Mechanistic determinants of bacterial growth, death, and spread within mammalian hosts cannot be fully resolved studying a single bacterial population. They are also currently poorly understood. Here, we report on the application of sophisticated experimental approaches to map spatiotemporal population dynamics of bacteria during an infection. We analyzed heterogeneous traits of simultaneous infections with tagged Salmonella enterica populations (wild-type isogenic tagged strains [WITS]) in wild-type and gene-targeted mice. WITS are phenotypically identical but can be distinguished and enumerated by quantitative PCR, making it possible, using probabilistic models, to estimate bacterial death rate based on the disappearance of strains through time. This multidisciplinary approach allowed us to establish the timing, relative occurrence, and immune control of key infection parameters in a true host-pathogen combination. Our analyses support a model in which shortly after infection, concomitant death and rapid bacterial replication lead to the establishment of independent bacterial subpopulations in different organs, a process controlled by host antimicrobial mechanisms. Later, decreased microbial mortality leads to an exponential increase in the number of bacteria that spread locally, with subsequent mixing of bacteria between organs via bacteraemia and further stochastic selection. This approach provides us with an unprecedented outlook on the pathogenesis of S. enterica infections, illustrating the complex spatial and stochastic effects that drive an infectious disease. The application of the novel method that we present in appropriate and diverse host-pathogen combinations, together with modelling of the data that result, will facilitate a comprehensive view of the spatial and stochastic nature of within-host dynamics. © 2008 Grant et al.

For the first time in its history, the International Symposium on Sea Turtle Biology and Conservation migrated to a site outside of the United States. Thus the Eighteenth edition was hosted by the Mazatlán Research Unit of the Instituto de Ciencias del Mar y Limnología of the Mexican National Autonomous University (UNAM) in Mazatlán, Sinaloa (Mexico) where it was held from 3-7, March, 1998. Above all, our symposium is prominent for its dynamism and enthusiasm in bringing together specialists from the world´s sea turtle populations. In an effort to extend this philosophy, and fully aware of how fast the interest in sea turtles has grown, the organizers paid special attention to bring together as many people as possible. With the tremendous efforts of the Travel Committee and coupled with a special interest by the Latin American region´s devotees, we managed to get 653 participants from 43 countries. The number of presentations increased significantly too, reaching a total of 265 papers, ranging from cutting-edge scientific reports based on highly sophisticated methods, to the experiences and successes of community-based and environmental education programs. A priority given by this symposium was the support and encouragement for the construction of "bridges" across cultural and discipline barriers. We found success in achieving a multinational dialogue among interest groups- scientists, resource managers, decision makers, ngo's, private industry. There was a broad representation of the broad interests that stretch across these sectors, yet everyone was able to listen and offer their own best contribution towards the central theme of the Symposium: the conservation of sea turtles and the diversity of marine and coastal environments in which they develop through their complicated and protracted life cycle. Our multidisciplinary approach is highly important at the present, finding ourselves at a cross roads of significant initiatives in the international arena of environmental law, where the conservation of sea turtles has a key role to play. Many, many people worked hard over the previous 12 months, to make the symposium a success. Faults can slip either aseismically or through episodic seismic ruptures, but we still do not understand the factors which determine the partitioning between these two modes of slip. This challenge can now be addressed thanks to the dense set of geodetic and seismological networks that have been deployed in various areas with active tectonics. The data from such networks, as well as modern remote sensing techniques, indeed allow documenting of the spatial and temporal variability of slip mode and give some insight. This is the approach taken in this study, which is focused on the Longitudinal Valley Fault (LVF) in Eastern Taiwan. This fault is particularly appropriate since the very fast slip rate (about 5 cm/yr) is accommodated by both seismic and aseismic slip. Deformation of anthropogenic features shows that aseismic creep accounts for a significant fraction of fault slip near the surface, but this fault also released energy seismically, since it has produced five M_w>6.8 earthquakes in 1951 and 2003. Moreover, owing to the thrust component of slip, the fault zone is exhumed which allows investigation of deformation mechanisms. In order to put constraint on the factors that control the mode of slip, we apply a multidisciplinary approach that combines modeling of geodetic observations, structural analysis and numerical simulation of the "seismic cycle". Analyzing a dense set of geodetic and seismological data across the Longitudinal Valley, including campaign-mode GPS, continuous GPS (cGPS), leveling, accelerometric, and InSAR data, we document the partitioning between seismic and aseismic slip on the fault. For the time period 1992 to 2011, we found that about 80-90% of slip on the LVF in the 0-26 km seismogenic depth range is actually aseismic. The clay-rich Lichi M\'elange is identified as the key factor promoting creep at shallow depth. Microstructural investigations show that deformation within the fault zone must have resulted from a combination of frictional sliding at grain boundaries, cataclasis and pressure solution creep. Numerical modeling of earthquake sequences have been performed to investigate the possibility of reproducing the results from the kinematic inversion of geodetic and seismological data on the LVF. We first investigate the different modeling strategy that was developed to explore the role and relative importance of different factors on the manner in which slip accumulates on faults. We compare the results of quasi dynamic simulations and fully dynamic ones, and we conclude that ignoring the transient wave-mediated stress transfers would be inappropriate. We therefore carry on fully dynamic simulations and succeed in qualitatively reproducing the wide range of observations for the southern segment of the LVF. We conclude that the spatio-temporal evolution of fault slip on the Longitudinal Valley Fault over 1997-2011 is consistent to first order with prediction from a simple model in which a velocity-weakening patch is embedded in a velocity-strengthening area.

