First published November 19, 2009; doi:10.1152/japplphysiol.00490.2009.-Skeletal muscle atrophy induced by denervation and metabolic diseases has been associated with increased ubiquitin ligase expression. In the present study, we evaluate the influence of androgens on muscle ubiquitin ligases atrogin-1/MAFbx/FBXO32 and Murf-1/Trim63 expression and its correlation with maintenance of muscle mass by using the testosterone-dependent fast-twitch levator ani muscle (LA) from normal or castrated adult male Wistar rats. Gene expression was determined by qRT-PCR and/or immunoblotting. Castration induced progressive loss of LA mass (30% of control, 90 days) and an exponential decrease of LA cytoplasm-to-nucleus ratio (nuclear domain; 22% of control after 60 days). Testosterone deprivation induced a 31-fold increase in LA atrogin-1 mRNA and an 18-fold increase in Murf-1 mRNA detected after 2 and 7 days of castration, respectively. Acute (24 h) testosterone administration fully repressed atrogin-1 and Murf-1 mRNA expression to control levels. Atrogin-1 protein was also increased by castration up to 170% after 30 days. Testosterone administration for 7 days restored atrogin-1 protein to control levels. In addition to the well known stimulus of protein synthesis, our results show that testosterone maintains muscle mass by repressing ubiquitin ligases, indicating that inhibition of ubiquitin-proteasome catabolic system is critical for trophic action of androgens in skeletal muscle. Besides, since neither castration nor androgen treatment had any effect on weight or ubiquitin ligases mRNA levels of extensor digitorum longus muscle, a fast-twitch muscle with low androgen sensitivity, our study shows that perineal muscle LA is a suitable in vivo model to evaluate regulation of muscle proteolysis, closely resembling human muscle responsiveness to androgens.

Objective To compare the quality of induction and recovery, degree of muscle relaxation, clinically apparent potency and cardiopulmonary effects of racemic ketamine or S(+)-ketamine when used for total intravenous anesthesia in horses. Study design Prospective randomized clinical trial Animals Sixteen healthy stallions (323 +/- 99 kg), with a mean age of 6.2 years, undergoing castration. Methods Horses were pre-medicated with romifidine IV, 15 minutes before induction of anesthesia. Each animal was then randomly allocated to receive either diazepam and ketamine (DK) or diazepam and S(+)-ketamine (DKS) at similar doses to induce anesthesia. For maintenance of anesthesia, 1/4 of the initial bolus of ketamine alone or S(+)-ketamine alone was administered, as required. Heart rate (HR), respiratory rate (RR) and systolic blood pressure were measured before and at 10-minute intervals during recumbency. Time from induction to lateral recumbency, time from induction to first additional dose, time from last additional dose to return to sternal posture and time from last additional dose to standing were recorded, and a subjective evaluation of quality of induction, endotracheal intubation, muscle relaxation and quality of recovery was recorded. Results The quality of the induction and duration of anesthesia were similar in both groups. HR, RR and systolic blood pressure were not significantly different between groups. Although some animals which received DKS showed some minor excitatory effects (25% of them) during the induction of anesthesia, these animals received 32% fewer doses for the maintenance of anesthesia and the recovery scores were better. Conclusions and clinical relevance S(+)-ketamine showed some advantages over racemic ketamine, such as less anesthetic agent being required and better overall recovery from anesthesia. Further studies are needed to obtain the optimum induction dose for the S(+)-ketamine.

The effects of castration on the primal joints and the cuts of the leg joint of Javan rusa (Cervus timorensis russa) stag carcases was investigated at three slaughter ages (13, 19 and 25 months). Castration reduced the weights of some primal joints in the 19- and 25-months age groups, but not at 13 months. At 19 months, the neck, and neck plus chuck, were heavier by 35 and 17% respectively in entires (P < 0.05), and at 25 months entires had heavier carcases, shoulder, forequarter and hindquarter (P < 0.05). The leg and saddle joints were approximately 39 and 18% of the side, respectively, for both treatments and all ages. The proportions of the neck, and neck plus chuck, were higher (P < 0.05) in 19-month old entires than castrates. There were few significant differences between treatments in the weight or proportion of the hind leg cuts at any slaughter age, but in the 25-months group the silverside was 8% (P < 0.05) heavier in entires. In both castrates and entires, there appeared to be an increase in the percentage of the rump as the animals grew from 13 to 19 months of age. (C) 2001 Elsevier Science Ltd. All rights reserved.

