The molar single ion activity coefficient (y(F)) of fluoride ions was determined at 25 degrees C and ionic strengths between 0.100 and 3.00 mol L(-1) NaClO(4) using an ion-selective electrode. The activity coefficient dependency on ionic strength was determined to be Phi(F) = log y(F) = 0.2315I-0.041I(2). The function Phi(F)(I), combined with functions obtained in previous work for copper (Phi(Cu)) and hydrogen (Phi(H)), allowed us to make the estimation of the stoichiometric and thermodynamic protonation constants of some halides and pseudo-halides as well as the formation constants of some pseudo-halides and fluoride 1:1 bivalent cation complexes. The calculation procedure proposed in this paper is consistent with critically-selected experimental data. It was demonstrated that it is possible to use Phi(F)(I) for predicting the thermodynamic equilibrium parameters independently of Pearson's hardness of acids and bases.

The speciose Brazilian Elateridae fauna is characterized by high karyotypic diversity, including one species (Chalcolepidius zonatus Eschscholtz, 1829) with the lowest diploid number within any Coleoptera order. Cytogenetic analysis of Conoderus dimidiatus Germar, 1839, C. scalaris (Germar, 1824,) C. ternarius Germar, 1839, and C. stigmosus Germar, 1839 by standard and differential staining was performed with the aim of establishing mechanisms of karyotypic differentiation in these species. Conoderus dimidiatus, C. scalaris, and C. ternarius have diploid numbers of 2n(male) = 17 and 2n(female) = 18, and a X0/XX sex determination system, similar to that encountered in the majority of Conoderini species. The karyotype of C. stigmosus was characterized by a diploid number of 2n=16 and a neoXY/neoXX sex determination system that was highly differentiated from other species of the genus. Some features of the mitotic and meiotic chromosomes suggest an autosome/ancestral X chromosome fusion as the cause of the neoXY system origin in C. stigmosus. C-banding and silver impregnation techniques showed that the four Conoderus species possess similar chromosomal characteristics to those registered in most Polyphaga species, including pericentromeric C band and autosomal NORs. Triple staining techniques including CMA(3)/DA/DAPI also provided useful information for differentiating these Conoderus species. These techniques revealed unique GC-rich heterochromatin associated with NORs in C. scalaris and C. stigmosus and CMA(3)-heteromorphism in C. scalaris and C. ternarius.

The first cytogenetic analysis of fireflies from Brazilian fauna was carried out in this work. The investigation of two species of the subfamily Lampyrinae, Aspisoma maculatum and Photinus sp. (aff. pyralis), showed the diploid number 2n = 19 and an X0 sex determination system in males. These observations are similar to those already described for all the Lampyrinae species previously studied. In contrast, Bicellonycha lividipennis (Photurinae) revealed the karyotype 2n = 16 + neoXY, which has not yet been registered for any firefly species. The neoXY sex determination system encountered in this species probably arose through fusion between an ancestral X sex chromosome, belonging to the X0 system, and an autosomal element. This event also reduced the diploid number from 2n = 19, which is more frequent in the family Lampyridae, to 2n = 18 in B. lividipennis. The analysis of meiotic cells showed that the neoXY sexual bivalent of B. lividipennis exhibited a prominent terminal chiasma, indicating that the sex chromosomes are not wholly differentiated and still retain a region of homology. A review of the cytogenetic data known for the family Lampyridae was also documented in this work, as well as a discussion on the main trends of chromosomal evolution that seem to have occurred in this group.

Synaptonemal complexes were analysed by electron microscopy in 2 bucks heterozygous for the 5/15 Robertsonian translocation. The cis configuration (free homologous 5 and 15 chromosomes on the same side of the 5/15 translocated chromosome) was found in all 50 cells examined. This feature is considered a prerequisite for the development of balanced gametes. No association between the sex bivalent and trivalent was observed.

