Um experimento em casa de vegetação foi conduzido entre novembro de 2005 e abril de 2006, na UNESP, campus de Jaboticabal, SP, objetivando estudar a produção de massa seca, a distribuição e o acúmulo de macronutrientes por Ipomoea nil, uma importante infestante de culturas anuais e perenes no Brasil. As plantas foram cultivadas em vasos de 7 litros com substrato de areia e irrigadas diariamente com solução nutritiva de Hoagland & Arnon. O delineamento experimental foi inteiramente casualizado, com quatro repetições. Os tratamentos corresponderam às épocas de avaliação, em intervalos de 14 dias, iniciandose 21 dias após a emergência (DAE). em cada avaliação, as plantas de quatro vasos foram analisadas quanto ao acúmulo de massa seca e à nutrição mineral. I. nil apresentou pequeno acúmulo de massa seca e de macronutrientes no início da fase experimental. Esses acúmulos se intensificaram após 77 DAE, atingindo o máximo valor teórico aos 123, 122, 124, 121, 119, 121 e 125 DAE, para massa seca, N, P, K, Ca, Mg e S, respectivamente. Folhas e caules foram os principais órgãos responsáveis pelo acúmulo de massa seca por plantas de I. nil. K e N foram os macronutrientes extraídos em maior quantidade por essa espécie.

As plantas foram cultivadas em vasos de 7 L com substrato de areia, que foram irrigados diariamente com solução nutritiva de Hoagland & Arnon. Os tratamentos corresponderam às épocas de avaliação, em intervalos de 14 dias, iniciando-se 21 dias após a emergência (DAE). em cada avaliação, as plantas de quatro vasos foram analisadas quanto à produção de massa seca e ao conteúdo de macronutrientes. I. hederifolia apresentou pequeno acúmulo de massa seca e de macronutrientes no início da fase experimental, o qual foi intensificado após 63 DAE. As folhas foram as principais estruturas acumuladoras de massa seca na primeira metade do ciclo de I. hederifolia, enquanto os caules o foram na segunda metade. N e K foram os macronutrientes mais acumulados em plantas dessa espécie. O período de acúmulo máximo de massa seca e macronutrientes por I. hederifolia ocorreu entre 133 e 146 DAE.

I. quamoclit apresentou pequeno acúmulo de massa seca e de macronutrientes no início da fase experimental. Esses acúmulos intensificaram-se após 77 DAE, atingindo o máximo valor teórico aos 146, 143, 140, 149, 142, 153 e 124 DAE, para massa seca, N, P, K, Ca, Mg e S, respectivamente. K e N foram os macronutrientes acumulados em maior quantidade por plantas de I. quamoclit.

Com o objetivo de avaliar a eficácia do diclosulam (doses de 21,8 e 25,2 g i.a. ha-1) associado à palha de sorgo (Sorghum bicolor) cultivar AG 1018 no controle de Ipomoea grandifolia e Sida rhombifolia, realizou-se um experimento em casa de vegetação, onde os tratamentos foram constituídos de diferentes posicionamentos do herbicida, aplicado sobre e sob a palha em diferentes condições de umidade. Após o preenchimento dos vasos com solo, as plantas daninhas (I. grandifolia e S. rhombifolia) foram semeadas superficialmente; em seguida, nos tratamentos com palha de sorgo, os vasos foram cobertos com 6 t ha-1. O delineamento experimental utilizado foi o inteiramente casualizado, com quatro repetições. Foram realizadas avaliações visuais de controle (0 a 100%) aos 7, 14, 21, 28 e 35 dias após a aplicação (DAA) e contagem de plantas e biomassa seca, ao final do estudo. Observaram-se elevados níveis de controle do herbicida diclosulam já para a menor dose utilizada (21,8 g i.a. ha-1) quando este foi aplicado diretamente sobre o solo, na ausência e presença de palha, ou sobre a palha e seguido da ocorrência de chuvas de 30 mm. A maior dose (25,2 g i.a. ha-1) promoveu elevados níveis de eficácia de controle das plantas daninhas nos diferentes posicionamentos do produto, exceto para a aplicação em palha seca e úmida sem ocorrência de chuvas.

