The present study investigated the germination and vigor of Caesalpinia echinata (Brazilwood) seeds stored at negative temperatures. Recently harvested seeds were cryopreserved at -18º or -196ºC and periodically evaluated for germination, seed vigor and carbohydrate composition. The temperatures did not influence the germination percentages or vigor. The germination percentage decreased from 88% in recently harvested seeds to 60% after 730 days of storage. The different temperature and storage times tested did not affect the vigor seed germination as indicated by the measures of plant growth and survival. The different temperatures used did not cause changes in the carbohydrate composition. The tegument cell walls were rich in lignin, arabinose and xylose. The cytoplasm of the cotyledons and embryos had high levels of glucose, fructose, and sucrose. The cryopreservation technique here presented was effective in the conservation of Brazilwood seeds for the medium term.

Este estudo foi conduzido para avaliar a seletividade de herbicidas aplicados em pós-emergência sobre plantas de Panicum maximum, cultivares Tanzânia e Mombaça, e os seus efeitos na qualidade das sementes. Os tratamentos testados (em g ha-1) foram: diclofopmethyl (284), ametryne (1.250), propanil (3.600), chlorimuron-ethyl (15), nicosulfuron (50), bentazon (720) e atrazine (3.000), além de uma testemunha sem aplicação de herbicidas. Foram instalados dois experimentos a campo, um para cada cultivar, no delineamento em blocos casualizados com quatro repetições, e um em laboratório, no delineamento inteiramente casualizado, com 16 repetições. O nicosulfuron foi o herbicida que mais causou injúrias visuais e redução de massa seca de plantas em ambos os cultivares. No cultivar Mombaça, todos os herbicidas reduziram a produção de sementes, mas não afetaram a germinação. Para o cultivar Tanzânia, todos os herbicidas foram seletivos, não afetando a produção e a qualidade fisiológica.

Smellmelon, an annual invasive weed of soybean production fields in the north of Iran, reproduces and spreads predominately through seed production. This makes seed bank survival and successful germination essential steps in the invasive process. To evaluate the potential of Smellmelon to invade water-stressed environments, laboratory studies were conducted to investigate the effect of desiccation and salinity at different temperatures on seed germination and seedling growth of Cucumis melo. Seeds were incubated at 25, 30, 35 and 40 ºC in the darkness in a solution (0, -0.2, -0.4, -0.6, -0.8, 1 and 1.2 MPa) of a salt (NaCl), and in a solution (0, -2, -4, -6, -8, -10, -12 bar) of PEG-6000 (Polyethylene glycol), in two separate experiments. The results showed that the highest percentage and rate of germination occurred at 35 ºC in salt concentrations of 0, -0.2, -0.4 MPa and PEG concentrations of 0, -2, -4 bar. Increasing the concentration of salt (NaCl) and PEG limited germination, seedling growth and water uptake but increased the sodium content in the seedlings. No significant difference was observed among 0, -0.2 and -0.4 MPa of NaCl and among 0, -2 and -4 bar of PEG concentration at 35 ºC. The negative effects of PEG were more than those of NaCl on germination percentage and germination rate. Increased stress levels lead to reduction of root and shoot length, and SVL of seedlings. Na+ content of seedling decreased with limited seedling growth of C. melo.

A laboratory study was conducted to investigate the allelopathic effect of aqueous extracts of plant parts of Alternanthera philoxeroides and A. sessilis and soil incorporated residues on germination and seedling growth of rice (Oryza sativa). Aqueous extracts prepared from different plant parts of Alternanthera species delayed rice germination. Alternanthera philoxeroides and A. sessilis inhibited rice germination by 9-100% and 4-49%, respectively. Germination of rice seeds was reduced with increasing concentration of aqueous leaf extracts of both weed species. Early seedling growth (root and shoot lengths) and seedling vigor index were significantly reduced by 5% aqueous leaf extract compared with distilled water treated control. Germination, root and shoot lengths, root and shoot dry weights and seedling vigor index of rice were drastically reduced by 3 and 4% in residue infested soil compared with residue free soil. The inhibitory effect of A. philoxeroides in terms of germination and seedling growth of rice was greater than that of A. sessilis. Five percent aqueous leaf extract and 4% residue infested soil of A. philoxeroides caused complete failure of rice seed germination. Alternanthera philoxeroides contained water soluble phenolics, namely 4 hydroxy-3-methoxy benzoic acid (16.19 mg L-1) and m-coumaric acid (1.48 mg L-1), whereas Alternanthera sessilis was rich in chlorogenic acid (17.85 mg L-1), gallic acid (11.03 mg L-1) and vanillic acid (9.88 mg L-1). The study indicates that the allelopathic potential of Alternanthera species may play an important role in enhancing the invasiveness of these species and may suppress rice plants in the vicinity.