Solid-state M-4-Cl-BP compounds, where M stands for bivalent Mg, Ca, Sr, Ba and 4-Cl-BP is 4-chlorobenzylidenepyruvate, have been synthesized. Simultaneous thermogravimetry-differential thermal analysis (TG-DTA), differential scanning calorimetry (DSC), infrared spectroscopy, elemental analysis and complexometry were used to characterize and to study the thermal behaviour of these compounds. The results led to informations about the composition, dehydration, thermal stability and thermal decomposition of the isolated complexes.

This work describes the first report about the occurrence of recombination nodules (RNs) in spread pachytene cells of two species of Coleoptera: Palembus dermestoides (Tenebrionidae) and Epicauta atomaria (Meloidae). The RNs were observed in preparations contrasted with phosphotungstic acid. Considering RN morphology and its occurrence in pachytene bivalents (one per autosome bivalent) these structures were interpreted to be late RNs. P. dermestoides and E. atolraria have 2n = 20 chromosomes including an Xyp sex determination system. In spite of most frequently subtelocentric morphology observed in the autosomes of both species, the occurrence of RNs is limited only to the synaptonemal complex (SC) structure of the long arms. These findings are in agreement with those obtained using light microscopy analysis in which only one chiasma or terminalization event is observed per autosomal bivalent in early or late metaphase I cells. The RNs have the same average width of the SC of each analyzed species, a circular shape, strong electron density, and are observed mainly between the lateral elements of the SC. The RNs of P. dermestoides and E. atomaria have approximately the same average size (width), 180 +/- 20 nm and 160 +/- 80 nm, respectively. The absence of RNs in the short arms and its occurrence in the long arms are discussed considering the short arm pericentromeric and pro-centric heterochromatin. Copyright (C) 2003 S. Currently, 60 species of harvestmen have been karyotyped and all of these are from the Nearctic and Palearctic regions. This work is the first cytogenetic report of three gonyleptid species of the suborder Laniatores: Goniosoina aff. badiuln, G. proxiinuni and G. spelaeum of the Neotropical region, from the southeastern region of Brazil. Conventional Giemsa stain chromosome preparations were obtained from embryonic cells and adult male testes. Embryo mitotic plates of G. aff. badium and G. proximum indicated 88 chromosomes, and mitotic spermatogonial plates of G. spelaeum males revealed intra- and interindividual variation of chromosome number, ranging from 92-109 chromosomes. In the three analyzed species, the mitotic chromosomes were meta- or subinetacentric with no obvious sex chromosomes being identified during mitosis. Prophase I spermatocytes of G. spelaeum also revealed intra- and interindividual bivalent number variation and furthermore indicated the presence of multivalence. The karyotypes of these three Goniosoina species exhibited the largest chromosome pair with a negative heteropycnosis in the distal region of the shortest arrn chromosomes of G. spelaeum submitted to silver impregnation evidenced this negative heteropycnotic region as nucleolus organizer region (NOR). These results, when compared with cytogenetic data of other Laniatores species from the Palearctic region, indicated that a new record for diploid chromosome number probably characterize the genus Goniosoma in the Neotropical region.
SYNAPTONEMAL COMPLEX-ANALYSIS IN SPERMATOCYTES OF TILAPIA, OREOCHROMIS-NILOTICUS (PISCES, CICHLIDAE)

Some adaptations of the synaptonemal complex (SC) whole-mounting technique first used in plants permitted its application to meiotic studies in tilapia, Oreochromis niloticus. Direct observation of the chromosome pairing process and bivalent structure during the meiotic prophase of this fish species by light and electron microscopy permitted the analysis of SCs in autosomes and the possible identification of sex chromosomes. The analysis of SCs in spermatocytes of 0. niloticus revealed that all 22 bivalent chromosomes completely paired, except for the occurrence of a size heteromorphism in the terminal region of the largest bivalent associated with the presence of an incompletely paired segment during the synapsis process, which may be the cytological visualization of an XX/XY sex chromosome system in this species.

