Recent molecular studies on langurs of the Indian subcontinent suggest that the widely-distributed and morphologically variable Hanuman langurs (Semnopithecus entellus) are polyphyletic with respect to Nilgiri and urple-faced langurs. To further investigate this scenario, we have analyzed additional sequences of mitochondrial cytochrome b as well as nuclear protamine P1 genes from these species. The results confirm Hanuman langur polyphyly in the mitochondrial tree and the nuclear markers suggest that the Hanuman langurs share protamine P1 alleles with Nilgiri and purple-faced langurs. We recommend provisional splitting of the so-called Hanuman langurs into three species such that the taxonomy is consistent with their evolutionary relationships.

BTK-2, a 32 residue scorpion toxin initially identified in the venom of red Indian scorpion Mesobuthus tamulus was cloned, overexpressed and purified using Cytochrome 155 fusion protein system developed in our laboratory. The synthetic gene coding for the peptide was designed taking into account optimal codon usage by Escherichia coli. High expression levels of the fusion protein enabled facile purification of this peptide. The presence of disulfide bonded isomers, occurring as distinctly populated states even in the fusion protein, were separated by gel filtration chromatography. The target peptide was liberated from the host protein by Tev protease cleavage and subsequent purification was achieved using RP-HPLC methods. Reverse phase HPLC clearly showed the presence of at least two isomeric forms of the peptide that were significantly populated. The oxidative folding of BTK-2 was achieved under ambient conditions during the course of purification. Structural characterization of the two forms, by solution homonuclear and heteronuclear NMR methods, has shown that these two forms exhibit significantly different structural properties, and represent the natively folded and a "misfolded" form of the peptide. The formation of properly folded BTK-2 as a major fraction without the use of in vitro oxidative refolding methods clearly indicate the versatility of the Cytochrome b(5) fusion protein system for the efficient production of peptides for high resolution NMR studies.
Molecular phylogeny and biogeography of langurs and leaf monkeys of South Asia (Primates: Colobinae)

The two recently proposed taxonomies of the langurs and leaf monkeys (Subfamily Colobinae) provide different implications to our understanding of the evolution of Nilgiri and purple-faced langurs. Groves (2001) [Groves, C.P., 2001. Primate Taxonomy. Smithsonian Institute Press, Washington], placed Nilgiri and purple-faced langurs in the genus Trachypithecus, thereby suggesting disjunct distribution of the genus Trachypithecus. [Brandon-Jones, D., Eudey, A.A., Geissmann, T., Groves, C.P., Melnick, D.J., Morales, J.C., Shekelle, M., Stewart, C.-B., 2003. Asian primate classification. Int. J. Primatol. 25, 97–162] placed these langurs in the genus Semnopithecus, which suggests convergence of morphological characters in Nilgiri and purple-faced langurs with Trachypithecus. To test these scenarios, we sequenced and analyzed the mitochondrial cytochrome b gene and two nuclear DNA-encoded genes, lysozyme and protamine P1, from a variety of colobine species. All three markers support the clustering of Nilgiri and purple-faced langurs with Hanuman langur (Semnopithecus), while leaf monkeys of Southeast Asian (Trachypithecus) form a distinct clade. The phylogenetic position of capped and golden leaf monkeys is still unresolved. It is likely that this species group might have evolved due to past hybridization between Semnopithecus and Trachypithecus clades.

Exposure of cold-acclimatized rats to heat (37 degrees C) for a short period decreased brown adipose tissue (BAT) mitochondrial substrate-dependent oxygen uptake and H2O2 generation. Both the concentration and substrate-dependent rate of cytochrome b reduction decreased as early as 3 h of heat exposure. These results identify cytochrome b as the locus of regulation of electron transport in BAT mitochondria under conditions of heat stress.

