BACKGROUND: Highway maintenance workers are constantly and simultaneously exposed to traffic-related particle and noise emissions, and both have been linked to increased cardiovascular morbidity and mortality in population-based epidemiology studies. OBJECTIVES: We aimed to investigate short-term health effects related to particle and noise exposure. METHODS: We monitored 18 maintenance workers, during as many as five 24-hour periods from a total of 50 observation days. We measured their exposure to fine particulate matter (PM2.5), ultrafine particles, noise, and the cardiopulmonary health endpoints: blood pressure, pro-inflammatory and pro-thrombotic markers in the blood, lung function and fractional exhaled nitric oxide (FeNO) measured approximately 15 hours post-work. Heart rate variability was assessed during a sleep period approximately 10 hours post-work. RESULTS: PM2.5 exposure was significantly associated with C-reactive protein and serum amyloid A, and negatively associated with tumor necrosis factor α. None of the particle metrics were significantly associated with von Willebrand factor or tissue factor expression. PM2.5 and work noise were associated with markers of increased heart rate variability, and with increased HF and LF power. Systolic and diastolic blood pressure on the following morning were significantly associated with noise exposure after work, and non-significantly associated with PM2.5. We observed no significant associations between any of the exposures and lung function or FeNO. CONCLUSIONS: Our findings suggest that exposure to particles and noise during highway maintenance work might pose a cardiovascular health risk. Actions to reduce these exposures could lead to better health for this population of workers.

BACKGROUND: Mitogen activated protein kinases (MAPK) play an essential role in integrating extra-cellular signals and intra-cellular cues to allow cells to grow, adapt to stresses, or undergo apoptosis. Budding yeast serves as a powerful system to understand the fundamental regulatory mechanisms that allow these pathways to combine multiple signals and deliver an appropriate response. To fully comprehend the variability and dynamics of these signaling cascades, dynamic and quantitative single cell measurements are required. Microscopy is an ideal technique to obtain these data; however, novel assays have to be developed to measure the activity of these cascades. RESULTS: We have generated fluorescent biosensors that allow the real-time measurement of kinase activity at the single cell level. Here, synthetic MAPK substrates were engineered to undergo nuclear-to-cytoplasmic relocation upon phosphorylation of a nuclear localization sequence. Combination of fluorescence microscopy and automated image analysis allows the quantification of the dynamics of kinase activity in hundreds of single cells. A large heterogeneity in the dynamics of MAPK activity between individual cells was measured. The variability in the mating pathway can be accounted for by differences in cell cycle stage, while, in the cell wall integrity pathway, the response to cell wall stress is independent of cell cycle stage. CONCLUSIONS: These synthetic kinase activity relocation sensors allow the quantification of kinase activity in live single cells. The modularity of the architecture of these reporters will allow their application in many other signaling cascades. These measurements will allow to uncover new dynamic behaviour that previously could not be observed in population level measurements.

Cette thèse explore la relation entre les littératures autochtones et multiculturelles du Canada. Même si les critiques littéraires examinent les littératures dites mineures de plus en plus, ces dernières sont rarement étudiées sans la présence médiatrice de la littérature canadienne considérée comme étant dominante. Afin de produire une telle analyse, cette thèse mobilise le concept d’hybridité en tant que catégorie d’analyse de texte qui, en plus de son histoire raciale et coloniale, décrit convenablement les formes d’expérimentations stylistiques que les écrivains autochtones et multiculturels emploient afin de représenter et questionner leur marginalisation. Ne voulant pas reproduire les interprétations fétichistes qui réduisent les littératures autochtones et multiculturelles à leurs représentations de concepts d’altérité, j’examine ces textes dans leurs relations avec différents discours et débats ayant marqué les études littéraires canadiennes, notamment, le long poème canadien, l’écriture des prairies canadiennes, la littérature urbaine, le multiculturalisme, et les premières nations. Ma méthode d’analyse repose sur la façon dont chaque texte étudié alimente ces catégories d’analyse littéraire tout en les modifiant radicalement. De plus, je développe un cadre conceptuel et théorique permettant l’étude de la relation entre les textes autochtones et multiculturels sans toutefois confondre ou réduire les contextes d’où proviennent ces littératures. Ma thèse et ma méthode d’analyse se concrétise par l’interprétation des textes écrits par Armand Garnet Ruffo, Suzette Mayr, Rawi Hage, et Jeannette Armstrong. Le chapitre d’introduction détaille la façon dont la relation entre les textes autochtones et multiculturels a été appréhendée jusqu’à présent. J’y élabore mon cadre théorique qui joint et réinterprète de manière critique diverses théories, dont celle du postcolonialisme, de l’hybridité, et de la mondialisation, et la façon dont ces théories se rapportent aux études littéraires canadiennes. Dans mon deuxième chapitre, j’analyse le long poème d’Armand Garnet Ruffo, Grey Owl: The Mystery of Archie Belaney, en m’attardant particulièrement aux stratégies d’expérimentations stylistiques et génériques que Ruffo développe afin de rendre le genre du long poème canadien autochtone et de questionner l’identité de Grey Owl. Mon troisième chapitre examine Venous Hum, un roman de Suzette Mayr. Ce texte remet en question la tradition de « prairie writing », le multiculturalisme canadien, et le conservatisme albertain à travers son style expérimental, son usage des métaphores et du réalisme magique. Mon quatrième chapitre interprète le roman montréalais Cockroach, de Rawi Hage, en examinant la façon dont ses unités locales, nationales, et globales rencontrent le colonialisme et contestent les discours nationaux une fois que sa critique de la mondialisation se trouve réarticulée dans une approbation des discours d’interventions humanitaires de l’occident. Mon dernier chapitre explore le roman de Jeannette Armstrong, Whispering in Shadows, afin de démontrer les limites de ma méthode d’analyse. Puisque l’hybridité sous-entend inévitablement la notion d’assimilation, son application dans le contexte de l’œuvre d’Armstrong s’avèrerait réductrice. Pour cette raison, ce chapitre utilise des concepts autochtones définis par Armstrong afin de développer une méthode de lecture non-hégémonique. Ma thèse examine donc la façon dont chaque texte déploie le concept d’hybridité pour à la fois contester et enrichir les discours critiques qui tentent de contenir ces textes. Elle contribue aux études postcoloniales de la littérature canadienne en élargissant leur champ habituel pour inclure les complexités des théories de la mondialisation, et en examinant quelles stratégies littéraires les textes autochtones et multiculturels partagent, mais mobilisent à des fins différentes.