Two experiments were carried out under greenhouse conditions to study the accumulation and distribution of dry mass and macronutrients in maize and Ipomoea hederifolia. Plants of both species had grown, separately, in pots with sand substrate and irrigation with nutrient solution. Treatments were represented by the times of evaluation, realized in intervals of 14 days, starting at 21 days after emergence (DAE). A maize plant showed slight growth up to 30 DAE, when dry mass allocation was higher in roots and leaves (80%); while an I. hederifolia plant, up to 50 DAE, when the allocation of dry mass was higher in offshoots and leaves (79). Dry mass accumulation was almost five times greater in maize (134 g per plant) than in I. hederifolia (29 g per plant). The average values of N and K contents were greater in I. hederifolia. Maximum accumulations of macronutrients by maize were 1,431; 474; 1,832; 594; 340, and 143 mg per plant, while by I. hederifolia, 727; 52; 810; 350; 148, and 65 mg per plant, for N, P, K, Ca, Mg, and S, respectively. Mean accumulation rate of dry mass and macronutrients by maize plants was crescent up to 87 DAE, reaching the maximum value at 103 DAE; while being crescent up to 121 DAE by I. hederifolia plants, reaching the maximum value at 138 DAE. Thus, beyond the interference on harvesting process, a population of I. hederifolia also can compete with maize crop for nutrients.

Ipomoea carnea is a toxic plant in Brazil and other tropical countries that often poisons livestock; its primary toxin is swainsonine. Some substances that interact with the endocrine system have been called endocrine disruptors (EDs). Considering swainsonine's mode of action, it is feasible to hypothesize that this compound could act as an ED. Bisphenol A (BPA), an estrogen-mimic, is considered a classical ED in rodents. This study aimed to evaluate the possible ED actions of I. cameo and BPA in male goats. Seventeen adult male goats were divided into three homogeneous groups: control (C, n = 5); IC (n = 6, received 5.0 g/kg body weight of freshly harvested I. cornea per day), and BPA (n = 6, received 25.0 mg/kg body weight of BPA per day). The experimental period was 120 days. During the experiment, blood samples were collected at 0, 30, 60, 90 and 120 days for biochemical and hormonal evaluations. On the same days, semen samples were collected for andrological evaluation, and scrotal circumference and testicular consistency were determined. The males were castrated on day 121, and fragments of testicle and epididymis were collected for histopathological evaluation. A decrease in serum T3 and T4 was observed in the IC group as well as an increase in the number of sperm with morphological alterations. In the BPA group, reduced serum 14 and a decreased percentage of sperm with plasma membrane integrates were observed. A histopathological examination revealed the vacuolar degeneration of Sertoli cells and areas of laminar patterns of calcium deposits in the IC group and vacuolar degeneration in the rete testis in the BPA group. These results indicate that both I. cameo and BPA are potential EDs in goats. This study emphasized the susceptibility of livestock to ED actions. We also demonstrated the action of I. cameo acting as EDs in the endocrine and reproductive systems. (C) 2014 Elsevier B.V. All rights reserved.