Oral administration (250 mg/kg) of menthofuran, a monoterpene furan, to rats once daily for 3 days caused hepatotoxicity as judged by a significant increase in serum glutamate pyruvate transaminase (SGPT) and decreases in glucose-6-phosphatase and aminopyrine N-demethylase activities. Administration of menthofuran also resulted in a decrease in the levels of liver microsomal cytochrome P-450, whereas cytochrome b(5) and NAD(P)H-cytochrome c reductase activities were not affected. These effects of menthofuran were both dose- and time-dependent. Pretreatment of rats with phenobarbital (PB) prior to menthofuran treatment potentiated hepatotoxicity suggesting that a PB-induced cytochrome P-450 catalyzed the formation of reactive metabolite(s) responsible for the hepatotoxicity.

Previous work has shown that irrespective of the route of exposure methyl isocyanate (MIC) caused acute lactic acidosis in rats (Jeevaratnam et al., Arch. Environ. Contam. Toxicol. 19, 314�319, 1990) and the hypoxia was of stagnant type due to tissue hypoperfusion resulting from hypovolemic hypotension in rabbits administered MIC subcutaneously (Jeevarathinam et al., Toxicology 51, 223�240, 1988). The present study was designed to investigate whether MIC could induce histotoxic hypoxia through its effects on mitochondrial respiration. Male Wistar rats were used for liver mitochondrial and submitochondrial particle (SMP) preparation. Addition of MIC to tightly coupled mitochondria in vitro resulted in stimulation of state 4 respiration, abolition of respiratory control, decrease in ADP/O ratio, and inhibition of state 3 oxidation. The oxidation of NAD+-linked substrates (glutamate + malate) was more sensitive (fiveto sixfold) to the inhibitory action of MIC than succinate while cytochrome oxidase remained unaffected. MIC induced twofold delay in the onset of anerobiosis, and cytochrome b reduction in SMP with NADH in vitro confirms inhibition of electron transport at complex I region. MIC also stimulated the ATPase activity in tightly coupled mitochondria while lipid peroxidation remained unaffected. As its hydrolysis products, methylamine and N,N?-dimethylurea failed to elicit any change in vitro; these effects reveal that MIC per se acts as an inhibitor of electron transport and a weak uncoupler. Administration of MIC sc at lethal dose caused a similar change only with NAD+-linked substrates, reflecting impairment of mitochondrial respiration at complex I region and thereby induction of histotoxic hypoxia in vivo.

1. Host-parasite interactions have the potential to influence broadscale ecological and evolutionary processes, levels of endemism, divergence patterns and distributions in host populations. Understanding the mechanisms involved requires identification of the factors that shape parasite distribution and prevalence. 2. A lack of comparative information on community-level host-parasite associations limits our understanding of the role of parasites in host population divergence processes. Avian malaria (haemosporidian) parasites in bird communities offer a tractable model system to examine the potential for pathogens to influence evolutionary processes in natural host populations. 3. Using cytochrome b variation, we characterized phylogenetic diversity and prevalence of two genera of avian haemosporidian parasites, Plasmodium and Haemoproteus, and analysed biogeographic patterns of lineages across islands and avian hosts, in southern Melanesian bird communities to identify factors that explain patterns of infection. 4. Plasmodium spp. displayed isolation-by-distance effects, a significant amount of genetic variation distributed among islands but insignificant amounts among host species and families, and strong local island effects with respect to prevalence. Haemoproteus spp. did not display isolation-by-distance patterns, showed marked structuring of genetic variation among avian host species and families, and significant host species prevalence patterns. 5. These differences suggest that Plasmodium spp. infection patterns were shaped by geography and the abiotic environment, whereas Haemoproteus spp. infection patterns were shaped predominantly by host associations. Heterogeneity in the complement and prevalence of parasite lineages infecting local bird communities likely exposes host species to a mosaic of spatially divergent disease selection pressures across their naturally fragmented distributions in southern Melanesia. Host associations for Haemoproteus spp. indicate a capacity for the formation of locally co-adapted host-parasite relationships, a feature that may limit intraspecific gene flow or range expansions of closely related host species.

