Objectif : Déterminer la prévalence de la Sclérose systémique (SSc) chez les patients atteints de cirrhose biliaire primitive (CBP) et décrire les caractéristiques cliniques, pronostiques, immunologiques et capillaroscopiques chez les patients avec et sans SSc. Méthode : Étude descriptive de 100 patients avec CBP naïfs de SSc référés par les gastroentérologues. Un examen physique ainsi qu’un prélèvement sanguin et une capillaroscopie ont été réalisés. Résultats : Parmi les 22 patients diagnostiqués avec SSc, 13 n’avaient pas d’atteinte cutanée. Trente-neuf patients présentaient un phénomène de Raynaud. Dix-neuf étaient anticentromères (ACAs) positifs dont 18 avaient une SSc. Le groupe de CBP avec SSc avait un score Mayo meilleur et une atteinte histologique moins sévère. Une capillaroscopie anormale était retrouvée chez 29/100 patients. Les patients sans ACAs avaient une hypertension portale légèrement plus fréquente 14/81 (17,28%, p= 0,876) et une atteinte histologique hépatique plus sévère (89,5%, p=0,125). Le score Mayo était globalement meilleur dans le groupe des ACAs positifs. La sensibilité et la spécificité du test immunologique sont 95,45% et 93,59% respectivement. La capillaroscopie a une sensibilité de 78% et une spécificité de 94% pour le diagnostic de la SSc dans la population de CBP. Conclusion : La SSc est fréquente dans la CBP, d’où l’intérêt de rechercher systématiquement les signes cliniques subtiles de la SSc, notamment le phénomène de Raynaud, et de demander une sérologie spécifique et une capillaroscopie pour identifier une SSc débutante.

This study examines the physical and chemical composition and the pharmacological effects of brown seaweed FRF 0.8 Lobophora variegata. Fractionation of the crude extract was done with the concentration of 0.8 volumes of acetone, obtaining the FRF 0.8. The physicochemical characterization showed that it was a fucana sulfated. Anti-inflammatory activity was assessed by paw edema model by the high rates of inhibition of the edema and the best results were in the fourth hour after induction (100 ± 1.4% at the dose of 75 mg / kg) and by the strong inhibitory activity of the enzyme myeloperoxidase (91.45% at the dose of 25 mg / kg). The hepataproteção was demonstrated by measurements of enzymatic and metabolic parameters indicative of liver damage, such as bilirubin (reduction in 68.81%, 70.68% and 68.21% for bilirubin total, direct and indirect, respectively at a dose of 75 mg / kg), ALT, AST and γ-GT (decrease of 76.93%, 44.58% and 50% respectively at a dose of 75 mg / kg) by analysis of histological slides of liver tissue, confirming that hepatoprotective effect the polymers of carbohydrates, showing a reduction in tissue damage caused by CCl4 and the inhibition of the enzyme complex of cytochrome P 450 (increasing sleep time in 54.6% and reducing the latency time in 71.43%). The effectiveness of the FRF 0.8 angiogenesis was examined in chorioallantoic membrane (CAM) of fertilized eggs, with the density of capillaries evaluated and scored, showing an effect proangigênico at all concentrations tested FRF (10 mg- 1000 mg). The FRF showed antioxidant activity on free radicals (by inhibiting Superoxide Radical in 55.62 ± 2.10%, Lipid Peroxidation in 100.15 ± 0.01%, Hydroxyl Radical in 41.84 ± 0.001% and 71.47 Peroxide in ± 2.69% at concentration of 0.62 mg / mL). The anticoagulant activity was observed with prolongation of activated partial thromboplastin time (aPTT) at 50 mg (> 240 s), showing that its action occurs in the intrinsic pathway of the coagulation cascade. Thus, our results indicate that these sulfated polysaccharides are an important pharmacological target

The study was conducted to evaluate the liver tissue weight, body fat weight, the fatty somatic index and the liver somatic index of bullfrogs subjected to three photoperiod during the growing phase (initial 30 days) and finishing (60 days). The treatments were 16 h light and 8 h dark, 12 h of light and dark and 8 h light and 16 h dark. The experimental design was completely randomized design with three replications. We used 36 bullfrogs/m2 weighing from 22 to 28 g. The results were submitted to ANOVA and Tukey Test at 5% probability. Differences were found for the weight of body fat, liver tissue weight, liver somatic index and fatty somatic index the bullfrog to the treatments and the study periods and there is greater accumulation of energy to ninety days. The photoperiod did not influence body weight and fat weight of the liver tissue of bullfrog at the end of the fattening period and the difference fatty somatic index and liver somatic index were influenced by body weight of animals.