The dissected carcass composition and fatty acid profiles of intermuscular fat from 110 male goat kids from six genotypes i.e. Boer x Angora (BA), Boer x Feral (BF), Boer x Saanen (BS), Feral x Feral (1717), Saanen x Angora (SA) and Saanen x Feral (SF) and two slaughter weight groups i.e. Capretto and Chevon (liveweight at slaughter 14-22 and 30-35 kg, respectively) were compared. Carcass tissue distribution for various genotypes was: muscle (63-66%), fat (10-13%) and bone (21-24%). Genotype significantly (P < 0.05) influenced the carcass composition; BA and FF carcasses had significantly higher muscle to bone ratio, while carcasses from BS kids were leaner compared to other genotypes. However, the two slaughter weight groups did not differ significantly (P > 0.05) in terms of carcass composition, when compared at the same carcass weight. In the present study, significant (P < 0.01) correlations were observed between percentage of muscle, fat and bone in most of the primal cuts and that in the carcass side. The main saturated fatty acids (SFAs) identified were palmitic (16:0) and stearic acid (18:0), while oleic acid (18: 1, omega9) was the main unsaturated fatty acid (UFA) in the intermuscular fat from goat kids. There were significant (P < 0.05) differences between genotypes in the proportions of individual fatty acids. Adipose tissue from BS kids had significantly higher UFAs (mainly oleic acid) and thus had a significantly lower melting point compared to other genotypes. There were significantly higher proportions of palmitic acid (35%) in the adipose tissue from Capretto kids compared to that from Chevon kids (22%). The authors summarize the results of former works, based on the technics of parabiosis. After parabiotic union of two infantile rats, normal + castrate, the normal fellow enters into precocious puberty in about 7 days (Kallas). In the case of pairs: castrated male + normal female, the implants of testicles, or injection of maceration or aqueous extracts of testis in the castrated fellow, prevents the induction of early puberty in the normal female. In the case: castrated female + normal female, no inhibiting effect is provoked by that treatment. There is therefore a testicular hormone that regulates the hypophysis. After castration, this gland manifests a hyper-function and shows histological alterations, the chief character of these being the appearing in the anterior lobe, of the so-called castration cells, probably originated from basophile cells. Implants or injections of testis material prevent those alterations. This is a useful test; the effect is controlled by estimating the castration cells in the microscopic field. The testicular hormone that regulates the anterior lobe is probably another one, quite different from that which regulates the accessory genitalia. On account of the facts and experiments, it may be assumed that this new hormone is elaborated by the germinal epithelium of the testicles.

