One-Carbon Metabolic Pathway Rewiring in Escherichia coli Reveals an Evolutionary Advantage of 10-Formyltetrahydrofolate Synthetase (Fhs) in Survival under Hypoxia

In cells, N-10-formyltetrahydrofolate (N-10-fTHF) is required for formylation of eubacterial/organellar initiator tRNA and purine nucleotide biosynthesis. Biosynthesis of N-10-fTHF is catalyzed by 5,10-methylene-tetrahydrofolate dehydrogenase/cyclohydrolase (FolD) and/or 10-formyltetrahydrofolate synthetase (Fhs). All eubacteria possess FolD, but some possess both FolD and Fhs. However, the reasons for possessing Fhs in addition to FolD have remained unclear. We used Escherichia coli, which naturally lacks fhs, as our model. We show that in E. coli, the essential function of folD could be replaced by Clostridium perfringens fhs when it was provided on a medium-copy-number plasmid or integrated as a single-copy gene in the chromosome. The fhs-supported folD deletion (Delta folD) strains grow well in a complex medium. However, these strains require purines and glycine as supplements for growth in M9 minimal medium. The in vivo levels of N-10-fTHF in the Delta folD strain (supported by plasmid-borne fhs) were limiting despite the high capacity of the available Fhs to synthesize N-10-fTHF in vitro. Auxotrophy for purines could be alleviated by supplementing formate to the medium, and that for glycine was alleviated by engineering THF import into the cells. The Delta folD strain (harboring fhs on the chromosome) showed a high NADP(+)-to-NADPH ratio and hypersensitivity to trimethoprim. The presence of fhs in E. coli was disadvantageous for its aerobic growth. However, under hypoxia, E. coli strains harboring fhs outcompeted those lacking it. The computational analysis revealed a predominant natural occurrence of fhs in anaerobic and facultative anaerobic bacteria.

Simultaneous presence of fhs and purT genes is disadvantageous for the fitness of Escherichia coli growth

In bacteria, alternate mechanisms are known to synthesize N-10-formyltetrahydrofolate (N10-formyl-THF) and formyl glycinamide ribotide (fGAR), which are important in purine biosynthesis. In one of the mechanisms, a direct transfer of one carbon unit from formate allows Fhs to convert tetrahydrofolate to N-10-formyl-THF, and PurT to convert glycinamide ribotide (GAR) to fGAR. Our bioinformatics analysis of fhs and purT genes (encoding Fhs and PurT) showed that in a majority of bacteria (similar to 94%), their presence was mutually exclusive. A large number of organisms possessing fhs lacked purT and vice versa. The phenomenon is so penetrating that even within a genus (Bacillus) if a species possessed fhs it lacked purT and vice versa. To investigate physiological importance of this phenomenon, we used Escherichia coli, which naturally lacks fhs (and possesses purT) as model. We generated strains, which possessed fhs and purT genes in singles or together. Deletion of purT from E. coli in the presence or absence of fhs did not confer a detectable growth disadvantage in pure cultures. However, growth competition assays revealed that the strains possessing either of the single genes outcompeted those possessing both the genes suggesting that mutual exclusion of purT and fhs in organisms confers fitness advantage in mixed cultures.

Los títulos de los reyes de León en los documentos medievales como reflejo de la continuidad del reino visigodo de Toledo

Resumen: Partiendo de la idea expuesta por S. Teillet de que en los últimos tiempos del reino visigodo de Toledo quedó configurada una plena identificación de esta formación política con Hispania, así como del hecho de que la continuidad entre el reino visigodo y el reino de Asturias fue real y evidente (si bien, el reino de Asturias es una realidad desde luego diferente del reino visigodo de Toledo), como creemos haber demostrado en trabajos anteriores, hemos considerado que la identificación de Hispania y el regnum gothorum permite una valoración más matizada de determinados títulos utilizados por los reyes de León, sucesores de los reyes de Asturias. A este tema en concreto dedicamos este trabajo, mediante el que hemos podido apreciar cómo la idea de continuidad de esta comunidad política con respecto al reino visigodo de Toledo, que se manifiesta en los más diversos aspectos, se plasma de alguna manera en la titulación de sus reyes que fueron, en efecto, los continuadores de los reyes del reino visigodo de Toledo. Por ello aspiraron al dominio de toda la Península, de Hispania, y tales aspiraciones quedaron reflejadas en la titulatura: rex o imperator Hispaniae, sin que se pueda afirmar, como se ha venido haciendo, que tales títulos eran un mero reflejo de los ideales neogóticos, elaborados a finales del siglo IX.