The aim of the present study was to extract vegetable oil from brown linseed (Linum usitatissimum L.), determine fatty acid levels, the antioxidant capacity of the extracted oil and perform a rapid economic assessment of the SFE process in the manufacture of oil. The experiments were conducted in a test bench extractor capable of operating with carbon dioxide and co-solvents, obeying 23 factorial planning with central point in triplicate, and having process yield as response variable and pressure, temperature and percentage of cosolvent as independent variables. The yield (mass of extracted oil/mass of raw material used) ranged from 2.2% to 28.8%, with the best results obtained at 250 bar and 50ºC, using 5% (v/v) ethanol co-solvent. The influence of the variables on extraction kinetics and on the composition of the linseed oil obtained was investigated. The extraction kinetic curves obtained were based on different mathematical models available in the literature. The Martínez et al. (2003) model and the Simple Single Plate (SSP) model discussed by Gaspar et al. (2003) represented the experimental data with the lowest mean square errors (MSE). A manufacturing cost of US$17.85/kgoil was estimated for the production of linseed oil using TECANALYSIS software and the Rosa and Meireles method (2005). To establish comparisons with SFE, conventional extraction tests were conducted with a Soxhlet device using petroleum ether. These tests obtained mean yields of 35.2% for an extraction time of 5h. All the oil samples were sterilized and characterized in terms of their composition in fatty acids (FA) using gas chromatography. The main fatty acids detected were: palmitic (C16:0), stearic (C18:0), oleic (C18:1), linoleic (C18:2n-6) and α-linolenic (C18:3n-3). The FA contents obtained with Soxhlet dif ered from those obtained with SFE, with higher percentages of saturated and monounsaturated FA with the Soxhlet technique using petroleum ether. With respect to α-linolenic content (main component of linseed oil) in the samples, SFE performed better than Soxhlet extraction, obtaining percentages between 51.18% and 52.71%, whereas with Soxhlet extraction it was 47.84%. The antioxidant activity of the oil was assessed in the β-carotene/linoleic acid system. The percentages of inhibition of the oxidative process reached 22.11% for the SFE oil, but only 6.09% for commercial oil (cold pressing), suggesting that the SFE technique better preserves the phenolic compounds present in the seed, which are likely responsible for the antioxidant nature of the oil. In vitro tests with the sample displaying the best antioxidant response were conducted in rat liver homogenate to investigate the inhibition of spontaneous lipid peroxidation or autooxidation of biological tissue. Linseed oil proved to be more efficient than fish oil (used as standard) in decreasing lipid peroxidation in the liver tissue of Wistar rats, yielding similar results to those obtained with the use of BHT (synthetic antioxidant). Inhibitory capacity may be explained by the presence of phenolic compounds with antioxidant activity in the linseed oil. The results obtained indicate the need for more detailed studies, given the importance of linseed oil as one of the greatest sources of ω3 among vegetable oils