The work of this thesis would like to investigate two particular questions about the care of underage: on the one hand, the maltreatment, and the mother's desire on the other hand. About the first, this proposal try to underscore the elaborate irregularity and disparity related to this phenomenon and his historical evolution. The objective's thesis propose to underline that the abuse crosses the crisis of educational dimension inside and outside the family. We are talking about a type of crisi that places at the origin of a specific clinic and that it is about the maltreatment designated, at the same time, to cope with the educational void and with the ill-treated under age's hardships. The so-called clinic of the maltreatment horns, therefore, with the purpose to join and to set in order the normative and educational aspects. However, the prescriptive requirement is intended to measure against the same one crisis in a paradoxical way; the laws and the juveniles court must make up for the lack absence of the rules that characterized the maltreatment, getting around to use the typical reference that traditionally were been at the base of the educational way's construction. The same crimes on underage often found in the father the responsibility. The requirements and the therapeutic needs has utilized the theoretical construct of the trauma as the key line of clinical practice. On the one hand, the trauma was been paired controversially to the psychoanalytical concepts as the notion of the phantom, in the same manner that the objective truth is been opposed to the notion of the imagination and fantasy; on the other hand, the trauma was gotten to be the mark and the characteristic that defined the individual identity of the mistreated persons, regardless of the person's clinical structure or, generally, regardless of the their individual position. Both the normative demand and therapeutic converge on the idealization of the imaginary figure that is able to supply to every type of maltreatment: the mother. This position is connected on theoretical construct well-established in the mythology, that associated the mother figure to a natural dimension; this is related to the second point at issue: the mother's desire. The awareness of the mother figure in natural and idealized terms describes accusingly and negatively the trauma, without seeing the difference between trauma that helps to give a structure to the individual position from the trauma solely devastating. The mother's mythology, therefore, is in an evident antagonism with the female figure and woman's sexuality. In these way the maltreatment's clinic suggests that a woman is able to be a mother only when she doesn't appreciate the rule of female. This situation preclusives a comparison with own sexuality and gives to the women a complex of castration. The clinic cases reported highlight as these difficulties are the expression of the familiarity's heredity that wasn't been sufficiently elaborated. This condition (into the relation between mother and his child) turn the infant in the incapacity to symbolized and it doesn't help him to accede to the positive trauma, to the language and to the edipic rule; this is the reason why the child toils difficulties to approach to own personality and to orient himself in the relation with his body, with the body of the adults and the same age children.

Boar taint is the off-odour or off flavour of cooked pork. Currently, the most common method of controlling boar taint is surgical castration. However, immunocastration has been used in some parts of the world as an alternative to surgical castration. The aim of this study was to evaluate the sensory acceptability of meat from immunocastrated pigs (IM) compared with meat from females (FE), surgically castrated (CM) and entire males (EM). Twenty animals of each type were evaluated by 201 consumers in 20 sessions. Longissimus thoracis muscle of the different animals was cooked in an oven at 180 °C for 10 min. Consumers scored the odour and the flavour of the meat in a 9-point category scale without an intermediate level. There were no significant differences in consumer’s evaluation of meat from IM, CM, and FE. In contrast, EM meat presented a higher percentage of dissatisfied scores and was significantly (P & 0.05) less accepted than meat from CM, IM and FE. Consumers’ acceptability of EM meat was always lower, independently of its androstenone levels. However meat with low levels of androstenone was more accepted that meat with medium or high levels of this substance. It can be concluded that immunocastration produced pork that was accepted by the consumers, and was indistinguishable from pork from CM or FE.

El objetivo de este estudio fue evaluar el efecto de la castración quirúrgica (MC), la inmunocastración (MI), la producción de hembras (HE) y machos enteros (ME) sobre la productividad, el comportamiento y el bienestar animal. La castración quirúrgica se realizó a los siete días de edad y la inmunocastración mediante la administración a los 77 y 146 días de edad de un producto compuesto por una forma modifi cada del GnRF conjugado a una proteína capaz de inducir la formación de anticuerpos frente al GnRF. Durante el engorde, se evaluó el consumo individual, peso, espesor de grasa y lomo de 120 cerdos (36 ME, 36 MI, 24 MC y 24 hembras HE). Asimismo, se registró el comportamiento de los animales durante dos días consecutivos en las semanas 9, 11, 20, 21, 23 y 25. Los MI presentaron un crecimiento e ingestión diarias superior a los ME y un mejor índice de conversión que los MC. El espesor de lomo de los MI fue superior al de los MC, pero inferior al de los ME. La actividad se vio reducida en los MI en comparación con los ME, especialmente después de la segunda administración. Estos resultados sugieren que la inmunocastración es una alternativa a la castración quirúrgica. Result of differet alternatives to surgical castration of pigs The objective of this study was to evaluate the effect of surgical castration (MC), immunocastration (MI), keeping females (HE) and entire males (ME) on performance, behaviour and welfare. The surgical castration was carried out at 7 days of age, and the immunocastration through the administration at 77 and 146 days of age of a modifi ed form of GnRF conjugated to protein to induce the formation of antibodies against GnRF. Individual consumption, weight and fat and loin depth of 120 pigs (36 ME, 36 MI, 24MC and 24 HE) was measured. Moreover, behaviour was recorded during two consecutive days in weeks 9, 11, 20, 21, 23 and 25. MI presented a higher growth and feed intake compared to ME and a better conversion ratio than MC. Loin depth was higher for MI than for MC, but lower than for ME. Activity was reduced in MI compared to ME, especially after the second administration. These results suggest that immunocastration is an alternative to surgical castration.