Hitherto, electron transfer (ET) between redox proteins has been deemed to occur via donor-acceptor binding, and diffusible reactive species are considered as deleterious side-products in such systems. Herein, ET from cytochrome P450 reductase (CPR, an animal membrane flavoprotein) and horseradish peroxidase (HRP, a plant hemoprotein) to cytochrome c (Cyt c, a soluble animal hemoprotein) was probed under diverse conditions, using standard assays. ET in the CPR-Cyt c system was critically inhibited by cyanide and sub-equivalent levels of polar one-electron cyclers like copper ions, vitamin C/Trolox and superoxide dismutase. In the presence of lipids, inhibition was also afforded by amphipathic molecules vitamin E, palmitoyl-vitamin C and the membrane hemoprotein, cytochrome b(5). Such nonspecific inhibition (by diverse agents in both aqueous and lipid phases) indicated that electron transfer/relay was effected by small diffusible agents, whose lifetimes are shortened by the diverse radical scavengers. When CPR was retained in a dialysis membrane and Cyt c presented outside in free solution, ET was still observed. Further, HRP (taken at nM levels) catalyzed oxidation of a phenolic substrate was significantly inhibited upon the incorporation of sub-nM levels of Cyt c. The findings imply that CPR-Cyt c or HRP-Cyt c binding is not crucial for ET. Further, fundamental quantitative arguments (based on diffusion/collision) challenge the erstwhile protein-protein binding-assisted ET hypothesis. It is proven beyond reasonable doubt that mobile and diffusible electron carriers (ions and radicals) serve as ``redox-relay agents'' in the biological ET models/setup studied.

A rapid and cost effective DNA test is described to identify European eel (Anguilla anguilla) and North American eel (Anguilla rostrata). By means of polymerase chain reaction (PCR) technique parts of the mitochondrial cytochrome b gene are amplified with species specific primers which are designed to produce PCR fragments of different characteristic sizes for European and American eel. The size differences can easily be made visible by agarose gel electrophoresis

In the last years farmed Pangasius (Tra-Pangasius, Pangasius hypophthalmus) from Vietnam has reached a considerable market share, whereas aquaculture of Asian Redtail Catfish (Hemibagrus wyckioides) is in its infancy. Recently it has been detected by food control authorities in Hamburg, that Pangasius fillets have been mislabelled and sold as fillets produced from Asian Redtail catfish. The necessity to improve the analytical methods for differentiation of Pangasius and Redtail Catfish prompted us to evaluate the suitability of isoelectric focusing (IEF) and DNA-analysis for identification of the two species. IEF of water soluble proteins was found to be a fast, reliable and economical method for differentiation of raw fillets of Pangasius and Redtail Catfish, as long as reference material is available. PCR-based DNA analysis was performed as follows: (i) amplification of a 464 bp segment of the cytochrome b gene; (ii) sequencing of the PCR product; (iii) comparison of the sequence with entries in GenBank using BLAST. The sequences of both species differed considerably, allowing the unequivocal differentiation between P. hypophthalmus and H. wyckioides. Kurzfassung Pangasius (Schlankwels, Tra-Pangasius, Pangasius hypophthalmus) hat sich innerhalb weniger Jahre zu einem bedeutenden Zuchtfisch entwickelt, während die Aquakultur des Asiatischen Rotflossenwelses (Hemibagrus wyckioides) in Vietnam noch in einem relativ kleinen Maßstab stattfindet. Kürzlich wurde von der Lebensmittelüberwachung in Hamburg nachgewiesen, dass im Handel erhältliche Filets mit der Deklaration „Rotflossenwels“ aus Pangasius hergestellt worden waren. Vor diesem Hintergrund wurden zwei Methoden auf ihre Eignung zur Differenzierung von Pangasius und Rotflossenwels geprüft. Es zeigte sich, dass sowohl die isoelektrische Fokussierung (IEF) wasserlöslicher Proteine als auch die PCR-basierte DNA-Analyse zur Unterscheidung beider Arten gut geeignet ist. Die IEF stellt eine schnelle und kostengünstige Untersuchungsmethode dar, die allerdings Referenzmaterial benötigt. Mit Hilfe der PCR (Polymerase-Kettenreaktion) wurde ein Abschnitt des Cytochrom b-Gens vervielfältigt und sequenziert. Die Sequenzen von P. hypophthalmus und H. wyckioides wiesen beträchtliche Unterschiede auf. Es wird diskutiert, wie sich durch Vergleich dieser Sequenzen mit Einträgen in Gendatenbanken unbekannte Proben beider Arten sicher zuordnen lassen.