CONTEXTO E OBJETIVO: em crianças, a esteatose hepática pode se relacionar a erros inatos do metabolismo (EIMs) ou à doença hepática gordurosa não-alcoólica (DHGNA). O objetivo deste estudo foi avaliar e caracterizar esteatose de causa indeterminada por meio de análises morfológica e morfométrica em tecido hepático. TIPO DE ESTUDO E LOCAL: Estudo transversal nos Departamentos de Patologia da Faculdade de Ciências Médicas da Universidade Estadual de Campinas (FCM-Unicamp) e Faculdade de Medicina de Botucatu da Universidade Estadual Paulista (FMB-Unesp). MÉTODOS: Foram utilizadas 18 biópsias hepáticas consecutivas obtidas de 16 pacientes com idade variando de 3 meses a 12 anos e 9 meses, inseridas num banco de dados no período do estudo, que foram analisadas por microscopia óptica e eletrônica. Na microscopia eletrônica, foi realizada determinação da densidade mitocondrial e da área superficial média das mitocôndrias nos hepatócitos. Dez pacientes com idade variando de 1 a 14 anos foram usados como grupo controle. RESULTADOS: Foi detectada esteatose pura, não acompanhada por fibrose ou outra alteração histológica. Foi verificado que, na predominância de esteatose microvesicular, houve aumento significativo da área mitocondrial média. CONCLUSÃO: A esteatose microvesicular pode estar relacionada à hepatopatia mitocondrial primária, principalmente devido à redução na β-oxidação ou parcial estagnação da fosforilação oxidativa. Por essas razões, esta forma de esteatose (que não pode ser chamada de pura) possivelmente represente uma fase inicial no amplo espectro da DHGNA. Chamamos a atenção para casos de esteatose no grupo pediátrico com predomínio da forma microvesicular, uma vez que pode haver associação com desordens mitocondriais.

The tegus increase in body mass after hatching until early autumn, when the energy intake becomes gradually reduced. Resting rates of oxygen consumption in winter drop to 20% of the values in the active season (Vo(2)=0.0636 ml g(-1) h(-1)) and are nearly temperature insensitive over the range of 17-25degreesC (Q(10)=1.55). During dormancy, plasma glucose levels are 60% lower than those in active animals, while total protein, total lipids and beta-hydroxybutyrate are elevated by 24%, 43% and 113%, respectively. In addition, a significant depletion of liver carbohydrate (50%) and of fat deposited in the visceral fat bodies (24%) and in the tail (25%) and a slight loss of skeletal muscle protein (14%) were measured halfway through the inactive period. Otherwise, glycogen content is increased 4-fold in the brain and 2.3-fold in the heart of dormant lizards, declining by the onset of arousal. During early arousal, the young tegus are still anorexic, although Vo(2) is significantly greater than winter rates. The fat deposits analysed are further reduced (62% and 45%, respectively) and there is a large decrease in tail muscle protein (50%) together with a significant increase in glycogen (2-3-fold) and an increase in plasma glucose (40%), which suggests a role for gluconeogenesis as a supplementary energy source in arousing animals. No change is detectable in citrate synthase activity, but beta-hydroxyacyl CoA dehydrogenase activities are strongly affected by season, reaching a Mold and 5-fold increase in the liver tissue of winter and arousing animals, respectively, and becoming reduced by half in skeletal muscle and heart of winter animals compared with late fall or spring active individuals. From hatching to late autumn, the increase of the fat body mass relatively to body mass is disproportionate (b=1.44), and the mass exponent changes significantly to close to 1.0 during the fasting period. The concomitant shift in the Vo(2) mass exponent in early autumn (b=0.75) to values significantly greater than 1.0 in late autumn and during winter dormancy indicates an allometric effect on the degree of metabolic depression related to the size of the fat stores and suggests greater energy conservation in the smaller young.

Um experimento foi conduzido com o objetivo de avaliar os desempenhos reprodutivo e zootécnico e a deposição de lipídios no tecido hepático de fêmeas de tilápia-do-nilo alimentadas com rações contendo diferentes níveis de energia digestível, obtidos pela inclusão de óleo de soja. Foram utilizados 100 reprodutores e 300 reprodutoras, distribuídos em delineamento inteiramente casualizado com cinco tratamentos e quatro repetições. Os reprodutores, machos e fêmeas, foram alimentados com rações contendo, por quilograma, 35% de proteína bruta e 2.700, 2.950, 3.200, 3.450 ou 3.700 kcal de energia digestível. Os animais foram mantidos em hapas por 93 dias. O desempenho reprodutivo e zootécnico não foi influenciado pelos níveis energéticos das rações, o que pode estar relacionado ao curto período de tempo de alimentação com as dietas experimentais. Contudo, o aumento nos níveis de energia teve efeito linear nas deposições lipídicas nos hepatócitos. Desta forma, os níveis crescentes de energia digestível nas rações fornecidas às fêmeas de tilápia-do-nilo, obtidos pela inclusão de óleo de soja, apenas aumentam o tempo de sobrevivência das larvas ao jejum e a deposição de lipídios nos hepatócitos das fêmeas.

