[ES] Los flavonoides son compuestos polifenólicos que están omnipresentes en las plantas y muestran un amplio espectro de actividades biológicas. Aquí hemos estudiado el efecto del derivado tetraacetilado (QD) del producto natural 3 metil éter quercetina, sobre la viabilidad celular en las líneas celulares de leucemia humana Hl-60 y u937. Los resultados muestran que QD es citotóxico e induce parada en la fase G2-M del ciclo celular en ambas líneas celulares y es un potente inductor de la apoptosis. La apoptosis inducida por QD (i) es mediada por la activación de las caspasas, (ii) está asociada con la liberación del citocromo c y (iii) es activada en células u937 que sobreexpresan bcl-2. El tratamiento de células Hl-60 y u937 con QD provoca la activación de la vía de las quinasas activadas por mitógenos (MaPKs), incluyendo JNK, p38 MaPK y ErK 1/2. La inhibición de JNK mediante el sP600125 y de p38 MaPK mediante sB203580 no tiene influencia en la apoptosis mediada por QD. Por el contrario, la inhibición de ErK 1/2 con inhibidores farmacológicos u0126 o PD98059 potenció el porcentaje de apoptosis inducida por QD y sugiere que la inhibición de esta vía es una estrategia valiosa para aumentar la sensibilidad de células de leucemia humana Hl-60 hacia la QD.

[EN] Phytochemical research of two Tolpis species, T. proustii and T. lagopoda, led to the isolation of three new compounds: 30-chloro-3beta-acetoxy-22alfa-hydroxyl-20(21)- taraxastene (1), 3beta,22alfa-diacetoxy-30-ethoxy-20(21)-taraxastene (2) and 3beta,28-dihydroxy- 11alfa-hydroperoxy-12-ursene (3). The structures of the new compounds were elucidated by means of extensive IR, NMR, and MS data and by comparison of data reported in the literature. The in vitro antioxidant activities of the extracts were assessed by the DPPH and ABTS scavenging methods. The cytotoxicity of several known compounds and its derivatives was also assessed against human myeloid leukemia K-562 and K-562/ADR cell lines.

In questo elaborato viene studiato il comportamento asintotico di un polimero diretto in ambiente aleatorio (o disordine) al variare della temperatura e della dimensione in cui lo si considera. Si dimostra che quando d=1,2 il polimero si comporta in modo super diffusivo, ovvero la catena polimerica dispone i suoi monomeri lontano dall'origine per raggiungere zone in cui l'ambiente risulta maggiormente favorevole; al contrario, quando d è > o = 3, si verifica una transizione tra una fase diffusiva(valori alti della temperatura), in cui le traiettorie della catena si comportano, su larga scala, come un moto browniano, ed una fase super diffusiva(basse temperature).

BACKGROUND There is great variability for the type of anaesthesia used during TAVI, with no clear consensus coming from comparative studies or guidelines. We sought to detect regional differences in the anaesthetic management of patients undergoing transcatheter aortic valve implantation (TAVI) in Europe and to evaluate the relationship between type of anaesthesia and in-hospital and 1year outcome. METHODS Between January 2011 and May 2012 the Sentinel European TAVI Pilot Registry enrolled 2807 patients treated via a transfemoral approach using either local (LA-group, 1095 patients, 39%) or general anaesthesia (GA-group, 1712 patients, 61%). RESULTS A wide variation in LA use was evident amongst the 10 participating countries. The use of LA has increased over time (from a mean of 37.5% of procedures in the first year, to 57% in last 6months, p<0.01). MI, major stroke as well as in-hospital death rate (7.0% LA vs 5.3% GA, p=0.053) had a similar incidence between groups, confirmed in multivariate regression analysis after adjusting for confounders. Dividing our population in tertiles according to the Log-EuroSCORE we found similar mortality under LA, whilst mortality was higher in the highest risk tertile under GA. Survival at 1year, compared by Kaplan-Meier analysis, was similar between groups (log-rank: p=0.1505). CONCLUSIONS Selection of anaesthesia appears to be more influenced by national practice and operator preference than patient characteristics. In the absence of an observed difference in outcomes for either approach, there is no compelling argument to suggest that operators and centres should change their anaesthetic practice.