Zusammenfassung Zur Identifizierung der folgenden vier Welsarten bzw. zwei Hybriden (Clarias gariepinus, Pangasius hypophthalmus, Pseudoplatystoma spp., Silurus glanis, Claresse® und Melander®) wurden die isolektrische Fokussierung (IEF) der wasserlöslichen Muskelproteine und die Polymerase-Kettenreaktion (PCR) zur Vervielfältigung und Sequenzierung eines Abschnittes aus dem Cytochrom b – Gen eingesetzt. Die IEF ergab artspezifische Proteinmuster mit hitzestabilen Proteinbanden im anodalen Gelbereich. Der afrikanische Wels (C. gariepinus) und das Hybriderzeugnis Melander® wiesen das gleiche Proteinmuster auf. Mittels DNA-Analyse ließen sich die Welsarten anhand ihrer Cytochrom b Gensequenzen eindeutig identifizieren. Auch hier zeigte der Welshybrid Melander® ein identisches Ergebnis wie der afrikanische Wels. Die Schwierigkeiten der Identifizierung von Tigerwelsen südamerikanischer Herkunft aus der Gattung Pseudoplatystoma werden diskutiert. Abstract Isoelectric focusing (IEF) of water soluble proteins and PCR-based DNA- analysis were used to differentiate between four catfish species (Clarias gariepinus, Pangasius hypophthalmus, Pseudoplatystoma spp., Silurus glanis) and two hybrids Claresse® and Melander®. Specific protein patterns have been obtained for all species and Claresse®, but in case of Melander® the identical pattern was observed as for the African catfish Clarias gariepinus. By sequencing the PCR products and application of BLAST, authenticity of the different catfish samples was confirmed. The cytochrome b gene sequences of Melander® and African catfish were identical. The difficulties of identifying catfishes of the genus Pseudoplatystoma are discussed.

Random Amplified Polymorphic DNA (RAPD) markers and cytochrome b (Cyt-b) gene sequences were utilized to fingerprint and construct phylogenetic relationships among four species of mackerel commonly found in the Straits of Malacca namely Rastrelliger kanagurta, R. brachysoma, Decapterus maruadsi and D. russelli. The UPGMA dendogram and genetic distance clearly showed that the individuals clustered into their own genus and species except for the Decapterus. These results were also supported by partial mtDNA cytochrome b gene sequences (279 bp) which found monotypic sequence for all Decapterus studied. Cytochrome b sequence phylogeny generated through Neighbor Joining (NJ) method was congruent with RAPD data. Results showed clear discrimination between both genera with average nucleotide divergence about 25.43%. This marker also demonstrated R. brachysoma and R. kanagurta as distinct species separated with average nucleotide divergence about 2.76%. However, based on BLAST analysis, this study indicated that the fish initially identified as D. maruadsi was actually D. russelli. The results highlighted the importance of genetic analysis for taxonomic validation, in addition to morphological traits.

