PURPOSE OF REVIEW: Current computational neuroanatomy based on MRI focuses on morphological measures of the brain. We present recent methodological developments in quantitative MRI (qMRI) that provide standardized measures of the brain, which go beyond morphology. We show how biophysical modelling of qMRI data can provide quantitative histological measures of brain tissue, leading to the emerging field of in-vivo histology using MRI (hMRI). RECENT FINDINGS: qMRI has greatly improved the sensitivity and specificity of computational neuroanatomy studies. qMRI metrics can also be used as direct indicators of the mechanisms driving observed morphological findings. For hMRI, biophysical models of the MRI signal are being developed to directly access histological information such as cortical myelination, axonal diameters or axonal g-ratio in white matter. Emerging results indicate promising prospects for the combined study of brain microstructure and function. SUMMARY: Non-invasive brain tissue characterization using qMRI or hMRI has significant implications for both research and clinics. Both approaches improve comparability across sites and time points, facilitating multicentre/longitudinal studies and standardized diagnostics. hMRI is expected to shed new light on the relationship between brain microstructure, function and behaviour, both in health and disease, and become an indispensable addition to computational neuroanatomy.

Levels of methylmalonic acid (MMA) comparable to those of human methylmalonic acidemia were achieved in blood (2-2.5 mmol/l) and brain (1.35 µmol/g) of rats by administering buffered MMA, pH 7.4, subcutaneously twice a day from the 5th to the 28th day of life. MMA doses ranged from 0.76 to 1.67 µmol/g as a function of animal age. Control rats were treated with saline in the same volumes. The animals were sacrificed by decapitation on the 28th day of age. Blood was taken and the brain was rapidly removed. Medulla, pons, the olfactory lobes and cerebellum were discarded and the rest of the brain ("cerebrum") was isolated. Body and "cerebrum" weight were measured, as well as the cholesterol and triglyceride concentrations in blood and the content of myelin, total lipids, and the concentrations of the lipid fractions (cholesterol, glycerolipids, phospholipids and ganglioside N-acetylneuraminic acid (ganglioside-NANA)) in the "cerebrum". Chronic MMA administration had no effect on body or "cerebrum" weight, suggesting that the metabolites per se neither affect the appetite of the rats nor cause malnutrition. In contrast, MMA caused a significant reduction of plasma triglycerides, but not of plasma cholesterol levels. A significant diminution of myelin content and of ganglioside-NANA concentration was also observed in the "cerebrum". We propose that the reduction of myelin content and ganglioside-NANA caused by MMA may be related to the delayed myelination/cerebral atrophy and neurological dysfunction found in methylmalonic acidemic children.

Different from most mammalian species, the optic nerve of the rabbit eye is initially formed inside the retina where myelination of the axons of the ganglion cells starts and vascularization occurs. Astrocytes are confined to these regions. The aforementioned nerve fibers known as medullated nerve fibers form two bundles that may be identified with the naked eye. The blood vessels run on the inner surface of these nerve fiber bundles (epivascularization) and, accordingly, the accompanying astrocytes lie mostly facing the vitreous body from which they are separated only by the inner limiting membrane of the retina. The arrangement of the astrocytes around blood vessels leads to the formation of structures known as glial tufts. Fragments (N = 3) or whole pieces (N = 3) of the medullated nerve fiber region of three-month-old male rabbits (Orictolagus cuniculus) were fixed in glutaraldehyde followed by osmium tetroxide, and their thin sections were examined with a transmission electron microscope. Randomly located discontinuities (up to a few micrometers long) of the basement membrane of the inner limiting membrane of the retina were observed in the glial tufts. As a consequence, a direct contact between the astrocyte plasma membrane and vitreous elements was demonstrated, making possible functional interactions such as macromolecular exchanges between this glial cell type and the components of the vitreous body.