In analysing the release of agricultural land to urban development, the urban fringe literature has not focused on whether farmers are able to relocate from the urban fringe to remoter rural areas. Through interviews with representatives from the poultry industry in two Australian states, this paper identifies that poultry farm relocation strategies are constrained by off-farm economic relations, the land-use planning system and financial considerations. Closely aligned to these constraints on relocation is the on-going process of poultry farm intensification, which is seen as presenting rising problems for land-use management around expanding metropolitan centres in Australia. Of particular concern is the potential for amenity complaints and associated land-use conflicts, which have not been comprehensively investigated. Recognising that existing environmental and land-use planning controls are ineffective in producing amicable solutions when conflict involving poultry farming is at its most intense, the paper calls for improvements to the regulatory system, including greater consideration for how the process of relocation can be encouraged. Here, we identify the Arabidopsis thaliana ortholog of the mammalian DEAD box helicase, eIF4A-III, the putative anchor protein of exon junction complex (EJC) on mRNA. Arabidopsis eIF4A-III interacts with an ortholog of the core EJC component, ALY/Ref, and colocalizes with other EJC components, such as Mago, Y14, and RNPS1, suggesting a similar function in EJC assembly to animal eIF4A-III. A green fluorescent protein (GFP)-eIF4A-III fusion protein showed localization to several subnuclear domains: to the nucleoplasm during normal growth and to the nucleolus and splicing speckles in response to hypoxia. Treatment with the respiratory inhibitor sodium azide produced an identical response to the hypoxia stress. Treatment with the proteasome inhibitor MG132 led to accumulation of GFP-eIF4A-III mainly in the nucleolus, suggesting that transition of eIF4A-III between subnuclear domains and/or accumulation in nuclear speckles is controlled by proteolysis-labile factors. As revealed by fluorescence recovery after photobleaching analysis, the nucleoplasmic fraction was highly mobile, while the speckles were the least mobile fractions, and the nucleolar fraction had an intermediate mobility. Sequestration of eIF4A-III into nuclear pools with different mobility is likely to reflect the transcriptional and mRNA processing state of the cell.

This paper is an engineer's appreciation of environmental assessment with particular reference to highway development. While scheme-related Environmental Assessment for individual development may identify particular potential impacts, and may avoid or minimise some of the problems, in many cases it may be too late to take such actions. Ideally, Environmental Assessment should commence at the Strategic Level to cover policies, plan and programmes, and the scheme-related Environmental Assessments for individual projects should supplement those in the framework of Strategic Level. The utimate target is to assess the policy for their contribution to effecting sustainable development. Whole Life Environmental Impacts should be considered. These are the full impact consideration from planning, design and choice of materials, construction, operation and finally decommission. Most of the Environmental Assessments have not included the Whole Life Environmental Impacts. There is only limited monitoring in the operation stage after the construction of the scheme is complete, therefore, subsequent Environmental Assessments cannot benefit from the feedback of the scheme. No development should cost the Earth, hence Environmental Assessments have to be carried out thoroughly to serve as one of the instruments to meet the need of sustainable development.

Older adult computer users often lose track of the mouse cursor and so resort to methods such as mouse shaking or searching the screen to find the cursor again. Hence, this paper describes how a standard optical mouse was modified to include a touch sensor, activated by releasing and touching the mouse, which automatically centers the mouse cursor to the screen, potentially making it easier to find a 'lost' cursor. Six older adult computer users and six younger computer users were asked to compare the touch sensitive mouse with cursor centering with two alternative techniques for locating the mouse cursor: manually shaking the mouse and using the Windows sonar facility. The time taken to click on a target following a distractor task was recorded, and results show that centering the mouse was the fastest to use, with a 35% improvement over shaking the mouse. Five out of six older participants ranked the touch sensitive mouse with cursor centering as the easiest to use.

The authors simulated the effects of Amazonian mesoscale deforestation in the boundary layer and in rainfall with the Brazilian Regional Atmospheric Modeling System (BRAMS) model. They found that both the area and shape (with respect to wind incidence) of deforestation and the soil moisture status contributed to the state of the atmosphere during the time scale of several weeks, with distinguishable patterns of temperature, humidity, and rainfall. Deforestation resulted in the development of a three-dimensional thermal cell, the so-called deforestation breeze, slightly shifted downwind to large-scale circulation. The boundary layer was warmer and drier above 1000-m height and was slightly wetter up to 2000-m height. Soil wetness affected the circulation energetics proportionally to the soil dryness (for soil wetness below similar to 0.6). The shape of the deforestation controlled the impact on rainfall. The horizontal strips lined up with the prevailing wind showed a dominant increase in rainfall, significant up to about 60 000 km(2). On the other hand, in the patches aligned in the opposite direction (north-south), there was both increase and decrease in precipitation in two distinct regions, as a result of clearly separated upward and downward branches, which caused the precipitation to increase for patches up to 15 000 km(2). The authors` estimates for the size of deforestation impacting the rainfall contributed to fill up the low spatial resolution in other previous studies.
