The cysteine cathepsins are a family of closely related thiol proteases, normally found in the endosomal and lysosomal compartments of cells. A growing body of evidence has clearly linked the dysregulated activity of these proteases with many diseases and pathological conditions, offering therapeutic, prognostic and diagnostic potential. However, these proteases are synthesised as inactive precursors and once activated, are controlled by factors such as pH and presence of endogenous inhibitors, meaning that overall protein and activity levels do not necessarily correlate. In order to fully appreciate the role and potential of these proteases, tools are required that can detect and quantify overall cathepsin activity. Two main strategies have evolved; synthetic substrates and protease-labelling with affinity-binding probes (or activity-based probes). This review examines recent innovations in these approaches as the field moves towards developing tools that could ultimately be used in patients for diagnostic or prognostic applications.

For physicians facing patients with organ-limited metastases from colorectal cancer, tumor shrinkage and sterilization of micrometastatic disease is the main goal, giving the opportunity for secondary surgical resection. At the same time, for the majority of patients who will not achieve a sufficient tumor response, disease control remains the predominant objective. Since FOLFOX or FOLFIRI have similar efficacies, the challenge is to define which could be the most effective targeted agent (anti-EGFR or anti-VEGF) to reach these goals. Therefore, a priori molecular identification of patients that could benefit from anti-EGFR or anti-VEGF monoclonal antibodies (i.e. the currently approved targeted therapies for metastatic colorectal cancer) is of critical importance. In this setting, the KRAS mutation status was the first identified predictive marker of response to anti-EGFR therapy. Since it has been demonstrated that tumors with KRAS mutation do not respond to anti-EGFR therapy, KRAS status must be determined prior to treatment. Thus, for KRAS wild-type patients, the choices that remain are either anti-VEGF or anti-EGFR. In this review, we present the most updated data from translational research programs dealing with the identification of biomarkers for response to targeted therapies.

Este trabalho pretende abordar a importância de um estudo geomecânico no apoio à otimização e estabilidade de explorações subterrâneas por subníveis, com criação de bancadas e posterior enchimento. O presente envolveu um estudo geológico-geotécnico em quinze galerias situadas a muro, teto e corpo da mineralização com o levantamento das características mais relevantes do maciço rochoso para aplicação das classificações geomecânicas, englobando uma amostragem de mais de 1780 descontinuidades, obtendo um modelo cartográfico subterrâneo com um panorama geral da qualidade do maciço rochoso intercetado pelas escavações nas diferentes zonas. Os dados dos levantamentos de campo levaram à criação de uma base de dados com a aplicação das classificações geomecânicas Q-System, RMR e GSI, por galeria e, em seguida, por zona, com proposta de classe de sustimento a aplicar em cada local, pelo ábaco de Barton, em conjunto com a determinação de parâmetros geomecânicos fundamentais ao refinamento do conhecimento geológico-geotécnico das unidades litológicas em estudo. Na parte final, focando a localização da massa mineralizada de Feitais é efetuada uma abordagem relativa à estabilidade das cavidades geradas pelo desmonte em bancada entre subníveis, com respetivo dimensionamento das larguras admissíveis, em condições de segurança, através da relação entre o número de estabilidade e raio hidráulico, pelo método do gráfico de estabilidade. Com esta metodologia de caracterização geológico geotécnica, é pretendido efetuar um ponto de partida à criação de um modelo geomecânico comportamental do jazigo de Feitais, Mina de Aljustrel, contando com um processo inicial de apoio ao planeamento mineiro aplicado ao método de desmonte em bancada e posterior enchimento por subníveis, atuando nos parâmetros de estabilidade e apoio à extração, favorecendo assim a segurança das operações de trabalho em conjunto com um apoio de otimização da extração.