The complex nature of spinal cord injury appears to demand a multifactorial repair strategy. One of the components that will likely be included is an implant that will fill the area of lost nervous tissue and provide a growth substrate for injured axons. Here we will discuss the role of Schwann cells (SCs) in cell-based, surgical repair strategies of the injured adult spinal cord. We will review key studies that showed that intraspinal SC grafts limit injury-induced tissue loss and promote axonal regeneration and myelination, and that this response can be improved by adding neurotrophic factors or anti-inflammatory agents. These results will be compared with several other approaches to the repair of the spinal cord. A general concern with repair strategies is the limited functional recovery, which is in large part due to the failure of axons to grow across the scar tissue at the distal graft-spinal cord interface. Consequently, new synaptic connections with spinal neurons involved in motor function are not formed. We will highlight repair approaches that did result in growth across the scar and discuss the necessity for more studies involving larger, clinically relevant types of injuries, addressing this specific issue. Finally, this review will reflect on the prospect of SCs for repair strategies in the clinic.

Afin de mieux comprendre l’évolution des fonctions du récepteur EphA4 pendant le développement du système nerveux central (SNC), nous avons étudié sa localisation cellulaire et subcellulaire dans l’hippocampe du rat, d’abord chez l’adulte, puis pendant le développement postnatal, ainsi que ses rôles potentiels dans la genèse, la migration ou la maturation des cellules granulaires dans l’hippocampe adulte. Pour ce faire, nous avons utilisé la méthode d’immunocytochimie en microscopie photonique, électronique et confocale. En microscopie photonique, une forte immunoréactivité (peroxydase/DAB) pour EphA4 est observée aux jours 1 et 7 suivant la naissance (P1 et P7) dans les couches de corps cellulaires, avec un marquage notamment associé à la surface des corps cellulaires des cellules granulaires et pyramidales, ainsi que dans les couches de neuropile du gyrus dentelé et des secteurs CA3 et CA1. L’intensité du marquage diminue progressivement dans les couches de corps cellulaires, entre P7 et P14, pour devenir faible à P21 et chez l’adulte, tandis qu’elle persiste dans les couches de neuropile, sauf celles qui reçoivent des afférences du cortex entorhinal. En microscopie électronique, après marquage à la peroxydase/DAB, EphA4 décore toute la surface des cellules pyramidales et granulaires, du corps cellulaire jusqu’aux extrémités distales, entre P1 et P14, pour devenir confiné aux extrémités synaptiques, c’est-à-dire les terminaisons axonales et les épines dendritiques, à P21 et chez l’adulte. À la membrane plasmique des astrocytes, EphA4 est redistribué comme dans les neurones, marquant le corps cellulaire et ses prolongements proximaux à distaux, à P1 et P7, pour devenir restreint aux prolongements périsynaptiques distaux, à partir de P14. D’autre part, des axones en cours de myélinisation présentent souvent une forte immunoréactivité punctiforme à leur membrane plasmique, à P14 et P21. En outre, dans les neurones et les astrocytes, le réticulum endoplasmique, l’appareil de Golgi et les vésicules de transport, organelles impliquées dans la synthèse, la modification posttraductionnelle et le transport des protéines glycosylées, sont aussi marqués, et plus intensément chez les jeunes animaux. Enfin, EphA4 est aussi localisé dans le corps cellulaire et les dendrites des cellules granulaires générées chez l’adulte, au stade de maturation où elles expriment la doublecortine (DCX). De plus, des souris adultes knockouts pour EphA4 présentent des cellules granulaires DCX-positives ectopiques, c’est-à-dire positionnées en dehors de la zone sous-granulaire, ce qui suggère un rôle d’EphA4 dans la régulation de leur migration. Ces travaux révèlent ainsi une redistribution d’EphA4 dans les cellules neuronales et gliales en maturation, suivant les sites cellulaires où un remodelage morphologique s’effectue : les corps cellulaires lorsqu’ils s’organisent en couches, les prolongements dendritiques et axonaux pendant leur croissance, guidage et maturation, puis les épines dendritiques, les terminaisons axonales et les prolongements astrocytaires distaux associés aux synapses excitatrices, jusque chez l’adulte, où la formation de nouvelles synapses et le renforcement des connexions synaptiques existantes sont exercés. Ces localisations pourraient ainsi correspondre à différents rôles d’EphA4, par lesquels il contribuerait à la régulation des capacités plastiques du SNC, selon le stade développemental, la région, l’état de santé, ou l’expérience comportementale de l’animal.

