Inhibition of NFkB by the compound Bay 11–7082 (Bay) induces tolerogenic properties in dendritic cells (DC). While activation of NFkB can be induced by reactive oxygen species (ROS) and thiol/disulfide redox states, the consequences of NFkB blockade on ROS/redox state is not known. To generate immature DC, monocytes were cultured in GM-CSF and IL-4 (with or without Bay) for 48 h. Genes potentially involved in redox regulation were determined using microarray technology and validated using FACS, real-time PCR or western blotting. ROS were measured using two fluorescent dyes DHR-123 and DHE (to detect H2O2 or O2 respectively). We found increased expression of genes associated with reductants such as thioredoxin reductase (TrxR1) and glutathione (GSH), although those associated with the breakdown of H2O2 such as glutathione peroxidase, peroxiredoxins and catalase were decreased. Interestingly, Bay-treated DC produced less ROS in comparison to control DC under basal conditions and following stimulation with various pro-oxidants. In conclusion, Bay-treated DC display not only tolerogenic properties but also an intracellular reducing environment and an impaired ability to produce ROS. We are currently investigating whether exogenous ROS can interfere with the tolerogenic properties of Bay-treated DC.

Michael H. [Henry] Hogue (1878-1963) was the son of Lemuel and Frances Hogue of Wainfleet Township, Welland County. In 1901 Michael Hogue was married to Margaret Case and they had five sons, William, Lemuel, Harry, Ross and Frank. In the 1911 Census of Canada he is residing in Welland and working as a blacksmith for Howard H. Lymburner. At Lymburner’s retirement, Hogue purchased the business and moved it from 15 Niagara Street, Welland to 12 Frazer Street, Welland. Mr. Hogue’s blacksmith activities included work with various industries in Welland including the expansion of blacksmithing work into the automobile industry. Mr. Hogue also worked for subcontractors Canadian Dredge & Dock and G.L. Campbell. These jobs included work related to the Welland Ship Canal in 1923. The business ceased operation in 1956.

There is very little information on the subtidal bottom fauna of the shelf regions in the seas around India. What little is known is restricted to macro benthos. The paucity of the work on bottom fauna and the importance of mud banks in the fishery of the South West Coast of India has initiated the present study. Attempts have been made to obtain a picture of the bottom fauna of a mud bank region of the Kerala Coast. The difficulties involved in the sampling and analysis, especially the availability of a suitable vessel during the S.W. Monsoon, resulted in the work being restricted mainly to the Narakal mud bank region 6 Km. north of Cochin Detailed sampling is conducted using grab, dredge and to a small extent beam trawl, to assess the qualitative and quantitative nature of the macro benthos. Important species contributing to the fauna are identified and the standing crop estimated for different seasons. The meiobenthos was studied using core samples taken from the grab. Animals were identified to the major taxa. Standing crop of meiobenthos and the quantitative importance of different groups were also studied. The data collected have been interpreted and discussed. As an understanding of the physico-chemical aspects of the environment is essential in order to obtain a true picture of the benthos, attempts were made in this direction. Environmental parameters such as temperature of the sediment, salinity, temperature, and dissolved oxygen in the overlying water were studied .during the period of benthos investigation. Monthly observations on the dissolved inorganic and organic phosphorus in the area of investigation have been made. The physico-chemical nature of the sediment was also studied. Influence of these ecological variables on the bottom fauna is discussed.