The number of eggs laid per snail in Bradybaena similaris and the nucleic acids (DNA and RNA) in the albumen gland and ovotestis were quantified in snails infected with sporocysts of the digenetic trematode Eurytrema coelomaticum. The total number of eggs laid per mollusc was reduced by 96.32% at the end of the larval development. The DNA concentration increased by 700% and the RNA concentration was reduced by 8,38% by the time when the daughter sporocysts of E. coelomaticum were released from B. similaris. The relation between these values and the inhibition of the reproduction observed in infected molluscs is discussed.

ABSTRACT: BACKGROUND: Upregulation of nuclear factor kappa B (NFκB) activity and neuroendocrine differentiation are two mechanisms known to be involved in prostate cancer (PC) progression to castration resistance. We have observed that major components of these pathways, including NFκB, proteasome, neutral endopeptidase (NEP) and endothelin 1 (ET-1), exhibit an inverse and mirror image pattern in androgen-dependent (AD) and -independent (AI) states in vitro. METHODS: We have now investigated for evidence of a direct mechanistic connection between these pathways with the use of immunocytochemistry (ICC), western blot analysis, electrophoretic mobility shift assay (EMSA) and proteasome activity assessment. RESULTS: Neuropeptide (NP) stimulation induced nuclear translocation of NFκB in a dose-dependent manner in AI cells, also evident as reduced total inhibitor κB (IκB) levels and increased DNA binding in EMSA. These effects were preceded by increased 20 S proteasome activity at lower doses and at earlier times and were at least partially reversed under conditions of NP deprivation induced by specific NP receptor inhibitors, as well as NFκB, IκB kinase (IKK) and proteasome inhibitors. AD cells showed no appreciable nuclear translocation upon NP stimulation, with less intense DNA binding signal on EMSA. CONCLUSIONS: Our results support evidence for a direct mechanistic connection between the NPs and NFκB/proteasome signaling pathways, with a distinct NP-induced profile in the more aggressive AI cancer state.

The tubero-infundibular and nigrostriatal DA neurone systems of rats respond to systemic (i.p.) injection of alpha-MSH (2-100 microgram/kg). The response of the tubero-infundibular (arcuate) DA neurones, an increase in cellular fluorescence intensity which can be interpreted as a sign of increased neuronal activity, is essentially the same in males, estrogen-progesterone-pretreated ovariectomized females and hypophysectomized males, whereas the type of response elicited by alpha-MSH in the nigral DA neurones depends upon the hormonal state of the animal. Differences between the two DA neurone groups exist also with regard to the effects of peptide fragments containing the two active sites of the alpha-MSH molecule. Results of lesion experiments in the lower brainstem (area postrema) and of blockade of muscarinic mechanisms by atropine further point to differences in the mechanisms underlying the peptide effects on the two neurone systems. The reaction of the tubero-infundibular DA system (which controls the pars intermedia of the pituitary) can be considered to reflect the activation of a feedback mechanism on MSH secretion, while the functional counterpart of the changes observed in the nigral system remains unknown at the present time.

Les patients métastatiques d'un cancer prostatique répondent initialement à un traitement antiandrogénique. Après un temps variable, la maladie progresse néanmoins. Le cancer est défini alors comme « hormono-résistant. ». Cependant des manipulations hormonales ultérieures, (antiandrogènes, oestrogènes), peuvent induire une nouvelle baisse du PSA contredisant l'appellation « hormono-résistant ». Certains auteurs préfèrent parler de maladie « résistante à la castration ». Cette étude rétrospective lausannoise rapporte l'expérience de traitement oestrogéniques chez des patients dits « hormono-résistants».