El dolor es un síntoma muy común en pacientes con Esclerosis Múltiple, pues del 42 al 65% de los enfermos lo presentan, y es calificado como el síntoma más severo entre el 8 y el 32%. Todos los síndromes dolorosos centrales se presentan por lesión o disfunción del sistema nervioso central, causando discapacidad severa y deterioro de la calidad de vida de los pacientes.

Schizophrenia is likely to be a consequence of serial alterations in a number of genes that, together with environmental factors, will lead to the establishment of the illness. The dorsolateral prefrontal cortex (Brodmann`s Area 46) is implicated in schizophrenia and executes high functions such as working memory, differentiation of conflicting thoughts, determination of right and wrong concepts, correct social behavior and personality expression. We performed a comparative proteome analysis using two-dimensional gel electrophoresis of pools from 9 schizophrenia and 7 healthy control patients` dorsolateral prefrontal cortex aiming to identify, by mass spectrometry, alterations in protein expression that could be related to the disease. In schizophrenia-derived samples, our analysis revealed 10 downregulated and 14 upregulated proteins. These included alterations previously implicated in schizophrenia, such as oligodendrocyte-related proteins (myelin basic protein and transferrin), as well as malate dehydrogenase, aconitase, ATP synthase subunits and cytoskeleton-related proteins. Also, six new putative disease markers were identified, including energy metabolism, cytoskeleton and cell signaling proteins. Our data not only reinforces the involvement of proteins previously implicated in schizophrenia, but also suggests new markers, providing further information to foster the comprehension of this important disease. Objectives: To establish normative amplitude values for relative difference measurements of the middle latency response (MLR) in normal-hearing pediatrics and to determine if these measurements provided a significant reduction of within-group variability when compared to raw, absolute amplitude measures. A relative amplitude difference is defined in the present paper as the difference in Na-Pa amplitude between two electrodes (e.g. vertical bar Na-Pa at C3 minus Na-Pa at C4 vertical bar, or electrode effects) or between two ears (e.g. vertical bar Na-Pa on left ear stimulation minus Na-Pa on right ear stimulation vertical bar, or ear effects). In contrast, an absolute amplitude is defined as a single Na-Pa measurement made at one electrode for stimulation of one ear (e.g. Na-Pa measured at C3 on left ear stimulation). Design: Cross-sectional study. Study sample: 155 pediatrics with normal peripheral and central hearing, and no history of psychological, neurological, or learning disability issues. Results: Within-group variability was significantly smaller for relative differences when compared to absolute amplitude measures. Electrode effects showed significantly less variability than ear effects. Normative values for ear and electrode effects were reported. Conclusions: Relative differences may provide better utility in the clinical diagnosis of central auditory pathology in pediatrics when compared to absolute amplitude measures because these difference measures show significantly lower variability when examined across subjects.

The induction of autoimmune encephalomyelitis (EAE) in Lewis rats results in a period of exacerbation followed by complete recovery. Therefore, this model is widely used for studying the evolution of multiple sclerosis. In the present investigation, differentially expressed proteins in the spinal cord of Lewis rats during the evolution of EAE were assessed using the combination of 2DE and MALDI-TOF MS. The majority of the differentially expressed proteins were identified during the acute phase of EAE, in relation to naive control animals. On the other hand, recovered rats presented a similar protein expression pattern in comparison with the naive ones. This observation can be explained, at least in part, by the intense catabolism existent in acute phase due to nervous tissue damage. In recovered rats, we have described the upregulation of proteins that are apparently involved in the recovery of damaged tissue, such as light and medium neurofilaments, glial fibrillary acidic protein, tubulins subunits, and quaking protein. These proteins are involved mainly in cell growth, myelination, and remyelination as well as in astrocyte and oligodendrocyte maturation. The present study has demonstrated that the inflammatory response, characterized by an increase of the proliferative response and infiltration of autoreactive T lymphocytes in the central nervous system, occurs simultaneously with neurodegeneration.

