One approach to reducing the yield losses caused by banana viral diseases is the use of genetic engineering and pathogen-derived resistance strategies to generate resistant cultivars. The development of transgenic virus resistance requires an efficient banana transformation method, particularly for commercially important 'Cavendish' type cultivars such as 'Grand Nain'. Prior to this study, only two examples of the stable transformation of banana had been reported, both of which demonstrated the principle of transformation but did not characterise transgenic plants in terms of the efficiency at which individual transgenic lines were generated, relative activities of promoters in stably transformed plants, and the stability of transgene expression. The aim of this study was to develop more efficient transformation methods for banana, assess the activity of some commonly used and also novel promoters in stably transformed plants, and transform banana with genes that could potentially confer resistance to banana bunchy top nanovirus (BBTV) and banana bract mosaic potyvirus (BBrMV). A regeneration system using immature male flowers as the explant was established. The frequency of somatic embryogenesis in male flower explants was influenced by the season in which the inflorescences were harvested. Further, the media requirements of various banana cultivars in respect to the 2,4-D concentration in the initiation media also differed. Following the optimisation of these and other parameters, embryogenic cell suspensions of several banana (Musa spp.) cultivars including 'Grand Nain' (AAA), 'Williams' (AAA), 'SH-3362' (AA), 'Goldfinger' (AAAB) and 'Bluggoe' (ABB) were successfully generated. Highly efficient transformation methods were developed for both 'Bluggoe' and 'Grand Nain'; this is the first report of microprojectile bombardment transformation of the commercially important 'Grand Nain' cultivar. Following bombardment of embryogenic suspension cells, regeneration was monitored from single transfom1ed cells to whole plants using a reporter gene encoding the green fluorescent protein (gfp). Selection with kanamycin enabled the regeneration of a greater number of plants than with geneticin, while still preventing the regeneration of non-transformed plants. Southern hybridisation confirmed the neomycin phosphotransferase gene (npt II) was stably integrated into the banana genome and that multiple transgenic lines were derived from single bombardments. The activity, stability and tissue specificity of the cauliflower mosaic virus 358 (CaMV 35S) and maize polyubiquitin-1 (Ubi-1) promoters were examined. In stably transformed banana, the Ubi-1 promoter provided approximately six-fold higher p-glucuronidase (GUS) activity than the CaMV 35S promoter, and both promoters remained active in glasshouse grown plants for the six months they were observed. The intergenic regions ofBBTV DNA-I to -6 were isolated and fused to either the uidA (GUS) or gfjJ reporter genes to assess their promoter activities. BBTV promoter activity was detected in banana embryogenic cells using the gfp reporter gene. Promoters derived from BBTV DNA-4 and -5 generated the highest levels of transient activity, which were greater than that generated by the maize Ubi-1 promoter. In transgenic banana plants, the activity of the BBTV DNA-6 promoter (BT6.1) was restricted to the phloem of leaves and roots, stomata and root meristems. The activity of the BT6.1 promoter was enhanced by the inclusion of intron-containing fragments derived from the maize Ubi-1, rice Act-1, and sugarcane rbcS 5' untranslated regions in GUS reporter gene constructs. In transient assays in banana, the rice Act-1 and maize Ubi-1 introns provided the most significant enhancement, increasing expression levels 300-fold and 100-fold, respectively. The sugarcane rbcS intron increased expression about 10-fold. In stably transformed banana plants, the maize Ubi-1 intron enhanced BT6.1 promoter activity to levels similar to that of the CaMV 35S promoter, but did not appear to alter the tissue specificity of the promoter. Both 'Grand Nain' and 'Bluggoe' were transformed with constructs that could potentially confer resistance to BBTV and BBrMV, including constructs containing BBTV DNA-1 major and internal genes, BBTV DNA-5 gene, and the BBrMV coat protein-coding region all under the control of the Ubi-1 promoter, while the BT6 promoter was used to drive the npt II selectable marker gene. At least 30 transgenic lines containing each construct were identified and replicates of each line are currently being generated by micropropagation in preparation for virus challenge.