Este estudo objetiva a sistematização do conhecimento, através de uma abordagem multidisciplinar, de forma a conformar uma proposta inicial de protocolo de gestão ocupacional e ambiental em grandes túneis urbanos em âmbito nacional. Há na presente pesquisa uma atenção particular aos túneis urbanos dedicados ao tráfego de veículos automotores, devido às suas características físico-espaciais que os tornam especialmente perigosos e tóxicos aos seus usuários e aos funcionários envolvidos. A relativa carência de material bibliográfico de caráter sistêmico e específico no Brasil enseja à busca por referências em documentos desenvolvidos em outros países. Ainda que em nível preliminar, a compilação crítica de dados, normas e referências pesquisadas, à luz do contexto e dos instrumentos normativos brasileiros referentes ao tema, traduz-se no aspecto original desta proposta dissertativa, a ser possivelmente utilizada como ponto de partida para estudos mais extensos. São abordadas as seguintes disciplinas relativas ao objeto de pesquisa: a) Características físico-espaciais; b) Formas de uso; c) Sistema de ventilação e exaustão; d) Sistema de iluminação; e) Sistema de segurança contra sinistros e incêndio; f) Sistema de fiscalização e orientação de tráfego, comunicação e sinalização; g) Sistema de drenagem; h) Sistema de gerenciamento de resíduos sólidos. O túnel Rebouças (1962-1965) e o túnel Engenheiro Raymundo de Paula Soares (Linha Amarela) (1997), distanciados por mais de três décadas, foram escolhidos como estudos de caso, não tão somente devido aos seus portes, fluxos intensos de veículos etc., mas também devido aos contextos das evoluções normativas em que se inserem.

O presente estudo, como investigação acadêmica de abordagem multidisciplinar, tem como objetivo geral contribuir para a identificação de possíveis caminhos que minimizem as discrepâncias verificadas entre os valores contábil e econômico das marcas. Neste contexto, em que o Brasil vivencia a transição para o International Financial Reporting Standards (IFRS), faz-se mister investigar a relação destas normas com a evidenciação da significância econômica desses bens, com o intuito de analisar as restrições normativas à contabilização das marcas, e identificar causas e impactos dessas discrepâncias. A pesquisa, de características essencialmente exploratória e qualitativa, aborda os conceitos teóricos de Ativos Intangíveis, Marcas, Brand Equity, Brand Value e Normativa Contábil, e utiliza a consulta bibliográfica e a entrevista presencial como formas de coleta de dados. No estudo, foram comparados os valores contábil e econômico das 31 marcas brasileiras listadas nos rankings de marcas mais valiosas divulgados em 2011 pelas três principais consultorias especializadas em avaliação de marcas; e entrevistadas 11 pessoas entre acadêmicos e profissionais das áreas Contábil, Econômica, Administrativa e Financeira, com o propósito primário de entender a essência da problemática da evidenciação contábil da marca. Como resultado, identificou-se que a questão basilar para a solução desse problema reside na confiabilidade da mensuração do valor monetário da marca, e que, como ainda não há consenso científico sobre a aplicabilidade dos métodos disponíveis na literatura aos diferentes propósitos de avaliação desse bem, é preciso que se desenvolvam novos e contínuos estudos a fim de identificar um padrão metodológico consistente, uniforme e objetivo que melhor se aplique aos interesses da Contabilidade como instrumento de gestão.