A Subcomissão dos Estados recebe Paulo Brossard, Ministro da Justiça, que defende maior participação dos Estados nos recursos da União. Elverlindo Renklein, representante do Departamento Intersindical de Estatística e Estudos Socioeconômicos (DIEESE), defende o fim do imposto de renda (IRRF) para assalariados na Subcomissão de Tributos, Participação e Distribuição das Receitas. Simão Sessim (PFL-RJ) aprova a palestra de Elverlindo e defende a fiscalização unificada dos impostos.

A nova Constituição e a melhoria do transporte coletivo. Cidadãos falam do transporte em Curitiba. Roberto Requião, prefeito de Curitiba, explica as melhorias do transporte coletivo na cidade e sugere que a Constituição garanta maior controle público sobre as frotas. Afonso Camargo (PMDB-PR) relata que é preciso o vale transporte para garantir o transporte público bom e barato. O substitutivo do Relator da Assembleia Nacional Constituinte (ANC) Bernardo Cabral garante eleições para governador do Distrito Federal. Mas o Presidente da República José Sarney considera que o Distrito Federal deve ter um prefeito nomeado. Cidadãos opinam sobre o tema. Bancada do Distrito Federal unida em favor das eleições. O Deputado Walmir Campelo (PFL-DF) informa que a eleição para governador é uma conquista que não pode ser alterada. O Deputado Sigmaringa Seixas (PT-SP) afirma que a tese de autonomia para o Distrito Federal conquistou os constituintes e que a opinião do Presidente não vai prevalecer. Rex Nazaré, Presidente do Conselho Nacional de Energia Nuclear, debate com os constituintes do Nordeste e Norte o uso pacífico da energia nuclear para resolver problemas como irrigação, conservação de alimentos e maior produção agrícola. O Deputado Jalles Fontoura (PFL-GO) declara que esse tipo de aplicação moderna da energia nuclear é perfeitamente viável. O Deputado Albérico Cordeiro (PFL-AL) opina sobre a questão.

Rex Nazaré, presidente da Comissão Nacional de Energia Nuclear (CNEN), discutiu com a bancada de Goiás na Constituinte sobre o acidente com uma bomba de césio em Goiânia. O Deputado Pedro Canedo (PFL-GO) fala sobre a necessidade de criar dispositivos para maior controle do material radioativo existente no Brasil. O Deputado Fernando Cunha (PMDB-GO) apresenta proposta para que haja um acordo entre CNEN, Secretarias de Estado e Municipais de Saúde e Meio Ambiente para melhor fiscalização. Lúcia Vânia (PMDB-GO) defende a democratização da política nuclear brasileira. O Deputado Fábio Feldmann (PMDB-SP) fala sobre a necessidade de criar mecanismos efetivos para evitar que a questão nuclear seja tratada exclusivamente pelas autoridades, dando à sociedade civil mais poder fiscalização. Rex Nazaré, presidente da CNEN, refere-se ao substitutivo que inclui no art. 31 a determinação de que as atividades nucleares serão realizadas exclusivamente com fins pacíficos e controladas pelo Congresso Nacional. Os sem-terra protestam alegando que a Comissão de Sistematização não considerou a emenda popular da reforma agrária. Não houve acordo entre empresários e líderes sindicais a respeito da estabilidade no emprego. O Senador Fernando Henrique informa que a questão será discutida em plenário. Foi aprovada a emenda do Deputado Paulo Paim (PT-RS), que altera o artigo 6º, onde se lê direito dos trabalhadores, foi acrescentada a expressão urbanos e rurais. O Deputado Luís Inácio Lula da Silva pede a aprovação do projeto que garantia plena estabilidade ao trabalhador. Após votação, a estabilidade não foi aprovada. O Deputado comenta sobre a situação da estabilidade no emprego no Brasil e como esse tema foi conduzido na Constituinte.