Background Adolescent Idiopathic Scoliosis is the most common type of spinal deformity, and whilst the risk of progression appears to be biomechanically mediated (larger deformities are more likely to progress), the detailed biomechanical mechanisms driving progression are not well understood. Gravitational forces in the upright position are the primary sustained loads experienced by the spine. In scoliosis they are asymmetrical, generating moments about the spinal joints which may promote asymmetrical growth and deformity progression. Using 3D imaging modalities to estimate segmental torso masses allows the gravitational loading on the scoliotic spine to be determined. The resulting distribution of joint moments aids understanding of the mechanics of scoliosis progression. Methods Existing low-dose CT scans were used to estimate torso segment masses and joint moments for 20 female scoliosis patients. Intervertebral joint moments at each vertebral level were found by summing the moments of each of the torso segment masses above the required joint. Results The patients’ mean age was 15.3 years (SD 2.3; range 11.9 – 22.3 years); mean thoracic major Cobb angle 52° (SD 5.9°; range 42°-63°) and mean weight 57.5 kg (SD 11.5 kg; range 41 – 84.7 kg). Joint moments of up to 7 Nm were estimated at the apical level. No significant correlation was found between the patients’ major Cobb angles and apical joint moments. Conclusions Patients with larger Cobb angles do not necessarily have higher joint moments, and curve shape is an important determinant of joint moment distribution. These findings may help to explain the variations in progression between individual patients. This study suggests that substantial corrective forces are required of either internal instrumentation or orthoses to effectively counter the gravity-induced moments acting to deform the spinal joints of idiopathic scoliosis patients.

Dynamic properties of proteins have crucial roles in understanding protein function and molecular mechanism within cells. In this paper, we combined total internal reflection fluorescence microscopy with oblique illumination fluorescence microscopy to observe directly the movement and localization of membrane-anchored green fluorescence proteins in living cells. Total internal reflect illumination allowed the observation of proteins in the cell membrane of living cells since the penetrate depth could be adjusted to about 80 nm, and oblique illumination allowed the observation of proteins both in the cytoplasm and apical membrane, which made this combination a promising tool to investigate the dynamics of proteins through the whole cell. Not only individual protein molecule tracks have been analyzed quantitatively but also cumulative probability distribution function analysis of ensemble trajectories has been done to reveal the mobility of proteins. Finally, single particle tracking has acted as a compensation for single molecule tracking. All the results exhibited green fluorescence protein dynamics within cytoplasm, on the membrane and from cytoplasm to plasma membrane.

O objetivo deste estudo foi avaliar a Influência da associação de catalisadores químicos junto a diferentes sistemas adesivos autocondicionantes e cimentos resinosos de dupla polimerização, na cimentação de pinos pré-fabricados de fibra de vidro, quanto a sua resistência ao cisalhamento por extrusão - push out, seu grau de conversão e nanoinfiltração. Foram utilizadas trinta raízes bovinas extraídas, que tiveram seus canais obturados com guta-percha termoplastificada, divididas em três grupos: G1 - Adper SE Plus /Rely X ARC; G2- Adper SE Plus / Catalisador Scotchbond + Rely X ARC ; G3- Clearfil SE Bond / ED Primer + Panavia F. Após a cimentação dos pinos foram obtidas fatias das raízes, com 1mm de espessura, dos terços cervical (C), médio (M) e apical (A). O ensaio de resistência ao cisalhamento por extrusão foi realizado em máquina de ensaio universal EMIC D500 com carga de 100KN à velocidade de 1,0 mm/min, até o deslocamento do pino. Os dados obtidos no ensaio foram tabulados e submetidos à análise estatística. A análise de variância a dois critérios mostrou que apenas os fatores grupo e profundidade foram significativos, não sendo significativa a sua interação. O resultado do teste de Tukey (ρ≥0,05), para o fator grupo, mostrou que a menor média de resistência ao cisalhamento por extrusão foi obtida pelo grupo 3, que apresentou diferença estatística significativa para os grupos 1 e 2 que não diferiram entre si. Para o fator Profundidade a maior média foi obtida no terço cervical que apresentou diferença estatística significativa para os terços médio e apical que não diferiram entre si: A análise do grau de conversão foi feita após vinte e quatro horas e os fatores estudados foram os cimentos resinosos Panavia F e Rely X ARC e os catalisadores químicos ED Primer e Catalisador Scotchbond, na forma incorporada ou aplicada superficialmente aos cimentos, formando 10 grupos experimentais. Para o grau de conversão, o teste de Tukey mostrou que o catalisador químico não aumentou o grau de conversão do RelyX ARC, já para o Panavia F, este aumentou significativamente seu grau de conversão. Quanto a análise em MEV da nanoinfiltração para o fator grupo, o resultado do teste de Tukey (ρ≥0,05) mostrou que a maior média foi obtida pelo grupo 3 que apresentou diferença estatística significativa para os grupos 1 e 2, que não diferiram entre si. Para o fator Profundidade a maior média foi obtida no terço apical que apresentou diferença estatística significativa para o terço cervical, que não diferiu do terço médio. Conclusões: 1- A associação de catalisadores químicos não aumentou a resistência ao cisalhamento por extrusão dos pinos de fibra de vidro. 2- O cimento Panavia F é dependente de catalisador aumentar o grau de conversão. 3- A associação de catalisadores químicos não foi capaz de alterar o padrão da camada híbrida, representado pela nanoinfiltração. 4- A nanoinfiltração está associada inversamente ao grau de conversão dos sistemas de cimentação, o que contribui negativamente para a resistência adesiva de pinos de fibra de vidro.