O objetivo desta pesquisa foi analisar os conteúdos dos relatos verbais de profissionais de enfermagem, vítimas de assédio moral durante o exercício de suas atividades laborais. Para tanto, foi utilizada uma amostra previamente selecionada, constituída de 31 profissionais de enfermagem, sendo todos atuantes em unidades de saúde, tanto da rede pública quanto da rede privada. Essa amostra foi extraída do universo de 3.498 profissionais de formações diversas, oriundos de vários Estados brasileiros. Trata-se de uma pesquisa quanti-qualitativa, sendo utilizado, como instrumento para a coleta de dados, o questionário específico de Assédio Moral, gentilmente cedido pela pesquisadora Margarida Maria Silveira Barreto e constituído de 48 questões, abertas e fechadas. Os itens desse instrumento compreendem a caracterização dos participantes, a identificação de situações específicas de assédio moral, de fatores relacionados à empresa, a organização e comunicação no trabalho. O tratamento dos dados estatísticos foi realizado com o apoio do teste de Qui-Quadrado, e os dados qualitativos foram obtidos com base em relatos verbais, os quais foram organizados em categorias de respostas dos participantes. Os resultados quantitativos da pesquisa apontaram diferenças significativas entre enfermeiros e auxiliares de enfermagem quanto a favorabilidade em relação aos objetivos pouco claros na organização do trabalho. A partir da análise qualitativa dos dados emergiram como resultados, três categorias de respostas: Hierarquia e Relação de Poder; Organização do Trabalho; e Comunicação no Trabalho. De posse dos resultados da presente pesquisa, a meta é contribuir para a melhoria das condições de trabalho, de forma a minimizar o aparecimento de humilhações psicológicas ou assédio moral no ambiente de trabalho de enfermagem. Diante desse quadro, é de importância aprofundar estudos sobre o assédio moral, considerando-se a adoção de legislação específica e a formulação de políticas públicas para o combate dessa prática na profissão de enfermagem. O que se propõe com esse estudo é auxiliar na formulação de futuras políticas públicas, de modo a evitar conseqüências nefastas ao trabalhador, ao próprio trabalho e à sociedade de um modo geral, pois o combate ao assédio moral precisa ser intensificado, exigindo uma abordagem multidisciplinar, com a participação de todos os segmentos da sociedade.

A análise em tela investiga a construção do projeto político pedagógico do Departamento de Psicologia da Universidade Estadual da Paraíba (2011), tendo como parâmetros o projeto político pedagógico inicial do curso (1999) e a produção de textos, discursos e documentos no cenário nacional que versam sobre a reforma curricular da graduação em Psicologia no Brasil. A abordagem do ciclo contínuo de políticas, proposta por Stephen Ball e a vertente analítica das comunidades epistêmicas são a base teórico-metodológica escolhida. Também há uma interlocução com o campo da Psicologia da Educação, em uma perspectiva multidisciplinar, apoiada nas contribuições de Michel Foucault sobre discurso e poder. Compreendemos, a partir desta perspectiva, que as políticas curriculares são pensadas e produzidas num processo complexo que envolve diversas ações nos contextos da influência, produção e da prática (BALL e BOWE, 1992). Ainda que o foco de nossa pesquisa seja no contexto da prática (reforma curricular do curso de Psicologia), a análise considera os outros contextos pelas relações de interdependência que há entre eles. O intervalo cronológico definido para compreensão das políticas curriculares para a formação em Psicologia (1999-2011) foi atento às definições ocorridas, neste período, as quais permitem a análise mais precisa do nosso objeto: as recontextualizações institucionais na reforma curricular da Psicologia da UEPB. Elas são analisadas a partir da identificação de dois projetos de formação em disputa no cenário nacional: um projeto generalista e outro especialista. O primeiro tem como características principais a defesa de um curso com denominação única (Curso de Psicologia), formação focada na profissionalização, substituição das antigas áreas de habilitação para ênfases curriculares, dissociação entre formação profissional e formação do professor e, por fim, extinção da terminologia de bacharel. Já o segundo defende dois aspectos que dizem respeito a reivindicação da formação do bacharel e a formação do professor. Identificamos que o projeto de formação generalista que tem como maior representante o CFP, tornou-se vencedor, no cenário nacional, tendo repercussões no contexto da prática na UEPB. Há algumas sintonias entre as diretrizes curriculares construídas para a Psicologia na universidade e as diretrizes do CFP que defendem a inclusão das práticas emergentes ao currículo da graduação em Psicologia no país. Esta constatação atesta a ação de uma comunidade epistêmica que defendeu um projeto de formação focado na profissionalização na área clínica. Assim, concluímos que, ainda que haja a definição da profissionalização como meta do curso de Psicologia da UEPB, é a área/ênfase de Psicologia Organizacional e do Trabalho que aparece como dominante; a área/ênfase de Psicologia Clínica tem suas horas diminuídas; as ênfases curriculares são correlatas das áreas de formação do PPPI (1999) e a formação do professor de Psicologia é excluída do NPPP (2011), ficando para o futuro a construção de um projeto específico para esta formação. Portanto, a reforma curricular de Psicologia da Universidade Estadual da Paraíba, apesar de pretender uma formação generalista, reproduz uma lógica especialista e o perfil do curso foca na profissionalização, mas com uma ascensão da área/ênfase Psicologia Organizacional e do Trabalho.