Jamun (Syzygium cumini L. Skeels) (Black plum, Damson plum) fruits weigh between 2-5 g at maturity. Fresh seeds represented 20-80% of the total fruit weight; the seed coat and cotyledons contributed 6% and 94% to the total seed weight respectively, while the weight of the embryonic axis was insignificant. Only the embryonic axis stained with Tetrazolium, not the cotyledons. The seeds are polyembryonic with up to four embryos, of which at most three germinate. Decoated seeds germinated faster than coated seeds under nursery conditions, with high significant germination percentages, dry matter production rates and vigor indices. The lack of staining of the cotyledon by tetrazolium was probably due to the presence of an impermeable layer. Decoating seeds for faster germination is recommended.

Sunflower crop was based, as yet, on high linoleic cultivars, but in the last years request for oil with higher content of oleic acid has increased, due to their dietary characteristics. At the beginning, high oleic cultivars were used to be sown in warm regions, but then the concern about growing it in temperate areas, as the south-east of Buenos Aires Province, was posed. In this region, early sowings are recommended, so that grain filling matches with a period of appropriate hydric and light conditions, as to result in greater yields. However, early sowings are limited by low soil temperature, that delays seedling emergence, resulting in heterogeneous stand establishment. The aim of this work was to evaluate seed performance of four high oleic cultivars in the southern area of Buenos Aires Province, by means of vigor tests and field trials. Germination, cold, tetrazolium viability, tetrazolium viability with cold, accelerated ageing tests and three field sowings at different soil temperatures were performed. Data were analyzed by Anova using generalised linear models, and tests and cultivars were contrasted among themselves. Similar seedling emergence under optimal and suboptimal temperatures for high oleic and high linoleic cultivars was recorded. The success of seedling establishment does not appear to be related to the acidic composition of seeds.

Excess salts in the root zone inhibit water uptake by plants, affect nutrient uptake and may result in toxicities due to individual salts in the soil solution. Excess exchangeable sodium in the soil may destroy the soil structure to a point where water penetration and root aeration become impossible. Sodium is also toxic to many plants. Beans (Phaseolus vulgaris L.) are consumed as protein source in northeastern Brazil, although little is known about common bean cultivar tolerance to salinity. The germination of bean cultivars under salt stress was studied. The cultivars 'Carioca' and 'Mulatinho' were submitted to germination test in a germinator at 25ºC, at the Seed Analysis Laboratory of the Brazilian Agricultural Research Corporation unit in the Semi- Arid region (Embrapa Semi Árido), Petrolina, Pernambuco State. These seeds were germinated on "germitest" papers imbibed in distilled water or in 10, 50, 100 e 200 mol.m-3sodium chloride (NaCl) solutions. At the first and second counts of the germination test, normal seedlings were counted, measured, weighed and dried, supplying data for vigor, total germination, fresh matter weight and dry matter weight and seedlings length. Total protein was quantified in cotyledons at 3, 6 and 9 days after sowing. The results indicated that the NaCl content influenced seed germination and concentrations above 50 mol.m-3 decreased germination and seedling growth.

Currently, the highest interest with respect to the assessment of seed physiological quality is to obtain reliable results in a relatively short period of time. This initiative allows for prompt decisions during different phases of seed production primarily after physiological maturity. This research was performed to verify the efficiency and rapidity of the method of Pettenkofer to determine the respiratory activity and to differentiate vigor levels of soybean seed lots. Three lots of soybean seeds cv. 8000 were used. Seed performance was determined by respiratory activity, compared to the following tests: standard germination, germination first count, electrical conductivity, seedling emergence, seedling shoot and root length and total dry mass. Results ranked seed lots according to defferences in physiological quality. Seed imbibition and conditioning period in Pettenkofer's equipment were enough to detect differences in vigor among seed lots, showing that the determination of the respiratory activity is a promising procedure to identify differences in vigor levels among soybean seed lots.

