, 1030 and 3900 year) indicating some external forcing by solar variation. Both mechanisms could explain the observed synchronous instability of the northwestern Fennoscandian Ice Sheet to other European Ice Sheets.

The northern Arabian Sea is one of the few regions in the open ocean where thermocline water is severely depleted in oxygen. The intensity of this oxygen minimum zone (OMZ) has been reconstructed over the past 225,000 years using proxies for surface water productivity, water column denitrification, winter mixing, and the aragonite compensation depth (ACD). Changes in OMZ intensity occurred on orbital and suborbital timescales. Lowest O2 levels correlate with productivity maxima and shallow winter mixing. Precession-related productivity maxima lag early summer insolation maxima by ~6 kyr, which we attribute to a prolonged summer monsoon season related to higher insolation at the end of the summer. Periods with a weakened or even non-existent OMZ are characterized by low productivity conditions and deep winter mixing attributed to strong and cold winter monsoonal winds. The timing of deep winter mixing events corresponds with that of periods of climatic cooling in the North Atlantic region.

Variations in the strength of coastal upwelling in the South East Atlantic Ocean and summer monsoonal rains over South Africa are controlled by the regional atmospheric circulation regime. Although information about these parameters exists for the last glacial period, little detailed information exists for older time periods. New information from ODP Site 1085 for Marine Isotope Stages (MIS) 12-10 shows that glacial-interglacial productivity trends linked to upwelling variability followed a pattern similar to the last glacial cycle, with maximums shortly before glacial maxima, and minimums shortly before glacial terminations. During the MIS-11/10 transition, several periodic oscillations in productivity and monsoonal proxies are best explained by southwards shifts in the southern sub-tropical high-pressure cells followed by abrupt northwards shifts. Comparison to coeval sea-surface temperature measurements suggests that these monsoonal cycles were tightly coupled to anti-phased hemispheric climate change, with an intensified summer monsoon during periods of Northern (Southern) Hemisphere cooling (warming). The timing of these events suggests a pacing by insolation over precession periods. A lack of similar regional circulation shifts during the MIS-13/12 transition is likely due to the large equatorwards shift in the tropical convection zone that occurred during this extreme glaciation.

Planktonic foraminiferal assemblages and artificial neural network estimates of sea-surface temperature (SST) at ODP Site 1123 (41°47.2'S, 171°29.9'W; 3290 m deep), east of New Zealand, reveal a high-resolution history of glacial-interglacial (G-I) variability at the Subtropical Front (STF) for the last 1.2 million years, including the Mid-Pleistocene climate transition (MPT). Most G-I cycles of ~100 kyr duration have short periods of cold glacial and warm deglacial climate centred on glacial terminations, followed by long temperate interglacial periods. During glacial-deglacial transitions, maximum abundances of subantarctic and subtropical taxa coincide with SST minima and maxima, and lead ice volume by up to 8 kyrs. Such relationships reflect the competing influence of subantarctic and subtropical surface inflows during glacial and deglacial periods, respectively, suggesting alternate polar and tropical forcing of southern mid-latitude ocean climate. The lead of SSTs and subtropical inflow over ice volume points to tropical forcing of southern mid-latitude ocean-climate during deglacial warming. This contrasts with the established hypothesis that southern hemisphere ocean climate is driven by the influence of continental glaciations. Based on wholesale changes in subantarctic and subtropical faunas, the last 1.2 million years are subdivided into 4-distinct periods of ocean climate. 1) The pre-MPT (1185-870 ka) has high amplitude 41-kyr fluctuations in SST, superimposed on a general cooling trend and heightened productivity, reflecting long-term strengthening of subantarctic inflow under an invigorated Antarctic Circumpolar Current. 2) The early MPT (870-620 ka) is marked by abrupt warming during MIS 21, followed by a period of unstable periodicities within the 40-100 kyr orbital bands, decreasing SST amplitudes, and long intervals of temperate interglacial climate punctuated by short glacial and deglacial phases, reflecting lower meridional temperature gradients. 3) The late MPT (620-435 ka) encompasses an abrupt decrease in the subantarctic inflow during MIS 15, followed by a period of warm equable climate. Poorly defined, low amplitude G-I variations in SSTs during this interval are consistent with a relatively stable STF and evenly balanced subantarctic and subtropical inflows, possibly in response to smaller, less dynamic polar icesheets. 4) The post-MPT (435-0 ka) is marked by a major climatic deterioration during MIS 12, and a return to higher amplitude 100 kyr-frequency SST variations, superimposed on a long term trend towards cooler SSTs and increased mixed-layer productivity as the subantarctic inflow strengthened and polar icesheets expanded.