Axes et clés de lecture. Ce travail est structuré sur trois axes de recherche qui sont restitués par chacune des parties proposées. Ces parties ont une structure narrative permettant une lecture relativement indépendante des autres, et contenant des renvois indiqués au cas par cas. A chacune de celle-ci, des introductions contextuelles plus spécifiques sont proposées. D'autre part, les citations originales sont signalées par des chiffres romains qui renvoient en fin d'ouvrage après la bibliographie. Enfin, la bibliographie ne contient que les références citées et prioritairement exploitées dans ce travail, en jugeant inutile d'y mentionner l'ensemble des lectures exploratoires. La première approche consiste à mettre en évidence une homosexualité «sociale », en envisageant le «milieu» homosexuel suisse au cours des années de guerre dans sa pluralité. Le premier chapitre s'attache à visibiliser la «scène homosexuelle», associative, commerciale et de rencontres entre hommes. Il fait ressortir la place éminente de Zurich, mais aussi éclaire d'autres aspects moins connus. Ainsi, l'association «Der Kreis - Le Cercle», par son existence unique au monde durant les années de guerre, mérite une analyse de son fonctionnement, de la forme de son militantisme et de l'influence de ses acteurs clés sur le développement de ce regroupement d'«homoérotes». Les données proviennent essentiellement des fonds conservés aux Archives gaies à Zurich, et sont complétées par des informations inédites provenant de certains dossiers militaires. Cependant, la scène homosexuelle ne se limite pas à l'association, et les publicités parues dans le «Kreis», les études contemporaines et les données contenues dans les dossiers de la justice militaire permettent de reconstituer, en partie, les formes de possibilité de rencontre entre homosexuels et les lieux fréquentés sur l'ensemble du pays. Le deuxième chapitre propose une étude résolument inédite sur les termes et les vécus de, et sur, les homosexualités, en se plaçant au niveau des hommes poursuivis par la justice militaire. Par leurs aveux ou leurs dénis, les prévenus se définissent comme étant «homosexuels », «bisexuels », «normaux » ou ne savent pas comment qualifier leurs actes. En outre, les témoins et les lésés emploient des termes plus ou moins grossiers pour qualifier l'homosexualité, tout comme la police et la justice optent pour des termes en apparence plus neutres. L'ensemble de ces termes relevés dans les dossiers de la justice militaire ouvre un champ d'analyse unique et précieux pour pouvoir mettre en évidence des variations symboliques sur l'homosexualité, et porteuses de significations différentes de part et d'autre de la Sarine. Enfin, la richesse des dossiers de la justice militaire permet d'explorer, au-delà des arguments conservateurs de l'époque, une infime partie de la réalité de la prostitution homosexuelle, en reprenant les catégories employées par Michel Dorais dans son étude sur les travailleurs du sexe masculins en Amérique du Nord au début du XXIe siècle.as Le deuxième axe développe l'aspect des normes pénales sur l'homosexualité, depuis leur élaboration par les chambres fédérales jusqu'aux applications concrètes et interprétations jurisprudentielles par la justice militaire. Le troisième chapitre, reprenant la chronologie des débats en commissions ad hoc et parlementaires, retrace l'adoption du droit pénal militaire, puis du droit pénal civil, en analysant les processus politiques à la base d'articles pénaux distincts sur la question de la pénalisation de l'homosexualité entre adultes consentants. En apparence, les articles pénaux sont contradictoires, mais ils se rejoignent profondément par leurs reprises des conceptions allemandes du droit, classique pour l'un et réformateur pour l'autre, et leurs interprétations s'interpénètrent durant les années de guerre. L'accès aux dossiers de la justice militaire permet de voir plus précisément la manière dont l'article pénal est appliqué et quels actes sont considérés comme étant «homosexuels ». Aussi, le quatrième chapitre retrace-t-il les affaires à l'origine des recours adressés au Tribunal Militaire de~ Cassation et motivant les principes jurisprudentiels de la primauté du pouvoir appréciateur des juges de première instance, du lien entre l'homosexualité et la «pédophilie » lorsque les victimes sont des garçons mineurs, de la pondération des conclusions d'un expert psychiatre dans les verdicts ou encore de la non automaticité de l'octroi d'un sursis en cas d'irresponsabilité ou d'une bonne conduite. En ratifiant le plus souvent les décisions des tribunaux de première instance, la cour de cassation valide les interprétations