O experimento foi conduzido com o objetivo de avaliar o desempenho reprodutivo e zootécnico e a deposição de lipídios no tecido hepático de machos de tilápia-do-nilo alimentados com rações contendo diferentes níveis de energia digestível, obtidos com a inclusão de óleo de soja. Foram utilizados 400 reprodutores (300 fêmeas e 100 machos) distribuídos em delineamento inteiramente casualizado, composto de cinco níveis de energia digestível (2.700, 2.950, 3.200, 3.450 e 3.700 kcal.kg de ração-1) e quatro repetições. Os reprodutores foram alimentados com rações contendo 35% de proteína bruta e submetidos ao manejo reprodutivo em hapas por 101 dias. O melhor resultado de concentração espermática e percentual de espermatozoides normais foram obtidos para reprodutores alimentados com rações contendo 3.465,56 e 3.443,43 kcal. kg de ração-1, que produziram 7,98 × 10(9) espermatozoides.mL de sêmen-1 e 38,98% de espermatozoides normais, respectivamente. A produção de sêmen, o pH seminal, o índice de sobrevivência espermática e o tempo de ativação espermática não foram afetados pelos níveis energéticos das rações. Os níveis de energia das rações não influenciaram o desempenho zootécnico dos peixes, mas promoveram aumento linear na deposição de lipídios nos hepatócitos e afetaram a qualidade seminal, estimulando a produção de espermatozoides e a melhora dos índices de normalidade da morfologia espermática em níveis energéticos próximos a 3.450 kcal. kg de ração-1.

Aim. To investigate the effects of physical training associated to dexamethasone administration in carbohydrate metabolism and adrenocorticotrophic hormone (ACTH) release. Materials and methods. Young Wistar rats were divided into four groups: sedentary control (CS), sedentary dexanzethasone (DxS), trained control (CT) and trained dexamethasone (DxT). The rats were submitted to swimming training associate to administration of dexamethasone for ten weekends. Before sacrifice the rats received Subcutaneous insulin to calculate the maximum decreased in blood glucose. Venous blood was sampled obtained at the end experiment period to determine glucose, insulin, free fatty acids (FFA) and ACTH. Gastrocnemius and liver tissue samples were used to determination glycogen, and adipose epididimal tissue was used to measured the weight. Results. Dexamethasone administration provoke insulin resistance and the physical training reverted this aspect. Training promoted increase in muscle and liver glycogen store and a high utilization of FFA. Moreover the dexamethasone provoke decreased of ACTH release in response to acute exercise, showing marked differences in the functioning of the hypothalamy pituitary-adrenal (HPA) axis between groups of rats. Conclusions. a) Low-dose of dexamethasone promote several side effects in metabolism intermediary and chronic exposure to steroid was associated with insulin resistance; b) the regular swimming exercise promoted increased insulin sensitiviry Therefore. exercise can override the dexametasone negative feedback of the HPA axis activation in rats.

This research was done to compare the effects of different zinc sources and doses in the Santa Ines sheep diet. Forty lambs at weaning, with 18,4kg BW were randomly allotted and fed 10 treatments: 1- base diet without zinc supplementation; 2- base diet + 200mg Zn/kg of DM as zinc oxide; 3- base diet + 400mg Zn/kg of DM as zinc oxide; 4- base diet + 600mg Zn/kg of DM as zinc oxide; 5- base diet + 200mg Zn/kg of DM as amino acid zinc; 6- base diet + 400mg Zn/kg of DM as amino acid zinc; 7- base diet + 600mg Zn/kg of DM as amino acid zinc; 8- base diet + 200mg Zn/kg of DM as proteinato zinc; 9- base diet + 400mg Zn/kg of DM as proteinato zinc; 10- base diet + 600mg Zn/kg of DM as proteinato zinc. The animals were weighed and sampled for blood zinc analysis, phosphatase alkaline analysis and immunoglobulins G and M analysis. At the end of the experiment liver samples were collected to study the zinc hepatic levels. There was no difference in phosphatase alkaline levels, hepatic zinc levels and weight gain (P>0,05) but differences (P<0,05) in plasmatic zinc levels and in IgG and IgM levels were observed. Based on liver tissue uptake, estimates of the zinc bioavailability, through the regression equations showed that the organic and inorganic sources of zinc did not differ.