NG2 is a transmembrane proteoglycan with two N-terminal LNS domains and a C-terminal PDZ-binding motif. It is expressed in the developing and adult CNS by oligodendroglial precursor cells and subpopulations of perisynaptic glia and elsewhere by many immature cell types. In order to elucidate the functions of the protein and the heterogenous cell population which expresses it, we undertook to identify and characterise interaction partners of the molecule. The presence of the C-terminal PDZ recognition site in NG2 suggested PDZ-domain proteins as intracellular binding partners. In this work, interaction between the PDZ protein Syntenin and NG2 has been characterised. Syntenin is known to be involved in plasma membrane dynamics, metastasis and adhesion. Syntenin may thus link NG2 to the cytoskeleton, mediating migration of developing oligodendrocytes to axonal tracts prior to myelination, as well as process movement of NG2+ perisynaptic glia. NG2 is involved in cell spreading and polyclonal antibodies against NG2 inhibit the migration of immature glia and cell lines expressing the molecule. In this work we have characterised the segments of the extracellular portion of NG2 that are involved in migration. We found that the extracellular region immediately preceding the transmembrane segment is most important for cell motility. As part of this thesis, biochemical approaches to identify a trans-binding ligand interacting with the extracellular part of NG2 was also explored.

Sono stati studiati gli effetti tossici dell’esposizione cronica a cobalto e cromo. In passato, questa tossicità, che colpiva lavoratori esposti per ragioni occupazionali, è stata un problema molto sentito. Tuttavia, recenti pubblicazioni hanno descritto una specifica tossicità mediata da elevati livelli di cobalto e cromo, anche in pazienti portatori di protesi metalliche, quali gli impianti d’anca. Anche se sintomi clinici tra cui, cecità, sordità e neuropatia periferica, suggeriscono uno specifico neurotropismo, ancora poco è conosciuto delle basi neuropatologiche di questo processo ed oltretutto non ne è ancora stata apportata un’evidenza sperimentale. In questo progetto di ricerca, quindi, si è voluto approfondire il meccanismo patogenetico da cui scaturiscono tali sintomi neurologici, utilizzando come modello sperimentale il coniglio. Conigli New Zealand White sono stati trattati con dosi endovenose ripetute di cobalto e cromo, inoculati singolarmente od in associazione tra loro. Nessuna evidente alterazione clinica o patologica è stata associata alla somministrazione di solo cromo, nonostante gli elevati livelli in sangue e tessuti, mentre i trattati con cobalto-cromo o solo cobalto hanno mostrato segni clinici gravanti sul sistema vestibolo-cocleare; il cobalto, quindi, è stato identificato come il maggiore elemento scatenante neurotossicità. Inoltre all’esame istopatologico gli animali hanno mostrato severa deplezione delle cellule gangliari retiniche e cocleari, assieme a danno al nervo ottico e perdita di cellule sensitive capellute dell’orecchio. È risultato infine evidente che la gravità delle alterazioni è stata correlata al dosaggio ed al tempo di esposizione; dati questi che confermano, quindi, le precedenti osservazioni fatte su pazienti umani esposti a rilascio abnorme di cobalto e cromo da usura di protesi d’anca. È stato ipotizzato che il cobalto agisca sui mitocondri provocando l’incremento di produzione di specie reattive dell’ossigeno e il rilascio di fattori proapoptotici, causando sulle cellule neuronali un danno proporzionale al loro fabbisogno energetico e grado di mielinizzazione.