Background: Ipomoea carnea (I. carnea) is a poisonous plant found in Brazil and other tropical countries that often poison livestock. The plant contains the alkaloids calystegines and mainly swainsonine, which inhibit cellular enzymes and cause systematic cell death. The objective of this study was to evaluate the perinatal effects of I. carnea in goats. Methods: Forty-seven pregnant goats were randomly allocated into 5 treatment groups and given the following doses (g/kg BW) of I. carnea: 0 (IC0), 1.0 (IC1), 3.0 (IC3), 5.0 (IC5) and 7.5 (IC7). The treatment animals were given fresh I. carnea from day 27 of gestation to parturition. Weight gains and serum biochemistry were evaluated. Fetuses were evaluated using ultrasonographic measurements. Results: Goats from the IC7 group showed clinical signs of poisoning. Ultrasound examination revealed that I. carnea feeding in all treatment groups reduced fetal movement compared to the controls. There was an increase in the total number of birth defects (retrognathia and arthrogyposis) in the IC7 and IC5 groups compared to the controls. Conclusion: The results show that I. carnea has teratogenic potential in goats. In addition, ultrasounds were useful in evaluating fetotoxicity and teratogenicity. Mobile element dynamics in seven alleles of the chalcone synthase D locus (CHS-D) of the common morning glory (Ipomoea purpurea) are analyzed in the context of synonymous nucleotide sequence distances for CHS-D exons. By using a nucleotide sequence of CHS-D from the sister species Ipomoea nil (Japanese morning glory [Johzuka-Hisatomi, Y., Hoshino, A., Mori, T., Habu, Y. & Iida, S. (1999) Genes Genet. Syst. 74, 141–147], it is also possible to determine the relative frequency of insertion and loss of elements within the CHS-D locus between these two species. At least four different types of transposable elements exist upstream of the coding region, or within the single intron of the CHS-D locus in I. purpurea. There are three distinct families of miniature inverted-repeat transposable elements (MITES), and some recent transpositions of Activator/Dissociation (Ac/Ds)-like elements (Tip100), of some short interspersed repetitive elements (SINEs), and of an insertion sequence (InsIpCHSD) found in the neighborhood of this locus. The data provide no compelling evidence of the transposition of the mites since the separation of I. nil and I. purpurea roughly 8 million years ago. Finally, it is shown that the number and frequency of mobile elements are highly heterogeneous among different duplicate CHS loci, suggesting that the dynamics observed at CHS-D are locus-specific.

The evolution of the chalcone synthase [CHS; malonyl-CoA:4-coumaroyl-CoA malonyltransferase (cyclizing), EC 2.3.1.74] multigene family in the genus Ipomoea is explored. Thirteen CHS genes from seven Ipomoea species (family Convolvulaceae) were sequenced--three from genomic clones and the remainder from PCR amplification with primers designed from the 5' flanking region and the end of the 3' coding region of Ipomoea purpurea Roth. Analysis of the data indicates a duplication of CHS that predates the divergence of the Ipomoea species in this study. The Ipomoea CHS genes are among the most rapidly evolving of the CHS genes sequenced to date. The CHS genes in this study are most closely related to the Petunia CHS-B gene, which is also rapidly evolving and highly divergent from the rest of the Petunia CHS sequences.

Common bean (Phaseolus vulgaris) is present in the daily diet of various countries and, as for other legumes, has been investigated for its nutraceutical potential. Thus, 16 genotypes from different gene pools, representing seven types of seed coats and different responses to pathogens and pests, were selected to verify their isoflavone contents. The isoflavonoids daidzein and genistein and the flavonols kaempferol, myricetin, and quercetin were found. Grains of the black type showed the highest concentrations of isoflavonoids and were the only ones to exhibit daidzein. IAC Formoso, with high protein content and source of resistance to anthracnose, showed the greatest concentration of genistein, representing around 11% of the content present in soybean, as well as high levels of kaempferol. Arc 1, Raz 55, and IAC Una genotypes showed high content of coumestrol. The results suggest the use of IAC Formoso to increase the nutraceutical characteristics in common bean.

Genipap fruits, native to the Amazon region, were classified in relation to their stage of ripeness according to firmness and peel color. The influence of the part of the genipap fruit and ripeness stage on the iridoid and phenolic compound profiles was evaluated by HPLC-DAD-MS(n), and a total of 17 compounds were identified. Geniposide was the major compound in both parts of the unripe genipap fruits, representing >70% of the total iridoids, whereas 5-caffeoylquinic acid was the major phenolic compound. In ripe fruits, genipin gentiobioside was the major compound in the endocarp (38%) and no phenolic compounds were detected. During ripening, the total iridoid content decreased by >90%, which could explain the absence of blue pigment formation in the ripe fruits after their injury. This is the first time that the phenolic compound composition and iridoid contents of genipap fruits have been reported in the literature.