Distribution, Abundance, and Biological Characteristics of Groundfish off the Coast of Washington, Oregon, and California, 1977-1986

We compare results of bottom trawl surveys off Washington, Oregon, and California in 1977, 1980, 1983, and 1986 to discern trends in population abundance, distribution, and biology. Catch per unit of effort, area-swept biomass estimates, and age and length compositions for 12 commercially important west coast groundfishes are presented to illustrate trends over the lO-year period. We discuss the precision, accuracy, and statistical significance of observed trends in abundance estimates. The influence of water temperature on the distribution of groundfishes is also briefly examined. Abundance estimates of canary rockfish, Sebastes pinniger, and yellowtail rockfish, S. Jlavidus, declined during the study period; greater declines were observed in Pacific ocean perch, S. alutus, lingcod, Ophiodon elongatus, and arrowtooth flounder, Atheresthes stomias. Biomass estimates of Pacific hake, Merluccius productus, and English, rex, and Dover soles (Pleuronectes vetulus, Errex zachirus, and Microstomus pacificus) increased, while bocaccio, S. paucispinis, and chilipepper, S. goodei, were stable. Sablefish, Anoplopoma fimbria, biomass estimates increased markedly from 1977 to 1980 and declined moderately thereafter. Precision was lowest for rockfishes, lingcod, and sablefish; it was highest for flatfishes because they were uniformly distributed. The accuracy of survey estimates could be gauged only for yellowtail and canary rockfish and sablefish. All fishery-based analyses produced much larger estimates of abundance than bottom trawl surveys-indicative of the true catchability of survey trawls. Population trends from all analyses compared well except in canary rockfish, the species that presents the greatest challenge to obtaining reasonable precision and one that casts doubts on the usefulness of bottom trawl surveys for estimating its abundance. Membrane Deformability and Membrane Tension of Single Isolated Mitochondria

Mitochondria dynamics is crucial to many biological processes such as mitochondria fusion and fission, which is highly correlated to the mechanics of single mitochondria. However, the mechanobiological coupling of mitochondria has been poorly understood. Here membrane deformability and membrane tension of individual mitochondria isolated from MtDsRed labeled human embryonic T-Rex-293 kidney cells were measured using a micropipette aspiration assay. The results demonstrated that membrane deformation of isolated mitochondria exhibited an elastic transition phase followed by an equilibrium phase, and mitochondrial membrane tension was proportional to the area compressibility. It was also indicated that mitochondrial membrane deformability was significantly affected by physical chemical factors such as osmotic pressure or pH value, and was further correlated to mitochondrial functionality in different respiratory states and Ca2+ regulation. These findings provide a new insight into understanding the mechanical regulation of mitochondrial physiology.

A versão portuguesa do mito de Édipo

Esta dissertação visa a leitura do texto dramático António Marinheiro (O Édipo de Alfama) de Bernardo Santareno, tendo como ponto de partida Édipo Rei de Sófocles. Os fundamentos teóricos para a interpretação desses textos são a obra freudiana e o ensino de Jacques Lacan. Santareno retira as suas personagens do interior das muralhas de Tebas para colocá-las em Alfama, bairro de Lisboa, na segunda metade do século XX. Nesse novo cenário, o fado, o vinho e a taberna compõem os costumes tipicamente portugueses. Escolhemos dois filmes, Édipo Rei de Pier Paolo Pasolini e Incêndios de Denis Villeneuve, para fazer uma leitura comparativa com as peças de Sófocles e de Bernardo Santareno. O início do filme de Pasolini ocorre em uma cidade pequena ao norte da Itália e depois se transfere para a Grécia antiga onde se passa a tragédia de Sófocles. Incêndios é uma adaptação da peça do libanês Wadji Mouawad e retrata a guerra civil do Líbano, a intolerância religiosa, estupro, incesto e genocídio

Whole-gear efficiency of a benthic survey trawl for flatfish

Whole-gear efficiency (the proportion of fish passing between the otter doors of a bottom trawl that are subsequently captured) was estimated from data collected during experiments to measure the herding efficiency of bridles and doors, the capture efficiency of the net, and the length of the bridles sufficiently close to the seafloor to elicit a herding response. The experiments were focused on four species of flatfish: arrowtooth flounder (Atheresthes stomias), flathead sole (Hippoglossoides elassodon), rex sole (Glyptocephalus zachirus), and Dover sole (Microstomus pacificus). Whole-gear efficiency varied with fish length and reached maximum values between 40% and 50% for arrowtooth flounder, flathead sole, and rex sole. For Dover sole, however, whole-gear efficiency declined from a maximum of 33% over the length range sampled. Such efficiency estimates can be used to determine catchability, which, in turn, can be used to improve the accuracy of stock assessment models when the time series of a survey is short.