O presente estudo teve como objetivo comparar a qualidade do selamento promovido por 3 cimentos reparadores endodônticos: Ceramicrete, iRoot-BP Plus e BioAggregate, com o ProRoot MTA branco, utilizando o modelo de infiltração de glicose sob pressão. 64 incisivos centrais superiores, recém-extraídos e sem tratamento endodôntico foram selecionados. A instrumentação do canal radicular foi realizada em todos os dentes com brocas Gates-Glidden e limas K-Flexofile (Dentsply Maillefer, Ballaigues Suíça), 1mm aquém do ápice. A patência foraminal foi confirmada a cada instrumento usado. Os dentes tiveram suas coroas removidas padronizando-os em 15mm de comprimento. Foi realizada apicetomia a 3mm do ápice e o preparo da cavidade retrógrada com o auxílio do ultrassom Various 350 (NSKNakanishi Inc., Tóquio, Japão) e da ponta E32D (NSKNakanishi Inc., Tóquio, Japão). As raízes foram distribuídas, randomicamente, em quatro grupos experimentais (n=15): G1Ceramicrete, G2iRoot BP Plus, G3BioAggregate e G4ProRoot MTA branco. Como controle negativo (n=2) foram utilizados dentes hígidos, e como controle positivo (n=2), dentes acessados e com patência confirmada. Os cimentos reparadores foram manipulados seguindo as recomendações do fabricante e inseridos na cavidade retrógrada utilizando a mesma ponta ultrassônica usada no preparo. As amostras foram mantidas na presença de umidade por 72h para o completo endurecimento dos materiais. As raízes foram montadas em um dispositivo de dupla-câmara selada para a infiltração da glicose. Foram utilizados 0,75ml de solução de glicose a 1Mol/L na câmara superior e 0,75ml de água destilada na câmara inferior. Os dispositivos foram conectados a um sistema de distribuição de pressão, que permitiu a infiltração de 32 amostras em uma mesma etapa. A solução de glicose foi forçada apicalmente sob uma pressão de 15psi durante 1 hora. Uma alíquota de 10l foi coletada da câmara inferior para quantificar a glicose infiltrada. A concentração de glicose foi determinada com o auxílio do Kit GlucoseHK (Megazyme, Wicklow, Irlanda) e de um espectrofotômetro de luz visível (Campsec M330, Cambridge, Reino Unido) em um comprimento de onda de 340nm. O teste não-paramétrico Kruskal-Wallis e o teste Dunns foram utilizados na análise estatística. Os resultados encontrados demonstraram que houve diferença significativa entre os grupos experimentais (p=0,0036). O BioAggregate apresentou a maior concentração de glicose-1,85(g/L), seguido do ProRoot MTA-1,2; IRoot BP-0,85 e Ceramicrete-0,75. Não houve diferença entre os três novos cimentos reparadores e o material padrão-ouro (p>0,05). Não houve diferença entre Ceramicrete e iRoot BP Plus (p>0,05), porém estes foram estatisticamente diferentes do BioAggregate (p<0,05). Diante dos resultados obtidos, pode-se concluir que: nenhum dos cimentos testados foi capaz de promover selamento hermético; os três novos cimentos testados não revelaram o mesmo padrão de selamento; os três novos cimentos testados revelaram um padrão de selamento semelhante a do ProRoot MTA branco; e o Ceramicrete e o iRoot BP Plus apresentaram padrão de selamento superior em comparação com o BioAggregate.