The effect of chemical and biological treatments on castor bean emergence, seedling vigor, dry matter production, and also the control of microorganisms associated with seeds of the AL Guarany 2002 and Lyra cultivars, was evaluated. The products tested were carbendazim + thiram, carboxin + thiram and a product based on Trichoderma. Total seed and seedling emergence were evaluated at 27 days after sowing whereas dry matter production was verified for plants removed 45 days after sowing. The Guarany 2002 AL cultivar had a higher incidence of microorganisms than the Lyra cultivar. The chemical treatment was 100% effective in controlling fungi but the biological treatment did not reduce microorganism incidence on the seeds. Chemical treatment resulted in plants with more dry matter and the best results were for carbendazim + thiram and carboxin + thiram at doses of 60 g + 140 g and 50 g + 50 g/100 kg of seeds, respectively. The carbendazim + thiram mixture was the only treatment which was statistically higher for total emergence whereas the biological treatment increased emergence only for the Lyra cultivar, thus demonstrating its lower efficiency. The importance of fungicides to control pathogens associated with seeds was discussed.

The objective of this study was to investigate the possibility of using hydric restriction as a method for evaluating vigor of soybean seeds. The soybean seeds, cultivar BRS 245RR, represented by four different seed lots, were characterized by germination and vigor. For the treatment of hydric restriction and temperature, the combination of substrate water potential and temperature were the following: deionized water (0.0 MPa); polyethylene glycol (PEG 6000) aqueous solution (-0.1, -0.3 and -0.5 MPa); and four temperatures (20 ºC, 25 ºC, 30 ºC, and 35 ºC), respectively. A completely randomized experimental design was used, with four replications per treatment, and the ANOVA was performed individually for each combination of temperature and water potential of substrate. According to results obtained, the test of hydric restriction has the same efficiency of the accelerated aging test in estimating vigor of soybean seeds, cv. BRS 245RR, when water potentials of -0.1 MPa or -0.3 MPa at a temperature of 25 ºC, or -0.3 MPa at a temperature of 30 ºC are used.

Mimosa caesalpiniaefolia Benth. is a forest species of the Mimosaceae family, recommended for recovery of degraded areas. The evaluation of vigor by biochemical tests have been an important tool in the control of seed quality programs, and the electrical conductivity and potassium leaching the most efficient in the verifying the physiological potential. The objective, therefore, to adjust the methodology of the electrical conductivity test for seeds of M. caesalpiniaefolia, for then compare the efficiency of this test with the potassium in the evaluation of seed vigor of different lots of seeds M. caesalpiniaefolia. To test the adequacy of the electrical conductivity were used different combinations of temperatures , 25 °C and 30 ºC, number of seeds , 25 and 50, periods of imbibition , 4 , 8 , 12 , 16 and 24 hours , and volumes deionized water, 50 mL and 75mL. For potassium leaching test, which was conducted from the results achieved by the methodology of the adequacy of the electrical conductivity test, to compare the efficiency of both tests , in the classification of seeds at different levels of vigor, and the period 4 hours also evaluated because the potassium leaching test can be more efficient in the shortest time . The best combination obtained in experiment of electrical conductivity is 25 seeds soaked in 50 mL deionized or distilled water for 8 hours at a temperature of 30 ° C. Data were subjected to analysis of variance, the means were compared with each other by F tests and Tukey at 5 % probability, and when necessary polynomial regression analysis was performed. The electrical conductivity test performed at period eight hour proved to be more efficient in the separation of seed lots M. caesalpiniaefolia at different levels of vigor compared to the potassium test

The accelerated ageing (AA) test is widely used to evaluate seed vigor in various species including vegetable crops. The objective of this work was to study procedures for conducting AA to evaluate seed vigor of pumpkin and zucchini seed lots. Five seed lots of commercially acceptable quality (germination) standards were selected from both cv. `Menina Brasileira' (zucchini) and `Barbara' Hybrid (pumpkin). The following tests were conducted: standard germination, the first count of germination, and AA with and without saline solution (periods of 48, 72 and 96 hours). For each combination of temperature and ageing period, seeds were placed in a single layer on a screen in a germination plastic box with either 40 mL deionized water or 40g NaCl/100mL of water. Both AA methods identified lots 2 and 5 of `Menina Brasileira' and lot 10 of `Barbara' hybrid as having the lowest physiological quality. The temperature 41 C promoted a more drastic reduction in germination than did 38 C. This observation was more marked after 96 hours. Based on the results obtained, it can be concluded that the combination 41 degrees C/96 hours should be used to evaluate the physiological potential of pumpkin and zucchini seeds; however, additional studies are necessary to evaluate other combinations and confirm this indication.