BACKGROUND The use of transcatheter mitral valve repair (TMVR) has gained widespread acceptance in Europe, but data on immediate success, safety, and long-term echocardiographic follow-up in real-world patients are still limited. OBJECTIVES The aim of this multinational registry is to present a real-world overview of TMVR use in Europe. METHODS The Transcatheter Valve Treatment Sentinel Pilot Registry is a prospective, independent, consecutive collection of individual patient data. RESULTS A total of 628 patients (mean age 74.2 ± 9.7 years, 63.1% men) underwent TMVR between January 2011 and December 2012 in 25 centers in 8 European countries. The prevalent pathogenesis was functional mitral regurgitation (FMR) (n = 452 [72.0%]). The majority of patients (85.5%) were highly symptomatic (New York Heart Association functional class III or higher), with a high logistic EuroSCORE (European System for Cardiac Operative Risk Evaluation) (20.4 ± 16.7%). Acute procedural success was high (95.4%) and similar in FMR and degenerative mitral regurgitation (p = 0.662). One clip was implanted in 61.4% of patients. In-hospital mortality was low (2.9%), without significant differences between groups. The estimated 1-year mortality was 15.3%, which was similar for FMR and degenerative mitral regurgitation. The estimated 1-year rate of rehospitalization because of heart failure was 22.8%, significantly higher in the FMR group (25.8% vs. 12.0%, p[log-rank] = 0.009). Paired echocardiographic data from the 1-year follow-up, available for 368 consecutive patients in 15 centers, showed a persistent reduction in the degree of mitral regurgitation at 1 year (6.0% of patients with severe mitral regurgitation). CONCLUSIONS This independent, contemporary registry shows that TMVR is associated with high immediate success, low complication rates, and sustained 1-year reduction of the severity of mitral regurgitation and improvement of clinical symptoms.

We investigate the thermal evolution of comet 67P/Churyumov-Gerasimenko's subsurface in the Seth_01 region, where active pits have been observed by the ESA/Rosetta mission. Our simulations show that clathrate destabilization and amorphous ice crystallization can occur at depths corresponding to those of the observed pits in a timescale shorter than 67P/Churyumov-Gerasimenko's lifetime in the comet's activity zone in the inner solar system. Sublimation of crystalline ice down to such depths is possible only in the absence of a dust mantle, which requires the presence of dust grains in the matrix small enough to be dragged out by gas from the pores. Our results are consistent with both pits formation via sinkholes or subsequent to outbursts, the dominant process depending on the status of the subsurface porosity. A sealed dust mantle would favor episodic and disruptive outgassing as a result of increasing gas pressure in the pores, while high porosity should allow the formation of large voids in the subsurface due to the continuous escape of volatiles. We finally conclude that the subsurface of 67P/Churyumov-Gerasimenko is not uniform at a spatial scale of similar to 100-200 m.

Pits have been observed on many cometary nuclei mapped by spacecraft(1-4). It has been argued that cometary pits are a signature of endogenic activity, rather than impact craters such as those on planetary and asteroid surfaces. Impact experiments(5,6) andmodels(7,8) cannot reproduce the shapes of most of the observed cometary pits, and the predicted collision rates imply that few of the pits are related to impacts(8,9). Alternative mechanisms like explosive activity(10) have been suggested, but the driving process remains unknown. Here we report that pits on comet 67P/Churyumov-Gerasimenko are active, and probably created by a sinkhole process, possibly accompanied by outbursts. We argue that after formation, pits expand slowly in diameter, owing to sublimation-driven retreat of the walls. Therefore, pits characterize how eroded the surface is: a fresh cometary surface will have a ragged structure with many pits, while an evolved surface will look smoother. The size and spatial distribution of pits imply that large heterogeneities exist in the physical, structural or compositional properties of the first few hundred metres below the current nucleus surface.