[1] Planktonic d18O and Mg/Ca-derived sea surface temperature (SST) records from the Agulhas Corridor off South Africa display a progressive increase of SST during glacial periods of the last three climatic cycles. The SST increases of up to 4°C coincide with increased abundance of subtropical planktonic foraminiferal marker species which indicates a progressive warming due to an increased influence of subtropical waters at the core sites. Mg/Ca-derived SST maximizes during glacial maxima and glacial Terminations to values about 2.5°C above full-interglacial SST. The paired planktonic d18O and Mg/Ca-derived SST records yield glacial seawater d18O anomalies of up to 0.8 per mill, indicating measurably higher surface salinities during these periods. The SST pattern along our record is markedly different from a UK'37-derived SST record at a nearby core location in the Agulhas Corridor that displays SST maxima only during glacial Terminations. Possible explanations are lateral alkenone advection by the vigorous regional ocean currents or the development of SST contrasts during glacials in association with seasonal changes of Agulhas water transports and lateral shifts of the Agulhas retroflection. The different SST reconstructions derived from UK'37 and Mg/Ca pose a significant challenge to the interpretation of the proxy records and demonstrate that the reconstruction of the Agulhas Current and interocean salt leakage is not as straightforward as previously suggested.

This paper analyzes the role of Computer Algebra Systems (CAS) in a model of learning based on competences. The proposal is an e-learning model Linear Algebra course for Engineering, which includes the use of a CAS (Maxima) and focuses on problem solving. A reference model has been taken from the Spanish Open University. The proper use of CAS is defined as an indicator of the generic ompetence: Use of Technology. Additionally, we show that using CAS could help to enhance the following generic competences: Self Learning, Planning and Organization, Communication and Writing, Mathematical and Technical Writing, Information Management and Critical Thinking.

The analysis of the interference modes has an increasing application, especially in the field of optical biosensors. In this type of sensors, the displacement Δν of the interference modes of the transduction signal is observed when a particular biological agent is placed over the biosensor. In order to measure this displacement, the position of a maximum (or a minimum) of the signal must be detected before and after placing the agent over the sensor. A parameter of great importance for this kind of sensors is the period Pν of the signal, which is inversely proportional to the optical thickness h0 of the sensor in the absence of the biological agent. The increase of this period improves the sensitivity of the sensor but it worsens the detection of the maximum. In this paper, authors analyze the propagation of uncertainties in these sensors when using least squares techniques for the detection of the maxima (or minima) of the signal. Techniques described in supplement 2 of the ISO-GUM Guide are used. The result of the analysis allows a metrological educated answer to the question of which is the optimal period Pν of the signal. El análisis del comportamiento de los modos de interferencia tiene una aplicación cada vez más amplia, especialmente en el campo de los biosensores ópticos. En este tipo de sensores se observa el desplazamiento Δν de los modos de interferencia de la señal de transducción al reconocer un de-terminado agente biológico. Para medir ese desplazamiento se debe detectar la posición de un máximo o mínimo de la señal antes y después de dicho desplazamiento. En este tipo de biosensores un parámetro de gran importancia es el periodo Pν de la señal el cual es inversamente proporcional al espesor óptico h0 del sensor en ausencia de agente biológico. El aumento de dicho periodo mejora la sensibilidad del sensor pero parece dificultar la detección del mínimo o máximo. Por tanto, su efecto sobre la incertidumbre del resultado de la medida presenta dos efectos contrapuestos: la mejora de la sensibilidad frente a la dificultad creciente en la detección del mínimo ó máximo. En este trabajo, los autores analizan la propagación de incertidumbres en estos sensores utilizando herramientas de ajuste por MM.CC. para la detección de los mínimos o máximos de la señal y técnicas de propagación de incertidumbres descritas en el suplemento 2 de la Guía ISO-GUM. El resultado del análisis permite dar una respuesta, justificada desde el punto de vista metrológico, de en que condiciones es conveniente o no aumentar el periodo Pν de la señal.

The level of mRNAs derived from the plastid-encoded psbD light-responsive promoter (LRP) is controlled by a circadian clock(s) in wheat (Triticum aestivum). The circadian oscillations in the psbD LRP mRNA level persisted for at least three cycles in continuous light and for one cycle in continuous dark, with maxima in subjective morning and minima in subjective early night. In vitro transcription in chloroplast extracts revealed that the circadian cycles in the psbD LRP mRNA level were dominantly attributed to the circadian-regulated transcription of the psbD LRP. The effects of various mutations introduced into the promoter region on the psbD LRP activity in vitro suggest the existence of two positive elements located between −54 and −36, which generally enhance the transcription activity, and an anomalous core promoter structure lacking the functional “−35” element, which plays a crucial role in the circadian fluctuation and light dependency of psbD LRP transcription activity.