As part of a program to understand the genetics of Amazonian ornamental fish, classical cytogenetics was used to analyze Symphysodon aequifasciatus, S. discus and S. haraldi, popular and expensive aquarium fishes that are endemic to the Amazon basin. Mitotic analyses in Symphysodon have shown some odd patterns compared with other Neotropical cichlids. We have confirmed that Symphysodon species are characterized by chromosomal diversity and meiotic complexity despite the fact that species share the same diploid number 2n = 60. An intriguing meiotic chromosomal chain, with up to 20 elements during diplotene/diakinesis, was observed in S. aequifasciatus and S. haraldi, whereas S. discus only contains typical bivalent chromosomes. Such chromosomal chains with a high number of elements have not been observed in any other vertebrates. We showed that the meiotic chromosomal chain was not sex related. This observation is unusual and we propose that the origin of meiotic multiples in males and females is based on a series of translocations that involved heterochromatic regions after hybridization of ancestor wild Discus species. Meiotic cells of triploid male rainbow trouts were analyzed by a surface-spreading SC technique in order to show the process of chromosome synapsis. At zygotene the formation of SCs involved, almost exclusively, two sets of lateral elements (LEs), and the remaining set of LEs presented different synaptic configurations involving one, two, three, or four LEs. The absence of SCs involving more than two LEs from mid- to late pachytene indicates that the multivalents produced by synapsis involving more than two LEs, are eliminated before the end of pachytene; thus, the mechanism of chromosome pairing in triploid male rainbow trouts produces, almost exclusively, bivalents with a probable extensive nonhomologous synapsis involving the extra set of chromosomes.

The purpose of this paper is to characterize the lability/inertness metal fractions complexed by aquatic humic substances (HS) in relation to pH, complexation time, and HS concentration. HS were preconcentrated by ultrafiltration and complexed with bivalent metal ions. These fractions were characterized by ion exchange with the chelating collector cellulose Hyphan by applying batch procedure. The metals were determined by atomic absorption spectrometry. The results show that the distribution coefficients, Kd, decreased with HS presence, and that the relative lability of metal fractions complexed by HS is dependent on variables such as pH, complexation time, and HS concentration. Until c.a. 15 min, the metal change between aquatic HS and ion exchanger occurs following a 2 order reaction. Afterwards, the remaining metal fraction in the HS reacts following a 1st order reaction. For traces of metal ions bound to dissolved HS, the lability orderPb > Mn > Cd, Ni > Cu is revealed. ©1997 Soc. Bras. Química.

Meiotic analysis performed on a sample of 10 specimens of Prochilodus lineatus revealed continuous filaments of different sizes stained by AgNO3, corresponding to the bivalent of the normal complement. Small supernumerary chromosomes were observed as isolated and well stained bodies, scattered among the other elements. Synaptonemal complex studies have shown that the beginning of chromosome pairing process in P. lineatus usually occurs from the telomeres to the pericentromeric region. At the end of the pachytene 27 bivalents are perfectly paired and the small supernumerary chromosomes of this species are seen as bivalents, trivalents, or tetravalents. The central region of these small chromosomes show a trick staining when they formed bivalents or tetravalents. This portion seems to correspond to the pericentromeric region of the regular chromosomes with the heterochromatic characteristic of B chromosomes.

A synaptonemal complex (SC) study of specimens of Nellore and Gyr breeds of Bos taurus indicus was performed with the main objective to identify and determinate the frequency of abnormalities of SC and the frequency of cells with abnormalities. All animals analyzed had 29 autosomal bivalents and one sexual bivalent. The Nellore breed had 30.00% of cells with SC abnormalities while the Gyr breed had only 11.11%. Statistical analyses showed that there were not significant differences for the number of cells with abnormalities among the breeds studied. The subspecies Bos taurus indicus had 16.92% of cells showing abnormalities, being 62.82% of these abnormalities in zygotene and 37.18% in pachytene. Some aspects regarding the frequency of cells with abnormalities and the fertility of Nellore and Gyr breeds are discussed.