This unhappy looking young girl is featured in an early black and white tintype photograph. The date, location and name of the photographer are unknown. This small tintype was in the possession of Iris Sloman Bell, of St. Catharines, Ontario. The Sloman - Bell families have descendants who are former slaves from the United States."Tintypes were the invention of Prof. Hamilton Smith of Ohio. They begin as thin sheets of iron, covered with a layer of black paint. This serves as the base for the same iodized collodion coating and silver nitrate bath used in the ambrotype process. First made in 1856, millions were produced well into the twentieth century. When tintypes were finished in the same sorts of mats and cases used for ambrotypes, it can be almost impossible to distinguish which process was used without removing the image to examine the substrate." The relationships between vine water status, soil texture, and vine size were observed in four Niagara, Ontario Pinot noir vineyards in 2008 and 2009. The vineyards were divided into water status zones using geographic information systems (GIS) software to map the seasonal mean midday leaf water potential (,P), and dormant pruning shoot weights following the 2008 season. Fruit was harvested from all sentinel vines, bulked by water status zones and made into wine. Sensory analysis included a multidimensional sorting (MDS) task and descriptive analysis (DA) of the 2008 wines. Airborne multispectral images, with a spatial resolution of 38 cm, were captured four times in 2008 and three times in 2009, with the final flights around veraison. A semi-automatic process was developed to extract NDVI from the images, and a masking procedure was identified to create a vine-only NDVI image. 2008 and 2009 were cooler and wetter than mean years, and the range of water status zones was narrow. Yield per vine, vine size, anthocyanins and phenols were the least consistent variables. Divided by water status or vine size, there were no variables with differences between zones in all four vineyards in either year. Wines were not different between water status zones in any chemical analysis, and HPLC revealed that there were no differences in individual anthocyanins or phenolic compounds between water status zones within the vineyard sites. There were some notable correlations between vineyard and grape composition variables, and spatial trends were observed to be qualitatively related for many of the variables. The MDS task revealed that wines from each vineyard were more affected by random fermentation effects than water status effects. This was confirmed by the DA; there were no differences between wines from the water status zones within vineyard sites for any attribute. Remotely sensed NDVI (normalized difference vegetation index) correlated reasonably well with a number of grape composition variables, as well as soil type. Resampling to a lower spatial resolution did not appreciably affect the strength of correlations, and corresponded to the information contained in the masked images, while maintaining the range of values of NDVI. This study showed that in cool climates, there is the potential for using precision viticulture techniques to understand the variability in vineyards, but the variable weather presents a challenge for understanding the driving forces of that variability.

Daily sunshine duration is commonly reported at weather stations. Beyond the basic duration report, more information is available from scorched cards of Campbell-Stokes sunshine recorders, such as the estimation of direct-beam solar irradiance. Sunshine cards therefore potentially provide information on sky state, as inferred from solar-radiation data. Some sites have been operational since the late 19th century, hence sunshine cards potentially provide underexploited historical data on sky state. Sunshine cards provide an example of an archive source yielding data beyond the measurements originally sought.

Dictionary compilers and designers use punctuation to structure and clarify entries and to encode information. Dictionaries with a relatively simple structure can have simple typography, and simple punctuation; as dictionaries grew more complex, and encountered the space constraints of the printed page, complex encoding systems were developed, using punctuation and symbols. Two recent trends have emerged in dictionary design: to eliminate punctuation, and sometimes to use a larger number of fonts, so that the boundaries between elements are indicated by font change, not punctuation.

Patients with mental health difficulties do not always receive appropriate and recommended psychological treatment for their difficulties, and clinicians are not always appropriately trained to deliver them. This paper considers why this might be the case and provides an overview of the Charlie Waller Institute, a not-for-profit organisation funded by the NHS, University of Reading, and the Charlie Waller Memorial Trust. The Institute seeks to address this problem by training clinicians in wide variety of evidence-based therapies and assessing the impact of this training on clinician knowledge and skill.

While the registrability of scents as Community trade marks has become source of much controversy, the possibility of trademarking scents represents a great potential for the industry. In the aftermath of the Sieckmann case, which has raised the threshold of registrability for scent marks, companies have refrained from submitting new smell-mark applications. Despite the difficulties in registering scents as trademarks, however, it is not impossible to meet the Sieckmann criteria and file successful scent mark applications. This article explains how this could be possible; it reviews all objections in registering scents as trademarks and brings new light into this topic by way of a comprehensive analysis of the conditions under which scents can be registered as Community trade marks.