This thesis explores the impact of Christianity on the landscape in Ireland from the conversion period to the coming of the Anglo-Normans. The premise is that ecclesiastical and secular settlement formed a cohesive whole which characterised the societal organisation of early medieval Ireland. The matter of the thesis is to isolate some of the agents of cohesion to see was this homogenous or did it vary in different areas. One of these agents was the ownership of land and the thesis undertakes to identify ecclesiastical landholding and examine the manner of settlement on it. A corollary is to explore the contribution of the genealogical link between kin-group, founding saint and territory to the construction of local identities. This necessitated a narrow focus; thus small study areas were chosen, which approximated to early medieval kingdoms in North Louth, Rathdown, Co Dublin and Ross, Co Cork. A multidisciplinary approach was taken using both archaeological and documentary evidence. The thesis found ecclesiastical sites were at the same density through the study areas, but there were considerable regional variations in related secular settlement. Ecclesiastical estates were identified in the three study areas and common settlement patterns were found in two of them. Settlement in all areas indicated the foundation of minor churches by local groups. Ecclesiastical sites were found to be integral to kin-group identity and status, but the manner in which each group negotiated this, was very different. Finally the thesis examined material evidence for a change from diffused to concentrated power in the political organisation of Irish society, a process entwined with developments of the Viking Age. This centralisation of power and associated re-formation of identity was still often mediated through the ecclesiastical sphere but the thesis demonstrates diversity in the materialising of the mediation.

A distinctive subset of metastatic breast cancer (MBC) is oligometastatic disease, which is characterized by single or few detectable metastatic lesions. The existing treatment guidelines for patients with localized MBC include surgery, radiotherapy, and regional chemotherapy. The European School of Oncology-Metastatic Breast Cancer Task Force addressed the management of these patients in its first consensus recommendations published in 2007. The Task Force endorsed the possibility of a more aggressive and multidisciplinary approach for patients with oligometastatic disease, stressing also the need for clinical trials in this patient population. At the sixth European Breast Cancer Conference, held in Berlin in March 2008, the second public session on MBC guidelines addressed the controversial issue of whether MBC can be cured. In this commentary, we summarize the discussion and related recommendations regarding the available therapeutic options that are possibly associated with cure in these patients. In particular, data on local (surgery and radiotherapy) and chemotherapy options are discussed. Large retrospective series show an association between surgical removal of the primary tumor or of lung metastases and improved long-term outcome in patients with oligometastatic disease. In the absence of data from prospective randomized studies, removal of the primary tumor or isolated metastatic lesions may be an attractive therapeutic strategy in this subset of patients, offering rapid disease control and potential for survival benefit. Some improvement in outcome may also be achieved with optimization of systemic therapies, possibly in combination with optimal local treatment. © 2010. Published by Oxford University Press.

Marine environments are greatly affected by climate change, and understanding how this perturbation affects marine vertebrates is a major issue. In this context, it is essential to identify the environmental drivers of animal distribution. Here, we focused on the little auk (Alle alle), one of the world’s most numerous seabirds and a major component in Arctic food webs. Using a multidisciplinary approach, we show how little auks adopt specific migratory strategies and balance environmental constraints to optimize their energy budgets. Miniature electronic loggers indicate that after breeding, birds from East Greenland migrate .2000 km to overwinter in a restricted area off Newfoundland. Synoptic data available from the Continuous Plankton Recorder (CPR) indicate that this region harbours some of the highest densities of the copepod Calanus finmarchicus found in the North Atlantic during winter. Examination of large-scale climatic and oceanographic data suggests that little auks favour patches of high copepod abundance in areas where air temperature ranges from 0uC to 5uC. These results greatly advance our understanding of animal responses to extreme environmental constraints, and highlight that information on habitat preference is key to identifying critical areas for marine conservation.