sur les actes commis et leur gravité. En peu d'année, il se produit une extension des actes tombant sous le coup de la «débauche contre nature », et les tentatives d'actes, les exhibitions, les attouchements plus ou moins brutaux des parties génitales d'un autre homme et la masturbation mutuelle deviennent punissables. Ces interprétations .marquent une convergence des logiques juridiques conservatrices françaises et allemandes, et la pénalisation de la masturbation mutuelle est également adoptée pour les civils. Le concours des droits français et allemand est ensuite envisagé sous l'angle de l'outrage public aux moeurs, ce qui permet de voir que le «vice» / «Unzucht», héritier du droit canonique, est transféré dans le droit «positif» comme une loi sociale en considérant l'outrage comme une notion objective en soi. La dernière partie propose une lecture sur les liens complexes entre la psychiatrie et l'homosexualité. Le cinquième chapitre débute en relatant la formation d'une psychiatrie militaire comme corps sanitaire distinct dans l'armée suisse. Avant la Seconde Guerre mondiale, les procédures de collaboration entre l'armée et la psychiatrie civile sont peu formalisées, et des directives instituant des services de consultation psychiatriques et des procédures d'appel à des psychiatres, en fonction de leur incorporation dans l'armée, sont introduites quelques mois après l'entrée en guerre. Ces ordres ont pour base les Instructions sur l'appréciation sanitaire des militaires, un livret contenant les diagnostics sur les maladies pouvant se rencontrer chez des militaires, les conditions de leur réforme sanitaire du service militaire, ainsi que des instructions de prise en charge des patients. Parmi les neuf codes diagnostics sur les pathologies psychiatriques, la procédure pour le chiffre 250/71 («perversion sexuelle») prévoit une exclusion de l'armée par la voie sanitaire. La non-conservation des dossiers sanitaires ne permet pas de mener une étude des liens entre l'homosexualité et la psychiatrie militaire en dehors des demandes de la justice militaire. Par contre -aspect tout à fait exceptionnel - 47 expertises psychiatriques sont accessibles, et permettent d'analyser en profondeur .les conceptions théoriques et les registres discursifs sur l'homosexualité. La période de la guerre se caractérise par une absence complète d'unité théorique sur la question. En effet, des variations théoriques significatives peuvent s'observer chez un même psychiatre, en fonction de la formulation de la demande par des juges d'instruction ou du grade et de la classe sociale de l'expertisé. Aussi, ce chapitre propose-t il d'explorer les différentes catégories d'homosexualité identifiées -innée, latente, bisexuelle, commise avec des mineurs ou en relation avec la consommation d'alcool - en ne reprenant que les expertises de quatre officiers psychiatres, et en les joignant de trois autres à fin de comparaison. Le sixième chapitre poursuit et rompt partiellement le rapport entre l'homosexualité et la psychiatrie, en envisageant les apports de la médecine somatique. Un unique cas de castration découvert dans les dossiers de la justice militaire permet de poursuivre l'analyse, essentiellement par les publications médicales de l'époque sur la castration «thérapeutique » des délinquants sexuels. En Suisse, la castration ne peut se faire que sur le consentement «éclairé », mais celui-ci apparaît fortement contraint dans les faits, et cette «thérapeutique » permet d'aborder une face sombre de la psychiatrie institutionnelle civile. Par la théorie des hormones sexuelle, la médecine somatique apparaît subitement comme une rivale sur la question de l'étiologie des déviances sexuelles. Toutefois, dans un accord médical dominant sur les bienfaits de la castration, ces deux sciences se confortent pour déterminer, tantôt à cause d'un surplus de testostérone, tantôt en raison d'une force psychique supérieure à la moyenne, une libido plus impérieuse chez les homosexuels délinquants. Enfin, quatre expertises menées par des médecins somaticiens en Suisse romande s'éloignent plus nettement de la psychiatrie et permettent d'entrevoir un dernier aspect complémentaire du rapport entre la médecine et l'homosexualité. Ces expertises démontrent la survivance d'une pratique médico-légale de l'homosexualité par l'examination des pénis et des anus, qui est ensuite mise en relation avec la moralité et la responsabilité des prévenus. Cette influence de la France sur la Suisse romande apparaît toutefois minime, et montre a contrario que l'objet juridique de la responsabilité pénale de l'homosexualité est devenu de la compétence de la psychiatrie sur l'ensemble du pays.