En 1976 se sembró E. lehmanniana Nees en la Reserva de Ñacuñán. Se evaluaron, después de 24 años de la siembra, la tasa lineal de dispersión, la abundancia y el impacto de esta especie exótica sobre la comunidad de gramíneas perennes nativas. Se compararon sitios dominados por la especie exótica con otros que incluían sólo vegetación nativa. Para comparar la cobertura de la canopia de las gramíneas, la cantidad de especies por parcela y la abundancia de la especie exótica se usó la prueba de Mann- Whitney. La especie exótica se dispersó 32 m año-1. Ella estaba presente en los caminos internos de la Reserva donde la vegetación nativa leñosa había sido eliminada y en el 45 % de los sitios ubicados en áreas no disturbadas adyacentes a los caminos. La cobertura total de las gramíneas nativas se redujo significativamente en los sitios donde la especie exótica estaba presente. La cantidad total de gramíneas por parcela no fue afectada por la presencia de E. lehmanniana. Pappophprum caespitosum, la gramínea dominante en Ñacuñán, y Sporobolus cryptandrus presentaron cobertura más baja en los sitios dominados por la especie exótica que en aquéllos no disturbados. La introducción de E. lehmanniana tuvo un impacto negativo en la composición de la comunidad de gramíneas perennes nativas en los sitios disturbados de la Reserva.

The Arabidopsis heterotrimeric G-protein controls defense responses to necrotrophic and vascular fungi. The agb1 mutant impaired in the Gβ subunit displays enhanced susceptibility to these pathogens. Gβ/AGB1 forms an obligate dimer with either one of the Arabidopsis Gγ subunits (γ1/AGG1 and γ2/AGG2). Accordingly, we now demonstrate that the agg1 agg2 double mutant is as susceptible as agb1 plants to the necrotrophic fungus Plectosphaerella cucumerina. To elucidate the molecular basis of heterotrimeric G-protein-mediated resistance, we performed a comparative transcriptomic analysis of agb1-1 mutant and wild-type plants upon inoculation with P. cucumerina. This analysis, together with metabolomic studies, demonstrated that G-protein-mediated resistance was independent of defensive pathways required for resistance to necrotrophic fungi, such as the salicylic acid, jasmonic acid, ethylene, abscisic acid, and tryptophan-derived metabolites signaling, as these pathways were not impaired in agb1 and agg1 agg2 mutants. Notably, many mis-regulated genes in agb1 plants were related with cell wall functions, which was also the case in agg1 agg2 mutant. Biochemical analyses and Fourier Transform InfraRed (FTIR) spectroscopy of cell walls from G-protein mutants revealed that the xylose content was lower in agb1 and agg1 agg2 mutants than in wild-type plants, and that mutant walls had similar FTIR spectratypes, which differed from that of wild-type plants. The data presented here suggest a canonical functionality of the Gβ and Gγ1/γ2 subunits in the control of Arabidopsis immune responses and the regulation of cell wall composition.

El número de cerdos por cuadra y la densidad influyen sobre el rendimiento productivo (Edmons et al., 1998). Además, la densidad de cría puede afectar a la calidad de la canal y de la carne (Estevez et al., 2003), ya que una reducción del espacio disponible da lugar a peleas y mordeduras de colas. Sin embargo, la producción de carne por m 2 aumenta con la densidad con un mejor aprovechamiento de la superficie disponible. Por otro lado, la densidad podría afectar de distinto modo a hembras (HE) y a machos castrados (MC) debido posiblemente al menor consumo voluntario de pienso de las HE. A este particular, Hamilton et al. (2003) observaron que a altas densidades de cría las HE fueron relativamente más magras mientras que los MC eran relativamente más grasos. Cambios en la densidad de cría, pueden afectar a la composición en ácidos grasos de la canal debido a cambios en el consumo y a la utilización del alimento (Nürnberg et al., 1998). El objetivo de este estudio fue comparar el efecto de la densidad de cebo de HE y MC con alto potencial de crecimiento, sobre el rendimiento productivo y la composición de la carne y el perfil de ácidos grasos en cerdos de 19 a 109 kg PV.