During recent decades anthropogenic activities have dramatically impacted the Black Sea ecosystem. High levels of riverine nutrient input during the 1970s and 1980s caused eutrophic conditions including intense algal blooms resulting in hypoxia and the subsequent collapse of benthic habitats on the northwestern shelf. Intense fishing pressure also depleted stocks of many apex predators, contributing to an increase in planktivorous fish that are now the focus of fishing efforts. Additionally, the Black Sea's ecosystem changed even further with the introduction of exotic species. Economic collapse of the surrounding socialist republics in the early 1990s resulted in decreased nutrient loading which has allowed the Black Sea ecosystem to start to recover, but under rapidly changing economic and political conditions, future recovery is uncertain. In this study we use a multidisciplinary approach to integrate information from socio-economic and ecological systems to model the effects of future development scenarios on the marine environment of the northwestern Black Sea shelf. The Driver–Pressure–State-Impact-Response framework was used to construct conceptual models, explicitly mapping impacts of socio-economic Drivers on the marine ecosystem. Bayesian belief networks (BBNs), a stochastic modelling technique, were used to quantify these causal relationships, operationalise models and assess the effects of alternative development paths on the Black Sea ecosystem. BBNs use probabilistic dependencies as a common metric, allowing the integration of quantitative and qualitative information. Under the Baseline Scenario, recovery of the Black Sea appears tenuous as the exploitation of environmental resources (agriculture, fishing and shipping) increases with continued economic development of post-Soviet countries. This results in the loss of wetlands through drainage and reclamation. Water transparency decreases as phytoplankton bloom and this deterioration in water quality leads to the degradation of coastal plant communities (Cystoseira, seagrass) and also Phyllophora habitat on the shelf. Decomposition of benthic plants results in hypoxia killing flora and fauna associated with these habitats. Ecological pressure from these factors along with constant levels of fishing activity results in target stocks remaining depleted. Of the four Alternative Scenarios, two show improvements on the Baseline ecosystem condition, with improved waste water treatment and reduced fishing pressure, while the other two show a worsening, due to increased natural resource exploitation leading to rapid reversal of any recent ecosystem recovery. From this we conclude that variations in economic policy have significant consequences for the health of the Black Sea, and ecosystem recovery is directly linked to social–economic choices.

Para ello, se analiza y contextualiza la web semántica desde una perspectiva general para posteriormente analizar las principales iniciativas que trabajan con lo audiovisual: Proyecto MuNCH, Proyecto S5T, Semantic Television y VideoActive.

Venom has only been recently discovered to be a basal trait of the Anguimorpha lizards. Consequently, very little is known about the timings of toxin recruitment events, venom protein molecular evolution, or even the relative physical diversifications of the venom system itself. A multidisciplinary approach was used to examine the evolution across the full taxonomical range of this similar to 130 million-year-old clade. Analysis of cDNA libraries revealed complex venom transcriptomes. Most notably, three new cardioactive peptide toxin types were discovered (celestoxin, cholecystokinin, and YY peptides). The latter two represent additional examples of convergent use of genes in toxic arsenals, both having previously been documented as components of frog skin defensive chemical secretions. Two other novel venom gland-overexpressed modified versions of other protein frameworks were also recovered from the libraries (epididymal secretory protein and ribonuclease). Lectin, hyaluronidase, and veficolin toxin types were sequenced for the first time from lizard venoms and shown to be homologous to the snake venom forms. In contrast, phylogenetic analyses demonstrated that the lizard natriuretic peptide toxins were recruited independently of the form in snake venoms. The de novo evolution of helokinestatin peptide toxin encoding do-mains within the lizard venom natriuretic gene was revealed to be exclusive to the helodermatid/anguid subclade. New isoforms were sequenced for cysteine-rich secretory protein, kallikrein, and phospholipase A 2 toxins. Venom gland morphological analysis revealed extensive evolutionary tinkering. Anguid glands are characterized by thin capsules and mixed glands, serous at the bottom of the lobule and mucous toward the apex. Twice, independently this arrangement was segregated into specialized serous protein-secreting glands with thick capsules with the mucous lobules now distinct (Heloderma and the Lanthanotus/Varanus clade). The results obtained highlight the importance of utilizing evolution-based search strategies for biodiscovery and emphasize the largely untapped drug design and development potential of lizard venoms.