Molecular characterization of radical prostatectomy specimens after systemic therapy may identify a gene expression profile for resistance to therapy. This study assessed tumor cells from patients with prostate cancer participating in a phase II neoadjuvant docetaxel and androgen deprivation trial to identify mediators of resistance. Transcriptional level of 93 genes from a docetaxel-resistant prostate cancer cell lines microarray study was analyzed by TaqMan low-density arrays in tumors from patients with high-risk localized prostate cancer (36 surgically treated, 28 with neoadjuvant docetaxel þ androgen deprivation). Gene expression was compared between groups and correlated with clinical outcome. VIM, AR and RELA were validated by immunohistochemistry. CD44 and ZEB1 expression was tested by immunofluorescence in cells and tumor samples. Parental and docetaxel-resistant castration-resistant prostate cancer cell lines were tested for epithelial-to-mesenchymal transition (EMT) markers before and after docetaxel exposure. Reversion of EMT phenotype was investigated as a docetaxel resistance reversion strategy. Expression of 63 (67.7%) genes differed between groups (P < 0.05), including genes related to androgen receptor, NF-k B transcription factor, and EMT. Increased expression of EMT markers correlated with radiologic relapse. Docetaxel-resistant cells had increased EMT and stem-like cell markers expression. ZEB1 siRNA transfection reverted docetaxel resistance and reduced CD44 expression in DU-145R and PC-3R. Before docetaxel exposure, a selected CD44 þ subpopulation of PC-3 cells exhibited EMT phenotype and intrinsic docetaxel resistance; ZEB1/CD44 þ subpopulations were found in tumor cell lines and primary tumors; this correlated with aggressive clinical behavior. This study identifies genes potentially related to chemotherapy resistance and supports evi-dence of the EMT role in docetaxel resistance and adverse clinical behavior in early prostate cancer.

The effect of caponisation on fat composition by parts (wing, breast, thigh, and drumstick) and tissues (skin, subcutaneous adipose tissue, intermuscular adipose tissue and muscle) was examined in the present study and fatty acid profiles of abdominal fat and edible meat by parts and tissue components were determined. The sample was made up of twenty-eight castrated and twenty male Penedesenca Negra chicks reared under free-range conditions and slaughtered at 28 wk of age; the birds were castrated at four or eight weeks. Caponisation significantly increased (P < 0.01) the chemical fat content in all parts (16.31% to 37.98% in breast; 21.98% to 34.13% in wing; 21.09% to 49.57% in thigh; 14.33% to 24.82% in drumstick) and led to minor modifications in fat haracteristics, particularly in the thigh and the drumstick, where the unsaturated vs. saturated fatty acid ratio increased from 1.31 to 1.76 ( P < 0.01) and from 1.48 to 2.07 (P < 0.01), respectively. Delaying the age of castration from 4 to 8 weeks increased this ratio by 0.35 in the edible meat. Even though the profile of the abdominal fat is less saturated in capons, all changes occurring on fat quality after caponisation indicate that increased fatness after castration does not imply worse fat nutritional properties.