During central nervous system myelination, oligodendrocytes extend membrane processes towards an axonal contact site which is followed by ensheathment resulting in a compacted multilamellar myelin sheath. The formation of this axon-glial unit facilitates rapid saltatory propagation of action potentials along the axon and requires the synthesis and transport of copious amounts of lipids and proteins to the axon-glial contact site. Fyn is a member of the Src family of non receptor tyrosine kinases and inserted into the inner leaflet of the oligodendrocyte membrane by acylation. Fyn activity plays a pivotal role in the maturation of oligodendrocytes and the myelination process. It was suggested previously that Fyn kinase can be stimulated by binding of a neuronal ligand to oligodendroglial F3/ contactin, a glycosyl-phosphatidyl-inositol anchored immunoglobulin superfamily (IgSF) member protein. It could be shown here, that neuronal cell adhesion molecule L1 binds to oligodendrocytes in an F3-dependent manner and activates glial Fyn. In the search for downstream participants of this novel axon-glial signalling cascade, heterogeneous nuclear ribonucleoprotein (hnRNP) A2 was identified as a novel Fyn target in oligodendrocytes. HnRNP A2 was known to be involved in the localisation of translationally repressed myelin basic protein (MBP) mRNA by binding to a cis acting A2 response element (A2RE) present in the 3’ untranslated region. Transport of MBP mRNAs occurs in RNA-protein complexes termed RNA granules and translational repression during transport is achieved by hnRNP A2-mediated recruitment of hnRNP E1 to the granules. It could be shown here, that Fyn activity leads to enhanced translation of reporter mRNA containing a part of the 3’ UTR of MBP including the A2RE. Furthermore hnRNP E1 seems to dissociate from RNA granules in response to Fyn activity and L1 binding. These findings suggest a novel form of neuron- glial communication: Axonal L1 binding to oligodendroglial F3 activates Fyn kinase. Activated Fyn phosphorylates hnRNP A2 leading to removal of hnRNP E1 from RNA granules initiating the translation of MBP mRNA. MBP is the second most abundant myelin protein and mice lacking this protein show a severe hypomyelination phenotype. Moreover, the brains of Fyn knock out mice contain reduced MBP levels and are hypomyelinated. Hence, L1-mediated MBP synthesis via Fyn as a central molecule could be part of a regulatory mechanism required for myelinogenesis in the central nervous system.

Die Bildung, Aufrechterhaltung und die Funktionalität von Oligodendrozyten, den myelinisierenden Zellen des ZNS, bedarf einer präzisen Regulation von Ereignissen wie Migration, Proliferation und Differenzierung. Die Src-Kinasen spielen in vielen Signalkaskaden eine zentrale Rolle. In murinen Oligodendrozyten werden die beiden Src-Kinasen Fyn und Lyn exprimiert, diese Arbeit konzentriert sich auf die Analyse der Lyn Kinase. Es konnte die Expression von Lyn in Oligodendrozyten und Myelin in vitro und in vivo bestätigen werden, sowie die Lokalisation innerhalb und außerhalb von rafts. Je älter die Mäuse, desto weniger Lyn wird in Myelin exprimiert und desto stärker ist das verbleibende Lyn in den rafts lokalisiert. Die Aktivität von Lyn ist im Myelin 12-Tage alter Mäuse am höchsten. Synchronisation des Zellzyklus in Oli-neu Zellen zeigte eine zyklische Expression von Lyn. Transfektion aktiver und inaktiver Konstrukte machte deutlich, dass eine Lyn Aktivität die Zellausläufer Bildung, sowie die Differenzierung der Zelle, hemmt. Es konnte eine Assoziation der GluRB/C Untereinheit des AMPA Rezeptors mit Lyn nachgewiesen werden. Stimulierung des AMPA-Rezeptors mit Glutamat führte zu einer Aktivierung der Lyn Kinase und daraus resultierend, zu einer Inhibierung der Zelldifferenzierung. Welche biologische Relevanz diese Ergebnisse haben könnten, wurde in drei Hypothesen festgehalten. Zum einen könnte die Lyn Kinase den zeitlichen Übergang von Proliferation und Differenzierung in Oligodendrozyten regulieren und zwar sowohl in perinatalen oligodendroglialen Vorläuferzellen, als auch, unter besonderen Umständen, in adulten oligodendroglialen Vorläuferzellen. Des Weiteren könnte die Lyn Kinase auch eine Rolle in der Neuron-Glia-Synapsenbildung spielen. Weitere Versuche, insbesondere in vivo, müssen folgen, um diese Hypothesen zu bestätigen oder zu widerlegen.