O gênero Steno pertence à Ordem Cetartiodactyla, Família Delphinidae, e compreende apenas uma espécie: o golfinho-de-dentes-rugosos, Steno bredanensis. O golfinho-de-dentes-rugosos é encontrado nos Oceanos Atlântico, Pacífico e Índico, em águas profundas tropicais, subtropicais e temperadas quentes. Entretanto, em algumas localidades como as regiões Sudeste e Sul do Brasil, esta espécie é conhecida por apresentar hábitos costeiros, o que a torna suscetível a ameaças antropogênicas como a degradação do hábitat, as capturas acidentais e diversos tipos de poluição. Conhecer a magnitude destes impactos e o grau de diferenciação genética das populações usando marcadores moleculares são aspectos importantes para a conservação da espécie. Os marcadores moleculares são segmentos específicos de DNA que podem ou não fazer parte de um gene e que apresentam grau de polimorfismo adequado para responder questões sobre as relações genéticas de indivíduos, populações ou diferentes espécies. O DNA mitocondrial é um dos marcadores moleculares mais utilizados em estudos sobre estrutura populacional, sistemática e filogenia de cetáceos. Estudos genéticos têm mostrado que várias espécies de delfinídeos apresentam estrutura populacional genética, entre e dentro das bacias oceânicas. No presente estudo foi investigada a diferenciação genética do golfinho-de-dentes-rugosos usando sequências da região controle mitocondrial de várias localidades em todo o mundo (Oceano Pacífico Centro-Sul: N=59; Pacífico Tropical Leste: N= 4; Pacífico Noroeste: N=1; Oceano Índico: N=1; Atlântico - Caribe: N=3; Atlântico Sudoeste: N=44; N total = 112). Análises preliminares indicaram grande diferenciação genética entre os Oceanos Atlântico e Pacífico/Índico (distância p = 0,031), que foram posteriormente investigadas utilizando sequências do citocromo b e mitogenomas completos. As análises filogenéticas de Neighbor-Joining e Bayesianas não foram conclusivas sobre a existência de especiação críptica em Steno. No entanto, a grande diferenciação entre as bacias oceânicas merece uma análise mais aprofundada, utilizando outros marcadores genéticos (por ex., sequências nucleares) bem como dados morfológicos. Não obstante, as análises AMOVA e FST par-a-par revelaram forte diferenciação populacional, não só entre os oceanos Atlântico e Pacífico, mas também no Atlântico, onde foram detectadas três populações: Caribe, região Sudeste e região Sul do Brasil. As populações detectadas no Atlântico Sudoeste devem ser aceitas como Unidades de Manejo (Management Units, MU) e dados demográficos básicos precisam ser levantados para essas MU, a fim de possibilitar uma melhor avaliação dos impactos antrópicos sobre elas. Este estudo fornece a primeira perspectiva sobre a diferenciação genética mundial de S. bredanensis.

To ensure the authentication of fishery products lacking biological characters, rapid species identification methods are required. Two DNA- and protein-based methods, PCR-SSCP (polymerase chain reaction - single strand conformation polymorphism) of a 464 bp segment of the cytochrome b – gene and isoelectric focusing (IEF) of water-soluble proteins from fish fillets, were applied to identify fillets of (sub-) tropical fish species available on the European market. Among the samples analysed were two taxonomically identified species from the family Sciaenidae and one from Sphyraenidae. By comparison of DNA- and protein patterns of different samples, information about intra-species variability of patterns, and homogeneity of batches (e.g. fillet blocks or bags) can be obtained. PCR-SSCP and IEF may be useful for pre-checking of a large number of samples by food control laboratories. Zusammenfassung Zur Sicherstellung der Authentizität von Fischerei-Erzeugnissen ohne biologische Merkmale sind schnelle Verfahren zur Speziesidentifizierung hilfreich. Zwei Methoden der DNA- bzw. Protein-Analyse wurden eingesetzt, um Filets (sub-) tropischer Fischarten, die auf dem europäischen Markt angeboten werden, zu identifizieren. Bei diesen Methoden handelt es sich um die PCR-SSCP (Polymerase-Kettenreaktion – Einzelstrang-Konformationspolymorphismus) – Analyse der PCR-Produkte und die IEF (isoelektrische Fokussierung) der wasserlöslichen Fischmuskelproteine. Unter den untersuchten Proben waren zwei taxonomisch bestimmte Arten aus der Familie Sciaenidae und eine Spezies aus der Familie Sphyraenidae. Durch Vergleich der DNA- bzw. Proteinmuster lassen sich Informationen über die intra-spezifische Variabilität solcher Muster und die Einheitlichkeit von Partien (beispielsweise Filetblöcke oder Filetbeutel) gewinnen. PCR-SSCP und IEF können in Laboratorien der Lebensmittelüberwachung als Vortest gerade bei hohen Probenzahlen sinnvoll eingesetzt werden.