O presente estudo visa avaliar a influência da espessura do filme de cimento sobre a resistência de união de pinos de fibra de vidro em diferentes regiões do conduto radicular (cervical, médio e apical), cimentados com cimento resinoso autoadesivo, com e sem a adição de Rodamina B, por meio do teste de push-out. Quarenta raízes foram incluídas em resina epóxi, submetidas a tratamento endodôntico e obturadas com guta percha e cimento endodôntico sem eugenol. Após sete dias, os condutos foram desobstruídos e aleatoriamente divididos em 4 grupos (n=10), de acordo com as brocas do sistema de pinos de fibra WhitePost DC (FGM) usadas: (G1) broca #2; (G2) broca #3; (G3) broca #4; (G4) broca #2. O preparo foi realizado a uma profundidade de 10 mm. A cimentação foi realizada com o cimento resinoso autoadesivo RelyX U100 (3M ESPE), e apenas nos grupos G1, G2 e G3 uma pequena quantidade de Rodamina B em pó foi usada como corante no cimento. Após uma semana, cada raiz foi seccionada em máquina de corte, obtendo-se 6 fatias de 1 0,1 mm de espessura. Antes do ensaio de push-out, imagens digitais foram obtidas, por meio de um estereomicroscópio, de ambas as faces de cada fatia, para determinação do raio dos pinos e da espessura do filme de cimento. Após o ensaio mecânico, novas imagens foram obtidas para determinação do modo de falha. Para determinar a espessura de cimento, foi desenvolvida uma rotina (macro) no software KS 400. Os dados foram estatisticamente analisados com análise de variância (ANOVA) 2 fatores (influência do diâmetro da broca e influência dos terços) e Kruskal-Wallis (influência da espessura do filme de cimento). Comparações múltiplas foram realizadas com o teste Duncan. Todos os testes foram aplicados com α=0,05. Houve diferenças significantes entre os grupos em relação ao diâmetro da broca (p<0,0001), sendo G2 (14,62 5,15 MPa) > G1 (10,04 5,13 MPa) > G3 (7,68 6,14 MPa). O terço do conduto exerceu influência significativa sobre a resistência adesiva (p<0,0001), sendo os maiores valores obtidos no terço apical. As espessuras do filme de cimento foram estatisticamente diferentes nos grupos. Os maiores valores de espessura de cimento foram obtidos no G3 (248,78 μm), seguido de G2 (185,91 μm) e G1 (110,16 μm), sendo o último estatisticamente semelhante ao G4 (119,99 μm). Os resultados de G1(10,04 5,13 MPa) e G4 (8,89 + 5,18 MPa) foram estatisticamente semelhantes, indicando que a presença da Rodamina B não influencia significativamente na resistência de união. O tipo de falha predominante no G1 foi mista, no G2, adesiva entre pino e cimento, e no G3 e G4, adesiva entre cimento e dentina. O diâmetro da broca influenciou significativamente nos resultados de resistência de união ao teste de push-out. O ligeiro aumento na espessura do filme de cimento promoveu um aumento nos valores de resistência, quando comparado com filmes de cimento muito finos ou muito espessos.

An iris tumor developed in a 37-year-old woman who had had a bronchial carcinoid tumor resected nine years previously. The iris tumor was locally excised with a modified trabeculectomy approach. Histologic studies showed it to be a metastatic carcinoid tumor. Electron microscopy demonstrated typical dark and pale carcinoid cells with neurosecretory granules, basal bodies, and apical microvilli. The cisternae of the granular endoplasmic reticulum were disposed in a series of concentric rings encapsulating a central core of mitochondria. This unusual type of subcellular organization and specialization is probably a reflection of the slow-growing and highly differentiated nature of the iris tumor.

Purpose: The aim of the study was to assess the in vitro bond strength (BS) of glass fiber posts (GF) and carbon fiber posts (CF) in the cervical, middle, and apical thirds of root canals cemented with RelyX-Unicem (RX) and Cement-Post (CP). Materials and Methods: Forty maxillary canines were divided into 4 groups (n = 10) according to the cement and post used: group 1: GF and RX; group 2: CF and RX; group 3: GF and CP; group 4: CF and CP. The push-out test was applied in the cervical, middle and apical thirds of each specimen to assess bond strength of the cement/post complex to the root canal wall. The data obtained were submitted to ANOVA (Bonferroni test, p < 0.05), and fracture analysis was done with SEM. Results: The GF posts presented the best results when cemented with RX and with CF (p < 0.05). RX presented the highest BS values for both GF and CF (p < 0.05). For all the groups, BS was higher in the cervical third, followed by the middle and apical thirds. Fracture analysis showed a predominance of cohesive fracture of posts for RX, and a predominance of adhesive fracture between dentin/cement, and mixed failure mode for CP. Conclusion: GF posts cemented with RX presented the highest BS values in all root thirds.