The androgynophore column, a distinctive floral feature in passion flowers, is strongly crooked or bent in many Passiflora species pollinated by bats. This is a floral feature that facilitates the adaptation to bat pollination. Crooking or bending of plant organs are generally caused by environmental stimulus (e.g. mechanical barriers) and might involve the differential distribution of auxin. Our aim was to study the role of the perianth organs and the effect of auxin in bending of the androgynophore of the bat-pollinated species Passiflora mucronata. Morpho-anatomical characterisation of the androgynophore, including measurements of curvature angles and cell sizes both at the dorsal (convex) and ventral (concave) sides of the androgynophore, was performed on control flowers, flowers from which perianth organs were partially removed and flowers treated either with auxin (2,4-dichlorophenoxyacetic acid; 2,4-D) or with an inhibitor of auxin polar transport (naphthylphthalamic acid; NPA). Asymmetric growth of the androgynophore column, leading to bending, occurs at a late stage of flower development. Removing the physical constraint exerted by perianth organs or treatment with NPA significantly reduced androgynophore bending. Additionally, the androgynophores of plants treated with 2,4-D were more curved when compared to controls. There was a larger cellular expansion at the dorsal side of the androgynophores of plants treated with 2,4-D and in both sides of the androgynophores of plants treated with NPA. This study suggests that the physical constraint exerted by perianth and auxin redistribution promotes androgynophore bending in P. mucronata and might be related to the evolution of chiropterophily in the genus Passiflora.

Extracts from malagueta pepper (Capsicum frutescens L.) were obtained using supercritical fluid extraction (SFE) assisted by ultrasound, with carbon dioxide as solvent at 15MPa and 40°C. The SFE global yield increased up to 77% when ultrasound waves were applied, and the best condition of ultrasound-assisted extraction was ultrasound power of 360W applied during 60min. Four capsaicinoids were identified in the extracts and quantified by high performance liquid chromatography. The use of ultrasonic waves did not influence significantly the capsaicinoid profiles and the phenolic content of the extracts. However, ultrasound has enhanced the SFE rate. A model based on the broken and intact cell concept was adequate to represent the extraction kinetics and estimate the mass transfer coefficients, which were increased with ultrasound. Images obtained by field emission scanning electron microscopy showed that the action of ultrasonic waves did not cause cracks on the cell wall surface. On the other hand, ultrasound promoted disturbances in the vegetable matrix, leading to the release of extractable material on the solid surface. The effects of ultrasound were more significant on SFE from larger solid particles.

To examine the influence of l-arginine supplementation in combination with physical training on mitochondrial biomarkers from gastrocnemius muscle and its relationship with physical performance. Male Wistar rats were divided into four groups: control sedentary (SD), sedentary supplemented with l-arginine (SDLA), trained (TR) and trained supplemented with l-arginine (TRLA). Supplementation of l-arginine was administered by gavage (62.5mg/ml/day/rat). Physical training consisted of 60min/day, 5days/week, 0% grade, speed of 1.2km/h. The study lasted 8weeks. Skeletal muscle mitochondrial enriched fraction as well as cytoplasmic fractions were obtained for Western blotting and biochemical analyses. Protein expressions of transcriptor coactivator (PGC-1α), transcriptor factors (mtTFA), ATP synthase subunit c, cytochrome oxidase (COXIV), constitutive nitric oxide synthases (eNOS and nNOS), Cu/Zn-superoxide dismutase (SOD) and manganese-SOD (Mn-SOD) were evaluated. We also assessed in plasma: lipid profile, glycemia and malondialdehyde (MDA) levels. The nitrite/nitrate (NOx(-)) levels were measured in both plasma and cytosol fraction of the gastrocnemius muscle. 8-week l-arginine supplementation associated with physical training was effective in promoting greater tolerance to exercise that was accompanied by up-regulation of the protein expressions of mtTFA, PGC-1α, ATP synthase subunit c, COXIV, Cu/Zn-SOD and Mn-SOD. The upstream pathway was associated with improvement of NO bioavailability, but not in NO production since no changes in nNOS or eNOS protein expressions were observed. This combination would be an alternative approach for preventing cardiometabolic diseases given that in overt diseases a profound impairment in the physical performance of the patients is observed.

Malaria is still one of the major diseases in the world, causing physical and economic problems in tropical regions. Artemisinin (Qinghaosu), a natural compound identified in Artemisia annua L. , is an effective drug mainly against cerebral malaria. The action of this drug is immediate and parasitaemia in the treatment of drug-resistant malaria is rapidily reduced, justifying the industrial production of artemisinin. This article focuses on the industrial production of this potent antimalarial drug, including strategies for enhancing yield using inexpensive and easy steps.