本文由两部分组成,一部分是关于一组新型除草剂(K-15,K-23)的抑制特性及作用位点的研究;另一部分是关于碳酸氢盐对细胞色素b-559高电势的保护作用的研究。 在第一部分,我们首先研究了抑制剂K-23对Psn放氧活性、DCIP光还原和可变荧光等光合特性的影响。研究发现,K-23在低浓度时刺激放氧活性,而在相对高浓度时抑制放氧活性。但是,K-23在低浓度时却有效地抑制了可变荧光。这些数据表明了新型抑制剂的抑制反应是基于氧化还原作用而不是猝灭作用。此外,通过采用胰蛋白酶消化类囊体膜的方式初步检测了新型抑制剂的作用部位,其结果表明:虽然新型抑制剂抑制受体侧电子传递,但它的抑制部位与DCMU不同。 其次,研究了新型抑制剂对光系统II色素蛋白复合体与多肽组分的影响及抑制剂的键合蛋白。应用SDS-PAGE技术,发现新型抑制剂主要作用于光系统II的反应中心蛋白。用温和的Deriphat-PAGE分析也证实了新型抑制剂作用于核心复合物,导致核心复合物二聚体消失。 进一步用SDS-PAGE分析新型抑制剂对Psn多肽组分的影响,发现新型抑制剂主要影响D.、D2、CP43和CP47。用荧光发射的方法确定了K-15键合在D2蛋白上。 最后,结合荧光动力学和HPLC方法,分别从定性和定量的角度,以核心复合物以及抑制剂存在下从BBY中分离的核心复合物为研究对象,详细研究了抑制剂对QA的取代作用。研究发现,在无去垢剂或低浓度去垢剂存在情况下,由于不能创造出适合于QA存在的疏水环境,我们没有得到QA被K-15取代的实验证据。而在抑制剂K-15存在下,用2.2% HTG从BBY分离的核心复合物的实验中,检测不到正的可变荧光Fv,而是得到了降低的FM,这个结果表明QA已被抑制剂在它的作用位点处所取代。 在第二部分,研究了pH5.0—7.0范围内碳酸氢盐对Cyt b-559氧化还原状态转变的影响。首先研究了pH5.0~7.0条件下碳酸氢盐对PSII Cyt b-559还原减氧化差异吸收光谱的影响,发现铁氰化钾还原的PSII Cyt b-559 HP的含量随介质pH值的降低而减少。然而,碳酸氢盐的加入阻止了由于介质pH降低而引起的Cyt b-559由高电势向低电势的转化。比如,当样品温育在pH5.0的介质中,铁氰化钾还原的Cyt b-559 HP含量占总量的25%-30%,当介质中加入2mmol/L碳酸氢盐后Cyt b-559 HP的含量上升,占总量的50%-56%。碳酸氢盐效应对氢醌还原的Cyt b-559HP含量的影响尤为显著。pH6.5时碳酸氢盐对Cyt b-559的还原氧化状态的影响不显著。其次,分别研究了PSII经Tris处理去除锰簇和三个外周蛋白及NH20H处理去除锰簇和17 kDa和23 kDa后,碳酸氢盐对Cytb一559 HP影响的pH依赖值,发现不论在pH5.0还是pH6.5的介质中碳酸氢盐效应都不存在。 综合以上实验结果,我们认为碳酸氢盐对酸化引起的Cyt b-559氧化还原状态的影响与它和锰的作用有关,但也不能排除钙离子与碳酸氢盐之间的协同作用。