Some recent articles have reported that mesenchymal stem cells (MSCs) can be induced to express hepatocyte markers by transplanting them into animal models of liver damage, or by in vitro culture with growth factors and cytokines. In this study, the aim is to evaluate the behavior of MSCs subjected to induction of hepatocyte differentiation. The MSCs were isolated from the bone marrow of 4 normal donors, characterized and subjected to both in vitro and in vivo induction of hepatocyte differentiation. The in vitro induced cells showed morphological changes, acquiring hepatocyte-like features. However, the immunophenotype of these cells was not modified. The induced cells exhibited no increase in albumin, cytokeratin 18 or cytokeratin 19 transcripts, when analyzed by real-time RT-PCR. The expression of albumin, cytokeratin 18 and alpha fetoprotein was also unchanged, according to immunofluorescence tests. In vivo, the MSC demonstrated a potential to migrate to damaged liver tissue in immunodeficient mice. Taken together, the results suggest that bone marrow MSCs are incapable of in vitro differentiation into hepatocytes by the approach used here, but are capable of homing to damaged hepatic tissue in vivo, suggesting a role for them in the repair of the liver. This contribution to tissue repair could be associated with a paracrine effect exerted by these cells.

The aims of this investigation was to evaluate the effect of hepatoprotective treatments with a compound prepared by the association of N-Acetyl DL-Methionine (5%) + Choline chloride (2%) + Caffeine (1%) + Thiamine hydrochloride (1%) + Nicotinamida (0,5%)+ Pyridoxine hydrochloride (0.04%), administered through intramuscular (IM) route, at doses of 0.2, 0.6 and 1.0 mL/kg of BW, through the study of leukocytes responses in rats submitted to acute intoxication with CCl4. 147 females were randomized into 21 groups, performing five different treatments, which were evaluated seven animals in four periods: two, four, six and eight days after CCl4-induced intoxication. In this study, it was observed absolute eosinophilia and monocytosis in animals untreated and treated with the lowest dose of 0.2 mL. These responses were significantly better in animals treated with 0.6 and 1.0 mL/ kg BW. The untreated animals showed thrombocytopenia, when compared to treated animals. Absolute neutropenia and lymphocytosis was observed in all rats intoxicated with CCl4, there is no difference among treatments. The analysis of white blood cells demonstrated that the hepatoprotective treatments favored the leukocyte response, by act beneficially on the population of these cells, supporting the hypothesis that these events may reduce the deleterious effects in liver tissue after intoxication by CCl4.

Alterations in liver functions are common among diabetic patients, and many symptoms in the liver have been reported, including changes in glycogen stores and in the amount of collagen fibers. The practice of physical training and its morphological effects in this organ, however, are scarcely studied. In order to observe the morphological effects of alloxan-induced diabetes and the alterations arising from the practice of long-term chronic physical training in the liver, samples were collected and processed, and then analyzed by means of the histochemical techniques Periodic Acid-Schiff and Picrosirius-Hematoxylin, and ultrastructural cytochemical test of Afzelius. Through evaluation of the tissue, it was observed a drastic reduction in hepatic glycogen stores of sedentary diabetics, recovered in trained diabetic rats. Furthermore, it was detected a decrease in the content of perisinusoidal collagen fibers in the diabetic liver, also recovered due to the development of a training protocol. On ultrastructural level, cytochemical analysis confirmed the loss of glycogen and the recovery obtained by training. In conclusion, the practice of a long-term chronic physical training protocol may be considered an important assistant in the treatment of diabetes, mitigating the occurrence of possible damages to liver tissue. © 2011 Elsevier Ltd.