Leaves comprise most of the vegetative body of tank bromeliads and are usually subjected to strong longitudinal gradients. For instance, while the leaf base is in contact with the water accumulated in the tank, the more light-exposed middle and upper leaf sections have no direct access to this water reservoir. Therefore, the present study attempted to investigate whether different leaf portions of Guzmania monostachia, a tank-forming C(3)-CAM bromeliad, play distinct physiological roles in response to water shortage, which is a major abiotic constraint in the epiphytic habitat. Internal and external morphological features, relative water content, pigment composition and the degree of CAM expression were evaluated in basal, middle and apical leaf portions in order to allow the establishment of correlations between the structure and the functional importance of each leaf region. Results indicated that besides marked structural differences, a high level of functional specialization is also present along the leaves of this bromeliad. When the tank water was depleted, the abundant hydrenchyma of basal leaf portions was the main reservoir for maintaining a stable water status in the photosynthetic tissues of the apical region. In contrast, the CAM pathway was intensified specifically in the upper leaf section, which is in agreement with the presence of features more suitable for the occurrence of photosynthesis at this portion. Gas exchange data indicated that internal recycling of respiratory CO(2) accounted for virtually all nighttime acid accumulation, characterizing a typical CAM-idling pathway in the drought-exposed plants. Altogether, these data reveal a remarkable physiological complexity along the leaves of G. monostachia, which might be a key adaptation to the intermittent water supply of the epiphytic niche. The purpose of this study was to evaluate the effectiveness of different light-curing units on the bond strength (push-out) of glass fiber posts in the different thirds of the root (cervical, middle and apical) with different adhesive luting resin systems (dual-cure total-etch; dual-cured and self-etch bonding system; and dual-cure self-adhesive cements), Disks of the samples (n = 144) were used, with approximately 1 mm of thickness of 48 bovine roots restored with glass fiber posts, that were luted with resin cements photo-activated by halogen LCU (QTH, Optilux 501) and blue LED (Ultraled), with power densities of 600 and 550 mW/cm(2), respectively. A universal testing machine (MTS 810 Material Test System) was used with a 1 mm diameter steel rod at cross-head speed of 0.5 mm/min until post extrusion, with load cell of 50 kg, for evaluation of the push-out strength in the different thirds of each sample. The push-out strength values in kgf were converted to MPa and analyzed through Analysis of Variance and Tukey`s test, at significance level of 5%. The results showed that there were no statistical differences between the QTH and LED LCUs. The self-adhesive resin cement had lower values of retention. The total-etch and self-adhesive system resin cements seem to be a possible alternative for glass fiber posts cementation into the radicular canal and the LED LCU can be applied as an alternative to halogen light on photo-activation of dual-cured resin cements.

The objective of this work was to evaluate the oviposition behavior of the whitefly Bemisia tabaci (Genn.) biotype B in relation to plant age, adult density and plant region on common beans plants. The genotype Perola was used to do the test. In free-choice tests 20, 30 and 40 day-old plants were used where evaluated the oviposition preference of the insect. In no-choice test, 30 day-old plants were used to evaluate densities of 50, 100 and 150 adults per plants. No-choice test for plant position (apex, median, base) 40 day-old plants were adopted for liberation of adults. The 30 and 40 day-old plants were preferred for oviposition, the densities of 100 adults per plants ensured the number of eggs enough to discriminate common beans genotypes with different whitefly resistance degrees, and apical region of plant were preferred for whitefly egg laying and therefore more suitable for egg sampling in common bean plants.

The objective was to study the phenology of Caryocar brasiliense Camb. (Caryocaraceae), a type of fruit of the Brazilian cerrado, in three areas (cerrado, pasture and in the campus of the Federal University of Minas Gerais) of the municipality of Montes Claros, State of Minas Gerais, Brazil, during 3 years. The plants of C brasiliense presented linear growth with about 50 cm of height and 35 cm of crown width per year. Soils with loamy texture, dystrophic, rich in aluminum and with low pH favor the production of this species. C. brasiliense produced higher quantity of flowers in the middle third, followed by the apical and basal parts of the crown, while the fruit production was higher in the basal third, followed by the middle and apical parts of the canopy of this species. C brasiliense bloomed during the dry period and produced fruits in the rainy season. Its fruits presented about 6, 8 and 6 cm of width, length and height, respectively, and 160 g of gross weight. The extrativism without control has strong impact on the propagation of C brasiliense because only about 7.96 and 10.65% of plants up to 1.0 m height and 59.58 and 44.73% higher than 3.0 m (reproductive phase) was observed in the cerrado and pasture, respectively. This indicates that fruit collectors, practically, remove all fruits of this tree and thus they reduce, considerably, the propagation of C. brasiliense in the cerrado areas of Brazil.