Dentro de um programa de controle de qualidade, a avaliação do vigor de sementes é fundamental e necessária para o sucesso da produção. O trabalho teve por objetivo comparar diferentes métodos para avaliação do potencial fisiológico de sementes de pepino. Para tanto, cinco lotes de sementes do híbrido Safira foram submetidos aos seguintes testes: germinação, primeira contagem de germinação, condutividade elétrica (4x50 sementes; 25ºC; 50 e 75mL; 12, 18 e 24h), germinação a baixa temperatura (4x50 sementes; 18ºC; contagens aos quatro e oito dias após a semeadura); envelhecimento acelerado tradicional (41ºC; 48, 72 e 96h) e com solução saturada de NaCl (41ºC; 72 e 96h). Pelos resultados obtidos, pode-se concluir que os testes de condutividade elétrica (50 e 75mL; 12, 18 e 24h) e de envelhecimento acelerado com o uso de solução salina (41ºC, por 72h), possibilitam a melhor identificação de lotes com diferentes níveis de qualidade fisiológica.

O teste de condutividade elétrica da solução de embebição é recomendado para avaliar o vigor de sementes de ervilha e indicado para outras espécies, mas ainda há necessidade da continuidade de estudos sobre o assunto. A presente pesquisa foi desenvolvida com o objetivo de comparar procedimentos no teste de condutividade elétrica para avaliar o potencial fisiológico de sementes de abobrinha. Utilizaram-se sementes de abobrinha (Cucurbita pepo L.), cultivar Menina Brasileira, representadas por quatro lotes na primeira etapa e por outros cinco, na segunda. Foram conduzidos os teste de germinação, primeira contagem de germinação, emergência de plântulas, índice de velocidade de emergência e de condutividade elétrica. Para o teste de condutividade foram estudadas variações no volume de água (50 e 75mL), na temperatura (20, 25 e 30ºC) e no tempo de embebição (1, 2, 4, 8, 12, 16, 20 e 24 horas). O tempo de embebição para o teste de condutividade elétrica pode ser reduzido para 8 horas. Independentemente do volume de água, a temperatura de 30ºC foi a mais favorável para a ordenação consistente dos lotes quanto ao vigor. Assim, conclue-se que a condição mais adequada para o teste de condutividade elétrica em sementes de abobrinha é a utilização de 50 sementes em 75mL de água por oito horas, a 30ºC.

A avaliação do vigor de sementes tem sido ferramenta fundamental dentro do programa de controle de qualidade, sendo o teste de envelhecimento acelerado (EA) parte importante do processo. O trabalho foi desenvolvido com o objetivo de confirmar metodologia específica para avaliação do vigor de sementes, em laboratório, tendo como base o EA. Utilizou sementes de milho e soja, seis lotes para cada espécie, com padrões de qualidade (germinação) comercialmente aceitas. As condições de envelhecimento préestabelecidas foram 42ºC/96h e 45ºC/72h para milho e 41ºC/48 e 72h e 42ºC/48 e 72h para soja. Para cada combinação de temperatura e período de envelhecimento, as sementes foram colocadas em caixas plásticas, sobre tela, com 40mL de água destilada, distribuídas em camada única e com base no peso das mesmas. O envelhecimento foi feito usando-se dois tipos de câmara, BOD e jaquetada de água (CJ). Foi determinado o teor de água (TA) e a germinação das sementes antes e após o EA. A absorção de água pelas sementes considerando-se todas as combinações de período de exposição e temperatura, as duas câmaras e as duas formas de determinação do TA, foi maior quando as sementes foram acondicionadas em camada única em relação aquelas pesadas (massa constante), usando-se a câmara jaquetada. Para o milho, a combinação de 45ºC/72h com o acondicionamento das sementes em camada única foi o método mais eficiente para a separação dos lotes quanto ao vigor. Para soja, a combinação 42ºC/48h foi mais eficiente na separação dos lotes, não havendo interferência do método de acondicionamento das sementes, na tela. Conclui-se que a camada única e a câmara tipo BOD devem ser preferencialmente utilizadas no teste EA face aos menores custos de aquisição e manutenção da BOD, visando à separação de lotes quanto aos níveis de vigor, para milho e soja.
